This study evaluated the levels of heavy metals in the soil and castor bean plant after fertilization with sewage sludge (SS) subjected to different stabilization processes. The study was conducted in a Haplic Cambisol with the following treatments: control (CO), fertilization with solarized sewage sludge (SSS), composted sewage sludge (CSS), vermi composted sewage sludge (VSS), limed sewage sludge (LSS), and mineral fertilizer (MF). The experimental design included a subdivided plot scheme with fertilization being the primary factor and soil layers or leaf parts being the secondary factor. The data obtained were analyzed using analysis of variance and the Scott-Knott test with 5% significance. Fertilization with SSS, CSS, and LSS led to the highest increase in Zn in the soil, mainly in the surface layer. In the leaf tissues, Zn and Cu levels were higher with the CO and SSS treatments, whereas the Ni level was higher with SSS, CSS, VSS, and MF treatments. In general, Zn and Cu levels were higher in the leaf limb than in the petiole, whereas Ni, Pb, and Cr levels were higher in the petiole than in the leaf limb. In any case, no symptoms of toxicity in the plants and no risk of soil contamination were observed with any of the fertilization treatments.

Purpose: Stroke is an acute cerebrovascular disease. Astragaloside IV (AS-IV) is an active ingredient extracted from Astragalus membranaceus with an established therapeutic effect on central nervous system diseases. This study examined the neuroprotective properties and possible mechanisms of AS-IV in stroke-triggered early brain injury (EBI) in a rat transient middle cerebral artery occlusion (MCAO) model. Methods: The neurological scores and brain water content were analyzed. 2,3,5-triphenyl tetrazolium chloride (TTC) staining was utilized to determine the infarct volume, neuroinflammatory cytokine levels, and ferroptosis-related genes and proteins, and neuronal damage and molecular mechanisms were evaluated by terminal deoxynucleotidyl transferase dutp nickend labeling (TUNEL) staining, western blotting, and real-time polymerase chain reaction. Results: AS-IV administration decreased the infarct volume, brain edema, neurological deficits, and inflammatory cytokines TNF-α, interleukin-1ß (IL-1ß), IL-6, and NF-κB, increased the levels of SLC7A11 and glutathione peroxidase 4 (GPX4), decreased lipid reactive oxygen species (ROS) levels, and prevented neuronal ferroptosis. Meanwhile, AS-IV triggered the Nrf2/HO-1 signaling pathway and alleviated ferroptosis due to the induction of stroke. Conclusion: Hence, the findings of this research illustrate that AS-IV administration can improve delayed ischemic neurological deficits and decrease neuronal death by modulating nuroinflammation and ferroptosis via the Nrf2/HO-1 signaling pathway.

Mammary neoplasia represents the most frequently diagnosed type of neoplasia in bitches. Although surgical removal is the procedure of choice for therapeutic management, chemotherapy protocols appear as important allies and adjuvants. Despite the great advances that have occurred in the field of cancer therapy, the systemic repercussions of these drugs still impose important limitations on their use. In this sense, the development of increasingly targeted therapeutic protocols and preventive monitoring of patients represent important strategies to avoid possible complications - among them, Acute Kidney Injury (AKI). Routinely, ultrasound evaluation is used to identify morphological or metastatic variations in abdominal cavity organs. Acting complementary to the B-mode evaluation, Doppler mapping proves to be efficient in recognizing alterations in vascular hemodynamics. Therefore, the objective of the present study was to evaluate the use of B-mode and Doppler ultrasound to identify renal morphological and hemodynamic alterations in bitches with mammary neoplasia submitted to adjuvant chemotherapy protocols that associate gemcitabine with carboplatin. Thirteen bitches were included, without distinction of breed and between seven and 13 years of age. The animals were evaluated ultrasonographically at two different times during three consecutive chemotherapy cycles: before (T0) and one and a half hours after each cycle (T1), for 42 days. No morphological changes were observed in B-mode throughout the chemotherapy protocol. However, the Doppler velocimetry indices demonstrated statistical differences before (T0) and after (T1) the administration of the drugs. It was concluded that Doppler ultrasound could be used as a complementary method for monitoring the renal response of patients exposed to nephrotoxic drugs and potentially causing renal injury.

