Resumo

Background: Hepatocellular carcinoma is a primary malignant tumor of the liver tissue and its occurrence in birds is considered rare. The tumor can occur as a single mass leading to hepatomegaly, or as multiple nodules in the liver. In animals of the genus Amazona, only 1 case of metastatic hepatocellular carcinoma has been reported in the United States, therefore, little is known about its epidemiology and clinicopathological aspects in these species. In this context, the aim of this work was to describe a case of metastatic hepatocellular carcinoma in an Amazona aestiva. Case: A blue-fronted amazon parrot (Amazona aestiva) was referred to necropsy after being found dead in its enclosure. On examination, it presented cachectic body score. Examination of the coelomic cavity, revealed a serous translucent fluid and adhesions between the liver and peritoneum.A red mass restricted to the right hepatic lobe and raised to the capsular surface, interspersed with whitish and dark red multifocal areas was observed. When cut, this mass was soft, protruding, multilobulated, whitish and with a friable reddish center. Additionally, on the dorsal surface of the left lung lobe, there was a rounded, well defined, whitish, and soft nodule. Microscopically, partial replacement of the hepatic parenchyma was observed by neoplastic proliferation of cuboidal epithelial cells, organized in mantle and supported by a scarce fibrovascular stroma. Cells have large, eosinophilic, well-delimited cytoplasm, with a central, oval nucleus, loose chromatin, and evident nucleolus. Moderate pleomorphism was characterized by anisocytosis, anisokaryosis, and aberrant nuclei. In the lung, a focally extensive mass with a pattern similar to that seen in the liver was observed. In the kidney, multifocal neoplastic emboli were noted. Liver immunohistochemistry was performed. Positive and negative controls were used to validate the reaction; however, there was no immunolabelling for the evaluated antibodies. Discussion: The histopathological characteristics observed in this study favored the diagnosis of hepatocellular carcinoma (HCC) with metastasis to kidney and lung. Primary liver tumors are rare in wild birds. In ducks, experimental studies have pointed aflatoxins and the duck hepatitis B virus as oncogenic agents, however, in birds of the genus Amazona, there are no studies that evaluate predisposing factors to the development of liver carcinoma. Macroscopically, hepatocellular carcinoma may present in massive, nodular or diffuse forms. In birds, the right lobe is the largest, which may suggest that this lobe is more prone to the development of HCC, as seen in the present case. The solid form, similar to that observed in this report, seems to be more commonly observed, as seen in the wild bird reports consulted. Metastases most often spread hematogenous, and in the present report there was metastasis to kidneys and lungs, which is a common feature for this neoplasm. In the present case, there was no labeling by any of the antibodies, perhaps because of their aggressiveness, associated with autolytic factors that prevent the labeling of antibodies, in addition to the specificity in the antibodyantigen relationship. This tumor must be differentiated from other liver tumors such as cholangiocarcinoma, and also the well-differentiated hepatocellular adenoma, in addition to non-neoplastic conditions. HCC should be considered as a differential diagnosis for Amazona aestiva found dead in the enclosure without previous clinical signs. This neoplasm is rare in Amazon parrots and reports should be encouraged in order to contribute to the understanding of the epidemiological and clinicopathological aspects of the tumor.

Resumo

Neoplasias are important causes of illness and death in domestic animals. However, in birds, they are less frequent compared with other species such as dogs and cats. Moreover, benign tumors of bone are even less frequent in captive birds being considered sporadic, which make it a poorly understood entity due to the lack of clinical and histopathological data. Thus, we present a case of an osteoma in a Peach-faced lovebird (Agapornis roseicollis) approaching clinical and morphological aspects. A two-years-old lovebird was referred to the veterinary hospital with a 4-month evolution round to oval tumoral mass on the wing. The nodule was firm, painful when manipulated. Radiographic examination revealed a radiopaque mass in the radius and ulna bones. The animal has died during the surgical procedures. The affected wing was submitted to pathology service and histopathology revealed an osteoma.


As neoplasias são importantes causas de doença e morte em animais domésticos. No entanto, em aves, são menos frequentes em comparação com outras espécies, como cães e gatos. Além disso, tumores ósseos benignos são ainda menos frequentes em aves de cativeiro sendo considerados esporádicos, o que o torna uma entidade pouco compreendida devido à falta de dados clínicos e histopatológicos. Assim, apresentamos um caso de osteoma em Periquito cara-de-pêssego (Agapornis roseicollis) abordando aspectos clínicos e morfológicos. Um periquito de dois anos foi encaminhado ao hospital veterinário com uma massa tumoral ovalada na asa com evolução de 4 meses. O nódulo era firme, doloroso quando manipulado. O exame radiográfico revelou uma massa radiopaca nos ossos rádio e ulna. O animal morreu durante os procedimentos cirúrgicos. A asa afetada foi submetida ao serviço de patologia e a histopatologia revelou um osteoma.

