Due to the complex nature of pain and the participation of physical, cognitive, psychological and behavioral aspects, pain management has several approaches. The use of medicinal plants in developing countries is quite expressive. Seeking new options for the treatment of emerging or debilitating diseases. Therefore, the present study seeks to elucidate the effects of the monoterpene, citronellal, differentiating its activity by isomers (R)-(+) and (S)-(-) citronellal. The study used several methods to evaluate the effects of citronellal isomers on motor coordination, nociceptive response, and the involvement of opioid, glutamatergic, and transient receptor pathways. The methods included rota-rod, hot-plate, and formalin tests, as well as the use of specific inhibitors and agonists. Data were analyzed using inferential statistics with a 95% confidence level. Both isomers did not significantly affect the motor coordination of the studied animals. The isomer (S)-(-) citronellal showed better results in relation to its structural counterpart, managing to have an antinociceptive effect in the formalin and hot plate tests with a lower concentration (100 mg/kg) and presenting fewer side effects, however, the this study was not able to elucidate the mechanism of action of this isomer despite having activity in studies with substances that act on specific targets such as glutamate and capsaicin, its activity was not reversed with the use of antagonists for pathways related to nociception. While the (R)-(+) citronellal isomer, despite showing total activity only at a concentration of 150 mg/kg, was able to determine its mechanism of action related to the opioid pathway by reversing its activity by the antagonist naloxone, being this is a pathway already correlated with nociception control treatments, however, it is also related to some unwanted side effects. In this way, new studies are sought to elucidate the mechanism related to the isomer (S)-(-) citronellal and a possibility of use in other areas related to the treatment of pain or inflammation.

Slash-and-burn is a traditional agricultural system still widely used in Brazil. The variation in temperature during fires results in different levels of physical, chemical, and biological changes in the soil, which makes it necessarily better to understand the dynamics of fire severity in this system. The aim of this study was to make an analytical comparison of the variation in the temperature reached in the soil during fires in slash-and-burn agriculture. Temperature data were measured in burnt areas with similar fallow times at the following soil depths 0; 2.5 and 5 cm and compared with secondary data from other studies in the same slash-and-burn system in southern Brazil. The peak temperature showed more significant variations in the surface (0 cm) of different soils and different types of regeneration vegetation in each area. Overall, the peak temperatures ranged from 32 to 673 ºC across the three depths. The peak temperature reduction in the 5 cm layer was 88 % compared to the surface and 30 % compared to the 2.5 cm layer. The sandier soils showed greater thermal conductivity at depth. The surface litter seems to exert a more significant influence on the burning severity than the biomass load cut for burning, suggesting that the characteristics of the litter layer on the surface should be better characterized in future studies on fire severity.(AU)

Ruminants, as all other animals, feel pain, which can be caused by diseases or frequently practiced procedures, such as dehorning and orchiectomy. However, in these animals the use of analgesics is still less common than ideal, requiring greater awareness, in addition to studies with drugs and protocols capable of effectively relieving pain in these species. In this context, opioids, potent analgesic drugs, are highlighted as a great option for pain control. Nevertheless, their use is still infrequent in ruminants, where studies are limited and show varied results. Therefore, the objective of this study was to review the benefits of pain control in ruminants through opioids, elucidating the analgesic possibilities with different drugs of this class and administration routes to encourage the use by veterinarians of ruminants, based on the literature already published on this subject. Even with some variable results, several studies have demonstrated the antinociceptive potential of opioids in ruminants, in addition to the sedative effects, highlighting the analgesic potential mainly through alternative routes of administration, such as transdermal fentanyl and the epidural route for various opioids, which become important options to be considered in analgesic protocols for ruminants.

This study aimed to determine the psychometric properties and clinical use of the Brazilian version of the Canine Brief Pain Inventory (CBPI) in dogs with hip dysplasia (HD). Forty-three dogs with HD and 16 clinically normal dogs were enrolled. The HD dogs were treated daily with 4.4mg/kg carprofen (GT = 21) or placebo (GP = 19), for four weeks. Owners completed the CBPI at two weeks (W-2) and immediately before the start of the treatment (W0), two (W2) and four (W4) weeks during treatment, and two weeks (W6) after the end of treatment. The internal structure was accessed, and the Cronbach's alpha coefficient was 0.97, indicating the high internal consistency of the instrument. Principal Component Analysis (PCA) suggested the retention of one component, which accounted for 78% of the variability. The ROC curve analysis concluded that the score 3 has an excellent performance to discriminate between normal and possible HD dogs (AUC of 0.973). There was no difference between dogs treated with carprofen versus placebo. The instrument in Portuguese showed construct and criterion validity and reliability to be used in dogs with HD.

