Resumo

An experiment was conducted to evaluate the effect of diets with reduced energy level content, supplemented with carbohydrase, on broiler performance and the coefficient of metabolizability of nutrients. A total of 720 one-day-old male Cobb-500 chicks were distributed in a completely randomized design, with six treatments, eight repetitions of 15 birds each. The treatments were: (1) a positive control, basal diet to meet the requirements of broiler chickens (PC); (2) a negative control, basal diet with a reduction of 80 kcal kg-1 (NC); (3) NC + alphagalactosydase; (4) NC + xylanase; (5) NC + xylanase and alphagalactosydase, and (6) NC + enzymatic blend (alphagalactosydase, xylanase, pectinase and amylase). The nutrient digestibility was not improved by use of enzymes. At 7 days of age, the broilers which were fed diets supplemented with enzymes showed a lower feed intake (FI) and better feed conversion ratio (FCR) than the broilers fed on PC. Both the NC and enzymatic blend resulted in a worse performance of the broilers at 21, 35 and 42 days old. The use of alphagalactosydase and xylanase, isolated or in combination, in a corn-soybean meal-based diet is effective in improving the growth performance of broilers fed energy-reduced diets.(AU)

Resumo

Polysaccharide storage myopathy (PSSM) is a genetic disorder or by undetermined cause characterized by the abnormal accumulation of glycogen and polysaccharides in skeletal muscle fibers. The present report describes two Percheron horses, from different properties in the municipality of Pouso Redondo -SC, with locomotor clinical signs that started after work. The clinical signs observed were rigid gait, mainly of the pelvic limbs, muscle weakness, and recumbency. In the first case, the animal was diagnosed with severe multifocal rhabdomyonecrosis, moderate multifocal myoglobin nephrosisand severe diffuse transmural fibrinonecrotic gastritis. PSSM was confirmed by the presence of polysaccharide inclusions in myofiber sarcolemma visualized by periodic acid-Schiff (PAS) staining. In the second case, the horse presented two episodes ofmuscle disorder after work. The therapeutic protocol was based on penicillin, flunixin meglumine and fluid therapy with ringer lactate. An increase in creatine kinase (669.0 UI/L) and aspartate aminotransferase (669.0 UI/L) was observed in the serum biochemical evaluation. Research for mutation in the GYS1 gene was performed, with a positive heterozygote result. After rest and gradual recovery, the horse was only submitted to light work, with no return of the clinical condition since then. Thus, this report describes two cases of PSSM with distinct clinical evolution and diagnostic methods.(AU)


A miopatia por acúmulo de polissacarídeos (PSSM) é uma desordem genética ou de causa indeterminada caracterizada pelo acúmulo anormal de glicogênio e polissacarídeos nas fibras musculares esqueléticas. O presente relato descreve dois cavalos Percheron, oriundos de diferentes propriedades do município de Pouso Redondo ­SC, com sinais clínicos locomotores que iniciaram após o trabalho. Os sinais clínicos observados foram marcha rígida, principalmente dos membros pélvicos, fraqueza muscular, e decúbito. No primeiro caso, o animal foi diagnosticado com rabdomionecrose multifocal acentuada, nefrose mioglobínica multifocal moderada e gastrite fibrinonecrótica transmural difusa acentuada. A PSSM foi confirmada pela presença de inclusões polissacarídicas no sarcolema de miofibras visualizadas pela coloração com ácido periódico de Schiff (PAS). No segundo caso, o equino apresentou dois episódios de disfunção muscular após o trabalho. O protocolo terapêutico foi baseado em penicilina, flunixin megluminee fluidoterapia com ringer lactato. Um aumento de creatina quinase (669,0 UI/L) e aspartato aminotransferase (669,0 UI/L) foi observado na avaliação bioquímica sérica. Foi realizada pesquisa de mutação nogene GYS1, com resultado heterozigoto positivo. Após repouso e recuperação gradual, o cavalo foi submetido apenas a trabalhos leves, sem retorno do quadro clínico desde então. Assim, este relato descreve dois casos com PSSM com evolução clínica e métodos diagnósticos distintos.(AU)

In dogs, Rhipicephalus linnaei transmits pathogens such as Ehrlichia canis, Babesia vogeli, and Hepatozoon canis. The veterinary market has synthetic acaricides to ticks control. Esters derived from castor oil are efficient. However, there is little information about their effects on non-target organisms. This work consisted of a clinical (AST, ALT, and ALP) and histological and histochemical analysis (liver and spleen) of female rabbits exposed to these esters and afoxolaner. The rabbits were divided into three groups: control group (CG) received Bandeirante® rabbit feed; the afoxolaner treatment (TG1) received rabbit feed and two doses of afoxolaner; castor oil esters treatment (TG2) received rabbit feed enriched with esters (1.75 g esters/kg). No alterations were observed in the AST, ALT, and ALP enzymes in exposure to esters TG2. Rabbits from TG1 showed changes in AST. The liver of rabbits exposed to afoxolaner underwent histological and histochemical changes, such as steatosis and vacuolation, as well as poor protein labeling. Polysaccharides were intensely observed in the group exposed to esters. The spleen showed no changes in any of the exposure. Esters from castor oil caused fewer liver changes when incorporated into the feed and fed to rabbits than exposure to afoxolaner.(AU)