Background: Industrial poultry farming has developed progressively in Brazil, conferring the country a prominent position on the national and international scene. Likewise, alternative poultry farming is an important economic activity for small-scale family farmers. However, shortcomings related to sanitary management lead to increased occurrence of avian diseases, such as those caused by Salmonella spp. Despite salmonellosis has been described in industrial establishments, reports in alternative farms are less common, therefore the objective of this study was to describe the occurrence of salmonellosis in free-range chickens in the municipality of Barra, Western Bahia, Brazil. Cases: The poultry farmer reported the occurrence of diarrhoea in his chicken flock since the acquisition of the batch of chicks. Initially, carrying out the medicinal treatment of the birds, there was clinical improvement, however, successive recurrences of clinical signs occurred, such as diarrhoea, apathy, anorexia, and death of some birds. Upon learning about the case, an epidemiological investigation of the flock was carried out, and it was noted that some of the birds were retracted, apathetic, anorexic, and a lot of diarrheic faeces of a yellowish-white appearance were also observed. The entire flock had a history of vaccination against diseases: newcastle disease, infectious bronchitis, gumboro disease, and fowlpox. For better evaluation, five birds were necropsied, enabling the observation that the animals had a good body score. However, the necropsy revealed lesions such as splenomegaly, hepatomegaly, and enteritis in the three birds initially analysed (Animals 1, 2, and 3). The other birds (Animals 4 and 5) were submitted to evaluation for Eimeria sp. oocysts by means of scrapings from the intestinal mucosa, and there were no structures compatible with oocysts. Faecal samples were collected from another six birds in the flock for coproparasitological examination, and the presence of oocysts was not detected. Finally, sera from 20 birds in the flock were collected for the Rapid Serum Agglutination Test (SAR) for the detection anti-Salmonella Pullorum antibodies. Discussion: The diagnosis was based on clinical evaluation, post mortem pathological findings of the necropsied birds, epidemiological data and confirmed with SAR testing, whereby 11 birds were seropositive for Salmonella enterica subspecies enterica serovar Pullorum. The prevalence of S. Pullorum is poorly described in alternative farming. Based on the farmer's report, it is believed that the chicks were purchased already infected, because birds from the same batch, also purchased by neighbouring producers, showed the same clinical signs. The unsatisfactory sanitation in the flock was another factor that may have favoured the persistence of the bacteria, since, the lack of removal of organic matter is a source of nutrients for microorganisms, and this may have favoured the multiplication and maintenance of the bacteria in the down feathers, feed, and water. The elimination of S. Pullorum through the faeces, in addition to the density of the birds, may have led to transmission to the other healthy birds. However, the knowledge and adoption of prophylactic measures in free-range chicken farms is a crucial factor in minimizing the occurrence of outbreaks and thus avoiding a future public Health Problem.

ABSTRACT: Avian colibacillosis is an acute and globally occurring infectious disease of domestic and wild birds caused by Escherichia coli, and it is associated with considerable economic losses mainly due to the morbidity and mortality associated. The present study aimed to describe the pathological, bacteriological and immunohistochemical aspects of avian colibacillosis in broiler chickens of Mozambique. Forty-nine broiler chicken presented anorexia, decreased weight gain, ataxia, diarrhea, dyspnea, and death in a clinical course of 3-5 days. The birds were raised in five farms (small, medium and large farms) with manual and automatic breeding system, with flocks ranging from 100 to 20,000 birds. At the necropsy, all birds had poor body condition, and the pericardium and the Glissons capsule of all avian exhibited different degrees of adherence often associated with severe fibrin deposition. The thoracic and abdominal air sacs were thickened and adhered to the costal wall. Mild, moderate or marked hepatomegaly associated with white pinpoint multifocal areas (100%, 49/49) and mild to moderate splenomegaly in 75.5% (37/49) with a mottled surface were observed. The lungs and kidney were enlarged and reddish. Histologically, a multiorgan fibrinoheterophilic polyserositis was observed in 75.5% of the cases (37/49), which were characterized by inflammatory infiltrates composed mainly of degenerative heterophils, macrophages and plasma cells, associated with fibrin deposits and intermixed by coccobacillary bacterial basophilic aggregates. These affected mainly the pericardium (28.6%, 14/49), the pleura (18.4%, 9/49), the Glissons capsule (10.2%, 5/49), the ventriculus (10.2, 5/33), and the proventriculus (8.2%, 4/49) serosa. Multifocal to coalescing areas of coagulative necrosis associated with similar inflammatory cells were observed mainly in the spleen (28.6%, 14/49), liver (24.5%, 12/49), and intestines (22.4%, 11/49). A similar infiltrate was also observed affecting the the lungs (16.3%, 8/49), the kidney (16.3%, 8/49) and the myocardium (14.3%, 7/49). Isolation and identification of E. coli was obtained in 12 cases through bacterial culture. Some organs (2 cases of each farms) were selected and submitted to immunohistochemistry anti-E. coli, and a positive stain was observed in all tested cases in liver (3/3), heart (4/4), spleen (1/1), lungs (4/4), intestines (4/4), bursa of Fabricius (1/1), ventriculus (1/1), and proventriculus (1/1) tissue sections. These results demonstrate that E. coli was the cause of mortality in these birds. Therefore, biosecurity and management measures should be employed to prevent and control the disease occurrence in Mozambiques poultry farming.


