Resumo

ABSTRACT: Allergic dogs are commonly sensitized to food allergens. We evaluated the use of the prick test to diagnose food allergies in dogs with pruritus and the efficacy of an exclusion diet based on the test. The prick test was performed in 10 healthy dogs and 34 dogs with pruritus, of which 25 received an exclusion diet for 60 days and was challenged with positive food on the test. pVAS and CADESI-4 were assessed on days 0, 30, and 60 after re-exposure. As a result, two control group dogs reacted to a single food allergen, milk and wheat. Of the 25 dogs with pruritus that reacted to food allergens, 24 (96%) reacted to more than one food allergen, and only one (4%) reacted to a single food protein: pork. In the test group (n=25), there was a significant improvement of pVAS and CADESI-4 after 30 and 60 days of dietary exclusion, with significant worsening of the scores with food allergen challenge. In conclusion, the prick test can be used for screening food allergens to make an exclusion diet.


RESUMO: Cães alérgicos são comumente sensibilizados a alérgenos alimentares. Nós avaliamos o uso do teste de puntura para o diagnóstico de alergias alimentares em cães com prurido e a eficácia de uma dieta de exclusão baseada no teste. O teste de puntura foi realizado em 10 cães saudáveis e em 34 cães com prurido, dos quais 25 receberam uma dieta de exclusão por 60 dias e depois desafiados com o alimento que foi positivo no teste. pVAS e CADESI-4 foram realizados nos dias 0, 30, 60 e depois da reexposição. Como resultados, dois cães do grupo controle reagiram a um único alérgeno alimentar, leite e trigo. Dos 25 cães com prurido que reagiram aos alérgenos alimentares, 24 (96%) reagiram a mais de um alérgeno alimentar e apenas um (4%) reagiu a uma única proteína, que foi porco. No grupo teste (n=25), houve uma significativa melhora do pVAS e CADESI-4 depois de 30 e 60 dias de dieta de exclusão, com significante piora dos scores com o desafio alimentar. Como conclusão, o teste de puntura pode ser usado para selecionar alérgenos alimentares para fazer uma dieta de exclusão.

A correria do dia-a-dia tem exigido cada vez mais agilidade e praticidade, o que tem resultado no aumento do consumo de alimentos industrializados e redução dos alimentos in natura. Além dos diversos problemas de saúde resultantes do excesso do consumo dos alimentos ultraprocessados, existe uma outra preocupação que envolve esse tipo de alimento, que é saber o que realmente está sendo oferecido naquele produto, visto que, na sua maioria, são originados a partir de vários outros ingredientes, e muitos deles podem conter os alergênicos, os quais, por sua vez resultam em quadro de alergia alimentar, graves ou não, mas já interferem na saúde do indivíduo. A preocupação com os consumidores alérgicos que fazem uso de alimentos industrializados, estimulou a criação de uma resolução, que é aplicada a alimentos, exclusivamente para o processo industrial, incluindo bebidas, ingredientes, aditivos e coadjuvantes que causam alergias. O presente trabalho refere-se a um estudo transversal com o objetivo de analisar e avaliar, por meio de uma lista de checagem, os rótulos de alimentos industrializados que continham, na sua lista de ingredientes, um ou mais alergênicos e apurar se os mesmos estão obedecendo às legislações vigentes. Foram selecionados 301 rótulos em variados mercados nas cidades de Lauro de Freitas e Salvador- BA. Constatou-se que não havia conformidade em 27% da rotulagem presente nesse estudo, nas quais as informações são apresentadas de forma deficiente, mesmo após sete anos da criação das normas para esse tipo de produto, o que reforça a necessidade de uma maior fiscalização dos órgãos responsáveis.(AU)


