Resumo

We described a case of cerebral infarction and thrombotic meningoencephalitis due to candidiasis in a seven-month-old calf. The death occurred three days after the onset of apathy, fever, and the head's lateral deviation to the left. Macroscopic changes in the brain consisted of asymmetry of telencephalic hemispheres; the right hemisphere was enlarged, causing cerebellar herniation. A focally extensive red area was observed on the surface of the right occipital lobe. At cross-sections of the fixed brain, the lesions revealed to be extensive, red-brown, soft or cavitated areas affecting the white and grey matter from the level of the thalamus to the cerebellum and compressing subjacent structures. Histologically, there was acute, coalescent, multifocal necrosupurative meningoencephalitis, associated with vasculitis, congestion, thrombosis, edema, infarction, and intralesional fungal hyphae. The diagnosis of cerebral infarction and thrombotic meningoencephalitis due to candidiasis was made by the pathological changes, the staining and morphological characteristics of the agent, and immunohistochemistry. The cerebral infarction and thrombotic meningoencephalitis in cattle can result from vascular lesions due to infection by Candida sp.; although uncommon, this case demonstrated that candidiasis should be part of a list of differential diagnoses of severe brain injuries in cattle.


Descreve-se um caso de infarto cerebral e meningoencefalite trombótica devido a candidíase em um bezerro de sete meses de idade. A morte ocorreu três dias após o início de apatia, febre e desvio lateral da cabeça para a esquerda. As alterações macroscópicas no cérebro consistiam em assimetria dos hemisférios telencefálicos; o hemisfério direito estava aumentado, causando herniação cerebelar. Uma extensa área vermelha focal foi observada na superfície do lobo occipital direito. Nos cortes transversais do encéfalo fixado, as lesões revelaram áreas extensas, marrom-avermelhadas, moles ou cavitadas, afetando a substância branca e cinzenta desde o nível do tálamo até o cerebelo e comprimindo as estruturas subjacentes. Histologicamente, havia meningoencefalite necrossupurativa multifocal aguda, coalescente, associada a vasculite, congestão, trombose, edema, infarto e hifas fúngicas intralesionais. O diagnóstico de infarto cerebral e meningoencefalite trombótica devido a candidíase foi feito pelas alterações patológicas, coloração e características morfológicas do agente e imuno-histoquímica. O infarto cerebral e meningoencefalite trombótica em bovinos pode resultar de lesões vasculares devido à infecção por Candida sp.; embora incomum, este caso demonstra que a candidíase deve fazer parte de uma lista de diagnósticos diferenciais de lesões cerebrais graves em bovinos.

Resumo

Purpose: Remote ischemic preconditioning (RIPC) confers cardioprotection against ischemia reperfusion (IR) injury. However, the precise mechanisms involved in RIPC-induced cardioprotection are not fully explored. The present study was aimed to identify the role of melatonin in RIPC-induced late cardioprotective effects in rats and to explore the role of H2 S, TNF-α and mitoKATP in melatoninmediated effects in RIPC. Methods: Wistar rats were subjected to RIPC in which hind limb was subjected to four alternate cycles of ischemia and reperfusion of 5 min duration by using a neonatal blood pressure cuff. After 24 h of RIPC or ramelteon-induced pharmacological preconditioning, hearts were isolated and subjected to IR injury on the Langendorff apparatus. Results: RIPC and ramelteon preconditioning protected the hearts from IR injury and it was assessed by a decrease in LDH-1, cTnT and increase in left ventricular developed pressure (LVDP). RIPC increased the melatonin levels (in plasma), H2 S (in heart) and decreased TNF-α levels. The effects of RIPC were abolished in the presence of melatonin receptor blocker (luzindole), ganglionic blocker (hexamethonium) and mitochondrial KATP blocker (5-hydroxydecanoic acid). Conclusion: RIPC produce delayed cardioprotection against IR injury through the activation of neuronal pathway, which may increase the plasma melatonin levels to activate the cardioprotective signaling pathway involving the opening of mitochondrial KATP channels, decrease in TNF-α production and increase in H2 S levels. Ramelteon-induced pharmacological preconditioning may also activate the cardioprotective signaling pathway involving the opening of mitochondrial KATP channels, decrease in TNF-α production and increase in H2 S levels.

