Resumo

Atrazine and nitrate have been shown to act as potent oxidative stressors in amphibians either alone or in combination under stable laboratory conditions, causing histopathological alternations in liver and kidney structures at the sub-lethal concentrations. A control group and three treatments groups were tested; atrazine, nitrate, atrazine-nitrate treatments with doses of 300 µg L-1, 200 mg L-1 and their combination respectively. Sever distortion in liver and kidney tissues were shown related to the different treatments. The most hepatic lesions were observed depletion in glycogen content, degeneration of hepatocytes, hemorrhage, necrosis, vasodilatation, congestion in blood vessels, cloudy swelling in the hepatocytes and aggregation of melanomacrophage cells in between the hepatocytes that increased in combination treatment group. In kidney, the most lesions were represented in degeneration of renal tubules, fibrosis, hemorrhage, leucocytes infiltration, thickness in the wall of the renal capsule, atrophy of glomerulus, deformation of Bowman's epithelium. These negative impacts may be a bioindicator alarming the ecosystem disrupting caused by the uncontrolled apply of these chemicals in agriculture.(AU)

Monensin is an ionophore antibiotic (IA) widely used for growth promotion and weight gain in the production of ruminants. However, it has caused intoxication in several species, including buffaloes, mainly because of the ignorance or disrespect of the recommendations for use in each animal species. The objective of this study was to describe, for the first time, clinical-epidemiological and anatomopathological data of an outbreak of accidental poisoning by monensin in buffalos and rediscuss the recommendation of the use of IA in the production of this species. The outbreak affected 21 adult buffaloes after consumption of remains from a feed formulated on the farm and whose constituents were mixed by hand. Clinical and first death signs were observed 24 hours after ingestion of this food. In general, the clinical picture was characterized by muscle weakness, tremors, difficulty in locomotion, and decubitus. Fifteen buffaloes presented clinical signs of poisoning (71.5% morbidity), followed by death (100% lethality), after acute to subacute evolution (<24h to 96h). Laboratory tests indicated elevated serum activity of creatine phosphokinase and aspartate aminotransferase enzymes. Three buffaloes underwent necropsy, and samples from several organs were collected for histopathological examination. The main injuries found were hyaline degeneration and multifocal segmental necrosis in the skeletal and cardiac striated muscles (myopathy and degenerative-necrotic multifocal multifocal-necrotic cardiopathy). The diagnosis was confirmed by the toxicological evaluation of suspected ration remains, which detected 461.67mg/kg of monensin. The death of 71.5% buffaloes in this lot occurred due to a succession of errors, which included faults in the formulation of the ration and, above all, due to the use of monensin in a highly sensitive species. Despite the possible beneficial effects of IA use as a dietary supplement for buffaloes, we are of the opinion that IAs should never be used in bubalinoculture since any increment in production does not compensate for the imminent risk of death due to a small safety margin for this species and the absence of antidotes.


Monensina é um antibiótico ionóforo (AI) amplamente empregado na produção de ruminantes para promoção de crescimento e ganho de peso, mas que tem causado intoxicação em diversas espécies, incluindo os búfalos, principalmente, pelo desconhecimento ou desrespeito das recomendações de uso e às particularidades de cada espécie animal. Objetivou-se descrever, pela primeira vez na Bahia, dados clínico-epidemiológicos e anatomopatológicos de um surto de intoxicação acidental por monensina em búfalos e rediscutir a recomendação do uso de AI na produção de bubalinos. O surto acometeu um lote de 21 búfalos adultos após consumo de sobras de uma ração para bovinos formulada na fazenda e cujos constituintes eram misturados à mão. Os sinais clínicos e primeiros óbitos foram observados 24 horas após a ingestão dessa ração. O quadro clínico, em geral, se caracterizou por fraqueza muscular, tremores, dificuldade de locomoção e decúbito. Quinze búfalos apresentaram sinais clínicos de intoxicação (morbidade 71,5%), seguido de morte (letalidade 100%), após evolução aguda a subaguda (<24h até 96h). Exames laboratoriais indicaram acentuada elevação na atividade sérica das enzimas CPK e AST. Três búfalos foram necropsiados, sendo coletadas amostras de diversos órgãos para exame histopatológico. A principal lesão encontrada foi degeneração hialina e necrose segmentar multifocal nos músculos estriados esqueléticos e cardíacos (miopatia e cardiopatia degenerativo-necrótica tóxica multifocal polifásica). O diagnóstico foi confirmado pela avaliação toxicológica das sobras da ração suspeita, que detectou 461,67mg/kg de monensina. A morte de 71,5% dos búfalos deste lote ocorreu devido a uma sucessão de erros, que incluíram falhas na formulação da ração e, sobretudo, devido ao uso da monensina em uma espécie altamente sensível. Enfatizamos que, apesar dos possíveis efeitos benéficos do uso AIs como suplemento dietético para bubalinos, somos da opinião que os AIs nunca devem ser empregados na bubalinocultura, uma vez que os eventuais incrementos na produção não compensam o risco iminente de morte, devido a pequena margem de segurança para essa espécie e a inexistência de antídotos.

