Resumo

Microbial contamination of the wort during the fermentation process causes significant losses in ethanol production worldwide and creates a dependence of the industry on chemicals and antibiotics to control contamination. Therefore, this study used electron beam (e-beam) to disinfect wort from sugarcane (Saccharum officinarum L.) molasses and investigate the bioethanol fermentation. Four treatments (T0 - T3) were carried out using ionizing doses of radiation through the electron accelerator: 0 (control), 10, 20, and 40 kGy. Total mesophiles, total bacteria, sucrose, glucose, fructose, phenolics, flavonoids, hydroxymethylfurfural (5-HMF), and Furfural were measured. An alcoholic fermentation assay was performed after the irradiation process. The irradiated treatments showed no inversion of sugars and formation of the inhibitory by-products flavonoids, furfural and 5-HMF, except for the phenolic compounds. The lower dose tested (10 kGy) reduced more than 99.9 % of the total mesophiles and more than 99.99 % of the total bacteria in the substrate. In the fermentation, the irradiated worts presented similar (p > 0.05) yields (92, 93, and 94 %) and ethanol productivity levels (0.89, 0.88, and 0.87 g L-1 h-1, for T1, T2, and T3 respectively). However, all treatments presented higher yields and productivity (p < 0.05) when compared to the control (88 % and 0.85 g L-1 h-1), highlighting the possible use of e-beam in wort fermentation at a lower dose (10 kGy). This allows reduction in losses caused by microbial contamination, besides increasing fermentation yield and productivity with lower energy consumption.(AU)

The phenolic extract was prepared with a solution of methanol/water/formic acid. The following analyses were performed every 15 days: contents of total phenolic compounds by Folin-Ciocauteau, cyanidin-3-glycoside and cyanidin-3-rutiniside by HPLC and antioxidant capacity (ABTS and DPPH). Total phenolics and anthocyanins decreased with the increasing irradiation dose and storage time. Pasteurization did not affect the phenolic concentration immediately after processing. However, the contents of TPC and cyanidin-3-rutiniside were reduced during storage of the pasteurized pulp. No processed samples presented characteristics similar to the control at the end of storage.


RESUMO: Os efeitos da radiação gama e pasteurização na estabilidade das antocianinas e na capacidade antioxidante durante o armazenamento da polpa de jussara foram avaliados. A polpa de jussara foi dividida em diferentes tratamentos: controle (sem tratamento), polpa irradiada (2, 4, 6, 8 kGy) e pasteurizada polpa (92 ºC / 1 minuto). Os tratamentos foram armazenados a 4ºC por 60 dias. O extrato fenólico foi preparado com uma solução de metanol / água / ácido fórmico. As seguintes análises foram realizadas a cada 15 dias: teores de compostos fenólicos totais por Folin-Ciocauteau, cianidina 51 3-glicosídeo e cianidina-3-rutinisida por HPLC e capacidade antioxidante (ABTS e DPPH). Os fenólicos totais e as antocianinas diminuíram com o aumento da dose de irradiação e do tempo de armazenamento. A pasteurização não afetou a concentração fenólica imediatamente após o processamento. No entanto, os teores de TPC e cianidina-3-rutinisida foram reduzidos durante o armazenamento da polpa pasteurizada. Nenhuma amostra processada apresentou características semelhantes ao controle ao final do armazenamento.

Resumo

The potential of chitosan as a blood lead chelator and an antioxidant had been proven, yet its ability was still not optimal. Chitosan had a relatively large molecular size, which reduced the effectivity of its distribution towards the tissues. Gamma Co-60 irradiation technique was presumably applicable to solve the issue. The antioxidant activity of chitosan could also be optimized by the addition of vitamin E. This study aimed to analyze the antioxidant activity of the combination of Gamma Co-60 irradiated chitosan and vitamin E in lead acetate-induced rats. Twenty-four rats, which were distributed in six groups, were treated using the combination of gamma Co-60 irradiated chitosan at a dose of 150 kGy and vitamin E 1000 IU. All groups, except for the naïve group, were induced with lead acetate. The positive control group was induced with only lead acetate, while treatment group 1 had an additional treatment of irradiated chitosan. The treatment groups 2-4 were treated using the combination of irradiated chitosan and vitamin E in increasing doses respectively for forty days. Blood serum was collected for measurement of Superoxide dismutase (SOD), Catalase (CAT), Glutathione peroxidase (GPx), and Malondialdehyde (MDA). The results showed that the providedtreatment increased the enzymatic activity of SOD, CAT, and GPx, and reduced the MDA level in lead acetate-induced rats. However, as the vitamin E dosage was increased, it posed several side effects. It was concluded that the combination Gamma Co-60 irradiated chitosan and vitamin E increased the activity of various endogenous antioxidant enzymes and decreased lipid peroxidation, dependent on the amount of vitamin E.(AU)

