Resumo

Background: The uncontrolled multiplication of Sertoli cells causes Sertoli cell tumor or Sertolioma. Because of this, the level of estrogen in the bloodstream increases rapidly and approximately 25% of dogs with this tumor develop feminization syndrome. Testicular neoplasms are more common in dogs than cats, and are often found in elderly patients. This work aims to describe the clinical signs of the feminization syndrome and the treatment instituted in a canine diagnosed with sertolioma. Case: A 18-year-old male canine, 19.5 kg of body mass, with an increase in testicular volume for about 2 years, was treated at the University Veterinary Hospital. On clinical examination, a matte and brittle coat, alopecia on the hind limbs and gynecomastia were observed. Also noted, non-harmonious aspect of the scrotum, pendular foreskin, atrophied right testicle and hyperplastic left, scrotal hyperthermia and absence of pain. In addition, as a result of the hyperestrogenism resulting from the neoplasm, the paraneoplastic syndrome of feminization, the patient also presented galactorrhea, pendular foreskin, atrophy of the penis and the contralateral testicle, dermatopathies, such as bilateral symmetrical alopecia of the flanks, easily removable hair and variable hyperpigmentation. Rectal body temperature of 38.6°C, clear lung auscultation and muffled cardiac auscultation. The results of laboratory tests showed changes such as thrombocytopenia, platelet counts below the reference levels, platelet count of 163,000/uL. There were no alterations that represented metastases in the imaging exams, such as in the chest X-ray in three incidences and in the abdominal ultrasonography. Then, we opted for the surgical procedure of orchiectomy, with the traditional technique of three clamps, associated with total ablation of the scrotum. Samples were sent to the histopathology laboratory and the diagnosis of sertolioma was confirmed. At 10, 30 and 90 days after the operation, the patient was reassessed for possible recurrences or alterations, but there were no complications or recurrence after the procedure. Discussion: Neoplasms of the male reproductive system are common in dogs. Sertolioma is considered one of the most frequent neoplasms in elderly dogs and that results in systemic clinical signs. This is in line with the 18-year-old dog described in the present report. In addition, it may result in clinical signs resulting from hyperestrogenism resulting from the neoplasm that is called paraneoplastic feminization syndrome. The characteristics of this syndrome are: gynecomastia, galactorrhea, pendular foreskin, atrophy of the penis and contralateral testicle, associated with dermatopathies, such as symmetrical bilateral alopecia. All these clinical signs were present. The diagnosis is made through complete anamnesis, complete clinical examination and complementary examination such as ultrasound help in the presumptive diagnosis, but only with histopathology can it be confirmed. In the clinical approach, histopathology was performed to close the diagnosis. Treatment is behind orchiectomy and total ablation of the scrotum, which was performed in the reported case. The treatment of choice was easy to apply, in addition to improving the patient's quality of life, promoting rapid post-surgical healing and an early return to normal life. However, for the effectiveness of the technique, the early diagnosis and collaboration of tutors is fundamental.