The study aimed to verify the water stress indicators of garlic (Allium sativum L.) submitted to water deficit levels, simulating the effects of possible climatic changes. Considering that A. sativum is a crop known worldwide for its medicinal, culinary properties and applications in the ecological management of agricultural pests and, knowing that the planet is undergoing constant climatic changes, this study becomes essential to understand the influence of these changes on this species cultivation. The treatments were established with water conditions of 100, 75, 50, and 25% of the pot capacity (PC). According to the pot capacity, three bulbils were planted per pot (5 L) containing Plantmax® substrate, irrigated every two days. During the first and last week of the greenhouse treatments, the chlorophyll index was evaluated using a porometer with a seven-day interval between the analysis. The first analysis was performed in the first week of water stress and the last one in the last days of stress. Garlic plants showed sensitivity to water deficit, resulting in lower conductance and transpiration compared to treatments 50% and 75% water deficit, apparently without causing changes in production. In view of the results of this study, it is concluded that Allium sativum L. will not be totally harmed by the climate changes expected in the future according to atmospheric changes in the region of Laranjeiras do Sul.

This study investigated the influence of the farm microclimate parameters in different seasons on the incidence of ketosis, foot diseases, reproductive diseases, and mastitis in three dairy farms of North Kazakhstan. Microclimate parameters were recorded in four seasons: summer, autumn, winter, and spring. The average temperature was 1.2 °C above the established norm in summer and 1.7 °C below the established norm in winter. The humidity was below the norm by 9.7% and 1.6% in summer and autumn, respectively, and above the norm by 9.6 % and 6.6 % in winter and spring, respectively. The airflow was 0.1 m/s lower than the norm in summer, winter, and spring. The general illumination was 3.8 and 2.6 LUX lower than the norm in winter and spring, respectively. The average morbidity (cumulative incidence of all four diseases) was 63.8% (highest) in the winter, followed by spring (60.0%). Average morbidity was considerably less in summer and autumn at 46.7% and 37.1%, respectively. Study results indicate that minor deviations in microclimate parameters from the norms, particularly in winter and spring, can considerably impact the disease incidence on dairy farms in Kazakhstan. The farm management should strive to maintain microclimate conditions on the farm as close to the norm as possible during different seasons to avoid losses from livestock morbidity.

Background: Ehlers Danlos Syndrome (EDS) is a rare genetic disease characterized by a deficiency in collagen synthesis, which can result in joint laxity. Patellar luxation is one of the main orthopedic conditions that affect the canine knee joint, with limited descriptions of its association with EDS in dogs. The purpose of this report is to describe the surgical management and postoperative evolution of a 1-year-old Chow Chow dog with grade II patellar luxation, tibial valgus and EDS.Case: A 1-year-old Chow Chow dog was referred to the University Veterinary Hospital due to lameness of the left pelvicfor 3 months. At the orthopedic examination were verified severe lameness and lateral deviation of the left stifle joint during the ambulation of the animal. Additionally, it was verified bilateral hyperextension of the tibiotarsal joint and grade IIpatellar luxation of both pelvic limbs with painful hyperextension of the left stifle joint. Radiographic evaluation showedlateral displacement of the patella from both femoral trochlear groove, and a valgus deviation of the proximal left tibialshaft. In addition, it was verified cutaneous hyperextensibility and an extensibility index suggestive of EDS. The animalwas submitted to trochlear block resection technique and medial imbrication, followed by corrective tibial osteotomy.Furthermore, skin biopsies of the scapular and lumbar folds were performed during the corrective tibial osteotomy. Thesamples were sent for histopathological examination, which revealed fragmented and unorganized collagen fibers in thedermis. Histopathological findings were compatible with EDS. The absence of lameness and correct positioning of thepatella in the trochlear sulcus were verified in the post-surgical follow-up. Complete bone consolidation of the closingwedge osteotomy to correct the tibial...

Partial fuel replacement strategies arising from fossil sources used in compression ignition engines involve mixtures of mineral Diesel oil, biodiesel and ethanol to minimize the gas emissions. In this study, experimental assessments were performed on a multi-cylinder, turbocharged aftercooler, compression-ignition, agricultural tractor engine provided with electronic injection management and an exhaust gas recirculation (EGR) gas treatment system. Diesel oil containing low (BS10 -10 ppm) and high sulfur concentrations (BS500 - 500 ppm) was utilized, with 10% of biodiesel as a constituent established by Brazilian legislation, in blends with 5, 10, 15 and 20% of the total volume, made up of anhydrous ethanol with additives. Thus, there were eight fuels blends and two reference conditions (without ethanol). The emissions of CO, HC, NOx and the HC+NOx gases were estimated, corresponding to the eight operating modes (M) of the ABNT NBR ISO 8178-4 standard. From the findings, it was evident that with the rise in the ethanol concentrations in the fuel blends there was a corresponding increasing in the CO, NOx and HC+NOx emissions. The HC, on the contrary, exhibited a pattern of higher emissions for the high-sulfur fuels (BS500) at low loads. No difference was observed for the NOx emissions at high loads. In the other operation modes, different behaviors were expressed for the BS10, which sometimes showed an increase, while at other times a reduction in the NOx emissions. Regarding the BS500, the NOx emission increased when the ethanol concentrations rose. As the specific emissions of the NOx were higher than those of the HC (in g.kW-¹.h-¹), the behavior exhibited by the HC+NOx showed similarity to that of the NOx. When the directly analysis of the operating modes was taken into consideration, the use of ethanol triggered an upswing in the emissions, exceeding the threshold of MAR-1 and EURO V standards.

