Purpose: To evaluate the in vivo efficiency of commercial polymeric membranes for guided bone regeneration. Methods: Rat calvarial critical size defects was treated with LuminaCoat (LC), Surgitime PTFE (SP), GenDerm (GD), Pratix (PR), Techgraft (TG) or control (C-) and histomorphometric analysis determined the percentage of new bone, connective tissue and biomaterial at 1 or 3 months. Statistical analysis used ANOVA with Tukey's post-test for means at same experimental time and the paired Student's t test between the two periods, considering p < 0.05. Results: New bone at 1 month was higher for SP, TG and C-, at 3 months there were no differences, and between 1 and 3 months PR had greater increase growthing. Connective tissue at 1 month was higher for C-, at 3 months for PR, TG and C-, and between 1 and 3 months C- had sharp decline. Biomaterial at 1 month was higher for LC, in 3 months for SP and TG, and between 1 and 3 months, LC, GD and TG had more decreasing mean. Conclusion: SP had greater osteopromotive capacity and limitation of connective ingrowth, but did not exhibit degradation. PR and TG had favorable osteopromotion, LC less connective tissue and GD more accelerated biodegradation.

Nidularium minutum is an ornamental bromeliad from the Brazilian Rainforest. The micropropagation of this species is essential for obtaining plants available for conservation programs or commercial use. Our study aimed to establish an efficient plant production method by in vitro sprouting. This bromeliad takes a long time to sprout in vitro, and 10% of the plants produce shoots in a culture medium without plant growth regulators (PGRs). When subcultured in a PGR-free medium, these individualized shoots can sprout like the mother plant. The Murashige and Skoog basal medium (MS) containing 1.0 mg L-1 of 6-benzylaminopurine (BAP) promoted the induction of adventitious shoots in greater than 90% of the plants after 240 days of culture with an average of more than eight shoots per plant. Approximately 100% of the in vitro-produced shoots survived after acclimatization, reaching the flowering stage. Therefore, our results showed that in vitro regeneration of N. minutum depends on the cultivation period and that plants with a higher sprouting capacity can be selected and used as micropropagation matrices, contributing to the production of this endangered bromeliad.(AU)

Purpose: Nontransmissible chronic diseases, such as diabetes mellitus (DM) and nephropathy, affect a significant portion of the population, often treated due to injuries that require healing and regeneration. To create an experimental model of associated comorbidities, for healing and regeneration studies, protocols for induction of nephropathy by ischemia and reperfusion (I/R) and induction of DM by injection of streptozotocin (STZ) were associated. Methods: Sixty-four mice (Mus musculus), female, adult, Swiss strain, weighing approximately 20 g, were divided into four groups: G1: control (n = 24), G2: nephropathy group (N) (n = 7), G3, DM (n = 9), and G4: N+DM (n = 24). Arteriovenous stenosis (I/R) of the left kidney was performed as the first protocol. The animals received a hyperlipidemic diet for 7 days after the injection of STZ (150 mg/kg, via i.p.) and an aqueous glucose solution (10%) for 24 h. The animals in the G3 and G4 groups were observed for 14 days before receiving the diet and STZ. The evolution of nephropathy was observed using a urine test strip and the DM, through the analysis of blood glucose with a reagent strip on a digital monitor. Results: The ischemic induction protocols of nephropathy and DM with STZ, associated, were sustainable, low-cost, and without deaths. There were alterations compatible with initial renal alterations, in the first 14 days, such as increased urinary density, pH alteration, presence of glucose, proteins and leukocytes, when compared to the control group. DM was confirmed by the presence of hyperglycemia 7 days after induction and its evolution after 14 days. The animals in the G4 group showed constant weight loss when compared to the other groups. It was possible to observe morphological alterations in the kidneys submitted to I/R, regarding coloration, during surgery and after the end of the observation period, in the volume and size of the left kidney, when compared to the contralateral kidney. Conclusion: It was possible to induce nephropathy and DM associated in the same animal, in a simple way, confirmed with rapid tests, without losses, providing a basis for future studies.

