Resumo

A criação comercial de gato doméstico tem aumentado assim como a realização de estudos para melhorar a eficiência reprodutiva é uma realidade. O interesse pela identificação das falhas reprodutivas tem crescido, entretanto, os casos de infertilidade nos machos são escassos na literatura disponível. O histórico de infertilidade de um gato deve ser investigado cuidadosamente, pois pode estar associado a mudanças no manejo ou doenças. O diagnóstico dessas 'falhas reprodutivas' geralmente é simples, se realizada uma boa anamnese, exames, físico e andrológico, minuciosos, assim como exames complementares, entre eles hemograma completo, perfil bioquímico e testes para doenças infecciosas como FIV, FELV e FIP.(AU)


Commercial breeding of domestic cats has increased, and studies to improve reproductive efficiency are a reality. Interest in identifying reproductive failures has increased, however, cases of infertility in men are scarce in the available literature. A cat's history of infertility should be investigated carefully as it may be associated with changes in management or illness. The diagnosis of these 'reproductive failures' is usually simple if a good anamnesis, thorough physical and andrological examinations are carried out, in addition to complementary tests, including complete blood count, biochemical profile, tests for infectious diseases such as FIV, FELV, FIP.(AU)

This study evaluated if the milk ring test (MRT) used to the brucellosis diagnostic at dairy herd level can be done in milk samples with conservative (Bronopol®) collected to the somatic cell count (SCC) that is used to mastitis monitoring. It were analyzed 38 bulk tank milk samples (BTMS) from 19 dairy herds artisanal cheese producers and 36 BTMS from a brucellosis free dairy herd, used as negative control (NC) and positive control (PC), when was added serum from vaccinated animals. The milk samples were collected in two different bottles (with or without Bronopol®). All the 38 BTMS from artisanal cheese producers (samples with and without Bronopol®) were non-reagent to MRT, as all NC. All PC were reagent to MRT, without interference of Bronopol®. Results showed that the milk sample used to SCC presented the proportion of agreement of results in all milk samples (100%), that is, false positive and false negative results were not observed. Results indicated that Bronopol® did not interfere with the color in the time of MRT reading suggesting that the same milk samples could be used to monitoring mastitis and brucellosis.


O objetivo deste estudo foi avaliar se o teste do anel do leite (TAL), utilizado para o diagnóstico da brucelose em nível de rebanho leiteiro, pode ser realizado em amostras de leite com conservador (Bronopol®) coletado para contagem de células somáticas (CCS) que é utilizado para monitoramento de mastite. Foram analisadas 38 amostras de leite de tanques de expansão (ALTE) de 19 rebanhos leiteiros produtores de queijo artesanal e 36 ALTE de rebanho leiteiro livre de brucelose, utilizadas como controle negativo (CN) e controle positivo (CP), quando foi adicionado soro de animais vacinados. As amostras de leite foram coletadas em dois frascos diferentes (com e sem Bronopol®). Todos as 38 ALTE de produtores artesanais de queijo (amostras com e sem Bronopol®) foram não reagentes ao TAL, como todos os CN. Todos os CP foram reagentes ao TAL, sem interferência do Bronopol®. Os resultados mostraram que a amostra de leite utilizada para CCS apresentaram a proporção de concordância dos resultados em todas as amostras de leite (100%), ou seja, não foram observados resultados falso-positivos e falso-negativos. Os resultados indicam que o Bronopol® não interferiu na cor no momento da leitura do TAL, sugerindo que as mesmas amostras de leite poderiam ser usadas para monitorar mastite e brucelose.

ABSTRACT: Allergic dogs are commonly sensitized to food allergens. We evaluated the use of the prick test to diagnose food allergies in dogs with pruritus and the efficacy of an exclusion diet based on the test. The prick test was performed in 10 healthy dogs and 34 dogs with pruritus, of which 25 received an exclusion diet for 60 days and was challenged with positive food on the test. pVAS and CADESI-4 were assessed on days 0, 30, and 60 after re-exposure. As a result, two control group dogs reacted to a single food allergen, milk and wheat. Of the 25 dogs with pruritus that reacted to food allergens, 24 (96%) reacted to more than one food allergen, and only one (4%) reacted to a single food protein: pork. In the test group (n=25), there was a significant improvement of pVAS and CADESI-4 after 30 and 60 days of dietary exclusion, with significant worsening of the scores with food allergen challenge. In conclusion, the prick test can be used for screening food allergens to make an exclusion diet.


RESUMO: Cães alérgicos são comumente sensibilizados a alérgenos alimentares. Nós avaliamos o uso do teste de puntura para o diagnóstico de alergias alimentares em cães com prurido e a eficácia de uma dieta de exclusão baseada no teste. O teste de puntura foi realizado em 10 cães saudáveis e em 34 cães com prurido, dos quais 25 receberam uma dieta de exclusão por 60 dias e depois desafiados com o alimento que foi positivo no teste. pVAS e CADESI-4 foram realizados nos dias 0, 30, 60 e depois da reexposição. Como resultados, dois cães do grupo controle reagiram a um único alérgeno alimentar, leite e trigo. Dos 25 cães com prurido que reagiram aos alérgenos alimentares, 24 (96%) reagiram a mais de um alérgeno alimentar e apenas um (4%) reagiu a uma única proteína, que foi porco. No grupo teste (n=25), houve uma significativa melhora do pVAS e CADESI-4 depois de 30 e 60 dias de dieta de exclusão, com significante piora dos scores com o desafio alimentar. Como conclusão, o teste de puntura pode ser usado para selecionar alérgenos alimentares para fazer uma dieta de exclusão.

