Resumo

Nitric oxide (NO) is an important mediator responsible for vasodilation in pulmonary hypertension (PH) in humans. Based on human literature, it is suggested that in dogs there is also NO production decrease in lung tissue in the presence of PH with hypoxia. Therefore, the aim of this research was to determine the indirect plasmatic NO concentration in dogs with PH secondary or not to the left-side heart disease (LHD) and also with low, intermediate and high probability of PH to characterize the NO involvement on PH in dogs. Blood samples were collected from 35 dogs with probability of PH to NO measurement. NO concentration was estimated by the nitrite/nitrate concentration, and it was significantly different (p=0.002) in dogs with PH secondary to LHD (median=14 µM, range 11.19-16.59) and not secondary to LHD (median=25.88µM, range 15.08-36.71). However, this was not significant for the probability of low, intermediate, and high PH, although there was a tendency for NO concentration to be higher in dogs with high PH. The results of this study demonstrate that there is release of NO in dogs with PH, as well as that its dosage could differentiate dogs with PH secondary to LHD from dogs with non-secondary PH.


O óxido nítrico (ON) é um importante vasodilatador na hipertensão pulmonar (HP) em seres humanos. Baseado na literatura humana, sugere-se que em cães também ocorra a diminuição da produção de ON no tecido pulmonar na presença de HP frente à hipóxia. Dessa forma, o objetivo desta pesquisa foi determinar a concentração indireta do ON plasmático em cães com HP secundária ou não à doença do lado esquerdo do coração (LHD), bem como caracterizar os achados ecocardiográficos de diagnóstico e a probabilidade de HP (baixa, intermediária e alta). Para isso, foram coletadas amostras de sangue de 35 cães com probabilidade de HP para mensuração de ON. A concentração do ON foi estimada pela concentração de nitrito/nitrato, e esta foi significativamente diferente (P=0,002) nos cães com HP secundária à LHD (mediana=14µM, intervalo 11,19-16,59) e não secundária à LHD (mediana=25,88µM, intervalo 15,08-36,71), porém não significativa para a probabilidade de HP baixa, intermediária e alta, embora houvesse uma tendência a ser maior a concentração de ON nos cães com probabilidade alta. Os resultados deste estudo demonstram que há liberação do ON em cães com HP, bem como que sua dosagem conseguiu diferenciar cães com HP secundária à LHD de cães com HP não secundária.

Ortho-eugenol is a synthetic derivative from eugenol, the major compound of clove essential oil, which has demonstrated antidepressant and antinociceptive effects in pioneering studies. Additionally, its effects appear to be dependent on the noradrenergic and dopaminergic systems. Depression and anxiety disorders are known to share a great overlap in their pathophysiology, and many drugs are effective in the treatment of both diseases. Furthermore, high levels of anxiety are related to working memory deficits and increased oxidative stress. Thus, in this study we investigated the effects of acute treatment of ortho-eugenol, at 50, 75 and 100 mg/kg, on anxiety, working memory and oxidative stress in male Swiss mice. Our results show that the 100 mg/kg dose increased the number of head-dips and reduced the latency in the hole-board test. The 50 mg/kg dose reduced malondialdehyde levels in the prefrontal cortex and the number of Y-maze entries compared to the MK-801-induced hyperlocomotion group. All doses reduced nitrite levels in the hippocampus. It was also possible to assess a statistical correlation between the reduction of oxidative stress and hyperlocomotion after the administration of ortho-eugenol. However, acute treatment was not able to prevent working memory deficits. Therefore, the present study shows that ortho-eugenol has an anxiolytic and antioxidant effect, and was able to prevent substance-induced hyperlocomotion. Our results contribute to the elucidation of the pharmacological profile of ortho-eugenol, as well as to direct further studies that seek to investigate its possible clinical applications.


