Resumo

ErpY-like protein (LIC11966) is an antigen from Leptospira spp., which is possibly involved in the infection process and, consequently, can be a promising solution for the development of new diagnostic tests and vaccines. Here, the presence of the erpY-like gene was evaluated in several Leptospira serovars by polymerase chain reaction (PCR), and the ErpY-like recombinant protein was produced and characterized in terms of antigenicity and immunogenicity in vivo. The erpY-like gene was detected by PCR in all Leptospira pathogenic serovars tested (n = 8) and was absent in the saprophytic ones. The rErpY-like protein was recognized by antibodies present in the sera of humans and animals (swine and canine) naturally infected, suggesting ErpY-like expression during natural infection. The rErpY-like protein used to immunize mice with Freund's adjuvant stimulated a mixed Th1/Th2 response, an important protective immunity against leptospirosis.(AU)

Resumo

Neste estudo, 308 amostras de fetos mumificados foram testadas para parvovírus suíno (PPV), circovírus suíno tipos 2 e 3 (PCV2 e PCV3) e leptospiras patogênicas. A idade gestacional no momento da perda gestacional e a frequência da mumificação fetal de acordo com a ordem de parto também foram investigadas. As amostras foram coletadas em granjas comerciais de criação de suínos da região sul do Brasil que apresentassem taxas de mumificação fetal igual ou maiores a 2,5%. Fragmentos de pulmão, rim, fígado e coração de fetos suínos mumificados foram coletados para análise molecular. Resultados da PCR foram classificados de acordo com a região de origem das amostras, tendo Santa Catarina, Paraná e Rio Grande do Sul contabilizado 87 (28,25%), 89 (28,90%) e 132 (42,86%) do total de amostras de fetos suínos mumificados, respectivamente. Coinfecções foram observadas na maioria dos casos e PCV3 foi o agente mais prevalente detectado, encontrado em 298 amostras (96,75%). A maioria das perdas gestacionais foi observada entre 50 e 70 dias de gestação (168; 54,5%) e a mumificação fetal não foi associada à ordem de parto das matrizes. Os achados sugerem que as altas taxas de fetos suínos mumificados na região Sul do Brasil podem ser explicadas pela infecção com esses agentes virais.(AU)

Blood and urine samples were obtained and analyzed by microscopic agglutinationtest (MAT) and isolation through culture, respectively. Among the altered reproductive indicators were identified: totalpigs born (TPB) in 72.5%, stillbirths (SB) by 70%, mummified pigs (MUM) in a 52.5%, pre-weaning death (PWD) by40% and the 24 h mortality (M24h) in the 20%. The 77.5% of the sows were positive by MAT. The predominant serovarsof Leptospira spp. included Grippotyphosa (67.5%), Canicola (22.5%), Icterohaemorrhagiae (20%), Hardjo (17.5%) andPomona (12.5%). Leptospirosis is a major public health threat, and swine are one of the most important reservoirs and sources of Leptospira infection for man. The aim of this study was to investigate the epidemiological situation of leptospirosis in swine in the state of Pernambuco, Brazil. Blood samples from 265 swine were collected and tested by the microscopic agglutination test (MAT), and an epidemiological form containing questions about animal production characteristics, reproductive management, and sanitary and hygiene aspects of the herd was applied to evaluate infection risk factors. The data were analyzed by means of a logistic regression model. An occurrence of 53.1% (143/265) of swine positives to Leptospira spp. was observed. The most commons serovars were Icterohaemorrhagiae (39.1%), Pomona (25.9%), and Shermani (14.0%). Factors associated with the infection were stagnant water source (p = 0.034, odds ratio - OR = 2.29; confidence interval of 95% - 95%CI 1.06-4.93), farms where the healthy animals are bred with sick ones (OR = 1.69; 95%CI 1.04-2.75), and the properties with flooded areas (OR = 1.65; 95%CI 1.01-2.68). The risk factors found in this study played an important role in the agent dissemination and should be avoided in a way to control the disease in the herds studied.(AU)