Resumo

Background: The most common primary brain neoplasm is meningioma. Dolichocephalic breeds are predisposed and there is no sexual predilection. Clinical signs depend on the location and size of the tumor and have a progressive course. Primary treatement includes surgery, radiotherapy or both. This study aimed to describe the treatment of a dog with suspected intracranial meningioma with definitive radiotherapy, which resulted in significant clinical improvement and prolonged survival. Case: A 9-year-old Shetland Shepherd bitch was diagnosed with a head tilt to the left side that progressed over a few weeks. She previously received corticosteroid therapy, which resulted a clinical improvement that worsened after treatment was discontinued. Computed tomography revealed an extra-axial brain mass in the caudal fossa, lateralized to the right, welldelimited, and measuring approximately 1.5 × 1.4 × 1.7 cm, suggestive of intracranial meningioma. The patient was treated with radiotherapy using Cobalt-60 equipment, with 18 daily fractions of 2.5 Gy at a total dose of 45 Gy using parallel and opposite technique fields. A new tomography performed 30 days after treatment showed a remission of 85% of the initial brain mass measuring approximately 0.9 × 1.0 × 0.5 cm, as well as complete resolution of the clinical signs initially presented. After 14 months, the patient presented with signs of lethargy and ataxia and was medicated with hydroxyurea at a dose of 50 mg/kg 3 times a week and corticosteroid therapy. However, the patient's neurological condition deteriorated, and she was subjected to reirradiation using the same protocol used previously, which resulted in clinical improvement and a 54% reduction in tumor volume on magnetic resonance. As a late side effect, only permanent alopecia in the irradiated region was observed. The patient died of disease 330 days after the second course of radiotherapy, with a total survival time of 1087 days. Discussion: Meningiomas are extra-axial neoplasms of the central nervous system that grow inside the dura mater. The literature shows that meningiomas are more common in dolichocephalic races with a mean age of 9 years, which supports our findings. Meningiomas most commonly affect the cortical thalamus and cerebellopontine region in dogs, which are normally associated with vestibular symptoms, as seen in this case. Diencephalic damage can result in vestibular signals since the thalamus functions as a relay station for vestibular afferent stimuli that are relayed to the cerebral cortex. In addition to the vestibular syndrome, common clinical signs associated with meningiomas in dogs include seizures, behavioral changes, and walking in circles, which are frequently misinterpreted due to tumor-induced side effects, such as cerebral edema, obstructive hydrocephalus, and cerebral hernia. Advanced imaging techniques should be used to diagnose intracranial neoplasms. In this case, computed tomography was critical for diagnosis and treatment planning. Meningioma treatment may comprise palliative measures, surgery, and radiotherapy. Radiotherapy as a single treatment can improve the quality of life with a decrease in clinical signs and a median survival time of approximately 250-536 days, as reported in the literature. Hydroxyurea can be a therapeutic option in inoperable cases and for patients with clinical limitations to undergo successive anesthesia during radiotherapy. Its most serious side effect is progressive myelosuppression. It can cause temporary partial tumor remission and improvement in clinical signs. As previously stated, radiotherapy can be an effective primary treatment option for treating intracranial meningiomas in dogs, with significant improvement in neurological clinical signs and mild side effects.

