Resumo

Purpose: Remote ischemic preconditioning (RIPC) confers cardioprotection against ischemia reperfusion (IR) injury. However, the precise mechanisms involved in RIPC-induced cardioprotection are not fully explored. The present study was aimed to identify the role of melatonin in RIPC-induced late cardioprotective effects in rats and to explore the role of H2 S, TNF-α and mitoKATP in melatoninmediated effects in RIPC. Methods: Wistar rats were subjected to RIPC in which hind limb was subjected to four alternate cycles of ischemia and reperfusion of 5 min duration by using a neonatal blood pressure cuff. After 24 h of RIPC or ramelteon-induced pharmacological preconditioning, hearts were isolated and subjected to IR injury on the Langendorff apparatus. Results: RIPC and ramelteon preconditioning protected the hearts from IR injury and it was assessed by a decrease in LDH-1, cTnT and increase in left ventricular developed pressure (LVDP). RIPC increased the melatonin levels (in plasma), H2 S (in heart) and decreased TNF-α levels. The effects of RIPC were abolished in the presence of melatonin receptor blocker (luzindole), ganglionic blocker (hexamethonium) and mitochondrial KATP blocker (5-hydroxydecanoic acid). Conclusion: RIPC produce delayed cardioprotection against IR injury through the activation of neuronal pathway, which may increase the plasma melatonin levels to activate the cardioprotective signaling pathway involving the opening of mitochondrial KATP channels, decrease in TNF-α production and increase in H2 S levels. Ramelteon-induced pharmacological preconditioning may also activate the cardioprotective signaling pathway involving the opening of mitochondrial KATP channels, decrease in TNF-α production and increase in H2 S levels.

Cattle are considered intermediate hosts of Sarcocystis, which can cause clinical signs and lower performance in the acute phase of infection. Sarcocystis spp. are usually not visible to the naked eye during the post mortem inspection. Moreover, fresh microscopic examination and transmission electron microscopy techniques are difficult to apply to large samples. Therefore, extensive studies on Sarcocystis infection in cattle using molecular and serological methods are required. Here, we investigated Sarcocystis spp. infection in cattle using fresh microscopic examination and polymerase chain reaction of myocardium samples and compared the results with the presence of antibodies against Sarcocystis spp. in corresponding serum samples detected using indirect fluorescent antibody test. Microscopic Sarcocystis were observed in 100% of the myocardial samples, and Sarcocystis DNA was present in 86% (43/50) of these samples. Antibodies against Sarcocystis spp. were detected in 96% (48/50) and 80% (40/50) of the serum samples at 1:25 and 1:200 dilutions, respectively. The three associated methods (fresh microscopic examination, PCR and serology) showed good sensitivity and detection for Sarcocystis spp. compared with fresh microscopic examination (only), and they may facilitate diagnosis in live animals on a large scale as well as monitoring of the herd status.


Os bovinos são considerados hospedeiros intermediários de Sarcocystis, podendo causar sinais clínicos e menor desempenho na fase aguda da infecção. Sarcocystis spp. geralmente não são visíveis a olho nu durante a inspeção post mortem. Além disso, o exame microscópico a fresco e as técnicas de microscopia eletrônica de transmissão são difíceis de aplicar a uma amostras de grande tamanho. Portanto, são necessários extensos estudos sobre a infecção por Sarcocystis em bovinos usando métodos moleculares e sorológicos. Aqui, investigamos a infecção de Sarcocystis spp. em bovinos por meio de exame microscópico a fresco e reação em cadeia da polimerase de amostras de miocárdio e comparado os resultados com a presença de anticorpos contra Sarcocystis spp. em amostras de soro correspondentes detectadas usando o teste de anticorpos fluorescentes indiretos. Sarcocistos microscópicos foram observados em 100% das amostras de miocárdio, e o DNA de Sarcocystis estava presente em 86% (43/50) dessas amostras. Anticorpos contra Sarcocystis spp. foram detectados em 96% (48/50) e 80% (40/50) das amostras de soro nas diluições 1:25 e 1:200, respectivamente. Os três métodos associados (exame microscópico a fresco, PCR e sorologia) mostraram boa sensibilidade e detecção para Sarcocystis spp. em comparação com o exame microscópico fresco (apenas) e podem facilitar o diagnóstico em animais vivos em larga escala, bem como o monitoramento do status do rebanho.