Background: Testicular neoplasms in dogs are more frequent than in other animal species, representing the most common tumors in elderly subjects after skin neoplasms. In cryptorchid subjects, the risk of neoplastic degeneration is high. The cytological examination is essential to differentiate the type of neoplasia and to determinate the best diagnostic approach. Aim of this report was to describe clinical and histopathological features of a dog with coexistence of Sertoli cell and interstitial endocrine cell tumors in a non-cryptorchid dog. Case: A 9-year-old non-neutered male dog, German Spitz breed, was presented to the veterinary clinic. On clinical examination, the dog had a body condition (BCS 6/9), pink mucous membranes, capillary refill time (CRT) < 2 sec, lymph nodes of normal size, afebrile, normal heart and respiratory rate. The abdomen was depressible to palpation, without pain, the skin appeared hyperpigmented, with generalized presence of comedones, pendular foreskin and absence of hair at the abdominal level, in the ventral portion of the trunk and neck, scant hair also at the level of the inner thighs and in perianal. At ultrasounds examinations, the right testicle presented a hypoechoic circular focal lesion, in the caudal pole, of 0.7 cm in diameter, well defined, echogenicity and a remnant of normal echostructure, smooth and regular margins; the left testicle showed an increase in size, irregular margins, with a heterogeneous echo structure, given by hypoechoic areas, referred to cysts, hemorrhagic or necrotic areas. The hemogram reported slightly microcytic and normochromic regenerative anemia. The leukogram showed monocytosis. The absence of the typical stress leukogram characterized by neutrophilia, lymphopenia and eosinopenia, and the reduction of ALP allows to rule out Cushing's disease. In order to rule out hypothyroidism due to the inhibitory effect of estrogens on the release of thyroid-stimulating hormone (TSH) the concentration of total thyroxine was analyzed, reporting normal values excluding hypothyroidism. Blood oestradiol 17-ß (E2) concentration was increased, with a normal testosterone (T) concentration of 0.30 ng/mL. Given the suspicion of the presence of testicular tumors, castration was performed by the surgical excision of both gonads, after ruling out the presence of abdominal or pulmonary metastases by chest and abdominal radiography. Discussion: The clinical, histopathological findings supported the diagnosis of testicular tumors. According to the pathological report, both gonads presented parenchymal nodular neoplastic nodular areas referring to the Sertoli sustentacular cells in the right testicle, to the proliferation of Leydig interstitial cells in the left one. The nodule in the left testicle was unencapsulated and showed a solid-diffuse pattern. Neoplastic cells were irregular polygonal, medium to large in size with moderate nucleus/cytoplasm ratio and moderate anisocytosis. In the right testicle, the nodule was heterogeneous in consistency and a diffuse pattern was present. Neoplastic cells were polygonal morphology, had a moderate nucleus/cytoplasm ratio and were organized tubules lined and obliterated the extensive cell growth. Bilateral orchiectomy allowed to improve the clinical signs, and 3 months after surgery, the animal was in good health, with evident improvement skin lesions. The E2 analysis was repeated, detecting normal values, demonstrating that testicular neoplasm in this patient were involved in E2 production; also T concentration decreased considerably from 0.30 to < 0.07 values.

O tumor de células de Sertoli, classificado como tumor do cordão sexual puro, é caracterizado por provocar manifestações clínicas endócrinas. É encontrado mais comumente em machos e é raro em fêmeas. O presente estudo objetivou relatar um caso de tumor ovariano de células de Sertoli em uma cadela. A paciente era uma fêmea canina, de cinco anos de idade da raça Shi Tzu, que apresentava queixa de "posição de prece" com frequência e anestro há dois anos. O exame físico determinou alopecia difusa e simétrica, hiperpigmentação de pele, pelos frágeis e comedões, com aumento de volume no flanco esquerdo e sensibilidade à palpação abdominal. O exame ultrassonográfico evidenciou estrutura arredondada, de bordas definidas em topografia de ovário esquerdo, medindo aproximadamente 5,27cm em seu maior eixo; além de piometra incipiente. Após ovariossalpingohisterectomia, realizou-se avaliação histopatológica do ovário, com diagnóstico de tumor do cordão sexual/estroma, com padrão de células de Sertoli infiltrativo. Conclui-se que, apesar de incomum, é importante incluir o tumor de células de Sertoli como diagnóstico diferencial, em casos de sinais de desregulação hormonal e alterações no ciclo estral e do pelame do animal. Ressalta-se a importância dos exames de imagem e histopatológicos para o diagnóstico, prognóstico e tratamento adequado.


Sertoli cells tumor, classified as a pure sexual cord tumor, is characterized by causing endocrine clinical manifestations. It is commonly found in males and is rare in females. The present study aimed to report a case of a Sertoli cell ovarian tumor in a female dog. The patient was a five-year-old female Shi Tzu dog, who complained of prayer position frequently and anestrus for two years. The physical examination determined diffuse and symmetrical alopecia, skin hyperpigmentation, fragile hair, and comedones, with increased volume in the left flank and sensitivity to abdominal palpation. The ultrasound examination evidenced the presence of a rounded structure, with defined edges in the topography of the left ovary, measuring approximately 5.27cm in its longest axis, in addition to incipient pyometra. After ovariosalpingohysterectomy, a histopathological evaluation of the ovary was performed with a diagnosis of sexual cord/stroma with an infiltrative Sertoli cell pattern. It is concluded that, although uncommon, it is important to include the Sertoli cell tumor as a differential diagnosis in cases of signs of hormonal dysregulation and changes in the estrous cycle, and animal's coat. The importance of imaging and histopathological exams for diagnosis, prognosis, and appropriate treatment is highlighted.

