Resumo

Vacas de alta produção de leite estão mais sujeitas a desequilíbrios metabólicos pós-parto, influenciando na performance reprodutiva subsequente. Alterações relacionadas a funcionalidade do ovário podem desencadear desequilíbrios hormonais e desordens reprodutivas, como a formação de cistos ovarianos, os quais podem interferir no processo de ovulação e impactar substancialmente na eficiência reprodutiva. Neste estudo de caso em específico, sete vacas leiteiras foram diagnosticadas com a presença de cistos foliculares ovarianos. Foram submetidas a terapia hormonal envolvendo o uso de análogos de Hormônio Liberador de Gonadotrofinas (GnRH) e prostaglandina-F2a (PGF-2a), a qual apresentou resultados satisfatórios. A abordagem terapêutica hormonal tem sido amplamente utilizada devido à sua efetividade na resolução dos cistos ováricos e melhoria da função reprodutiva. É fundamental que o médico veterinário tenha conhecimento clínico sobre a abordagem adequada de cistos ovarianos, a fim de proporcionar saúde e bem-estar aos animais, garantindo bons índices em produção de leite e desempenho reprodutivo.


High milk production cows are more likely to postpartum metabolic imbalances, which may influence subsequent reproductive performance. Changes in ovarian functionality can trigger hormonal alterations and reproductive disorders, such as the formation of ovarian cysts, which can interfere with the ovulation process and substantially impact reproductive efficiency. In this specific case study, seven dairy cows were diagnosed with the presence of ovarian follicular cysts. They were submitted to hormonal therapy involving the use of Gonadotropin-Releasing Hormone (GnRH) and prostaglandin F2a (PGF-2a) analogs, which presented satisfactory results. Hormonal therapeutic approaches have been widely employed due to their effectiveness in resolving ovarian cysts and improving reproductive function. It is essential for the veterinarian to have clinical knowledge about the appropriate management of ovarian cysts in order to provide the health and welfare of the animals, ensuring suitable rates in milk production and reproductive performance.


Las vacas de alta producción de leche tienen mayor probabilidad de desequilibrios metabólicos posparto, lo cual puede influir en el rendimiento reproductivo posterior. Los cambios en la funcionalidad ovárica pueden desencadenar alteraciones hormonales y trastornos reproductivos, como la formación de quistes ováricos, que pueden interferir en el proceso de ovulación y afectar sustancialmente la eficiencia reproductiva. En este estudio de caso específico, se diagnosticó la presencia de quistes foliculares ováricos en siete vacas lecheras. Se sometieron a terapia hormonal que incluyó el uso de análogos de Hormona Liberadora de Gonadotropinas (GnRH) y prostaglandina F2α (PGF-2a), lo cual arrojó resultados satisfactorios. Los enfoques terapéuticos hormonales se han utilizado ampliamente debido a su efectividad en la resolución de quistes ováricos y la mejora de la función reproductiva. Es fundamental que el médico veterinario tenga conocimiento clínico sobre el manejo adecuado de los quistes ováricos para garantizar la salud y el bienestar de los animales, asegurando adecuadas tasas de producción de leche y rendimiento reproductivo.

Background: Measurement of pregnancy-associated glycoproteins (PAGs) by radioimmunoassay or enzyme-linked immunosorbent assay has been commonly used for early pregnancy diagnosis in ruminants. However, an accurately pregnancy detection depends on test antibody, breed and number of embryos. Only few studies have conducted to detect or predict animals at risk of late embrionic mortality (LEM) and to use hormonal interventions for embryo losses reducing, but this area is still open in sheep. The current study aimed to evaluate the effect of gonadotropin releasing hormone (GnRH) administration on Day 4 after artificial insemination on the pregnancy-associated glycoproteins in dairy sheep with different reproductive status in relation to pregnancy and late embrionic mortality detection. Materials, Methods & Results: Sixty-five East Friesian sheep were divided in 2 groups - I (Control group, n = 35) and II (GnRH group, n = 30) and sublected to estrus synchronization and artificial insemination (AI). Group I was not treated and Group II received 50 µg GnRH on Day 4 after AI. PAGs in blood serum were measured by Alertys Ruminant Pregnancy test on Days 4, 12, 20, 25 and 35 after AI and ultrasound pregnancy test was conducted on Days 20, 25, 35 and 60. Reproductive status (non-pregnant, pregnant and animals with LEM) was determined by ultrasound and the results between different groups were compared. The PAGs mean values according to reproductive status and Day after AI were analysed. Accuracy, sensitivity and specificity of the PAGs test for pregnancy diagnosis on Day 35 were calculated. On Day 20 after AI the pregnat sheep (83.3%) in group I tended to be higher than those (77.1%) in group II with decrease of 25.7% and 20% for the same groups on Day 25. On day 25 LEM was recorded in 33.3% and 24% in group I and II, respectively. The ultrasound exams on Day 60 confirmed the results from Day 25 after AI. A total value of non-pegnant, pregnant animals and LEM was 20%, 56.9% and 28.8%, respectively. The mean values of PAGs in animals with the same reproductive status in group I and II no differed statistically between Days 4 and 35 after AI. On Day 25 the PAGs values in LEM groups (0.126 ± 0.072 and 0.179 ± 0.029) were higher than those (0.062 ± 0.038 and 0.083 ± 0.023) in the non-pregnant groups, but no significan difference was deternined. On Day 35 after AI the mean values of PAGs (0.414 ± 0.125 and 0.421 ± 0.121) for the pregnant groups were significantly (P < 0.05) higher than those (0.078 ± 0.053 and 0.093 ± 0.034) for the non-pregnant groups. The values of PAGs in LEM groups on Day 25 (0.099 ± 0.062 and 0.113 ± 0.058) were decreased and close to the mean values in non-pregnant sheep. A significant effect of the day after AI on the PAGs values was evidenced in pregnant animals only (control r = 0.97 and GnRH r = 0.98; P < 0.05). The PAGs started to rise rapidly between Days 25 and 35 after AI. On Day 35 the accuracy (98%), the sensitivity (97.3%) and the specificity (100%) of the PAG test for pregnancy diagnosis were similar to the values (100%) for ultrasound method. Discussion: The gonadotropin treatment on Day 4 after AI tended to improve the reproductive performance in the sheep, but no significant effect of GnRH on the PAGs values in the animals with different reproductive status was found. The PAGs profiles in non-pregnant and LEM sheep were close and distinguishing between non-pregnant and LEM ewes on Day 35 was impossible. The PAGs values in pregnant sheep showed significantly (P < 0.05) increase between Days 25 and 35 after AI with higher mean values in pregnant than non-pregnant groups on Day 35 (P < 0.05). Alertys Ruminant Pregnancy test was a reliable for pregnancy detection in sheep on Day 35 after artificial insemination.