Recently, some portions of the Atlantic Forest biome have been suffering an increase in forest fires, possibly changing its vegetation cover, composition, structure and functioning. Understanding these changes is critical to evaluate the present and future response of tropical forests to fire. Thus, the purpose of our study was to evaluate how diversity, structure and functioning of tree communities differed between burned and unburned sites. Two unburned and two burned forest patches were selected for floristic and phytosociological surveys. Then, we calculated species richness, Shannon diversity index, tree density and basal area, Importance Value Index for trees in each site and we assessed community weighted mean of six functional traits (maximum tree height, wood density, leaf length, leaf deciduousness, shade tolerance and dispersal mode). Diversity, species richness, tree density and basal area were similar between sites. We found changes in floristic composition, but did not verified variations in functional traits. Results indicate that recovery may be fast and that pioneer and early secondary species are occupying post burned sites (nine years old). One-time anthropogenic, superficial and low intensity fires might disrupt advanced stages of succession and start again the dynamics of species substitution.

Purpose: To investigate the active ingredients of walnut ointment (WO) and its mechanism in repairing wounds. Methods: The ingredients of WO were detected by gas chromatographymass spectrometry. The effect of linoleic acid (LA) was tested by in vitro Alamar Blue (AB) reagent. Image J software, histological and immunohistochemical analysis were used to confirm the healing effect of LA in the porcine skin model. The animals were euthanized after the experiment by injection of pentobarbital sodium. Results: LA, 24% in WO, promotes keratinocytes and fibroblasts proliferation, which were 50.09% and 15.07% respectively higher than control (p < 0.05). The healing rate of the LA group (96.02% ± 2%, 98.58% ± 0.78%) was higher than the saline group (82.11% ± 3.37%, 88.72% ± 1.73%) at week 3 and week 4 (p < 0.05). The epidermal thickness of the LA was 0.16 ± 0.04 mm greater and the expression of the P63 and CK10 proteins was stronger in the LA group than the control (p < 0.05). Conclusions: LA, which is the main components in WO can promote full-thickness burning wounds (FBWs) by stimulating cell proliferation and differentiation.

We report two outbreaks of nitrate and nitrite poisoning in Paraíba, Northeast Brazil. The first, due to Pennisetum purpureum (elephant grass), and the second, due to P. purpureum and Brachiaria spp. (brachiaria grass), both occurred during a prolonged drought. In the first outbreak, the irrigation of the pastures with wastewater and sewage contributed to nitrate accumulation. The second outbreak occurred in pastures cultivated in the border of a dam, that had been submerged for long time accumulating large amounts of organic matter in the soil. Other probably risk factors for nitrate accumulation included the use of chemical fertilizers and herbicides and burning of the vegetation. In the first outbreak, four calves out of a total of 42 cattle died, and in the second outbreak 49 out of 243 cattle, including adults, yearlings, and a 2-day-old calf died. The clinical signs included dyspnea, cyanosis, ataxia, and falls, leading to death. The presence of nitrates was detected in both outbreaks using the diphenylamine test. Quantitative tests were performed in the second outbreak using a portable nitrate meter, and high nitrate concentrations were found. The characteristic macroscopic findings and absence of microscopic lesions and response to treatment with methylene blue were key to the diagnosis of poisoning by nitrates and nitrites. We conclude that poisoning by nitrates and nitrites in ruminants in the semiarid region of Northeastern Brazil is frequent due to the cultivation of grasses in the border of dams that had been covered by water for long periods or in areas irrigated by wastewater and/or sewage. In addition, the use of a portable measuring device is an effective alternative for the quantification of nitrates in pastures.