La hormona anti-Mülleriana (HAM) es una glicoproteína dimérica de 140 kDa que pertenece a la superfamilia del factor de crecimiento transformante ß. La HAM es producida por las células de Sertoli de los testículos fetales siendo responsable de la regresión de los conductos müllerianos durante la diferenciación sexual masculina. En la hembra la HAM circula a niveles casi indetectable al nacer, con un ligero aumento en los primeros años de vida hasta la pubertad cuando se expresa en las células de la granulosa de folículos preantrales y antrales pequeños del ovario. En hembras adultas, la HAM actúa como un regulador autocrino y paracrino inhibiendo el reclutamiento de folículos primordiales y reduciendo la respuesta a la hormona folículo estimulante. En el gato doméstico el número de estudios de HAM es muy escaso quedando aún abundantes aspectos por definir en la especie. También la validación de un único inmunoensayo que permita la comparación de valores séricos entre los trabajos es necesaria. El objetivo de esta revisión fue explicar aspectos básicos de la fisiología de la HAM y resumir las principales publicaciones en el gato doméstico. Finalmente, se proponen aplicaciones prácticas para esta hormona en la especie.(AU)

O HAM é produzido pelas células de Sertoli dos testículos fetais, sendo responsável pela regressão dos ductos de Müller durante a diferenciação sexual masculina. Na fêmea, o HAM circula a níveis quase indetectáveis ao nascimento, com um ligeiro aumento nos primeiros anos de vida até a puberdade, quando se expressa nas células da granulosa de folículos préantrais e antrais pequenos do ovário. Nas fêmeas adultas, o HAM atua como um regulador autócrino e parácrino inibindo o recrutamento de folículos primordiais e reduzindo a resposta ao hormônio folículoestimulante. No gato doméstico o número de estudos do HAM é muito escasso, faltando vários aspectos a serem definidos na espécie. Também é necessária a validação de um único imunoensaio que permita a comparação de valores séricos entre trabalhos. O objetivo desta revisão foi explicar aspectos básicos da fisiologia do HAM e resumir as principais publicações no gato doméstico. Finalmente, se propõe aplicações práticas para este hormônio na espécie.(AU)

Abstract Neuroendocrine substances play essential roles in regulating the normal physiological functions of testicles. The purpose of this study is to explore the localization and effects of four neuroendocrine markers (NSE, SP, NFH and DβH) in normal and cryptorchid testes of Bactrian camels using western blotting, transmission electron microscopy, immunohistochemistry, and immunofluorescence methods. The results showed that cryptorchidism caused a reduction in layers of spermatogenic epithelium and decreased glycogen positivity in the basement membrane. The ultrastructure revealed that macrophages were always found around the Leydig cells, crowded with swelling mitochondria in cryptorchidism. Expression of NSE in the Leydig cells of cryptorchidism was significantly weakened compared to that in the normal group(p<0.01). We found that SP was always distributed along the nerve fibers in normal testes and was expressed in the Leydig cells of cryptorchidism. However, expression of NFH in the cryptorchidic tissue was strongly positive in the spermatogenic epithelium, with limited expression in Leydig cells and no expression in peritubular myoid cells. Therefore, the expression of DβH in the Sertoli cells was comparatively strong in both the normal and cryptorchidism groups. NFH and DβH expression was significantly increased in the cryptorchidism group compared with the normal group (p<0.01). These findings indicated that the underdeveloped seminiferous epithelium and pathological changes in cryptorchid tissue in Bactrian camels were potentially related to a disorder in glycoprotein metabolism. Our results suggest that NSE and SP could help judge the pathological changes of cryptorchidism. The present study provides the first evidence at the protein level for the existence of NFH and DβH in Sertoli and Leydig cells in Bactrian camel cryptorchidism and provides a more in-depth understanding of neuroendocrine regulation is crucial for animal cryptorchidism.

