Resumo

A criação comercial de gato doméstico tem aumentado assim como a realização de estudos para melhorar a eficiência reprodutiva é uma realidade. O interesse pela identificação das falhas reprodutivas tem crescido, entretanto, os casos de infertilidade nos machos são escassos na literatura disponível. O histórico de infertilidade de um gato deve ser investigado cuidadosamente, pois pode estar associado a mudanças no manejo ou doenças. O diagnóstico dessas 'falhas reprodutivas' geralmente é simples, se realizada uma boa anamnese, exames, físico e andrológico, minuciosos, assim como exames complementares, entre eles hemograma completo, perfil bioquímico e testes para doenças infecciosas como FIV, FELV e FIP.(AU)


Commercial breeding of domestic cats has increased, and studies to improve reproductive efficiency are a reality. Interest in identifying reproductive failures has increased, however, cases of infertility in men are scarce in the available literature. A cat's history of infertility should be investigated carefully as it may be associated with changes in management or illness. The diagnosis of these 'reproductive failures' is usually simple if a good anamnesis, thorough physical and andrological examinations are carried out, in addition to complementary tests, including complete blood count, biochemical profile, tests for infectious diseases such as FIV, FELV, FIP.(AU)

Resumo

Background: Dermatophytes, fungi of universal distribution, invade semi or fully keratinized structures, such as skin, fur/ hair and nails. The various species of dermatophytes are classified into three genera anamorphic: Microsporum, Trichophyton and Epidermophyton. The genus Epidermophyton includes only E. floccosum, that rarely affects animals. The main species responsible for the disease in dogs and cats are Microsporum canis, M. gypseum and Trichophyton mentagrophytes, which were characterized through conventional mycological methodology (microscopic examination with KOH and culture). Molecular methodologies, such as real-time PCR, can contribute to a rapid laboratory diagnosis, helping clinicians to initiate an early antifungal treatment. This case report describes a case of canine dermatophytosis due to Trichophyton mentagrophytes detected from a clinical sample by SYBR-Green real-time PCR. Case: A 8-year-old dog, rescued from the street, was referred to a private veterinary clinic in the city of Canoas, RS, Brazil, presenting generalized lymphadenomegaly, crusted lesions all over the body, generalized alopecia, signs of excoriation and epistaxis. Initially, were administered prednisone [1 mg/kg every 48 h, BID] and cephalexin [30 mg/kg, BID]. Weekly baths with benzoyl peroxide were also given. The therapy was not clinically successful. Wood's Lamp Test was negative. As a differential diagnosis, PCR for detection of Leishmania was negative. Complete blood count and serum biochemical assay were also performed. For mycological diagnosis, hair specimen was clarified and examined microscopically using 10% potassium hydroxide (KOH) for the visualization of chains of arthroconidia (ectothrix invasion of hair). The infected hair was plated onto MycoselTM Agar, incubated at 28°C for 15 days. Microscopy of hyphae/ conidia and macroscopic colony characteristics (colors and texture) were conducted for the differentiation of the species within the genus Microsporum and Trichophyton. In addition, real-time PCR was applied for direct analysis of the fungal DNA obtained from the hair sample. Microscopic examination was negative. The dermatophyte present in the hair sample was confirmed as Trichophyton mentagrophytes by culture and qPCR (melting-point analysis). The patient was treated with systemic itraconazole [10 mg/ kg SID - 90 days]. Twice-weekly application of 2.5 % miconazole and 2% chlorhexidine shampoo until complete cure. Discussion: Dermatophytosis is often listed as self-limiting infection; however, animal dermatophytosis can spread between pets, as well as a zoonotic transmission to humans. The literature on dermatophytosis indicates that Microsporum canis is the predominant etiological agent, followed by M. gypseum. Trichophyon mentagrophytes that appear in a lower percentage of isolation. The culture of hair, even with specific medium containing chloramphenicol and cyclohexamide, may present contaminating fungi, not related to dermatophytosis, which can inhibit or override the growth of dermatophytes. The use of real-time PCR provided a faster and specific diagnosis of dermatophytosis when compared to the conventional mycological methodology for detection and identification of T. mentagrophytes, which takes around 10 to 15 days for culture. It is possible to use this technique as an alternative diagnosis for dermatophytes associated to clinical hair samples of dogs.

