Resumo

ABSTRACT Optimization of the use of water and the possibility of using residues as substrate in the black pepper production chain can yield economic and environmental benefits. The objective of this study was to evaluate the biometric and physiological traits of black pepper plantlets (Piper nigrum L.), cv. Bragantina, at different irrigation levels and soilless substrates. Plantlets were grown for 120 days on three substrates [biosolid + crushed coconut fiber (3:1, by volume); biosolid + granitic rock powder (3:1, by volume); and biosolid + crushed coconut fiber + granitic rock powder (2:1:1, by volume)] and at five irrigation levels [36 % (very low); 55 % (low); 85 % (moderate); 100 % (well-irrigated); and 126 % of the reference irrigation level (over-irrigated)]. The biometric traits evaluated increased as the irrigation levels increased, with the exception of root dry mass, which was not influenced by this factor, nor water use efficiency, which decreased as the irrigation levels increased. In general, plants grown with a mixture of the three wastes (biosolid + crushed coconut fiber + granitic rock powder) exhibited higher values for biometric variables and chlorophyll content when they were moderately, well or over-irrigated, in comparison with the other two substrates. It was possible to achieve the plantlet commercial standard using this three waste mixture and the moderate irrigation level.

Resumo

The characterization of substrate properties supports the correct choice of components and directs management decisions in container cultivation. There are several systems of analysis, however, the routine procedures to determine the water retention curve (WRC) are time consuming and inaccurate. This study proposed a reproducible, less time-consuming, and easy to perform method for physical characterization of substrates. We used Sphagnum peat, coconut powder, carbonized rice husk, expanded vermiculite, and expanded clay in isolation. The WRC was obtained at tensions 0, 10, 50, and 100 hPa using the method of Büchner funnel, the European standard (CEN-EN13041), and by the centrifugation method of samples. In centrifugation, tensions were simulated by combining the angular velocity, sample height, and radius measured from the sample outermost point to the central point of the centrifuge. Three rotations were applied (230; 520 and 730 rpm), testing the centrifugation time for each tension and then the volumetric water content was compared between the methods. The results showed a positive correlation (p < 0.001) between the centrifugation and standard methods, considering that the centrifugation time required for moisture stabilization varied according to the material analyzed and the tension applied. However, the time required to obtain a report was reduced by half. Sample centrifugation is a promising method, ensuring that the tensile force actually acts on the entire sample to determine the water retention curve of the substrate also reducing the duration of the analyses.

Background: Semen extenders are required to protect and preserve semen, and the development of suitable extenders iskey for artificial insemination. Although the use of Tris-based diluent is widespread, new diluents such as powdered coconut water have been developed for better sperm protection. One way to evaluate the effectiveness of diluents is throughmicroscopic analyses that evaluate sperm motility, vigor, and concentration. However, these analyses are limited, and maynot provide accurate results. New evaluation techniques have been studied, and one of the tests that can be used to addreliability to these analyses is mitochondrial activity evaluation, which can sum all the parameters, and provide a moreaccurate evaluation. Thus, the present study aimed to evaluate the efficacy of ACP-102c in cryopreserved ram semen.Materials, Methods & Results: Five semen samples were collected from two ram breeders using artificial vagina (n = 10).Each ejaculate was divided into the following two treatments: T1 - ACP-102c + 20% egg yolk + 7% glycerol and T2 - TRIS+ 20% egg yolk + 7% glycerol. Extended semen samples were then packed in 0.5 mL plastic straws, subjected through therefrigeration curve up to 4°C (0.35°C/min), and equilibrated for 2 h at 4°C. Subsequently, the straws were placed at 4 cmabove liquid nitrogen level (-60°C) for 15 min, immersed, and then finally stored in the liquid nitrogen at -196°C. Bothfresh and thawed samples were evaluated for total and progressive sperm motility using conventional microscopy (40x),and the same evaluator on each occasion. O objetivo deste trabalho foi avaliar a relação entre a concentração de proteínas no plasma seminal ovino com parâmetros de qualidade do sêmen criopreservado em diluentes TRIS e em Água de Coco em Pó (ACP102c). Um total de 12 coletas/animal em ovinos Santa Inês (n=2) e Dorper (n=2) foram realizadas para obtenção do plasma seminal e para criopreservação nos diluentes de trabalho. Foi utilizado o método de Bradford para determinação da concentração de proteína. Para avaliação do sêmen fresco e criopreservado foram utilizados os testes de viabilidade por eosina-nigrosina e teste hiposmótico (HOST), além dos parâmetros de motilidade em sistema de análise de sêmen auxiliada por computador (CASA). Baseado no valor médio da concentração de proteína, os ejaculados foram agrupados em alta (AP) e baixa (BP) concentração proteica. Correlações significativamente positivas foram encontradas entre a concentração de proteína com os parâmetros motilidade progressiva (PROG) e congelabilidade (CONGEL), e negativa para deslocamento lateral de cabeça (ALH) nas amostras criopreservadas em ambos os diluentes, além de correlação positiva para motilidade total (MOT) em TRIS. Sêmen de ejaculados do grupo AP criopreservados em TRIS apresentaram MOT, PROG e CONGEL significativamente superior ao grupo BP e inferiores para ALH. Quando criopreservado em ACP102c, sêmen do grupo AP foi superior ao BP para PROG e CONGEL. Variações na qualidade do sêmen criopreservado entre ejaculados podem estar associados à concentração de proteínas do plasma seminal, e estas podem atuar na manutenção da progressividade de espermatozoides ovinos criopreservados em diluentes TRIS e ACP102c.