Resumo

Purpose: Myocardial ischemia/reperfusion (MI/R) injury refers to a pathological condition of treatment of myocardial infarction. Oxidative stress and inflammation are believed to be important mechanisms mediating MI/R injury. Kukoamine A (KuA), a sperm, is the main bioactive component extracted from the bark of goji berries. In this study, we wanted to investigate the possible effects of KuA on MI/R injury. Methods: In this experiment, all rats were divided into sham operation group, MI/R group, KuA 10 mg + MI/R group, KuA 20 mg + MI/R group. After 120 min of ischemia/reperfusion treatment, left ventricular systolic pressure (LVSP), left ventricular end-diastolic pressure (LVEDP), maximal rates of rising and fall of left ventricular pressure (±dp/dtmax), and ischemic area were detected. Serum samples of rats in each group were collected. The enzyme activities of catalase (CAT), glutathione peroxidase (GSH-PX), superoxide dismutase (SOD), levels of malondialdehyde (MDA), CK muscle/brain (CK-MB), tumor necrosis factor (TNF), interleukin-1ß (IL-1ß), and interleukin-6 (IL-6) were detected using enzyme-linked immunosorbent assay (ELISA). The apoptosis of myocardium in each group was detected according to the instructions of the terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay. The expressions of mammalian target of glycogen synthase kinase-3ß (GSH-3ß) and protein kinase B (Akt) mRNA level in myocardial tissues were detected via reverse transcription-polymerase chain reaction (RT-PCR). Results: MI/R rats showed a significant increase in oxidative stress and inflammation. In addition, we showed that KuA significantly improved the myocardial function such as LVSP, left ventricular ejection fraction, +dp/dt, and -dp/dt. Here, it attenuated dose-dependent histological damage in ischemia-reperfused myocardium, which is associated with the enzyme activities of SOD, GSH-PX, and levels of MDA, IL-6, TNF-α, L-1ß. Conclusions: KuA inhibited gene expression of Akt/GSK-3ß, inflammation, oxidative stress and improved MR/I injury. Taken together, our results allowed us to better understand the pharmacological activity of KuA against MR/I injury.

Purpose: To explore the mechanism of jatrorrhizine on apoptosis and fibrosis induced by myocardial infarction (MI) in an animal model. Methods: The left anterior descending branch of coronary artery was surgically ligated to duplicate the mouse model of MI. The sham and infarcted mice were treated with normal saline once a day, while mice in experimental groups received low-dose (LD) and high-dose (HD) jatrorrhizine once a day respectively. Two weeks later, cardiac function was detected by echocardiography, and histopathological examination was performed using hematoxylin and eosin (H&E) and Masson staining. The expressions of p53, TGF-ß1, Smad/2/3, Bax, Bcl-2, collagen I and collagen III were quantified using qRT-PCR and western blot assays. Results: Jatrorrhizine significantly improved left ventricular ejection fraction (LVEF) and left ventricle end-systolic (LVES) in mice. Histopathological, administration of jatrorrhizine weakened infiltration of inflammatory cells and cardiac fibrosis in myocardium of mice caused by MI. Additionally, jatrorrhizine suppressed cardiomyocyte apoptosis exhibited as its capability to reverse changes of Bax and Bcl-2 levels in myocardium caused by MI. Jatrorrhizine statistically significantly downregulated expression of collagen I and collagen III, as well as TGF-ß1, Smad2/3 and p53. Conclusions: Jatrorrhizine reduce cardiomyocyte apoptosis and fibrosis through inhibiting p53/Bax/Bcl-2 and TGF-ß1/Smad2/3 signaling pathways.

Resumo

Ionóforos são substâncias que foram inicialmente utilizadas como coccidiostáticos na produção de frangos de corte e, posteriormente, passaram a ser adicionados como aditivos na dieta de ruminantes. A intoxicação por ionóforos pode ocorrer em diversas espécies animais, no entanto, a espécie equina é especialmente sensível. O quadro clínico e anatomopatológico relacionado à intoxicação em equinos é caracterizado por sinais clínicos decorrentes de cardiomiopatia e miopatia da musculatura esquelética devido a necrose causada nestes tecidos. Não existe tratamento específico para os quadros de intoxicação por ionóforos, sendo assim, a prevenção deve ser preconizada de modo a evitar exposição dos equinos a alimentos que contenham ionóforos, especialmente nas criações em que bovinos e equinos compartilham do mesmo ambiente de criação. O objetivo deste artigo é abordar os principais aspectos relacionados aos ionóforos, bem como a intoxicação por estas substâncias em equinos.(AU)