Tetanus toxin blocks the release of the inhibitory neurotransmitters in the central nervous system and causes tetanus and its main form of prevention is through vaccination. The vaccine is produced by inactivation of tetanus toxin with formaldehyde, which may cause side effects. An alternative way is the use of ionizing radiation for inactivation of the toxin and also to improve the potential immunogenic response and to reduce the post-vaccination side effects. Therefore, the aim of this study was to characterize the tetanus toxin structure after different doses of ionizing radiation of 60Co. Methods Irradiated and native tetanus toxin was characterized by SDS PAGE in reducing and non-reducing conditions and MALD-TOF. Enzymatic activity was measured by FRET substrate. Also, antigenic properties were assessed by ELISA and Western Blot data. Results Characterization analysis revealed gradual modification on the tetanus toxin structure according to doses increase. Also, fragmentation and possible aggregations of the protein fragments were observed in higher doses. In the analysis of peptide preservation by enzymatic digestion and mass spectrometry, there was a slight modification in the identification up to the dose of 4 kGy. At subsequent doses, peptide identification was minimal. The analysis of the enzymatic activity by fluorescence showed 35 % attenuation in the activity even at higher doses. In the antigenic evaluation, anti-tetanus toxin antibodies were detected against the irradiated toxins at the different doses, with a gradual decrease as the dose increased, but remaining at satisfactory levels. Conclusion Ionizing radiation promoted structural changes in the tetanus toxin such as fragmentation and/or aggregation and attenuation of enzymatic activity as the dose increased, but antigenic recognition of the toxin remained at good levels indicating its possible use as an immunogen. However, studies of enzymatic activity of tetanus toxin irradiated with doses above 8 kGy should be further analyzed.(AU)

Phytochemical and antioxidant activity of quinoa flour was evaluated after subjected to gamma irradiation processes at dose 3 and 6 kGy. Both non-irradiated and irradiated quinoa samples were subjected to successive extractions in ethanol solvent. The antioxidant activity after gamma irradiation treatment was investigated via Ferric reducing antioxidant power (FRAP) and radical-scavenging activity (RSA) using 2,2-diphenyl-1-picrylhydrazyl (DPPH).Total phenolic and flavonoid content were analyzed using FolinCiocalteu micro-method, aluminium chloride (AlCl3) method and High Performance Liquid Chromatography (HPLC). As well as, effect of irradiation treatment on saponin was also evaluated. Irradiation treatment showed slight differences in the saponin content after exposure to 3 and 6 kGy. Irradiation process enhanced both total phenolic content (TPC) and Total flavonoid content (TFC), TPC were 34.52 and 30.92 mg Gallic Acid Equivalent (GAE)/100g compared to 26.25 mg GAE/100g in non-irradiated quinoa. TFC were 67.44 and 62.89 mg Quercetin Equivalents (QE)/100g compared to 53.15 mg QE/100g. Irradiation dose 3 kGy significantly (p> 0.05) decreased the IC50 as DPPH-RSA and increased the FRAP.(AU)


Atividades fitoquímica e antioxidante da farinha de quinoa foram analisadas após submissão a processos de irradiação gama nas doses 3 e 6 kGy. As amostras de quinoa não irradiadas e irradiadas foram submetidas a extrações sucessivas em solvente etanol. A atividade antioxidante, após o tratamento com irradiação gama, foi investigada por meio do poder antioxidante redutor férrico (ARF) e da atividade de eliminação de radicais (AER) usando 2,2-difenil-1-picril-hidrazil (DPPH). O conteúdo fenólico total e o teor de flavonoide foram analisados usando o método de Folin-Ciocalteu, método de cloreto de alumínio (AlCl3) e cromatografia líquida de alta eficiência (CLAE). Além disso, o efeito do tratamento de irradiação na saponina também foi avaliado. O tratamento por irradiação não mostrou diferenças significativas no conteúdo de saponina após exposição a 3 e 6 kGy. O processo de irradiação aumentou o conteúdo fenólico total (CFT) e o teor total de flavonoides (TTF); o CFT foi de 34,52 e 30,92 mg de equivalente de ácido gálico (EAG) / 100 g em comparação com 26,25 mg de EAG / 100 g na quinoa não irradiada. Os TTF foram 67,44 e 62,89 mg de equivalentes de quercetina (EQ) / 100 g em comparação com 53,15 mg de EQ / 100 g. A dose de irradiação de 3 kGy diminuiu significativamente (p > 0,05) o IC50 como DPPH-AER e aumentou o ARF.(AU)