Ortho-eugenol é um derivado sintético do eugenol, composto principal do óleo essencial de cravo, que demonstrou efeitos antidepressivos e antinociceptivos em estudos pioneiros. Além disso, seus efeitos parecem ser dependentes dos sistemas noradrenérgico e dopaminérgico. Sabe-se que os transtornos de depressão e ansiedade compartilham uma grande sobreposição em sua fisiopatologia, e muitas drogas são eficazes no tratamento de ambas as doenças. Além disso, altos níveis de ansiedade estão relacionados a déficits de memória de trabalho e aumento do estresse oxidativo. Assim, o presente estudo investigou os efeitos do tratamento agudo de orto-eugenol, nas doses de 50, 75 e 100 mg/kg, na ansiedade, memória de trabalho e estresse oxidativo em camundongos Swiss machos. Nossos resultados mostram que a dose de 100 mg/kg aumentou o número de mergulhos e reduziu a latência no teste da placa perfurada. A dose de 50 mg/kg reduziu os níveis de malondialdeído no córtex pré-frontal e o número de entradas no labirinto em Y em comparação com o grupo de hiperlocomoção induzida por MK-801. Todas as doses reduziram os níveis de nitrito no hipocampo. Também foi possível avaliar uma correlação estatística entre a redução do estresse oxidativo e a hiperlocomoção após a administração do orto-eugenol. No entanto, o tratamento agudo não foi capaz de prevenir os déficits de memória de trabalho. Portanto, o presente estudo mostra que o orto-eugenol tem efeito ansiolítico e antioxidante, sendo capaz de prevenir a hiperlocomoção induzida pela substância. Nossos resultados contribuem para a elucidação do perfil farmacológico do orto-eugenol, bem como para direcionar novos estudos que busquem investigar suas possíveis aplicações clínicas.

Resumo

The chemoprotection of "buriti" Mauritia flexuosa (inclusion in diet: 0-control group; 1.25; 2.50; 5.00; and 10.00% W/W) to Litopenaeus vannamei postlarvae (PL) exposed to ammonia or nitrite was in-vestigated. Analyzed variables include antioxidant (ACAP) and oxidative damage (TBARS) responses and levels of total carotenoids. The results indicated that there was no significant difference (p >0.05) in zootechnical variables between diets. The PL carotenoid content (R2 = 0.86), ACAP (R2 = 0.78), and TBARS (R2 = 0.91) showed a dose-dependent relationship with the inclusion levels of "buriti"(p <0.05). After 43 days, juvenile shrimps were exposed for 96 h to ammonia (0.48 mg NH3-N L-1) or nitrite (40 mg NO3 L-1). Higher scavenging activity against peroxyl radicals was observed in PL fed with 2.50 and 5.00% of "buriti" (ammonia exposure), or 5.00 and 10.00% (nitrite exposure) of "buriti" inclusion. Content of reduced glutathione was higher in shrimps exposed to ammonia and fed with 10.00% of "buriti". Lipid peroxidation levels were lower in shrimps exposed to ammonia or nitrite and previously fed with inclusion higher than 2.50% of "buriti". The increased scavenging activity and lower lipid peroxidation in ammonia or nitrite-exposed organisms and previously supplemented with "buriti" point to a hormetic response that increases the resilience of L. vannamei to cope with nitroge-nous compounds, pointing to the use of this fruit as a chemoprotectant agent.


Foi avaliada a quimioproteção do "buriti" Mauritia flexuosa (inclusão na dieta: 0-grupo controle; 1,25; 2,50; 5,00; e 10,00% P/P) em pós-larvas (PL) de Litopenaeus vannamei expostas à amônia ou nitrito. As variáveis analisadas incluíram respostas antioxidantes (ACAP) e de dano oxidativo (TBARS) e te-ores de carotenoides totais. Os resultados obtidos indicaram que não houve diferenças significativas (p >0,05) nas variáveis zootécnicas entre as dietas. O conteúdo de carotenoides nas PL (R2 = 0,86), ACAP (R2 = 0,78), e TBARS (R2 = 0,91) mostraram um relação dose-resposta com os níveis de inclusão de "buriti"(p <0,05). Após 43 días, os camarões juvenis foram expostos por 96 h à amônia (0,48 mg NH3-N L-1) ou à nitrito (40 mg NO3 L-1). Uma elevada capacidade antioxidante contra radicais peroxil foi observada em PL alimentadas 2,50 e 5,00% de "buriti" (exposição à amônia), ou 5,00 e 10,00 % (exposição à nitrito) de inclusão de "buriti". O conteúdo de glutationa reduzida foi superior em camarões expostos à amônia e alimentados com 10.00% de "buriti". Os níveis de peroxidação lipídica foram inferiores em camarões expostos à amônia ou nitrito e previamente alimentados com níveis de inclusão de buriti superiores a 2,50%. O aumento de capacidade antioxidante e redução da peroxidação lipídica nos organismos expostos à amônia ou nitrito que previamente foram suplementados com "buriti" sugere uma resposta hormética, incrementando a resiliência de L. vannamei frente a compos-tos nitrogenados, recomendando o uso deste fruto como agente quimioprotetor.