Ionophores are substances that were initially used as coccidiostats in the production of broilers and later were added as additives in the diet of ruminants. Ionophores poisoning can occur in several animal species, however, horses are especially sensitive. The clinical and anatomopathological features related to poisoning in horses are characterized by clinical signs resulting from cardiomyopathy and myopathy of the skeletal musculature due to necrosis caused in these tissues. There is no specific treatment for ionophores poisoning conditions, therefore, prevention must be advocated in order to avoid exposure of horses to feed containing ionophores, especially in breeding where cattle and horses share the same rearing environment. The aim of this article is to address the main aspects related to ionophores, as well as poisoning by these substances in horses.(AU)

Purpose: The present study explored the role and mechanism involved in aprepitant-induced cardioprotective effects in rat model of ischemia-reperfusion injury. Methods: The isolated hearts of Wistar male albino rats were subjected to ischemia-reperfusion injury on Langendorff apparatus. The extent of myocardial injury was assessed by measuring lactate dehydrogenase 1 and CK-MB release in the coronary effluent. The rats were treated with aprepitant (5, 10 and 20 mg/kg) before isolating hearts. After injury, the levels of HIF-1α, p-AkT, p-GSK-3ß/GSK-3ß were measured in heart homogenates. LY294002 was employed as PI3K inhibitor. Results: Ischemia-reperfusion led to significant myocardial injury and decreased the levels of HIF-1α, p-AkT and ratio of p-GSK-3ß/GSK-3ß. Aprepitant attenuated myocardial injury and restored the biochemical changes in a dose-dependent manner. Pre-treatment with LY294002 (10 and 20 mg/kg) abolished aprepitant-mediated cardioprotective effects and restored the biochemical parameters in the heart homogenate. Conclusions: Aprepitant may be effective in preventing ischemia-reperfusion-induced myocardial injury, which may be due to activation of PI3K-AkT-GSK-3ß and HIF-1α signaling pathway.

Mazama gouazoubira is a widely distributed deer in the South American continent. However, studies on its anatomical characteristics are still scarce. The objective of this study was to elucidate the coronary circulation of M. gouazoubira. With this aim, after collecting cadavers on highways, twenty-four hearts injected with latex and preserved in formaldehyde at 10% were dissected. The dissections revealed that all specimens presented left coronary dominance, characterized by the origin of the subsinuous interventricular branch from the circumflex branch of the left coronary artery. The number of ventricular branches originated from branches of the left coronary artery was approximately five times higher than that of the right coronary artery. The occurrence of myocardial bridges was registered in 91.7% of the individuals, with bridges predominating over the paraconal interventricular branch. The mean length of the paraconal interventricular sulcus was significantly higher in males (p = 0.02). The number of atrial branches and ventricular branches, length of myocardial bridges and length of subsinuous interventricular sulcus did not vary significantly between sex. Left coronary dominance and high frequency of myocardial bridges in M. gouazoubira are preponderant anatomical traits also present in different species of Ruminantia suborder. Coronary dominance studies in other South American deer species may confirm the correlation of this trait with the evolutionary history of these animals.


Mazama gouazoubira é um cervídeo com ampla distribuição no continente sul americano. Entretanto, estudos sobre suas características anatômicas, ainda, são escassos. Objetivou-se elucidar a circulação coronariana do M. gouazoubira. Para tal, foram dissecados 24 corações injetados com látex e conservados em formaldeído a 10%, obtidos de cadáveres recolhidos em rodovias. As dissecações revelaram que todos os espécimes apresentaram dominância coronariana esquerda, caracterizada pela origem do ramo interventricular subsinuoso, a partir do ramo circunflexo da artéria coronária esquerda. O número de ramos ventriculares originados, a partir de ramos da artéria coronária esquerda, foi aproximadamente cinco vezes maior do que aquele da artéria coronária direita. A ocorrência de pontes de miocárdio foi registrada em 91,7% dos indivíduos, predominando as pontes sobre o ramo interventricular paraconal. O comprimento médio do sulco interventricular paraconal foi significativa-mente maior em machos (p = 0,02). O número de ramos atriais e ramos ventriculares, comprimento das pontes de miocárdio e comprimento do sulco interventricular subsinuoso, não variaram, significativamente, entre os sexos. A dominância corona-riana esquerda e a elevada frequência de pontes de miocárdio no M. gouazoubira são traços anatômicos preponderantes, tam-bém, em diferentes espécies da subordem Ruminantia. Estudos sobre dominância coronariana em outras espécies de cervídeos sul americanos poderão sugerir a correlação desta característica com a história evolutiva destes animais.

