Resumo

Our study aimed to compare the cecum of German mast geese with other poultry species through macroanatomy and scanning electron microscope (SEM). Adult German geese were used as research material, which were obtained from its breeders and died of various causes. In the study, after determining the macroanatomical features of the German mast goose cecum, it was examined with a scanning electron microscope (JEOL JSM-5600LV) and the general structures were photographed. The craniodorsal of the rectum of German mast geese was shaped with two ceca, giving right and left arms to the cranial symmetrical to each other near the end of the ileum. The macroanatomy of the apex, corpus, and basis cecum parts was examined in the cecum divided into two. In the study, it was determined that villus extensions were sparse in the right and left apex ceci parts in the cecum SEM images of the German mast geese, while the number of the corpus and basis ceci parts was higher. As a result, the macroanatomy and scanning electron microscopic findings of German mast geese cecum were examined in detail and its differences from other poultry species were revealed.(AU)

The use of antimicrobials as growth promoters and disease prevention is being constantly reduced in several animal production systems, including in the swine industry. Therefore, this study aimed to evaluate the effectiveness of using acidifiers to control Salmonella Typhimurium in 65-day-old pigs by detecting the pathogen in organs at euthanasia. For this, 24 piglets were divided into two experimental groups consisting of 12 piglets each. An untreated control group (G1) and a treatment group (G2) received a liquid organic acidifier in the drinking water for 10 days (D-5 to D5). Five days after the start of treatment (D0), all piglets were challenged with 106 CFU of Salmonella Typhimurium and assessed for 12 days (D12). Every three days (D3, D6, D9, and D12), three animals from each experimental group were euthanized and then submitted for necropsy. Samples from the intestines (ileum, cecum, mesenteric lymph nodes, and ileocolic lymph nodes), liver, spleen, and lungs were collected to isolate Salmonella. The results show that, numerically, Salmonella isolation in the organs of G2 was lower than in G1 and that the number of positive cecum samples in G1 (66.7%; 8/12) was statistically different from the number of positive models in G2 (16.7%; 2/12), with a reduction of 28.6% of the total cecum positive samples in the treated group compared to the control. Therefore, it was observed that the liquid organic acidifier product could reduce the colonization of organs by Salmonella Typhimurium. (AU)

O uso de antimicrobianos como promotores de crescimento e prevenção de doenças vem sendo constantemente reduzido em diversos sistemas de produção animal, inclusive na suinocultura. Portanto, o objetivo do presente estudo foi avaliar a eficácia do uso de acidificantes no controle de Salmonella Typhimurium em suínos de 65 dias de idade, detectando o patógeno em órgãos após a eutanásia. Para isso, 24 leitões foram divididos em dois grupos experimentais constituídos por 12 leitões cada. Um grupo controle não tratado (G1) e um grupo de tratamento (G2) que recebeu um acidificante orgânico líquido na água de beber por 10 dias (D-5 a D5). Cinco dias após o início do tratamento (D0), todos os animais foram inoculados oralmente com 106 UFC de Salmonella Typhimurium e avaliados por 12 dias (D12). A cada três dias (D3, D6, D9 e D12), três leitões de cada grupo experimental foram eutanasiados e posteriormente submetidos à necropsia. Amostras de intestino (íleo, ceco, linfonodos mesentéricos e linfonodos ileocólicos), fígado, baço e pulmões foram coletadas para o isolamento de Salmonella. Os resultados mostram que, numericamente, o isolamento de Salmonella nos órgãos do G2 foi inferior ao G1, e que o número de amostras positivas de ceco no G1 (66,7%; 8/12) foi estatisticamente diferente do número de amostras positivas no G2 (16,7%; 2/12), com redução de 28,6% do total de amostras positivas de ceco no grupo tratado em relação ao controle. Portanto, observou-se que o ácido orgânico líquido foi capaz de reduzir a colonização de órgãos por Salmonella Typhimurium.(AU)

Resumo

This study aimed to redescribe two species of Ozolaimus, parasites of free-living green iguanas native to Marajó Island. The gastrointestinal system of four iguana specimens was evaluated for the presence of helminths. Altogether, 12,028 nematodes were found, with a prevalence of 100%, an infection range of 780 to 7,736 nematodes, an infection intensity of 3.007, and a mean abundance of 3,007. Light microscopy and scanning electron microscopy were used to determine the species of nematodes found. The cecum was the site of infection that had the highest parasitic load. Morphologically, the nematodes were compatible with the genus Ozolaimus Dujardin, 1844, with the species Ozolaimus megatyphlon (Rudolphi, 1819) Dujardin, 1845, and Ozolaimus cirratus Linstow, 1906. Scanning electron microscopy showed the presence of small structures (serrated in Ozolaimus cirratus and rounded in Ozolaimus megatyphlon) located below the esophageal leaves. We also evidenced the phasmids in both species; this is the first record of these structures in nematodes of the genus Ozolaimus. In addition, this work expands the records on the geographic distribution of these parasites.(AU)