Carrapatos Rhipicephalus linnaei transmitem patógenos como Ehrlichia canis, Babesia vogeli e Hepatozoon canis, para os cães. O mercado veterinário dispõe de acaricidas sintéticos para o controle desses ectoparasitas. Ésteres derivados do óleo de mamona são eficientes, mas há pouca informação sobre seus efeitos em organismos não-alvos. Esse trabalho objetivou analisar clínica (AST, ALT e ALP), histológica e histoquímicamente (fígado e baço) coelhas expostas aos ésteres e ao afoxolaner, tendo sido as mesmas alocadas nos grupos: Controle (GC) que recebeu ração Bandeirante®; Tratamento 1 (TG1), além da ração foram expostas ao afoxolaner, Tratamento 2 (TG2) receberam ração enriquecida com os ésteres (1,75 g ésteres/kg). Não houve alterações nas enzimas AST, ALT e ALP na exposição aos ésteres (TG2). No TG1 houve alterações na AST. O fígado das coelhas do TG1 apresentou alterações histológicas e histoquímicas, tais como esteatose e vacuolização, bem como fraca marcação de proteínas. Polissacarídeos foi intensamente marcados no fígado do TG2. O baço não apresentou alterações em nenhum dos grupos. Os ésteres do óleo de mamona causaram menos alterações hepáticas nas coelhas do que quando elas foram expostas ao afoxolaner.(AU)

Resumo

Purpose: To investigate the mechanism of polysaccharides from aloe vera (PAV), a main active ingredient of Aloe vera, treatment in pulpitis rats. Methods: Pulpitis were modeled by drilling the occlusal central fossa with Sprague Dawley rats. Next, the rats were treated with 20, 40, and 80 mg/kg PAV for three weeks, respectively. Computed tomography scanning assay, hematoxylin and eosin staining, and tartrate-resistant acid phosphatase staining were used to detect the pathology change. Then, levels of tumor necrosis factor-α, interleukin-1ß, prostaglandin E2, and ciclooxigenase 2 were detected by enzyme-linked immunosorbent assay. The expressions of bone morphogenetic protein 2 human (BMP-2), osteocalcin, osterix, and runt-related transcription factor 2 (Runx2) were quantified by quantitative real-time polymerase chain reaction and Western blotting (WB). Finally, Wnt3a expression, p-GSK3ß/GSK3ß and p-ß-catenin/ß-catenin ratio were analyzed by WB. Results: PAV up regulated the bone mineral density, and reduced the breakage of the crown and cervical structures, and the necrosis of the crown and root pulp of pulpitis rats. In addition, results indicated that PAV could inhibit osteoblast formation. While osteoblasts' number was decreased, proteins of BMP-2, osteocalcin, osterix, and Runx2 were up-regulated by PAV. Furthermore, PAV increased the Wnt3a expression and the p-ß-catenin/ß-catenin ratio, and decreased p-GSK3ß/GSK3ß ratio. Interestingly, these effects were all in dose dependence. Conclusions: PAV could inhibit pulp inflammation and promote osteoblasts differentiation via suppressing the activation of the Wnt/ß-catenin signaling, enhancing the dental bone density.

Microbial polysaccharides are of great biotechnological and commercial interest and have wide application in the food, cosmetic and medicine industries. Exopolysaccharide (EPS) production by the yeast Cryptococcus laurentii SD7, isolated from fresh water molluscs, was studied using agro-industrial byproducts as substrates in the submerged fermentation. The Central Composite Design (CCD) 23 was used to study the influence of pH, different concentrations on sugarcane molasses and corn steep liquor (CSL), for 48 hours. According to the results, the highest EPS production occurred at the initial pH 5 and at 8.4% concentration of sugarcane molasses, which were statistically significant variable at 10% (p < 0.1). The concentration of CSL had no influence in the studied range, thus, it can be used lowest concentration (0.3%). The time course of EPS production showed that while cell growth peaked within 48 hours, the highest EPS production (6.61 g L-1) occurred at 168 hours, with a productivity of about 0.04 g L-1 h-1. The pH of the medium remained approximately constant throughout the fermentation process. The yeast C. laurentii SD7 showed great potential for EPS production at a low cost and with sustainable substrates.(AU)

Resumo

The addition of natural molecules such as microbial exopolysaccharides in cosmetics is a trend in the current market, adding properties and improving the product quality. Therefore, the aim of this study was to develop a facial biocosmetic formulation containing microbial levan, almond and cinnamon oils. The centroid-simplex design was used to evaluate the spreadability, antioxidant activity, moisture retention capacity and viscosity of formulations. Since it is a facial cosmetic, the formulation was optimized using the intermediate viscosity. The optimized formulation with intermediate viscosity was 75% (0.75 g) levan and 25% (2 mL) almond oil, without the addition of cinnamon oil. This formulation was submitted to 90 days under different exposure conditions, and the results showed a spreadability of 805 mm2, pH and density ideal for the facial area, with an antioxidant activity of 72%, hydration capacity of 100.3%, viscosity with no-Newtonian behavior, and normal organoleptic properties when stored at room and low temperature. The formulation with levan associated with almond oil showed potential for application in the facial area, with high antioxidant properties, moisturizing intermediate viscosity and stability for 90 days. The utilization of centroid-simplex design allowed the development of a biocosmetic with desired characteristics just by adjusting the concentrations of the bioactive.(AU)