RESUMO: A colibacilose aviária é uma doença aguda de ocorrência mundial que acomete aves domésticas e silvestres, causada por Escherichia coli e resulta em perdas econômicas consideráveis devido à elevada morbidade e mortalidade das aves. O presente estudo teve o objetivo de descrever os aspectos patológicos, bacteriológicos e imuno-histoquímicos de colibacilose aviária em frangos de corte de Moçambique. Um total de 49 frangos de corte apresentaram anorexia, baixo ganho de peso, ataxia, diarreia, dispneia e morte em um curso clínico de 3 a 5 dias. As aves eram provenientes de 5 granjas (pequenas, média e grandes), com sistema de criação manual e automático, com rebanhos que variavam de 100 a 20.000 aves. À necropsia, todas as aves exibiam condição corporal ruim a caquética, além de pericárdio e cápsula de Glisson de todas aves (100%; n=49) com diferentes graus de aderência e deposição de fibrina de forma difusa acentuada. Os sacos aéreos torácicos e abdominais estavam espessados e aderidos à parede costal. Foi observado ainda hepatomegalia discreta, moderada a severa frequentemente associada com áreas multifocais puntiformes brancacentas (100%; 49/49), e esplenomegalia discreta a moderada, associado a áreas multifocais moteadas (75,5%; 37/49). Os pulmões e rins estavam aumentados e com coloração avermelhada. Histologicamente, observou-se majoritariamente serosite fibrinoheterofílica em 75,5% dos casos (37/49), caracterizadas por infiltrado inflamatório composto por heterófilos degenerados, macrófagos, linfócitos e plasmócitos, com deposição de fibrina entremeada por uma miríade de estruturas bacterianas cocobacilares. Esta lesão foi observada principalmente em pericárdio (28,6%; 14/49), pleura (18,4%; 9/49), cápsula de Glisson (10,2%; 5/49), ventrículo (10,2; 5/33) e em proventrículo (8,2%; 4/49). Áreas multifocais a coalescentes de necrose de coagulação associada a infiltrado inflamatório semelhante ao descrito foi observado principalmente no baço (28,6%; 14/49), fígado (24.5%; 12/49), e intestinos (8,2%; 4/49). Um infiltrado inflamatório semelhante também foi visualizado em pulmões (16,3%; 8/49), rins (16,3%; 8/49) e miocárdio (14,3%; 7/49), Colônias puras de E. coli foram identificadas e isoladas em 12 casos. Alguns órgãos (2 de cada granja) foram submetidos ao exame imuno-histoquímico anti-E. coli e marcação positiva foi visualizada em todos casos testados, como em fígado (3/3), coração (4/4), baço (1/1), pulmão (4/4), intestinos (4/4), bursa de Fabricius (1/1), rim (1/1), ventrículo (1/1) e proventrículo (1/1). Estes resultados demonstram que E. coli foi a causa de morte destas aves. Sendo assim, a adoção de boas medidas de biosseguridade e de manejo são indispensáveis para a prevenção e controle da ocorrência da doença nas granjas de frango de corte de Moçambique.

The genus Brachyspira corresponds to the group of bacteria formerly classified into the genus Serpulina and includes several commensal and pathogenic intestinal spirochetes that affect pigs, poultry, and other animal species, including humans. In birds, some pathogenic species of this genus causes a condition known as avian intestinal spirochetosis, which remains under diagnosed, thereby causing serious economic losses. Brachyspira is a fastidious organism that necessitates the employment of fast and efficient identification techniques. The aim of this study was to identify Brachyspira spp. using histology, immunohistochemistry (IHC) and fluorescence in situ hybridization (FISH) in formalin-fixed paraffin embedded (FFPE) tissue samples from the cecum of commercial poultry. Samples were collected from129 birds aged between 35 and 45 days from commercial broiler farms. For evaluation, routine histology processing (H&E) and the histochemical technique, periodic acid–Schiff (PAS) were done. Additionally, FFPE tissue samples were evaluated for FISH and IHC. The histological lesions were analyzed and graded after H&E staining, and the goblet cells were counted and compared using PAS staining with the positive and negative samples obtained through FISH and IHC. For FISH, probes labeled with Brachyspira spp., B. pilosicoli, B. hyodysenteriae, and B. intermedia were used, where as rabbit polyclonal antibody specific for Brachyspira spp. was used for IHC. Of 129 samples, 82 were positive with IHC and 86 were positive with FISH. The samples positive for the genus Brachyspira in the FISH technique were tested for B. pilosicoli, B. hyodysenteriae, and B. intermedia in which 56 were positive for B. pilosicoli, 75 for B. hyodysenteriae and 80 for B. intermedia. There was an increase in goblet cells in the samples positive for FISH and IHC. The techniques used were effective and gave corresponding results, thus serving as a fast and efficient tool for diagnosis.