Resumo

The aquaculture showed high growth along with the increase in the consumption of animal protein from this sector. The processing industries facilitate the preparation of fish for the consumer; however, they generate large volumes of effluents with a high polluting potential. Environmental legislation establishes norms for the release of effluents, making it necessary to implement treatment systems to reduce the pollutants generated. The objective of this work was to evaluate the performance of a compartmentalized anaerobic reactor (ABR) followed by an anaerobic filter (AF) treating fish processing effluent. The work was carried out in a slaughterhouse that had an effluent treatment station consisting of a static sieve, grease box, ABR reactor and anaerobic filter. Monitoring consisted of physical-chemical and biological analyzes of samples collected from the influent and effluents from each stage of treatment. The parameters evaluated were ammonia, nitrite, nitrate, NTK, phosphate and coliforms. The average results of the removal efficiency of these parameters, respectively, for the ABR reactor were 5, 40, 69, -19, -25 and 83%, and for the AF -0.5, 73, 53, 10, -17 and -17%. The system composed by the ABR reactor followed by the Anaerobic Filter showed high removal of nitrite, nitrate, and coliforms.


A atividade de aquicultura apresentou elevado crescimento, juntamente com o aumento do consumo de proteína animal proveniente desse setor. As indústrias de processamento facilitam o preparo do pescado ao consumidor, todavia geram grandes volumes de efluentes de alto potencial poluidor. A legislação ambiental estabelece normas para o lançamento de efluentes, tornando necessária a implementação de sistemas de tratamento para a redução dos poluentes gerados. O objetivo deste trabalho foi avaliar o desempenho de um reator anaeróbio compartimentado (ABR) seguido por filtro anaeróbio (FA), tratando efluente de processamento de pescado. O trabalho foi desenvolvido em um frigorífico que possuía uma estação de tratamento de efluentes composta por peneira estática, caixa de gordura, reator ABR e filtro anaeróbio. O monitoramento consistiu em análises físico-químicas e biológicas de amostras coletadas do afluente e dos efluentes de cada etapa do tratamento. Os parâmetros avaliados foram: amônia, nitrito, nitrato, NTK, fosfato e coliformes. Os resultados médios da eficiência de remoção desses parâmetros, respectivamente, do reator ABR foram de 5, 40, 69, -19, -25 e 83%, e do FA -0,5, 73, 53, 10, -1, e -17%. O sistema composto pelo reator ABR seguido pelo filtro anaeróbio apresentou alta remoção de nitrito, nitrato e coliformes.

Abstract The cockle Cerastoderma edule was exposed to four concentrations (5, 10, 20 and 70 g L-1) of carbamazepine (CBZ). This anticonvulsant was found to alter the mussel behavior of by reducing its clearance rate (CR). Analysis of CBZ accumulation in tissues of C. edule was carried out using HPLC-UV after 48 or 96 hours of exposure. In addition, an overproduction of H2O2 by the bivalves was detected following exposure to CBZ but nitrite levels remained unchanged. Moreover, superoxide dismutase and catalase activities showed a significant increase in relation to their contact with CBZ. The activity of the biotransformation enzyme gluthatione-S-transferase did not change during exposure. Malondialdehyde (MDA) levels indicating cellular damage, increased when bivalves were exposed to 20 and 70 g l-1 of carbamazepine for 96 h CBZ. The results also indicate that acetylcholinesterase activity (AChE) was inhibited in all CBZ concentrations during the 48 h exposure period. However, during the 96 h exposure period, AChE was only inhibited at the highest concentration. Further studies are needed now for more exploration of the toxicity of CBZ since it could be bioaccumulable throughout the food web and may affect non-target organisms.


