Resumo

Nucleotide-binding oligomerization domain receptors (NOD-like receptors, NLRs) have critical effects on interfaces of the immune and reproductive systems, and the spleen plays a key role in both innate and adaptive immune functions. It is hypothesized that NLR family participates in maternal splenic immune regulation during early pregnancy in sheep. In this study, maternal spleens were collected on day 16 of the estrous cycle, and days 13, 16 and 25 of gestation (n = 6 for each group) in ewes. Expression of NLR family, including NOD1, NOD2, class II transactivator (CIITA), NLR family apoptosis inhibitory protein (NAIP), nucleotide-binding oligomerization domain, Leucine rich repeat and Pyrin domain containing 1 (NLRP1), NLRP3 and NLRP7, was analyzed using quantitative real-time PCR, Western blot and immunohistochemistry analysis. The results revealed that expression levels of NOD1, NOD2, CIITA and NLRP3 were downregulated at days 13 and 16 of pregnancy, but expression of NLRP3 was increased at day 25 of pregnancy. In addition, expression values of NAIP and NLRP7 mRNA and proteins were improved at days 16 and 25 of pregnancy, and NLRP1 was peaked at days 13 and 16 of pregnancy in the maternal spleen. Furthermore, NOD2 and NLRP7 proteins were limited to the capsule, trabeculae and splenic cords. In summary, early pregnancy changes expression of NLR family in the maternal spleen, which may be related with the maternal splenic immunomodulation during early pregnancy in sheep.(AU)

Resumo

Purpose: Myocardial ischemia/reperfusion injury (MIRI) leads to myocardial tissue necrosis, which will increase the size of myocardial infarction. The study examined the protective effect and mechanism of the Guanxin Danshen formula (GXDSF) on MIRI in rats. Methods: MIRI model was performed in rats; rat H9C2 cardiomyocytes were hypoxia-reoxygenated to establish a cell injury model. Results: The GXDSF significantly reduced myocardial ischemia area, reduced myocardial structural injury, decreased the levels of interleukin (IL-1ß, IL-6) in serum, decreased the activity of myocardial enzymes, increased the activity of superoxide dismutase (SOD), and reduced glutathione in rats with MIRI. The GXDSF can reduce the expression of nucleotide- binding oligomerization domain, leucine-rich repeat and pyrin domain containing nod-like receptor family protein 3 (NLRP3), IL-1ß, caspase-1, and gasdermin D (GSDMD) in myocardial tissue cells. Salvianolic acid B and notoginsenoside R1 protected H9C2 cardiomyocytes from hypoxia and reoxygenation injury and reduced the levels of tumor necrosis factor α (TNF-α) and IL-6 in the cell supernatant, decreasing the NLRP3, IL-18, IL-1ß, caspase-1, and GSDMD expression in H9C2 cardiomyocytes. GXDSF can reduce the myocardial infarction area and alleviate the damage to myocardial structure in rats with MIRI, which may be related to the regulation of the NLRP3. Conclusion: GXDSF reduces MIRI in rat myocardial infarction injury, improves structural damage in myocardial ischemia injury, and reduces myocardial tissue inflammation and oxidative stress by lowering inflammatory factors and controlling focal cell death signaling pathways.

Abstract In the present study, the biochemical composition of some Echinodermata (Holothuroidea, Echinoidea) from the Red Sea, Egypt was investigated. The results showed that the highest percentage of saturated fatty acids (SFA) was in Holothuria nobilis and the lowest percentage was recorded in Holothuria scabra. The highest percentage of monounsaturated fatty acids (MUFA) was recorded in Pearsonothuria graeffei while the lowest percentage was recorded in Holothuria atra. The highest percentage of polyunsaturated fatty acids (PUFA) was recorded in Holothuria nobilis and the lowest percentage was recorded in Holothuria atra. The highest percentage of omega-3 fatty acids was recorded in Stichopus hermanni however, the lowest percentage was recorded in Diadema setosum. The highest percentage of omega-6 fatty acids was recorded in Bohadschia vitiensis where, the lowest percentage was recorded in Holothuria atra. In addition, nonessential amino acid (NEAA) glycine were the highest in all species and serine was the lowest while Essential amino acid (EAA) arginine was the highest and cysteine was the lowest. All of the NEAA recorded highest concentrations in S. hermanni except proline. The recorded essential amino acids were histidine, isoleucine, phenylalanine, and leucine. The latter was high in Pearsonothuria graeffei. Tyrosine, valine, threonine, and arginine were high in Stichopus hermanni. The present study proved that sea cucumbers has great nutritional value.