The present study explored the predictive values of milk leukocyte differentials (MLD) as a basis for improving the diagnosis of intramammary infections (IMIs) and subclinical mastitis. Quarter milk samples were collected for bacteriological analysis, quarter somatic cell count (qSCC), and MLD. The MLD were assessed using the cytospin technique, direct microscopic smears, and flow cytometry. The predictive values of each single leukocyte population and useful potential indices that could better reflect immune complexity were also calculated. Changes in the percentage of any leukocyte alone failed to substantially improve the predictive value of qSCC in diagnosing IMIs. Although certain parameters increased the area under the receiver operating characteristic curve (ROC curve) as a result of increased specificity values, a slight reduction in sensitivity was observed. The so-called CD8 complex was a unique parameter which improved both the sensitivity (78.79 %) and the specificity (80.77 %) in IMI diagnosis, resulting in the highest area under the ROC curve (0.87). To diagnose subclinical mastitis, the percentage of macrophages and the sum of the percentage PMNLs and T CD8+ cells divided by the percentage of macrophages showed the highest predictive values (sensitivity = 79.63, specificity = 73.68, and area under the ROC curve = 0.83) in the differentiation of the inflammatory condition status of cows. In conclusion, this study provides further insights into using T CD8+ lymphocytes in diagnosing bovine IMIs, combined with PMNLs and macrophages. The antidromic trend of macrophages vs. PMNLs and T CD8+ lymphocytes due to the increasing qSCCs was crucial to differentiating quarters under both inflammatory and non-inflammatory conditions.

Sunflower (Helianthus annuus) plants showing symptoms of chlorosis, mosaic, chlorotic ringspot, and necrosis on younger leaves were found in a small experimental plot in Piracicaba, in the state of São Paulo, Brazil. Preliminary examinations by transmission electron microscopy of symptomatic leaf tissue revealed flexuous filamentous particles 13-15 nm wide and 700-750 nm long, and cytoplasmatic cylindrical inclusions typical of those found in plant cells infected by members of the Potyvirus genus. Total RNA extracted from symptomatic leaves and subjected to RT-PCR followed by partial nucleotide sequencing confirmed the presence of a potyvirus in the affected plants, which was identified as sunflower chlorotic mottle virus (SuCMoV), a member of the Sunflower chlorotic mottle virus (genus Potyvirus, family Potyviridae) species. Mechanical transmission assays with extracts of symptomatic sunflower leaves reproduced the original symptoms in sunflowers, mosaic symptoms in Zinnia elegans, and chlorotic local lesions in Chenopodium amaranticolor and C. quinoa. Sunflower and zinnia plants became infected after aphid transmission experiments with Myzus persicae. RT-PCR tests using specific primers for SuCMoV confirmed the presence of this virus in experimentally infected plants, meeting the criteria of Koch's postulate. This is the first report of SuCMoV infecting sunflower plants in Brazil.

Resumo

Among phytosanitary problems of the grapevine, viruses stand out for their capacity of reducing the quality and yield of grapes. However, detecting and identifying viral infections in grapevines can be challenging. This study performed a high throughput sequencing (HTS) of the viral pathogens present in a vine showing virus-like symptoms to elucidate the etiology. HTS analysis reported in a hybrid grapevine with mild curling down of leaf edges, the presence of four viruses and viroids, which were probably implicated in the observed symptoms. The determined complete genomes showed high genetic identities with previously characterized isolates of homologous pathogens.


Dentre os problemas fitossanitários da videira, os vírus se destacam pela capacidade de reduzir a qualidade e o rendimento da uva. No entanto, detectar e identificar infecções virais em videiras pode ser um desafio. O objetivo do estudo foi realizar um sequenciamento de alto rendimento (HTS) para determinar os patógenos virais presentes em uma videira com sintomas de virose e elucidar a etiologia. Com o HTS foi detectada, em uma videira híbrida com leve enrolamento dos bordos foliares, a presença de quatro vírus e viroides, os quais provavelmente estavam implicados com os sintomas observados. Os genomas completos determinados mostraram altas identidades genéticas com isolados previamente caracterizados de patógenos homólogos.

Bovine rabies is endemic in most Brazilian States, including Rio Grande do Sul (RS), which has faced an unprecedented rabies outbreak between 2011 and 2018. We described a real-time reverse transcription quantitative PCR (RT-rtPCR) for detection of rabies virus (RABV) in bovine samples. The primers were designed targeting a highly conserved region of the nucleoprotein (N) gene of RABV obtained from cattle. The detection limit corresponded to 13 DNA copies and the intra- and inter-run repeatability was adequate (CV<9%) in all dilutions tested. Amplification of other pathogens associated with neurological disease in cattle or cross-contamination was not observed. Brain samples from cattle suspicious of rabies (n=21) were tested in triplicate by the RT-rtPCR and by the gold-standard direct fluorescent antibody test (DFAT), resulting in 100% of sensitivity and specificity of the RT-rtPCR. Testing of additional 41 bovine brain samples submitted to the routine DFAT testing yielded 37 (90.2%) concordant results (30 positive/7 negative) and 4 (9.7%) inconclusive in DFAT and RT-rtPCR positive. These results showed a good concordance between the tests and a higher sensitivity of the RT-rtPCR. This assay represents an alternative for RABV detection, either as a confirmatory test or for large-scale diagnosis in endemic regions.


A raiva bovina é endêmica na maioria dos estados brasileiros, inclusive no Rio Grande do Sul (RS), que enfrentou um surto de raiva sem precedentes entre 2011 e 2018. Descrevemos um PCR quantitativo de transcrição reversa em tempo real (RT-rtPCR) para detecção do vírus da raiva (RABV) em bovinos. Os primers foram desenhados visando uma região altamente conservada do gene da nucleoproteína (N) de RABV obtido de bovinos. O limite de detecção correspondeu a 13 cópias de DNA e a repetibilidade intra e inter-ensaios foi adequada (CV <9%) em todas as diluições testadas. Não foi observada amplificação de outros patógenos associados a doenças neurológicas em bovinos ou contaminação cruzada. Amostras de cérebro de bovinos com suspeita de raiva (n = 21) foram testadas em triplicata no RT-rtPCR e pelo teste de anticorpo fluorescente padrão ouro (DFAT), resultando em 100% de sensibilidade e especificidade do RT-rtPCR. O teste de 41 amostras de cérebro bovino adicionais submetidas ao teste de DFAT de rotina rendeu 37 (90,2%) resultados concordantes (30 positivos / sete negativos) e quatro (9,7%) inconclusivos em DFAT e RT-rtPCR positivo. Esses resultados mostraram boa concordância entre os testes e maior sensibilidade do RT-rtPCR. Este ensaio representa uma alternativa para a detecção do vírus da raiva, seja como teste confirmatório ou para diagnóstico em larga escala em regiões endêmicas.

