Resumo

Linoleic (LA, 18:2) and alpha-linolenic (ALA, 18:3) acids are omega-6 and omega-3 polyunsaturated fatty acids considered essential to human and animal health. The optimum LA/ALA ratio for the production of fertile eggs in Japanese quail breeders has been little investigated. Thus, this study aimed to assess the effects of LA/ALA ratio on productive performance, egg quality, serum biochemical profile, body composition, and organ weight in Japanese quail. A completely randomized design was used. Birds were distributed into five treatments consisting of different LA/ALA ratios (1.48:1, 4.57:1, 7.63:1, 10.69:1, and 13.75:1), obtained by adding soybean (LA) and flaxseed (ALA) oils to the feed. Productive performance and egg quality were assessed in three cycles of 28 days each. At the end of the experiment, the birds were slaughtered and evaluated for biochemical profile, genital organ weights, and body composition. Data were subjected to analysis of variance and regression (p<0.05). LA/ALA ratio had no effect on productive performance or egg quality. Blood cholesterol in females and males and total triglycerides in females showed a quadratic response. Breeders fed the highest level of flaxseed oil (1.48:1) had the best serum levels of cholesterol and total triglycerides. Live weight and relative organ weights were not influenced by LA/ALA ratio. Differences in ash and crude protein levels were observed between groups, with LA increasing the deposition of these nutrients. It was concluded that LA/ALA ratio didn't affect productive performance or egg quality. However, based on the results of body composition, serum triglycerides, and cholesterol, it is recommended to use an LA/ALA ratio of 1.48:1 in diets.(AU)

This study aimed to evaluate the effects of partial replacement (750g/kg) of Tifton hay by two cactus cladodes (Nopalea or Opuntia) on the metabolic profile of lambs. Thirty-six uncastrated male Santa Inês lambs (22.0±2.9kg initial body weight) were distributed in a completely randomized design, with three treatments and 12 repetitions. The animals were fed a control diet (Tifton hay as exclusive roughage), Miúda cactus cladodes-based diet or Orelha de Elefante Mexicana (O.E.M.) cactus cladodes-based diet. Blood samples were collected one day before (baseline) and 45 days after the introduction of the tested diets. The Miúda cactus cladodes caused an increase (P=0.055) in the serum activity of the gamma-glutamyl transferase enzyme (53.66U/L) and in the blood content of glucose and fructosamine. The O.E.M. cactus cladodes caused lower (P=0.038) serum cholesterol content (41.33mg/dL). Regardless of the variety, there was a decrease (P=0.001) in the serum content of indirect bilirubin, urea, and sodium, and increase in the serum magnesium concentration. The partial replacement of the Tifton hay by Miúda or O.E.M. cactus cladodes in lamb feeding increases the enzyme activity, indicating liver and/or kidney changes, but does not cause relevant damage to energy, protein, and mineral metabolism.


Este trabalho teve como objetivo avaliar os efeitos da substituição parcial (750g/kg) do feno de Tifton por duas variedades de palma forrageira (Nopalea ou Opuntia) no perfil metabólico de cordeiros. Trinta e seis cordeiros Santa Inês, machos, não castrados (22,0 ± 2,9kg de peso corporal inicial), foram distribuídos em delineamento inteiramente ao acaso, com três tratamentos e 12 repetições. Os animais foram alimentados com dieta controle (feno de Tifton como volumoso exclusivo), dieta à base de palma forrageira Miúda ou dieta à base de palma forrageira Orelha de Elefante Mexicana (OEM). Amostras de sangue foram coletadas um dia antes (baseline) e 45 dias após a introdução das dietas testadas. A palma Miúda causou aumento (P=0,055) na atividade sérica da enzima gamaglutamiltransferase (53,66U/L) e no teor sanguíneo de glicose e frutosamina. A palma OEM causou menor teor (P=0,038) de colesterol sérico (41,33mg/dL). Independentemente da variedade, houve diminuição (P=0,001) do teor sérico de bilirrubina indireta, ureia e sódio, e aumento na concentração sérica de magnésio. A substituição parcial do feno de Tifton por palma Miúda ou por OEM na alimentação de cordeiros aumenta a atividade enzimática, o que indica alterações hepáticas e/ou renais, mas não causa danos relevantes nos metabolismos energético, proteico e mineral.

We analyzed the influence of the season, the environment, and the sex, as well as the relation of body mass (BM) in the serum albumin (ALB), aspartate aminotransferase (AST), creatinine (C), creatine kinase (CK), phosphorus (P), total calcium (tCa), total protein (TP), urea (U), uric acid (UA), calcium:phosphorus ratio (Ca:P), and the globulin value (GV) of thirty individuals of Phrynops geoffroanus of the urban area of Cuiabá, Mato Grosso, Brazil. The modeling of biochemical parameters was performed using the Generalized Additive Models for Location, Scale and Shape (GAMLSS) to verify the influence of variables considered in this study on each of the biochemical parameters analyzed. The season influenced AST, CK, C, tCa, Ca:P and UA. The environment influenced tCa, Ca:P, U and UA. On the other hand, CK, tCa, P, Ca:P and U differed significantly between males and females. Regarding the BM, a relationship of this variable was observed with CK, C, tCa, P, U, UA and Ca:P. We concluded that the season, environment, sex, and body mass can influence the biochemical parameters of P. geoffroanus, and these factors should be routinely considered in the interpretation of laboratory results.