O gênero Brachyspira corresponde ao grupo de bactérias anteriormente classificadas no gênero Serpulinae inclui várias espiroquetas intestinais comensais e patogênicas que afetam suínos, aves e outras espécies animais, incluindo humanos. Em aves, algumas espécies patogênicas desse gênero causam uma condição conhecida como espiroquetose intestinal aviária, que permanece sub-diagnosticada, causando sérios prejuízos econômicos. Brachyspira é um organismo fastidioso que necessita do emprego de técnicas de identificação rápidas e eficientes. O objetivo deste estudo foi identificar Brachyspira spp. usando histologia, imunohistoquímica (IHQ) e hibridização fluorescente in situ (FISH) em amostras de tecido fixado em formalina e embebido em parafina (TFEP) do ceco de aves comerciais. As amostras foram coletadas de 129 aves com idades entre 35 e 45 dias em granjas comerciais. Para avaliação, o processamento histológico de rotina (H&E) e a técnica histoquímica, ácido periódico-Schiff (PAS) foram realizados. Além disso, as amostras de tecido TFEP foram avaliadas para FISH e IHC. As lesões histológicas foram analisadas e graduadas após coloração H&E, e as células caliciformes contadas e comparadas pela coloração PAS com as amostras positivas e negativas obtidas por FISH e IHC. Some organs (2 cases of each farms) were selected and submitted to immunohistochemistry anti-E. coli, and a positive stain was observed in all tested cases in liver (3/3), heart (4/4), spleen (1/1), lungs (4/4), intestines (4/4), bursa of Fabricius (1/1), ventriculus (1/1), and proventriculus (1/1) tissue sections. These results demonstrate that E. coli was the cause of mortality in these birds. Therefore, biosecurity and management measures should be employed to prevent and control the disease occurrence in Mozambique's poultry farming.(AU)


A colibacilose aviária é uma doença aguda de ocorrência mundial que acomete aves domésticas e silvestres, causada por Escherichia coli e resulta em perdas econômicas consideráveis devido à elevada morbidade e mortalidade das aves. O presente estudo teve o objetivo de descrever os aspectos patológicos, bacteriológicos e imuno-histoquímicos de colibacilose aviária em frangos de corte de Moçambique. Um total de 49 frangos de corte apresentaram anorexia, baixo ganho de peso, ataxia, diarreia, dispneia e morte em um curso clínico de 3 a 5 dias. As aves eram provenientes de 5 granjas (pequenas, média e grandes), com sistema de criação manual e automático, com rebanhos que variavam de 100 a 20.000 aves. À necropsia, todas as aves exibiam condição corporal ruim a caquética, além de pericárdio e cápsula de Glisson de todas aves (100%; n=49) com diferentes graus de aderência e deposição de fibrina de forma difusa acentuada. Os sacos aéreos torácicos e abdominais estavam espessados e aderidos à parede costal. Foi observado ainda hepatomegalia discreta, moderada a severa frequentemente associada com áreas multifocais puntiformes brancacentas (100%; 49/49), e esplenomegalia discreta a moderada, associado a áreas multifocais moteadas (75,5%; 37/49). Os pulmões e rins estavam aumentados e com coloração avermelhada. Histologicamente, observou-se majoritariamente serosite fibrinoheterofílica em 75,5% dos casos (37/49), caracterizadas por infiltrado inflamatório composto por heterófilos degenerados, macrófagos, linfócitos e plasmócitos, com deposição de fibrina entremeada por uma miríade de estruturas bacterianas cocobacilares. Esta lesão foi observada principalmente em pericárdio (28,6%; 14/49), pleura (18,4%; 9/49), cápsula de Glisson (10,2%; 5/49), ventrículo (10,2; 5/33) e em proventrículo (8,2%; 4/49). Áreas multifocais a coalescentes de necrose de coagulação associada a infiltrado inflamatório semelhante ao descrito foi observado principalmente no baço (28,6%; 14/49), fígado (24.5%; 12/49), e intestinos (8,2%; 4/49). Um infiltrado inflamatório semelhante também foi visualizado em pulmões (16,3%; 8/49), rins (16,3%; 8/49) e miocárdio (14,3%; 7/49), Colônias puras de E. coli foram identificadas e isoladas em 12 casos. Alguns órgãos (2 de cada granja) foram submetidos ao exame imuno-histoquímico anti-E. coli e marcação positiva foi visualizada em todos casos testados, como em fígado (3/3), coração (4/4), baço (1/1), pulmão (4/4), intestinos (4/4), bursa de Fabricius (1/1), rim (1/1), ventrículo (1/1) e proventrículo (1/1). Estes resultados demonstram que E. coli foi a causa de morte destas aves. Sendo assim, a adoção de boas medidas de biosseguridade e de manejo são indispensáveis para a prevenção e controle da ocorrência da doença nas granjas de frango de corte de Moçambique.(AU)