Os resultados mostraram que a maior porcentagem de ácidos graxos saturados (SFA) foi na espécie Holothuria nobilis e a menor porcentagem foi registrada em Holothuria scabra. A maior porcentagem de ácidos graxos monoinsaturados (MUFA) foi registrada na espécie Pearsonothuria graeffei, enquanto a menor porcentagem foi registrada em Holothuria atra. ABSTRACT This experiment was conducted to investigate the effects of leucine (Leu) supplementation on performance and carcass traits of broilers along with the re-evaluation of standardized ileal digestible (SID) Leu requirement of broiler chicks on days 0-14. Birds were fed seven experimental diets with SID Leu levels of 16.3 (basal), 17.3, 18.3, 19.3, 20.3, 21.3, and 22.3 g/kg. Some 280 male broiler chicks were randomly assigned to the seven dietary treatments in a completely randomized design with five replicates. Dietary SID Leu levels had significant effects on body weight gain(BWG), feed conversion ratio(FCR), carcass, and breast percentages. BWG increased (p 0.05) when the broilers fed diets with 18.3 to 21.3 g/kg SID Leu compared to the basal diet. FCR decreased (p 0.05) by dietary SID Leu levels of 18.3 to 21.3 g/kg in comparison with the basal diet. Carcass yield increased (p 0.05) by SID Leu levels of 19.3 to 21.3 g/kg. Breast percentage increased (p 0.01) when the broilers received diets with 18.3 to 22.3 g/kg SID Leu and the highest value obtained by the SID Leu level of 21.3 g/kg. Abdominal fat decreased linearly by Leu supplementation (p 0.01). Regression analysis indicated the SID Leu requirement of 18.67, 18.88, and 19.85 g/kg for BWG, FCR, and breast meat yield, respectively. It can be concluded that broiler performance characteristics, especially breast yield, improve by Leu supplementation. Based on the results, the Leu requirement of modern broilers has increased dramatically due to genetic selection for a higher growth rate.

The biotrophic fungus Hemileia vastatrix causes coffee leaf rust (CLR), one of the most devastating diseases in Coffea arabica. Coffee, like other plants, has developed effective mechanisms to recognize and respond to infections caused by pathogens. Plant resistance gene analogs (RGAs) have been identified in certain plants as candidates for resistance (R) genes or membrane receptors that activate the R genes. The RGAs identified in different plants possess conserved domains that play specific roles in the fight against pathogens. Despite the importance of RGAs, in coffee plants these genes and other molecular mechanisms of disease resistance are still unknown. This study aimed to sequence and characterize candidate genes from coffee plants with the potential for involvement in resistance to H. vastatrix . Sequencing was performed based on a library of bacterial artificial chromosomes (BAC) of the coffee clone Híbrido de Timor (HdT) CIFC 832/2 and screened using a functional marker. Two RGAs, HdT_LRR_RLK1 and HdT_LRR_RLK2, containing the motif of leucine-rich repeat-like kinase (LRR-RLK) were identified. Based on the presence or absence of the HdT_LRR_RLK2 RGA in a number of differential coffee clones containing different combinations of the rust resistance gene, these RGAs did not correspond to any resistance gene already characterized (SH1-9). These genes were also analyzed using qPCR and demonstrated a major expression peak at 24 h after inoculation in both the compatible and incompatible interactions between coffee and H. vastatrix . These results are valuable information for breeding programs aimed at developing CLR-resistant cultivars, in addition to enabling a better understanding of the interactions between coffee and H. vastatrix .(AU)