ABSTRACT Purpose: To evaluate the low-level laser therapy (LLLT) on the membrane induced by the Masquelet technique in rabbits. Methods: Twelve Norfolk rabbits at approximately 3 months of age were used. A 1-cm segmental defect was induced in both radii, which were filled with polymethylmethacrylate cylinder. LLLT was used postoperatively in the bone defect of one of the forelimbs every 48 hours for 15 days. Six rabbits were euthanatized on third and sixth postoperative weeks. Results: In both forelimbs, radiographs showed new bone growth from radius cut ends on the third postoperative week and more advanced stage on the sixth postoperative week. Ultrasound showed induced membrane one week after the surgery. Histologically, there were no significant differences in the semi-quantitative score of inflammation intensity, total number of blood vessels, bone metaplasia, and collagen. The average thicknesses were 2,050.17 and 1,451.96 μm for control membranes and 2,724.26 and 2,081.03 μm for irradiated membranes, respectively, on third and sixth postoperative weeks. Vascular endothelial growth factor A (VEGF-A) and platelet derived growth factor (PDGF) expression were present in the induced membranes of control and irradiated forelimbs, but there was no significant difference. Conclusions: Based on assessment methods, it was not possible to demonstrate the effect of LLLT on the induced membrane.

Resumo

Six different bread wheat genotypes; two Egyptian commercial varieties (control); Giza-168 and Gemmeiza-11, and four promising lines; L84 and L148, resulted via hybridization and M10 and M34 via radiation mutation program) were rheologically evaluated using extensograph and for protein, analysis using sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE). The radiation mutant M10 and M34 had the highest maximum resistance which is a very good indicator of strong gluten. The amount of gluten content was higher in M10, L148, and M34 compared to the control samples Gz168 and Gm11. Sulfide amino acids (CYS and MET) are slightly higher in M10. The electrophoretic results and amino acid analyzers show that the best technological quality was exhibited by M10. Radiation mutants wheat genotypes have a protein with good characteristics, mainly gluten which is significantly higher compared to control samples. The rheological properties measured as extensograph and gel electrophoresis were much better in irradiated lines M10 and M34.(AU)


Seis diferentes genótipos de trigo de pão, duas variedades comerciais egípcias (controle) Giza-168 e Gemmeiza-11 e quatro linhas promissoras L84 e L148, obtidas via hibridação, e M10 e M34, via programa de mutação por radiação foram avaliados reologicamente por meio de extensógrafo, enquanto, para proteínas, foram feitas análises utilizando eletroforese em gel de poliacrilamida com dodecilsulfato de sódio (SDS-PAGE). Os mutantes de radiação M10 e M34 apresentaram a maior resistência máxima, o que é um indicador muito bom de glúten forte. A quantidade de glúten foi maior em M10, L148 e M34 em comparação com as amostras de controle Gz168 e Gm11. Os aminoácidos sulfurados (CYS e MET) são um pouco mais altos no M10. Os resultados eletroforéticos e analisadores de aminoácidos mostram que a melhor qualidade tecnológica foi exibida pelo M10. Os genótipos de trigo mutantes da radiação possuem uma proteína com boas características, principalmente o glúten, que é significativamente maior em comparação às amostras do grupo controle. As propriedades reológicas medidas, como extensógrafo e eletroforese em gel, foram muito melhores nas linhas irradiadas M10 e M34.(AU)