Avian colibacillosis is an acute and globally occurring infectious disease of domestic and wild birds caused by Escherichia coli, and it is associated with considerable economic losses mainly due to the morbidity and mortality associated. The present study aimed to describe the pathological, bacteriological and immunohistochemical aspects of avian colibacillosis in broiler chickens of Mozambique. Forty-nine broiler chicken presented anorexia, decreased weight gain, ataxia, diarrhea, dyspnea, and death in a clinical course of 3-5 days. The birds were raised in five farms (small, medium and large farms) with manual and automatic breeding system, with flocks ranging from 100 to 20,000 birds. At the necropsy, all birds had poor body condition, and the pericardium and the Glisson's capsule of all avian exhibited different degrees of adherence often associated with severe fibrin deposition. The thoracic and abdominal air sacs were thickened and adhered to the costal wall. Mild, moderate or marked hepatomegaly associated with white pinpoint multifocal areas (100%, 49/49) and mild to moderate splenomegaly in 75.5% (37/49) with a mottled surface were observed. The lungs and kidney were enlarged and reddish. Histologically, a multiorgan fibrinoheterophilic polyserositis was observed in 75.5% of the cases (37/49), which were characterized by inflammatory infiltrates composed mainly of degenerative heterophils, macrophages and plasma cells, associated with fibrin deposits and intermixed by coccobacillary bacterial basophilic aggregates. These affected mainly the pericardium (28.6%, 14/49), the pleura (18.4%, 9/49), the Glisson's capsule (10.2%, 5/49), the ventriculus (10.2, 5/33), and the proventriculus (8.2%, 4/49) serosa. Multifocal to coalescing areas of coagulative necrosis associated with similar inflammatory cells were observed mainly in the spleen (28.6%, 14/49), liver (24.5%, 12/49), and intestines (22.4%, 11/49). A similar infiltrate was also observed affecting the the lungs (16.3%, 8/49), the kidney (16.3%, 8/49) and the myocardium (14.3%, 7/49). Isolation and identification of E. coli was obtained in 12 cases through bacterial culture. Background: Free-range chickens are quite common in Brazil. In this alternative rearing system, the animals are rustic and raised in an extensive system. Free access to "bare soil" results in the increased occurrence of intestinal parasites since larvae and / or eggs of helminths and protozoa oocysts find favorable conditions for their survival and dissemination in the soil. Although the occurrence and importance of parasitic infections in free-range chickens is well known, the objective of this study is to report an outbreak of endoparasites in free-range chickens in the municipality of Barra - BA, in view of bird susceptibility associated with scarcity of studies in western Bahia. Cases: The chickens were kept free, in a bare soil yard in a household at the urban perimeter of the municipality of Barra - BA. Feeding consisted of whole corn grains, thrown directly in the soil. The drinking fountains were dirty and the animals had no history of vaccination or deworming. Symptoms Anorexia, difficulty in eating and in locomotion, presence of seromucous secretion in the oral cavity, emaciation and diarrhea were all observed symptoms. One of the birds presented excessive vocalization, drowsiness and flaccid paralysis of the neck. Necropsy was performed on 3 chickens: 2 females (cases 1 and 2) and 1 male (case 3). Macroscopic analysis revealed the presence of seromucous secretion in the upper respiratory tract of all animals. Specimens of Ascaridia galli were observed in cases 1 and 2, Heterakis gallinarum in cases 2 and 3, Raillietina sp. in cases 2 and 3 and Davainea proglottina in case 1. Microscopically, the animals had an inflammatory infiltrate in the liver and intestines. Some animals presented necrosis of the tracheal epithelial cells, as well as of the epithelial cells present at the apex of the villi. No significant results were found in the coproparasitological exam. Discussion: The diagnosis of endoparasitosis in this outbreak was based on epidemiological, clinical and pathological findings. The prevalence of intestinal parasitosis in free-range chickens is linked to factors such as age, high animal density, absence of sanitary hygienic measures, as well as environmental temperature and humidity. The appearance of injuries in the intestinal mucosa is influenced by characteristics such as parasitic load, concomitant infections, age and the host's immune status. During necropsy of the birds were found 2 helminths of the Nematoda class (Ascaridia galli and Heterakis gallinarum) and 2 of the Cestoda class (Davainea proglottina and Raillietina sp.). A. galli, seen in 2 cases, is considered low pathogenicity for adult chickens, however young birds are susceptible and can die due to intestinal obstruction and hemorrhages. H. gallinarum is responsible for causing typhlitis, with diarrhea and weight loss, this helminth was found in 2 animals in the present study, however only 1 had changes in the digestive tract. Davainea proglotina and Raillietina spp. might cause, respectively, severe hemorrhagic enteritis and nodule formation in the small intestine mucosa. In Brazil, even though it is notable that intestinal parasitism is one of the key problems in alternative poultry farming, there are few studies that evaluate the presence of endoparasites in chickens raised in alternative production systems, with animals being more frequently exposed to nematodes and cestodes. The multiparasitism observed in this study probably stems from flaws in the rearing system, mainly related to sanitary hygiene management. Therefore, the reduction in the occurrence of these helminths is closely related to the performance of basic prophylactic measures, such as offering good quality food and water in clean containers, separating lots by age, performing sanitary emptiness and deworming.(AU)

In India, increasing incidence of Marek’s Disease Virus (MDV) outbreaks are being reported even in vaccinated poultry farms. Hence identifying the new emerging pathotype of MDV is necessary for successful control through vaccination. Birds received in the post mortem section of The Avian Disease Laboratory, Indian Veterinary Research Institute, were screened for the presence of MDV by collecting neoplastic tissues, spleen and feather follicles. Screening was carried out by polymerase chain reaction (PCR) and histopathological examination. Among the tested 150 birds’ tissue samples, 35 bird tissue samples were found positive for MDV. Based on pathotyping specific PCR, it was found that 34 birds tissues were affected virulent MDV and one birds tissue was affected with very virulent MDV. Since, HVT vaccine will not protect the very virulent pathotype, combined vaccine of SB-1 and HVT can be administered to control the very virulent MDV. Among the MD infected birds, neoplastic liver is most commonly encountered. Spleen tissue samples was found to be more suitable for the DNA isolation for PCR.