The Ziwuling black goat is an indigenously in China, their offspring are frequently affected by congenital cryptorchidism. The extracellular matrix (ECM) contains cytokines and growth factors that regulate the development of the testis, and component changes often result in pathological changes. Cryptorchidism is closely related to structural changes in ECM. In this study, the histochemical staining, immunohistochemical, immunofluorescence and Western blot combined with semi-quantitative analysis was used to describe the distribution of the important ECM components Collagen type IV (Col IV), laminin (LN)and heparan sulfate proteoglycans (HSPG) in the normal and cryptorchid testes of Ziwuling black goats. Results showed that: The histochemical staining showed that the dysplasia of seminiferous tubules and decreased number of Sertoli cells in cryptorchidism, as well as sparse collagen fiber. Meanwhile, the distribution of reticular fibers is relatively rich. Furthermore, the PAS and AB staining in the interstitial vessels and lamina propria of seminiferous tubules is weak. The immunohistochemical and immunofluorescence revealed that Col IV, LN was strongly expressed in Leydig, Sertoli cells of normal testes and moderately positive in the spermatogonia and spermatids, but HSPG was not expressed in the spermatogonia. However, cryptorchidism, the expression of Col IV, LN and HPSG in Leydig, Sertoli cells significantly decreased, as well as the expression of Col IV and LN in capillary endothelial cells, but HSPG was moderately expressed in spermatogonia. Based on these data, the underdevelopment of spermatogenic epithelium, decreased synthesis function of collagen fibers and Leydig cells develop usually in the cryptorchidism were shown to be closely related to the abnormal metabolism of Col IV and LN. The positive expressed of HSPG in the spermatogonia of cryptorchid testes is related to the compensatory development of spermatogonia.(AU)

O procedimento de imunocastração vem sendo utilizado como técnica alternativa amplamente favorável ao bem-estar animal, por ser indolor, pouco invasiva e com eficácia semelhante à da castração cirúrgica, que causa maior estresse aos animais, principalmente quando realizada de forma inadequada. A imunocastração estimula a produção de anticorpos contra o Hormônio Liberador de Gonadotrofina (GnRH), bloqueando temporariamente a produção da testosterona pelas gônadas masculinas. Face a escassez de informações sobre este procedimento em touros bubalinos, o estudo teve como objetivo analisar os possíveis efeitos da imunocastração no parênquima testicular de búfalos. Vinte touros bubalinos, com idade entre 2 a 3 anos, oriundos da ilha do Marajó ­ PA, foram avaliados e divididos aleatoriamente em dois grupos composto por dez animais no grupo controle (GC) e dez do grupo imunocastrado (GIM). O produto utilizado foi a vacina anti-GnRH Bopriva ® (Zoetis, SP, Brasil). O GIM recebeu duas doses de 1,0ml contendo 400µg da vacina Bopriva ® com intervalo de 8 semanas entre as aplicações e o grupo controle recebeu 1,0 ml de solução fisiológica. Após 14 dias da última dose, os animais foram abatidos e coletado os 20 pares de testículos para analises de parâmetros macroscópicos como: comprimento, largura, circunferência e peso. Além disso, foi realizada a retirada de fragmentos do parênquima testicular para confecção de lâminas histológicas. Para comparação dos dados foi aplicado o teste t de Student (Nonparametric Test) considerando a significância com p<0,05 e intervalo de confiança de 95%. Foi observado diferença significativa entre os dois grupos para grupos para peso (direto p<0001; esquerdo p≤ 0,0001), circunferência (direita p< <0,0001), 1; esquerda p<0,0001), ), largura (somente lado direito p<0,0372) e comprimento (direita p≤ 0,0013; esquerda p<0,0437). Na avaliação microscópica, os animais do GC não apresenteram alterações. Nos animais do GIM houve degeneração em todas as amostras, sendo visualizado descamação, tortuosidade e espessamento da membrana basal do túbulo seminífero, assim como vacuolização e atrofia das células de Sertoli. Foi observado também uma redução do número de células de Leydig, fibrose intertubular pronunciada, núcleos picnóticos, azoospermia e células multinucleadas no interior dos túbulos. Conclui-se a vacina anti-GnRH mostrou-se eficaz em provocar lesões no parênquima testicular, comprometendo significativamente a espermatogênese a partir da possível supressão de testosterona.(AU)