Currently, gonadotropin products (follicle stimulating hormone, FSH, and luteinizing hormone, LH) used in animal reproduction are produced by extraction and purification from abattoir-derived pituitary glands. This method, relying on animal-derived materials, carries the potential risk of hormone contamination and pathogen transmission. Additionally, chorionic gonadotropins are extracted from the blood of pregnant mares (equine chorionic gonadotropin; eCG) or the urine of pregnant women (human chorionic gonadotropin; hCG). However, recent advancements have introduced recombinant gonadotropins for assisted animal reproduction therapies. The traditional use of FSH for superovulation has limitations, including labor requirements and variability in superovulation response, affecting the success of in vivo (SOV) and in vitro (OPU/IVEP) embryo production. FSH treatment for superstimulation before OPU can promote the growth of a homogenous follicular population and the recovery of competent oocytes suitable for IVEP procedures. At present, a single injection of a preparation of long-acting bovine recombinant FSH (rFSH) produced similar superovulation responses resulting in the production of good-quality in vivo and in vitro embryos. Furthermore, the treatment with eCG at FTAI protocol has demonstrated its efficacy in promoting follicular growth, ovulation, and P/AI, mainly in heifers and anestrous cows. Currently, treatment with recombinant glycoproteins with eCG-like activity (r-eCG) have shown promising results in increasing follicular growth, ovulation, and P/AI in cows submitted to P4/E2 -based protocols. Bovine somatotropin (bST) is a naturally occurring hormone found in cows. Recombinant bovine somatotropin (rbST), produced through genetic engineering techniques, has shown potential in enhancing reproductive outcomes in ruminants. Treatment with rbST has been found to improve P/IA, increase donor embryo production, and enhance P/ET in recipients. The use of recombinant hormones allows to produce non-animal-derived products, offering several advantages in assisted reproductive technologies for ruminants. This advancement opens up new possibilities for improving reproductive efficiency and success rates in the field of animal reproduction.(AU)

This study was conducted to investigate the effects of amylin, a pancreatic hormone polypeptide, on egg quality traits in laying hens. A total of 40 birds (14 wk of age) were randomly divided into two treatments with two replicates, 10 birds in each replicate. Twenty birds were subcutaneously injected with amylin at 75 µg/kg every other day (treatment 1), and the remaining animals (20 hens) were given only water as the control group (treatment 2). Eggs collected from the two groups (140 eggs per group) were examined for several quality performance traits such as egg weight, specific gravity, shape index, eggshell strength, eggshell thickness, eggshell ash, eggshell weight, Haugh unit (HU), albumen index, yolk index, yolk color, and albumen height. The results indicated that amylin had a positive effect on some egg quality traits in laying hens. Specific gravity, eggshell thickness, eggshell weight, HU, albumen index, yolk index, and albumen height, which are all considered as important quality parameters from the consumer point of view, especially HU, were relatively improved in the amylin treatment group. These results showed that the injection of 75 µg/kg amylin has a significant effect on some egg quality traits. Considering that egg quality characteristics are crucial for the egg industry, this study can be a reference for the detailed investigation of the use of amylin in the different stages of egg production.

The females of yellowtail tetra (Astyanax lacustris), known as the freshwater sardine, are approximately 1.33 times larger than males, and thus, all-female monosex culture would increase production and reduce size variability. The present work aimed to identify the optimal dose of 17α-methyltestosterone (MT) to be used in the masculinization of A. lacustris for indirect sex reversal. Three different concentrations of MT (20, 40, and 60 mg/kg of feed in the diet) were fed to the fry for 30 days. Thirty adult individuals from each treatment, including the control (0 mg MT/kg), were evaluated for gonadal development, morphological and histological sexual identification, zootechnical performance, and the possible genotoxic effect caused by prolonged exposure to MT. MT significantly (P<0.01) affected the differentiation of the gonads, with the presence of possible inhibitory effects in all treatments. Intersex individuals were present in the 20 and 60 mg MT/kg treatments. All treatments were able to masculinize A. lacustris and the treatment with the lowest hormone concentration produced the highest percentage of males 76.7%, while the control had 46.7% males. The presence of erythrocyte nuclear alterations indicated a possible cytotoxic effect of MT in treatments 40 and 60 mg MT/kg, however, the use of the hormone did not affect the growth and the survival of the individuals. Thus, the use of MT is a viable option for obtaining neomales as a first step into the production of all-female progenies.(AU)