Resumo

Purpose: Myocardial ischemia/reperfusion (MI/R) injury refers to a pathological condition of treatment of myocardial infarction. Oxidative stress and inflammation are believed to be important mechanisms mediating MI/R injury. Kukoamine A (KuA), a sperm, is the main bioactive component extracted from the bark of goji berries. In this study, we wanted to investigate the possible effects of KuA on MI/R injury. Methods: In this experiment, all rats were divided into sham operation group, MI/R group, KuA 10 mg + MI/R group, KuA 20 mg + MI/R group. After 120 min of ischemia/reperfusion treatment, left ventricular systolic pressure (LVSP), left ventricular end-diastolic pressure (LVEDP), maximal rates of rising and fall of left ventricular pressure (±dp/dtmax), and ischemic area were detected. Serum samples of rats in each group were collected. The enzyme activities of catalase (CAT), glutathione peroxidase (GSH-PX), superoxide dismutase (SOD), levels of malondialdehyde (MDA), CK muscle/brain (CK-MB), tumor necrosis factor (TNF), interleukin-1ß (IL-1ß), and interleukin-6 (IL-6) were detected using enzyme-linked immunosorbent assay (ELISA). The apoptosis of myocardium in each group was detected according to the instructions of the terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay. The expressions of mammalian target of glycogen synthase kinase-3ß (GSH-3ß) and protein kinase B (Akt) mRNA level in myocardial tissues were detected via reverse transcription-polymerase chain reaction (RT-PCR). Results: MI/R rats showed a significant increase in oxidative stress and inflammation. In addition, we showed that KuA significantly improved the myocardial function such as LVSP, left ventricular ejection fraction, +dp/dt, and -dp/dt. Here, it attenuated dose-dependent histological damage in ischemia-reperfused myocardium, which is associated with the enzyme activities of SOD, GSH-PX, and levels of MDA, IL-6, TNF-α, L-1ß. Conclusions: KuA inhibited gene expression of Akt/GSK-3ß, inflammation, oxidative stress and improved MR/I injury. Taken together, our results allowed us to better understand the pharmacological activity of KuA against MR/I injury.

Resumo

Purpose: To explore the mechanism of jatrorrhizine on apoptosis and fibrosis induced by myocardial infarction (MI) in an animal model. Methods: The left anterior descending branch of coronary artery was surgically ligated to duplicate the mouse model of MI. The sham and infarcted mice were treated with normal saline once a day, while mice in experimental groups received low-dose (LD) and high-dose (HD) jatrorrhizine once a day respectively. Two weeks later, cardiac function was detected by echocardiography, and histopathological examination was performed using hematoxylin and eosin (H&E) and Masson staining. The expressions of p53, TGF-ß1, Smad/2/3, Bax, Bcl-2, collagen I and collagen III were quantified using qRT-PCR and western blot assays. Results: Jatrorrhizine significantly improved left ventricular ejection fraction (LVEF) and left ventricle end-systolic (LVES) in mice. Histopathological, administration of jatrorrhizine weakened infiltration of inflammatory cells and cardiac fibrosis in myocardium of mice caused by MI. Additionally, jatrorrhizine suppressed cardiomyocyte apoptosis exhibited as its capability to reverse changes of Bax and Bcl-2 levels in myocardium caused by MI. Jatrorrhizine statistically significantly downregulated expression of collagen I and collagen III, as well as TGF-ß1, Smad2/3 and p53. Conclusions: Jatrorrhizine reduce cardiomyocyte apoptosis and fibrosis through inhibiting p53/Bax/Bcl-2 and TGF-ß1/Smad2/3 signaling pathways.