Purpose: To evaluate the protective effect of Cuscuta chinensis Lam. polysaccharides (PCCL) on 5-fluorouracil-(5-FU)-induced intestinal mucositis (IM) in mice. Methods: PCCL was orally administered at a dose of 20 mg·kg­1 for 7 days and its protective effect on 5-FU-induced IM (5-FU, 50 mg·kg­1 for 5 days) was evaluated by monitoring changes in body weight, degree of diarrhea, levels of tissue inflammatory factors (tumor necrosis factor α, interleukin 6, and interleukin 1ß levels), apoptosis rates, and the expression levels of caspase-3, Bax and Bcl-2. Results: The severity of mucosal injury (as reflected by body weight changes, degree of diarrhea, height of villi, and damage to crypts) was significantly attenuated by PCCL administration. PCCL also reduced the levels of tissue inflammatory factors, the apoptosis rate, and the expression of caspase-3 and Bax, and increased Bcl-2 expression. Conclusions: PCCL administration may be significantly protective against 5-FU-induced IM by inhibiting apoptosis and regulating the abnormal inflammation associated with it.

Broiler diets are based on corn, soybean and wheat production; however, some protein ingredients have many antinutritional factors and low digestibility. The objective of this study was to add a blend of exogenous enzymes to the feed with low nutritional value for broilers to reduce production costs and improve digestibility while maintaining good zootechnical performance. The experimental design was completely randomized, including three treatments with five replications (n=15) each: a) positive control (PC), diet calculated for males with average performance; b) negative control + exogenous enzymes (NC+EE), minimum nutrient diet according to the requirements of the production phase, and the enzymatic blend was added; and c) negative control (NC), minimum levels of nutrients for each phase. At 21 days, the PC group showed greater weight gain and lower feed conversion than the NC (P<0.05). At 42 days, PC had lower feed intake than NC (P=0.040), while lower feed conversion was observed in groups PC and NC+EE than NC (P=0.001). The production efficiency index was higher in the PC treatment, but the NC+EE treatment was higher than the NC (P=0.001). Considering production costs and body weight, we found that NC+EE birds had greater profitability. Therefore, we conclude that the blend of exogenous enzymes added to a diet with minimal nutritional levels has practical application in the broiler production system.(AU)


As dietas de frangos de corte são baseadas na produção de milho, soja e trigo; no entanto, alguns ingredientes proteicos possuem muitos fatores antinutricionais e baixa digestibilidade. O objetivo deste estudo foi adicionar à ração uma mistura de enzimas exógenas com baixo valor nutricional para frangos de corte para reduzir os custos de produção e melhorar a digestibilidade, mantendo o bom desempenho zootécnico. O delineamento experimental foi inteiramente casualizado, incluindo três tratamentos com cinco repetições (n=15) cada: a) controle positivo (PC), dieta calculada para machos com desempenho médio; b) controle negativo + enzimas exógenas (CN+EE), dieta mínima de nutrientes de acordo com as exigências da fase de produção, e foi adicionada a mistura enzimática; c) controle negativo (CN), teores mínimos de nutrientes para cada fase. Aos 21 dias, o grupo PC apresentou maior ganho de peso e menor conversão alimentar que o NC (P<0,05). Aos 42 dias, PC apresentou menor consumo de ração que NC (P=0,040), enquanto foi observada menor conversão alimentar nos grupos PC e NC+EE que NC (P=0,001). O índice de eficiência de produção foi maior no tratamento PC, mas o tratamento NC+EE foi maior que o NC (P=0,001). Considerando os custos de produção e peso corporal, verificamos que as aves NC+EE tiveram maior rentabilidade. Portanto, concluímos que a mistura de enzimas exógenas adicionada a uma dieta com níveis nutricionais mínimos tem aplicação prática no sistema de produção de frangos de corte.(AU)

Resumo

This research valorized Pachira aquatica Aubl.'s fruit shells (PAS) through its energetic characterization and flash pyrolysis for biofuels or chemicals production. The characterization was performed through proximate and ultimate analysis, bulk density, higher heating value (HHV), hemicellulose, cellulose and lignin content, thermogravimetric analysis and absorption spectra in the infrared region obtained by Fourier-transform infrared spectroscopy technique (FTIR). The analytical flash pyrolysis was performed at 500°C in a Py-5200 HP-R coupled to a gas chromatograph (Py-GC/MS). The PAS biomass presents potential for thermochemical energy conversion processes due to its low moisture and ash content, 76.90% of volatile matter, bulk density of 252.6 kg/m3 and HHV of 16.24 MJ/kg. Flash pyrolysis products are mostly phenols or light organic acids derived from the decomposition of polysaccharides. Results confirmed the potential of PAS to produce bio-phenolics, such as 4-methoxyphenol which is an important active ingredient for skin depigmentation used in drugs and cosmetics, and as phenolic extract that can be used as a precursor to resins, applications that convert this forest waste into bio products for industry into a green circular economy.