ABSTRACT: Methyl jasmonate (MeJA) is recognized as a plant hormone that induces several biochemical changes related to improving fruit quality, but it is evident that the effect of MeJA during postharvest is very variable upon concentration, plant species, and even cultivars. The objective of this research was to relate the application of this regulator at low concentration (10 µmol L-1 for 24 h) to changes in postharvest physiology, on the incidence of anthracnose and quality of 'Golden' papaya fruit during storage at 24 ± 1°C. From the seventh day of storage, anthracnose incidence was reduced by more than 50% with MeJA treatment. The lesion area after infection was also reduced. Although, MeJA reduced fruit acidity, the ascorbic acid and soluble solids content were not altered during storage with the hormonal treatment. The skin color and pulp firmness showed slight retention. The reduction in ethylene production was accompanied by a reduction of respiration in treated fruits. MeJa increased CAT activity only in the skin whereas SOD activity was not induced by MeJA in both skin and pulp. Although, the increase of CAT from the third day of storage may have contributed to the reduction of lipid peroxidation in the skin, the MDA reduction in the pulp cannot be explained only by CAT activity. In summary, the application of MeJA in 'Golden' papaya reduced the incidence and severity of anthracnose, decreased respiration, ethylene production and lipid peroxidation. It is concluded that the application of MeJA at a low concentration (10 µmol L-1) may contribute to anthracnose control in ´Golden`papaya and slows the ripening of fruits.


RESUMO: O metil jasmonato (MeJA) é reconhecido como um hormônio vegetal que induz várias alterações bioquímicas relacionadas à qualidade dos frutos, mas é evidente que o efeito do MeJA durante a pós-colheita é muito variável quanto à concentração, espécies de plantas e até mesmo cultivares. O objetivo deste trabalho foi relacionar a aplicação deste regulador em baixa concentração (10 µmol L-1 por 24 h) às mudanças na fisiologia pós-colheita, na incidência de antracnose e na qualidade do mamão 'Golden' durante o armazenamento a 24 ± 1 ° C. A partir do sétimo dia de armazenamento, a incidência de antracnose foi reduzida em mais de 50% com o tratamento com MeJA. Incidência da doença também foi reduzida. Embora o MeJA tenha reduzido a acidez dos frutos, os teores de ácido ascórbico e sólidos solúveis não foram alterados durante o armazenamento com o tratamento hormonal. A cor da casca e a firmeza da polpa apresentaram ligeira retenção. A redução na produção de etileno foi acompanhada pela redução da respiração nos frutos tratados. MeJa aumentou a atividade da CAT apenas na casca, enquanto a atividade da SOD não foi induzida por MeJA na casca e na polpa. Embora o aumento da CAT a partir do terceiro dia de armazenamento possa ter contribuído para a redução da peroxidação lipídica na casca, a redução do MDA na polpa não pode ser explicada apenas pela atividade da CAT. Em resumo, a aplicação de MeJA em mamão 'Golden' reduziu a incidência e severidade da antracnose, diminuiu a respiração, a produção de etileno e a peroxidação lipídica. Conclui-se que a aplicação de MeJA em baixa concentração (10 µmol L-1) pode contribuir para o controle da antracnose em mamão ´Golden` e retardar o amadurecimento dos frutos.

Background: Hypertrophic cardiomyopathy phenotype (HCM) is the most common cardiac disease in domestic cats but is rarely described in wild species. This phenotype is characterized by concentric hypertrophy of the left ventricle and may be of familial inheritance or secondary to other diseases such as hyperthyroidism, chronic kidney disease, systemic arterial hypertension, and hyperaldosteronism. HCM can cause diastolic and systolic dysfunction and may cause congestive heart failure in affected animals. The present work aims to describe the first report of cardiomyopathy of the hypertrophic phenotype in a specimen of Leopardus pardalis, kept under human care. Case: A 11-year-old female ocelot (Leopardus pardalis) kept under human care, during a preventive care visit, had hypertrophic cardiomyopathy phenotype detected by cardiological evaluation with echocardiogram and a murmur grade III/ VI could be detected on cardiac auscultation. This preventive care occurred under chemical restraint with ketamine [6 mg/kg, i.m] associates with midazolam [0.5 mg/kg, i.m] and other evaluations have been done like complete blood count (CBC), biochemistry (alanine aminotransferase, aspartate aminotransferase, total proteins, albumin, albumin:globulin ratio, creatinine, urea, sodium, potassium, calcium, phosphorus, and globulin) and tyroid hormones [free tyroxine (T4) and thyroid-stimulating hormone (TSH)]. Medical management based on clopidogrel, and atenolol was administered for 3 months until the patient showed manifestations of congestive heart failure (CHF) 80 days later the initial evaluation. In this moment the patient presented with dyspnea, so a cardiological and laboratory evaluation was requested. On pulmonary auscultation crackling was identified, suggesting pulmonary edema and, on echocardiographic examination, some parameters had worsened. The CBC and biochemistry were all within reference ranges. Then, the beta-blocker was discontinued and replaced by pimobendan combined with furosemide as treatment of CHF, and the condition stabilized. After one year, the patient was re-evaluated and showed a slight improvement in the condition but still remained stable. Also, feline proBNP levels was tested (SNAP Feline proBNP® IDEXX) in this moment and it was increased. Discussion: The findings on echocardiography associated with the subjective evaluation associated with progressive worsening and clinical manifestation of CHF, as well as the response to treatment, even though there are no reference values for the species, reinforce the diagnosis. There is no evidence to suggest diseases that may contribute to secondary left ventricular hypertrophy. It is believed that pimobendan plays a key role in maintaining hemodynamic balance, since this has already been observed in other mammalian species. The use of beta blockers is commonly employed in domestic cats with HCM, and they have been prescribed in an attempt to promote greater ventricular relaxation, decrease left ventricular outflow obstruction, thus improving ventricular filling for maintenance of cardiac output. In view of the atrial enlargement and possible risk of thrombus formation, clopidogrel was prescribed, extrapolating what is known from domestic cats. It is reasonable to conclude that in this case, the cardiomyopathy behaved similarly to what is observed in domestic cats, both in its clinical evolution and in the means of diagnosis, and in its response to the therapy instituted.