This study aims to investigate the effect of different periods of fasting and re-feeding on compensatory responses in Nile tilapia fingerlings and the frequency of muscle fiber distribution. A total of 108 Nile tilapia fingerlings with initial weight of 1.64 ± 0.41 g and mean initial length of 3.60 ± 0.39 cm were used for 55 days. The fish were distributed in a water recirculation system in a completely randomized design with three treatments and four replications: Control - CO - (fish fed until apparent satiation throughout the experimental period); fasting 10 - J10 - (fish fed to apparent satiation for 15 days, followed by 10 days of fasting and re-feeding to satiation for 30 days); and fasting 15 - J15 - (fish fed to apparent satiation for 15 days, followed by 15 days of fasting and re-feeding to satiation for 25 days). Fish from the J15 treatment showed unsatisfactory results in terms of productive performance (p<0.05), such as lower final weight, apparent feed conversion, protein efficiency ratio, and survival, while fish from the J10 treatment achieved the same results as those animals kept in the CO treatment with the exception of the variables of relative weight gain and feed intake. Furthermore, food restriction directly influenced the growth of muscle fibers with a diameter smaller than 20 µm (p<0.05), and fish from the J15 treatment had the lowest frequency of fibers in this diameter class. Therefore, this study concludes that food restriction in short periods (10 days) and at low temperatures can present a compensatory growth, as they alter the process of hyperplasia and hypertrophy of muscle fibers without affecting the morphology of the fibers; however, 15 days of fasting under low temperatures do not compensate for growth and delays the hypertrophic growth of muscle fibers.(AU)

Background: Demodicosis is a parasitic dermatopathy often found in dogs and considered rare in felines. It is caused by the mite of the genus Demodex. In cats, it can be caused by 3 species: Demodex cati, Demodex gatoi, and another Demodex species not yet named. Demodicosis can be associated with systemic diseases, which can compromise the animal's immune system, as is the case of demodicosis caused by Demodex cati. The present case report describes 3 cases of demodicosis, 2 by D. cati and 1 by D. gatoi, with pruritic lesions and abrasions in different parts of the animals' bodies, but which showed improvement with common treatment. Cases: The first 2 cases refer to feline patients treated in Curitibanos and the third in Blumenau, both municipalities located in Santa Catarina. The first patient, a 6-year-old female, SRD, IVF/FeLV-negative, presented abrasions and alopecia in the cervical region, at the base of the auricle and submandibular region, with pruritus for 3 months. Skin scraping was performed revealing presence of Demodex cati. The second patient, a 11-year-old male, SRD, FIV-negative and FeLVpositive, presented alopecic lesions with mild pruritus, one on the dorsal region of the head and 2 others slightly oval on the tail. A skin scraping was performed for parasitological examination which demonstrated the presence of mites of the species Demodex cati. The third patient, a 10-month-old male Persian, IVF/FeLV-negative, had alopecic and pruritic lesions on the back and head near the auditory canal, as well as signs of external otitis. The lesions on the skin were scaly and presented a blackish appearance, with evolution of a few weeks. Skin scraping was performed for parasitological examination, confirming the presence of Demodex gatoi. In all cases, fungal cultures were negatives. For the 3 patients, fluralaner was used transdermally, as a pipette with a single application, and all showed complete improvement within 30 days after administration. Discussion: Demodicosis is considered a rare disease among felines and the literature suggests involvement when linked to immunosuppressive causes, such as viral diseases or systemic comorbidities. Still, it can become the cause of bacterial or fungal co-infections due to immunological impairment. In the 3 cases reported, only 1 patient was positive for FeLV. Parasitological examination of the skin by deep or superficial skin scraping, considered as the diagnostic method of choice for demodicosis, was performed in the 3 patients and demonstrated the presence of mites. Although the literature does not provide treatment considered to be of choice for felines, fluralaner was prescribed transdermally for the 3 patients reported here due to its practicality and prolonged time of action, administered in single application as antiparasitic therapy, achieving success and complete improvement up to 30 days after its use. The animals that presented co-infections were treated according to the type of infections they presented and their therapies of choice.