Background: Anaplastic carcinoma of the mammary gland is considered a highly invasive and malignant neoplasia, whichusually leads to neoplastic thrombosis and epidermal ulceration due to invasiveness of lymphatic vessels by tumor emboli.Loss of epidermal barrier predisposes to secondary bacterial infection, mainly by commensal bacteria of the skin, andsepticemia can be observed in cases of systemic dissemination. Neoplastic emboli and septic thrombi can lead to vascularobstruction, which can predispose to infarction in multiple organs. The aim of this case report is to describe a case ofhemorrhagic stroke in a dog with anaplastic carcinoma of the mammary gland.Case: A 12-year-old, mixed breed, female spayed canine presented with mammary tumor with plaque formation, associatedwith skin ulceration which extended to the medial region of pelvic limbs. The animal also displayed neurologic clinicalsigns characterized by stupor, opisthotonos and apathy. Due to poor prognosis, the canine was euthanized and submittedto necropsy. During external examination, the animal was in a good body condition, had moderately pale mucosae, andit was noted a nodular plaque in the abdominal ventral area extending from thoracic to inguinal mammary gland. Themass was firm and white, extensively ulcerated, and extending from the thoracic to inguinal mammary gland. There wasa well circumscribed area of infarction in the spleen and kidney. In the cerebral cortex it was observed an extensive areaof hemorrhage with adhered blood clot from frontal to temporal lobe. There were multiple well circumscribed, firm, andwhite areas in the myocardium. Histologically, the neoplastic proliferation was composed by epithelial cells with high pleomorphism, features of malignancy, and no delimitations, leading to the diagnosis of anaplastic carcinoma of the mammarygland. Admixed with tumor cells there were multiple areas of hemorrhage, necrosis and thrombosis...

A doença renal é a enfermidade mais frequentemente diagnosticada em animais de companhia e a segunda causa de morte em gatos, sendo o distúrbio mais comum envolvendo os rins de gatos e cães. O diagnóstico é realizado com base no histórico do animal, sinais clínicos associados à exames séricos e de imagem. A complexidade em se realizar o diagnóstico precoce reside na ausência de sinais clínicos nos estágios iniciais da doença. Geralmente, o diagnóstico ocorre em fases mais avançadas da doença renal em que, devido à elevada perda da função renal, culmina no aparecimento dos sinais clínicos. Neste relato de caso, dois gatos sem raça definida foram atendidos em um hospital veterinário particular em Fortaleza/CE para realização da consulta de rotina e, por meio da dos achados observados através dos exames complementares - bioquímica sérica, urinálise e ultrassonografia - foi possível realizar precocemente o diagnóstico de doença renal crônica.


Kidney disease is the most frequently diagnosed disease in companion animals and the second leading cause of death in cats, being the most common disorder involving the kidneys of cats and dogs. The diagnosis is made based on the animal's history, clinical signs associated with serum and imaging tests. The complexity in performing the early diagnosis is in the absence of clinical signs at the initial stages of the disease. Generally, the diagnosis occurs in more advanced stages of renal disease in which, due to high loss of renal function, culminates in the appearance of clinical signs. In this case report, two undefined cats were treated at a private veterinary hospital in Fortaleza / CE for a routine visit and, through the findings observed through complementary tests - serum biochemistry, urinalysis and ultrasonography - it was possible early diagnosis of chronic kidney disease.

Resumo

Purpose: To investigate the safety and clinical, hemodynamic and tissue improvement ability in mini pigs undergoing cell and gene therapy for the treatment of acute myocardial infarction. Methods: Thirty-two mini pigs Br1 lineage, 12 months old, undergoing induction of acute myocardial infarction by occlusion of the diagonal branch of the paraconal coronary. They were divided into 4 groups: one control group and 3 treatment groups (cell therapy and gene cell therapy). Echocardiography reviews were performed on three occasions and histopathological analysis was performed after 4 weeks. Analysis of variance (ANOVA), Tukey and Wilcoxon tests, were performed. Results: Association of vascular endothelial growth factor (VEGF) with angiopoietin1 (Ang1) presented satisfactory results in the improvement of ventricular function following ischemic cardiomyopathy in mini pigs when compared to the results of the other treated groups. Conclusion: The therapy with VEGF and the combination of VEGF with Ang1, promoted recovered function of the myocardium, characterized by reduced akinetic area and induction of neovascularization.(AU)