Resumo

This study describes an outbreak of acute necrotic hepatopathy associated with spontaneous poisoning by Lantana camara L. in dairy cattle. A herd of 15 cows and heifers was introduced into a native pasture with limited food supply, and, sixteen days later, eight animals had spontaneous nasal hemorrhage, fever, lethargy, jaundice, and dry, dark stools with mucus and blood. The clinical course varied from two to five days. In the pasture where the cattle were kept, abundant adult specimens of L. camara L. with evident signs of consumption were observed. In total, seven cattle died and necropsy was performed in three of them. All animals had moderate jaundice, hemorrhage in the subcutaneous tissue and on the surface of different organs. The liver was slightly enlarged, with orange discoloration and enhanced lobular pattern. Histologically, multifocal areas of coagulative necrosis of hepatocytes in the centrilobular area, occasionally extending to the midzonal area, were observed, as well as marked hepatocellular degeneration and prominent cholestasis. The current study suggests that L. camara L. poisoning should be considered a differential diagnosis of acute and necrotic hepatotoxicity in cattle, despite the absence of photosensitization.(AU)


Esse estudo descreve um surto de hepatopatia necrótica aguda associada a intoxicação espontânea por Lantana camara L. em bovinos leiteiros. Um lote de 15 vacas e novilhas foram introduzidas em um piquete com campo nativo, com escassa oferta de alimento. Após dezesseis dias, oito animais manifestaram epistaxe, febre, apatia, icterícia, fezes ressecadas e escuras com muco e sangue. A evolução do quadro clínico variou de dois a cinco dias. No piquete em que os bovinos estavam alojados havia grande quantidade de L. camara L. com sinais evidentes de consumo. No total, sete bovinos morreram, e destes, o exame post mortem foi realizado em três. Os bovinos exibiam moderada icterícia, hemorragias no tecido subcutâneo e na superfície de diferentes órgãos. O fígado estava discretamente aumentado, com coloração alaranjada e padrão lobular evidente. As lesões histológicas consistiam em acentuada necrose de coagulação de hepatócitos em região centrolobular, por vezes se estendendo a região mediozonal, além de acentuada degeneração dos hepatócitos e evidente Colestase. O presente trabalho alerta para que intoxicação por L. camara L. seja levada em consideração nos diagnósticos diferenciais de hepatotoxicidade necrótica aguda em bovinos, mesmo sem indícios de fotossensibilização.(AU)

Background: Xanthium spp., the cockleburs, are invasive plants found on riverbanks, lakeshores, and floodplains. Ingestion of Xanthium sprouts or fruit causes cocklebur toxicosis, which is characterized clinically and pathologically by acuteliver failure. The main lesion observed is an accentuation of the lobular pattern of the liver (nutmeg appearance), whichis microscopically characterized by coagulation necrosis in the centrilobular region, hepatocyte degeneration in otherregions of the hepatic parenchyma, and sinusoidal congestion. The objective of this study was to describe an outbreak ofspontaneous cattle poisoning by ingestion of Xanthium spp. sprouts in the southern region of Rio Grande do Sul, Brazil.Case: In September 2018, a beef cattle farm located in Rio Grande county, Rio Grande do Sul, Brazil, reported significantmortality of cattle in a short period of time. There were 700 cattle on the property, divided by class into three differentherds, all kept under extensive conditions in native wetlands. All fields were heavily infested by cocklebur, with mostplants in the budding stage. Twenty animals developed muscle tremors, salivation, aggression, and recumbency. Theclinical course lasted 24 to 48 h, with a lethal outcome in all cases. An 18-month-old male crossbreed bovine was sent tothe Regional Diagnostic Laboratory of the Universidade Federal de Pelotas (UFPel) School of Veterinary Medicine fornecropsy. Gross examination revealed ascites and accentuation of the lobular pattern of the liver. Organ fragments wereharvested and fixed in 10% buffered formalin. Microscopically, the liver exhibited marked coagulation necrosis in thecentrilobular region and vacuolization of the hepatocyte cytoplasm in the midzonal and periportal regions. In the brain,there was perineuronal edema, cytoplasmic shrinkage and basophilia, gliosis, and satellitosis...