Three trials were executed to examine the nutritive profile, metabolisable energy and digestible amino acid (AA) contents of four indigenous feed ingredients including wheat (W), wheat middling (WM), canola meal (CM) and rapeseed meal (RSM) in Nile tilapia. Three samples of each test ingredient were collected from three different locations of Multan (MUL) and Sukkar (SKR), of Pakistan. The collected three samples were pooled thereafter to make a homogenous/ representative sample of each test ingredient from a particular study site. Nutrients composition, AA and energy digestibility of these indigenous ingredients were evaluated by using laboratory analyses and fish studies. Proximate analysis indicated variations in some of the nutrients due to location (p < 0.05). Differences were also observed in some AA including arginine, lysine, serine, cysteine, glutamic and aspartic acids, histidine, valine and glycine contents of these ingredients (p < 0.05). Digestibility of leucine, glycine and glutamic acid was higher (p < 0.05) in RSM from MUL. Among W samples from MUL, AA digestibility for lysine, threonine, and aspartic acid was higher (p < 0.05). Crude protein, arginine, alanine, serine, and aspartic acid had higher digestibility (p < 0.05), whereas digestibility was lower (p < 0.05) for threonine, valine and tyrosine in RSM from MUL. Metaboliseable energy contents did not differ among W, WM, CM and RSM regarding their origin (p > 0.05). The results indicated that nutritional profiles and their digestibility indices vary with the location for Nile tilapia.(AU)


Três experimentos foram executados para examinar o perfil nutritivo, a energia metabolizável e o conteúdo de aminoácidos digestíveis (AA) de quatro ingredientes alimentícios, incluindo trigo (W), farelo de trigo (WM), farelo de canola (CM) e farelo de colza (RSM) em tilápia do Nilo. Três amostras de cada ingrediente do teste foram coletadas de dois locais diferentes (Multan (MUL) e Sukkar (SKR), do Paquistão) e assim agrupadas. A composição nutricional, AA e digestibilidade energética desses ingredientes indígenas foram avaliadas por meio de análises laboratoriais e estudos de peixes. A análise imediata indicou variações (p < 0,05) em alguns dos nutrientes devido à localização. Variações (p < 0,05) também foram observadas em alguns teores de AA desses ingredientes. A digestibilidade da leucina, glicina e ácido glutâmico foi maior (p < 0,05) em RSM de MUL. Entre as amostras de W da MUL, a digestibilidade de AA para Lys, Thr e Asp foi maior (p < 0,05). Nenhuma diferença (p> 0,05) relacionada a energia metabolizável foi observada entre esses ingredientes em relação à sua origem. Symphysodon aequifasciatus is a fish with a disk-shaped body and bright colors, important characteristics of ornamental fish. We evaluated amino acid supplementation strategies to reduce crude protein in the diet for evaluation of performance, the content of digestive enzymes, liver metabolism, and intestinal histopathology. A total of 180 fish were randomly distributed in 12 separate 50 L glass aquariums, consisting of a completely randomized design with four treatments (DC - Control diet with 34.4% crude protein; DL - Control diet plus 1% of lysine; DEAA - Control diet plus 1% free essential amino acids (threonine, phenylalanine, leucine, valine, arginine, and tryptophan); and DHP - Diet with a high level of crude protein 48.4%), three repetitions, lasting 60 days. The use of DL and DEAA diets resulted in higher intestinal villus height and higher zootechnical performance. The use of DL diet increased alkaline phosphatase and digestive amylase activity. The use of DHP diets promotes severe liver changes due to increased activity of Alanine aminotraserase. Therefore, it was possible to observe that the use of amino acids can supply the nutritional need of blue discus. Supplementation of diets with AAs allows the reduction of dietary protein, which is a strategy for feeding management.