Purpose: To evaluate in vivo animal model of cardiac ischemia/reperfusion the cardioprotective activity of pancreatic lipase inhibitor of the orlistat. Methods: Adult male Wistar rats were anesthetized, placed on mechanical ventilation and underwent surgery to induce cardiac I/R by obstructing left descending coronary artery followed by reperfusion to evaluation of ventricular arrhythmias (VA), atrioventricular block (AVB) and lethality (LET) with pancreatic lipase inhibitor orlistat (ORL). At the end of reperfusion, blood samples were collected for determination of triglycerides (TG), very low-density lipoprotein (VLDL), low-density lipoprotein (LDL), high-density lipoprotein (HDL), lactate dehydrogenase (LDH), creatine kinase (CK), and creatine kinase-MB (CK-MB). Results: Treatment with ORL has been able to decrease the incidence of VA, AVB and LET. Besides that, treatment with ORL reduced serum concentrations of CK and LDL, but did not alter the levels of serum concentration of TG, VLDL and HDL. Conclusion: The reduction of ventricular arrhythmias, atrioventricular block, and lethality and serum levels of creatine kinase produced by treatment with orlistat in animal model of cardiac isquemia/reperfusion injury suggest that ORL could be used as an efficient cardioprotective therapeutic strategy to attenuate myocardial damage related to acute myocardial infarction.(AU)

Background: Hypothyroidism in dogs can cause lethargy, weight gain, dermatological signs and, uncommonly, neurological signs. These may involve the peripheral or central nervous system, leading to a decreased level of consciousness, central or peripheral vestibular disease, epileptic seizures, cognitive dysfunction, facial nerve paralysis, laryngeal paralysis, and polyneuropathy. There are few cases reported in the literature relating hypothyroidism and central vestibular disease. The aim of this study was to report a case of a stroke secondary to hypothyroidism that resulted in central vestibular syndrome in a dog. Case: A 13-year-old female Pitbull, weighting 37 kg, was referred to a Veterinary University Hospital with a two weeks history of dyspnea, coughing and walking difficulty. On neurological examination, the animal presented vertical nystagmus, somnolence, non-ambulatory tetraparesis, decreased postural reaction in all limbs and normal spinal reflexes. The laboratory exams revealed a mild leukocytosis and increased creatinine, glucose, cholesterol and triglycerides levels. Due to historical, clinical, neurological and laboratory tests findings, the animal was diagnosed with central vestibular syndrome secondary to an intracranial neoplasm. Clinical treatment was instituted with corticosteroids, however the animal did not respond and died. On necropsy, the heart was enlarged and coronary arteries were thicker. No macroscopic changes were found in the brain. On histological analysis, the coronary arteries were congest by lipids that almost occluded the arterial lumen. The same alteration were found in splenic and renal arterioles, and in the brain’s leptomeninges. Also in the brain, it was observed perivascular infiltration of mononuclear cells and focal gliosis. The thyroid was atrophied and fibrosed bilaterally. Therefore, those changes were compatible with atherosclerosis secondary to hypothyroidism...

Resumo

ABSTRACT: Monosodium methanearsonate (MSMA) is an organic form of arsenic present in the formulations of some herbicides. Accidental ingestion of pasture contaminated with arsenic may lead to toxicosis in cattle. Almost 200 head of cattle maintained in an area sprayed with MSMA presented with intense diarrhea and dehydration after grazing. Subsequently, 16 of these animals died. Toxic levels of arsenic (>1.5g/g) were detected in the kidney, liver, urine, and skeletal muscle of 6 animals. At gross inspection were observed multifocal to coalescent ulcers in the mucosa from on the forestomachs associated with hemorrhagic areas and marked wall edema. Microscopic examination mainly showed fibrinoid necrosis of vessels with multifocal thrombosis associated with ischemic infarction that were characterized by large transmural necrotic areas in the forestomachs. The clinical and pathological changes interestingly showed that this form of arsenic although considered less toxic, has caused severe vascular injury in forestomachs of cattle.