Resumo

Purpose: To evaluate the in vivo efficiency of commercial polymeric membranes for guided bone regeneration. Methods: Rat calvarial critical size defects was treated with LuminaCoat (LC), Surgitime PTFE (SP), GenDerm (GD), Pratix (PR), Techgraft (TG) or control (C-) and histomorphometric analysis determined the percentage of new bone, connective tissue and biomaterial at 1 or 3 months. Statistical analysis used ANOVA with Tukey's post-test for means at same experimental time and the paired Student's t test between the two periods, considering p < 0.05. Results: New bone at 1 month was higher for SP, TG and C-, at 3 months there were no differences, and between 1 and 3 months PR had greater increase growthing. Connective tissue at 1 month was higher for C-, at 3 months for PR, TG and C-, and between 1 and 3 months C- had sharp decline. Biomaterial at 1 month was higher for LC, in 3 months for SP and TG, and between 1 and 3 months, LC, GD and TG had more decreasing mean. Conclusion: SP had greater osteopromotive capacity and limitation of connective ingrowth, but did not exhibit degradation. PR and TG had favorable osteopromotion, LC less connective tissue and GD more accelerated biodegradation.

Abstract The -lactam/lactamase inhibitors (BLBLIs) combination drugs are considered an effective alternative to carbapenems. However, there is a growing concern that the increased use of BLBLIs may lead to increased resistance. This study determined the temporal association between the consumption of BLBLI and the antimicrobial resistance in Gram-negative bacteria. In this retrospective study, electronic data on the Gram-negative bacterial isolates, including A. baumannii, P. aeruginosa, E. coli, and K. pneumoniae from in-patients and susceptibility testing results were retrieved from the medical records of the clinical laboratory. A linear regression and cross-correlation analysis were performed on the acquired data. Increasing trends (p 0.05) in the consumption of BIBLI and carbapenem with a median use of 27.68 and 34.46 DDD/1000 PD per quarter were observed, respectively. A decreased trend (p=0.023) in the consumption of fluoroquinolones with a median use of 29.13 DDD/1000 PD per quarter was observed. The resistance rate of K. pneumoniae was synchronized with the BIBLI and carbapenem consumptions with a correlation coefficient of 0.893 (p=0.012) and 0.951 (p=0.016), respectively. The cross-correlation analysis against the consumption of BIBLI and meropenem resistant K. pneumoniae was peaked at 0-quarter lag (r=951, p=0.016). There was an increasing trend in the consumption of BLBLI and carbapenems. The increasing trend in the rates of resistance to piperacillin/tazobactam, in line with the increasing consumption of BLBLI, suggests that BLBLI has to be used with caution and cannot be directly considered as a long-term alternative to carbapenems.

Uma regressão linear e análise de correlação cruzada foram realizadas nos dados adquiridos. Foram observadas tendências crescentes (p 0,05) no consumo de BIBLI e carbapenem com uma mediana de uso de 27,68 e 34,46 DDD/1000 PD por trimestre, respectivamente. Foi observada uma tendência de diminuição (p = 0,023) no consumo de fluoroquinolonas com uma mediana de uso de 29,13 DDD/1000 PD por trimestre. Resumo

Purpose: Bone repair aims to restore the anatomical, biomechanical, and functional integrity of the affected structure. Here we study the effects of ascorbic acid (AA) and epidermal growth factor (EGF) applied in a single dose and in combination on the repair of a noncritical bone defect model. Methods: Twenty-four rats were divided into four groups: an intact G-1 control group, and three groups that underwent a noncritical bone defect in the right tibia: G-2 treated with AA, G-3 treated with EGF, and G-4 treated with AA in combination with EGF. After 21 days of treatment, rats were sacrificed, the tibias were dissected and a destructive biomechanical analysis of three-point flexion test was performed in a universal testing machine; the values of stiffness, resistance, maximum energy, and energy at maximum load were statistically compared. Results: G-3 and G-4 recovered the biomechanical properties of strength and stiffness of an intact tibia 3 weeks after their application. Not so the energy and energy at maximum load. For G-2, only the stiffness of an intact tibia was recovered. Conclusion: EGF and AA-EGF applied to a noncritical bone defect in the rat tibia favors the recovery of bone resistance and stiffness.