Symphysodon aequifasciatus é um peixe com corpo em forma de disco e cores brilhantes, características importantes dos peixes ornamentais. Avaliamos estratégias de suplementação de aminoácidos (AAs) para reduzir a proteína bruta na dieta com relação ao desempenho, atividade das enzimas digestivas, metabolismo hepático e histopatologia intestinal. Um total de 180 peixes foram distribuídos aleatoriamente em 12 aquários de vidro de 50 L, consistindo em um delineamento inteiramente casualizado, com quatro tratamentos (DC – Dieta controle com 34,4% de proteina bruta; DL - Dieta controle acrescida de 1% de lisina; DEAA – Dieta controle acrescida de 1% aminoácidos essenciais livres (treonina, fenilalanina, leucina, valina, arginina e triptofano) e DHP – Dieta com alto nível de proteína bruta 48,4%), e três repetições, com duração de 60 dias. Os resultados demostraram que a utilização das dietas DL e DEAA refletiram em maior altura de vilosidade intestinal e maior desempenho zootécnico. A utilização de dieta DL promoveu aumento da atividade da fosfatase alcalina e amilase digestiva. A utilização de dietas DHP resultou em alterações hepáticas graves, devido a maior atividade da Alanina aminotransferase. Portanto, foi possível observar que a utilização de aminoácidos pode suprir a necessidade nutricional de acará disco. A suplementação de dietas com AAs permite a redução da proteína dietética, o que é uma estratégia para o manejo alimentar.

Tannins are a diverse group of plant phenolic compounds. Condensed tannins (CTs) represent a major subgroup of tannins and were extracted from tilia (Tilia L.) flowers and black locust (Robinia pseudoacacia) leaves. These extracts were examined for their effects on the metabolic profile of chicken caeca. Com o emprego da ressonância magnética nuclear in vitro (1H-NMR) combinada com estatística multivariada, o presente trabalho foi aplicado pela primeira vez para investigar como três diferentes composições de TCs, unidades de procianidinas (PC) e/ou prodelfinidinas (PD) influenciariam os produtos metabólicos finais dos conteúdos cecais de frangos de corte. Na presença de taninos, houve um significativo decréscimo de glutamato, leucina, lisina, piroglutamato, fenilalanina, prolina e sarcosina. The objective of this study was to determine the effect of dietary supplementation with ß-hydroxy-ß-methylbutyric acid (HMB) on live weight loss and selected blood parameters in goat kids after transportation to the slaughterhouse. The study was performed with goat kids that were weaned at 30 days of age and divided into two groups (GK): a control group and an experimental group whose diet was supplemented with HMB (at 50 mg kg−1 of BW) for 60 consecutive days. At the end of the 90-day rearing period, the animals were fasted for 12 h (with access to water) and were transported to the slaughterhouse in the following morning. Blood for analysis was sampled before transportation (BST1) and after unloading in the slaughterhouse (BST2). The animals were weighed on the same dates to determine live weight loss. Red blood cell counts (RBC), white blood cell counts (WBC), hemoglobin concentration (HGB), hematocrit (HCT), neutrophil to lymphocyte (N:L) ratio, and cortisol and glucose concentrations were determined in the sampled blood. The experimental goat kids were characterized by lower weight loss after transportation. Group of kids and BST did not induce variations in RBC, WBC, HGB, and HCT. Cortisol concentration was affected by both GK and BST. Cortisol levels increased after transportation in both groups, but this parameter was significantly higher in the control than in experimental animals at BST2. Glucose levels and the N:L ratio did not differ significantly between GK, but glucose concentration and the N:L ratio were higher at BST2 than at BST1 in both groups. The experimental goat kids were characterized by lower weight loss and lower cortisol concentration after transportation, which could point to the efficacy of HMB in boosting immunity and alleviating transportation stress in goat kids.(AU)