Este estudo tem como objetivo redescrever duas espécies de Ozolaimus, parasitas de iguanas verdes de vida livre nativas da Ilha de Marajó. O sistema gastrointestinal de quatro espécimes de iguana foi avaliado quanto à presença de helmintos. Ao todo, foram encontrados 12.028 nematoides, com prevalência de 100%, intervalo de infecção de 780 a 7.736 nematoides, intensidade de infecção de 3.007 e abundância média de 3.007. Microscopia de luz e microscopia eletrônica de varredura foram utilizadas para determinar as espécies de nematoides encontradas. O ceco foi o local de infecção que apresentou maior carga parasitária. Morfologicamente, os nematoides eram compatíveis com o gênero Ozolaimus Dujardin, 1844, com as espécies Ozolaimus megatyphlon (Rudolphi, 1819) Dujardin, 1845 e Ozolaimus cirratus Linstow, 1906. A microscopia eletrônica de varredura mostrou a presença de pequenas estruturas (serrilhadas em Ozolaimus cirratus e arredondadas em Ozolaimus megatyphlon) localizado abaixo das folhas esofágicas. Também foram evidenciados os fasmídeos em ambas as espécies; este é o primeiro registro dessas estruturas em nematoides do gênero Ozolaimus. Além disso, este trabalho amplia os registros sobre a distribuição geográfica desses parasitos.(AU)

This was a retrospective case study of colic syndrome of cecal origin in 159 horses treated at a reference Veterinary Center between June 2015 and July 2021. The data studied were breed, age, sex, clinical examinations of the initial emergency care, treatment protocols (clinical and/or surgical) and resolution (hospital discharge, euthanasia or death). It was observed that 7.55% (n = 12) of the cases corresponded to colic syndrome of cecal origin, where 75% (n = 9) had heart rate within the physiological range for the species, as well as 50% (n = 6) for capillary refill time values, 33.33% (n = 4) for pink mucosa, 41.67% (n = 5) for rectal temperature, and 16.67% (n = 2) on abdominal auscultation. In 58.34% (n = 7) of patients, pain could be controlled with analgesics, with recurrence in 41.67% (n = 5). All treated cases were cecal impaction, where 58.33% (n = 7) ruptured the cecum. In 75% (n = 9) of the cases, surgical treatment was performed. Regarding the general resolution of cases, 41.67% (n = 5) of the animals were discharged. Therefore, it is concluded that despite the increase in the diagnosis of colic of cecal origin, this disease is still rare (7.55%) and has a high potential for death since there are few clinical alterations, making its early diagnosis difficult.(AU)


Este foi um estudo retrospectivo dos casos de síndrome cólica de origem cecal em 159 cavalos atendidos em um Centro Veterinário de referência entre junho de 2015 e julho de 2021. Os dados estudados foram raça, idade, sexo, exames clínicos do atendimento inicial de emergência, protocolos de tratamento (clínico e/ou cirúrgica) e resolução (alta hospitalar, eutanásia ou óbito). Observou-se que 7,55% (n = 12) dos casos correspondiam à síndrome cólica de origem cecal, onde 75% (n = 9) apresentavam frequência cardíaca dentro da faixa fisiológica para a espécie, assim como 50% (n = 6) para valores de tempo de enchimento capilar, 33,33% (n = 4) para mucosa rosada, 41,67% (n = 5) para temperatura retal e 16,67% (n = 2) para ausculta abdominal. Em 58,34% (n = 7) dos pacientes, a dor pôde ser controlada com analgésicos, com recidiva em 41,67% (n = 5). Todos os casos tratados foram de impactação cecal, onde 58,33% (n = 7) romperam o ceco. Em 75% (n = 9) dos casos, o tratamento cirúrgico foi realizado. Em relação à resolução geral dos casos, 41,67% (n = 5) dos animais tiveram alta. Portanto, conclui-se que apesar do aumento do diagnóstico de cólica de origem cecal, esta enfermidade ainda é rara (7,55%) e apresenta um alto potencial de óbito visto que há poucas alterações clínicas dificultando seu diagnóstico precoce.(AU)