Abstract Some studies report the positive effect of organic residues from ant nests on soil properties and on the structure of the adjacent plant community in field experiments, but there is a gap about the effect on individual species. The purpose of the present study was to compare the soil nutrient content and the development of Turnera subulata Smith, an ornamental species, in the presence of the nest refuse (basically composed of fragments of grass leaves and the symbiotic fungus) produced by the leaf-cutting ant Acromyrmex balzani (Emery, 1890) or in control soil through a greenhouse pot experiment. The experiment was carried out with two treatments: control soil and soil with 25% of nest refuse. The plants were kept in 1L pots for 90 days. We evaluated the parameters: plant height, stem diameter, root length, number of leaves, dry weight of the root, dry and fresh aboveground biomass. Additionally, the relative chlorophyll content and leaf nutrients were used as nutritional parameters. As a result, plants that grew in the soil with nest refuse showed significant higher values of all parameters evaluated when compared to the control treatment (p 0.001). We conclude that this biofertilizer contributed to the production of more vigorous plants, being able to act on the local dynamics of nutrients in the ecosystems where A. balzani occurs. As it is relatively abundant and easy to collect, the refuse of A. balzani has the potential to be used as an alternative substrate in the production of shortlife cycle plants.

To gain insights into the forage morphological and anatomical characteristics in a silvopastoral system (SPS) with Bolsa de Pastor (Zeyheria tuberculosa) and palisadegrass 'Marandu' (Urochloa brizantha) monoculture (MONO). The SPS was established through natural regeneration of the tree species. Treatments were a SPS and MONO distributed in a completely randomized design with six replicates and repeated measures were the harvest periods. Response variables were morpho-physiological and anatomical characteristicss: green: dead material ratio, leaf blade: stem+sheath ratio, leaf area index, chlorophyll and carotenoid concentrations, proportions of non-lignified and achlorophyllous areas, lignified areas in stems, proportions of non-lignified and achlorophyllous areas, lignified and chlorophyllous areas in leaves, as well as cell length in longitudinal section of stem. Morpho-physiological patterns were altered (p < 0.05) under natural shading conditions due to higher photosynthetic efficiency in the SPS. There was no effect (p > 0.05) of the systems on anatomical patterns, proportions of non-lignified and achlorophyllous, lignified and chlorophyllous tissues, these proportions were influenced only by the periods of the year, both for stems and leaves. Cells of the internodes of the grasses of the studied systems had the same length. The SPS alters morpho-physiological characteristics of palisadegrass and increases the concentration of chlorophyll a and b.(AU)

Slash-and-burn is a traditional agricultural system still widely used in Brazil. The variation in temperature during fires results in different levels of physical, chemical, and biological changes in the soil, which makes it necessarily better to understand the dynamics of fire severity in this system. The aim of this study was to make an analytical comparison of the variation in the temperature reached in the soil during fires in slash-and-burn agriculture. Temperature data were measured in burnt areas with similar fallow times at the following soil depths 0; 2.5 and 5 cm and compared with secondary data from other studies in the same slash-and-burn system in southern Brazil. The peak temperature showed more significant variations in the surface (0 cm) of different soils and different types of regeneration vegetation in each area. Overall, the peak temperatures ranged from 32 to 673 ºC across the three depths. The peak temperature reduction in the 5 cm layer was 88 % compared to the surface and 30 % compared to the 2.5 cm layer. The sandier soils showed greater thermal conductivity at depth. The surface litter seems to exert a more significant influence on the burning severity than the biomass load cut for burning, suggesting that the characteristics of the litter layer on the surface should be better characterized in future studies on fire severity.(AU)

Logging and agricultural exploitation have led to the degradation of Araucaria Forest remnants and the alteration of its last preserved patches. This forest typology contains many endangered plant species, as is the case of the tree Oreopanax fulvus Marchal. To support conservation of this species and understand how different landscape matrices can influence its populations, we evaluated the demographic structure and spatial distribution of O. fulvus in two Araucaria Forest remnants in Paraná state. We delimited two plots (urban and rural population), each with 1 ha, subdivided them into 100 subplots (10 x 10 m), and recorded diameter at ground level (DGL), height, and coordinates of two post-germinative developmental stage of individuals. In each subplot, we measured slope, luminosity, and canopy height. We used Ripley's K function analysis to describe distribution patterns of the species and the spatial relationship between mature and juvenile trees. We performed correlations between abundance and environmental and structural variables of the O. fulvus populations. Abundance varied between remnants, from 183 individuals/ha (12 mature and 171 juvenile) to 1306 individuals/ha (10 and 1296). The remnants varied in abundance and plant frequency. The species showed an investment in seedling banks. Most juvenile had DGL up to 3.0 cm and height up to 1.0 m and presented aggregated spatial distribution, while adults had random distribution. In the rural population juvenile abundance were correlated with canopy height (positively) and distance to mature trees (negatively). The slope was correlated for both sites, but oppositely, indicating that other factors might have interfered in the regeneration abundance. The urban remnant showed a high abundance of this endangered species, calling attention for potential studies in urban arborization, management and conservation of these remnants.