ABSTRACT: This study reported the epidemiological data and the clinical and pathological conditions of spontaneous poisoning by Ateleia glazioviana in sheep and goats in the West region of Santa Catarina. The small ruminants were located in a place where there was a large amount of young plants of A. glazioviana and showing signs of consumption. The stock of sheep was composed of 250 animals, of which 45 died. In the goat herd, there were 28 animals, and of these, 27 died, and one was sacrificed. The main clinical signs were weight loss, fatigue, slow-walk, and eventually submandibular and facial edema. Some animals were found dead, and others died suddenly after being moved. Abortions and the birth of weak lambs were also observed. In total, five sheep and one goat were necropsied. The main macroscopic lesions were associated to eccentric cardiac hypertrophy and moderate, multifocal white areas in the epicardium and myocardium. Two sheep showed marked edema in the subcutaneous tissue of the head and neck and it could be noted hydrothorax and hydroperitoneum and liver with a nutmeg-like appearance. Histological changes were relevant in the myocardium. They consisted of fibrosis and myofiber necrosis associated with macrophage infiltrate, multifocal, low and marked swelling of cardiomyocytes with loss of fibrillarystriae, and increased nuclear volume. In two sheep, there was still congenital hepatic, as well as marked centrilobular and diffuse areas.


RESUMO: Descrevem-se dados epidemiológicos, sinais clínicos e lesões da intoxicação espontânea por Ateleia glazioviana em ovinos e caprinos no Oeste de Santa Catarina. Os ovinos e caprinos se encontravam em um local que havia grande quantidade de A. glazioviana jovem e com sinais de consumo. O plantel de ovinos era composto por 250 animais, dos quais 45 morreram. No rebanho caprino haviam 28 animais e destes, 27 morreram e um foi sacrificado. Os principais sinais clínicos se caracterizaram por perda de peso, cansaço, caminhar lento e eventualmente edema submandibular e facial. Alguns foram encontrados mortos e outros morreram subitamente após serem movimentados. Abortos e nascimento de cordeiros fracos também foi observado. No total, cinco ovinos e um caprino foram necropsiados. As principais lesões macroscópicas foram hipertrofia excêntrica do coração e áreas brancas multifocais, moderadas, no epicárdio e miocárdio. Dois ovinos apresentaram edema acentuado no tecido subcutâneo da cabeça e pescoço, bem como hidrotórax, hidroperitôneo e fígado com aspecto de noz-moscada. As alterações histológicas foram relevantes no miocárdio e consistiram de áreas de fibrose e necrose de miofibras associada à infiltrado de macrófagos, multifocal, leve, além de tumefação acentuada de cardiomiócitos com perda das estrias fibrilares e aumento do volume nuclear. Em dois ovinos, havia ainda, congestão hepática, centrolobular, difusa.

Resumo

A nine-year-old, male, Pekingese dog was presented with a history of dyspnea, lethargy, syncope, polyuria, polydipsia, and selective appetite over two years. When the clinical signs first began, a radiographic exam revealed a radiopaque mass in the cranial mediastinum. An adequate treatment was not performed and the dog was presented to the hospital with severe tachycardia, dyspnea, and tachypnea, and died during physical examination. On necropsy, the heart presented bilateral auricular aneurysms. The pericardial sac was intact, and no other defects were found within the heart. Microscopically, the cardiomyocytes were marked degenerated and proliferation of connective tissue. This is the first case report of a bilateral auricular aneurysm with intact pericardial sac in a dog.~ien

As miocardiopatias são as cardiopatias mais comumente diagnosticadas em felinos domésticos. A cardiomiopatia hipertrófica (CMH) soma a maioria dos casos de cardiomiopatias em pacientes atendidos na rotina da medicina interna de felinos. Gatos com doenças cardíacas podem não apresentar manifestações clínicas evidentes podendo apresentar sinais sutis de insuficiência cardíaca (IC) ao longo do tempo de modo que em um dado momento descompensam e evidenciam sinais clínicos. O pimobendan é um fármaco denominado inodilatador devido a sua capacidade de promover inotropismo positivo e vasodilatação. Apesar de escassos, os estudos existentes com a utilização de pimobendan demonstram boa tolerância em gatos saudáveis e com cardiopatias diversas. Atualmente, o uso do pimobendan em gatos é extra bula e a dose baseia-se em estudos realizados com cães. No entanto, sabe-se que as concentrações plasmáticas diferem entre as espécies o que pode requerer possíveis ajustes. A terapia da IC em gatos é bastante discutida e evidências recentes demonstram que o pimobendan pode ser um aliado no tratamento de diversas doenças cardíacas que levem à disfunção sistólica sendo capaz de prover melhora dos sinais clínicos e até aumento da expectativa de vida desses pacientes. Contudo, pesquisas são necessárias para melhor entendimento dos efeitos hemodinâmicos do medicamento em gatos com disfunção diastólica.