O cimento ósseo de polimetilmetacrilato é considerado um material padrão como carreador de antibiótico em cirurgias ortopédicas. A energia ultrassônica é um método capaz de gerar efeitos biológicos por mecanismos térmicos e não térmicos. Este estudo teve como objetivo analisar a dispersão de azul de metileno em pérolas de polimetilmetacrilato combinada com a exposição em campo acústico gerado por ultrassom não térmico. Foram utilizados 49 corpos-de-prova, cada um composto por uma pérola de polimetilmetacrilato (0,6 mm de diâmetro) dopada com azul de metileno e depositada em amostra de gelatina. Quarenta espécimes foram divididos em quatro grupos de 10 amostras com base nas intensidades de ultrassom (Grupo 1: 1,0 W/cm2; Grupo 2: 1,5 W/cm2) e profundidade (A - 2 cm; B - 3 cm) dos grânulos de polimetilmetacrilato na amostra de gelatina. Os outros nove espécimes constituíram o grupo controle. O grupo controle diferiu estatisticamente dos outros grupos. Todos os grupos irradiados com ultrassom apresentaram diferenças significativas, exceto os Grupos 2A e 2B. A dispersão de azul de metileno na gelatina entre os grupos foi: 1A> 1B; 2A> 1A; 2B> 1A; 2A> 1B; 2B> 1B. Ultrassom de baixa intensidade permitiu maior dispersão de azul de metileno com polimetilmetacrilato posicionado mais superficialmente, e não ocorreu influência da profundidade da pérola com ultrassom de alta intensidade.


Polymethylmethacrylate bone cement is a standard material used as antibiotic carrier in the orthopedic surgery. The ultrasonic energy method is capable of triggering biological effects based on both thermal and non-thermal mechanisms. The aim of the current study is to analyze methylene blue dispersion in polymethylmethacrylate beads, in association with the acoustic field generated by non-thermal ultrasound. Forty-nine specimens were used, and each specimen comprised one polymethylmethacrylate bead (0.6-mm diameter) doped with methylene blue and deposited in gelatin sample. Forty test specimens were divided into four groups comprising 10 samples, each, based on different ultrasound intensities (Group 1: 1.0 W/cm2; Group 2: 1.5 W/cm2) and polymethylmethacrylate bead depths (A - 2 cm; B - 3 cm) in gelatin sample. The control group comprised other nine specimens and statistically differed from the other groups. All groups irradiated with ultrasound have shown statistically significant differences in methylene blue dispersion, except for Groups 2A and 2B. Methylene blue dispersion in gelatin among groups was 1A> 1B; 2A> 1A; 2B> 1A; 2A> 1B; and 2B> 1B. Low-intensity ultrasound enabled the highest methylene blue dispersion when polymethylmethacrylate bead was positioned superficial; bead depth associated with high-intensity ultrasound did not influence methylene blue dispersion.

Background: Laser photobiomodulation has been used in the treatment of various injuries and diseases. This promotes modulation of the inflammatory process, edema reduction and devitalized tissue regeneration. The advantages of Antimicrobial photodynamic therapy are its easy application and the absence of side effects. Other advantages are the cost of the therapy, minimal damage to animal tissue, the broad spectrum of action, and efficiency against strains resistant to antimicrobials. The aim of this study was to report the clinical and their resolution in a female dog with a traumatic, infected wound treated with laser phototherapy as an alternative therapy method. Case: A 3-year-old bitch Border Collie, weighing 18 kg, from the municipality of Ilhéus, Bahia, Brazil, waspresented for examination with a history of traumatic laceration of the left thoracic member. On the anamnesis, it was reported that the patient presented with laceration of the left thoracic member. The wound was cleaned and an antibiotic [30 mg/kg of 12/12 h] and anti-inflammatory [0.1 mg/kg every 24 h were prescribed, both for 5 days]. Twenty-four h after the surgical procedure, there was dehiscence of the sutures, with daily topical therapy based on zinc oxide for secondary intention healing. Upon physical examination, the wound was found to be contaminated with swollen and erythematous edges, an ulcerated area with devitalized tissue, serous exudate, and 8.8 cm2 of wounded area. Given the macroscopic characteristics of the lesion, phototherapy was associated with conventional therapy until complete healing of the wound, with three weekly applications at 48 h intervals. Initially, the wound was cleaned with saline solution at 0.9% and a single treatment with aPDT was scheduled due to the high degree of contamination. The dosimetry parameters of irradiation were calculated according to the wounded area with a diode laser of 0.1W of power, continuous emission, spot area of 0.028 cm2, and energy of 9 J per application point. A gauze imbibed with 1 mL of methylene blue aqueous solution (300 µM), which was the photosensitizer was applied to the lesion, with a pre-irradiation time of 5 min, after which it was irradiated with red laser (RL) (λ = 660nm) for 90 s per point, using the sweeping technique. The edge of the lesion was irradiated with infrared laser (IRL) (λ = 808 nm), total energy of 5 J, using the technique of specific points and 1 J of energy/point. After aPDT, low-intensity laser therapy (LILT) sessions were set up with RL and IRL, with energies of 0.5 J/point and 1 J/point, respectively. The wound was cleaned daily, protected with a bandage, and clinically evaluated until complete regression. Discussion: In the present case, methylene blue was used as a photosensitizer, but it is noteworthy that, apparently, each microorganism responds differently to photosensitizers. Thus, the therapy becomes specific for each application, for example: the type of photosensitizer, its concentration, pre-irradiation time, type of light used in photosensitization, wavelength, energy, power, mode delivery of light. Thus, for the best result, the specific protocol in each application should be used Low-intensity laser therapy is an easily executed technique with effective results. The use of PDT associated with photobiomodulation therapy enabled rapid healing of the cutaneous wound, in addition to an improvement in clinical signs and pain caused by the lesion. The technique proved to be an efficient alternative in the treatment of wounds, whether used in isolation or associated with conventional therapy.