Semen samples from AP group cryopreserved in TRIS extender were significantly better than BP group for MOT, PROG and CONGEL, and inferior for ALH. When criopreserved in ACP102c extender, the AP group were better than BP group for PROG and CONGEL. Variations in the quality of cryopreserved semen from different ejaculates may be associated with the concentration of seminal plasma proteins and may act to maintain the progressiveness of ram sperm cryopreserved in TRIS and ACP102c extenders.

Abstract Semen cryopreservation is associated with low productivity results. This study aimed to test the Coconut Water Powder (ACP®-103) as a ressuspension diluent after thawing semen, also evaluate sperm quality during the cooling curve until thawing of the semen. For this, the semen was collected from 15 boars once a week, incubated at 30 ºC for 15 minutes, and afterwards, the samples were diluted in the diluent Beltsville Thawing Solution - BTS (Control) or ACP-103®, and subjected to a slow cooling curve, where the force and the motility were analyzed in each step. The thawed semen was ressuspended in its respective solvents and analyzed in the characteristics of vigor, motility, vitality, acrosome integrity and functionality of the membrane. During the analysis of vigor and motility that make up the cooling curve, and thawing, for analysis of vitality and intact acrosomal membrane, it was observed that there was no significant difference between treatments. Besides, after thawing, the BTS showed better results of sperm vigor, sperm motility and membrane functionality. However, the cooling curve and coconut water powder can be used in the protocol for cryopreservation of boar semen, as both ensured quality of sperm viability that may be used in artificial insemination.

Muitas tentativas são feitas para se melhorar a conservação do sêmen suíno, sendo a gema de ovo conhecida por suas propriedades crioprotetoras. Este trabalho teve por objetivo testar diferentes concentrações de gema de ovo em pó (GOP), adicionada ao diluente água de coco em pó (ACP-103®), e verificar qual mantém melhor a viabilidade espermática. Foram diluídos 36 ejaculados em ACP-103®, acrescidos de diferentes concentrações de GOP (0%, 1%, 3%, 5% e 7%) e conservados a 17 °C. Diariamente, foram realizadas análises de vigor e motilidade e nos dias 1 (D0), 3 (D2) e 5 (D4) de conservação foram feitas as de vitalidade, morfologia e resistência osmótica. Utilizou-se o teste estatístico de Kruskal-Wallis e de Dunn's para dados não paramétricos e ANOVA e Tukey para os paramétricos. Queda de vigor e motilidade foi observada em todos os tratamentos. Os GOP-3%, GOP-5% e GOP-7% foram os que melhor mantiveram o vigor espermático em D0 (2,4±0,8; 2,5±1,1 e 2,8±0,9, respectivamente), sem diferenças significativas entre si. O mesmo ocorreu para a motilidade (77±15%, 74±23% e 81±16% em D0). Os resultados das análises de vitalidade, morfologia e resistência osmótica não diferiram. Em conclusão, as concentrações entre 5 e 7% de GOP adicionado ao ACP-103® permitem sua utilização como diluente eficiente para manter a qualidade espermática


Many attempts have been made to improve the conservation of boar semen. Egg yolk is known to have cryoprotectant properties. This study aimed to test different concentrations of egg yolk added to the coconut milk powder extender (ACP-103®), and verify which one is better to maintain sperm viability. The ejaculated (36) were diluted in ACP-103® supplemented with different concentrations of egg yolk (0%, 1%, 3%, 5% and 7%). The conservation occurred at 17 °C, and vigor and motility analysis were carried out daily. On days 1 (D0), 3 (D2) and 5 (D4) the semen was evaluated for vitality, morphology and osmotic resistance. For statistical nonparametric analysis Kruskal-Wallis and Dunn's tests were performed, and for parametric data ANOVA and Tukey test were used. A decrease in vigor and motility was observed in all treatments. Treatments T2, T3 and T4 better maintained spermatic on D0 (2.4±0.8, 2.5±1.1, and 2.8±0.9, respectively), with no significant differences among them. The same was observed for motility (77±15%, 74±23%, and 81±16% on D0). Analyses of vitality, morphology and osmotic resistance did not show statistical difference among treatments. In conclusion, the concentration of egg yolk (7%) added to the ACP-103® can be effectively used as extender to maintain sperm viability