Resumo

O hipoadrenocorticismo primário ou doença de Addison é uma endocrinopatia que envolve a deficiência de glicocorticoides, comumente associada a deficiência mineralocorticoide. O tratamento mais utilizado para reposição hormonal envolve a associação de fludrocortisona e prednisona. O pivalato de desoxicorticosterona (DOCP) tem sido demonstrado na literatura como superior no controle eletrolítico no paciente com hipoadrenocorticismo, e sempre deve ser associado a prednisona/prednisolona. Neste trabalho, relatamos o benefício do DOCP e o protocolo de ajuste de dose desse fármaco em duas etapas em dois casos distintos. Houve uma eficácia superior no controle eletrolítico e redução dos efeitos colaterais com o protocolo de DOCP e prednisolona em relação ao uso de fludrocortisona em ambos os pacientes.(AU)


Primary hypoadrenocorticism or Addison's disease is an endocrinopathy involving glucocorticoid deficiency, commonly associated with mineralocorticoid deficiency. The most commonly used treatment for hormone replacement involves the combination of fludrocortisone and prednisone. Deoxycorticosterone pivalate (DOCP) has been shown in the literature to be superior in electrolyte control in patients with hypoadrenocorticism, and should always be associated with prednisone/prednisolona. In this study, we report the benefit of DOCP and the two-stage dose adjustment protocol for this drug in 2 different cases. There was superior efficacy in electrolyte control and reduction of side effects with the DOCP and prednisolone protocol compared to the use of fludrocortisone in both patients.(AU)


El hipoadrenocorticismo primario o enfermedad de Addison es una endocrinopatía que cursa con deficiencia de glucocorticoides, comúnmente asociada con deficiencia de mineralocorticoides. El tratamiento más utilizado para el reemplazo hormonal consiste en la combinación de fludrocortisona y prednisona. El pivalato de desoxicorticosterona (DOCP) ha demostrado en la literatura ser superior en el control de electrolitos en pacientes con hipoadrenocorticismo, y siempre debe asociarse con prednisona/prednisolona. En este trabajo, reportamos el beneficio de DOCP y el protocolo de ajuste de dosis en dos etapas para este fármaco en 2 casos diferentes. Hubo una eficacia superior en el control de electrolitos y la reducción de los efectos secundarios con el protocolo DOCP y prednisolona en comparación con el uso de fludrocortisona en ambos pacientes.(AU)

Background: Pyometra is a bacterial and hormone-induced reproductive disease that occurs in the post-estrus luteal phase in intact queens. Pyometra is more common in the diestrus period due to the high progesterone concentration (in queens that mated, spontaneously ovulated, or were induced to ovulate). However, it can also be seen due to the use of exogenous hormones such as progesterone for the suppression of estrus. More research is needed in cases of pyometra in queens, as well as in bitches. Because, considering that the pathogenesis and characteristics of feline pyometra is similar to bitches, studies on pyometra-affected bitches are taken as reference in studies and applications on queens. From this point of view, the aims of this study were to reveal the changes in complete blood count, blood gas, and serum biochemistry parameters in feline pyometra cases and to determine the correlation between the mentioned parameters. Materials, Methods & Results: In the study, a total of 25 female cats of different breeds were used, between the ages of 6 months and 7 years, 15 were diagnosed with pyometra, and 10 healthy. Anamnesis, clinical findings, and ultrasonographic examinations were used in the diagnosis of pyometra. Abdominal ultrasonography was performed on queens brought to the clinic with complaints such as anorexia, polydipsia, polyuria, abdominal tension, and fever. The control group (n =10) consisted of queens that were introduced to the clinic and were reproductively healthy. Before any treatment in queens with pyometra and the control group, 1 mL blood samples were taken from v. cephalica to evaluate complete blood count, blood gases and serum biochemistry parameters. In complete WBC, Lym, Mon, Gra, RBC, Hb, HCT, MCV, MCH and PLT parameters and, blood gas parameters such as pH, pCO2 , pO2 , sO2 , Na, K, Cl, lactate, glucose, HCO3 , and BE were also evaluated in taken blood samples. Biochemical parameters BUN, creatinine, ALT, AST, ALP, amylase, T.BIL, D.BIL, P, CHOL, TG, LDH, TP, CPK, ALP, Ca, GGT were measured in serum samples. After examination and laboratory analysis, ovariohysterectomy was performed on queens as a treatment. Granulocyte, WBC, HCT and MCH levels of the pyometra group were higher (P < 0.05) and Lym levels were lower (P < 0.05) compared to the control group. According to these results, pH, HCO3 , and BE were lower (P < 0.05) in queens with pyometra than those in the control group, while Na and lactate parameters were higher (P < 0.05). According to the results of biochemical analysis, it was determined that BUN, creatinine levels, GGT, and LDH enzyme activities were found to be higher in the pyometra group compared to the control group, while the Ca level was found to be low (P < 0.05). A positive correlation was observed between BUN and creatinine and LDH, WBC, granulocyte, HCT, and lactate, and a negative correlation between lymphocytes, pH, and BE in the correlation analysis performed on queens with pyometra and control group. However, a positive correlation was observed between creatinine and LDH and HCT, and a negative correlation between lymphocyte, pH and BE. Discussion: There is not enough information about pyometra in queens. As a result, it was determined that there were significant changes in complete blood count, blood gases and serum biochemical parameters in queens with pyometra in this study. These changes were generally thought to be related to dehydration and sepsis or endotoxemia. In addition, it was evaluated that prerenal azotemia occurring in pyometra affected queens may cause renal dysfunction. For this reason, it is thought that the results obtained in the presented study may contribute to the diagnosis, treatment, and prognosis of pyometra cases in queens.