The immunocastration procedure has been used as an alternative technique widely favorable to animal welfare, as it is painless, minimally invasive and with similar efficacy to surgical castration, which causes greater stress to animals, especially when performed improperly. Immunocastration stimulates the production of antibodies against Gonadotropin Releasing Hormone (GnRH), temporarily blocking the production of testosterone by the male gonads. Due to the scarcity of information about this procedure in buffalo bulls, the study aimed to analyze the possible effects of immunocastration on the testicular parenchyma of buffaloes. Twenty buffalo bulls, aged between 2 and 3 years, from the island of Marajó - PA, were evaluated and randomly divided into two groups consisting of ten animals in the control group (CG) and ten from the immunocastrated group (GIM). The product used was the anti-GnRH vaccine Bopriva ® (Zoetis, SP, Brazil). The GIM received two doses of 1.0ml containing 400µg of Bopriva ® vaccine with an interval of 8 weeks between applications and the control group received 1.0 ml of saline solution. After 14 days of the last dose, the animals were slaughtered and the 20 pairs of testes were collected for analysis of macroscopic parameters such as: length, width, circumference and weight. In addition, fragments of the testicular parenchyma were removed for the preparation of histological slides. To compare the data, Student's t test (Nonparametric Test) was applied, considering the significance with p< 0.0372) and length (right p≤ 0.0013; left p<0.0437). In the microscopic evaluation, the animals of the CG showed no alterations. In the GIM animals there was degeneration in all samples, with desquamation, tortuosity and thickening of the seminiferous tubule basement membrane, as well as vacuolization and atrophy of Sertoli cells. A reduction in the number of Leydig cells, pronounced intertubular fibrosis, pyknotic nuclei, azoospermia and multinucleated cells within the tubules were also observed. In conclusion, the anti-GnRH vaccine proved to be effective in causing lesions in the testicular parenchyma, significantly compromising spermatogenesis from the possible suppression of testosterone.(AU)

Resumo

Infertility is one of the most prevalent health disorders in reproductive-age males and females. Ficus carica (Fc), an herbal plant, has been used traditionally for the treatment of different diseases such as infertility especially in Iranian folk medicine. This study examined the effects of Fc leaf extract on the proliferation of mice spermatogonial stem cells (SSCs). Phenolic, flavonoid content, major polyphenolic compounds and antioxidant activity of the extract was evaluated respectively by Folin-Ciocateu, aluminum chloride, HPLC and the FRAP and DPPH methods. Testicular cells of neonate mice were extracted and their identity was confirmed using cytokeratin for Sertoli and Oct-4, CDHI and PLZF for SSCs. Effects of Fc (0.0875, 0.175, 0.35, 0.71 and 1.42 mg/ml) was evaluated at third, 7th, 9th and 14th days of culture by colony assay. The expression of the Mvh, GFRα1 and Oct-4 genes and the viability and proliferation of cultured cells was assessed at the end of the culture period. The extract has a rich phenolic and flavonoid content such as Rutin, Psoralen, Bergapten and Caffeoylmalic acid using HPLC analysis. It also had a potent reducing and radical scavenging activity. Morphology of colonies was similar in all groups. Higher viability, proliferation, colony number and diameter of SSCs was seen in the presence of Fc leaf extract in a dosedependent manner so that higher number and diameter of colonies were observed in two higher doses of 0.71 and 1.42 mg/ml, separately for each time point relative to other groups. The Mvh, Oct-4 and GFRα1 genes expression had no significant differences between groups. It seems that Fc leaf extract not only had no any cytotoxic effects on the viability and proliferation of SSCs but also support their stemness state. So, this culture system can be employed for enrichment of germ stem cells for use in clinical applications.(AU)

Resumo

This review addresses advances, directions and opportunities for research on sheep reproduction in the context of the global challenges of food security and climate change, and demand for 'clean, green and ethical' (CGE) animal management. The foundation of CGE management is an understanding of the physiological processes through which the reproductive system responds to changes in the animal's environment. These days, to the main environmental factors (photoperiod, nutrition, pheromones), we need to add stress from extreme weather events. With respect to nutrition in rams, we now have a deeper understanding of the responses of the brain centres that control gonadotrophin secretion (the kisspeptin system). At testis level, we have found that nutrition affects non-coding RNAs in Sertoli cells and germ cells, thus affecting the balance between cell proliferation and apoptosis. This proliferation-apoptosis balance is also affected during prenatal development, when undernutrition or stress in pregnant ewes seems to elicit epigenetic changes in developing gonads that could affect offspring fertility in adult life. With respect to nutrition in ewes, metabolic signals act directly on ovarian follicles, and thus change ovulation rate, but the variety of signals now includes the adipokines. An early concern was that nutritional supplements that increase ovulation rate would also increase embryo mortality but we now know that embryo survival is improved under field conditions. Finally, we had always thought that the efficiency gains from early puberty in lambs could only be achieved by accelerating fat accumulation, but we now know that faster muscle growth will achieve the same goal, offering two advantages in meat production systems. With respect to pheromones ('ram effect'), we have a deeper understanding of the brain responses (kisspeptin system) but, most importantly, a realization that the response of ewes to the ram signal involves cell division in memory centres. Many opportunities remain.(AU)