Bidens pilosa L. is a medicinal plant popularly used for treatment of liver diseases. In this study, the dry extract of aerial parts of Bidens pilosa and Silymarin, a phytocomplex obtained from the Silybum marianum fruits and marketed as hepatoprotective, were tested in dogs experimentally acutely intoxicated with carbon tetrachloride. The liver activity was evaluated by hematological and biochemical profiles, and histological and ultrasound analyzes. It was observed that the lowest serum activities of ALT and serum concentrations of total bilirubin occurred in the groups treated with the dry extract of Bidens pilosa, while only decreased serum concentrations of total bilirubin occurred in the group treated with Silymarin. Best liver recovery was also observed for the dry extract of B. pilosa at a 400mg/Kg dose by ultrasonography. This study showed that the dry extract of Bidens pilosa acted more efficiently in the treatment of acute toxic hepatitis induced in dogs than Silymarin.(AU)

Neste trabalho, o extrato seco das partes aéreas da Bidens pilosa e a silimarina, um fitocomplexo obtido dos frutos da Silybum marianum e comercializado como hepatoprotetor, foram testados em cães intoxicados experimentalmente de forma aguda com tetracloreto de carbono. A atividade hepática foi avaliada por meio dos perfis hematológico e bioquímico, análises histológica e ultrassonográfica. Crotalaria lanceolata E. Mey. and Crotalaria pallida Aiton. are leguminous plants of family Fabaceae found in most of the Brazilian territory. They were initially used as green manure and due their easy spread they are currently considered weeds in crops. Soybean and corn contamination can occur through the mechanical harvesting of these grains along with seeds of the Crotalaria species, which end up in the formulation of feed for production animals. Crotalaria spp. genus has toxic pyrrolizidine alkaloids (PA). Most plant species belonging to this genus can cause acute or chronic liver injury. In a first stage, one-day old broilers were divided into three groups: Group A (C. pallida seeds), Group B (C. lanceolata seeds), and Group C (Control). Groups A and B were divided into five subgroups, each with eight broilers, which received the following doses of the respective seeds in feed as of the 7th day of age: daily doses of 0.4%, 0.8% and 2.5%, and single doses of 15% and 25%. Four broilers in each study group were euthanized at 28 days of age - completing 21 days of seed consumption, and the four remaining broilers were euthanized at 42 days of age - completing 35 days of seed consumption. In a second stage, experiments were conducted using seeds of both the aforementioned plants with 28-day old broilers. These were divided into three groups of four animals each: Group D (C. pallida seeds) and Group E (C. lanceolata seeds), which received the respective seeds at daily doses of 1% and 2% in feed for 20 days, and Group F (Control). These broilers were euthanized when they were 80 days old. C. lanceolata seeds showed higher toxicity to broilers than C. pallida seeds, both supplied as of the 7th day of life. Clinical signs included inappetence, ruffled feathers, and brown diarrhea. The following gross lesions were observed: subcutaneous edema, ascites, hydropericardium, yellowish liver with hypertrophy or atrophy and enhanced lobular pattern, and distended gallbladder. Histologic lesions present in all birds in varying degrees were characterized by tumefaction and vacuolar degeneration of hepatocytes. The following clinical conditions and gross lesions were observed in the broilers: hepatocyte megalocytosis and karyomegaly, slight biliary epithelial hyperplasia, eosinophilic spheroids, and nuclear invagination with loss of hepatocyte cord architecture.(AU)