Resumo

This study verified the in vivo biocompatibility of bovine elastic cartilage decellularized with alkaline solution in relation to the non-decellularized cartilage implanted in rats. Fifty Wistar rats were divided into two groups, with the experimental group (EG) receiving subcutaneous implants of cartilage treated in alkaline solution and the control group (CG) receiving subcutaneous implants of untreated cartilage. In both groups, the implants were removed on days 3, 7, 14, 21 and 42 with the preparation of histological slides stained with Hematoxylin and Eosin for the quantification of inflammatory cells, fibroblasts, angiogenesis, and cartilage degradation. The results showed that EG presented a less intense inflammatory infiltrate and better organization of collagen fibers compared to CG. It was concluded that the alkaline treatment provided better biocompatibility for elastic cartilage when implanted subcutaneously in rats.


Este estudo verificou a biocompatibilidade in vivo de cartilagem elástica bovina descelularizada com solução alcalina em relação à cartilagem não descelularizada implantada em ratos. Cinquenta ratos Wistar foram divididos em dois grupos; o grupo experimental (GE) recebeu implantes subcutâneos de cartilagem tratada em solução alcalina, e o grupo controle (GC) recebeu implantes subcutâneos de cartilagem não tratada. Em ambos os grupos, os implantes foram retirados nos dias três, sete, 14, 21 e 42, com a preparação de lâminas histológicas coradas com hematoxilina e eosina para a quantificação de células inflamatórias, de fibroblastos, de angiogênese e para a degradação da cartilagem. Os resultados mostraram que o GE apresentou infiltrado inflamatório menos intenso e melhor organização das fibras de colágeno em relação ao GC. Concluiu-se que o tratamento alcalino proporcionou melhor biocompatibilidade para a cartilagem elástica quando implantado por via subcutânea em ratos.

The objective of this study was to assess a training protocol employed in the Brazilian Northeast region for fitness conditioning of vaquejada horses. For 12 months, 24 Quarter Horses were evaluated under a completely randomized split-plot experimental design in which the plots comprised three age groups: horses at two, three, and four years of age. The split plots were made up of six fitness tests carried out every other month. The fitness test protocol consisted of five levels of protocol exercises on a standard vaquejada track. Prior to the tests with fasted animals, we collected blood samples to determine muscle enzymes (aspartate aminotransferase, creatine kinase, and lactate dehydrogenase). During the tests, heart rate, speed, and distance run were recorded using a heart rate monitor. Next, the results were used to calculate speed at which each horse reached 150 bpm (V150), speed at which each horse reached 200 bpm (V200), maximum heart rate (HRmax), maximum speed (Vmax), recovery time needed for the heart rate of horses to return to half the maximum value reached during the fitness tests (HR50%), and recovery time needed for the heart rate of horses to return to baseline values (HRbasal). No difference was found among the age groups for V150, HRmax, Vmax, HR50%, HRbasal, or muscle enzymes. By the final stage of training, the V200 of the three-year-old horses was higher than that of the four-year-old foals. During training, all groups exhibited increases in serum concentrations of muscle enzymes and reductions in efficiency to recover heart rate after exercise. The training protocol assessed is unable to maintain proper fitness for competitions throughout the year.(AU)

Oeceoclades maculata is a terrestrial orchid species that has potential for commercial purposes. Taking that into consideration, the present investigation aimed at studying its germination and initial development in vitro as well as its acclimatization. The influence of Murashige and Skoog (MS), Knudson C (KC), and Vacin and Went (VW) media in the presence and absence of 0.3% activated charcoal on in vitro germination and protocorm development were investigated. The effects of different concentrations of BA in combination with 0.5 mg L-1 NAA on seedling multiplication and growth were evaluated. The possibility of using dark-grown stem segments for micropropagation and acclimatization under laboratory and field conditions was also assessed. The results indicated that the most adequate media for germination were full-strength MS enriched with activated charcoal or KC supplemented with 1.5 mg L-1 BA in combination with 0.5 mg L-1 NAA. In terms of protocorm development, KC supplemented with 1.5 mg L-1 BA alone or in combination with 0.5 mg L-1 NAA provided the best results. The addition of 1.5 mg L-1 BA in combination with 0.5 mg L-1 NAA to KC medium favored the best results for seedling multiplication and development. The use of dark-grown stem segments is a viable alternative for the micropropagation of O. maculata. Regarding acclimatization, 100% survival of plants was observed during the initial phases and under field conditions average survival was 53.33%.(AU)