Resumo

An experiment was carried out to study the effect of different eubiotics on productive characteristics, intestinal integrity, as well as the content of enterobacteria in the cecum of broiler chickens. A completely randomized design with five treatments and 8 replicates of 25 birds each was used. In total 1000 mixed broiler chickens from Ross308 strain, one day old were obtained from a commercial hatchery. The birds were housed on concrete floors in a conventional house. A sorghum+soybean meal control diet was used, to which the additives under study were added. The treatments were distributed as follows: T1 = Control diet without antibiotic or eubiotic; T2 = T1 + bacteriophages; T3 = T1 + antibiotic; T4 = T1 + probiotic; T5 = T1 + symbiotic. The results obtained at 49 days of age for weight gain and feed conversion rate improved (p<0.05) with the addition of the antibiotic and eubiotics. A lower (p<0.05) intestinal density was observed with the probiotic. The height, width, and area of villi in duodenum was higher (p<0.05) when antibiotic and eubiotics were included. In the histological score, in duodenum, the antibiotic and eubiotics resulted with a higher score (p<0.05), associated to a physiological and controlled inflammation response that allowed improving productivity. Finally, the relative expression of enterobacteria, such as Lactobacillus salivarius, allowed associating positive changes in the microbiome and better productive parameters when including the symbiotic, with comparable results to the antibiotic when including the eubiotics.(AU)

This study performed the characterization of coccidiosis in broilers and evaluated the occurrence of suggestive cases of necrotic enteritis (NE), seeking if there is an association between the diseases in Brazilian flocks. Two hundred and fifty-six birds from 32 flocks were evaluated. Macroscopic and histopathological lesions were graduated for coccidiosis and NE. Intestinal content was investigated by polymerase chain reaction (PCR) for seven species of Eimeria and by selective anaerobic culture for Clostridium perfringens and identification of the NetB gene. Flocks positive for coccidiosis represented 93.8%. Macroscopic lesions of coccidiosis were Grade 1 for E. acervulina (27%); E. tenella (9.7%) and E. maxima (8.9%). Histopathological evaluation showed Grade 1 in duodenum (38.2%); jejunum (21.4%); cecum (9.3%) and ileum (5%). PCR demonstrated positivity for E. tenella (21.9%), E. maxima (18.8%), and E. acervulina (3.1%). Suggestive macroscopic lesions of necrotic enteritis ranged from Grade 1 (16%), 2 (23%) and 3 (10,9%). Histopathology indicated the absence of necrosis, showing only hemorrhage in the mucosa and submucosa, with the presence of Eimeria spp. Clostridium perfringens type A netB+ was not isolated, demonstrating that macroscopic lesions found mostly in the jejunum did not characterize NE, based on histopathology and negativity of the NetB gene. The study suggests that, due to the high occurrence of coccidiosis, many macroscopic findings suggestive of NE are, in fact, attributed to atypical lesions caused by the reproduction of Eimeria spp.


Este estudo realizou a caracterização de coccidiose em frangos de corte e avaliou a ocorrência de casos sugestivos de enterite necrótica (EN), buscando se há alguma associação entre estas duas enfermidades em lotes de frango de corte no Brasil. Foram avaliadas 256 aves de 32 lotes. Lesões macroscópicas e histopatológicas foram graduadas para coccidiose e EN. O conteúdo intestinal foi investigado por reação em cadeia da polimerase (PCR) para sete espécies de Eimeria e por cultura anaeróbia seletiva para Clostridium perfringens e identificação do gene NetB. Os lotes positivos para coccidiose representaram 93,8%. Lesões macroscópicas de coccidiose foram de Grau 1 para E. acervulina (27%); E. tenella (9,7%) e E. maxima (8,9%). A avaliação histopatológica mostrou Grau 1 no duodeno (38,2%); jejuno (21,4%); ceco (9,3%) e íleo (5%). A PCR demonstrou positividade para E. tenella (21,9%), E. maxima (18,8%) e E. acervulina (3,1%). Lesões macroscópicas sugestivas de enterite necrótica variaram de grau 1 (16%), 2 (23%) e 3 (10,9%). A histopatologia indicou ausência de necrose, apresentando apenas hemorragia em mucosa e submucosa, com presença de Eimeria spp. Clostridium perfringens tipo A netB + não foi isolado, demonstrando que lesões macroscópicas encontradas principalmente no jejuno não caracterizaram NE, com base na histopatologia e negatividade do gene NetB. O estudo sugere que, em virtude da alta ocorrência de coccidiose nos lotes, muitos achados macroscópicos sugestivos de EN são, na verdade, atribuídos a lesões atípicas provocadas pela reprodução de Eimeria spp.