Abstract The use of appropriate extenders is important for the success of an artificial insemination program. The objective of this study was to evaluate the efficiency of alternative extenders for swine semen at different temperatures (17 to 10 °C). The following extenders were used: Beltsville Thawing Solution (BTS), powdered coconut water (ACP-103®), and skimmed milk powder (LPD). The 50 ejaculates were analyzed daily, in natura and after dilution, during the 5-day period of semen preservation (D0 to D4), regarding spermatic vigor and motility. Acrosome integrity and sperm viability were evaluated on D0 and D4. Data were analyzed using Mann-Whitney, Students, Tukey and chi-square tests (p 0.05). The LPD extender at 10 °C presented higher motility and sperm vigor compared to BTS and ACP until D2, and to treatments stored at 17 °C. Acrosome vitality and integrity remained higher (p 0.001) with LPD at 10 °C on D0 and D4. LPD showed to be a good extender for the swine semen at lower temperature (10 °C). Furthermore, it provided better protection to sperm cells, by allowing greater integrity and vitality of the acrosome.

O objetivo deste experimento foi avaliar a eficiência de diluentes alternativos para o sêmen suíno, em diferentes temperaturas (17 e 10 °C). Foram utilizados os diluentes: Beltsville Thawing Solution (BTS); água de coco em pó (ACP®-103) e leite em pó desnatado (LPD). Os 50 ejaculados, foram analisados in natura e após diluição, diariamente, durante os 5 dias de conservação do sêmen (D0 a D4), quanto ao vigor e motilidade espermática. Avaliou-se a integridade do acrossoma e a viabilidade espermática no D0 e D4. Na análise estatística, foram utilizados os testes de Mann-Whitney, Students, Tukey e o qui-quadrado, com intervalo de confiança de 5%. O diluente LPD a 10 °C apresentou motilidade e vigor espermático melhores em relação ao BTS e ACP (10 °C) até D2 e aos tratamentos conservados a 17 °C. A vitalidade e integridade acrossomal, no D0 e D4, foram melhores com LPD a 10 °C, que mostrou ser um bom meio diluente para o sêmen suíno em temperatura mais baixa (10 °C), conferindo uma maior proteção à célula espermática, através de maiores taxas de viabilidade e integridade de acrossoma.

Abstract The conservation of boar semen at lower temperatures might contribute to the further expansion of artificial insemination in this species. Egg yolk cryoprotectant properties have already been extensively tested on sperm cryopreservation of several species. This study aimed to test different temperature curves for the conservation of boar semen diluted with coconut milk powdered (ACP®-103) add 7% egg yolk and to verify which one better maintains sperm viability. For this, 36 ejaculates were diluted and stored at 17, 10 and 5 °C. Daily analysis of vigor and motility were performed, and on days D0, D2, and D4 semen was evaluated regarding vitality, morphology, and osmotic resistance. For the statistical analysis we performed the tests of Kruska-Wallis with Dunns post-test (nonparametric data) and ANOVA and Tukey test (parametric data). The storage temperature of 10 °C was the best one to maintain spermatic motility at appropriate levels to be used in an artificial insemination program. Analyses of viability, morphology, and hypoosmotic test did not show statistical difference among the treatments. In conclusion, the best temperature curve was 10 °C with diluted semen previously kept at 17 °C to maintain the viability of sperm cells in pigs for a longer period.


Resumo A conservação do sêmen suíno em temperaturas mais baixas pode permitir uma maior expansão da inseminação artificial nessa espécie. A gema de ovo apresenta propriedades crioprotetoras já amplamente testadas na conservação seminal de diversas espécies. Este trabalho teve por objetivo testar diferentes curvas de temperatura na conservação do sêmen suíno diluído em água de coco em pó (ACP®-103) acrescido de 7% de gema de ovo e verificar qual delas mantém por mais tempo a viabilidade espermática. Para tanto, o sêmen de 36 ejaculados foi diluído e conservado a 17, 10 e 5 °C. Diariamente foram realizadas análises de vigor e motilidade e nos dias D0, D2 e D4 o sêmen foi avaliado quanto à sua viabilidade, morfologia acrossomal e resistência osmótica. Para a análise estatística foram utilizados os testes de Kruska-Wallis, com pós-teste de Dunns (dados não paramétricos) e Anova com teste de Tukey (dados paramétricos). A conservação à temperatura de 10 °C foi a que melhor manteve o vigor espermático e a motilidade em níveis adequados para ser utilizado em um programa de inseminação artificial. As análises de vitalidade, morfologia e teste hiposmótico não apresentaram diferença estatística entre os tratamentos avaliados. Em conclusão, a melhor curva de temperatura foi a de 10 °C com sêmen diluído por manter por um período maior a viabilidade da célula espermática suína.