Myocardiopathies are the most diagnosed heart disease in domestic cats. Hypertrophic cardiomyopathy (HCM) adds up to most cases of cardiomyopathies in patients seen in the routine of feline internal medicine. Cats with heart disease may not have evident clinical manifestations and may show subtle signs of heart failure (HF) over time so that at a given moment they decompensate and show clinical symptoms. Pimobendan is an inodilator drug due to its ability to promote positive inotropism and vasodilation. Although scarce, the existing studies with the use of pimobendan demonstrate good tolerance in healthy cats with various heart diseases. Currently, the use of pimobendan in cats is off-label and the dose is based on studies conducted in dogs. However, it is known that plasma concentrations differ between species, which may require possible adjustments. The therapy of HF in cats is widely discussed and recent evidence shows that pimobendan can be an ally in the treatment of various heart diseases that lead to systolic dysfunction, being able to provide improvement in clinical signs and even increase the life expectancy of these patients. However, research is necessary to better understand the hemodynamic effects of the drug in cats with diastolic dysfunction.

This study reported the epidemiological data and the clinical and pathological conditions of spontaneous poisoning by Ateleia glazioviana in sheep and goats in the West region of Santa Catarina. The small ruminants were located in a place where there was a large amount of young plants of A. glazioviana and showing signs of consumption. The stock of sheep was composed of 250 animals, of which 45 died. In the goat herd, there were 28 animals, and of these, 27 died, and one was sacrificed. The main clinical signs were weight loss, fatigue, slow-walk, and eventually submandibular and facial edema. Some animals were found dead, and others died suddenly after being moved. Abortions and the birth of weak lambs were also observed. In total, five sheep and one goat were necropsied. The main macroscopic lesions were associated to eccentric cardiac hypertrophy and moderate, multifocal white areas in the epicardium and myocardium. Two sheep showed marked edema in the subcutaneous tissue of the head and neck and it could be noted hydrothorax and hydroperitoneum and liver with a nutmeg-like appearance. Histological changes were relevant in the myocardium. They consisted of fibrosis and myofiber necrosis associated with macrophage infiltrate, multifocal, low and marked swelling of cardiomyocytes with loss of fibrillarystriae, and increased nuclear volume. In two sheep, there was still congenital hepatic, as well as marked centrilobular and diffuse areas.(AU)


Descrevem-se dados epidemiológicos, sinais clínicos e lesões da intoxicação espontânea por Ateleia glazioviana em ovinos e caprinos no Oeste de Santa Catarina. Os ovinos e caprinos se encontravam em um local que havia grande quantidade de A. glazioviana jovem e com sinais de consumo. O plantel de ovinos era composto por 250 animais, dos quais 45 morreram. No rebanho caprino haviam 28 animais e destes, 27 morreram e um foi sacrificado. Os principais sinais clínicos se caracterizaram por perda de peso, cansaço, caminhar lento e eventualmente edema submandibular e facial. Alguns foram encontrados mortos e outros morreram subitamente após serem movimentados. Abortos e nascimento de cordeiros fracos também foi observado. No total, cinco ovinos e um caprino foram necropsiados. As principais lesões macroscópicas foram hipertrofia excêntrica do coração e áreas brancas multifocais, moderadas, no epicárdio e miocárdio. Dois ovinos apresentaram edema acentuado no tecido subcutâneo da cabeça e pescoço, bem como hidrotórax, hidroperitôneo e fígado com aspecto de noz-moscada. As alterações histológicas foram relevantes no miocárdio e consistiram de áreas de fibrose e necrose de miofibras associada à infiltrado de macrófagos, multifocal, leve, além de tumefação acentuada de cardiomiócitos com perda das estrias fibrilares e aumento do volume nuclear. O gênero Aspergillus compreende espécies de fungos saprotróficos, globalmente distribuídos, sendo os Aspergillus seção Fumigati conhecidos por causarem doença em humanos e animais imunocomprometidos. Os sinais clínicos dependem do sistema acometido, podendo apresentar febre branda, cólicas intensas, pneumonias, micose de bolsa gutural, e, ocasionalmente, lesões no sistema nervoso central. O presente trabalho objetiva relatar os sinais clínicos, os achados de necropsia e a histologia de um caso de aspergilose sistêmica em um potro de oito meses de idade. O animal apresentava refluxo enterogástrico, peristaltismo aumentado, fezes pastosas, dor abdominal e desidratação intensa. O quadro clínico evoluiu para óbito e o cadáver foi encaminhado para a necropsia. Macroscopicamente, havia sufusões e equimoses na serosa do trato gastrointestinal e ulcerações na mucosa do intestino delgado. No pulmão, havia nódulos multifocais, esbranquiçados e firmes que, ao corte, apresentavam-se císticos, com revestimento interno vinhoso e friável. O coração exibia áreas esbranquiçadas multifocais no miocárdio e os rins continham nódulos avermelhados nas regiões cortical e medular. No encéfalo, havia áreas multifocais amareladas e hemorrágicas com bordos avermelhados. Microscopicamente, observaram-se áreas multifocais de necrose com infiltrado inflamatório granulomatoso no intestino, pulmão, encéfalo, miocárdio e nos rins. Hifas fúngicas intralesionais e intravasculares foram observadas no encéfalo e no pulmão. Estruturas de fenótipo compatível com Aspergillus fumigatus foram observadas na cultura fúngica. Amostras encaminhadas para virologia apresentaram-se positivas para Herpesvírus Equino (HVE) tipo 1 e 4. O diagnóstico de aspergilose sistêmica foi determinado pelo histórico clínico, imunossupressão, achados macroscópicos, histológicos, isolamento do fungo e à infecção concomitante por HVE.