A leptospirose é um grande problema de saúde pública, e os suínos são um dos mais importantes reservatórios e fonte de infecção por Leptospira para o homem. O objetivo deste estudo foi investigar a situação epidemiológica da leptospirose em suínos no estado de Pernambuco, Brasil. Amostras de sangue de 265 suínos foram coletadas e testadas pelo teste de microaglutinação (MAT), e foi aplicado um questionário epidemiológico contendo perguntas sobre características da produção, manejo reprodutivo e aspectos sanitários e de higiene do rebanho para avaliar fatores de risco de infecção. Os dados foram analisados por meio de um modelo de regressão logística. Foi observada ocorrência de 53,1% (143/265) de suínos positivos para Leptospira spp. Os sorovares mais comuns foram Icterohaemorrhagiae (39,1%), Pomona (25,9%) e Shermani (14,0%). Os fatores associados à infecção foram fonte de água parada (p = 0,034; odds ratio - OR = 2,29; intervalo de confiança de 95% - IC95% 1,06-4,93), fazendas onde animais saudáveis são criados com os doentes (OR = 1,69; IC95% 1,04-2,75) e áreas inundadas (OR = 1,65; IC95% 1,01-2,68). Os fatores de risco encontrados neste estudo desempenharam papel importante na disseminação do agente e devem ser evitados, de forma a controlar a doença nos rebanhos estudados.(AU)

Abstract In swine and bovines, leptospirosis prevention and control is carried out via vaccination of susceptible animals using bacterins. However, the efficiency of leptospirosis vaccines has been questioned. This work aimed to investigate the potency of five leptospirosis vaccines sold commercially in Brazil, challenging the animals with one autochthonous strain of Leptospira, Canicola serovar, denoted LO4, isolated from swine. The standard protocol was followed, and renal carriers of Leptospira were identified among the surviving animals by culture and PCR. Of the five vaccines tested, only two proved effective. None of the surviving animals was positive by culture; however, one animal was positive by PCR. Three of the five vaccines sold commercially in Brazil for the immunization of swine or bovines failed the test of the efficacy to protect the vaccinated animals following challenge with an autochthonous Leptospira strain, Canicola serovar. The two vaccines provided protection against the renal carrier state in the surviving animals. The criteria used to produce leptospirosis bacterins sold commercially in Brazil must be reviewed. The industry should support researches on leptospiral vaccinology to improve the quality of the present vaccines and discover new immunogenic strains, because it is known that vaccination is one of the most important tools to increase the reproduction rates in livestock.

Este estudo foi realizado em suínos do município de Mossoró, Rio Grande do Norte, com o objetivo de caracterizar epidemiologicamente a leptospirose suína em criações não tecnificadas do semiárido brasileiro. Para isso, foi realizado diagnóstico sorológico da infecção pela técnica de soroaglutinação microscópica (SAM), sendo as amostras submetidas a uma coleção de 24 variantes de leptospira, e foram obtidas informações, por meio de visitas às criações e entrevistas com os proprietários, abordando variáveis referentes a criador, características gerais da propriedade, manejo produtivo, reprodutivo e sanitário, com o propósito de realizar análise de fatores de risco. Obteve-se uma prevalência de 78,6% (324/412) de suínos soropositivos. As amostras foram positivas para 15 sorovares, com predominância do sorovar Icterohaemorrhagiae, seguido do Pomona, Shermani e Tarassovi. Os animais foram positivos a até oito sorovares. Os títulos variaram de 100 a 12.800, sendo as maiores titulações contra o sorovar Pomona. Em todas as 20 propriedades trabalhadas (100%) existiam animais reagentes, e a prevalência da infecção em cada propriedade variou de 25% a 100%. Os fatores de risco que estavam associados à leptospirose foram o sistema de confinamento dos animais e o saneamento ruim das instalações de criação. O uso de água tratada na dessedentação dos animais se mostrou fator de proteção. Os resultados mostraram que a Leptospira spp. encontrava-se bastante disseminada nas criações de suínos de Mossoró, com grande variação de sorovares circulantes, representando um problema na produção animal e riscos para a saúde pública.(AU)