The success of in vitro cultivation of plant species depends on factors associated with the induction and control of morphogenesis regarding the regeneration of shoots and roots in the organogenesis process, such as the culture medium composition. Thus, the objective of this study was to investigate if theconcentration of growth regulators in culture medium interferes with the in vitro morphogenesis of gypsophila. 'Golan' cultivar was subjected to three culture media (M), which constituted the study treatments: M1) Murashige and Skoog (MS) medium without the addition of growth regulators; M2) MS + 1 mg L-1of benzylaminopurine (BAP) + 0.05 mg L-1of naphthaleneacetic acid (NAA); M3) MS + 0.05 mg L-1BAP + 1 mg L-1NAA. After 45 days, the multiplication rate, plant height and root length were evaluated. The results showed that plantlets produced in M2 medium had a higher multiplication rate. Also, plantlets produced in M1 and M3 media showed higher shoot height and root length. It is concluded that there is a difference in the in vitromorphogenesis of gypsophila according to the concentration of growth regulators in the micropropagation medium.(AU)

The Brazilian Atlantic Forest is considered one of the most threatened tropical forests in the world due to the extensive environmental impact it has endured throughout history. Only 12.4% of its original vegetation is estimated to remain. Even though reduced and highly fragmented, it houses enormous biodiversity, and its preservation is paramount to the maintenance of the country's fauna, flora, funga and microbiota. One of the most efficient measures adopted by public agencies aimed at protecting biodiversity has been the creation of conservation units. To evaluate the preservation state of protected areas, several environmental studies have been performed; species inventories are one among them. Ants are excellent bioindicators, for they are not only sensitive to environmental changes, but they also have a history of being used in impact assessment (i.e., fragmentation). In this study we assessed the ant communities inhabiting the leaf litter in areas with different regeneration states at the RPPN Botujuru Serra do Itapety (Mogi das Cruzes, São Paulo Brazil). Mini-Winkler traps were used in the ant survey, and diversity analyses were performed. In total, we recorded 86 species of ants, with a highlight to Camponotus cillae Forel, 1912, a species that remained unrecorded for the state of São Paulo for over 100 years, and a possible new species of Octostruma Forel, 1912. Overall, the species found show that the areas are in the process of natural regeneration. Our data on RPPN Botujuru is unprecedented, and our species list has the potential of being used as an effective monitoring tool for this conservation unit.(AU)

Purpose: To compare the effect of vein conduit filled with adipose tissue stem cells (ASC) on peripheral nerve injury regeneration. Methods: We analyzed 30 male Wistar rats surgically submitted to a 5-mm gap on the sciatic nerve. Then, the animals were divided into three groups: nerve autografting (AG, n=10), autogenous inverted glycerol-conserved vein (VG, n=10), and autogenous inverted glycerol-conserved vein + ASC (VASCG, n=10). The study endpoints were neuromotor functional analysis, gastrocnemius muscle weight, and sciatic nerve graft histomorphometry analysis. In the histologic analysis, we added a control group (naïve nerve). Results: Regarding functional analysis (Walking tract- score), the findings at week 3 showed a difference between the AG and the VG (-96.6 vs. -59.6, p=0.01, respectively) and between the VG and the inverted vein + VASCG (-59.9 vs. -88.92, p=0.02). At week 12, this study showed a difference between the AG and the VG (-64.8 vs. -47.3, p=0.004, respectively), and also a difference between the VG and the VASCG (-47.3 vs. -57.4, p=0.02, respectively). There was no difference in the histomorphometry analysis (nerve diameter, Schwann cells counting). The gastrocnemius muscles on the intervention side were more atrophic when compared to the gastrocnemius muscles on the control side. Conclusions: Our results suggested better functional recovery in the inverted vein group when compared to control group, and inverted vein + ASC group.