Na macroscopia observou-se edema subcutâneo, ascite, hidropericárdio, fígado de coloração amarelada com hipertrofia e, ou, atrofia, evidenciação do padrão lobular e vesícula biliar distendida. As lesões histológicas presentes em todas as aves, em diferentes graus caracterizaram-se por tumefação e degeneração vacuolar de hepatócitos. Background: Cases of plant intoxication in small animals are observed frequently in the domestic environment, mainlybecause most dogs and cats live in households and occasionally have access to streets and rural areas. Among such toxicagents, ornamental plants of the genus Lilium and Hemerocallis, which are potentially nephrotoxic to the feline species,are highlighted. Affected cats start presenting clinical signs 1-6 h after plant ingestion. Renal failure takes place in 12-72h, and death may occur in an interval ranging from three to seven days. The objective of this article is to describe theepidemiological, clinical and pathological findings of lily (Lilium sp.) poisoning in two cats.Case: The aspects of lily poisoning in two cats are described (cat #1 and cat #2). Cat #1 was a 3-year-old, mixed breedfemale cat, which presented a clinical history of anorexia, apathy, drooling, vomiting and polydipsia. Serum biochemicalanalysis revealed creatinine elevation (21.2 mg/dL), as well as hyperphosphatemia (19 mg/dL). Seventy-two h after theonset of clinical signs, renal failure progressed to anuria, followed by death. The second animal of this report (cat #2) wasa 2-year-old, mixed-breed male cat. The animal was found dead by the owner without displaying any previous clinicalsigns. Cats #1 and #2 ingested leaves of lily, which were present in their households as ornamental plants. At necropsy, thekidneys of both cats presented mild enlargement. Moderate perirenal edema was also noted. Cat #1 showed morphologicextrarenal uremic lesions, characterized by ulcers in the oral mucosa and in the margin of the tongue ventral surface. Microscopic lesions observed in both cases were similar and compatible with acute toxic nephropathy. The properties of oil-resin of copaiba, Copaifera multijuga are commonly mentioned in the literature, but there are few studies on extracts from its stem bark. We evaluated the antioxidant effects of ethanolic (EE) and ethyl acetate (EA) crude stem bark extracts from copaiba and compared them to rutin in a paracetamol (PCM)-induced oxidative stress model in mice. All test comparisons differed significantly. Hepatic catalase (CAT) and glutathione-S-transferase (GST) activity decreased in the PCM group, and there was an increase of protein carbonyls in the liver, kidney and brain. However, the protein carbonyls decreased in the liver for the PCM + EE group, in the kidneys for the PCM + EA and PCM + Rutin groups, and in the brain for all treatments. Hepatic GSH decreased in the PCM group and increased in the PCM + EE group. The extracts showed a positive effect on ascorbic acid (ASA), since they were able to restore the levels of parameters that had been changed by PCM. There was an increase of ALT and AST activity in the plasma within the PCM group. Even though ALT decreased in the PCM + Rutin, PCM + EE and PCM + EA groups, EE and EA did not have an effect on AST. The strongest antioxidant effect was observed for EE, due to the presence of the phenolic compounds epicatechin and epiafzelechin, as well as the highest concentration of total phenols and an excellent antioxidant potential observed in the DPPH· test.