Resumo

In 2022, Salvador and co-authors published an up-to-date list of gastropod species recorded from Brazilian caves. That list included 18 species (15 land and three freshwater species) that were considered by the authors to be restricted to caves (troglobitic). In this contribution we argue that only six of the species listed by them as truly troglobites can be classified in that category. We disagree with some characteristics listed by Salvador and co-authors to diagnose the species they classify as cave-restricted and argue that those species do not have truly troglomorphic traits (characteristic of subterranean habitats). Since the Brazilian legislation considers the degree of association with caves as a decisive attribute to preserve them, it is important to clarify the characteristics are sufficient and necessary for a species to be classified as cave-dependent.

ABSTRACT: Leishmaniosis is a great public health problem affecting both humans and animals. The disease is caused by the protozoan Leishmania spp., which has a complex cycle involving a phlebotomine vector. The ELISA test (Enzyme-Linked Immunosorbent Assay) along with a chromatographic immunoassay was defined by the Brazil Health Ministry as the confirmatory screening protocol in 2011. Uruguaiana city is 630 km away from Porto Alegre, which makes it difficult to send samples and diagnose leishmaniasis, as well as receive quick results. In view of this, the present study evaluated an in-house indirect ELISA method compared to indirect immunofluorescence assay (IFA) and dual-path platform chromatographic immunoassay (DPP-BioManguinhos®) for the detection of an immune response to Leishmania spp. in canine species. The serological evaluation included 48 canines from the western border of Brazil (Uruguaiana and Barra do Quaraí city). Among the 48 canine samples tested, 18 were positive when using the ELISA technique, 19 were positive with IFA, and 17 were positive with rapid test DPP®. The ELISA technique showed a sensitivity/specificity of 83.3%/86.7% when compared to IFA and 100%/96.8% compared to DPP®. The present study showed a prevalence of 37.5%, demonstrating that the infection circulates in the studied population. It can be concluded that the ELISA technique was valuable for use in field conditions when performing screening tests in endemic areas.


RESUMO: A leishmaniose é um grande problema de saúde pública que afeta tanto humanos quanto animais. A doença é causada pelo protozoário Leishmania spp., que possui um ciclo complexo envolvendo um vetor flebotomíneo. O teste ELISA (Enzyme-Linked Immunosorbent Assay), juntamente com um imunoensaio cromatográfico, foi definido pelo Ministério da Saúde do Brasil como protocolo de triagem confirmatória em 2011. A cidade de Uruguaiana fica a 630 km de Porto Alegre, o que dificulta o envio de amostras e o diagnóstico da leishmaniose, além da demora para obter os resultados. Diante disso, o presente estudo teve como objetivo avaliar um método de ELISA indireto in-house comparado ao ensaio de imunofluorescência indireta (IFA) e imunoensaio cromatográfico de plataforma de dupla via (DPP-BioManguinhos®) para a detecção de uma resposta imune contra Leishmania spp. na espécie canina. A avaliação sorológica incluiu 48 caninos da fronteira oeste do Brasil (cidade de Uruguaiana e Barra do Quaraí). Das 48 amostras caninas testadas, 18 foram positivas na técnica de ELISA, 19 foram positivas com IFA e 17 foram positivas com o teste rápido DPP®. A técnica de ELISA apresentou sensibilidade/especificidade de 83,3%/86,7% quando comparada ao IFA e 100%/96,8% em relação ao DPP®. O presente estudo apresentou prevalência de 37,5%, demonstrando que a infecção circula na população estudada. Pode-se concluir que a técnica de ELISA foi valiosa para uso em condições de campo na realização de testes de triagem em áreas endêmicas.

Glyphosate is a herbicide commonly used in agriculture for weed control. Current agricultural production demands vast amounts of this product, which are applied by ground or aerial spraying. The concomitant aerial currents promote glyphosate drift to vegetated or urban areas. In this context, we hypothesized that the lichens, Parmotrema tinctorum and Usnea barbata, could be sensitive to the action of glyphosate and therefore be used to bio-indicate the presence of this herbicide in areas affected by drift. Since living organisms respond in different ways to the action of herbicides, our interest was also to indicate biological markers responsive to the action of glyphosate, through concentrations and exposure times of the thallus, besides identifying the most sensitive species. We evaluated the effect of different concentrations (0.0, 4.8, 9.6, and 19.2 mg L-1) and exposure times (24, 48, and 72 hours) to glyphosate on the morphoanatomy, photobiont vitality, photosynthetic efficiency, and oxidative metabolism of the thalli. We found that the lichens, P. tinctorum and U. barbata, respond to glyphosate stress, with prospects for use in the biomonitoring of pollutant dispersal from plantation areas. When using P. tinctorum as a bioindicator, lichen morphoanatomy, photobiont vitality, and photosynthetic pigment concentration were efficient biomarkers for the effect of concentration and exposure time. For U. barbata, the lichenic morphoanatomy and the activity of SOD and APX enzymes were essential tools to indicate the herbicide action. Parmotrema tinctotum, however, was characterized as more sensitive in bio-indicating the presence of this herbicide to diagnose the air quality in urban areas or vegetation sectors adjacent to agricultural environments.