The day-to-day rush has demanded more and more agility and practicality, which has resulted in an increase in the consumption of processed foods and a reduction in in natura foods. In addition to the various health problems resulting from the excessive consumption of ultra-processed foods, there is another concern that involves this type of food, which is to know what is actually being offered in that product, since, for the most part, they originate from several other ingredients, and in many of them they can contain allergens that in turn result in food allergy, serious or not, but already interfere with the safety of the individual's health. The concern with allergic consumers who use processed foods has stimulated the creation of a resolution, which is applied to foods exclusively for the industrial process, including beverages, ingredients, additives and adjuvants that cause food allergies. The present work refers to a cross-sectional study with the objective of analyzing and evaluating, through a checklist, the labels of industrialized foods that contained one or more allergens in their list of ingredients and to determine if they are complying with the current legislation. 301 labels were selected in various markets in the cities of Lauro de Freitas and Salvador. It was found that there was no compliance in 27% of the labeling present in this study, in which the information is presented in a deficient way, even after six years of the creation of the norms for this type of product, which reinforces the need for greater inspection of Organs responsible agency.(AU)

Resumo

Abstract Plant-derived products can assist in the healing process of dermal wounds. It has been demonstrated that Hancornia speciosa latex present angiogenic, osteogenic, anti-inflammatory, and antioxidant activities. Then, it could contribute to the wound healing process. However, natural products in contact with skin may cause dermatitis. The objective of this work was to evaluate the allergic and irritant potential of H. speciosa serum fraction latex using in vitro assays. The obtained results showed that the H. speciosa serum fraction latex has a slightly irritant potential and is not cytotoxic neither allergenic for human cells. Moreover, we identified a remarkable low amount of proteins in this material in comparison to Hevea brasiliensis latex. This result could explain the non-allergenic potential of H. speciosa serum fraction latex because proteins present in latex are the main responsible for allergy. This biomaterial could be used as a non-allergenic source for development of new medicines.


Resumo Produtos derivados de plantas podem auxiliar no processo de cicatrização de feridas cutâneas. Foi demonstrado que o látex de Hancornia speciosa apresenta atividades angiogênicas, osteogênicas, antiinflamatórias e antioxidantes. Então, este biomaterial pode contribuir para o processo de cicatrização de feridas. No entanto, produtos naturais em contato com a pele podem causar dermatites. O objetivo deste trabalho foi avaliar o potencial alérgico e irritante do látex da fração soro de H. speciosa por meio de ensaios in vitro. Os resultados obtidos mostraram que o látex da fração do soro de H. speciosa possui um potencial pouco irritante e não é citotóxico nem alergênico para células humanas. Além disso, foi identificado uma notável baixa quantidade de proteínas neste material em comparação ao látex de Hevea brasiliensis. Esse resultado poderia explicar o potencial não alergênico do látex da fração soro de H. speciosa, pois as proteínas presentes no látex são as principais responsáveis ​​pela alergia. Este biomaterial pode ser utilizado como fonte não alergênica para desenvolvimento de novos medicamentos.

Resumo

Canine atopic dermatitis is an inflammatory, genetic, pruritic and chronic dermatosis that affects between 10 and 30% of dogs and one of the most important allergens is grass pollen. The objective of this study was to evaluate the sensitization to grass pollen allergens in dogs with canine atopic dermatitis and to compare intradermal skin test (IDT) with percutaneous test (PT). For this study, ten healthy dogs and 39 dogs with atopic dermatitis were tested. Dogs were submitted to IDT and PT for Lolium multiflorum, Cynodon dactylon and Paspalum notatum. The IDT and PT tests were compared using the Proportion Test. All healthy dogs were negative to both tests. Ten atopic dogs (25.6%) responded positively to the PT and none were positive in IDT. C. dactylon, L. multiflorum and P. notatum were responsible for positive reactions in 70%, 70% and 30% of positive dogs, respectively. The number of positive reactions in PT were statistically higher than IDT (P<0.05). In conclusion, grass pollen can be important source of allergens for dogs in Paraná state (Brazil) and the PT showed higher sensitization to grass pollen in dogs with atopic dermatitis than IDT.(AU)