Progesterone (P4) can participate in the development of female mammalian antral follicles through nuclear receptor (PGR). In this experiment, the differences of P4 synthesis and PGR expression in different developmental stages of sheep antral follicles (large > 5mm, medium 2-5mm, small < 2mm) were detected by enzyme-linked immunosorbent assay, immunohistochemistry, qRT-PCR and Western blotting. Secondly, sheep follicular granulosa cells were cultured in vitro. The effects of different concentrations of FSH and LH on P4 synthesis and PGR expression were studied. The results showed that acute steroid regulatory protein (StAR), cholesterol side chain lyase (P450scc) and 3β Hydroxysteroid dehydrogenase (3β-HSD) and PGR were expressed in antral follicles, and with the development of antral follicles in sheep, StAR, P450scc and the expression of 3β-HSD and PGR increased significantly. In vitro experiments showed that FSH and LH alone or together treatment could regulate P4 secretion and PGR expression in sheep follicular granulosa cells to varying degrees, hint P4 and PGR by FSH and LH, and LH was the main factor. Our results supplement the effects of FSH and LH on the regulation of P4 synthesis during follicular development, which provides new data for further study of steroid synthesis and function in follicular development.(AU)

There is still no consensus regarding the best protocol for in vivo embryo production in sheep despite increasing studies in this area. Moreover, there is variability in the response of ewes to superovulation (SOV). An approach to mitigate this inconsistency is to initiate gonadotropin administration under favorable ovarian conditions. The present study compared three treatments in a crossover design: a traditional SOV protocol (TRAD) and "Day 0" D0 SOV protocol with (D0+GnRH), or without Lecilerin (D0-GnRH). Fifteen Santa Inês ewes received 200 mg of FSH at six decreasing doses and PGF2α with the fifth dose of FSH. They were naturally mated with fertile rams and subjected to surgical embryo collection. The number of viable embryos was similar among the different treatments (TRAD = 6.0 ± 4.7; D0-GnRH = 3.8 ± 6.4; D0+GnRH = 7.5 ± 6.5). Regardless of the treatment method, ewes with follicles ≤ 4 mm, at the first FSH dose, produced more viable embryos (9.6 ± 6.0, P < 0.05) compared to ewes that had follicles > 4 mm at the beginning of the SOV (2.9 ± 3.1, viable embryos). Both the TRAD and D0+GnRH groups had fewer animals with large follicles (> 4 mm) at the first FSH dose than the D0-GnRH group (P < 0.05). In conclusion, both the TRAD and D0+GnRH treatments induced a more favorable ovarian condition (follicles ≤ 4 mm) for adequate SOV; although, all three treatments exhibited similar efficacies in Santa Inês sheep.


Ainda não há consenso sobre qual é o protocolo mais apropriado para a produção in vivo de embriões em ovinos, apesar do crescente conhecimento. Uma abordagem para mitigar a variabilidade de resposta de ovelhas à superovulação (SOV) é iniciar a aplicação de gonadotrofinas em uma condição ovariana favorável. O presente estudo comparou três tratamentos em delineamento do tipo crossover: protocolo de SOV tradicional (TRAD) e "Dia 0" D0 SOV sem (D0-GnRH) ou com GnRH (D0+GnRH). Quinze ovelhas Santa Inês foram superovuladas com 200 mg de FSH em seis doses decrescentes e receberam PGF2α na quinta dose de FSH. As ovelhas foram submetidas a monta natural com carneiros férteis e os embriões colhidos por via cirúrgica. O número de embriões viáveis não diferiu entre os tratamentos (TRAD = 6,0 ± 4,7; D0-GnRH = 3,8 ± 6,4; D0+GnRH = 7,5 ± 6,5). Independentemente do tratamento, ovelhas com folículos ≤ 4 mm na primeira dose de FSH produziram mais embriões viáveis (9,6 ± 6,0; P < 0.05) quando comparadas aos animais que apresentavam folículos > 4 mm no início da SOV (2,9 ± 3,1 embriões viáveis). Os grupos TRAD e D0+GnRH apresentaram menor número de animais com folículos grandes (> 4 mm), no momento da primeira dose de FSH, quando comparados ao grupo D0-GnRH (P < 0,05). Em conclusão, os protocolos TRAD e D0+GnRH induziram uma condição ovariana mais favorável (folículos ≤ 4 mm) para a SOV. No entanto, os três tratamentos apresentaram eficiência semelhante em ovelhas Santa Inês.

Estradiol cypionate (EC) or GnRH have been widely used for ovulation induction in timed embryo transfer (TET). EC administration increases the proportion of cows that show estrus, whereas GnRH promotes more synchronized ovulations. The aim of the present study was to evaluate the potential beneficial effects of combining EC and GnRH in TET. In experiment 1, no difference was observed on serum progesterone concentrations on Day 6 and 13 after GnRH treatment between GnRH and EC+GnRH groups. In experiment 2, pregnancy per embryo transfer (P/ET) did not differ (p = 0.69) between GnRH (62.8%) and EC+GnRH (58.7%) groups. In conclusion, combining EC and GnRH for ovulation induction does not increase progesterone secretion and pregnancy rate after TET in cattle.(AU)

