Resumo

Zinc (Zn) is a micronutrient considered essential to plants which can be supplied through seed treatment. The treatment of soybean [Glycine max (L.) Merr.] seeds with Zn, however, is still not well known as regards the uptake and mobilization dynamics of the nutrient during the germination process. This study aimed to evaluate the uptake and distribution dynamics of Zn applied to soybean seeds at two levels of vigor during germination using X-ray micro-fluorescence spectroscopy (µ-XRF). Zinc treatments corresponded to 0, 2, 4 and 8 g of Zn per kg of seeds. High and low vigor seeds that had been treated were located appropriately so as to promote germination. Zn intensity measurements with µ-XRF were taken in different parts ("regions") of the seeds (seed coat, cotyledon, and embryonic axis) after 8, 16 and 24 h of imbibition and seedlings (primary root, hypocotyl, plumule, cotyledon, and seed coat) after 48, 72 and 96 h of germination. High vigor seeds showed higher Zn intensity in the embryonic axis in the first 16 h, while low vigor seeds showed higher intensity after 24 h. After 48, 72 and 96 h of germination low vigor seedlings showed higher Zn intensity than high vigor seedlings in the primary root. It was concluded that µ-XRF is an efficient technique for identifying variances in the dynamics of Zn uptake and mobilization during the germination of soybean seeds with different vigor levels.

ABSTRACT: Spermatozoa experience oxidative, osmotic, chemical, and thermal stresses when cooled, which degrade the quality and fertilizing capacity of the cells. Adding antioxidants to the sperm extender mitigates these alterations. This study evaluated the effect of isoespintanol (ISO) on boar semen subjected to cooling. Fifteen ejaculates from five boars (Susscrofadomestica) were extended in Beltsville thawing solution (BTS) supplemented with 0 µM (control), 5 µM (ISO5), 10 µM (ISO10), 15 µM (ISO15), 20 µM (ISO20), 25 µM (ISO25), and 30 µM (ISO30) of ISO, which were then cooled for five days at 16 °C. Sperm kinetics, total motility (TM), and progressive motility (PM) were evaluated every 24 h using an IVOS computer-assisted sperm analysis (CASA) system. On day 1 and day 5 of cooling, a hypoosmotic test, spectrofluorometry, and flow cytometry were performed to evaluate the following: membrane functionality, measured as a function of hypoosmotic swelling (HOS); total antioxidant capacity (TAC); reactive oxygen species (ROS); and mitochondrial membrane potential (Δ¥M). Regression analysis and comparison of means using the Duncan test were performed. The ISO added had a slight impact on sperm motility, as evidenced by a reduction in TM at 24 h of cooling (but not prior) with the addition of 20 µM of ISO. Similarly, no effect of the ISO on the kinetics and functional integrity of the sperm membrane was observed at 96 h of cooling; however, the regression coefficients indicated that the ISO lowered the rate of decrease in sperm motility and the proportion of rapid spermatozoa relative to the concentration of ISO used. The ISO did not affect the TAC of the cooled semen; however, different concentrations of ISO lowered ROS production in the semen after 96 h of cooling. ISO also impacted the Δ¥M of the spermatozoa at 0 h of cooling, increasing the proportion of low Δ¥M cells and decreasing the proportion of high Δ¥M cells. In conclusion, ISO can reduce the loss of quality and oxidative stress occurring in boar semen during cooling and can modulate the mitochondrial activity of sperm.


RESUMO: Durante a refrigeração, os espermatozoides sofrem estresse oxidativo, osmótico, químico e térmico, que diminuem sua qualidade e afetam sua capacidade de fertilização. A adição de antioxidantes ao diluente espermático é uma alternativa para mitigar essas alterações. O objetivo desta pesquisa foi avaliar o efeito do isospintanol (ISO) na refrigeração do sêmen suíno. Quinze ejaculados de cinco varrascos (Sus scrofa domestica) foram diluídos em BTS suplementado com ISO a 0 (controle), 5 (ISO5), 10 (ISO10), 15 (ISO15), 20 (ISO20), 25 (ISO25) e 30 (ISO30) µM e foram refrigerados por cinco dias a 16 °C. A motilidade total (MT), motilidade progressiva (MP) e cinética dos espermatozóides foram avaliadas a cada 24 h com um sistema CASA IVOS. Nos dias um e cinco de refrigeração, foram avaliadas a funcionalidade da membrana, a capacidade antioxidante total (CAT), as espécies reativas de oxigênio (ROS) e o potencial de membrana mitocondrial (Δ¥M), através do teste hiposmótico (HOS), espectrofluorimetría e citometria de fluxo. Foram realizadas análises de regressão e comparação de médias, pelo teste de Duncan. A adição de ISO teve pouca influência na motilidade espermática, apresentando apenas redução na MT em 24 h de refrigeração, devido à adição de 20 µM. Da mesma forma, não foi observada influência de ISO na cinética e integridade funcional da membrana em 96 horas de refrigeração; porém, os coeficientes de regressão mostraram que ISO produziu menor taxa de diminuição da motilidade e proporção de espermatozoides rápidos dependendo da concentração utilizada. ISO não influenciou significativamente na CAT do sêmen refrigerado; entretanto, diferentes concentrações de ISO reduziram a produção de EROs a partir do sêmen após de 96 h de refrigeração. ISO também influenciou o Δ¥M dos espermatozóides em 0 h de refrigeração, com aumento das células de baixo Δ¥M e diminuição das células de alto Δ¥M. Em conclusão, o isospintanol pode reduzir a perda da qualidade e o estresse oxidativo do sêmen suíno durante a refrigeração e pode modular a atividade mitocondrial do esperma.