O exame clínico andrológico é a ferramenta de eleição utilizada para avaliar a fertilidade de um reprodutor. Normalmente utilizado em tourinhos jovens na pré-comercialização, como uma forma de garantia reprodutiva; no período pré-estação de monta; quando existem falhas ou desordens reprodutivas nos rebanhos; prévio ao ingresso nas centrais de inseminação; na determinação da puberdade, dentre outras. O exame clínico andrológico deve ser realizado por profissional com experiência suficiente para tomada de decisões sobre o real potencial reprodutivo dos animais, porém, em muitas ocasiões mesmo um profissional capacitado defronta-se com dúvidas, entre elas destacam-se as questões relacionadas ao valor cobrado e "tipo de exame"; a biometria testicular; exigências relacionadas a impotencia coeundi; e o que é necessário no certificado do exame. Buscou-se elencar, explanar e discutir essas questões em vista de diminuí-las e/ou encontrar possíveis soluções. O exame clínico andrológico é uma etapa fundamental e complexa para a seleção de reprodutores, e apesar de constantemente debatida, ainda permanecem lacunas a serem respondidas, visto que os profissionais necessitam de constante atualização.(AU)


The clinical andrological examination is the tool of choice used to assess the fertility of a sire. Usually used in young bulls in pre-marketing, as a form of reproductive guarantee; in the pre-mounting season period; when there are reproductive failures/disorders in herds; before entering insemination centers, in determining puberty, among others. The clinical andrological examination must be carried out by a professional with sufficient expertise to make decisions about the real reproductive potential of these animals, however, on many occasions even a trained professional is faced with doubts, among them the issues related to the amount charged and "type of exam"; testicular biometry; requirements related to impotencia coeundi; and what is required in the exam certificate. Based on the above context, we sought to list some possible doubts, explaining them and discussing them among veterinarians, so that such issues could be reduced and/or possible alternative solutions could be defined. The clinical andrological examination is a fundamental and complex step for the selection of sires, and despite being constantly debated, there are still gaps to be answered, since professionals need constant updating.(AU)

Purpose: Various postoperative protocols have been proposed to improve outcomes and accelerate nerve regeneration. Recently, the use of physical exercise in a post-surgical neurorraphy procedure has shown good results when started early. We experimentally investigated the hypothesis that post-operative exercise speeds up results and improves clinical and morphologic parameters. Methods: Isogenic rats were randomly divided into four groups: 1 SHAM; 2 SHAM submitted to the exercise protocol (EP); 3 Grafting of the sciatic nerve; and 4 Grafting of the sciatic nerve associated with the EP. The EP was based on aerobic activities with a treadmill, with a progressive increase in time and intensity during 6 weeks. The results were evaluated by the sciatic functional index (SFI), morphometric and morphologic analysis of nerve distal to the lesion, and the number of spinal cord motor neurons, positive to the marker Fluoro-Gold (FG), captured retrogradely through neurorraphy. Results: Functional analysis (SFI) did not show a statistical difference between the group grafted with (­50.94) and without exercise (-65.79) after 90 days. The motoneurons count (Spinal cord histology) also showed no diference between these groups (834.5 × 833 respectively). Although functionally there is no difference between these groups, morphometric study showed a greater density (53.62) and larger fibers (7.762) in GRAFT group. When comparing both operated groups with both SHAM groups, all values were much lower. Conclusions: The experimental model that this aerobic treadmill exercises protocol did not modify nerve regeneration after sciatic nerve injury and repair with nerve graft.