Avaliamos os efeitos antioxidantes de extratos brutos etanólico (EE) e acetato de etila (EA) da casca do caule da copaíba e os comparamos à rutina no modelo de estresse oxidativo induzido por paracetamol (PCM) em camundongos. Todas as comparações de teste diferiram significativamente. A atividade da catalase hepática (CAT) e da glutationa-S-transferase (GST) diminuiu no grupo PCM, e houve um aumento de proteínas carboniladas no fígado, rim e cérebro. Background: Stryphnodendron fissuratum is a tree from the Brazilian Cerrado. Its fruit is toxic to cattle and can cause clinical digestive signs, hepatogenous photosensitization, and abortion. Cases of poisoning in cattle, goats and guinea pigs have been experimentally reproduced; however, photosensitization could not be reproduced. The aim of this work was to describe an outbreak of natural poisoning and experimental reproduction in cattle, both with hepatogenous photosensitization.Materials, Methods & Results: Its described and natural outbreak and an experimental poisoning. In the outbreak, three bovines in the acute phase and three in the chronic phase were examined. Blood samples were collected from all of these animals in order to measure serum levels of aspartate aminotransferase (AST), gamma-glutamyl transferase (GGT), urea, and creatinine. The first three animals underwent necropsy and histopathological evaluation. The experiment was conducted with two nine-month-old calves that received an oral paste made with crushed S. fissuratum fruits mixed with water. These fruits were collected at a farm at which cattle poisoning cases had occurred. Blood samples were collected in order to measure serum levels of AST, GGT, urea, and creatinine, before plant administration and then daily during the experimental period. Skin biopsies were taken before plant administration and new one after the first signs of skin lesions. The natural outbreak affected 52 of 160 bovine (31 calves and 21 cows) in the lot. Two calves and 14 cows died. Clinical signs consisted of depression, ataxia, incoordination, behavioral changes, decubitus, and death. One animal that died and 36 others that recovered had photodermatitis. Necropsy findings in the animals consisted of bad corporal condition, pale kidneys, evidence of liver lobular pattern, dry rumen contents, and full bladder.[...]

Bacillus thuringiensis subsp. israelensis has been used to control the Aedes aegypti (Diptera: Culicidae) mosquito larvae, the vector of virus diseases such as dengue, Chikungunya and Zika fever, which have become a major public health problem in Brazil and other tropical countries since the climate favors the proliferation and development of the transmitting vector. Because B. thuringiensis has shown potential for controlling insects of the Diptera order, this work aimed at testing the Bacillus thuringiensis subsp. thuringiensis strain T01-328 and its proteins Cry2Aa and Cry2Ab for control A. aegypti and at comparing the results to the B. thuringiensis subsp. israelensis specific dipteran strain. To this end, bioassays using spore-crystal of both strains, and Cry2Aa and Cry2Ab proteins from the heterologous expression in Escherichia coli, were performed against A. aegypti larvae. The results showed that the B. thuringiensis thuringiensis ­T01-328 has insecticidal activity against the larvae, but it is less toxic than B. thuringiensis subsp. israelensis. Cry2Aa and Cry2Ab proteins expressed heterologously were effective for controlling A. aegypti larvae. Therefore, the results indicate that the Cry2Aa and Cry2Ab proteins of the B. thuringiensis thuringiensis T01-328 can be used as an alternative to assist in the control of A. aegypti.(AU)


Bacillus thuringiensis subsp. israelensis vem sendo empregada no controle do díptero Aedes aegypti, vetor do vírus causador de doenças como dengue, febre Chikungunya e Zika, que se tornou um dos grandes problemas de saúde pública no Brasil e em outros países de clima tropical, que favorece a proliferação e o desenvolvimento do transmissor. Em virtude do potencial de B. thuringiensis no controle de dípteros, a proposta deste trabalho foi testar as proteínas Cry2Aa e Cry2Ab da linhagem de Bacillus thuringiensis subsp. thuringiensis T01-328 no controle de A. aegypti, em comparação à linhagem díptero específica B. thuringiensis subsp. israelensis. Para tanto, foram realizados bioensaios com larvas de A. aegypti com o esporo-cristal de ambas as linhagens, bem como com as proteínas Cry2Aa e Cry2Ab com expressão heteróloga em Escherichia coli. A linhagem B. thuringiensis thuringiensis T01-328 apresentou atividade inseticida contra as larvas, porém foi menos tóxica que a B. thuringiensis subsp. israelensis. As proteínas Cry2Aa e Cry2Ab expressas de forma heteróloga foram eficazes no controle de A. aegypti. Os resultados obtidos sugerem as proteínas Cry2Aa e Cry2Ab da linhagem B. thuringiensis thuringiensis T01-328 como alternativas para contribuir no controle do A. aegypti.(AU)