O glifosato é um herbicida comumente utilizado na agricultura para o controle de ervas daninhas, contudo, a produção agrícola atual demanda quantidades gigantescas deste produto, que são aplicadas por pulverizações terrestres ou aéreas, que acompanhadas de correntes aéreas, promovem a deriva do glifosato para áreas vegetacionais ou urbanas. Neste contexto, nós levantamos a hipótese de que os líquens Parmotrema tinctorum e Usnea barbata pudessem ser sensíveis à ação do glifosato e, portanto, bioindicar a presença deste herbicida em áreas afetadas por deriva. Como os organismos vivos respondem de diferentes formas à ação dos herbicidas, nosso interesse foi também de indicar marcadores biológicos responsivos à ação do glifosato, por meio de concentrações e tempos de exposição dos talos, além de identificar a espécie mais sensível. Para isso, nós avaliamos o efeito de diferentes concentrações (0.0, 4.8, 9.6 e 19.2 mg L-1) e tempos de aexposição (24, 48 e 72 horas) ao glifosato, sobre a morfoanatomia, vitalidade do fotobionte, eficiência fotossintética e metabolismo oxidativo dos talos. Nós verificamos que os liquens P. tinctorum e U. barbata respondem ao estresse por glifosato, com perspectivas para serem utilizados no biomonitoramento da dispersão de poluentes a partir de áreas de plantio. Contudo, para o uso de P. tinctorum como bioindicador, a morfoanatomia liquênica, a vitalidade do fotobionte e a concentração de pigmentos fotossintéticos consistiram em biomarcadores eficientes para o efeito de concentração e tempo de exposição. Para U. barbata, a morfoanatomia liquênica, bem como a atividade das enzimas SOD e APX constituíram ferramentas importantes para evidenciar a ação do herbicida. A espécie P. tinctotum, contudo, foi caracterizada como mais sensível, sendo indicada para bioindicar a presença deste herbicida e para diagnosticar a qualidade do ar em áreas urbanas ou fragmentos vegetacionais imersos em matrizes agrícolas.

Background: Thermography is a noninvasive, non-contact, painless, and non-ionizing imaging technique that records cutaneous thermal patterns generated by infrared emission of the surface. The surface heat is closely related to dermal microcirculation. Thromboembolism is responsible for important changes in the thermal pattern of the body surface due to physical obstruction of blood flow, being the main complication in immune-mediated hemolytic anemia. The aim of this paper is to report a dog with thrombus in his left forelimb secondary to idiopathic hemolytic anemia, whose diagnostic screening was performed through infrared thermography. Case: A 9-year-old mixed breed bitch was referred to a veterinary hospital with a history of emesis, diarrhea and dark-colored urine for 2 days. The complete blood count showed hypochromic macrocytic anemia (hematocrit [HTC] 28%, reference: 37 to 55%) with the presence of nucleated erythrocyte (14/100 leukocytes, reference: 0 to 5/100 leukocytes), polychromasia and spherocytes. Leukocytosis (28,300 mm³, reference: 6,000 to 17,000 mm³) by neutrophilia with left deviation and toxic granulations was also present, in addition to hemoglobinuria at urine exam. Therefore, treatment for immune-mediated hemolytic anemia (IMHA) was instituted. After 2 days, the animal returned with acute functional impairment of the left forelimb. Physical examination revealed that the limb was cold, without pulse, proprioception, reflexes, and deep pain. New blood analyses revealed decreased hematocrit (HTC 17%, reference: 37 to 55%), and increased total leukocyte number (57,000 mm3, reference: 6,000 to 17,000 mm³). Infrared thermography revealed an important temperature difference between the limbs, with the affected limb temperature considerably lower (31.3ºC) when compared to the contralateral limb (35.0ºC). Thermography showed the site of the thrombus in the medial portion of the limb (cephalic vein), where the catheter had been placed for fluid therapy. Due to the severity of the condition, the bitch was submitted to amputation surgery, which occurred without complications. The patient had a good response to treatment, with decreased signs of hemolysis and hypercoagulability. The medications were slowly withdrawn, and the clinical discharge occurred after 4 weeks. Discussion: In humans, thermography has been widely used in the assessment of thrombotic diseases, contributing to diagnosis, localization, and prognosis. In veterinary medicine, however, the use of this tool in the diagnosis of thromboembolism is still rare. The difference of 3.7°C between the affected and contralateral limb was objectively verified using thermography. A minimum difference of 2.4°C between limbs has high sensitivity and specificity for diagnosing thromboembolism and occurs due to the reduction in local blood flow. In the present case this tool was essential for the anatomical location of the thrombus, which was in the middle third of the forearm, and allowed an adequate surgical planning. It is known that the main complication of IMHA is thromboembolism. Its predisposing factors include venous stasis, endothelial damage, and hypercoagulability, being exacerbated by cage confinement, decubitus and presence of a peripheral venous catheter. The reported case corroborates at least one of these factors since it had a peripheral venous catheter in the left forelimb, which later showed absence of pulse, spinal reflexes, pain and proprioception. The thermography showed to be an objective, rapid and non-invasive tool to diagnose and precisely locate the thrombus, which allowed for adequate treatment and surgical planning for the case. To the best of our knowledge, this is the first report about use of thermography to diagnose thromboembolism secondary to immune-mediated hemolytic anemia in a dog.

A high frequency of feline leishmaniasis has been reported in several countries. However, much information about disease progression in cats still needs to be clarified. This study aimed to verify the occurrence of clinicopathological changes in cats infected with Leishmania infantum. A total of 60 cats were divided into three groups of 20 animals each: control, suspects, and infected. All 60 cats underwent blood count and biochemical analyses. Serum samples from 20 animals with leishmaniasis were also used to diagnose feline immunodeficiency virus and feline leukemia virus. A total of five of the infected animals underwent necropsy for a histopathological study. The main clinical findings in cats with leishmaniasis were lymphadenomegaly (65%), alopecia (55%), ulcerative skin lesions and weight loss (40%), skin nodules (25%), a significant reduction in red blood cells (p=0.0005) and hematocrit (p=0.0007), hyperplasia in spleen 4/5(80%), presence of Leishmania in the spleen 2/5(40%), hepatitis 3/5(60%), liver degeneration 4/5(80%) and inflammatory nephropathy 3/5(60%). It was concluded that cats with leishmaniasis presented significant clinical, hematological, and histopathological alterations compatible with L. infantum infection. The observation of lymphadenomegaly, weight loss, skin lesions and low concentration of red blood cells, contributes significantly to the diagnosis and analysis of progression of feline leishmaniasis.(AU)