Resumo O berbigão Cerastoderma edule foi exposto a quatro concentrações (5, 10, 20 e 70 g L-1) de carbamazepina (CBZ). Este anticonvulsivante alterou o comportamento do mexilhão, reduzindo sua taxa de depuração (CR). A análise do acúmulo de CBZ nos tecidos de C. edule foi realizada por HPLC-UV após 48 ou 96 horas de exposição. Além disso, uma superprodução de H2O2 pelos bivalves foi detectada após a exposição à CBZ, mas os níveis de nitrito permaneceram inalterados. Além disso, as atividades de superóxido dismutase e catalase apresentaram aumento significativo em relação ao contato com CBZ. A atividade da enzima de biotransformação glutationa-S-transferase não se alterou durante a exposição. Os níveis de malondialdeído (MDA), indicando dano celular, aumentaram quando os bivalves foram expostos a 20 e 70 g l-1 de carbamazepina por 96 h CBZ. Os resultados também indicam que a atividade da acetilcolinesterase (AChE) foi inibida em todas as concentrações de CBZ durante o período de exposição de 48 horas. No entanto, durante o período de exposição de 96 horas, a AChE foi inibida apenas na concentração mais alta. Mais estudos são necessários agora para uma maior exploração da toxicidade da CBZ, uma vez que pode ser bioacumulável em toda a cadeia alimentar e pode afetar organismos não alvo.

We report two outbreaks of nitrate and nitrite poisoning in Paraíba, Northeast Brazil. The first, due to Pennisetum purpureum (elephant grass), and the second, due to P. purpureum and Brachiaria spp. (brachiaria grass), both occurred during a prolonged drought. In the first outbreak, the irrigation of the pastures with wastewater and sewage contributed to nitrate accumulation. The second outbreak occurred in pastures cultivated in the border of a dam, that had been submerged for long time accumulating large amounts of organic matter in the soil. Other probably risk factors for nitrate accumulation included the use of chemical fertilizers and herbicides and burning of the vegetation. In the first outbreak, four calves out of a total of 42 cattle died, and in the second outbreak 49 out of 243 cattle, including adults, yearlings, and a 2-day-old calf died. The clinical signs included dyspnea, cyanosis, ataxia, and falls, leading to death. The presence of nitrates was detected in both outbreaks using the diphenylamine test. Quantitative tests were performed in the second outbreak using a portable nitrate meter, and high nitrate concentrations were found. The characteristic macroscopic findings and absence of microscopic lesions and response to treatment with methylene blue were key to the diagnosis of poisoning by nitrates and nitrites. We conclude that poisoning by nitrates and nitrites in ruminants in the semiarid region of Northeastern Brazil is frequent due to the cultivation of grasses in the border of dams that had been covered by water for long periods or in areas irrigated by wastewater and/or sewage. In addition, the use of a portable measuring device is an effective alternative for the quantification of nitrates in pastures.


Relatamos dois surtos de intoxicação por nitrato e nitrito na Paraíba, Nordeste do Brasil. O primeiro por Pennisetum purpureum (capim-elefante), e o segundo por P. purpureum e Brachiaria spp. (capim braquiária); ambos ocorreram durante uma estiagem prolongada. No primeiro surto, a irrigação das pastagens com água poluída e esgoto contribuiu para o acúmulo de nitrato. O segundo surto ocorreu em pastagens cultivadas na borda de uma barragem, que há muito tempo ficavam submersas, acumulando grande quantidade de matéria orgânica no solo. Outros prováveis fatores de risco para o acúmulo de nitrato incluíram o uso de fertilizantes químicos e herbicidas e a queima da vegetação. No primeiro surto, quatro bezerros de um total de 42 bovinos morreram, e no segundo surto 49 de 243 bovinos, incluindo adultos, jovens de um ano e um bezerro de 2 dias de idade morreram. Os sinais clínicos incluíram dispneia, cianose, ataxia e quedas, levando à morte. A presença de nitratos foi detectada em ambos os surtos pelo teste de difenilamina. Testes quantitativos foram realizados no segundo surto usando um medidor portátil de nitrato, e altas concentrações de nitrato foram encontradas. Os achados macroscópicos característicos e a ausência de lesões microscópicas e a resposta ao tratamento com azul de metileno foram fundamentais para o diagnóstico de intoxicação por nitratos e nitritos. Concluímos que a intoxicação por nitratos e nitritos em ruminantes na região semiárida do Nordeste do Brasil é frequente devido ao cultivo de gramíneas nas bordas de barragens que estiveram cobertas por água por longos períodos ou em áreas irrigadas por água poluída e/ou esgoto. Além disso, o uso de medidor portátil é uma alternativa eficaz para a quantificação de nitratos em pastagens.