ABSTRACT: Coccidiosis is a disease of great importance in industrial poultry. The correct diagnosis directs the poultry industry to its best treatment and control. Thus, a survey of Eimeria spp. was carried out in intestines of 64 broiler flocks, with an average age of 29 days. Eight broilers from each flock were randomly removed from the slaughter line, in a total of 512 samples. Macroscopic and histopathological lesions in the intestine were classified into Scores 0 to 4. Polymerase chain reaction (PCR) was used to research the oocysts from the seven species of Eimeria spp. in the intestinal content. The macroscopic evaluations showed that 59.4% (38/64) of the flocks were positive for E. acervulina, 32.8% (21/64) for E. maxima, 29.7% (19/64) for E. tenella, and 34.4% (22/64) for E. brunetti. The histopathological evaluation showed that 87.5% (56/64) of the flocks had at least one broiler with parasitic structures compatible with Eimeria spp. in the duodenum, 70.3% (45/64) in the jejunum, 18.8% (12/64) in the ileum, 46.9% (30/64) in the cecum, and 4.7% (3/64) in the colon. In PCR, 21.9% (14/64) of the flocks were positive for E. acervulina, 12.5% (8/64) for E. maxima, 3.1% (2/64) for E. mitis, and 32.8% (21/64) for E. tenella. The Kappa Cohen test between macroscopy, histopathology, and PCR demonstrated concordance ranging from weak to moderate with the exception of histopathology and PCR of the cecum, which was strong. In the comparison between macroscopy and histopathology, there were significative differences between Scores 0 and 1 (apart from the cecum). For Score 3, there were significative differences in duodenum, jejunum and cecum (p 0.05). In conclusion, the macroscopic diagnosis and PCR can generate false-negative results, and the histopathological exam proved to be effective, making it essential to associate different techniques for the correct diagnosis of Eimeria spp. in broiler chickens.


RESUMO: A coccidiose é uma doença de grande importância na avicultura industrial. O diagnóstico correto direciona a indústria avícola ao seu melhor tratamento e controle. Desta forma, realizou-se a pesquisa de Eimeria spp. em intestinos de 64 lotes de frangos de corte, com idade média de 29 dias. Em cada lote foram retirados aleatoriamente oito frangos da linha de abate, totalizando 512. Os intestinos foram classificados na macroscopia e na histopatologia em Grau de 0 a 4. No conteúdo intestinal pesquisou-se por reação em cadeia da polimerase (PCR) oocistos das sete espécies de Eimeria. As avaliações macroscópicas demonstraram que 59,4% (38/64) dos lotes foram positivos para E. acervulina, 32,8% (21/64) para E. maxima, 29,7% (19/64) para E. tenella e 34,4% (22/64) para E. brunetti. Na avaliação histopatológica, 87,5% (56/64) dos lotes apresentaram pelo menos um frango com estruturas parasitárias compatíveis com Eimeria spp. no duodeno, 70,3% (45/64) no jejuno, 18,8% (12/64) no íleo, 46,9% (30/64) no ceco e 4,7% (3/64) no cólon. O teste de concordância de Kappa Cohen entre macroscopia, histopatologia e PCR demonstrou concordância variando de fraca a moderada com exceção da histopatologia e PCR do ceco que foi forte. Na comparação dos graus de macroscopia e histopatologia, foram encontradas diferenças significativas entre o Grau 0 e 1 (exceto no ceco) e no Grau 3 houve diferença para duodeno, jejuno e cecos (p 0,05). Conclui-se que o diagnóstico macroscópico e a PCR podem gerar resultados falsos negativos e que o exame histopatológico se demostrou eficaz, tornando fundamental a associação de diferentes técnicas para o correto diagnóstico de Eimeria spp. em frangos de corte.

Chicken coccidiosis is a common and severe parasitic disease caused by infection from Eimeria spp., which affects the integrity of the intestinal mucosa. TGF-ß has been shown to play an important role in the healing of intestinal mucosas, immunity, and the maintenance of bowel mucosa integrity. Very little is known about the presence of the components of TGF-ß/Smads signaling pathway of chicken at different times following coccidian infection. In the present study, we measured the levels of TGF-ß2, 3, 4, receptor TßRI, II, down-stream Smad 2, 3, 7 in cecum and spleen of chicken at different times after inoculation with Eimeria tenella using quantitative real-time PCR. The results showed that the TGF-ß/Smads signaling pathway was not activated in cecum in the early stage of infection. However, on the 8th day, the expression of TGF-ß2, 4, down-stream protein Smad 2, 7 were significant up-regulated in the spleen, which indicated that the TGF-ß/Smads signaling was changed in the E. tenella infection and was differentially expressed in various tissues in the early stages of infection.(AU)