This study was conducted in pigs of Mossoró, Rio Grande do Norte, with the objective of epidemiological characterization of swine leptospirosis in herds reared under poor technical and sanitary conditions in the Brazilian semiarid. Serological diagnosis of infection was carried out by means of microscopic agglutination test (MAT), and the samples were submitted to a collection of 24 Leptospira serovars. Information about, variables referring to the farm owner, general property characteristics, productive, reproductive and health management were obtained, by means of visits to the farms and interviews, in order to perform an analysis of risk factors. A prevalence of 78.6% (324/412) of seropositive pigs was observed. The samples were positive to 15 serovars, with predominance of serovar Icterohaemorrhagiae, followed by Pomona, Shermani and Tarassovi. The animals were positive for up to eight serovars. The titles ranged from 100 to 12,800, and the highest titers were observed to serovar Pomona. In all the 20 examined herds there were reactor animals, and the prevalence in each herd ranged from 25% to 100%. The risk factors that were associated with leptospirosis were the confinement system of animals and bad sanitation of breeding facilities. The use of treated water for watering the animals showed a protective factor. The results showed that Leptospira spp. was found to be quite widespread in swine farms of Mossoró, with wide variation of circulating serovars, representing a problem in animal production and risks to public health.(AU)

Para isso, foi realizado diagnóstico sorológico da infecção pela técnica de soroaglutinação microscópica (SAM), sendo as amostras submetidas a uma coleção de 24 variantes de leptospira, e foram obtidas informações, por meio de visitas às criações e entrevistas com os proprietários, abordando variáveis referentes a criador, características gerais da propriedade, manejo produtivo, reprodutivo e sanitário, com o propósito de realizar análise de fatores de risco. Obteve-se uma prevalência de 78,6% (324/412) de suínos soropositivos. As amostras foram positivas para 15 sorovares, com predominância do sorovar Icterohaemorrhagiae, seguido do Pomona, Shermani e Tarassovi. Os animais foram positivos a até oito sorovares. Os títulos variaram de 100 a 12.800, sendo as maiores titulações contra o sorovar Pomona. Em todas as 20 propriedades trabalhadas (100%) existiam animais reagentes, e a prevalência da infecção em cada propriedade variou de 25% a 100%. Os fatores de risco que estavam associados à leptospirose foram o sistema de confinamento dos animais e o saneamento ruim das instalações de criação. O uso de água tratada na dessedentação dos animais se mostrou fator de proteção. Os resultados mostraram que a Leptospira spp. encontrava-se bastante disseminada nas criações de suínos de Mossoró, com grande variação de sorovares circulantes, representando um problema na produção animal e riscos para a saúde pública.(AU)

Reactions were considered positive with minimum titration of 1:100. Considering all serovars, 54.9% of sheep were positive, and the serovar more frequently detected was Celledoni (22.8%), followed by Javanica (19.4%), Castellonis (16.5%), and Norma (15.3%), with predominance of titration of 100 for these serovars, while only one sheep had a titration of 1:800 detected for serovar Hardjo. Rural settlements are defined as family-run agrarian spaces. Their purpose, based on public policies, is to strengthen family agriculture. They are peculiar environments that can predispose to the risk of diseases, especially zoonotic diseases, due to close natural contact with animals that can be sources of infection. The aim of this study was to determine the frequency of anti-Leptospira antibodies in dogs from rural settlements in the state of Paraíba, Northeast Brazil, and to identify the risk factors associated with infection. A total of 306 dogs were sampled and Leptospira screening was performed using the microscopic agglutination test. In all, 27 of 306 (8.82%; 95% CI = 6.13-12.5%) dogs were seropositive for Leptospira spp. The most frequent serotype was Pomona. Street access (odds ratio = 3.53, p = 0.012) was indicated as a risk factor for infection. The results indicate that leptospirosis is present in dogs from rural settlements in the state of Paraíba, Northeast Brazil. This reinforces the need to limit contact between dogs and wild reservoirs and perform serological monitoring of swine. It also reinforces the need for implementation of control measures such as improvements in hygienic-sanitary conditions in the settlements, including the construction of pigsties away from homes, and the introduction of vaccination to reduce the occurrence of this disease, which will prevent transmission to humans andanimals.

Os resultados do presente estudo indicam que a leptospirose está presente em cães de áreas de assentamentos rurais do estado da Paraíba, Nordeste do Brasil. Isso reforça a necessidade de limitar o contato de cães com reservatórios silvestres, bem como fazer o monitoramento sorológico dos suínos e implantação de medidas de controle como melhorias nas condições higiênico-sanitárias nos assentamentos, destacando-se a construção de pocilgas afastadas das residências e a introdução de vacinação com o intuito de reduzir a ocorrência dessa doença e, consequentemente, prevenir a transmissão da infecção aos sereshumanos e animais.