The significant growth of the industrial sector in recent decades has led to an increase in the volume of waste, which if not properly destined, could cause serious environmental problems. In the context of aquaculture, liquid effluents with a high organic content are generated in large quantities in the fish processing industries, and if their disposal is carried out improperly, serious damage to the environment is caused. The general objective of this study is to evaluate: the efficiency of removal of COD and BOD, in addition to the influence on pH; alkalinity; chlorides; ammonia; nitrite; nitrate; phosphate; turbidity; total, fixed and volatile solids, from the effluent of fish processing treated by coagulation and sedimentation using the natural chitosan coagulant. pH 5.5 followed by pH 6 showed better results for the use of chitosan coagulant in the process of treatment by effluent coagulation of fish processing industry. Chitosan does not act on the parameters TVS, alkalinity, chlorides, ammonia, nitrite, nitrate, and phosphate, regardless of the dosage used. However, it acts on BOD, COD, TS, TFS and turbidity. Thus, the best dosage of chitosan is 0.25 g L-1 in optimized activity at pH of 5.5.


O crescimento expressivo do setor industrial nas últimas décadas acarretou o aumento do volume de resíduos, que, se não forem destinados adequadamente, poderão causar sérios problemas ambientais. No contexto da aquacultura, efluentes líquidos com um alto teor orgânico são gerados em grandes quantidades nas indústrias de processamento de pescado, e, se seu descarte for realizado de maneira inadequada, há sérios prejuízos ao ambiente. O objetivo geral deste estudo foi avaliar: a eficiência de remoção de DQO e DBO, além da influência sobre o pH, a alcalinidade, os cloretos, a amônia, o nitrito, o nitrato, o fosfato, a turbidez, os sólidos totais, fixos e voláteis, bem como do efluente do processamento de pescado tratado por coagulação e da sedimentação, utilizando-se o coagulante natural quitosana. O pH 5,5, seguido pelo pH 6, apresentou melhores resultados para uso do coagulante quitosana no processo de tratamento por coagulação de efluente de indústria de processamento de pescado. A quitosana não atua sobre os parâmetros STV, alcalinidade, cloretos, amônia, nitrito, nitrato e fosfato, independentemente da dosagem utilizada. Mas atua sobre DBO, DQO, ST, STF e turbidez. Assim, a melhor dosagem de quitosana é 0,25 g L-1 em atividade otimizada no pH de 5,5.

The detection of beta-lactamase production and biofilm formation genes by polymerase chain reaction (PCR). Two multidrug resistant (MDR) and 3 single-drug resistant (SDR) strains of P. aeruginosa were identified. The cockle Cerastoderma edule was exposed to four concentrations (5, 10, 20 and 70 µg L-1) of carbamazepine (CBZ). This anticonvulsant was found to alter the mussel behavior of by reducing its clearance rate (CR). Analysis of CBZ accumulation in tissues of C. edule was carried out using HPLC-UV after 48 or 96 hours of exposure. In addition, an overproduction of H2O2 by the bivalves was detected following exposure to CBZ but nitrite levels remained unchanged. Moreover, superoxide dismutase and catalase activities showed a significant increase in relation to their contact with CBZ. The activity of the biotransformation enzyme gluthatione-S-transferase did not change during exposure. Malondialdehyde (MDA) levels indicating cellular damage, increased when bivalves were exposed to 20 and 70 µg l-1 of carbamazepine for 96 h CBZ. The results also indicate that acetylcholinesterase activity (AChE) was inhibited in all CBZ concentrations during the 48 h exposure period. However, during the 96 h exposure period, AChE was only inhibited at the highest concentration. Further studies are needed now for more exploration of the toxicity of CBZ since it could be bioaccumulable throughout the food web and may affect non-target organisms.