Purpose: To explore the mechanism and investigate the protective effect of hydroxysafflor yellow A (HSYA) on renal oxidative stress, which cyclosporine A (CsA) induces. Methods: HK-2 cells were treated with CsA to get CsA-induced oxidative stress. The effects on oxidative stress and apoptosis of HK-2 cells were detected. The contents of SOD, MDA, GSH-Px, ROS, and CAT in serum were measured, and the expression of apoptosis-related proteins was detected by western blot. Then, established the renal oxidative stress injury rats to verify the efficacy of HSYA. Results: HSYA could reduce the ROS and MDA contents induced by CsA. Compared with the CsA group, the activities of SOD, CAT, and GSH-Px increased significantly when treated with HSYA. HSYA could inhibit CsA-induced apoptosis in HK-2 cells, and promote the protein of Bcl-2 and inhibit the expression of Bax. Animal experiments showed that HSYA could reduce CsA-induced renal cell injury by reducing glomerular cell vacuoles and inflammatory factors in tissues. It also decreased serum creatinine (Crea) and blood urea nitrogen, increased Crea clearance significantly. Conclusions: HSYA could significantly improve the antioxidant capacity of the kidney cells and inhibit cell apoptosis, thereby effectively ameliorating CsA-induced oxidative stress in vitro and in vivo.

Purpose: Poultry by-products can contribute as an innovative natural source for the development of composites based on polymers and minerals aiming at bone regeneration. The objective of this study was the physicochemical and biological characterization of collagen-based hydrogels crosslinked with ultraviolet (UV)-riboflavin. Methods: Pure hydrogels of 100% collagen (G1) or hybrid hydrogels, 90% collagen:10% apatite (G2), 90% collagen:10% nanokeratin (G3), and 90% collagen:5% apatite:5% nanokeratin (G4) were characterized by scanning electron microscope, Fourier-transform infrared spectroscopy, differential scanning calorimetry, swelling degree and quali-quantitative histological analysis. Ectopic implantation in subcutaneous tissue in mice at one, three and nine weeks allowed to assess the inflammation (neutrophils, lymphocytes, macrophages, and giant cells) and repair (neovascularization, and connective tissue) to determine biocompatibility and the integrity of biomaterials to score their biodegradability. Histomorphometry on critical size defects in rat calvaria at one and three months evaluated the percentage of bone, connective tissue, and biomaterials in all groups. Results: The hydrogels presented porous microstructure, water absorption and physicochemical characteristics compatible with their polymeric and/or mineral composition. All materials exhibited biocompatibility, biodegradability, and low osteoconductivity. G2 showed greater density of new bone and biomaterial than the G1, G3 and G4. Conclusions: The collagen-apatite group formulation suggests potential for development as osteopromoting membrane.

Abscisic acid (ABA) is associated with bud dormancy, leaf abscission, and germplasm growth inhibition in in vitro conservation. We evaluated the effects of ABA in four wild Manihot accessions and one cassava accession (M. esculenta Crantz) to refine in vitro conservation methods for these species. The experiment was performed at the Laboratory for Tissue Culture from Embrapa, Cruz das Almas, Bahia State, Brazil. The statistical design was completely random in a 5 × 5 factorial scheme [(5 ABA dosages (0, 0.25, 0.50, 0.75, and 1 mg L-1) and 5 Manihot species (M. pseudoglaziovii, M. tristis, M. flabellifolia, M. chlorosticta, and M. esculenta)], with 15 replicates. Mini-cuttings of 1 cm were used, each inoculated in 10 mL of modified Murashige and Skoog medium, solidified with Phytagel® (2.4 g L-1) containing the respective ABA dosages. Tubes containing these mini-cuttings were placed in a germplasm conservation room with an irradiance of 30 µmol m-2 s-1, temperature of 22 ± 1°C, and photoperiod of 12 hours. Plant height (cm), the number of living and senescent leaves, shoots, and mini-cuttings (1 cm), and fresh and dry weights of shoots and roots (mg) were evaluated after 150 days. Growth reduction was prominent in M. pseudoglaziovii, M. tristis, and M. flabellifolia during the in vitro conservation period. In the present study, the addition of ABA did not promote the expected reduction in plant growth.(AU)