Este trabalho teve como objetivo a valorização das cascas dos frutos da Pachira aquatica Aubl. (PAC) através da sua caracterização energética e pirólise flash para produção de biocombustíveis ou produtos químicos. A caracterização foi realizada através de análises imediata e final, densidade aparente, poder calorífico superior (PCS), conteúdos de hemicelulose, celulose e lignina, análise termogravimétrica e espectros de absorção na região do infravermelho obtidos pela técnica de espectroscopia no infravermelho com transformada de Fourier (FTIR). A pirólise flash analítica foi realizada a 500 °C em equipamento Py-5200 HP-R acoplado a um cromatógrafo à gás (Py-GC/MS). A biomassa das PAC apresenta potencial para processos de conversão termoquímica de energia devido ao seu baixo teor de umidade e cinzas, além de 76,90% de materiais voláteis, densidade aparente de 252,6 kg/m3 e PCS igual a 16,24 MJ / kg. Os produtos da pirólise rápida são principalmente fenóis ou ácidos orgânicos leves derivados da decomposição de polissacarídeos. Os resultados confirmam o potencial das PAC para produzir bio-fenólicos, como o 4-metoxifenol que é um importante ingrediente ativo para despigmentação da pele usado em medicamentos e cosméticos, e como extrato fenólico que pode ser usado como precursor de resinas. Estas aplicações convertem esses resíduos florestais em produtos biológicos para a indústria em uma economia circular verde.

This study proposes to investigate the addition of sulfated polysaccharides (SP) extracted from two species of green seaweeds, Ulva lactuca and Caulerpa racemosa, to Colossoma macropomum semen cryodiluent medium. Four concentrations of SP (1.0, 2.0, 3.0, or 4.0 mg mL-1) of each seaweed were evaluated. Semen was collected during the month of September in Fortaleza - CE, Brazil. Fresh semen samples were analyzed for the parameters of total sperm motility, curvilinear velocity (VCL), straight line velocity (VSL), average path velocity (VAP), sperm morphology, membrane integrity, and DNA integrity. Then, the samples were cryopreserved in freezing medium containing 10% dimethyl sulfoxide (DMSO) + 5% glucose, which was supplemented with different concentrations of SP. An unsupplemented treatment was used as control. After 15 days, they were thawed in a water bath at 45 ºC for eight seconds and the same analyses of fresh semen were performed. Statistical analysis revealed that there were no significant differences (p > 0.05) between the different tested concentrations of SP for any of the evaluated parameters. Compared with the control, there was no difference in concentrations (p > 0.05) for total motility; however, for VCL, VSL, and VAP, the U. lactuca concentrations of 3.0 and 4.0 mg mL-1 were detrimental (p < 0.05). The same was observed with 4.0 mg mL-1 of C. racemosa for VSL and VAP. In terms of morphology, 1.0 and 4.0 mg mL-1 of C. racemosa reduced normal sperm (p < 0.05), whereas for the other concentrations there was no difference (p > 0.05). All concentrations of both seaweeds maintained plasma membrane integrity (p > 0.05). As for DNA integrity, only 4.0 mg mL-1 of U. lactuca produced lower results than the control (p < 0.05), whereas the other concentrations maintained the number of spermatozoa with intact DNA (p > 0.05). Based on the results, higher concentrations of SP are harmful to tambaqui sperm in the freezing medium, whereas lower concentrations maintain sperm parameters. Further research is warranted to better investigate the antioxidant potential of these polymers in cryodiluent medium for C. macropomum as well as other fish species.


O estudo teve como objetivo avaliar a adição de polissacarídeos sulfatados (PS) extraídos de duas espécies de macroalgas verdes, Ulva lactuca e Caulerpa racemosa, no meio criodiluidor do sêmen de Colossoma macropomum. Para isso, foram avaliadas quatro concentrações de PS (1,0; 2,0; 3,0 ou 4,0 mg mL-1), de cada macroalga. A coleta de sêmen foi realizada durante o mês de setembro, em Fortaleza, Ceará, Brasil. As amostras de sêmen fresco foram analisadas quanto aos parâmetros de motilidade total dos espermatozoides, velocidade curvilinear (VCL), velocidade em linha reta (VSL), velocidade média do trajeto (VAP), morfologia espermática, integridade de membrana e integridade de DNA. Em seguida, foram criopreservadas em meio de congelação contendo dimetilsulfóxido (DMSO) 10% + glicose 5%, e suplementadas com as diferentes concentrações de PS, tendo ainda um tratamento não suplementado como controle. Após 15 dias, foram descongeladas em banho-maria a 45 ºC por oito segundos, e as mesmas análises do sêmen fresco foram realizadas. Através da análise estatística, os resultados mostraram que não houveram diferenças significativas (p > 0,05) entre as diferentes concentrações de PS testadas para nenhum dos parâmetros avaliados. Já em relação ao controle, não houve diferença nas concentrações (p > 0,05) para a motilidade total, no entanto, para VCL, VSL e VAP, as concentrações de 3,0 e 4,0 mg mL-1 de U. lactuca foram prejudiciais (p < 0,05). O mesmo foi observado em 4,0 mg mL-1 de C. racemosa para VSL e VAP. Para a morfologia, 1,0 e 4,0 mg mL-1 de C. racemosa reduziram os espermatozoides normais (p < 0,05), enquanto para as demais concentrações não houve diferença (p > 0,05). Para todas as concentrações de ambas as macroalgas, a integridade de membrana plasmática foi mantida (p > 0,05). Quanto à integridade do DNA, apenas 4,0 mg mL-1 de U. lactuca foi inferior ao controle (p < 0,05), enquanto as demais concentrações mantiveram o número de espermatozoides com DNA íntegro (p > 0,05). De acordo com os resultados obtidos, concentrações mais elevadas de PS são prejudiciais aos espermatozoides de tambaqui no meio de congelação, enquanto concentrações mais baixas mantiveram parâmetros espermáticos. Estudos posteriores são indicados para melhor avaliar o potencial antioxidante destes polímeros no meio criodiluidor do sêmen de C. macropomum, bem como de outras espécies de peixes.