The disease affected 23,409 layers (from day one until 100 weeks) leading to mortality of 16.5% (5,049/30,600) of the flock. Twenty laying hens were clinically examined, and blood samples were collected and tested by Enzyme-Linked Immunosorbent Assay (ELISA). Five laying hens with neurological signs were necropsied and samples of the brain, cerebellum, spinal cord, sciatic nerve, heart, ceca tonsils, proventriculus and liver were collected, fixed in formalin buffered solution and processed according to routine methods for histological examination. Clinical signs were observed in approximately 60% of the chicken flock and included depression, ataxia, head and neck tremors, head tilt, motor incoordination and corneal opacity and of lens. All laying hens presented seroconversion for AE, with a geometric mean titer (GMT) of 6,854 and CV of 9.6%. At necropsy, there were bilateral opacity of the cornea and lens and the blood vessels of the cerebral cortex were hyperemic. The main microscopic lesions in the central nervous system consisted of red and shrunken neurons in the cerebral cortex, cerebellum and spinal cord. Significant areas of gliosis were observed in the cerebral cortex and in the molecular layer of the cerebellum. Hyperplasia of the proventricular lymphoid tissue and the ceca tonsils were observed with different degrees of intensity. In Northeastern Brazil, there havent been previous reports of AE in commercial laying hens and this outbreak possibly occurred by inefficient immunization of laying breeders at the genetic supply company.(AU)

This study describes an outbreak of avian poxvirus disease in previously pox-vaccinated turkeys in Brazil. The turkeys had suggestive gross lesions of cutaneous avian poxvirus in the skin of the head and cervical area without changes in the flock mortality rates. In the slaughterhouse, 30 carcasses were removed from the slaughter line to collect tissue from cutaneous lesions for histological analyses and characterization of the virus. The virus was identified by conventional polymerase chain reaction (PCR) and subsequent gene sequencing. Acanthosis, hyperkeratosis, and hydropic degeneration were seen on skin histopathology. Eosinophilic intracytoplasmic inclusion bodies (Bollinger) on keratinocytes were observed in 46.6% of the samples. Avian poxvirus DNA was detected on PCR in 83.3% of the total samples. PCR associated with histopathology had 93.3% of positivity for avian poxvirus. In the phylogenetic study, samples show 100% matching suggesting that the outbreak occurred by a single viral strain and was different from those strains affecting other wild birds such as canaries and sparrows. A single mutation (Adenine for Guanine) was detected in our study's strain and in the strains of turkey, chickens, and vaccine strains published in GenBank. Also, when the sequence strain of the present study and sequences from GenBank of canarypox and sparrowpox strains were aligned, a Thymine was found replacing the Adenine or Guanine. The in ovo vaccination method as single-use in turkeys of this study apparently did not provide adequate protection against avianpox disease, but additional vaccination administered by wing-web when turkeys were 45-60 days old in the new flocks controlled the disease. In the subsequent year, new cases of this disease were not found. It was not possible to confirm the source of the virus strain, but infection with a field strain derived from chickens is one possibility, considering the poultry farm population in the area and biosecurity aspects. For wide characterization of avipoxvirus and differentiation among strains, the complete sequence of the viral genome is required.(AU)


Este estudo descreve um surto de bouba aviária em perus previamente vacinados contra poxvirus aviário no Brasil. Os perus apresentaram lesões macroscópicas, sugestivas de bouba aviaria cutânea, na pele da cabeça e região cervical sem alteração nas taxas de mortalidade do lote. No abatedouro, 30 carcaças foram retiradas da linha de abate para coleta de dois fragmentos de pele com lesões para análise histológica e caracterização do vírus. A identificação do vírus foi realizada por PCR convencional e posterior sequenciamento. No exame histopatológico das lesões de pele, houve acantose, hiperqueratose e degeneração hidrópica. Corpúsculos de inclusão intracitoplasmáticos eosinofílicos (Bollinger) foram encontrados em 46,6% das amostras. A técnica de PCR detectou o DNA do vírus da bouba aviária em 83,3% do total de amostras. PCR associado com a histopatologia resultou em 93,3% de positividade para o vírus da bouba aviária. No estudo filogenético, as sequências resultaram em 100% de identidade, sugerindo que o surto ocorreu por uma única estirpe de vírus diferenciada das outras estirpes que acometem canários e pardais. Uma única mutação (Adenina para Guanina) foi detectada nas estirpes deste estudo e nas sequências de perus, galinhas e estirpes vacinais publicadas no GenBank. Além disso, quando a sequência da estirpe do presente estudo e as sequências das estirpes de canarypox e sparrowpox foram comparadas, a Timina foi encontrada em substituição a Adenina ou Guanina. A vacinação in ovo em dose única utilizada nos perus deste estudo aparentemente não forneceu proteção adequada contra a doença causada pelo poxvirus aviário. Entretanto, a revacinação na membrana da asa em perus com 45-60 dias de idade dos novos lotes controlou a doença. No ano subsequente, novos casos desta doença não foram registrados. Não foi possível confirmar a origem da estirpe viral, mas estirpes de campo oriundas de galinhas seria uma possibilidade, considerando a população na área e os aspectos de biosseguridade. Para caracterização ampla do avipoxvirus e diferenciação entre as estirpes, a sequência completa do genoma viral é requerida.(AU)