O trabalho propôs o desenvolvimento de plataforma informatizada para apoio ao gerenciamento da inspeção e análise laboratorial de produtos de origem animal fornecidos à tropa do Exército Brasileiro. A pesquisa aplicada corresponde à modalidade de produção tecnológica, em três fases. A fase I foi realizada por meio do levantamento dos arquivos de Laudos Fiscais dos artigos de origem animal, impressos e emitidos pelo LIAB (Laboratório de Inspeção de Alimentos e Bromatologia) do 12º Batalhão de Suprimento, durante o período de janeiro a novembro de 2021, e dos arquivos referentes aos Boletins Técnicos do Exército Brasileiro (BT30.404- EB). A fase II correspondeu ao projeto e desenvolvimento da "Plataforma Alimentar - Rastreabilidade e Confiabilidade Garantida". Toda a programação desenvolvida foi armazenada em um servidor web terceirizado e pode ser acessada pelos LIABs pelo link: www.pmppa.com.br/alimentar. A plataforma informatizada utilizou a linguagem PHP (Hypertext Preprocessor) associada às linguagens CSS (Cascading Style Sheet), HTML (Hyper Text Markup Language), JS (Java script) e bootstrap. A fase III consistiu na realização do teste de Caixa Preta. Os módulos disponíveis para inclusão das informações no sistema são: "cadastro dos artigos de origem animal"; "lançamento das análises"; "cadastro de Boletim Técnico" e "relatórios", os quais permitem atribuir ao artigo de origem animal a qualidade satisfatória ou insatisfatória para o consumo seguro da tropa, de acordo com a avaliação laboratorial das amostras. Houve, com eficácia, o desenvolvimento da plataforma, garantindo ao oficial médico veterinário uma ferramenta segura para apoio ao gerenciamento de qualidade dos produtos destinados à alimentação da tropa do Exército Brasileiro.


The study proposed the development of a computerized platform to support the inspection management and laboratory analysis of animal products supplied to the Brazilian Army troops. The applied research corresponds to the modality of technological production in three phases. Phase I consisted in survey from Fiscal Report Files of animal products, which was printed and issued by the FIBL (Food Inspection and Bromatology Laboratory) from the 12th Supply Battalion, from January to November 2021, and the Brazilian Army's Technical Bulletins (BT30.404-EB) Files. Phase II corresponded to design and development of the "Food Platform - Guaranteed Traceability and Reliability". All the developed programming stored at an outsourced web server by accessing the FIBLS through the link: www.pmppa.com.br/alimentar. The supportive computerized platform used the PHP (Hypertext Preprocessor) language associated with the CSS (Cascading Style Sheet), HTML (Hyper Text Markup Language), JS (Java script) and bootstrap. Phase III consisted in performing a Black Box testing. The available modules for information inclusion in the system are the following issues: "Animal products registration"; "Analyses release"; "Technical Bulletin registration" and "Reports", which allow attributing the satisfactory or unsatisfactory quality to the animal products providing safe consumption for the troops, according to the evaluation of laboratory samples. The platform was effectively developed, guaranteeing the veterinary officer a safe tool to support the quality management of products intended for feeding the Brazilian Army troops.

Background: Oocytes and embryos produce energy through mitochondrial oxidative phosphorylation by using oxygen. The membrane structure of the embryo is mostly composed of unsaturated fatty acids, for this reason DNA fragmentation, apoptosis, and abnormal gene expression are shaped as a result of the lipid peroxidation during culture. Oxidative stress (OS) is one of the most important problems affecting the in vitro embryo development. Antioxidant supplementation to the culture medium has been an alternative way to reduce cell damage caused by oxidative stress in in vitro embryo production systems. In this study, it was aimed to determine the effect of L-ergothioneine on blastocyst development when added to the culture medium. Materials, Methods & Results: The material of the study consisted of oocytes aspirated from the ovaries of Holstein cows which were collected from the local slaughterhouse. The ovaries were delivered to the laboratory within 2-3 h in a thermos which provided a constant temperature of 25-30o C with physiological saline (0.9%) containing antibiotics. All follicles in the 3-8 mm range on the ovaries were aspirated using 20 G needle. The collected follicle fluid was filtered through filters with a pore diameter of 70 micrometers. Cells remaining in the filter were washed with OPU medium and transferred to the petri dishes. Fluids were examined under a stereomicroscope. The cumulus-oocyte complexes were classified, and A and B quality oocytes were included to the study (A, B, C, and D quality COC). Oocytes aspirated from the ovaries and collected later on were incubated in IVM medium for 22 h. After maturation, it was taken into IVF medium, semen was added and incubated for 20-22 h. Possible zygotes to be taken to the culture stage were transferred to culture (IVC) drops with (L-ergothioneine 100 µL/mL (n:121) added and without antioxidant (control (n:124)), and kept in the incubator for 6-7 days. Evaluated on the 7th day differences in in vitro embryo production stages were evaluated with the Chi-square test. The study was run in 5 replicates each time, with at least 20 possible zygotes for per group being cultured. It was determined that 262 (87.33%) of a total of 300 oocytes undergoing in vitro maturation were matured. It was determined that 245 of the mature oocytes were fertilized (93.51%). The cleavage rates of the groups were determined as 87.60% and 86.29%, respectively. Eighty-two (33.47%) blastocysts were obtained from 245 zygotes taken into the culture stage, and the blastocyst rates in the groups were found to be 40.50% and 26.61%, respectively. After the study, it was determined that the statistical difference between L-ergothioneine and control in cleavage rates was insignificant (P > 0.05) and blastocyst rates was significant (P < 0.05) Discussion: Oxygen content above normal ratios can increase the formation of reactive oxygen species (ROS), particularly hydrogen peroxide (H2 O2 ), hydroxyl radical (HO·), and peroxyl radicals (ROO·). The increased rate of ROS negatively affects the success of IVP in mammalian embryos. It was observed that L-ergothioneine, which has high antioxidant activity, improved blastocyst development rates, and higher blastocyst rates could be achieved compared to the control group. By investigating the use of L-ergothioneine in different doses, it was thought that the dose with the highest antioxidant activity could be added to the culture medium in in vitro embryo production and more blastocysts could be produced.