Thousands of families live in agrarian reform rural settlements, these existing in Brazil since the 1980s. Factors such as agglomerations of families living in the same environment and the production of domestic animals can promote the transmission and maintenance of zoonosis in these areas. The epidemiology and geographic distribution of zoonotic diseases in settlement communities need to be highlighted. The present study aimed to investigate the prevalence, risk factors and spatial distribution of the teniasis-cysticercosis complex in the agrarian reform rural settlements in the state of Minas Gerais. A total of 497 family farming properties, distributed in 52 settlements, were randomly selected and sampled. Biological samples of humans, cattle and pigs were collected and processed. Survey questionnaires were applied in each family farming property to collect data on animal production as well as sanitary, hygienic and social conditions of each family. Human fecal samples were analyzed for detection of teniasis infection, while animal blood samples were collected and subjected to serological testing to detect the cysticercosis infection. A total of three (0.35%) positive cases of human teniasis were identified, 64 (4.2%) of bovine cysticercosis and 17 (3.3%) of swine cysticercosis. Prevalence per family farming unit was 0.6% (3/497) of taeniasis through fecal test, 5.8% (17/294) for swine cysticercosis and 11.1% (52/469) for bovine cysticercosis. The spatial profile for human teniasis and swine cysticercosis were clustered in two different regions, while bovine cysticercosis showed a dispersed geographical distribution. Two risk factors were associated with the occurrence of bovine cysticercosis: stream as source of water (p=0.009) and the environment as destination of sewage (p=0.031), while burning of garbage was shown to be a significant protective factor (p<0.001). Risk factor for swine cysticercosis was associated with the presence of free range pigs (p=0.008) and the environment as the destination of sewage (p≤0.024). The low number of positive human taeniasis did not allow statistical analysis. These zoonotic diseases represent a significant risk to public health because of their occurrence in livestock which are produced for both beef consumption and for commercialization. Significant endemic areas in the state of Minas Gerais have been determined in this study, and these discoverments suggest the importance of further investment in public health education about teniasis-cysticercosis transmission, the improvement of sanitary facilities for the settlements such as source and treatment of water and adequate destination of sewage. Integrated actions between the human, animal and environmental health sectors, at local and regional levels are needed, aiming at the adoption of effective public policies for the control and eradication of the teniasis-cysticercosis complex where the disease occurs.

Phα1ß is a neurotoxin purified from spider venom that acts as a high-voltage-activated (HVA) calcium channel blocker. This spider peptide has shown a high selectivity for N-type HVA calcium channels (NVACC) and an analgesic effect in several animal models of pain. Its activity was associated with a reduction in calcium transients, glutamate release, and reactive oxygen species production from the spinal cord tissue and dorsal ganglia root (DRG) in rats and mice. It has been reported that intrathecal (i.t.) administration of Phα1ß to treat chronic pain reverted opioid tolerance with a safer profile than ω-conotoxin MVIIA, a highly selective NVACC blocker. Following a recent development of recombinant Phα1ß (CTK 01512-2), a new molecular target, TRPA1, the structural arrangement of disulphide bridges, and an effect on glial plasticity have been identified. CTK 01512-2 reproduced the antinociceptive effects of the native toxin not only after the intrathecal but also after the intravenous administration. Herein, we review the Phα1ß antinociceptive activity in the most relevant pain models and its mechanisms of action, highlighting the impact of CTK 01512-2 synthesis and its potential for multimodal analgesia.

Pain is a common symptom induced during envenomation by spiders and scorpions. Toxins isolated from their venom have become essential tools for studying the functioning and physiopathological role of ion channels, as they modulate their activity. In particular, toxins that induce pain relief effects can serve as a molecular basis for the development of future analgesics in humans. This review provides a summary of the different scorpion and spider toxins that directly interact with pain-related ion channels, with inhibitory or stimulatory effects. Some of these toxins were shown to affect pain modalities in different animal models providing information on the role played by these channels in the pain process. The close interaction of certain gating-modifier toxins with membrane phospholipids close to ion channels is examined along with molecular approaches to improve selectivity, affinity or bioavailability in vivo for therapeutic purposes.(AU)