Uma alta frequência de leishmaniose felina tem sido relatada em vários países. No entanto, muitas informações sobre a progressão da doença em gatos ainda precisam ser esclarecidas. Este estudo teve como objetivo verificar a ocorrência de alterações clinicopatológicas em gatos infectados por Leishmania infantum. Um total de 60 gatos foi dividido em três grupos de 20 animais cada: controle, suspeitos e infectados. Todos os 60 gatos foram submetidos à hemograma e testes bioquímicos. Amostras de soro de 20 animais com leishmaniose também foram utilizadas para diagnosticar o vírus da imunodeficiência felina e o vírus da leucemia felina. Um total de cinco dos animais infectados foram necropsiados para estudo histopatológico. Os principais achados clínicos nos gatos com leishmaniose foram linfadenomegalia (65%), alopecia (55%), lesões ulcerativas na pele e emagrecimento (40%), nódulos cutâneos (25%), redução significativa de hemácias (p=0,0005) e hematócrito (p=0,0007), hiperplasia no baço 4/5(80%), presença de Leishmania no baço 2/5(40%), hepatite 3/5(60%), degeneração hepática 4/5(80%) e nefropatia inflamatória 3/5(60%). Concluiu-se que gatos com leishmaniose apresentaram alterações clínicas, hematológicas e histopatológicas significativas compatíveis com infecção por L. infantum. A observação de linfadenomegalia, emagrecimento, lesões cutâneas e baixa concentração de hemácias, contribui significativamente para o diagnóstico e análise da progressão da leishmaniose felina.(AU)

A produção de potros é o objetivo final de todas as atividades voltadas para a reprodução equina. Para tanto, o diagnóstico de anormalidades gestacionais deve ser realizado precocemente para que a instalação da terapêutica adequada seja garantida, visando manter a sobrevivência da égua e o nascimento de indivíduos vivos e saudáveis. Aqui encontram-se descritos os principais achados relacionados às anormalidades gestacionais de éguas mais frequentemente encontradas em atendimentos realizados a campo e nas rotinas hospitalares. São elas: gestação gemelar, placentite, separação prematura da placenta, torção uterina, hidropsias e ruptura de tendão pré-púbico. O objetivo é caracterizar os sinais clínicos, métodos diagnósticos, tratamento e prognóstico dessas enfermidades. A compreensão desses aspectos é essencial para o desenvolvimento de estratégias de prevenção e gestão das intercorrências obstétricas, visando reduzir seu impacto na criação de equinos.(AU)


The primary objective of equine reproduction is to produce healthy foals. To achieve this, it is crucial to diagnose gestational abnormalities at an early stage and administer appropriate therapy. This will increase the chances of survival for the mare and the birth of live foals. Here we outlines the most frequently observed gestational abnormalities in mares during field visits and hospital routines. The following abnormalities discussed are: twin pregnancy, placenta problems, premature separation of the placenta, uterine torsion, hydrops, and rupture of the prepubic tendon. The objective is to characterize the clinical signs, diagnostic methods, treatment, and prognosis for each of these conditions. It is essential to understand these aspects of gestational abnormalities to develop effective prevention and management strategies for obstetric complications. aiming to reduce their impact on equine breeding.(AU)

This study aimed to investigate the epidemiological and pathological findings of type 1 and 2 abomasal ulcers in cattle with different primary comorbidities. A total of 201 animals; 40/201 (20%) were young cattle under the age of two years and 161/201 (80%) were adult cattle over the age of two years, which were hospitalized for clinical care 152/201 (75,62%), 19/201 (9,45%) obstetric care, 17/201 (8,46%) surgical care and 13/201 (6,47%) for anatomopathological diagnosis, being euthanized or had natural death. The diagnosis of ulcers was based on the result of the post-mortem examination (macroscopic and histopathological analysis). Histopathological examination was performed on 201 fragments of ulcers and classified as type 1 or type 2. Of these, 193/201 (96.01%) corresponded to type 1 ulcers, of which 12/193 (5.97%) corresponded to subtype 1a lesions, 101/193 (50.25%) to subtype 1b, 77/ 193 (38.31%) to subtype 1c, 03/193 (1.49%) to subtype 1d, while 08/201 (3.98%) were type 2 ulcers. The ulcers were characterized by a focal, focally extensive, multifocal or diffuse inflammatory process, mainly by mononuclear cells. Abomasitis associated with ulcerated mucosa was found in 160/201 (79.60%). In 26/201 (12.93%) the abomasitis had diffuse foci of multifocal lymphocytic proliferation by atypical lymphocytes. Digestive and reproductive comorbidities were seen more frequently in cattle with type 1 or type 2 ulcers. The Subtype 1b focal ulcers and subtype 1a and 1b multifocal ulcers were more prevalent. In addition to the presence of comorbidities, most cases occur in the dry period, associated with feeding with higher amounts of concentrates and silages.(AU)


Objetivou-se estudar os achados epidemiológico e anatomopatológico de úlceras do abomaso tipo 1 e 2 em bovinos com diferentes comorbidades primárias. Um total de 201 animais; 40/201 (20%) eram bovinos jovens com idade inferior a dois anos e 161/201 (80%) eram bovinos adultos com idade superior a dois anos, os quais foram internados para atendimento clínico 152/201 (75,62%), 19/201 (9,45%) obstétrico, 17/201 (8,46%) para atendimento clínico-cirúrgico e 13/201 (6,47%) para diagnóstico anatomopatológico, sendo eutanasiados ou tiveram morte natural. O diagnóstico das úlceras foi baseado no exame post-mortem (análise macroscópica e histopatológica). O exame histopatológico foi realizado em 201 fragmentos de úlceras e classificado como tipo 1 ou do tipo 2. Destes, 193/201 (96,01%) corresponderam a úlceras tipo 1, das quais, 12/193 (5,97%) corresponderam a lesões subtipo 1a, 101/193 (50,25%) a subtipo 1b, 77/193 (38,31%) a subtipo 1c, 03/193 (1,49%) ao subtipo 1d, enquanto 08/201 (3,98%) foram úlceras tipo 2. As úlceras foram caracterizadas por processo inflamatório focal, focalmente extenso, multifocais ou difusos, principalmente por células mononucleares. Abomasite associada à mucosa ulcerada foi encontrada em 160/201 (79,60%). Em 26/201 (12,93%) a abomasite apresentava focos difusos de proliferação linfocítica multifocal por linfócitos atípicos. As comorbidades digestivas e reprodutivas foram observadas com maior frequência em bovinos com úlceras tipo 1 ou tipo 2. As úlceras focais subtipo 1b e úlceras multifocais subtipo 1a e 1b foram mais prevalentes. Além da presença de comorbidades, a maioria dos casos ocorrerem no período seco, associados à alimentação com maiores aportes de concentrados e silagens.(AU)