The ultrasonic energy method is capable of triggering biological effects based on both thermal and non-thermal mechanisms. The aim of the current study is to analyze methylene blue dispersion in polymethylmethacrylate beads, in association with the acoustic field generated by non-thermal ultrasound. O cimento ósseo de polimetilmetacrilato é considerado um material padrão como carreador de antibiótico em cirurgias ortopédicas. A energia ultrassônica é um método capaz de gerar efeitos biológicos por mecanismos térmicos e não térmicos. Este estudo teve como objetivo analisar a dispersão de azul de metileno em pérolas de polimetilmetacrilato combinada com a exposição em campo acústico gerado por ultrassom não térmico. Foram utilizados 49 corpos-de-prova, cada um composto por uma pérola de polimetilmetacrilato (0,6 mm de diâmetro) dopada com azul de metileno e depositada em amostra de gelatina. Quarenta espécimes foram divididos em quatro grupos de 10 amostras com base nas intensidades de ultrassom (Grupo 1: 1,0 W/cm2; Grupo 2: 1,5 W/cm2) e profundidade (A - 2 cm; B - 3 cm) dos grânulos de polimetilmetacrilato na amostra de gelatina. Os outros nove espécimes constituíram o grupo controle. O grupo controle diferiu estatisticamente dos outros grupos. Todos os grupos irradiados com ultrassom apresentaram diferenças significativas, exceto os Grupos 2A e 2B. A dispersão de azul de metileno na gelatina entre os grupos foi: 1A> 1B; 2A> 1A; 2B> 1A; 2A> 1B; 2B> 1B. Ultrassom de baixa intensidade permitiu maior dispersão de azul de metileno com polimetilmetacrilato posicionado mais superficialmente, e não ocorreu influência da profundidade da pérola com ultrassom de alta intensidade.

Background: Otitis is a severe inflammation of the skin of the auditory canal which can impact animals of all ages. Insheep, this disease can occur in isolated cases or in the entire flock. Laser photobiomodulation can be used in combination with medication or as single therapy and the effects are analgesia, modulation of the inflammatory process, edemareduction, tissue restoration and stimulation of local microcirculation. Antimicrobial photodynamic therapy stands out asa promising alternative to antimicrobial drugs for the treatment of localized infections. This study aimed to report the useof laser phototherapies on a sheep with bacterial otitis.Case: A case of bacterial otitis in a 4-year-old sheep, Dorper, was treated at the Veterinary Hospital of the State Universityof Santa Cruz. In the anamnesis, the owner reported that the patient presented crusts in the auricular pavilions, and hadbeen medicated with oxytetracycline 20%, administered intramuscularly, for two days and the crusts cleaned with iodine10%, but the animal did not present clinical improvement. On physical examination, the animal presented constant headswaying, edema of the ears, otalgia, enlarged parotid lymph nodes and ear wounds. No ear discharge was observed, andthe initial diagnosis was aural hematoma. The treatment prescribed was laser photobiomodulation for three consecutivedays. The lesions were irradiated with a diode laser with a power of 0.1 W, irradiance of 3.5 W/cm2, continuous emission,spot area of 0.028 cm2, wavelength of 808 nm, energy of 4J/point, and fluency/point of 142.8J/cm2, with seven pointson the external surface and four points on the internal surface of both ears. Two points were also targeted on the parotidlymph nodes with infrared laser (λ = 808 nm), with energy of 2J/point. On the fourth day of hospitalization, there was areduction in lesions, decreased edema and absence of pain and on the sixth day...