Neoplasias ovarianas de origem primária são incomuns em cadelas, exigindo métodos diagnósticos e terapêuticos mais precisos. Fêmeas também possuem células intersticiais secretoras de esteróides, contudo o aparecimento do tumor de células de Leydig é considerado raro. Uma cadela sem raça definida e 12 anos apresentava sinais de estro após ovariohisterectomia pré-púbere. Na avaliação ultrassonográfica, foi identificada uma massa localizada na região abdominal lateral direita, sugestiva de ovário remanescente. Durante a inspeção laparoscópica foi identificada a massa distante caudalmente do coto ovariano direito, inserida em região de mesentério jejunal. O segmento intestinal foi apreendido e exteriorizado de maneira videoassistida, optando-se pela remoção do tumor associado à enterectomia do segmento afetado. A avaliação do coto ovariano esquerdo, revelou um cisto de 0,5cm de diâmetro que também foi removido. A análise histopatológica foi sugestiva de neoplasia de células intersticiais (tumor de células de Leydig) e um cisto epitelial ovariano. O tumor de células de Leydig é relatado com freqüência em machos, entretanto, sua aparição em fêmeas é considerada extremamente incomum acometendo principalmente animais não esterilizados. A neoplasia não aderida ao coto ovariano comprova ainda que resquícios de tecido ovariano que porventura fiquem livres na cavidade abdominal podem ser responsáveis pelo desenvolvimento da síndrome do ovário remanescente e neoplasias. O prognóstico para neoplasias ovarianas sem metástases é favorável, principalmente quando aplicada a intervenção cirúrgica como método terapêutico. O uso da videocirurgia para auxiliar no diagnóstico e tratamento demonstrou-se adequado para o caso descrito, mas que deve ser complementado pela análise histopatológica.


Primary ovarian neoplasms are uncommon in bitches, requiring more accurate diagnostic and therapeutic methods. Females also have steroid-secreting interstitial cells, however, the appearance of the Leydig cell tumor is considered rare. A 12-year-old mongrel bitch showed signs of estrus after prepubertal ovariohysterectomy. Ultrasound evaluation located a mass in the right lateral abdominal region, suggestive of a remaining ovary. During laparoscopic exploration, the mass was identified caudally distant from the right ovarian stump, adhered to the jejunal mesentery. The intestinal segment was apprehended and exteriorized by video-assisted technique, and removal of the tumor was performed associated with enterectomy of the affected segment. Evaluation of the left ovarian stump revealed a 0.5cm diameter cyst that was also removed. Histopathological analysis was suggestive of interstitial cell neoplasia (Leydig cell tumor) and an ovarian epithelial cyst. The Leydig cell tumor is frequently related in males, however, its appearance in bitches is considered uncommon, affecting mainly nonsterile animals. The mass non-adhered to the ovarian stump proves that remnants of ovarian tissue that may be free in the abdominal cavity and may be responsible for the development of the remaining ovary syndrome and neoplasms. The prognosis for ovarian neoplasms without metastasis is favorable, especially when surgical intervention is applied as a therapeutic method. The use of video surgery to assist in the diagnosis and treatment was considered appropriate for the case described, but it must be complemented by histopathological analysis.

Abstract Glutamine is often used to treat metabolic changes associated with anorexia-cachexia syndrome in patients with malignant neoplasms. Walker 256 tumor is an excellent model for studying these changes associated with cancer in different organs, including injuries in testicular functions. However, the effects of supplementing glutamine on testicular morphometry in this model have not yet been investigated. Thus, the objective of this study was to evaluate the effect of L-glutamine supplementation on testicular morphometry in rats transplanted with Walker 256 tumor cells. Forty puberty Wistar rats were divided into four groups: control without L-glutamine (C); control supplemented with L-glutamine (CG); inoculated with Walker 256 tumor cells (WT) and inoculated with Walker 256 tumor cells and supplemented with L-glutamine (WTG). The testicles were removed, weighed, fixed in Bouin, and included in paraffin for histomorphometric analysis. Walker 256 tumor caused quantitative changes in the tubular and intertubular compartments and tunica albuginea, with reductions in the percentages of lumen and tunica albuginea, number of Sertoli cells per gram of testis; number of Leydig cells; percentage of blood vessels and connective tissue in intertubule. However, glutamine supplementation prevented part of these changes caused by the tumor, presenting mainly a protective effect on the tunica albuginea and percentage of blood and lymph vessels in the intertubule. These results indicate the potential of L-glutamine was able to recover for testicular dysfunction associated with cancer.