The objectives of this study were to investigate the effects of different concentrations of Suaeda rigida polysaccharides (SRPs) on the physiological characteristics of the frog heart and gastrocnemius muscle, compare their similarities and differences, and analyze the mechanisms. CaCl2 and acetylcholine (Ach) were selected respectively to be co-incubated with the high concentration SRPs to observe the effects on the heart contraction of frog. The effects of different concentrations of the SRPs on the activities of acetylcholinesterase (A-CHE), Na+-K+-ATPase and Ca2+-Mg2+-ATPase in the isolated frog heart were detected by UV-Vis spectrophotometry. The gastrocnemius muscle was immersed in the high concentration of SRPs for 10 min, and the systolic indexes were recorded. The effects of SRPs on the Ach content and A-CHE activity at the sciatic nerve-gastrocnemius junction were determined by UV-Vis spectrophotometry and enzyme-linked immunosorbent assay (ELISA). The results showed that the SRPs had significant inhibitory effects on the contractile amplitude of isolated heart and the contractile amplitude induced by CaCl2 and Ach, respectively (P<0.01). The activity of Ca2+-Mg2+-ATPase was significantly promoted, and the activity of A-CHE was significantly inhibited (P<0.01). The contraction amplitude, contraction rate, relaxation rate of gastrocnemius muscle and the Ach content at the junction of sciatic nerve-gastrocnemius muscle were significantly increased (P<0.01), and the activity of A-CHE at the junction was significantly inhibited (P<0.01) by the SRPs. All the results suggested that the SRPs could inhibit the contraction of heart and promote the contraction and relaxation of skeletal muscle. The mechanism was related to blocking the fast INa channel, inhibiting the ICa-L and activating the M receptors of myocardial membrane and then inhibiting external Ca2+ influx, increasing Ca2+-Mg2+-ATPase activity, decreasing a-che activity.


Os objetivos deste estudo foram investigar os efeitos de diferentes concentrações de Suaeda rigida polissacarídeos (SRPs) sobre as características fisiológicas do coração de rã e do músculo gastrocnêmio, comparar suas semelhanças e diferenças, e analisar os mecanismos. CaCl2 e acetilcolina (Ach) foram selecionados respectivamente para serem co-incubados com os SRPs de alta concentração para observar os efeitos sobre a contração do coração de rã. Os efeitos das diferentes concentrações dos SRPs sobre as atividades da acetilcolinesterase (A-CHE), Na+-K+-ATPase e Ca2+-Mg2+-ATPase no coração isolado de rã foram detectados pela espectrofotometria UV-Vis. O músculo gastrocnêmio foi imerso na alta concentração de SRP por 10 min, e os índices sistólicos foram registrados. Os efeitos das SRPs no conteúdo de Ach e na atividade de A-CHE na junção nervo-gastrocnêmio ciático foram determinados pela espectrofotometria UV-Vis e pelo ensaio de imunoabsorção enzimática (ELISA). Os resultados mostraram que os SRPs tiveram efeitos inibidores significativos sobre a amplitude contrátil do coração isolado e a amplitude contrátil induzida por CaCl2 e Ach, respectivamente (P<0,01). A atividade do Ca2+-Mg2+-ATPase foi significativamente promovida e a atividade do A-CHE foi significativamente inibida (P<0,01). A amplitude de contração, a taxa de contração, a taxa de relaxamento do músculo gastrocnêmio e o conteúdo de Ach na junção do músculo nervo ciático-gastrocnêmio foram significativamente aumentados (P<0,01), e a atividade do A-CHE na junção foi significativamente inibida (P<0,01) pelas SRPs. Todos os resultados sugeriram que os SRPs poderiam inibir a contração do coração e promover a contração e o relaxamento do músculo esquelético. O mecanismo estava relacionado ao bloqueio do canal INa rápido, inibindo a ICa-L e ativando os receptores M da membrana miocárdica e depois inibindo o influxo externo de Ca2+, aumentando a atividade de Ca2+-Mg2+-ATPase, diminuindo a atividade a-che.