Background: Juvenile panhypopituitarism is an endocrinopathy that affects young dogs and must be differentiated from other causes of height disorders, as one could have a deficiency of one or more adenohypophysis hormones, besides growth hormone. Diagnosis consists of excluding endocrine and non-endocrine causes. Treatment requires administration of heterologous porcine growth hormone or progestins, as well as supplementing other hormones that are deficient in the circulation. The prognosis for these patients tends to be unfavorable and they have a shorter life expectancy. The present case aimed to report the therapeutic challenges in a dog diagnosed with juvenile panhypopituitarism. Case: This study presents the case of a 6-month-old crossbred Labrador/Brazilian Fila dog, not neutered, 6.7 kg of body weight (body condition score 5/9), with a growth disorder and persistence of the puppy coat, when compared to other members of the same litter. During inspection, it was possible to observe an undersized dog and soft puppy coat, besides no bone irregularities or joint tenderness was noted. The other physical examination parameters were within the normal range for the species. No changes in complete blood count and only increases in urea, cholesterol and alkaline phosphatase activity were observed. Thyroid and abdominal ultrasound (US) examination did not reveal any remarkable changes. After serum dosage of insulin-like growth factor, thyroxine, thyrostimulating hormone, and cortisol, the patient was diagnosed with juvenile panhypopituitarism and underwent therapy with medroxyprogesterone and thyroid hormone supplementation. Monitoring was instituted at intervals of 3, 6, and 12 weeks, and currently every 3 or 4 months and the IGF-1 values normalized after 6 months with. After 8 months of therapy, the patient had good body growth and bone mineralization compared to the time of diagnosis. However, skeletal development was completed only 12 months after hormone replacement, accompanied by the presence of vertebral osteophytes and coxofemoral osteoarticular alterations. Considering chronic use of progestins, ovariosalpingohysterectomy (OSH) was recommended, but for personal reasons, the owners chose to do not submit her to surgery. After 18 months of treatment, the dog starts to exhibit prostration, selective appetite, and increased abdominal volume. After imaging exam, she was forwarded for OSH, due to consistent findings of pyometra. Subsequently, even the supervision of possible comorbidities involved in the chronic use of progestins was maintained at half-yearly intervals, the bitch returned to the service with skin thickening, increased limb volume, and macroglossia after 60 months of therapy. At this point IGF-1 values were higher when compared to the previous measurements and the application of medroxyprogesterone was suspended. Its supplementation was reintroduced only after 8 months when IGF-1 was significantly reduced. To date, the patient is close to 6 years of age and with a good quality of life. Discussion: The greatest therapeutic challenge for these patients involves dealing with the adverse effects of progestins, mainly related to reproductive disorders and alopecia at the application site, as well as maintaining adequate hormone replacement in order to avoid hypersomatotropism. Although longevity is lower in these individuals, the patient in this report has achieved 6 years of age and has had an excellent quality of life so far.

This study evaluated (a) the efficacy of an association between injectable antibiotic therapy and sealant (ATBS) on milk yield (MY), somatic cell count (SCC), and prevalence of intramammary infections (IMI); and (b) the efficacy of gonadotropin-releasing hormone (GnRH) on follicular cyst (FCs) resolution (cyclicity at the 45th day in milk; DIM) and cumulative pregnancy rate (CPR) in heifers submitted to a lactation induction protocol (LIP). A total of 114 crossbred (Holstein × Jersey) heifers, with 34.7 ± 4.8 months and 439 ± 56.35 kg were submitted to LIP. On the 5th day of the LIP, the heifers were assigned to (i) ATBS (n = 57) with 7 mg/kg of norfloxacin associated with sealant and (ii) Control 1 (n = 57; CONT1) with no treatments. Lactation began on the 21st day of LIP and the 15th DIM, FCs were diagnosed and 106 heifers were randomized into two treatment groups with 53 heifers each: (i) GnRH (5 mL injectable GnRH) and (ii) Control 2 (CONT2; no treatment). Of the 114 heifers initially induced, 83.33% (n = 95) responded to LIP with an average MY of 15.19 kg/milk/day during 22 weeks of lactation. In the first 14 DIM, the IMI prevalence was 18% and 28% for heifers ATBS and CONT1 treated, respectively. Additionally, coagulase-negative Staphylococcus was the most frequently isolated group of pathogens. Mammary quarters that received ATBS treatment had a lower risk of IMI and SCC than CONT1. The cyclicity at 45 DIM was 68% (ATBS) and 35% (CONT1), and 57% and 46% for animals in the GnRH and CONT2. CPR was 60% in the ATBS group and 89% in CONT1, but GnRH treatment did not affect the CPR. In conclusion, LIP was effective in stimulating MY in heifers, and the IMI prevalence decreased with ATBS treatment. Also, the use of GnRH did not affect the FC regression, cyclicity at 45 DIM, and CPR.(AU)


Este estudo avaliou a (i) eficácia da associação entre antibioticoterapia injetável e selante interno de tetos (ATBS) na produção de leite (PL), contagem de células somáticas (CCS), e prevalência de infecções intramamárias (IIM); e (ii) eficácia do hormônio liberador de gonadotrofina (GnRH) na resolução de cistos foliculares (CFs), ciclicidade ao 45º dia em lactação (DEL) e taxa de prenhez cumulativa (TPC) em novilhas submetidas a um protocolo de indução de lactação (PIL). Um total de 114 novilhas mestiças (Holandês × Jersey), com 34,7 ± 4,8 meses e 439 ± 56,35 kg foram submetidas ao PIL. No 5º dia do PIL, as novilhas receberam: (i) ATBS (n = 57) com 7 mg/kg de norfloxacina associada ao selante interno de tetos e (ii) Controle 1 (n = 57; CONT1) sem tratamento. A lactação teve início no 21º dia do PIL e no 15º DEL, foram diagnosticados CFs e 106 novilhas foram agrupadas em dois grupos de tratamento com 53 novilhas em cada: (i) GnRH (5 mL de GnRH injetável) e (ii) Controle 2 (CONT2; sem tratamento). Em conclusão, o PIL foi eficaz em estimular a PL em novilhas tardias e a prevalência de IIM diminiuiu com o tratamento ATBS. Além disso, o uso de GnRH não afetou a regressão de CF, ciclicidade em 45 DEL e a TPC.(AU)