Resumo

Purpose: Fluorescence spectroscopy techniques have been investigated aiming to reduce the invasiveness of methods for investigation of tissue. In transplantation procedures, it may offer the possibility of a complementary technique for the monitoring of liver grafts' conditions prior to and during the transplantation procedure stages involving cold perfusion. The objective of this study was to evaluate fluorescence spectroscopy under violet light excitation (408 nm) for the monitoring of clinical hypothermic liver transplantation procedures. Methods: Organ grafts were monitored from before the removal of the donor's body to 1 h after the implant into the receptor's body. Fluorescence spectroscopy was assessed over five stages within these transplant stages. Results: The study provided evidence of a correlation between fluorescence information collected during liver grafts transplantation and the survival of patients. Conclusions: Fluorescence spectroscopy can become a tool to monitor transplantation grafts, providing objective information for the final decision of surgeons to use organs.

Intensive management of tropical pastures has shown potential for greenhouse gas (GHG) mitigation due to high forage production and C accumulation in the soil. This study aimed to evaluate different pasture management options in relation to their effect on soil C stocks and soil organic matter (SOM) humification. Pastures in four beef cattle production systems were assessed: intensive and irrigated pasture with high stocking rate (IHS); dryland pasture with high stocking rate (DHS); dryland pasture with moderate stocking rate (DMS); degraded pasture (DP). The soil under the native forest was also evaluated and soil carbon stocks from the 0-100 and 0-30 cm layers were assessed. Carbon stocks (0-100 cm) ranged from 99.88 to 142.33 Mg ha1 in DP and DMS, respectively and were, respectively, 14 % and 24 % higher compared to the soil under the forest and indicate the capacity of adequately managed tropical pastures to mitigate GHG emissions from livestock production. Humification indexes indicated the presence of more labile C in pastures with greater C accumulation (DHS and DMS), mainly in the upper soil layers, indicating recent C accumulation resulting from correct management. However, more labile C can be easily lost to the atmosphere as CO2, depending on pasture management. Low C stocks associated with high humification indexes are characteristics of DP in which significant amounts of SOM are lost. It is necessary to develop technologies to improve C sequestration in IHS and results indicate the importance of quantifying C stocks in association with C stability.

The Brazilian coffee production plays important role both in the national and oversea trade. The quality and sanity of coffee depend on fungal contamination, with emphasis on ochratoxin A (OTA) producing species, which is a nephrotoxin with hazardous effect in public health. The exigency on continuous monitoring and control of OTA in agricultural commodity requires a simple, rapid and accessible analytical methodology. The objective of this research was to set up the spectrofluorimetric method for OTA detection intended for monitoring in coffee, which was developed in data-correlation with high efficiency liquid chromatography (HPLC). The performance of spectrofluorimetric analysis was evaluated using artificially contaminated coffee (10, 20 and 50 ng/g), and the data compared with HPLC. Although of the fault of former technique, with recovery of 40.14 % and correlation coefficient of 0.79 when compared to HPLC, i.e. low reliability and inadequate efficiency, the data showed perspective to follow the improvement inserting new tool, and further standardization and certification.


A cafeicultura brasileira apresenta grande importância ao mercado interno e comércio exterior. A qualidade e sanidade do café dependem da contaminação fúngica, com ênfase as espécies produtoras de ocratoxina A (OTA), uma nefrotoxina de efeito deletério à saúde pública. A necessidade de contínuo monitoramento e controle de OTA em produtos agrícolas requer metodologia analítica simples, rápida e acessível. O trabalho teve como objetivo padronizar um método espectrofluorimétrico para detecção de OTA visando monitoramento em grãos de café, correlacionando-o com a cromatografia líquida de alta eficiência (CLAE). A aplicabilidade da espectrofluorimetria foi avaliada em café artificialmente contaminado com OTA (10, 20 e 50 ng/g), comparando os resultados obtidos com a CLAE. Embora a fluorimetria tenha apresentado resultados insatisfatórios, com recuperação de 40,14 % e coeficiente de correlação de 0,79 em relação a CLAE, i.e. baixa confiabilidade e eficiência inadequada, a técnica constitui perspectiva para prosseguir o estudo inserindo novos ensaios referentes à sua padronização e certificação.