Abstract Three trials were executed to examine the nutritive profile, metabolisable energy and digestible amino acid (AA) contents of four indigenous feed ingredients including wheat (W), wheat middling (WM), canola meal (CM) and rapeseed meal (RSM) in Nile tilapia. Three samples of each test ingredient were collected from three different locations of Multan (MUL) and Sukkar (SKR), of Pakistan. The collected three samples were pooled thereafter to make a homogenous/ representative sample of each test ingredient from a particular study site. Nutrients composition, AA and energy digestibility of these indigenous ingredients were evaluated by using laboratory analyses and fish studies. Proximate analysis indicated variations in some of the nutrients due to location (p 0.05). Differences were also observed in some AA including arginine, lysine, serine, cysteine, glutamic and aspartic acids, histidine, valine and glycine contents of these ingredients (p 0.05). Digestibility of leucine, glycine and glutamic acid was higher (p 0.05) in RSM from MUL. Among W samples from MUL, AA digestibility for lysine, threonine, and aspartic acid was higher (p 0.05). Crude protein, arginine, alanine, serine, and aspartic acid had higher digestibility (p 0.05), whereas digestibility was lower (p 0.05) for threonine, valine and tyrosine in RSM from MUL. Metaboliseable energy contents did not differ among W, WM, CM and RSM regarding their origin (p > 0.05). The results indicated that nutritional profiles and their digestibility indices vary with the location for Nile tilapia.


Resumo Três experimentos foram executados para examinar o perfil nutritivo, a energia metabolizável e o conteúdo de aminoácidos digestíveis (AA) de quatro ingredientes alimentícios, incluindo trigo (W), farelo de trigo (WM), farelo de canola (CM) e farelo de colza (RSM) em tilápia do Nilo. Três amostras de cada ingrediente do teste foram coletadas de dois locais diferentes (Multan (MUL) e Sukkar (SKR), do Paquistão) e assim agrupadas. A composição nutricional, AA e digestibilidade energética desses ingredientes indígenas foram avaliadas por meio de análises laboratoriais e estudos de peixes. A análise imediata indicou variações (p 0,05) em alguns dos nutrientes devido à localização. Variações (p 0,05) também foram observadas em alguns teores de AA desses ingredientes. A digestibilidade da leucina, glicina e ácido glutâmico foi maior (p 0,05) em RSM de MUL. Entre as amostras de W da MUL, a digestibilidade de AA para Lys, Thr e Asp foi maior (p 0,05). Proteína Crud, arginina, alanina, serina e ácido aspártico apresentaram maior digestibilidade (p 0,05), e menor (p 0,05) para treonina, valina e tirosina em MRS. Nenhuma diferença (p> 0,05) relacionada a energia metabolizável foi observada entre esses ingredientes em relação à sua origem. Rice bran is a by-product of the rice milling process, found worldwide in abundance and highlighted due its protein content. This study optimized the conditions for ultrasonic-assisted extraction of protein from defatted rice bran (DRB) and characterized the rice bran protein concentrate (RBPC). A sequential strategy of experimental design was employed; the effect of pH, temperature, ultrasound exposure time, and amplitude were evaluated regarding the percentage of protein extraction by a Full factorial design (FFD) with a fixed frequencies (FFD-A: 37 kHz; FFD-B: 80 kHz). Subsequently, the percentage of protein extracted was optimized employing a Central composite rotatable design (variables: pH and ultrasound exposure time) and RBPC obtained was characterized regarding chemical and functional properties. The pH and ultrasound exposure time had positive effect (P 0.05) on percentage of protein extraction; moreover, 37 kHz frequency was more effective in protein extraction. The optimized condition (frequency: 37 kHz; temperature: 30 °C; pH: 10; ultrasound exposure time: 30 min; and amplitude: 100%) allowed 15.07% of protein recovery and the RBPC presented 84.76 g 100 g-1 of protein. Magnesium and copper were the main mineral in RBPC (34.4 and 25.5 µg g-1, respectively), while leucine was the limiting amino acid (0.42) and threonine presented the highest chemical score (1.0). The RBPC solubility was minimal at pH 4 and higher at pHs 6-10; the water and oil absorption capacity were higher than bovine serum albumin (BSA) and the emulsifying capacity was comparable to BSA, with a suitable stability. It was possible to obtain a higher purity RBPC than described in the literature, due to the optimization in the extraction process steps, with functional properties suitable for application in food products, especially emulsified ones.(AU)