Leishmaniosis is a great public health problem affecting both humans and animals. The disease is caused by the protozoan Leishmania spp., which has a complex cycle involving a phlebotomine vector. The ELISA test (Enzyme-Linked Immunosorbent Assay) along with a chromatographic immunoassay was defined by the Brazil Health Ministry as the confirmatory screening protocol in 2011. Uruguaiana city is 630 km away from Porto Alegre, which makes it difficult to send samples and diagnose leishmaniasis, as well as receive quick results. In view of this, the present study evaluated an in-house indirect ELISA method compared to indirect immunofluorescence assay (IFA) and dual-path platform chromatographic immunoassay (DPP-BioManguinhos®) for the detection of an immune response to Leishmania spp. in canine species. The serological evaluation included 48 canines from the western border of Brazil (Uruguaiana and Barra do Quaraí city). Among the 48 canine samples tested, 18 were positive when using the ELISA technique, 19 were positive with IFA, and 17 were positive with rapid test DPP®. The ELISA technique showed a sensitivity/specificity of 83.3%/86.7% when compared to IFA and 100%/96.8% compared to DPP®. The present study showed a prevalence of 37.5%, demonstrating that the infection circulates in the studied population. It can be concluded that the ELISA technique was valuable for use in field conditions when performing screening tests in endemic areas.


A leishmaniose é um grande problema de saúde pública que afeta tanto humanos quanto animais. A doença é causada pelo protozoário Leishmania spp., que possui um ciclo complexo envolvendo um vetor flebotomíneo. O teste ELISA (Enzyme-Linked Immunosorbent Assay), juntamente com um imunoensaio cromatográfico, foi definido pelo Ministério da Saúde do Brasil como protocolo de triagem confirmatória em 2011. A cidade de Uruguaiana fica a 630 km de Porto Alegre, o que dificulta o envio de amostras e o diagnóstico da leishmaniose, além da demora para obter os resultados. Diante disso, o presente estudo teve como objetivo avaliar um método de ELISA indireto in-house comparado ao ensaio de imunofluorescência indireta (IFA) e imunoensaio cromatográfico de plataforma de dupla via (DPP-BioManguinhos®) para a detecção de uma resposta imune contra Leishmania spp. na espécie canina. A avaliação sorológica incluiu 48 caninos da fronteira oeste do Brasil (cidade de Uruguaiana e Barra do Quaraí). Das 48 amostras caninas testadas, 18 foram positivas na técnica de ELISA, 19 foram positivas com IFA e 17 foram positivas com o teste rápido DPP®. A técnica de ELISA apresentou sensibilidade/especificidade de 83,3%/86,7% quando comparada ao IFA e 100%/96,8% em relação ao DPP®. O presente estudo apresentou prevalência de 37,5%, demonstrando que a infecção circula na população estudada. Pode-se concluir que a técnica de ELISA foi valiosa para uso em condições de campo na realização de testes de triagem em áreas endêmicas.

Relatos de ingestão de corpos estranhos dos mais diferentes materiais são comuns em aves, especialmente nas mais jovens. Os corpos estranhos podem causar intoxicação (dependendo da composição do material) ou mesmo perfuração do canal alimentar ou obstrução gastrintestinal. Quando há suspeita de ingestão de corpos estranhos, exames de imagem como a radiologia, ultrassonografia e endoscopia são ferramentas valiosas para o diagnóstico. Objetivou-se relatar um caso de uma ave da espécie Gallus gallus domesticus, raça Brahma, sete meses de idade, macho, pesando 4,3 Kg com quadro de sensibilidade na cavidade corporal e histórico de regurgitação, hiporexia e prostração há cinco dias. As radiografias simples indicaram a presença de corpo estranho radiopaco (parafuso) alojado na região do ventrículo (moela). Após tratamento clínico com lavagens gástricas por cinco dias sem êxito, optou-se por realizar uma endoscopia digestiva alta sob anestesia geral com quetamina e isoflurano. O corpo estranho foi satisfatoriamente removido com pinça de alça de polipectomia e a ave apresentou rápida melhora clínica sem complicações. A endoscopia mostrou-se um procedimento pouco invasivo e eficaz para a resolução do presente caso. Os clínicos veterinários de aves devem considerar a possibilidade de usar a endoscopia como ferramenta para diagnóstico e resolução de corpos estranhos no canal alimentar das aves.(AU)


Reports of ingestion of foreign bodies from most different materials are common in birds, especially younger ones. Foreign bodies can cause intoxication (depending on the composition of the material) or even perforation of the alimentary canal and gastrointestinal obstruction. When foreign body ingestion is suspected, imaging tests such as radiology, ultrasound, and endoscopy are valuable diagnostic tools. The objective of this paper was to report a case of an avian of the species Gallus gallus domesticus, Brahma breed, seven months old, male, weighing 4.3 kg, with sensitivity in the body cavity, and a history of regurgitation, hyporexia, and prostration for five days. Plain radiographs indicated the presence of a radiopaque foreign body (screw) lodged in the ventricle region (gizzard). After clinical treatment with gastric lavages for five days without success, it was decided to perform an upper digestive endoscopy under general anesthesia with ketamine and isoflurane. The foreign body was satisfactorily removed with polypectomy loop forceps, and the bird showed rapid clinical improvement without complications. Endoscopy proved to be a minimally invasive and effective procedure for resolving the present case. Avian veterinary practitioners may consider using endoscopy to diagnose and resolve foreign bodies in the alimentary canal of birds.(AU)