A dermatite atópica canina é uma dermatose inflamatória, genética, prurítica e crônica que afeta entre 10% e 30% dos cães, e um dos alérgenos mais importantes são os polens de gramíneas. O objetivo deste estudo é avaliar a sensibilização a alérgenos de polens de gramíneas em cães com dermatite atópica e comparar o teste intradérmico (TID) com o teste percutâneo (TP). Para o estudo, 10 cães hígidos e 39 cães com dermatite atópica foram testados. Estes foram submetidos ao TID e ao TP para Lolium multiflorum, Cynodon dactylon e Paspalum notatum. TID e TP foram comparados usando-se o teste de proporção. Todos os cães hígidos foram negativos em ambos os testes. Dez cães atópicos (25,6%) responderam positivamente ao TP e nenhum ao TID. C. dactylon, L. multiflorum e P. notatum foram responsáveis por reações positivas de 70%, 70% e 30% dos cães positivos, respectivamente. O número de reações positivas no TP foi estatisticamente maior que no TID (P<0,05). Foi concluído que os polens de gramíneas podem ser importantes fontes de alérgenos para cães no estado do Paraná (Brasil) e que o TP mostrou maior sensibilização a polens em cães com dermatite atópica que o TID.(AU)

Accidents caused by the bites of brown spiders (Loxosceles) generate a clinical condition that often includes a threatening necrotic skin lesion near the bite site along with a remarkable inflammatory response. Systemic disorders such as hemolysis, thrombocytopenia, and acute renal failure may occur, but are much less frequent than the local damage. It is already known that phospholipases D, highly expressed toxins in Loxosceles venom, can induce most of these injuries. However, this spider venom has a great range of toxins that probably act synergistically to enhance toxicity. The other protein classes remain poorly explored due to the difficulty in obtaining sufficient amounts of them for a thorough investigation. They include astacins (metalloproteases), serine proteases, knottins, translationally controlled tumor proteins (TCTP), hyaluronidases, allergens and serpins. It has already been shown that some of them, according to their characteristics, may participate to some extent in the development of loxoscelism. In addition, all of these toxins present potential application in several areas. The present review article summarizes information regarding some functional aspects of the protein classes listed above, discusses the directions that could be taken to materialize a comprehensive investigation on each of these toxins as well as highlights the importance of exploring the full venom repertoire.(AU)

Background: Canine atopic dermatitis (CAD) is a chronic and inflammatory disease present in veterinary dermatologicalpractice. The inflammation in CAD is triggered by environmental allergens and skin microorganisms, which are responsiblefor the worsening of cutaneous lesions. This continuous activation of skin inflammatory process can induce the productionof free radicals that also contribute to cellular damage and ultimately leads to changes in blood parameters in dogs withCAD. Although there are reports of inflammatory parameters in CAD, there are a lack of studies correlating skin lesions,blood leukocytes and oxidative stress. Based on that, this study aimed to evaluate the integumentary and systemic inflammatory response in dogs with atopic dermatitis.Materials, Methods & Results: Dogs with confirmed diagnosis of canine atopic dermatitis (n = 10) were divided in twogroups according to CADESI-IV: AI, with CADESI between 0-10, AII, with CADESI between 10-34, and control group (n= 5). Blood-biochemical and histological analysis were performed to access systemic and cutaneous inflammatory response.AII group tended to higher neutrophil and eosinophil counts, as well as neutrophil/lymphocyte ratio (NLR) when comparedto AI. The albumin was lower in AII compared to AI and control (P < 0.05), while total bilirubin and malondialdehyde(MDA) did not differ between groups. NLR (r = 0.64 and P = 0.04) and MDA (r = 0.54 and P = 0.1) were positively correlated with CADESI, while albumin was negatively correlated with CADESI (r = -0.79 and P = 0.005). Histopathologicalanalysis revealed a larger number of neutrophils, macrophages and mast cells in AI and AII than in control group (P < 0.05).Discussion: In this study it was possible to evaluate the systemic and cutaneous leukocyte dynamics in CAD. The relationship between slow loris (Nycticebus spp.) venom (BGE protein) and the major cat allergen (Fel d 1) from domestic cat (Felis catus) is known for about two decades. Along this time, evidence was accumulated regarding convergences between them, including their almost identical mode of action. Methods: Large-scale database mining for Fel d 1 and BGE proteins in Felidae and Nycticebus spp., alignment, phylogeny proposition and molecular modelling, associated with directed literature review were assessed. Results: Fel d 1 sequences for 28 non-domestic felids were identified, along with two additional loris BGE protein sequences. Dimer interfaces are less conserved among sequences, and the chain 1 shows more sequence similarity than chain 2. Post-translational modification similarities are highly probable. Conclusions: Fel d 1 functions beyond allergy are discussed, considering the great conservation of felid orthologs of this protein. Reasons for toxicity being found only in domestic cats are proposed in the context of domestication. The combination of the literature review, genome-derived sequence data, and comparisons with the venomous primate slow loris may point to domestic cats as potentially poisonous mammals.(AU)