Resumo

Broilers maintenance at high stocking density impacts uncomfortable conditions; this increases the potential for stress, reduces feed consumption, and reduces production performance and health in broiler chickens. This study aims to evaluate the effect of clove leaf extract on growth performance and microbial population in the digestive tract of broiler chickens reared at high density. From the eighth day, five treatment groups, T0 (negative control with a typical density of 10 birds/m2 ), T1 (positive control with a high density of 16 birds/m2 ), T2, T3, and T4 with a high density of 16 birds/m2, were randomly assigned from 444 broilers (bodyweight of 129.78±0.75 g) with six repetitions each on the eighth day. The feed includes clove leave extract as much as 0.5, 0.75, and 1 ml/kg for T2, T3, and T4, respectively. The weight gain, feed intake, and feed conversion ratio were tracked weekly. One chicken from each replicate was taken randomly, slaughtered, and feathered on day 35. The digesta was taken from the ileum and cecum to measure the intestinal bacterial population and put into a sterile container. Digesta from the duodenum, jejunum, ileum, and cecum were also collected to determine pH levels. MRS agar was used to determine the amount of LAB in the digesta. The findings revealed that giving clove leaf extract of feed to broiler chickens kept at high stocking density could higher body weight gain (P < 0.05), higher feed consumption (P < 0.05), decreased the pH of the ileum and cecum (P < 0.05), increased lactic acid bacteria and decreased coliform bacteria in the ileum and cecum (P < 0.05).

The objective was to evaluate the productivity, microbiology and histopathology of the jejunum, ceca and liver in broilers fed a control or a high non-starch polysaccharide (HNSPs) diet added with an extract of humic substances (EHS). 240 broilers individually allocated, from 21-42 d of age were assigned to a factorial arrangement of 2 types of diets: 1) A corn/soybean meal diet (Control) and 2) As Control plus 7% distillers dried grain with solubles and 12% of wheat bran (HNSPs), and 3 growth promoters: 1) Antibiotic growth promoters (AGP); 2) without AGP (NAGP) and C) with 0.5% of EHS. At the end of the trial, the breast and carcass were weighed and samples of the intestine, ceca and liver were taken for microbial and histopathology analysis. Results were subjected to ANOVA. EHS-fed broilers had lower feed conversion ratio (FCR) and total aerobic bacterial (TAB) counts in the liver and higher ashes digestibility with the Control diet, but negative responses were seen with the HNSPs diet (Type of diet and growth promoter interaction, p≤0.05). The lesion scores in the jejunum were lower in EHS-fed broilers with the Control and HNSPs diet (p≤0.05). The TAB and E. coli were lower in the jejunum and ceca of AGP-fed broilers (p<0.01) compared to NAGP and EHS groups. EHS-fed broilers showed improved FCR and ileal ashes digestibility and lower TAB in the liver with the Control diet and had lower lesion score in the jejunum and similar weight gain compared to the AGP-fed broilers.(AU)

Thus, a survey of Eimeria spp. was carried out in intestines of 64 broiler flocks, with an average age of 29 days. Eight broilers from each flock were randomly removed from the slaughter line, in a total of 512 samples. Macroscopic and histopathological lesions in the intestine were classified into Scores 0 to 4. Polymerase chain reaction (PCR) was used to research the oocysts from the seven species of Eimeria spp. in the intestinal content. The macroscopic evaluations showed that 59.4% (38/64) of the flocks were positive for E. acervulina, 32.8% (21/64) for E. maxima, 29.7% (19/64) for E. tenella, and 34.4% (22/64) for E. brunetti. The histopathological evaluation showed that 87.5% (56/64) of the flocks had at least one broiler with parasitic structures compatible with Eimeria spp. in the duodenum, 70.3% (45/64) in the jejunum, 18.8% (12/64) in the ileum, 46.9% (30/64) in the cecum, and 4.7% (3/64) in the colon. In PCR, 21.9% (14/64) of the flocks were positive for E. acervulina, 12.5% (8/64) for E. maxima, 3.1% (2/64) for E. mitis, and 32.8% (21/64) for E. tenella. The Kappa Cohen test between macroscopy, histopathology, and PCR demonstrated concordance ranging from weak to moderate with the exception of histopathology and PCR of the cecum, which was strong. In the comparison between macroscopy and histopathology, there were significative differences between Scores 0 and 1 (apart from the cecum). For Score 3, there were significative differences in duodenum, jejunum and cecum (p<0.05). In conclusion, the macroscopic diagnosis and PCR can generate false-negative results, and the histopathological exam proved to be effective, making it essential to associate different techniques for the correct diagnosis of Eimeria spp. in broiler chickens.

Desta forma, realizou-se a pesquisa de Eimeria spp. em intestinos de 64 lotes de frangos de corte, com idade média de 29 dias. Em cada lote foram retirados aleatoriamente oito frangos da linha de abate, totalizando 512. Os intestinos foram classificados na macroscopia e na histopatologia em Grau de 0 a 4. No conteúdo intestinal pesquisou-se por reação em cadeia da polimerase (PCR) oocistos das sete espécies de Eimeria. As avaliações macroscópicas demonstraram que 59,4% (38/64) dos lotes foram positivos para E. acervulina, 32,8% (21/64) para E. maxima, 29,7% (19/64) para E. tenella e 34,4% (22/64) para E. brunetti. Na avaliação histopatológica, 87,5% (56/64) dos lotes apresentaram pelo menos um frango com estruturas parasitárias compatíveis com Eimeria spp. no duodeno, 70,3% (45/64) no jejuno, 18,8% (12/64) no íleo, 46,9% (30/64) no ceco e 4,7% (3/64) no cólon. Conclui-se que o diagnóstico macroscópico e a PCR podem gerar resultados falsos negativos e que o exame histopatológico se demostrou eficaz, tornando fundamental a associação de diferentes técnicas para o correto diagnóstico de Eimeria spp. em frangos de corte.