RESUMO: Metano-arseniato ácido monossódico (MSMA) é uma forma orgânica de arsênio, presente nas formulações de alguns herbicidas. A ingestão acidental de pasto contaminado por arsênio pode levar a toxicose em bovinos. Aproximadamente 200 bovinos que estavam em uma pastagem pulverizada com MSMA manifestaram intensa diarreia e desidratação após o pastejo. Subsequentemente, 16 animais morreram. Níveis tóxicos de arsênio (>1.5g/g) foram detectados no rim, fígado, urina e músculo esquelético de 6 animais. A inspeção macroscópica revelou úlceras multifocais a coalescentes na mucosa de pré-estômagos, adjacentes a focos de hemorragia e intenso edema de parede. A avaliação microscópica revelou, predominantemente, necrose fibrinoide de vasos com trombose multifocal associada a infarto, caracterizado por grandes áreas de necrose transmural em pré-estômagos. As alterações clínicas e patológicas, interessantemente, demonstraram que esta forma de arsênio, apesar de ser considerada menos tóxica, causou severa injúria vascular em pré-estômagos de bovinos.

Background: Fungi of the genus Aspergillus are ubiquitous microorganisms of opportunistic character, of which animals and humans are continually exposed. Are described three main forms of aspergillosis in dogs: nasal, disseminated and bronchopulmonary. This form which is restricted to the lung parenchyma is considered rare in dogs. The aim of this paper is to report a case of pulmonary aspergillosis associated to acute myocardial infarction in a dog, focusing on epidemiological, clinical, pathological and immunohistochemical characteristics. Case: A male dog, 4-year-old, Pinscher, clinically presented dyspnea, anorexia, vomiting, concentrated urine and dark and mollified faeces. On physical examination, there were congested oral and conjunctival mucosas, dehydration and petechiae in the outer ear. The animal died five days after the clinical attendance and was sent for necropsy. The macroscopic lesions were restricted to the lung, heart and liver. There were multifocal areas of consolidation in the lung parenchyma and yellowish content in the trachea and bronchi lumen. The heart was rounded and with pale areas in the myocardium. Liver was discreet accentuation of the lobular pattern. Histologically the lesion was characterized by pyogranulomatous and necrotizing bronchopneumonia, multifocal to coalescing, moderate, associated with myriads of intralesional hyphae consistent with [...]

PURPOSE:To investigate the myocardial ischemia-reperfusion with sevoflurane anesthetic preconditioning (APC) would present beneficial effects on autonomic and cardiac function indexes after the acute phase of a myocardial ischemia-reperfusion.METHODS:Twenty Wistar rats were allocated in three groups: control (CON, n=10), myocardial infarction with sevoflurane (SEV, n=5) and infarcted without sevoflurane (INF, n=5). Myocardial ischemia (60 min) and reperfusion were performed by temporary coronary occlusion. Twenty-one days later, the systolic and diastolic function were evaluated by echocardiography; spectral analysis of the systolic arterial pressure (SAPV) and heart rate variability (HRV) were assessed. After the recording period, the infarct size (IS) was evaluated.RESULTS:The INF group presented greater cardiac dysfunction and increased sympathetic modulation of the SAPV, as well as decreased alpha index and worse vagal modulation of the HRV. The SEV group exhibited attenuation of the systolic and diastolic dysfunction and preserved vagal modulation (square root of the mean squared differences of successive R-R intervals and high frequency) of HRV, as well as a smaller IS.CONCLUSION:Sevoflurane preconditioning better preserved the cardiac function and autonomic modulation of the heart in post-acute myocardial infarction period.(AU)