Mulching use in agriculture has been known since 1802 as the practice of spreading dry leaves and straw on the soil to prevent erosion and water loss. Our study evaluated the mulch effectiveness in the establishment of regenerating seedlings and its contribution to the control of exotic grasses. The studies were carried out in an ecological restoration area in the municipality of Itapira-SP. The treatment with mulching consisted in chemical desiccation with glyphosate herbicide application, keeping the dry grass on the ground. In the control treatment, after chemical weeding, the grass was removed with manual mowing, exposing the soil. Eight months after implantation, we sampled all regenerating seedlings in 100 plots of 50 x 50 cm in each treatment. We considered as seedlings all individuals of tree species less than 100 cm tall. We calculated richness, abundance, similarity, and the relationship of the frequency of seedlings to the height of the mulch. We sampled eight species with 42 seedlings, with only one not identified. The highest abundance and species richness were found in the treatment with mulch (n = 34 individuals; eight species), the most abundant being Schinus terebinthifoliaRaddi (14 individuals) followed by Solanum mauritianumScop. (11 individuals), and Platypodium elegansVog. (three individuals). The presence of exoticgrasses was lower in the plots of the mulching (13%) compared to the control treatment (67%). The highest frequency of seedlings was obtained with mulch height from 21 to 37 cm. We suggest that adaptive management practices, such as the use of the mulching technique, can be implemented in ecological restoration areas, because they favor the natural regeneration of native seedlings and can contribute to the control of exotic grasses, but the height of the layer must be controlled.(AU)

Purpose: Poultry by-products can contribute as an innovative natural source for the development of composites based on polymers and minerals aiming at bone regeneration. The objective of this study was the physicochemical and biological characterization of collagen-based hydrogels crosslinked with ultraviolet (UV)-riboflavin. Methods: Pure hydrogels of 100% collagen (G1) or hybrid hydrogels, 90% collagen:10% apatite (G2), 90% collagen:10% nanokeratin (G3), and 90% collagen:5% apatite:5% nanokeratin (G4) were characterized by scanning electron microscope, Fourier-transform infrared spectroscopy, differential scanning calorimetry, swelling degree and quali-quantitative histological analysis. Ectopic implantation in subcutaneous tissue in mice at one, three and nine weeks allowed to assess the inflammation (neutrophils, lymphocytes, macrophages, and giant cells) and repair (neovascularization, and connective tissue) to determine biocompatibility and the integrity of biomaterials to score their biodegradability. Histomorphometry on critical size defects in rat calvaria at one and three months evaluated the percentage of bone, connective tissue, and biomaterials in all groups. Results: The hydrogels presented porous microstructure, water absorption and physicochemical characteristics compatible with their polymeric and/or mineral composition. All materials exhibited biocompatibility, biodegradability, and low osteoconductivity. G2 showed greater density of new bone and biomaterial than the G1, G3 and G4. Conclusions: The collagen-apatite group formulation suggests potential for development as osteopromoting membrane.

Purpose: To evaluate the histopathological, immunohistochemical, and biochemical effects of liver changes after mancozeb administration. Methods: Rats were divided into groupsthe control group (n=7) and the mancozeb group (n=7), given 500 mg/kg mancozeb dissolved in corn oil daily for four weeks by an orogastric tube. Caspase-3 and tumor necrosis factor-alpha (TNF-α) primary antibodies were used for immunohistochemical analysis. Results: Serum aspartate aminotransferase (AST) and alanine aminotransferase (ALT) values of the mancozeb group increased significantly than ones of the control group. Venous dilatation, inflammation, hepatocyte degeneration, TNF-α, and caspase-3 expression scores increased significantly in the mancozeb group. In the mancozeb group, intensive caspase-3 expression was observed in hepatocyte cells around the central vein in the center of the liver lobule, and there was an increase in TNF-α expression in the inflammatory cells around the enlarged central vein and Kupffer cells and apoptotic hepatocyte cells. Conclusions: Subacute mancozeb exposure in rats leads to elevated toxicity with impaired liver function, increased inflammation in tissue and increased apoptosis due to cellular damage in the liver, and decreased liver regeneration ability due to congestion and degeneration of blood vessels.

Purpose: To analyze the cytotoxicity and cell in porcine-derived decellularized skin matrix. Methods: We analyzed the effect of multiple decellularization processes by histological analysis, DNA quantification, and flow cytometry. Subsequently, we analyzed the most appropriate hydrogel concentration to minimize cytotoxicity on fibroblast culture and to maximize cell proliferation. Results: After the fourth decellularization, the DNA quantification showed the lowest DNA concentration (< 50 ng/mg). Histological analysis showed no cell components in the hydrogel. Moreover, hematoxylin and eosin showed a heterogeneous structure of collagen fibers. The best hydrogel concentration ranged from 3 to 25%, and there was no significant difference between the 24 hours and seven days. Conclusions: The process of hydrogel production was effective for removing cells and DNA elements. The best hydrogel concentration ranged from 3 to 25%.