Resumo

Spirulina platensis is a microalga with biological activity used to produce nutritional supplements rich in proteins, essential fatty acids, vitamins, and minerals. The current study aims to prove the impact of the product ZooBioR2 (obtained from Spirulina platensis) on health, and on the histology of duodenum, jejunum, ileum, and cecum in laying hens. The experiment was performed in duplicate on 5 groups of hens (14 birds/group), one control and four experimental, in which the feed was supplemented with ZooBioR2 in different doses (5.0; 10.0; 15.0; 20.0mg active substance/kg of fodder). Inclusion of the supplement in the diet have determined a significant decrease of height of intestinal villi, the depth of the crypts and the thickness of muscular layer of mucosa in the experimental groups receiving 15.0 and 20.0mg/kg. The decrease of crypts depth is an indicator of a higher rate of turn-over of epithelial tissue. The morphological changes of intestinal mucosa could be involved in a better digestibility of the fodder.


Spirulina platensis é uma microalga com atividade biológica utilizada para produzir suplementos nutricionais ricos em proteínas, ácidos graxos essenciais, vitaminas e minerais. O estudo atual visa provar o impacto do produto ZooBioR2 (obtido da Spirulina platensis) na saúde e na histologia do duodeno, jejuno, íleo e ceco em galinhas poedeiras. O experimento foi realizado em duplicata em cinco grupos de galinhas (14 aves/grupo), um controle e quatro experimentais, nos quais a ração foi suplementada com ZooBioR2 em diferentes doses (5,0; 10,0; 15,0; 20,0mg de substância ativa/kg de forragem). A inclusão do suplemento na ração determinou uma diminuição significativa da altura das vilosidades intestinais, da profundidade dos crypts e da espessura da camada muscular da mucosa nos grupos experimentais que receberam15,0 e 20,0mg/kg. A diminuição da profundidade das criptas é um indicador de maior taxa de rotação do tecido epitelial. As alterações morfológicas da mucosa intestinal podem estar envolvidas em uma melhor digestibilidade da forragem.

Clinical History: Two 18-month-old female bison and a heifer died suddenly in the same ranch. The three animals were submitted for postmortem examination and diagnostic workup. The changes of one of the bison are presented here. Necropsy Findings: The carcass was in fair nutritional condition, with small amount of fat reserves, but still well fleshed, and in mild state of post-mortem decomposition. There was a small amount of clotted dark-red blood in the nostrils and anus. The heart presented extensive sub-endocardial hemorrhages in all four chambers. These lesions were most pronounced in both ventricles where they extended deep into the underlying myocardium (Fig. 1 and Fig. 2). The mucosa of the abomasum was diffusely and mildly congested, and the mucosa of the small and large intestine was diffusely and severely congested (Fig. 3). There was a moderate amount of red fluid in the lumen of the small intestine, cecum, and colon. Both lungs had a few, small, well delineated, deep, firm, multifocal dark-red areas in all pulmonary lobes, but lung tissue samples floated when placed into formalin. Chicken meat is an important source of foodborne pathogens, including Salmonella, Campylobacter, and Clostridium perfringens. These bacteria can occur in the intestinal microbiota of broilers and contaminate chicken carcasses in industrial meat processing. This study aimed to develop and evaluate a procedure based on real-time PCRs for the direct detection and quantification of these three bacteria in broilers' ceca collected in poultry slaughter houses and demonstrate the occurrence of these important foodborne pathogens in Brazilian poultry production flocks. Cecal contents were collected from 45 different broiler flocks in three different slaughterhouses in the state of Paraná, Brazil, totaling 45 samples (in pools of 10 different ceca/chickens per broiler flock). Then, these samples were tested for the detection and quantification of Salmonella, Campylobacter, and Clostridium perfringens by real-time PCRs. The results demonstrated the occurrence of three (6.7%) positive pools for Salmonella, 20 (44.4%) for Campylobacter, and 32 (71.1%) for C. perfringens. Mean bacterial concentrations in the positive samples were 4.3log10 cells/g for Salmonella, 6.4 log10 cells/g for Campylobacter, and 5.5 log10 cells/g for C. perfringens. In conclusion, Salmonella, Campylobacter, and C. perfringens could be detected and quantified directly from the broilers cecal contents collected in the slaughter line. This procedure will be certainly useful to more quickly detect these foodborne pathogens and prevent their occurrence in chicken meat and other poultry food products.(AU)