Background: Hypothyroidism is characterized by hypofunction of the thyroid gland. It results in deficient production of thyroid hormones. Neurological disorders resulting from hypothyroidism are rare, which highlights the importance of this study. This study reports a case of hypothyroidism in a dog with neurological clinical signs, that was treated at the Universidade Estadual de Santa Cruz's Veterinary Hospital (HV-UESC). Case: A 4-year-old male intact Dogo Argentino breed dog, weighing 64 kg, presenting obesity, anorexia, prostration, walking in circles, and chronic dermatopathy was presented at HV-UESC. Upon physical examination, the animal presented a deficit of proprioception in the 4 limbs, with preserved superficial and deep pain. No alteration was observed in the ears, that could explained the clinical signs. In terms of dermatopathy, the animal presented symmetrical alopecia in the lateral region of the thighs and tail. Blood samples were collected for a complete blood count and biochemical tests of urea, creatinine, ALT, AST and cholesterol. Imaging radiography and ultrasonography were performed, which ruled out thoracic and abdominal changes that could be related to the case. Prior to receiving the blood test results, idiopathic encephalitis was suspected and enrofloxacin and prednisone were prescribed for 7 days. During the medication period, previous exams were provided, which indicated only increased cholesterol (500 mg/dL). The animal showed no improvement with the prescribed medication. In view of the clinical signs presented by the patient and the results of the additional tests, hormonal disease was suspected, compatible with hypothyroidism. Thus, hormonal tests of total T4, free T4, and TSH were requested, leading to verification of reduced total T4 (0.3 ng/dL) and free T4 (0.15 ng/dL) levels, and confirming the dysfunction of the thyroid gland. The previous treatment was suspended and thyroid hormone replacement was initiated. After 3 days of treatment, the neurological signs regressed and the animal became more active; after 30 days, the areas of alopecia decreased. Although the patient did not receive the recommended clinical follow-up for such cases, it was possible to establish the ideal levothyroxine dosage for the dog after appropriate adjustments, which permitted thyroid hormone levels to return to normal. Discussion: This report refers to a case of hypothyroidism in a giant dog breed. The dog in the report showed clinical signs of a dermatological, metabolic, and neurological nature, which is consistent with a lack of thyroid hormone. The main signs presented by the animal were neurological, such as walking in circles and a deficit of proprioception in the four limbs. These clinical signs are rarely mentioned in the literature associated with hypothyroidism. Laboratory abnormalities are correlated with the severity and chronicity of the disease. The animal showed a decrease in total T4 and free T4, which is to be expected in a hypothyroid animal. As a result, levothyroxine replacement treatment was initiated. The dose used for the dog, which is the recommended dose in the literature, greatly increased its total T4 levels. As a result, the dose was readjusted after a new clinical evaluation. The rate of metabolism and absorption of levothyroxine varies widely and is independent of weight. The patient showed quickly improvement in neurological signs, activity level, and serum cholesterol rate. Regarding dermatological signs and body condition, there was a more gradual improvement. This corroborates what is mentioned in the literature, which indicates that improvements can take many months

Hipertireoidismo é uma doença causada pela disfunção do eixo hipotálamo-hipófise-tireoide, na qual há um aumento na produção e secreção de T3 (triiodotironina) e T4 (tiroxina). É comum em gatos de meia idade a idosos, mas raro em cães, nos quais normalmente são causados por iatrogenia ou neoplasias da tireoide. O presente trabalho relata um caso de hipertireoidismo em um cão da raça golden retriever, secundário a um carcinoma folicular-compacto de tireoide. A enfermidade causa alterações em diversos sistemas, portanto é importante que o diagnóstico precoce seja firmado para o sucesso do tratamento e a avaliação do tamanho, da invasividade tumoral, da presença de metástases, bem como, das condições clínicas do paciente que são essenciais para a escolha da terapêutica.(AU)


Hyperthyroidism is a disease caused by the hypothalamic-pituitary-thyroid axis dysfunction, in which there is an increased production and secretion of T3 (triiodothyronine) and T4 (thyroxine). It is common in middle aged to old cats but is rare in dogs, in which are usually a result of iatrogenic cause or thyroid neoplasia. The present paper reports a case of hyperthyroidism in a golden retriever dog secondary to a thyroid follicular-compact carcinoma. The disease causes changes in several systems, so to achieve the treatment's success it must be performed an early diagnoses. The analyzis of the tumor size and invasion, presence of metastasis and the patient's clinical conditions are essential to the settlement of the therapeutic choice.(AU)

The objective of this study was to investigate the in vitro action of triiodothyronine (T3) on the chondrogenic differentiation of adipose tissue-derived stem cells (ASCs) of female rats, with different time periods and doses. ASCs were extracted from female Wistar rats and were cultured in chondrogenic medium with and without the presence of T3. Five groups were established: 1) ASCs without T3; and 2,3,4,5) ASCs with 0.01, 1, 100 and 1,000 nM T3, respectively). After 7, 14 and 21 days, cell morphology, chondrogenic matrix formation, and expression of Sox9, aggrecan, collagen II, and collagen X were evaluated. The Student-Newman-Keuls test was used. ASCs showed CD54, CD73, and CD90 before chondrogenic differentiation. The hormone treatment did not alter chondrogenic matrix formation, Sox9 expression at 14 or 21 days, or expression of collagen II or collagen X at any time. However, the 0.01, 1, and 1000 nM T3 doses decreased Sox9 expression at 7 days. In conclusion, chondrogenic differentiation of ASCs of female rats is not influenced by T3.