ß-Glucans (ßG) are polysaccharides widely distributed in nature with chemopreventive properties. The aim of this study was to investigate the effects of ßG and the combined treatment with doxorubicin (Dox) on cell viability and mRNA levels of genes involved in cell cycle, apoptosis and antioxidant response. ßG was not cytotoxic. The mRNA levels of CCNA2of cells exposed to ß-glucan was upregulated and the exposure to Dox decreased the expression, while the combination led to an upregulation. Modulation of mRNA levels of CASP9suggest that ßG could inhibit promotion and progression steps of carcinogenesis, eliminatingneoplastic cells. The upregulation of CCNA2gene in combined treatment could be occurred due to ability of ßG in restoring the cell cycle distribution pattern after treatment with Dox. The upregulation of SOD1suggests that ßG can enhance the intracellular antioxidant defense, reducing the levels of superoxide dismutase induced by Dox. This response could reduce oxidative damage and attenuate tissue damage during chemotherapeutic treatment. Our data suggest that the drug combination may be less effective in killing tumor cells than the treatment with Dox alone. Some authors showed its significant efficacy in preventing mutagenic effects caused by doxorubicin, cyclophosphamide and cisplatin (Tohamy, El-Ghor, El-Nahas, & Noshy, 2003), methyl methanesulfonate (Oliveira et al., 2007)and hydrogen peroxide (Slamenová, 2003). Moreover, some studies have related the antioxidant ability of ßGagainst reactive free radicals formed by endogenous metabolic processes or exogenous chemicals (Tsiapali et al., 2001; Slamenová,2003; Sener, Eksioglu-Demiralp, Cetiner, Ercan, & Yegen, 2006; Guerra Dore et al., 2007; Kofuji et al., 2012; Lei et al., 2015). Yeast-derived ßGhave modulating action of humoral and cellular immune responses (Vetvicka et al., 2007).This activity provides protection to the organism against infections and cancer development (Samuelsen, Schrezenmeir, & Knutsen, 2014; Roudbary, Daneshmand, Hajimorad, Roudbarmohammadip, & Hassan, 2015). Despite postulated modes of action by which ß-glucan works are lacking information about the molecular mechanisms involved in the chemopreventive activity of this polysaccharide. In addition, compounds with chemopreventive properties can contribute to reduce side effects and toxicity during the chemotherapeutic treatment. Therefore, the aim of this study was to investigate the effects of ßG and the combined treatment with doxorubicin (Dox) on the expression of genes related with apoptosis (CASP9), cell cycle control (CCNA2)and antioxidant defense (SOD1)in human breast cancer MCF-7 cells. Doxorubicin (Dox) was chosen because it is one of the most used chemotherapeutic agent for cancer treatment. The limitation on the use of Dox in cancer treatment is the lack of selectivity against cancer cells and, consequently, its toxicity to patients.(AU)

The objective was to evaluate the productivity, microbiology and histopathology of the jejunum, ceca and liver in broilers fed a control or a high non-starch polysaccharide (HNSPs) diet added with an extract of humic substances (EHS). 240 broilers individually allocated, from 21-42 d of age were assigned to a factorial arrangement of 2 types of diets: 1) A corn/soybean meal diet (Control) and 2) As Control plus 7% distillers dried grain with solubles and 12% of wheat bran (HNSPs), and 3 growth promoters: 1) Antibiotic growth promoters (AGP); 2) without AGP (NAGP) and C) with 0.5% of EHS. At the end of the trial, the breast and carcass were weighed and samples of the intestine, ceca and liver were taken for microbial and histopathology analysis. Results were subjected to ANOVA. EHS-fed broilers had lower feed conversion ratio (FCR) and total aerobic bacterial (TAB) counts in the liver and higher ashes digestibility with the Control diet, but negative responses were seen with the HNSPs diet (Type of diet and growth promoter interaction, p≤0.05). The lesion scores in the jejunum were lower in EHS-fed broilers with the Control and HNSPs diet (p≤0.05). The TAB and E. coli were lower in the jejunum and ceca of AGP-fed broilers (p<0.01) compared to NAGP and EHS groups. EHS-fed broilers showed improved FCR and ileal ashes digestibility and lower TAB in the liver with the Control diet and had lower lesion score in the jejunum and similar weight gain compared to the AGP-fed broilers.(AU)

Edible coatings based on polysaccharides have been applied on pieces of fruits and vegetables before drying because of their potential to improve physical and nutritional characteristics of dehydrated plant foods. In the present study, physical and thermal properties of pectin-based films, with and without the incorporation of by-products obtained from the processing of red guava, were determined. These properties allow one to predict the likely behaviour of these films when used as edible coatings on guava pieces, during and after their dehydration. Thus the structural and morphological characteristics and the physical, thermal and sorption properties of the films and of the fruit pulp were determined, using scanning electronic microscopy (SEM), differential scanning calorimetry (DSC), X-ray diffraction (XRD) and the static gravimetric method to determine the water-sorption curves. The addition of by-products provided crystallinity to the pectin film, attributed to their cellulose content, and hardly altered the water retention capacity of the pectin films. However, the pectin contributed to presenting a slightly higher sorption moisture at equilibrium, as compared to the guava pulp. The glass transition temperatures reported at extremely low moisture contents, both in the pulp and in the films with added by-products, indicated that at intermediate moisture contents, the fruit/film ensemble was in the rubbery state at room temperatures, providing softness to the dehydrated product. The images showed good integration of the edible films with the surface of the guava pieces.