The objective was to evaluate the effect of replacing soybean meal with the detoxified castor bean cake on testicular morphometry and spermatogenesis of sheep. Were used 24 uncastrated, 9-month old sheep weighing 29±0.8 kg they were randomly distributed among three treatments: T1 = 0%, T2 = 50%, and T3 = 100% substitution of soybean meal with detoxified castor bean cake. The animals were fed with Aruana grass pastage (Panicum maximum Aruana) and a ration for 90 days. After slaughtering, the testicles were collected and histological slides were prepared with tissue fragments. The data were evaluated for normality using the Shapiro-Wilk test, and analysis of variance was carried out at 5% level of significance. Substitution of soybean meal with detoxified castor bean cake had no effect on any of the assessed variables at the tested levels (P >0.05). The mean yield of spermatogenesis was 72.91 rounded spermatids per spermatogonium; the mean of total number of germ cells held by a Sertoli cell was 12.09; the mean of the testicular spermatic reserve was 31.82×109 and that per testicular gram was 238.28×106; the mean of daily spermatic production was 3.03×109 and that per testicular gram was 22.69×106; and the total number of Sertoli cells was 4.15×109 and that per testicular gram was 34.51×106. The results show that it is possible to replace 100% of the soybean meal with detoxified castor bean cake in sheep diet without any effects on spermatogenesis; however, it is important to perform seminal evaluations in future studies.(AU)


O objetivo com este estudo foi avaliar o efeito da substituição do farelo de soja pela torta de mamona destoxificada sobre a espermatogênese em ovinos. Foram utilizados 24 ovinos, com peso médio de 29±0,8kg e 9 meses de idade, distribuídos aleatoriamente em três tratamentos: T1= 0%, T2= 50% e T3= 100% de substituição do farelo de soja pela torta de mamona destoxificada. Our objective was to investigate whether the pulp and paper mill industry effluent could affect the testis and Sertoli cells in a fast exposure period. For this, the present study was carried out in immature rats at 10-day-old. Testis treated in vitro with 4% effluent for 1 h presented changes in energy metabolism in terms of a decrease in lactate content and glucose uptake. Elevation in GSH content, as an antioxidant defense mechanism, was also detected. Sertoli cells treated with 4% effluent for 1 hour showed alterations in the mitochondrial metabolism that favor the decoupling of oxidative phosphorylation and the generation of oxygen reactive species and also a time and concentration-dependent delay secretion of acidic vesicles. Our results showed that pollutants present in the pulp and paper mill effluents, in a short time of exposure, are capable of inducing alterations in important metabolic functions in the testis and in Sertoli cells that are crucial for the correct progression of spermatogenesis and fertility.

Sertoliomas are among the most common testicular neoplasms in dogs, and one of its main risk factors is disorders in the process of testicular dehiscence, which causes the retention of one or both testicles out of the scrotum. Thus, the present study aims to report a case of a tumor of Sertoli cells (Sertolioma) in an ectopic testicle in the right inguinal region in an SRD dog at Hospital Veterinário Sylvio Barbosa Cardoso, of the State University of Ceará. A 5-year-old, non-neutered male canine, SRD, weighing 18 kg, came to attend an increase in the right inguinal region. On physical examination, the presence of a firm mass located in the subcutaneous tissue in the right inguinal region was identified, in addition to absence of right testicle in the scrotal pouch and a left testicle decreased in size and with softened consistency. In ultrasound evaluation, the left testicle was located in usual topography, with regular contours and standard echogenicity. In the right inguinal region, an echogenic image of heterogeneous echotexture was perceived, indicating the presence of an ectopic testis. After bilateral orchiectomy, both parts were referred for histopathological analysis, presenting a benign Sertolioma in the ectopic testis and testicular degeneration in the topical testicle was reported. The case reinforces the importance of bilateral orchiectomy and ultrasound for abdominal and gonadal evaluation of non-neutered males.