A study was carried out to evaluate the inclusion of increasing levels of a residue from the rendering plant after obtaining meat and bone meal for growing pigs on performance, carcass characteristics, incidence of diarrhea and economic viability of the diets. Forty-eight pigs were allotted in a completely randomized design with four treatments (0, 2, 4 and 6% of residue inclusion), six replicates and two animals per experimental unit (barrows and females) with 30 ± 0.662 kg of initial weight in a 24-day trial period. Statistical assumptions of normality and homogeneity were verified using Shapiro-Wilk and Barllet tests, respectively, at 5% probability. The assumptions were met and the data were subjected to analysis of variance at 5% significance. The performance variables, carcass characteristics and diarrhea incidence did not show significant differences (P<0.05) between the levels of inclusion of residue in the diet. The residue can be used up to 6% in the feed without affecting performance, carcass characteristics and diarrhea incidence in growing pigs, however, the inclusion of 4% of the residue presents greater economic viability.

ABSTRACT: This study assessed the effect of cover crop biomass on the epigeal fauna of an organic eggplant (Solanum melongena L.) cropping system. A randomized block design was used, with four cover crop treatments: brachiaria, crotalaria, millet, and cocktail (brachiaria, crotalaria and millet). The epigeal faunal indices were determined at each plot. The epigeal fauna of a secondary forest site was also assessed for comparison. Epigeal individuals were collected using pitfall traps at two sampling periods (110 and 180 days after mowing cover crops). Soil samples were collected for determination of physical and chemic properties. 2032 individuals were captured in the first sampling and 3806 individuals in the second. The highest values of wealth in the first collection were observed in the millet and cocktail areas. Cluster analysis showed similarity of epigeal faunal groups between millet and brachiaria plots at the first sampling and between cocktail and crotalaria sites at the second sampling. Co-inertia analysis showed a significant covariance between epigeal fauna at the second sampling and soil properties. The cover crops management had a beneficial influence on the activity, wealth and ecological indexes, in both sampling periods. The positive association observed between epigeal faunal groups and soil physical and chemical properties demonstrates the efficiency of cover crops in improving soil quality in organic cropping systems.

Background: Canine vector borne diseases (CVBD) are common in tropical countries where the climate favors arthropods abundance. Comorbidity with one or more CVBD are reported as clinical complication and worsen prognostic. Canine visceral leishmaniosis (CanL) is an endemic zoonotic disease in Brazil caused by Leishmania infantum, with several restrictions to canine treatment and suggestion of reservoirs euthanasia for disease control. Heartworm (HW) is a helminthic disease caused by Dirofilaria immitis infection in dogs. It is a chronic heart disease, which can lead to death by congestive heart failure. Canine ehrlichiosis (CE) is caused by Ehrlichia canis bacterial infection with a zoonotic potential and fatal to dogs in acute and chronic presentations. Exposed the above, this study aims to describe a successful treatment and management of a dog with CanL, CE, and HW comorbidity. Case: A 3-year-old male uncastrated black Labrador dog, weighing 35 kg, was admitted to the veterinary clinic due to immunochromatographic CanL positive test performed by municipal zoonosis control center active surveillance. Clinical exam showed a mild shedding, intermittent eye white/yellow discharge and popliteal lymph nodes enlargement. After positive for CanL, veterinary requested more laboratorial exams. IFAT and ELISA were positive for CanL, blood smear showed presence of microfilaria, and bone marrow cytology showed Ehrlichia spp. morulae and microfilaria. Initial treatment prescribed was oral doxycycline, omeprazole, ranitidine, and domperidone for 30 days, and allopurinol and ivermectin until further recommendation. Additionally, repellent collar, repellent spray and vitamin supplement was indicated. After first month, marbofloxacin for 30 days and three doses of immunostimulant drug were administrated. After three months of treatment, dog still positive for heartworm, ehrlichiosis, and CanL. Doxycycline protocol was repeated. Dog became consistently negative for all pathogens one year later with persistent thrombocytopenia but without clinical signs, ergo allopurinol and ivermectin were discontinued. After 4 years of follow up, the animal had an acute pancreatitis and died, with unremarkable total blood count and negative for all pathogens. Discussion: CVBD coinfections are commonly reported as worsen prognostic in endemic regions. The pathogens reported here share a host immunomodulation competence. L. infantum and Ehrlichia spp. downregulates Th1 response, whereas D. immitis increase as Th2 profile. The therapeutic protocol was iniciated by staging CanL. Since the patient had clinical signs, allopurinol was prescribed as a well-established drug for CanL. Marbofloxacin was added due to its high safety drug in clinical improvement of infected dogs with and without renal disease and in vitro effectiveness against L. infantum. Domperidone was used to promote Th1 cytokine profile as INF-γ, IL-2, IL-12, and TNF-α. We used an immunostimulant protocol to favor polarization to the Th1 profile comprised by 30 days of domperidone protocol followed by a vaccine and an immunomodulator. Doxycycline was used successfully for Ehrlichia spp. and HE clearance after 2 treatment courses and 1 year of ivermectin every 15 days. The animal presented intermittent coughing episodes on the first treatment course, but no medical intervention was needed besides exercise restriction. Our report shows the successful management of one dog with CanL, CE and HE comorbidity. This success was possible due to early detection and good therapeutic choice.(AU)