Ricinus communis is a shrub of the family Euphorbiaceae popularly known in Brazil as "mamona" or "carrapateira". It is an oleaginous plant whose seeds have been used mainly in biodiesel production. Plant seed oil can be extracted mechanically or using solvents, generating castor bean cake and castor bean meal as by-products, respectively. Accidental ingestion of these by-products can cause poisoning in animals and humans, characterized by digestive signs resulting from the presence of a toxalbumin called ricin. Seed toxicity varies among animal species; in horses, the lethal dose of seeds is 0.1 g/kg of body weight. The literature presents plenty of studies addressing poisoning by R. communis in different animal species and in humans; however, reports are scarce and little information is available on the pathological aspects of poisoning regarding equines. Therefore, the present study aimed to describe the epidemiological, clinical and pathological aspects observed in an outbreak of accidental poisoning with castor bean cake in horses. The equines were supplemented with palm kernel (Elaeis guianeesis) cake; however, in the purchase of a new batch, there was an error in the order, and castor bean cake was requested instead. Four horses that received castor bean cake presented clinical signs indicative of colic, which started about 21 hours after administration. Three of these horses died, with clinical evolution from 2 to 4.5 hours; the other animal was treated with intravenous fluid and antibiotic therapy and non-steroidal anti-inflammatory drugs and recovered after five days. In the necropsy of two of these horses, the main lesions were found in the small intestine, where mucosae with pronounced redness and covered with a thin layer of yellow fibrous material were observed. In the intestinal lumen, there was a large amount of bloody liquid. The stomach was full, and dark lumps similar to crushed castor bean seeds were observed in the contents. The adrenal glands of both horses presented congestion and hemorrhage in the cortex. In microscopy, the main lesion was an acute, diffuse and accentuated fibrin-necrotic enteropathy affecting the jejunum. The diagnosis of castor bean cake poisoning was based on the circumstantial evidence of by-product consumption supported by clinical and pathological aspects. The results indicate that castor bean cake commercially available as fertilizer is extremely toxic when ingested, and there is a need for appropriate detoxification or labeling informing of its toxicity.(AU)


Ricinus communis é um arbusto da família Euphorbiaceae conhecido popularmente como "mamona" ou "carrapateira". A planta é considerada oleaginosa e suas sementes têm sido utilizadas, principalmente, na produção de biodiesel. A extração do óleo pode ser mecânica ou com solventes, gerando, como subprodutos, a torta de mamona e a farinha de mamona, respectivamente. A ingestão acidental desses subprodutos pode causar intoxicação em animais e humanos, caracterizada por sinais digestivos devido a toxalbumina ricina. A toxidez das sementes varia entre as espécies animais, em equinos a dose letal de sementes é de 0,1g/kg de peso vivo. Há vasta literatura sobre a intoxicação por R. communis em diferentes espécies animais e humanos. Em relação aos equinos os relatos são escassos e com poucas informações sobre os aspectos patológicos da intoxicação. Assim, os objetivos do presente estudo foram descrever os aspectos epidemiológicos, clínicos e patológicos observados em um surto de intoxicação acidental por torta de mamona em equinos. Os equinos eram suplementados com torta de dendê (Elaeis guianeesis), no entanto, na compra de uma nova partida, houve um erro no pedido sendo solicitada torta de mamona. Os quatro equinos que receberam a torta de mamona apresentaram sinais clínicos indicativos de cólica, que iniciaram cerca de 21 horas após administração. Três destes morreram, com evolução clínica 2 a 4,5 horas, o outro animal foi tratado com fluidoterapia intravenosa, antibioticoterapia e anti-inflamatório não esteroidal, recuperando-se em 5 dias. Na necropsia de dois destes equinos, as principais lesões foram encontradas no intestino delgado, sendo observadas mucosas com avermelhamento acentuado e recobertas por fina camada de material amarelado fibrinoso. No lúmen intestinal, havia grande quantidade de líquido sanguinolento. O estômago estava repleto de grumos escuros semelhantes às sementes trituradas da mamona em meio ao conteúdo. As adrenais de ambos equinos apresentavam congestão e hemorragias corticais. Na microscopia, a principal lesão foi uma enteropatia fibrino-necrótica, aguda, difusa e acentuada, afetando com maior intensidade o jejuno. O diagnóstico de intoxicação por torta de mamona foi baseado na evidência circunstancial de consumo do subproduto, sendo corroborado pelos aspectos clínicos e patológicos. Os resultados indicam que a torta de mamona vendida para fertilização do solo é extremamente tóxica quando ingerida, havendo a necessidade de detoxicação ou rotulagem adequada informando sobre a sua toxicidade.(AU)