In the phylogenetic study, samples show 100% matching suggesting that the outbreak occurred by a single viral strain and was different from those strains affecting other wild birds such as canaries and sparrows. A single mutation (Adenine for Guanine) was detected in our study's strain and in the strains of turkey, chickens, and vaccine strains published in GenBank. Also, when the sequence strain of the present study and sequences from GenBank of canarypox and sparrowpox strains were aligned, a Thymine was found replacing the Adenine or Guanine. The in ovo vaccination method as single-use in turkeys of this study apparently did not provide adequate protection against avianpox disease, but additional vaccination administered by wing-web when turkeys were 45-60 days old in the new flocks controlled the disease. In the subsequent year, new cases of this disease were not found. It was not possible to confirm the source of the virus strain, but infection with a field strain derived from chickens is one possibility...(AU)


Este estudo descreve um surto de bouba aviária em perus previamente vacinados contra poxvirus aviário no Brasil. Os perus apresentaram lesões macroscópicas, sugestivas de bouba aviaria cutânea, na pele da cabeça e região cervical sem alteração nas taxas de mortalidade do lote. No abatedouro, 30 carcaças foram retiradas da linha de abate para coleta de dois fragmentos de pele com lesões para análise histológica e caracterização do vírus. A identificação do vírus foi realizada por PCR convencional e posterior sequenciamento. No exame histopatológico das lesões de pele, houve acantose, hiperqueratose e degeneração hidrópica. Corpúsculos de inclusão intracitoplasmáticos eosinofílicos (Bollinger) foram encontrados em 46,6% das amostras. A técnica de PCR detectou o DNA do vírus da bouba aviária em 83,3% do total de amostras. PCR associado com a histopatologia resultou em 93,3% de positividade para o vírus da bouba aviária. No estudo filogenético, as sequências resultaram em 100% de identidade, sugerindo que o surto ocorreu por uma única estirpe de vírus diferenciada das outras estirpes que acometem canários e pardais. Uma única mutação (Adenina para Guanina) foi detectada nas estirpes deste estudo e nas sequências de perus, galinhas e estirpes vacinais publicadas no GenBank. Além disso, quando a sequência da estirpe do presente estudo e as sequências das estirpes de canarypox e sparrowpox foram comparadas, a Timina foi encontrada em substituição a Adenina ou Guanina. A vacinação in ovo em dose única utilizada nos perus deste estudo aparentemente não forneceu proteção adequada contra a doença causada pelo poxvirus aviário. Entretanto, a revacinação na membrana da asa em perus com 45-60 dias de idade dos novos lotes controlou a doença. No ano subsequente, novos casos desta doença não foram registrados. Poultry production is an activity of great importance in Brazilian economy, both due to the domestic consumption and the large amount of chicken meat exportation. Poultry activity modernization allowed the creation of animals in high density facilities, however, it facilitates the rapid dissemination of pathogens, which reduces the productivity rates. This review aims to highlight the avian reovirus, an important agent of arthritis in birds that has a worldwide distribution. The affected birds present a reduction in weight gain due to movement difficulties. In addition to arthritis, the virus may be related to a variety of pathological conditions, such as enteric and respiratory disorders, Hepatitis and myocarditis. The main prevention and control measure is the flock vaccination. Nevertheless, due to the avian reovirus great genetic variability, the vaccine may not be effective against circulating strains. This article aims to overview the virus biology, its variability and classification, and the infection pathology and diagnosis.


A avicultura é um setor de grande importância na economia brasileira tanto pelo aumento do consumo interno quanto pelo crescimento na exportação de carne de frango. A modernização da atividade avícola permitiu a criação adensada de animais, facilitando, no entanto, a rápida disseminação de patógenos que reduzem os índices de produtividade dos plantéis. Nesta revisão, é destacado o reovírus aviário, importante agente de artrite em aves que apresenta distribuição mundial. As aves acometidas apresentam redução no ganho de peso devido à dificuldade de locomoção. Além da artrite, o vírus pode estar relacionado a uma variedade de condições patológicas, como distúrbios entéricos e respiratórios, hepatite e miocardite. A principal forma de prevenção e controle é a vacinação do plantel. No entanto, devido à grande variabilidade genética do reovírus aviário, a vacina utilizada pode não ser eficiente contra estirpes que circulam no campo. O artigo traz uma visão geral sobre a biologia do vírus, sua variabilidade e propostas de classificação dos isolados, patologia da doença e diagnóstico da infecção.