Arthropod venoms are potential sources of bioactive substances, providing tools for the validation of popular use and new drugs design. Ants belonging to the genus Dinoponera are used in the folk medicine to treat inflammatory conditions. It was previously demonstrated that the venom of the giant ant Dinoponera quadriceps (DqV), containing a mixture of polypeptides, elicit antinociceptive effect in mice models of chemical, mechanical and thermal nociception. The aim of this study was to evaluate DqV antiinflammatory and antihypernociceptive effects in a mice model of traumatic cutaneous wound. Colonies of D. quadriceps were collected in the 'Serra de Maranguape' (State of Ceará, northeastern Brazil), a small mountain range located on the coastal zone, and the venom secreted by the ant glands was extracted with capillary tubes, further lyophilized and maintained at -20 ± 1ºC until use. Wounds were performed in the dorsum of Swiss mice. Animals received intravenous (i.v.) injection of DqV (50 µg kg-1 day-1) during 3 days for evaluation of inflammatory parameters present in the wounds: hypernociception, leukocyte infiltrate, myeloperoxidase activity, nitrite/nitrate content. Data was tested by two-way ANOVA and Bonferroni's post-hoc test. DqV reduced (2.7 folds) hypernociception at 48 hours, leukocyte infiltration by 65% at 6 hours and myeloperoxidase activity by 60% at 0.5 hour after wound induction. In conclusion, the venom extracted from D. quadriceps glands attenuates inflammation and hypernociception in mice cutaneous wounds.(AU)

The use of natural colorants in meat products can keep its coloring attractive as well as meet the demand from consumers interested in products that are healthier. Nitrite is added as a preservative to these products and it is also a precursor of the generation of a specifically desired color in meat products. However, its use has been associated with the development of certain kinds of cancer. The objective of this study was to evaluate the effect of reduced levels of nitrite in Brazilian sausage (linguiça) with natural colorants on the physicochemical and sensorial properties following frozen storage. Nine treatments were tested: 0.015 % nitrite (C150), 0.0075 % nitrite (C75) and 0 % nitrite (C0), 0.0075 % nitrite and 1.5 % microencapsulated betalain (BME75), 0 % nitrite and 1.5 % microencapsulated betalain (BME0), 0.0075 % nitrite and 0.19 % commercial betalain (BCP75), 0 % nitrite and 0.19 % commercial betalain (BCP0), 0.0075 % nitrite and 0.02 % of cochineal carmine (CC75) and 0 % nitrite and 0.02 % of cochineal carmine (CC0). Oxidative stability, residual nitrite and instrumental color were the determinant factors. Sensory analysis was directed at color, texture, taste and global acceptance. Natural colorants did not have any antioxidant effect. Sausages prepared with betalain and 0.0075 % nitrite showed the highest a* values. The addition of colorants improved both color and overall acceptance. The addition of betalain and cochineal carmine with 0.0075 % nitrite proved to be effective in achieving high acceptability in Brazilian sausage (linguiça).

Total ammonia nitrogen (TAN) was significantly higher in NT+ than in T+. In the C:N-ratio adjusted tanks, the presence of fish shortened the control of TAN in several days. As nitrite declined in T+, it increased in NT+. The final concentrations of TSS in T+, T- and NT+ were 236 ± 29 mg L-1, 100 ± 32 mg L-1, and 40 ± 22 mg L-1, respectively (p < 0.05). It can be concluded that stable nitrifying and heterotrophic bacterial communities could be attained in BFT aquaculture tanks, before the end of the 4th week of culture, if the bacterial growth is fostered by early Nile tilapia stocking.(AU)

Among treatments, E50 and E75 obtained higher maximum cell densities and presented an exponential growth rate similar to the control treatment. Regarding the concentrations of nitrogen compounds, a significant reduction was observed in all treatments, with an NO3-Nuptake of 99%, followed by 80% of TAN and 90% of NO2-N. Thus, giving the results obtained, besides being able to grow in wastewater, A. platensis can also be used in bioremediation processes, confirming the potential of this species.