Resumo

Abstract Phytochemical and antioxidant activity of quinoa flour was evaluated after subjected to gamma irradiation processes at dose 3 and 6 kGy. Both non-irradiated and irradiated quinoa samples were subjected to successive extractions in ethanol solvent. The antioxidant activity after gamma irradiation treatment was investigated via Ferric reducing antioxidant power (FRAP) and radical-scavenging activity (RSA) using 2,2-diphenyl-1-picrylhydrazyl (DPPH).Total phenolic and flavonoid content were analyzed using FolinCiocalteu micro-method, aluminium chloride (AlCl3) method and High Performance Liquid Chromatography (HPLC). As well as, effect of irradiation treatment on saponin was also evaluated. Irradiation treatment showed slight differences in the saponin content after exposure to 3 and 6 kGy. Irradiation process enhanced both total phenolic content (TPC) and Total flavonoid content (TFC), TPC were 34.52 and 30.92 mg Gallic Acid Equivalent (GAE)/100g compared to 26.25 mg GAE/100g in non-irradiated quinoa. TFC were 67.44 and 62.89 mg Quercetin Equivalents (QE)/100g compared to 53.15 mg QE/100g. Irradiation dose 3 kGy significantly (p> 0.05) decreased the IC50 as DPPH-RSA and increased the FRAP.


Resumo Atividades fitoquímica e antioxidante da farinha de quinoa foram analisadas após submissão a processos de irradiação gama nas doses 3 e 6 kGy. As amostras de quinoa não irradiadas e irradiadas foram submetidas a extrações sucessivas em solvente etanol. A atividade antioxidante, após o tratamento com irradiação gama, foi investigada por meio do poder antioxidante redutor férrico (ARF) e da atividade de eliminação de radicais (AER) usando 2,2-difenil-1-picril-hidrazil (DPPH). O conteúdo fenólico total e o teor de flavonoide foram analisados usando o método de Folin-Ciocalteu, método de cloreto de alumínio (AlCl3) e cromatografia líquida de alta eficiência (CLAE). Além disso, o efeito do tratamento de irradiação na saponina também foi avaliado. O tratamento por irradiação não mostrou diferenças significativas no conteúdo de saponina após exposição a 3 e 6 kGy. O processo de irradiação aumentou o conteúdo fenólico total (CFT) e o teor total de flavonoides (TTF); o CFT foi de 34,52 e 30,92 mg de equivalente de ácido gálico (EAG) / 100 g em comparação com 26,25 mg de EAG / 100 g na quinoa não irradiada. Os TTF foram 67,44 e 62,89 mg de equivalentes de quercetina (EQ) / 100 g em comparação com 53,15 mg de EQ / 100 g. A dose de irradiação de 3 kGy diminuiu significativamente (p > 0,05) o IC50 como DPPH-AER e aumentou o ARF.

Objetivou-se com esse estudo avaliar os efeitos de diferentes métodos de esterilização e fontes de óleo (salmão e linhaça) em formulação de rações sobre os parâmetros bioquímicos e hematológicos de ratos Wistar e camundongos NIH em diferentes idades. Foram avaliados 336 animais, sendo 168 de cada biomodelo. Houve efeito significativo das fontes de óleo sobre os parâmetros bioquímicos plasmáticos em camundongos NIH e ratos Wistar (P 0,05). Conclui-se que o óleo de salmão e linhaça podem ser utilizados nas rações para camundongos NIH e ratos Wistar, independente do método de esterilização adotado.