Purpose: To evaluate the effect of preoperative intravenous chemotherapy with 5-fluorouracil on liver regeneration in an experimental model of major hepatectomy in rats. Methods: Wistar rats were divided into two groups of 20 animals each and submitted to 70% hepatectomy 24 h after intravenous injection of 5-fluorouracil 20 mg/kg (fluorouracil group, FG) or 0.9% saline (control group, CG). After hepatectomy, each group was subdivided into two subgroups of 10 animals each according to the day of sacrifice (24 h or 7 days). Liver weight during regeneration, liver regeneration rate using Kwon formula, and the immunohistochemical markers proliferating cell nuclear antigen (PCNA) and Ki-67 were used to assess liver regeneration. Results: At early phase (24 h after hepatectomy) it was demonstrated the negative effect of 5-fluorouracil on liver regeneration when assessed by Kwon formula (p < 0.0001), PCNA analysis (p = 0.02). With regeneration process complete (7 days), it was possible to demonstrate the sustained impairment of chemotherapy with 5-fluorouracil on hepatocytes regeneration phenomenon when measured by Kwon formula (p = 0.009), PCNA analysis (p = 0.0001) and Ki-67 analysis (0.001). Conclusions: Preoperative chemotherapy with intravenous 5-fluorouracil negatively affected the mechanisms of liver regeneration after major hepatectomy in rats.

Purpose: Various postoperative protocols have been proposed to improve outcomes and accelerate nerve regeneration. Recently, the use of physical exercise in a post-surgical neurorraphy procedure has shown good results when started early. We experimentally investigated the hypothesis that post-operative exercise speeds up results and improves clinical and morphologic parameters. Methods: Isogenic rats were randomly divided into four groups: 1 SHAM; 2 SHAM submitted to the exercise protocol (EP); 3 Grafting of the sciatic nerve; and 4 Grafting of the sciatic nerve associated with the EP. The EP was based on aerobic activities with a treadmill, with a progressive increase in time and intensity during 6 weeks. The results were evaluated by the sciatic functional index (SFI), morphometric and morphologic analysis of nerve distal to the lesion, and the number of spinal cord motor neurons, positive to the marker Fluoro-Gold (FG), captured retrogradely through neurorraphy. Results: Functional analysis (SFI) did not show a statistical difference between the group grafted with (50.94) and without exercise (-65.79) after 90 days. The motoneurons count (Spinal cord histology) also showed no diference between these groups (834.5 × 833 respectively). Although functionally there is no difference between these groups, morphometric study showed a greater density (53.62) and larger fibers (7.762) in GRAFT group. When comparing both operated groups with both SHAM groups, all values were much lower. Conclusions: The experimental model that this aerobic treadmill exercises protocol did not modify nerve regeneration after sciatic nerve injury and repair with nerve graft.

Purpose: To study the influence of silver nanoparticles (AgNP) on tissue reaction when incorporated into a polymeric matrix of polyacrylic acid-based (Carbopol®) gel as a proposal for a new low-cost type of biomaterial that is simple to manufacture for use as an antimicrobial and antioxidative dressing. Methods: In-vivo tests of implantation in the subcutaneous tissue of the back of rats were performed using polyethylene tubes in three situations: empty, only the gel, and gel incorporated with AgNP. Then, the tissue reaction was studied by counting inflammatory cells. Additionally, in-vitro tests of the antioxidative and antimicrobial activity of AgNP were performed. The radical 2,2 diphenyl-1 picrylhydrazyl (DPPH) was used to test the antioxidative activity of AgNP using electron spin resonance. The antimicrobial activity of AgNP was determined by minimum inhibitory concentration against the microorganisms: Staphylococcus aureus, Pseudomonas aeruginosa, and Escherichia coli. Results: The results indicated that AgNP presents antioxidative activity and was able to inhibit the growth of the microorganisms tested. The addition of AgNP in Carbopol® did not alter the tissue inflammatory response (p>0.05, Kruskal-Wallis's test). Conclusions: The new biomaterial is promising for future use as a dressing for its beneficial properties for regenerative processes