Clinical signs depend on the affected system, exhibiting mild fever, severe colic, pneumonia, guttural bag mycosis and occasionally lesions in the central nervous system. The present work aims to report the clinical signs, necropsy findings and histology of a case of systemic aspergillosis in an eight-months-old foal. The animal had enterogastric reflux, increased peristalsis, pasty feces, abdominal pain and severe dehydration. The condition evolved to death and the cadaver was sent for necropsy. Macroscopically, there were suffusions and ecchymosis in the serosa of the gastrointestinal tract and ulcerations in the mucosa of the small intestines. In the lung, there were multifocal, whitish and firm nodules that were cystic when cut with a friable winey internal lining. The heart had whitish multifocal areas in the myocardium and the kidneys contained reddish nodules in the cortical and medullary regions. In the brain there were yellowish and hemorrhagic multifocal areas with reddish borders. Microscopically, multifocal areas of necrosis with granulomatous inflammatory infiltrate were observed in the intestine, lung, brain, myocardium and kidneys. Intralesional and intravascular fungal were observed in the brain and lung. Structures with phenotype compatible with Aspergillus fumigatus were observed in the fungal culture. In virology, samples were positive for equine herpesvirus (HVE) types 1 and 4. The diagnosis of systemic aspergillosis was determined due to the clinical history, immunosuppression, macroscopic and histological findings, fungal isolation and concomitant HVE infection.

This report describes clinical, ultrasonographic and anatomopathological findings in a case of metastatic melanoma in an adult Saanen goat. Clinically, the goat had apathy, an intra-abdominal palpable firm structure, and exophytic keratinized areas on the skin of the udder. Ultrasound revealed non-encapsulated oval structures, with heterogeneous echogenicity and marked central and peripheral vascularization, and hypoechoic hepatic multifocal to coalescent areas. In the udder, there were non-encapsulated oval structures with heterogeneous echogenicity and hyperechoic center surrounded by hypoechogenic tissue. Grossly, there were black multifocal to coalescent areas in the liver, as well as black nodules in mammary and mesenteric lymph nodes, uterus, spleen, and myocardium. Microscopically, multifocal melanocytic neoplastic proliferation was observed in the dermis and junction of the udder epidermis. Most of the neoplastic cells had cytoplasmic granules of melanin. In the liver there were areas of neoplastic tissue compressing the adjacent parenchyma, with central foci of necrosis, mild desmoplasia, and multifocal infiltration of malignant cells into the adjacent tissues. Similar findings were observed in the mammary and mesenteric lymph nodes, uterus, spleen, and myocardium, which characterized metastatic melanoma. Ultrasonography played a key role for establishing the diagnosis of a metastatic melanoma and helped establish a proper clinical management protocol.(AU)


Este relato descreve os achados clínicos, ultrassonográficos e anatomopatológicos em um caso de melanoma metastático em uma cabra Saanen adulta. Clinicamente, a cabra apresentava apatia, estrutura firme palpável intra-abdominal e áreas exofíticas queratinizadas na pele do úbere. A ultrassonografia revelou estruturas ovais não encapsuladas, com ecogenicidade heterogênea e marcada vascularização central e periférica, além de áreas hepáticas multifocais a coalescentes hipoecoicas. No úbere, havia estruturas ovais não encapsuladas, com ecogenicidade heterogênea e centro hiperecogênico circundado por tecido hipoecogênico. Macroscopicamente, havia áreas pretas multifocais a coalescentes no fígado, bem como nódulos pretos nos linfonodos mamários e mesentéricos, no útero, no baço e no miocárdio; microscopicamente, proliferação neoplásica melanocítica multifocal foi observada na derme e na junção da epiderme do úbere. A maioria das células neoplásicas apresentava grânulos citoplasmáticos de melanina. No fígado, havia áreas de tecido neoplásico comprimindo o parênquima adjacente, com focos centrais de necrose, desmoplasia leve e infiltração multifocal de células malignas nos tecidos adjacentes. Achados semelhantes foram observados nos nódulos linfáticos mamários e mesentéricos, no útero, no baço e no miocárdio, que caracterizaram o melanoma metastático. A ultrassonografia desempenhou um papel fundamental para estipular o diagnóstico de um melanoma metastático e ajudou a estabelecer um protocolo de manejo clínico adequado.(AU)