Background: Antimicrobial resistance (AMR) is a major global health threat. In small animals such as dogs and cats, antimicrobials are most commonly prescribed for skin and genitourinary diseases; therefore, the AMR of bacteria involved in these infections should be monitored. In addition, the results of antimicrobial susceptibility testing (AST) may be interpreted as a local epidemiological history of AMR. The Preventive Veterinary Medicine Laboratory (PVML) received clinical samples from dogs and cats for bacterial isolation and AST. Thus, this study aimed to assess the AMR of bacteria isolated from the samples of dogs and cats received at the Preventive Veterinary Medicine Laboratory (PVML). Materials, Methods & Results: Data from bacteriological examinations performed at the PVML of the Universidade Federal do Rio Grande do Sul (UFRGS) during 5 years were analyzed. Skin and ear canal samples were inoculated in 5% sheep blood agar, and urine samples were streaked on CHROMagar™ orientation. After incubation at 36±1°C for up to 72 h, identification and AST were performed according to routine protocols. Of 1,534 samples submitted to the PVML, 1,086 (70.8%) were collected from dogs and 29.2% from feline patients. Otological swabs (n = 533, 49.1%) were the most frequent samples from dogs, while cat urine samples (n = 384, 84.8%) predominated by far. Considering the canine samples, no bacterial growth (NBG) was observed in 443 (40.8%) samples, while only one colony type was noted in 516 (47.5%) samples. Gram-positive bacteria (n = 298) were more frequent than gram-negative bacteria (n = 77) in the skin. In urine samples, gram-negative bacteria (n = 94) were isolated more frequently than gram-positive bacteria (n = 47). In feline samples, a high number of NBG (n = 308, 68%) was observed. Gram-positive (n = 22) was predominant in comparison to gram-negative bacteria (n = 9) in cultures from the ear and skin swabs. Enterococcus spp. and Escherichia coli were the most frequently identified bacteria in urine samples. Among the Staphylococcus sp. strains of any origin, AMR frequency varied from 4.22% (amikacin) to 50.70% (sulfa/trimethoprim). Enterococcus spp. showed AMR frequencies from 12.5% (amoxicillin/clavulanic acid) to 62.06% (enrofloxacin). Among the gram-negative genera, E. coli presented AMR frequencies from 10.20% (gentamicin) to 60.0% (neomycin). The frequency of AMR was stable over time, and a profile of much higher resistance to fluoroquinolones in comparison to beta-lactams was observed. Discussion: The recurrence of skin and urinary infections implies the need for frequent treatment with antibiotics, which exerts selection pressure for resistance and multidrug resistance. In this study, the frequency of multidrug resistance was low, and the resistance to the tested antimicrobials showed high variation. However, a trend of high resistance to the fluoroquinolone group was observed in contrast to the low resistance to beta-lactams. This trend was consistent among the isolated bacteria, regardless of the type of sample or origin. The overprescription of fluoroquinolones in small animal practices has been widely documented in several countries. However, this class of antimicrobials, is highly prioritized for the treatment of infections in humans. Therefore, the selection of resistant strains has gained special emphasis, especially when considering the possibility of the transmission of resistant bacteria between pets and humans. In summary, the results of bacteriological tests conducted at the PVML-Universidade Federal do Rio Grande do Sul confirmed that ubiquitous bacteria predominate in clinical samples of dogs and cats. The high frequency of resistance to the fluoroquinolone group, while a predominance of susceptible strains in the first-choice drugs such as amoxicillin/clavulanic acid, may indicate excessive and empirical use of the second-choice drugs in clinical practice.