Conglycinin from soybean has been recognized as one of the major feed allergens. This study investigated the effects of -conglycinin-induced allergic sensitization on chicks small intestines. A total of 40 7-day-old (100 g) chicks were divided into four groups as control, -conglycinin 1 h, -conglycinin 6 h, and -conglycinin 12 h. All treatment groups were administered 60 mg of -conglycinin/chick and small intestine samples were collected. -conglycinin-induced allergic sensitization marginally damages the epithelium lining of the duodenum villi and, in addition, significantly increases the accumulation of mast cells in the lamina propria and crypt of the duodenum. Moreover, the TNF- level significantly increased in all -conglycinin groups. IL-8 and IL-2 were significantly downregulated in the 1 h group; however, there were increases for the 6 h and 12 h groups. These results suggest that -conglycinin may lead to an inflammatory response in the chicks small intestines.(AU)

Background:The feline atopic syndrome (FAS) associated to environmental allergens is the third most common allergic dermatosis in domestic cats. In general, clinical signs are not pathognomonic and the exclusion of other pruritus causes is necessary to reach the diagnosis of FAS. The treatment is based on the use of drugs to control the pruritus, such as glucocorticoids, cyclosporine and, recently, oclacitinib, a Janus kinase inhibitor. This study aimed to report the efficacy of oclacitinib on the treatment of FAS associated to environmental allergens. Case:A 10-year-old female feline, crossbred, presented a history of pruritic dermatitis during ten months and diarrhea. The animal had been submitted to treatment for ectoparasites with pour-on selamectin and was fed with a commercial hypoallergenic diet in the last eight weeks or so. However, no improvement on the skin condition was observed. Physical examination revealed disseminated furfuraceous desquamation, excoriation and erythema on the right supraorbital region. Bilateral conjunctivitis was also observed. Complete blood cell count, biochemistry profile, urinalysis, immunochroma-thographic test for feline immunodeffiency virus (FIV) and feline leukemia virus (FeLV), fungic culture and abdominal ultrasonography were requested. The abnormalities observed were reduced urinary density and discrete loss of renal corticomedullary differentiation. Thus, based on physical examination and complementary exams, the animal was diag-nosis with FAS, since the main other causes of pruritus (hypersensitivity to ectoparasites and alimentary allergens) were excluded. The animal was also diagnosed with stage 1 chronic kidney disease. Therapy based on oclacitinib was instituted with an induction dose of 1 mg/kg twice daily for 14 days, followed by a maintenance dose of 1 mg/kg once daily. After 30 days of treatment, a satisfactory therapeutic response was observed, with complete remission of pruritus. The...(AU)