ABSTRACT: Avian colibacillosis is an acute and globally occurring infectious disease of domestic and wild birds caused by Escherichia coli, and it is associated with considerable economic losses mainly due to the morbidity and mortality associated. The present study aimed to describe the pathological, bacteriological and immunohistochemical aspects of avian colibacillosis in broiler chickens of Mozambique. Forty-nine broiler chicken presented anorexia, decreased weight gain, ataxia, diarrhea, dyspnea, and death in a clinical course of 3-5 days. The birds were raised in five farms (small, medium and large farms) with manual and automatic breeding system, with flocks ranging from 100 to 20,000 birds. At the necropsy, all birds had poor body condition, and the pericardium and the Glissons capsule of all avian exhibited different degrees of adherence often associated with severe fibrin deposition. The thoracic and abdominal air sacs were thickened and adhered to the costal wall. Mild, moderate or marked hepatomegaly associated with white pinpoint multifocal areas (100%, 49/49) and mild to moderate splenomegaly in 75.5% (37/49) with a mottled surface were observed. The lungs and kidney were enlarged and reddish. Histologically, a multiorgan fibrinoheterophilic polyserositis was observed in 75.5% of the cases (37/49), which were characterized by inflammatory infiltrates composed mainly of degenerative heterophils, macrophages and plasma cells, associated with fibrin deposits and intermixed by coccobacillary bacterial basophilic aggregates. These affected mainly the pericardium (28.6%, 14/49), the pleura (18.4%, 9/49), the Glissons capsule (10.2%, 5/49), the ventriculus (10.2, 5/33), and the proventriculus (8.2%, 4/49) serosa. Multifocal to coalescing areas of coagulative necrosis associated with similar inflammatory cells were observed mainly in the spleen (28.6%, 14/49), liver (24.5%, 12/49), and intestines (22.4%, 11/49). A similar infiltrate was also observed affecting the the lungs (16.3%, 8/49), the kidney (16.3%, 8/49) and the myocardium (14.3%, 7/49). Isolation and identification of E. coli was obtained in 12 cases through bacterial culture. Some organs (2 cases of each farms) were selected and submitted to immunohistochemistry anti-E. coli, and a positive stain was observed in all tested cases in liver (3/3), heart (4/4), spleen (1/1), lungs (4/4), intestines (4/4), bursa of Fabricius (1/1), ventriculus (1/1), and proventriculus (1/1) tissue sections. These results demonstrate that E. coli was the cause of mortality in these birds. Therefore, biosecurity and management measures should be employed to prevent and control the disease occurrence in Mozambiques poultry farming.