Resumo

Effects of dietary Agaricus bisporus mixture or stalk or cap on growth performance, carcass components and some meat quality parameters, mesophilic aerobic bacterial counts, and intestinal histomorphology in broiler chickens were investigated. Two hundred and forty one-day-old male Ross 308 broiler chickens were divided into 4 experimental groups with 4 replicates, each including 15 birds. Chickens were fed with basal diet (C), mushroom mixture (MM, 10 g stalk+10 g cap/kg diet), mushroom stalk (MS, 20 g/kg diet) and mushroom cap (MC, 20 g/kg diet). Feed conversion ratio (FCR) was improved (P < 0.01) by dietary MS while feed intake (FI) decreased (P < 0.01) in all treatment groups compared to control. However, body weight gain (BWG) was decreased (P < 0.01) by MC inclusion. Mushroom supplemented groups had higher (P < 0.05) mesophilic aerobic bacteria in the cecum. MS inclusion increased villus height to crypt depth ratio in the jejunum (P < 0.01) and villus height in the ileum (P < 0.01) and jejunum (P < 0.05). MC increased (P < 0.01) crypt depth in the jejunum. There were no statistical differences among groups for carcass components (P > 0.05). The L* values were decreased (P < 0.01) in thigh meat but increased (P < 0.01) in breast meat by mushroom inclusion. The a* values were decreased (P < 0.01) and b* values were increased (P < 0.05) in both thigh and breast meat with mushroom addition to diet. The obtained results indicated that A. bisporus stalk meal at an inclusion level of 20 g/kg of diet had favourable effects on growth performance with higher feed efficiency, improved intestinal morphology with higher villus height and increased meat quality of broiler chickens.


Foram investigados os efeitos da mistura dietética de Agaricus bisporus ou talo ou tampa sobre o desempenho de crescimento, componentes da carcaça e alguns parâmetros de qualidade da carne, contagens de bactérias aeróbias mesofílicas e histomorfologia intestinal em frangos de corte. Duzentos e quarenta frangos de corte Ross 308 de um dia de idade foram divididos em quatro grupos experimentais com quatro repetições, cada um incluindo 15 aves. Galinhas alimentadas com dieta basal (C), mistura de cogumelos (MM, 10 g caule + 10 g cap / kg de dieta), caule de cogumelo (MS, 20 g / kg dieta) e cogumelo (MC, 20 g / kg dieta). A taxa de conversão alimentar (FCR) foi melhorada (P < 0.01) por MS dietético enquanto o consumo de ração (FI) diminuiu (P < 0.01) em todos os grupos de tratamento em comparação com o controle. No entanto, o ganho de peso corporal (BWG) foi diminuído (P < 0.01) pela inclusão de MC. Os grupos suplementados com cogumelos apresentaram maiores (P < 0.05) bactérias aeróbias mesofílicas no ceco. A inclusão de MS aumentou a proporção entre a largura das vilosidades e a profundidade da cripta no jejuno (P < 0.01) e o comprimento das vilosidades no íleo (P < 0.01) e jejuno (P < 0.05). MC aumentou (P < 0.01) a profundidade de cripta no jejuno. Não houve diferença estatística entre os grupos nos componentes da carcaça (P > 0.05). Os valores L * diminuíram (P < 0.01) na carne da coxa, mas aumentaram (P < 0.01) na carne de peito com a inclusão de cogumelos. Os valores de a* diminuíram (P < 0.01) e os valores de b* aumentaram (P < 0.05) em ambas as coxas e peitos pela adição de cogumelos à dieta. Os resultados obtidos indicaram que a farinha de colmo de A. bisporus com um nível de inclusão de 20 g / kg de dieta teve efeitos favoráveis no desempenho do crescimento com maior eficiência alimentar, melhorou a morfologia intestinal com maior comprimento de vilosidade e aumentou a qualidade da carne de frangos de corte.

Geese (Anser cygnoides) possess stronger ability of roughage digestion and utilization than other poultries, hence, it has become the focus of attention of scientists. Duodenal, jejunum and ileum were mainly participated in food digestion and nutrient absorption, while the cecum was responsible for biological fermentation. Effects on the geese's cecal microbiota community by feeding with the all-grass diet have been investigated, however, whether it had an influence on the geese's duodenal microbiota community remains unexplored. To address this problem, geese feeding with the basal diet for 28 days (G1), the basal diet for 28 days and the all-grass diet for the following 14 days (G2), the basal diet for 42 days (G3) were selected, respectively. The duodenal segments of geese were collected and the hypervariable V3-V4 region of the bacterial 16S rRNA gene was sequencing. A total of 4 main phyla and 16 main genera were identified. Moreover, we also successfully identified that two taxa including the Helcococcus and Clostridium could be used as distinguishing biomarkers specific to G2. The functional profiles of the duodenum microbiota were mainly involved in the membrane transport (e.g. ABC transporters), amino acid metabolism, energy metabolism, metabolism of cofactors and vitamins, and cellular processes and signaling pathways in geese feeding with the all-grass diet. In conclusion, the all-grass diet could impact the composition of duodenal microbiota. However, to resolve the underlying mechanism of the fiber digesting and utilization in geese's gut microbiota, the whole intestinal system needs to be assessed by further studies.(AU)