Los informes de ingestión de cuerpos extraños de los más diferentes materiales son comunes en las aves, especialmente en las más jóvenes. Los cuerpos extraños pueden causar intoxicación (dependiendo de la composición del material) o incluso perforación del tubo digestivo u obstrucción gastrointestinal. Cuando se sospecha la ingestión de un cuerpo extraño, las pruebas de imagen como la radiología, la ecografía y la endoscopia son valiosas herramientas diagnósticas. El objetivo fue reportar un caso de un ave de la especie Gallus gallus domesticus, raza Brahma, de siete meses de edad, macho, con un peso de 4,3 kg, con sensibilidad en la cavidad corporal y antecedentes de regurgitación, hiporexia y postración de cinco días de evolución. Las radiografías simples indicaron la presencia de un cuerpo extraño radiopaco (tornillo) alojado en la región del ventrículo (molleja). Tras tratamiento clínico con lavados gástricos durante cinco días sin éxito, se decide realizar endoscopia digestiva alta bajo anestesia general con ketamina e isoflurano. El cuerpo extraño se extrajo satisfactoriamente con pinzas de asa de polipectomía y el ave mostró una rápida mejoría clínica sin complicaciones. La endoscopia demostró ser un procedimiento mínimamente invasivo y efectivo para la resolución del presente caso. Los veterinarios aviares deberían considerar la posibilidad de utilizar la endoscopia como herramienta para el diagnóstico y resolución de cuerpos extraños en el tubo digestivo de las aves.(AU)

Tuberculosis is a communicable disease with high morbidity and mortality rates in developing countries. The study's primary objective is to compare conventional methods such as acid-fast bacillus (AFB) culture and microscopy with rapid diagnostic methods. The secondary objective is to compare histopathological and microbiological findings in suspected patients with tubercular lymphadenitis. A total of 111 samples (August 2018 to September 2019) of lymph nodes were processed for AFB microscopy, AFB cultures, drug-susceptibility testing (DST), histopathology, and Xpert Mycobacterium Tuberculosis (MTB)/resistance to Rifampin (RIF) assays. Out of 111 lymph node samples, 6 (5.4%) were positive for AFB smear microscopy, 84 (75.6%) were positive for AFB culture, 80 (70.7%) were positive on Gene Xpert, and 102 (91.8%) were indicative of tuberculosis for histopathology studies. Mycobacteria growth indicator tube (MGIT) culture positivity was 84 (75.6%) higher than solid Lowenstein-Jensen (LJ) culture 74 (66.6%). Positive cultures underwent phenotypic DST. Two cases were Multidrug-resistant (MDR) on DST, while three cases were Rifampicin resistant on Gene Xpert. The sensitivity of Genexpert was (62%) against the conventional AFB culture method. The poor performance of conventional lymphadenitis diagnostic methods requires early and accurate diagnostic methodology. Xpert MTB/RIF test can help in the treatment of multidrug-resistant TB cases. Nonetheless, rapid and conventional methods should be used for complete isolation of Mycobacterium tuberculosis.


A tuberculose é uma doença transmissível com altas taxas de morbimortalidade nos países em desenvolvimento. O objetivo principal do estudo é comparar métodos convencionais, como cultura de bacilo álcool-ácido resistente (BAAR) e microscopia, com métodos de diagnóstico rápido. O objetivo secundário é comparar os achados histopatológicos e microbiológicos em pacientes com suspeita de linfadenite tubercular. Um total de 111 amostras (agosto de 2018 a setembro de 2019) de gânglios linfáticos foi processado para microscopia de AFB, culturas de AFB, teste de susceptibilidade a drogas (DST), histopatologia e Xpert Mycobacterium tuberculosis (MTB)/ensaios de resistência à rifampicina (RIF). Das 111 amostras de linfonodos, 6 (5,4%) foram positivas para baciloscopia de AFB, 84 (75,6%) foram positivas para cultura de AFB, 80 (70,7%) foram positivas para o GeneXpert e 102 (91,8%) foram indicativas de tuberculose para estudos histopatológicos. A positividade da cultura do tubo indicador de crescimento de micobactérias (MGIT) foi 84 (75,6%), maior que a cultura sólida de Lowenstein-Jensen (LJ), 74 (66,6%). As culturas positivas foram submetidas a DST fenotípico. Dois casos eram multirresistentes (MDR) ao DST, enquanto três casos eram resistentes à rifampicina no GeneXpert. A sensibilidade do GeneXpert foi 62% contra o método convencional de cultura AFB. O fraco desempenho dos métodos convencionais de diagnóstico de linfadenite requer metodologia de diagnóstico precoce e precisa. O teste Xpert MTB/RIF pode ajudar no tratamento de casos de tuberculose multirresistente. No entanto, métodos rápidos e convencionais devem ser usados para o isolamento completo do Mycobacterium tuberculosis.

Advancements in assisted reproduction (AR) methodologies have allowed significant improvements in live birth rates of women who otherwise would not be able to conceive. One of the tools that allowed this improvement is the possibility of embryo selection based on genetic status, performed via preimplantation genetic testing (PGT). Even though the widespread use of PGT from TE biopsy helped to decrease the interval from the beginning of the AR intervention to pregnancy, especially in older patients, in AR, there are still many concerns about the application of this invasive methodology in all cycles. Therefore, recently, researchers started to study the use of cell free DNA (cfDNA) released by the blastocyst in its culture medium to perform PGT, in a method called non-invasive PGT (niPGT). The development of a niPGT would bring the diagnostics power of conventional PGT, but with the advantage of being potentially less harmful to the embryo. Its implementation in clinical practice, however, is under heavy discussion since there are many unknowns about the technique, such as the origin of the cfDNA or if this genetic material is a true representative of the actual ploidy status of the embryo. Available data indicates that there is high correspondence between results observed in TE biopsies and the ones observed from cfDNA, but these results are still contradictory and highly debatable. In the present review, the advantages and disadvantages of niPGT are presented and discussed in relation to tradition TE biopsy-based PGT. Furthermore, there are also presented some other possible non-invasive tools that could be applied in the selection of the best embryo, such as quantification of other molecules as quality biomarkers, or the use artificial intelligence (AI) to identify the best embryos based on morphological and/or morphokitetic parameters.(AU)