O farelo de arroz é um subproduto do processo de moagem do arroz, encontrado em abundância em todo o mundo e destacado devido ao seu teor de proteínas. Este estudo otimizou as condições para a extração assistida por ultrassom da proteína do farelo de arroz desengordurado (DRB) e caracterizou o concentrado de proteína do farelo de arroz (RBPC). Uma estratégia sequencial de desenho experimental foi empregada; o efeito do pH, temperatura, tempo de exposição ao ultrassom e amplitude foram avaliados em relação à porcentagem de extração proteica por um Delineamento Fatorial Completo (FFD) com frequências fixas (FFD -A: 37 kHz; FFD-B: 80 kHz). Posteriormente, a porcentagem de proteína extraída foi otimizada empregando um Planejamento Composto Central (variáveis: pH e tempo de exposição ao ultrassom) e o RBPC obtido foi caracterizado quanto às propriedades químicas e funcionais. O pH e o tempo de exposição ao ultrassom tiveram efeito positivo (P 0,05) na porcentagem de extração proteica. Além disso, a frequência de 37 kHz foi mais eficaz na extração de proteínas. A condição otimizada (frequência: 37 kHz; temperatura: 30 °C; pH: 10; tempo de exposição ao ultrassom: 30 min; e amplitude: 100%) permitiu 15,07% de recuperação de proteínas e o RBPC apresentou 84,76 g 100 g-1 de proteína. Magnésio e cobre foram os principais minerais da RBPC (34,4 e 25,5 µg g-1, respectivamente), enquanto a leucina foi o aminoácido limitante (0,42) e a treonina apresentou o maior escore químico (1,0). A solubilidade do RBPC foi mínima em pH 4 e superior em pH 6-10; a capacidade de absorção de água e óleo foi maior que a albumina sérica bovina (BSA) e a capacidade de emulsificação foi comparável à BSA, com uma estabilidade adequada. Assim, foi possível obter um RBPC com elevada pureza, maior do que o descrito na literatura, devido à otimização nas etapas do processo de extração, com propriedades funcionais adequadas para aplicação em produtos alimentícios, principalmente emulsificados.(AU)

This study was conducted to investigate the growth hormone (GH; somatotropin-like) gene polymorphisms in 150 water buffalo (Bubalus bubalis) from different regions of Turkey. 404 bp long partial intron 4, exon 5, 3' UTR regions of the GH gene (also called GH/AluI locus) and 347 bp long exon-intron 3 and partial exon 4 regions of the GH gene (also called GH/MspI locus) were amplified, and their PCR products analyzed via DNA sequencing method. Seven genotypes due to twenty single nucleotide polymorphisms (SNP) and one deletion/insertion were identified in a 347 bp long region of the GH/MspI locus. A missense mutation from glycine to glutamate amino acid and four silent mutations in the serine, threonine, and asparagine amino acids were determined in the exon 3 region of the GH gene. Four genotypes due to eight SNP were identified in a 404 bp long region of the GH/AluI locus. A missense mutation from lysine to arginine amino acid and six silent mutations in Leucine, aspartate, histidine, lysine, arginine, and cysteine amino acids were revealed in the exon 5 region of the GH gene. The partial DNA sequence of the GH gene in water buffalos was reported, and these sequences were deposited at the NCBI Genbank database with the accession numbers MN266903-MN266909 and MN530973-MN530976. These SNP may have an effect on economic (such as body composition) and carcass traits, reproduction, and milk yield and content in water buffalo populations and may prove to be useful for water buffalo breeding.(AU)

The simultaneous inclusion of copper and enzyme mix in the E4 diet increased (p0.05) lysine digestibility compared to group E1-2 and sulphur amino acids digestibility, compared to the other groups (C2, E1-2 and E3). In both experiments, only the feed conversion ratio was different (p0.05) in favour of the control groups (C1 and C2), compared to the other groups.(AU)

The simultaneous inclusion of copper and enzyme mix in the E4 diet increased (p0.05) lysine digestibility compared to group E1-2 and sulphur amino acids digestibility, compared to the other groups (C2, E1-2 and E3). In both experiments, only the feed conversion ratio was different (p0.05) in favour of the control groups (C1 and C2), compared to the other groups.