O objetivo do presente trabalho foi verificar o efeito in vitro da triiodotironina (T3) na diferenciação condrogênica de células tronco mesenquimais do tecido adiposo (CTM-TA) de ratas, durante vários períodos e em várias doses. CTM-TA foram coletadas de ratas Wistar e cultivadas em meio condrogênico com ou sem a presença de T3. Constitui-se cinco grupos: 1) CTM-TA sem T3; e 2,3,4,5) CTM-TA com T3 (0,01; 1; 100 e 1000 nM, respectivamente). Após sete, 14 e 21 dias, foram avaliados morfologia celular, formação de matriz condrogênica e expressão de Sox9, agrecano, colágeno II e colágeno X. Para as análises foi utilizado o teste de Student Newman Keuls. CTM-TA expressaram CD54, CD73 e CD90 antes da diferenciação condrogênica. O tratamento hormonal não alterou a formação de matriz condrogênica e a expressão de Sox9 aos 14 e 21 dias e expressão dos colágenos II e X em nenhum dos períodos avaliados. No entanto, as doses de 0,01; 1 e 1000 nM T3 diminuíram a expressão de Sox9 aos 7 dias. Conclui-se que a diferenciação condrogênica de CTM-TA de ratas não é influenciada pela T3.

Resumo

Elevadas taxas de descarte de vacas leiteiras relacionadas a baixos índices reprodutivos são observadas nos rebanhos comerciais. A utilização terapêutica de hormônios no intuito de induzir a lactação em animais hígidos é uma realidade. As doses hormonais utilizadas são elevadas e há a necessidade de experimentos que comprovem a possibilidade de redução. Nesse contexto, o objetivo desse projeto piloto foi reduzir as doses hormonais em vacas leiteiras submetidas a um protocolo de indução de lactação de 0,71mg/kg de peso vivo (PV) de benzoato de estradiol para 0,005mg/kg PV. Quatro vacas leiteiras receberam tratamento hormonal por 18 dias e iniciaram a lactação induzida após esse período. As vacas apresentaram produção de 32±4,3kg no pico lactacional induzido. A resposta lactacional das vacas foi considerada satisfatória e motivadora para experimentação de maior escala. Entretanto, são necessários estudos futuros que comprovem cientificamente o que foi encontrado nesse projeto piloto.


High culling rates of dairy cows related to low reproductive indices are found in commercial herds. The therapeutic use of hormones to induce lactation in healthy animals is a reality. The hormonal doses used are high and there is a need for experiments to prove the possibility of reduction. In this context, this study aimed to reduce the hormonal doses in dairy cows submitted to a lactation induction protocol from 0.71mg/kg of body weight (BW) of estradiol benzoate to 0.005mg/kg BW. Four dairy cows received hormone treatment for 18 days and started the induced lactation after this period. The cows had a production of 32±4.3kg at the induced lactational peak. The lactational response of the cows was considered satisfactory and motivating for larger scale experimentation. However, further studies are needed to scientifically prove what was observed in this pilot project.

Resumo

This study aimed to describe a case of compact thyroid carcinoma in a dog, emphasizing the diagnosis, treatment, and clinical evolution. An 11-year-old mixed-breed male canine was attended at a private clinic, complaining of swelling in the ventral cervical region and difficulty breathing. A mass close to the thyroid gland was observed in the cervical ultrasound and a mixed tumor was identified by cytology. Based on follow-up exams, the therapeutic approach to be adopted was determined: total surgical resection of the right portion of the thyroid gland, together with the parathyroid glands, followed by chemotherapy with doxorubicin. The histopathology of the tissue removed confirmed the compact thyroid carcinoma. After thyroid removal, signs of hypothyroidism were confirmed by the hormonal dosage and the canine received hormone replacement with Levothyroxine as treatment, ceasing the symptoms. During routine follow-up, the presence of miliary pulmonary metastases was identified. The patient continued with the chemotherapy sessions, presenting a stable condition for almost 6 months after starting the treatment. However, clinical signs of dyspnea were manifested, resulting in a gradual worsening. Thus, the tutors chose to euthanize the animal after 9 months of treatment. According to the case described, thyroidectomy is an option of treatment for cases of thyroid carcinoma with wide dimensions. However, the technique's success depends on the effective and complete removal of the affected tissue due to the high frequency of metastases and the possible occurrence of secondary hypothyroidism.


Objetivou-se, neste trabalho, descrever um caso de carcinoma compacto de tireoide em cão, enfatizando o diagnóstico, o tratamento e a evolução clínica. O paciente foi atendido em uma clínica particular, sendo um canino macho, sem raça definida, de 11 anos, com queixa de edema cervical ventral e dificuldade respiratória. Pela ultrassonografia cervical, foi observada uma massa próxima à glândula tireoide e pela citologia, um tumor misto. A partir dos exames de acompanhamento, foi determinada a conduta terapêutica a ser adotada: ressecção cirúrgica total da porção direita da glândula tireoide, junto às paratireoides, seguida de quimioterapia com doxorrubicina. A histopatologia do tecido removido confirmou o carcinoma compacto da tireoide. Após a remoção da tireoide, sinais de hipotireoidismo foram confirmados pela dosagem hormonal, sendo feita reposição hormonal com Levotiroxina, cessando os sintomas. Durante o acompanhamento de rotina, foi identificada a presença de metástase pulmonar miliar. O paciente continuou suas sessões de quimioterapia, mantendo um quadro estável por quase seis meses após o início do tratamento. No entanto, sinais clínicos de dispneia foram manifestados, com piora gradativa do quadro, tendo os tutores optado pela eutanásia do animal após nove meses do início do tratamento. De acordo com o exposto, conclui-se que a tireoidectomia é um tratamento de escolha para os casos de carcinoma de tireoide com amplas dimensões. Entretanto, o sucesso da técnica depende da remoção efetiva e completa do tecido afetado, devido à alta frequência de metástases, além de poder ser acompanhada por um quadro de hipotireoidismo secundário.