Poultry products may be a source of foodborne human salmonellosis. The use of alternatives to antimicrobials that are not harmful to humans may reduce the presence of Salmonella spp. in poultry production. Among the products used, organic acids stand out. In the present study, three different organic acid (OA) blends were evaluated for the control of Salmonella Heidelberg (SH) in commercial broilers. Day-old chicks (n = 114) were randomly assigned to four treatments, with three replicates of 12 birds each. Birds in treatments A and B received SCFA (0.2mL/L) and SCFA + MCFA (0.2mL/L), respectively, in the drinking water, while birds in treatment C received SCFA + MCFA in the feed (2g/Kg of feed). Birds from treatment D did not receive OAs (control group). At 8 days of age, each bird was orally inoculated with SH at 108 CFU/mL, and cloacal swabs and SH enumeration of the cecal content were performed 24-, 48-, and 72-hours post-inoculation (hpi). The results show a reduction of both SH shedding and counts in the birds fed OAs at all pi times relative to the control birds. Fecal shedding was significantly lower in the OA-treated groups compared with the control group. As for SH presence in the cecum, significant differences were detected between groups C and D at 24 and 72 hpi, and between groups B and D at 72 hpi. The results of this study indicate that the use of feeding OAs to broilers may contribute to reduce the incidence of SH in the poultry production chain, allowing better flock health management, provided an efficient biosecurity program is employed.(AU)

Currently, the fish farming with Arapaima gigas has suffered from technological obstacles in the fields of reproduction, health and nutrition, making it necessary to know the morphology of its structures, so that it can advance in more technified research in scope of production in Rondônia state, as well as in aquaculture nutrition and health. Therefore, the aimed is to characterize the macro and microscopic morphology of posterior digestive system of A. gigas. The intestine of six specimens A. gigas in ideal slaughter size was analyzed. The analyzes were performed using light-sheet microscopy (LM) and scanning electron (SEM) techniques. The intestine basically showes similar histological characteristics in three analyzed portions (proximal, middle and distal). Same type of simple columnar epithelium with goblet cells was evidenced, with subtle variations in pattern of villi in each segment, and in number of goblet cells. In the rectum, the amount of goblet cells and evident longitudinal villi was expressive. Macroscopic anatomy and histology of the intestine A. gigas analyzed showes characteristics of adaptation to cultivation, according to their diet and habitat. The intestinal mucosa can divided into three distinct portions: proximal, middle and final intestine, in addition to the rectum and anus. In the pyloric cecum, the folds are slightly higher and poorly branched. The rectum, compared to the midgut, showed a higher occurrence of goblet cells in the mucosa. This increase in goblet cells observed in the posterior portion may related to the assimilation of ions and fluids that occur at this location.


Atualmente, a piscicultura do Arapaima gigas tem sofrido com entraves tecnológicos nos campos de reprodução, saúde e nutrição, fazendo-se necessário conhecer a morfologia de suas estruturas, para que se possa avançar em pesquisas mais tecnificadas no âmbito da produção piscícola no estado de Rondônia, bem como na nutrição e na sanidade aquícola. Por isso, objetiva-se caracterizar a morfologia macro e microscópica do sistema digestivo posterior do A. gigas. Foi analisado o intestino de seis espécimes de A. gigas em porte de abate. As análises foram realizadas por meio de técnicas de microscopia de luz (ML) e eletrônica de varredura (MEV). O intestino apresenta basicamente características histológicas similares nas três porções analisadas (proximal, médio e distal). Evidenciou-se um mesmo tipo de epitélio colunar simples com células caliciformes, variando o padrão de vilos em cada segmento de forma sutil, e o número de células caliciformes. No reto foi expressiva a quantidade de células caliciformes e as vilos longitudinais evidentes. A anatomia macroscópica e a histologia do intestino de A. gigas analisadas apresentam características de adaptação ao cultivo, conforme sua alimentação e habitat. A mucosa intestinal pode ser dividida em três porções distintas: intestino proximal, médio e final, além de reto e ânus. No ceco pilórico, as dobras são ligeiramente mais altas e pouco ramificadas. O reto, comparado ao intestino médio, apresentou maior ocorrência de células caliciformes na mucosa. Esse aumento de células caliciformes observado na porção posterior pode estar relacionado com a assimilação de íons e fluidos que ocorrem neste local.