Several studies have reported the beneficial effects of inoculation of Azospirillum brasilense in wheat, but only a few of them have related the co-inoculation of A. brasilense and Rhizobium sp. and the evaluation of different inoculation methods. This study aimed i) to verify the efficiency of plant growth-promoting bacteria (PGPB) in subtropical environments, ii) to verify the efficiency of co-inoculation of A. brasilense and Rhizobium sp., and iii) to verify the efficiency of the management of different inoculation methods in the wheat crop. The experiments were carried out in Londrina and Apucarana, State of Paraná, Brazil, under a complete randomized block design, with four replications and nine treatments: T1) absence of nitrogen (N) topdressing, T2) 30 kg ha−1 of N topdressing, T3) 60 kg ha−1 of N topdressing, T4) A. brasilense Ab-V5 in the seeds, T5) A. brasilense Ab-V5 in post-emergence, T6) A. brasilense Ab-V5 + Rhizobium sp. 53GRM1 in the seeds, T7) A. brasilense Ab-V5 + Rhizobium sp. 53GRM1 in post-emergence, T8) commercial inoculant (A. brasilense Ab-V5 + Ab-V6) in the seeds, and T9) commercial inoculant (A. brasilense Ab-V5 + Ab-V6) in postemergence. The number of ears per linear meter, number of spikelets, number of grains per spikelet, number of grains per ear, thousand-grain weight, number of spikelets to ears ratio, leaf nitrogen content, and grain yield were evaluated. Leaf N content and yield components showed no alterations due to the inoculation and co-inoculation performed both in the seed and in the post-emergence of seedlings. Treatments T3, T7, and T9 showed the highest means of grain yield (2077.50, 1743.12, and 1660.62 kg ha−1, respectively), demonstrating that co-inoculation with A. brasilense Ab-V5 + Rhizobium sp. 53GRM1 and inoculation with A. brasilense Ab-V5 + Ab-V6, both in post-emergence of seedlings, have the potential to replace the topdressing nitrogen fertilization in wheat.(AU)