Revestimentos comestíveis à base de polissacarídeos têm sido aplicados em pedaços de frutas e vegetais antes da secagem devido ao seu potencial de melhorar as características físicas e nutricionais de alimentos vegetais desidratados. No presente estudo, foram determinadas as propriedades físicas e térmicas de filmes à base de pectina, com e sem a incorporação de subprodutos obtidos do processamento da goiaba vermelha. Essas propriedades permitem prever o provável comportamento desses filmes quando usados ​​como cobertura comestível em pedaços de goiaba, durante e após a sua desidratação. Assim, foram determinadas as características estruturais e morfológicas e as propriedades físicas, térmicas e de sorção dos filmes e da polpa do fruto, utilizando microscopia eletrônica de varredura (MEV), calorimetria exploratória diferencial (DSC), difração de raios X (DRX) e método gravimétrico estático para determinação das curvas de sorção de água. A adição de subprodutos proporcionou cristalinidade ao filme de pectina, atribuída ao seu teor de celulose, e dificilmente alterou a capacidade de retenção de água dos filmes de pectina. No entanto, a pectina contribuiu para apresentar uma umidade de sorção ligeiramente superior no equilíbrio, quando comparada à polpa de goiaba. As temperaturas de transição vítrea encontradas em teores de umidade extremamente baixos, tanto na polpa quanto nos filmes com subprodutos adicionados, indicaram que em teores de umidade intermediários, o conjunto fruta / filme estava no estado borracha em temperatura ambiente, proporcionando maciez ao produto desidratado. As imagens mostraram boa integração dos filmes comestíveis com a superfície dos pedaços de goiaba.

Resumo

Glycosaminoglycans (GAGs) are long-chain polysaccharides that are divided into sulphates and non-sulphates, these being chondroitin sulphate, heparan sulphate, dermatan sulphate, heparin sulphate and the only non-sulphate in the group is hyaluronic acid. GAGs are obtained from animal tissue and by an expensive low-yield extraction process; however, they are highly commercially valued polysaccharides and exploited in the biomedical market. Their disaccharidic composition, chain length and sulfation pattern present great variability depending on the species and extraction factors. GAGs possess immunomodulatory, antioxidant, antiviral, anti-inflammatory, neuroprotective, antiproliferative and anticoagulant properties, functioning as therapeutic agents modulating an array of biological processes. This report presents the general aspects of each GAG, source and extraction process, in addition to the characteristics that give them the most varied therapeutic properties and pharmacological applications.(AU)


Os glicosaminoglicanos (GAGs) são polissacarídeos de cadeias longas que se dividem em sulfatados e não sulfatados, sendo estes, sulfato de condroitina, sulfato de heparana, sulfato de dermatana, sulfato de heparina e o único não sulfatado do grupo que é o ácido hialurônico. Os GAGs são encontrados em todo tecido animal, são extraídos por um processo de alto custo e baixo rendimento, no entanto, o material obtido é valorizado comercialmente e amplamente explorado no mercado biomédico. Sua composição dissacarídica, comprimento da cadeia e padrão de sulfatação apresentam grande variabilidade dependendo das espécies e fatores de extração. Os GAGs possuem propriedades imunomoduladoras, antioxidantes, antivirais, anti-inflamatórias, neuroprotetoras, antiproliferativas e anticoagulantes, além de farmacológicas, funcionando como agentes terapêuticos moduladores de uma série de processos biológicos. Este relatório apresenta os aspectos gerais de cada GAG, fonte e processo de extração, além das características que lhes conferem as mais variadas propriedades terapêuticas e aplicações farmacológicas.(AU)

Marine algae are natural sources of macromolecules known as sulfated polysaccharides. This class of compounds has attracted the interest of Pharmaceutical Sciences due to its pharmacological anticoagulant, antiplatelet and antithrombotic properties. Therefore, this study evaluated the anticoagulant potential of sulfated polysaccharides extracted from the algae Penicillus capitatus. The extracted sulfated polysaccharides were purified, partially characterized and their anticoagulant activity was evaluated. The extraction process followed by ethanol precipitation resulted in five fractions. Among the analyzed fractions, F44 contained highest concentration of sulfated polysaccharides. After the purified fraction F23, F44 displayed in vitro anticoagulant activity in a time testing for activated partial thromboplastin time and pro-thrombin time. The preferential mechanism effect was based on interactions between thrombin and factor Xa. Additional studies on structure pharmacological are required to test the viability of the use of sulfated polysaccharides as therapeutic agents.