Resumo

Alergia alimentar é uma reação manifestada pelo sistema imunológico após a ingestão de determinado alimento. No Brasil, a legislação vigente não contempla a obrigatoriedade de informações sobre ingredientes alergênicos em serviços de alimentação. Tendo em vista a gravidade da alergia alimentar, este trabalho teve como objetivo coletar dados sobre o fornecimento de informações acerca da presença de ingredientes alergênicos em alimentos comercializados em dois shoppings centers de Aracaju/SE. Dos dezesseis estabelecimentos visitados, observou-se que nenhum possuía cardápio de bancada e apenas 31,25 % deles possuía menu com os ingredientes do alimento, dos quais, nenhuma continha informações sobre a presença de produtos alergênicos. A falta desse tipo de informação pode trazer danos ao consumidor, desde um possível desconforto até o óbito.(AU)

Resumo

This research investigated the potential of beef products with acid whey to release bioactive peptides and thereby emphasize their health-promoting potential. Peptide sequences were isolated and identified by liquid chromatography-electrospray ionization-mass spectrometry (LC-ESI-MS). Firstly, the antihealth properties (toxicity, allergenicity) of the peptides were estimated based on the peptide sequences. Next, their health-promoting potential was demonstrated based on an in silico analysis by determining their bioactivity scores (PeptideRanker). Their various biological actions were also determined using BIOPEP-UWM tools. We presented peptide sequences with properties relevant to ensuring good health and well-being, including cardiovascular system, nervous and immune systems, or their support for the maintenance of general homeostasis. We obtained information on generation of biologically active peptides in uncured beef with acid whey and it can be considered as a new knowledge as it contributes to science development of functional and nutraceutical foods. In the long term, this information can be used in designing products with desired nutritional and health-promoting properties that are important for the well-being and for preventing the occurrence of noncommunicable diseases.

Alergias e intolerâncias alimentares relacionadas ao consumo de alimentos industrializados podem decorrer de componentes suprimidos ou informações inadequadas nos rótulos. Avaliou-se o cumprimento das legislações atuais para rotulagem de alimentos para informações obrigatórias, alergênicos, glúten e de tartrazina. Rótulos de 165 produtos alimentícios industrializados foram avaliados quanto às informações gerais e obrigatórias para rotulagem de alimentos. Os resultados para as informações obrigatórias mostraram conformidade de 94% para alimentos em geral e 33% de não conformidade para os de origem animal. Para a lista de alergênicos, 1% apresentou não conformidade. Os rótulos avaliados apresentaram conformidade para glúten. Verificaram-se irregularidades nas informações da rotulagem que podem lesar a saúde do consumidor.(AU)

Background: Fire ant venom is a complex mixture consisting of basic piperidine alkaloids, various biologically active peptides and protein components, including a variety of major allergenic proteins. Tropical fire ant Solenopsis geminata is an important stinging ant species that causes anaphylaxis and serious medical problems. Although the biological activities of allergenic venom proteins that are unique to ant venom, particularly Solenopsis 2 and 4, are still unknown, these proteins are believed to play important roles in mediating the effects of the piperidine derivatives in the venom. Methods: In the present study, the cDNA cloning, sequencing and three-dimensional structure of Sol g 4.1 venom protein are described. The recombinant Sol g 4.1 protein (rSol g 4.1) was produced in E. coli , and its possible function as a hydrophobic binding protein was characterized by paralyzing crickets using the 50% piperidine dose (PD50). Moreover, an antiserum was produced in mice to determine the allergenic properties of Sol g 4.1, and the antiserum was capable of binding to Sol g 4.1, as determined by Western blotting. Results: The molecular weight of Sol g 4.1 protein is 16 kDa, as determined by SDS-PAGE. The complete cDNA is 414 bp in length and contains a leader sequence of 19 amino acids. The protein consists of six cysteines that presumably form three disulfide bonds, based on a predicted three-dimensional model, creating the interior hydrophobic pocket and stabilizing the structure. The rSol g 4.1 protein was expressed in inclusion bodies, as determined by SDS-PAGE. Dialysis techniques were used to refold the recombinant protein into the native form. Its secondary structure, which primarily consists of -helices, was confirmed by circular dichroism analysis, and the three-dimensional model was also verified.