RESUMO: A colibacilose aviária é uma doença aguda de ocorrência mundial que acomete aves domésticas e silvestres, causada por Escherichia coli e resulta em perdas econômicas consideráveis devido à elevada morbidade e mortalidade das aves. O presente estudo teve o objetivo de descrever os aspectos patológicos, bacteriológicos e imuno-histoquímicos de colibacilose aviária em frangos de corte de Moçambique. Um total de 49 frangos de corte apresentaram anorexia, baixo ganho de peso, ataxia, diarreia, dispneia e morte em um curso clínico de 3 a 5 dias. As aves eram provenientes de 5 granjas (pequenas, média e grandes), com sistema de criação manual e automático, com rebanhos que variavam de 100 a 20.000 aves. À necropsia, todas as aves exibiam condição corporal ruim a caquética, além de pericárdio e cápsula de Glisson de todas aves (100%; n=49) com diferentes graus de aderência e deposição de fibrina de forma difusa acentuada. Os sacos aéreos torácicos e abdominais estavam espessados e aderidos à parede costal. Foi observado ainda hepatomegalia discreta, moderada a severa frequentemente associada com áreas multifocais puntiformes brancacentas (100%; 49/49), e esplenomegalia discreta a moderada, associado a áreas multifocais moteadas (75,5%; 37/49). Os pulmões e rins estavam aumentados e com coloração avermelhada. Histologicamente, observou-se majoritariamente serosite fibrinoheterofílica em 75,5% dos casos (37/49), caracterizadas por infiltrado inflamatório composto por heterófilos degenerados, macrófagos, linfócitos e plasmócitos, com deposição de fibrina entremeada por uma miríade de estruturas bacterianas cocobacilares. Esta lesão foi observada principalmente em pericárdio (28,6%; 14/49), pleura (18,4%; 9/49), cápsula de Glisson (10,2%; 5/49), ventrículo (10,2; 5/33) e em proventrículo (8,2%; 4/49). Áreas multifocais a coalescentes de necrose de coagulação associada a infiltrado inflamatório semelhante ao descrito foi observado principalmente no baço (28,6%; 14/49), fígado (24.5%; 12/49), e intestinos (8,2%; 4/49). Um infiltrado inflamatório semelhante também foi visualizado em pulmões (16,3%; 8/49), rins (16,3%; 8/49) e miocárdio (14,3%; 7/49), Colônias puras de E. coli foram identificadas e isoladas em 12 casos. Alguns órgãos (2 de cada granja) foram submetidos ao exame imuno-histoquímico anti-E. coli e marcação positiva foi visualizada em todos casos testados, como em fígado (3/3), coração (4/4), baço (1/1), pulmão (4/4), intestinos (4/4), bursa de Fabricius (1/1), rim (1/1), ventrículo (1/1) e proventrículo (1/1). Estes resultados demonstram que E. coli foi a causa de morte destas aves. Sendo assim, a adoção de boas medidas de biosseguridade e de manejo são indispensáveis para a prevenção e controle da ocorrência da doença nas granjas de frango de corte de Moçambique.

Purpose The present study explored the influence of liraglutide on remote preconditioning-mediated cardioprotection in diabetes mellitus along with the role of nuclear factor erythroid 2-related factor 2 (Nrf2), hypoxia inducible factor (HIF-1) and hydrogen sulfide (H2S). Methods Streptozotocin was given to rats to induce diabetes mellitus and rats were kept for eight weeks. Four cycles of ischemia and reperfusion were given to hind limb to induce remote preconditioning. After 24 h, hearts were isolated and subjected to 30 min of ischemia and 120 min of reperfusion on Langendorff system. Liraglutide was administered along with remote preconditioning. Cardiac injury was assessed by measuring the release of creatine kinase (CK-MB), cardiac troponin (cTnT) and development of left ventricular developed pressure. After ischemia-reperfusion, hearts were homogenized to measure the nuclear cytoplasmic ratio of Nrf2, H2S and HIF-1 levels. Results In diabetic rats, there was more pronounced injury and the cardioprotective effects of remote preconditioning were not observed. Administration of liraglutide restored the cardioprotective effects of remote preconditioning in a dose-dependent manner. Moreover, liraglutide increased the Nrf2, H2S and HIF-1 levels in remote preconditioning-subjected diabetic rats. Conclusions Liraglutide restores the lost cardioprotective effects of remote preconditioning in diabetes by increasing the expression of Nrf2, H2S and HIF-1.(AU)

Resumo

Purpose: To evaluate the preventive cardioprotective effects of resveratrol and grape products, such as grape juice and red wine, in animal model of cardiac ischemia and reperfusion. Methods: Male Wistar rats orally pretreated for 21-days with resveratrol and grape products were anesthetized and placed on mechanical ventilation to surgically induce cardiac ischemia and reperfusion by obstruction (ischemia) followed by liberation (reperfusion) of blood circulation in left descending coronary artery. These rats were submitted to the electrocardiogram (ECG) analysis to evaluate the effects of pretreatment with resveratrol and grape products on the incidence of ventricular arrhythmias (VA), atrioventricular block (AVB) and lethality (LET) resulting from cardiac ischemia and reperfusion. Results: It was observed that the incidence of AVB was significantly lower in rats pretreated with resveratrol (25%), grape juice (37.5%) or red wine (12.5%) than in rats treated with saline solution (80%) or ethanol (80%). Similarly, incidence of LET was also significantly lower in rats pretreated with resveratrol (25%), grape juice (25%) or red wine (0%) than in rats treated with saline solution (62.5%) or ethanol (75%). Conclusion: These results indicate that the cardioprotective response stimulated by resveratrol and grape products prevents the lethal cardiac arrhythmias in animal model of ischemia and reperfusion, supporting the idea that this treatment can be beneficial for prevention of severe cardiac arrhythmias in patients with ischemic heart disease.(AU)