Background Naja mandalayensis is a spitting cobra from Myanmar. To the best of our knowledge, no studies on this venom composition have been conducted so far. On the other hand, few envenomation descriptions state that it elicits mainly local inflammation in the victims' eyes, the preferred target of this spiting cobra. Symptoms would typically include burning and painful sensation, conjunctivitis, edema and temporary loss of vision. Methods We have performed a liquid-chromatography (C18-RP-HPLC) mass spectrometry (ESI-IT-TOF/MS) based approach in order to biochemically characterize N. mandalayensis venom. Results A wide variety of three-finger toxins (cardiotoxins) and metallopeptidases were detected. Less abundant, but still representative, were cysteine-rich secretory proteins, L-amino-acid oxidases, phospholipases A2, venom 5'-nucleotidase and a serine peptidase inhibitor. Other proteins were present, but were detected in a relatively small concentration. Conclusion The present study set the basis for a better comprehension of the envenomation from a molecular perspective and, by increasing the interest and information available for this species, allows future venom comparisons among cobras and their diverse venom proteins.(AU)

The giant anteater is one of the species classified as vulnerable to extinction. Burning and being run over are among important causes in the decrease of individuals of this species and a better knowledge of the anatomy of these animals can contribute to the treatment of injured animals and their restoration to the environment. Thus, the objective of this work was to describe aspects of the anatomy of the muscles of the forearm of M. tridactyla. For this purpose, six adult specimens were used, three females and three males. The corpses were fixed with a 10% formaldehyde solution and preserved in vats containing the same solution. The thoracic limbs were dissected by routine dissection techniques. The forearm muscles of M. tridactyla were: brachioradialis; radial carpal extensor; common finger extensor; lateral finger extensor, ulnar carpal extensor; finger extensor I and II; long abductor of finger I; supinator, radial carpal flexor; ulnar flexor of the carpus, superficial flexor of the fingers, deep flexor of the fingers, pronator teres and square pronator, which were innervated by the radial, ulnar and median nerves. These muscles give a large volume to the forearm, are robust and have highly developed tendons, especially those involved with the flexion of the carpus, digits and elbow, actions that are fundamental to your defense habits and search for food.(AU)

Background: The giant anteater (Myrmecophaga tridactyla) returned to the category of vulnerable by the 2014 IUCN /SSC, due to the population reduction, less than 30%. The main reasons are: environmental degradation and fragmentation, being run over, burning, attacks by hunting dogs, illegal trade and diseases. It is very common to find these fractured patientsand the treatment is mostly surgical. Amputation is indicated when there are extensive lacerations and when reconstructivesurgery is not possible. The objective is to report a viable treatment option in fractured wild patients, contributing to thepreservation of the species.Case: It was received at the Veterinary Hospital Dr. Halim Atique - UNIRP an young female giant anteater (M. tridactyla)weighing 5.75 kg, brought by the Environmental Forestry Police of the municipality of São José do Rio Preto - SP, foundin the region of Olímpia at a gas station. The animal was traumatized by an agricultural harvester, with partial traumaticamputation in the left pelvic limb and tail. In the preoperative period, the patient was admitted to hospital under assistedcare, analgesia and all the necessary support for stabilization. High amputation was performed, with hip disarticulationand treatment of the tail wound, which was initially opted for healing in the first intention, where sutures were used toapproach the edges of the wound, but presented dehiscence and was treated as a second intention. There were almost 2months of care with specific food, cleaning of the enclosures, medications and dressings. After 57 days of hospitalization, the patient was transferred to the Municipal Zoo of São José do Rio Preto - SP, where it...(AU)

We analysed the frequency of symptoms and degree of muscle pain in selected body parts of racing horses assessed during classic massage sessions. The influence of horse's sex on obtained results was considered. The potential for the early determination of pain in horses by analysing their behaviour and cardiac parameters during a massage session was also evaluated. The study was conducted on 20 three-year-old purebred Arabian horses during one racing season. In the racing season, cyclic classic massage sessions were performed, during which the frequency of symptoms and the degree of pain in the neck, back, croup, front limbs, and hind limbs were analysed. A behavioural assessment of the horses was conducted, and cardiac parameters were analysed. During massage, the frequency of pain symptoms in front limbs amounted to 26, while in croup, it did not exceed 6. The studied horses were most susceptible to pain in the front limbs and in the back, with greater severity in stallions than in mares. An assessment of the frequency and severity of pain symptoms should not be based on changes in behaviour of horses or on cardiac parameters (HR and LF:HF ratio) during massage sessions. However, these methods can be applied after pain reactions intensify. Meanwhile, qualified masseurs can diagnose slight muscle pain during massage sessions.(AU)