Today, blood tests are an indispensable tool in avian medicine. This study aimed to describe and compare hematological and morphometric data of erythrocytes, leukocytes and thrombocytes of rhea, Rhea americana. To do so, 58 rheas of both sexes on two farms, one in Cachoeiro do Itapemirim, Espيrito Santo (ES), and the other in Sمo Carlos, Sمo Paulo (SP), were selected. Blood samples were taken and their RBC counts, PCV and Hb levels were determined and used in hematimetric index calculations. The total and differentiated leukocyte counts and the TPP and fibrinogen serum levels were also obtained. The results from the two farms analyzed were compared by means of the t test. There were differences in the parameters of the erythrocyte series between the two flocks because of a process of hypochromic macrocytic anemia observed among the rheas in ES. The values for the erythrocyte series and indexes were: RBC, 2.81 ± 0.15 x106/l; PCV, 44.20 ± 2.86%; Hb, 12.12 ± 0.74 g/dL; MCV, 15.75 ± 0.89 fL; MCH, 43.18 ± 1.82 pg; and MCHC, 27.44 ± 0.80 g/dL. The values for the leukocyte series were: WBC, 12,072 ± 4116 /L; heterophils, 64.10 ± 9.90%; eosinophils, 2.05 ± 2.06%; monocytes, 6.40 ± 2.99%; lymphocytes, 26.93 ± 9.62%; and basophils, 0.52 ± 1.27%. These can all be suggested as references for rheas reared in Brazil. Statistical differences were observed in erythrocyte length measurements, which were higher in the rheas in SP than in those in ES. Regarding width, the heterophils, lymphocytes and monocytes of the birds in ES were statistically greater than those of the birds in SP. It was concluded that rheas may exhibit morphometric alterations to blood cells and differences in blood elements according to the type of management to which they are subjected.(AU)


Atualmente exames de sangue são ferramentas indispensáveis na medicina aviária. Este estudo teve como objetivo descrever e comparar valores hematológicos e morfométricos de eritrócitos, leucócitos e trombócitos de emas, Rhea americana. Para isso, foram selecionadas 58 emas de ambos os sexos de dois criatórios, um em Cachoeiro do Itapemirim, Espírito Santo (ES) e o outro em São Carlos, São Paulo (SP). Amostras de sangue foram coletadas e realizadas a hematimetria, Hto e dosagem de Hb que foram utilizados nos cálculos dos índices hematimétricos. Também foram efetuadas as contagens total e diferencial de leucócitos, além da dosagem de proteínas total e do fibrinogênio séricos. Os resultados referentes às duas propriedades analisadas foram comparados pelo Teste t. Ocorreram diferenças nos parâmetros da série vermelha entre as aves dos dois criatórios devido a um processo de anemia macrocítica hipocrômica observado nas emas do ES. Os valores para a série vermelha e índices hematimétricos como: hematimetria, 2,81±0,15 x106/μl; Hto, 44,20±2,86%; Hb, 12,12±0,74 g/dL; VCM, 15,75±0,89 fL; HCM, 43,18±1,82 pg; CHCM, 27,44±0,80, além dos valores da série branca como: leucometria total, 12.072±4.116/μL, heterofilo, 64,10±9,90%, eosinófilo, 2,05±2,06%, monócito, 6,40±2,99%, linfócito, 26,93±9,62%, basófilo, 0,52±1,27% foram sugeridos como os de referência para as emas criadas no Brasil. Diferenças estatísticas foram observadas no comprimento de eritrócitos, o qual estava maior nas emas de SP do que aquelas do ES. Em relação à largura, os heterófilos, linfócitos e monócitos das aves do ES foram estatisticamente maior do que aquelas das aves de SP. Concluiu-se que emas podem apresentar alterações morfométricas de células sanguíneas e diferenças nos elementos do sangue de acordo com o tipo de manejo a que são submetidos.(AU)

Background: Infection of IBDV was reported to be endemic in worldwide including Malaysia and can be spread orally thru polluted fodder and water source, thus causing economic losses especially in commercial poultry industry. The infection resulted in depletion of B lymphocytes and subsequently destruction of the bursa which leaded to immunosuppression of the bird and it was postulated that the depletion of cells in the bursa was due to induction of apoptosis. In the current study, the infection of Malaysia isolated very virulent IBDV UPM0081 on IgM bearing B lymphocytes (IgM+ cells) from chicken spleen and bursa was compared. Materials, Methods & Results: A total of sixty eggs were obtained and raised until the age of 3 weeks old. The birds were divided into two groups (n = 30), which one of them served as control while IBDV strain UPM0081 was used to infect another group of birds at the concentration of 103 ELD50. The birds were observed and sacrificed at day 2, 4 and 5 post infections. Spleen and bursa of Fabricius were harvested and subjected to IgM+ cell enrichment using microbeads. The cell viability of enriched cells was assayed using MTT and cell cycle was analyzed using propidium iodide.