Background: Persistent hyperplastic primary vitreous is a rare ocular condition that is usually unilateral, formed when normal regression of the hyaloid vascular system does not occur. Diagnosis is possible through ultrasonography, by obtaining images that provide information and also serve as a differential diagnosis. Clinically the condition presents with signs of leukocoria, microphthalmia, and cataracts, and it can be further classified into 6 degrees according to the evolution. The objective of this work is to describe the case and treatment of a canine of the Pointer breed that presented opacity in both eyes, with diagnosis confirmed through ocular ultrasonography. Case: A 6-year-old male pointer dog, weighing 27 kg, was referred to the Veterinary Medical Teaching Hospital of the Federal University of Santa Maria (UFRGS), Santa Maria, RS - Brazil with a history of blindness. According to the owner, the animal's right eye has always been "white", since it was a puppy and the left eye began to present the same characteristic a short time ago. On ophthalmological examination, neither eye showed signs of ocular discomfort, the Schirmer's tear test was within the range expected for the species, fluorescein and threat tests were negative, and a positive pupillary reflex was observed in the left eye. The right eye had total corneal opacity associated with a chronic lesion and the presence of pigment cells in the medial portion of the eyeball, the opacity making it impossible to visualize the other internal structures of the eyeball. The left eye presented lens opacity. In the ultrasound of the right eye, there were several alterations, the presence of a triangular-shaped hyperechogenic structure in the vitreous chamber, a decrease in the depth of the anterior chamber, and the presence of abnormality in the development of the lens. The findings are compatible with and confirmed the diagnosis of PHPV. In the left eye, the ultrasound images indicated hyperechogenicity of the posterior and anterior poles of the lens, findings compatible with cataracts. Through the animal's history, complete ophthalmic examination, and use of ocular ultrasonography, it was possible to diagnose and differentiate the causes of the white eye in the patient. The preoperative screening was continued with an electroretinography examination, which showed a satisfactory electrical response for the retina of the left eye and an unsatisfactory electrical response for the retina of the right eye. Phacoemulsification surgery was performed to remove the cataract in the left eye with intraocular lens implantation. The positive threat reflex returned in this eye, with a return of visual acuity. Discussion: Persistent hyperplastic tunica lentis and persistent hyperplastic primary vitreous (PHTVL/PHPV) are congenital anomalies caused by a failure to regress the fetal hyaloid vasculature. These conditions are not common, but occur sporadically in dogs. The PHPV classification can be classified into degrees, based on the morphological aspect of the lesion. Following this classification scheme, the PHPV of the animal in question refers to the 6th degree of evolution. The total opacity of the cornea in the right eye was associated with the presence of a hyperplastic primary vitreous and the leukocoria in the left eye was due to the presence of a mature cataract. The conclusion of the diagnosis was only possible through ocular ultrasonography, which is very useful to differentiate the causes of ocular opacity and leukocoria, in addition to allowing complete evaluation of the intraocular structures and being considered an essential exam to confirm the diagnosis of PHPV.

Resumo

Tuberculosis is a communicable disease with high morbidity and mortality rates in developing countries. The study's primary objective is to compare conventional methods such as acid-fast bacillus (AFB) culture and microscopy with rapid diagnostic methods. The secondary objective is to compare histopathological and microbiological findings in suspected patients with tubercular lymphadenitis. A total of 111 samples (August 2018 to September 2019) of lymph nodes were processed for AFB microscopy, AFB cultures, drug-susceptibility testing (DST), histopathology, and Xpert Mycobacterium Tuberculosis (MTB)/resistance to Rifampin (RIF) assays. Out of 111 lymph node samples, 6 (5.4%) were positive for AFB smear microscopy, 84 (75.6%) were positive for AFB culture, 80 (70.7%) were positive on Gene Xpert, and 102 (91.8%) were indicative of tuberculosis for histopathology studies. Mycobacteria growth indicator tube (MGIT) culture positivity was 84 (75.6%) higher than solid Lowenstein-Jensen (LJ) culture 74 (66.6%). Positive cultures underwent phenotypic DST. Two cases were Multidrug-resistant (MDR) on DST, while three cases were Rifampicin resistant on Gene Xpert. The sensitivity of Genexpert was (62%) against the conventional AFB culture method. The poor performance of conventional lymphadenitis diagnostic methods requires early and accurate diagnostic methodology. Xpert MTB/RIF test can help in the treatment of multidrug-resistant TB cases. Nonetheless, rapid and conventional methods should be used for complete isolation of Mycobacterium tuberculosis.(AU)

O objetivo principal do estudo é comparar métodos convencionais, como cultura de bacilo álcool-ácido resistente (BAAR) e microscopia, com métodos de diagnóstico rápido. O objetivo secundário é comparar os achados histopatológicos e microbiológicos em pacientes com suspeita de linfadenite tubercular. Um total de 111 amostras (agosto de 2018 a setembro de 2019) de gânglios linfáticos foi processado para microscopia de AFB, culturas de AFB, teste de susceptibilidade a drogas (DST), histopatologia e Xpert Mycobacterium tuberculosis (MTB)/ensaios de resistência à rifampicina (RIF). Das 111 amostras de linfonodos, 6 (5,4%) foram positivas para baciloscopia de AFB, 84 (75,6%) foram positivas para cultura de AFB, 80 (70,7%) foram positivas para o GeneXpert e 102 (91,8%) foram indicativas de tuberculose para estudos histopatológicos. A positividade da cultura do tubo indicador de crescimento de micobactérias (MGIT) foi 84 (75,6%), maior que a cultura sólida de Lowenstein-Jensen (LJ), 74 (66,6%). As culturas positivas foram submetidas a DST fenotípico. Dois casos eram multirresistentes (MDR) ao DST, enquanto três casos eram resistentes à rifampicina no GeneXpert. A sensibilidade do GeneXpert foi 62% contra o método convencional de cultura AFB. O fraco desempenho dos métodos convencionais de diagnóstico de linfadenite requer metodologia de diagnóstico precoce e precisa. O teste Xpert MTB/RIF pode ajudar no tratamento de casos de tuberculose multirresistente. No entanto, métodos rápidos e convencionais devem ser usados para o isolamento completo do Mycobacterium tuberculosis.(AU)