Background: The gray brocket deer (Mazama gouazoubira) is a specie that shows great adaptability in different habitats and it is the most abundant deer specie in South America. The present work describes for the first time a case of abortion followed by death associated with colloid goiter, massive hemonchosis and necrotizing rumenitis in a captive female gray brocket deer. Case: A 4-year-old female gray brocket deer (M. gouazoubira) raised in captivity had a history of abortion during the last third of gestation. The animal was kept in an enclosure together with 3 other gray brockets deers, being 1 male of the same age and 2 juvenile brocket deer of approximately 1 and 2 years old. The animals were fed with concentrated used as cattle feed and dewormed annually with 1% Ivermectin. The animals' enclosure had vegetation cover formed by grasses and soil. The animals appeared healthy with no behavioral changes. The day after the stillbirth, the mother was found dead in the enclosure and sent to the animal pathology sector of the University of Vila Velha (UVV), Brazil. Necropsy revealed that thyroid lobules were highly increased in volume and histopathological findings were compatible with colloid goiter. A large number of nematodes were found in the abomasal content, totalizing 11,626 helminths, which were morphologically characterized as Haemonchus contortus. Grossly, the serous and ruminal mucosa exhibited an extensively reddish focal area with irregular contour, surface ulceration and a firm consistency. Microscopically, a severe necrotizing rumenitis was diagnosed. The liver showed pale multifocal areas on the subcapsular surface, friable to the touch which deepened when cut. Histopathological analysis revealed an accentuated multifocal panlobular coagulative necrosis, characterizing an acute liver necrosis. Discussion: Iodine is a mineral of great importance for thyroid hormones synthesis and your requirements are higher during pregnancy and lactation. Diets deficient in iodine causes a reduction in the basal activity of the hormones triiodothyronine (T3) and thyroxine (T4) and over-stimulation of the thyroid by thyroid stimulating hormone (TSH), resulting in goiter. In the present case, it is possible that the shortage of iodine in diet caused a goiter and, as a consequence, triggered the abortion. Haemonchus contortus is a pathogenic nematode of small ruminants, leading to decreased productivity and death in some cases as a result of anemia and hypoxia. The contact between domestic and wild animals, resulting in the emergence of infectious diseases and the spread of pathogens among species. In the present case, manual counting accounted for 11,626 H. contortus larvae, characterizing a massive infection and justifying the condition of severe anemia. The high parasitic load shown in this case points out this parasite's importance related to this species in captivity. In general, inflammatory lesions in the rumen are results of excessive intake of fermentable carbohydrates, which leads to a considerable decrease in ruminal pH and leads to a high proliferation of lactic acid bacteria. This lesion has been previously reported in cervids. This case of comorbidities demonstrates that failures in nutritional and health handling, may cause simultaneous multiple diseases leading to death. Preventive measures for helminth parasite control and a proper feeding management with an adequate diet must be provided in order to preserve the species in captivity.

The objective of this study was to estimate the components of variance and genetic parameters of test-day milk yield in first lactation Girolando cows, using a random regression model. A total of 126,892 test-day milk yield (TDMY) records of 15,351 first-parity Holstein, Gyr, and Girolando breed cows were used, obtained from the Associação Brasileira dos Criadores de Girolando. To estimate the components of (co) variance, the additive genetic functions and permanent environmental covariance were estimated by random regression in three functions: Wilmink, Legendre Polynomials (third order) and Linear spline Polynomials (three knots). The Legendre polynomial function showed better fit quality. The genetic and permanent environment variances for TDMY ranged from 2.67 to 5.14 and from 9.31 to 12.04, respectively. Heritability estimates gradually increased from the beginning (0.13) to mid-lactation (0.19). The genetic correlations between the days of the control ranged from 0.37 to 1.00. The correlations of permanent environment followed the same trend as genetic correlations. The use of Legendre polynomials via random regression model can be considered as a good tool for estimating genetic parameters for test-day milk yield records.(AU)

Abstract Background: Phoneutria nigriventer venom contains Ph1. This toxin and its recombinant form have a remarkable analgesic potential that is associated with blockage of voltage-gated calcium channels and TRPA1 receptors. Although morphine is a mainstay drug to treat moderate and severe pain related to cancer, it has serious and dose-limiting side effects. Combining recombinant Ph1 and morphine to treat pain is an interesting approach that has been gaining attention. Therefore, a quantitative and reliable method to establish the strength of the antinociceptive interaction between these two substances is necessary. The present study was designed to investigate the nature of the functional antinociceptive (analgesic) interaction between Ph1 recombinant toxin and morphine in a model of cancer pain. Methods: Melanoma was produced by intraplantar inoculation of B16-F10 cells into the right paw of C57BL/6J mice. Von Frey filaments measured the paw-withdrawal threshold after intrathecal administration of morphine, recombinant Ph1, and their combination. Thermal hyperalgesia was assessed using Hargreaves apparatus. The degree of interaction was evaluated using isobolographic analysis. Spontaneous and forced motor performance was assessed with the open-field and rotarod tests, respectively. Results: Co-administration of recombinant Ph1 and morphine synergistically reverses the melanoma-induced mechanical hyperalgesia. The potency of the mixture, measured as the effective dose to reach 50% of maximum possible effect (MPE) in ameliorating mechanical hyperalgesia, was about twice fold higher than expected if the interaction between morphine and recombinant Ph1 was merely additive. Treatment with the combination at doses necessary to reach 50% of MPE caused no spontaneous nor forced motor alterations. Conclusion: The combinatorial use of recombinant Ph1 and morphine allows significant and effective dose reduction of both agents, which has translational potential for opioid-sparing approaches in pain management related to cancer.