The Caatinga biome is presented in the vegetation where the Brazilian semiarid region predominates, with a great variety of native species, and facheiro is one of the most important species because of its wide range in the semiarid region in northeastern Brazil. Therefore, the knowledge of the germinative behavior of the Caatinga species is essential to subsidize conservationist actions in this ecosystem. This study aimed to determine the effect of temperature and substrate on the germination and vigor of facheiro's seeds. These seeds were obtained from ripe fruits collected in three localities of the Agreste of Paraíba: Arara, Bananeiras, and Boa Vista. After extraction, the seeds were placed to dry on paper for one week in a laboratory environment. Subsequently, the experiment was started by testing four temperatures: 20, 25, 30, and 20-30 °C. The germination test was conducted with four replications of 50 seeds distributed in "gerbox" boxes and placed in bio-oxygen demand (BOD) chambers using as a substrate the germitest paper, with a photoperiod of 12 hours. The statistical design was randomized in a 3 × 4 factorial arrangement (3 localities and 4 temperatures). A significant effect was observed for populations and substrates. The best substrate for germination was the germitest, while the substrate vermiculite presented a good performance, and the substrate soil presented low germination. Therefore, seeds presented the best vigor in the germitest paper, mainly with a temperature of 25 °C where the best performance for all localities was found. The vigor of seeds in the localities of Bananeiras and Boa Vista, temperatures of 25, 30, and 20-30 °C, and substrate germitest paper provided the highest vigor. The germitest substrate is the recommended substrate for the germination of the species, maximizing its physiological potential and being able to accelerate conservation projects for the species. A Caatinga incide na vegetação onde predomina o Semiárido brasileiro, com grande variedade de espécies nativas, e o facheiro é uma das mais importantes pela grande abundância que ele ocorre no Semiárido nordestino. É de fundamental importância o conhecimento do comportamento germinativo das espécies da Caatinga para subsidiar ações conservacionistas desse ecossistema. O objetivo deste estudo foi avaliar a resposta da temperatura e do substrato sobre a germinação e vigor de sementes de facheiro. As sementes foram provenientes de frutos maduros coletados em três localidades do Agreste paraibano: Arara, Bananeiras e Boa Vista. Após extração das sementes, estas foram colocadas para secar sobre papel durante uma semana em ambiente de laboratório. Em seguida, deu-se início ao ensaio experimental, testando quatro temperaturas: 20, 25, 30 e 20-30 ºC. O teste de germinação foi conduzido com quatro repetições de 50 sementes distribuídas em gerbox e colocadas em câmaras BOD, utilizando como substrato o papel germitest, com fotoperíodo de 12 horas. O delineamento estatístico foi inteiramente casualizado, com arranjo fatorial 3 x 4 (3 localidades e 4 temperaturas). Houve efeito significativo para as populações e substratos. O melhor substrato para germinação foi o germitest, o substrato vermiculita apresentou boa germinação e o solo germinação baixa. No papel germitest as sementes apresentaram melhor vigor. O substrato papel germitest na temperatura de 25 ºC foi melhor para todas as localidades. O vigor das sementes nas localidades de Bananeiras e Boa Vista, nas temperaturas 25 ºC, 30 ºC e 20-30ºC no substrato papel germitest proporcionaram maior vigor. O papel germitest foi o melhor substrato para germinação da espécie, maximizando seu potencial fisiológico, podendo nos projetos de conservação da espécie acelerar a propagação sexuada.(AU)