PURPOSE:To investigate the effects of Borage oil on cardiac remodeling after myocardial infarction (MI).METHODS:Male Wistar rats underwent ligation of the left coronary artery and divided into three groups: MI (control), BO-18 (18 mg/kg of borage oil) and BO-180 (180 mg/kg of borage oil). After seven days, heart was arrested in diastole and processed for histological evaluation of: MI size, LV dilation, myocyte hypertrophy, inflammatory infiltration and fibrosis in MI region and in remote region. The relative weight of the lung was used as a marker of heart failure. The MI size was comparable among groups.RESULTS:Compared to control, BO treated groups showed lower weight of heart and lungs, reduced LV dilation and myocyte hypertrophy. Hemodynamic measurements were comparable. The treatment attenuated the inflammatory infiltration and fibrosis in remote myocardium.CONCLUSION:Borage oil attenuates progression of cardiac remodeling after myocardial infarction and congestive heart failure.(AU)

The spleen of dogs is frequently affected by disorders that vary from local and systemic origin. The difficulty in associating clinical and gross findings contributes for the choice of total splenectomy as the main treatment, leading to an impairment of the immune and hematopoietic functions. The aim of this study was to evaluate the pathological findings in the spleen of splenectomized dogs during 2008 to 2014 at a Veterinary Teaching Hospital. From the 71 cases analyzed, 97% (69/71) of the dogs were submitted to total splenectomy and 3% (2/71) to partial splenectomy. In 45 (63.4%) of these cases, the histopathological diagnosis was non-neoplastic alterations; only 36.6% (26/71) had a splenic neoplasia. The main non-neoplastic lesions observed were nodular hyperplasia 24.4% (11/45), infarction 22.3% (10/45), and hematoma 20% (9/45). The most frequent tumors were hemangiosarcoma 50% (13/26), histiocytic sarcoma 23% (6/26), and lymphoma 11.5% (3/26). The clinical methods used to diagnose splenic lesions were ultrasonography 88% (63/71), radiography 2.8% (2/71) and exploratory laparotomy 4.2% (3/71). In 4.2% (3/71) the spleen changes were observed during the therapeutic ovariohysterectomy. The results of the present study showed a prevalence of benign disorders in the spleen of splenectomized dogs associated with a high incidence of total splenectomy performed, indicating a difficulty in recognizing the different lesions that can affect the spleen by theveterinarian medical.


O baço de cães é frequentemente afetado por alterações locais ou de origem sistêmica. A dificuldade em associar achados clínicos e macroscópicos contribuem para a escolha da esplenectomia total com principal tratamento, levando a um comprometimento das funções imune e hematopoiética. O principal objetivo deste estudo foi avaliar os achados patológicos no baço de cães esplenectomizados durante 2008 a 2014 em um Hospital Escola Veterinário. Dos 71 casos analisados, 97% (69/71) dos cães foram submetidos à esplenectomia total e 3% (2/71) à esplenectomia parcial. Em 45 (63.4%) casos, o diagnóstico histopatológico foi de alterações não-neoplásicas; somente 36,6% (26/71) tinham neoplasia esplênica. As principais alterações não-neoplásicas observadas foram hiperplasia nodular 24,4% (11/45), infarto 22,3% (10/45) e hematoma 20% (9/45). Os tumores mais frequentes foram hemangiossarcoma 50% (13/26), sarcoma histiocítico 23% (6/26) e linfoma esplênico 11.5% (3/26). Os métodos clínicos utilizados para diagnóstico foram ultrassonografia 88% (63/71), radiografia 2.8% (2/71) e laparatomia exploratória. Em 4,2% (3/71), as alterações esplênicas foram observadas durante ovariohisterectomia terapêutica. Os resultados do presente estudo evidenciam uma prevalência de alterações benignas no baço de cães esplenectomizados, associada com uma alta incidência de esplenectomias totais realizadas, indicam uma dificuldade dos médicos veterinários em reconhecer asdiferentes lesões que podem acometer o baço.