Esta classe de compostos atraiu o interesse das Ciências Farmacêuticas devido às suas propriedades farmacológicas como anticoagulante, antiplaquetária e antitrombótica. Portanto, este estudo tem como objetivo avaliar o potencial anticoagulante de polissacarídeos sulfatados extraídos de algas de Penicillus capitatus. The objective of this study was to determine the chemical composition, non-starch polysaccharides (NSP) content, in vitro digestibility of starch, apparent metabolizable energy (AME), nitrogen-corrected AME (AMEn ), and apparent and apparent nitrogen-corrected coefficient of metabolizable energy (ACME and ACMEn ) of three corn hybrids dried at different temperatures in the diets of broiler chickens. The energetic values were determined by the total collection of excreta method, using broilers from 11 to 21 d, placed in a completely randomized design in a factorial scheme (three corn hybrids and two drying temperatures: 80 and 110 °C), totalizing six treatments, seven replicates, and four birds/replicate. The data were subjected to two-way ANOVA. There was an interaction between the two factors tested wherein the AME and AMEn were increased when using corn Hybrid 1 dried at 80 °C. As main effects, the use of corn Hybrid 3 increased ACME and ACMEn . The drying temperature of 110 °C increased amylose, resistant starch, and nitrogen bound to the fiber contents, and reduced soluble:total NSP ratio. These changes may be responsible for the differences in digestibility of nutrients. The mean values of AME and AMEn of corn dried at 80 and 110 °C were 14.03 and 13.84 MJ/kg, respectively, and 81.62 and 80.53% for ACME and ACMEn , respectively.

Microbial polysaccharides are of great biotechnological and commercial interest and have wide application in the food, cosmetic and medicine industries. Exopolysaccharide (EPS) production by the yeast Cryptococcus laurentii SD7, isolated from fresh water molluscs, was studied using agro-industrial byproducts as substrates in the submerged fermentation. The Central Composite Design (CCD) 23 was used to study the influence of pH, different concentrations on sugarcane molasses and corn steep liquor (CSL), for 48 hours. According to the results, the highest EPS production occurred at the initial pH 5 and at 8.4% concentration of sugarcane molasses, which were statistically significant variable at 10% (p < 0.1). The concentration of CSL had no influence in the studied range, thus, it can be used lowest concentration (0.3%). The time course of EPS production showed that while cell growth peaked within 48 hours, the highest EPS production (6.61 g L-1) occurred at 168 hours, with a productivity of about 0.04 g L-1 h-1. The pH of the medium remained approximately constant throughout the fermentation process. The yeast C. laurentii SD7 showed great potential for EPS production at a low cost and with sustainable substrates.

The use of alternative ingredients has been increasing in a continuing attempt to reduce production costs, along with the use of additives, such as carbohydrases, for their possible positive effects on nutrient metabolization by layers. Thus, this study aimed to evaluate the effects of different xylanases in diets with reduced metabolizable energy (ME) and the inclusion of alternative ingredients on the metabolizability of nutrients in diets for commercial laying hens, by conducting two metabolism tests. In the first trial, 100 layers hens were distributed in a fully randomized 2 × 2 + 1 factorial design (two metabolizable energy reductions × two xylanases, plus one control diet). In the second assay, 140 layer hens were distributed in a fully randomized 2 × 3 + 2 factor design (two xylanases× three ingredients, plus two control diets, positive and negative). It was concluded that supplementation with pantanal xylanase improved the metabolizability of nutrients in corn-based diets and soybean meal for light commercial layers, allowing for a reduction of up to 200 kcal/kg of ME in the diets. Xylanases are more effective on wheat bran enabling a decrease of 150 kcal/kg of ME in commercial laying diets without affecting nutrient metabolizability. The action of pantanal xylanase in diets containing fibrous ingredients was similar to that of commercial xylanase.


O uso de ingredientes alternativos vem aumentando em uma tentativa contínua de reduzir os custos de produção, juntamente com o uso de aditivos, como os carboidrases, para seus possíveis efeitos positivos na metabolização de nutrientes por poedeiras. Assim, este estudo teve como objetivo avaliar os efeitos de diferentes xilanases em dietas com energia metabolizável reduzida (ME) e a inclusão de ingredientes alternativos sobre a metabolizabilidade de nutrientes em dietas para galinhas poedeiras comerciais, realizando dois testes de metabolismo. No primeiro ensaio, 100 aves foram distribuídas em delineamento inteiramente casualizado em esquema fatorial 2 x 2 + 1 (duas reduções de energia metabolizável x duas xilanases, mais uma dieta controle). No segundo ensaio, 140 aves foram distribuídas em delineamento inteiramente casualizado em esquema fatorial 2 x 3 + 2 (duas xilanases x três ingredientes -, mais duas dietas controle positivo e controle negativo). Conclui-se que a suplementação com xilanase do Pantanal melhora a metabolizabilidade dos nutrientes de dietas a base de milho e farelo de soja para poedeiras comerciais leves, possibilitando a redução em até 200 Kcal/kg de energia metabolizável das dietas. As xilanases são mais efetivas sobre o farelo de trigo e possibilitam a redução de 150 Kcal/kg de energia metabolizável nas dietas de poedeiras comerciais, sem prejuízo na metabolizabilidade dos nutrientes. Em dietas contendo ingredientes fibrosos a ação da xilanase do Pantanal é similar a xilanase comercial.