Analisou-se a influência da estação, do ambiente e do sexo, assim como a relação da massa corporal (BM) nos níveis sorológicos de albumina (ALB), aspartato aminotransferase (AST), creatinina (C), creatina quinase (CK), fósforo (P), cálcio total (tCa), sólidos totais (TS), ureia (U), ácido úrico (UA), relação cálcio:fósforo (Ca:P), e do valor da globulina (GV) de Phrynops geoffroanus da área urbana de Cuiabá, Mato Grosso, Brasil. A modelagem dos parâmetros bioquímicos foi realizada utilizando-se os modelos aditivos generalizados para locação, escala e forma (GAMLSS) para verificar a influência das variáveis consideradas neste estudo em cada um dos parâmetros bioquímicos analisados. A estação sazonal influenciou os níveis de AST, CK, C, tCa, Ca:P e UA. O ambiente foi capaz de influenciar tCa, Ca:P, U e UA. Por outro lado, CK, tCa, P, Ca:P e U diferiram significativamente entre machos e fêmeas. Em relação à BM, observou-se relação dessa variável com CK, C, tCa, P, U, UA e Ca:P. Concluiu-se que a estação sazonal, o ambiente, o sexo e a massa corporal são capazes de influenciar os parâmetros bioquímicos de P. geoffroanus e que esses fatores devem ser rotineiramente considerados na interpretação dos resultados laboratoriais.

Identifying the metabolic profile of farm animals constitutes an important tool for the clinical diagnosis of metabolic diseases that can affect animal production performance. This study investigates the prepartum metabolic profile of ewes reared in two feeding systems native pasture and cultivated pasture of black oat and ryegrass and their effects on lamb development. Forty adult Texel ewes and their lambs were used and evaluated for metabolites representative of energy, protein, and mineral metabolism in the prepartum period. Ewes kept on cultivated pasture exhibited higher glucose levels than those kept on native pasture (59.67 vs. 31.98 mg/dL, respectively), whereas those kept on native pasture had higher serum Ca and P levels (7.62 and 4.58 mg/dL, respectively) than the ewes on cultivated pasture (6.21 and 3.73 mg/dL, respectively). Albumin was higher in the ewes with single pregnancy (2.92 vs. 2.76 g/dL), while urea levels were affected by the interaction between feeding system and type of pregnancy. Prepartum levels of glucose, phosphorus, and calcium in the blood of the ewes were correlated with lamb weight at 30 days. The feeding system influenced the metabolic profile of the ewes in the prepartum period, with lamb weight at 30 days of age being mainly associated with the mother's glucose level at the end of the gestation period.(AU)


A identificação do perfil metabólico em animais de produção apresenta-se como uma importante ferramenta no diagnóstico clínico de doenças do metabolismo que podem afetar no desempenho produtivo dos animais. Avaliou-se o perfil metabólico no pré-parto de ovelhas criadas em dois sistemas alimentares, a pasto (pastagem nativa) e pastagem cultivada de aveia preta e azevém e seus efeitos sobre o desenvolvimento dos cordeiros. Foram utilizadas 40 ovelhas adultas, da raça Texel, e seus respectivos cordeiros, nas quais foram mensurados no pré-parto os metabólitos representativos do metabolismo energético, proteico e mineral. Animais mantidos em pastagem cultivada apresentaram maiores valores de glicose em relação aos mantidos em pastagem nativa (59,67 vs 31,98 mg/dL, respectivamente), e os animais mantidos em pastagem nativa apresentaram maiores níveis séricos de Ca e P (7,62 e 4,58 mg/dL, respectivamente) do que os em pastagem cultivada (6,21e 3,73 mg/dL, respectivamente). A albumina foi maior nas ovelhas com gestação simples (2,92 vs 2,76 g/dL) enquanto os níveis de ureia sofreram efeito da interação sistema alimentar vs tipo de gestação. Os teores de glicose, fósforo e cálcio no sangue das ovelhas no pré-parto se mostraram correlacionados com o peso dos cordeiros aos 30 dias. O sistema alimentar influenciou o perfil metabólico das ovelhas no pré-parto sendo o peso dos cordeiros aos 30 dias de idade principalmente associado com o nível de glicose da mãe no final do período gestacional.(AU)

Background: Ketosis is the most important metabolic disease with prevalence from 15 to 45%. Ketosis is diagnosed using a metabolic profile. Due to the high prevalence, it is necessary to determine a large number of metabolic profiles within farm, which represents an additional cost, so the implementation of pooled serum in assessing the metabolic status of cows was examined. The aim of this study was to validate and evaluate the influence of the relative position (Z-score) of the value of pooled sample metabolic parameters within the known reference value of healthy cows in the detection of ketosis in herd during early lactation. Materials, Methods & Results: The experiment has been carried out using blood samples collected by puncture of coccygeal vein from 50 ketotic and 50 healthy cows. Laboratory analysis includes determination of beta-hydroxybutyrate-BHB, non-esterified fatty acids-NEFA, cholesterol-CHOL, triglycerides-TGC, glucose-GLU, albumin-ALB, total protein-TPROT, UREA, Ca, P, total bilirubin-TBIL and aspartat aminotransferase-AST. The pooled serum was made from 10 individual samples originating from 10 different cows. A serum aliquot of 0.1 mL was taken from each sample, and a 1 mL volume of pooled serum was finally formed. Three types of serum pools were made: 1) 30 pooled sample were from ketosis; 2) 30 pooled sample were from healthy cows and 3) 60 pooled samples containing mixed sera of healthy cows and cows with ketosis were made as follows: 10 pools contain 10% to 60% of ketotic cows (1/10 to 6/10 samples). Statistical analysis includes: a) difference in metabolite concentration and Z-score in pooled sample and arithmetic mean individual sample in healthy and ketotic cow, b) correlation between Z-score of pooled sample and arithmetic mean of individual sample, c) ability of Z-score of metabolite to divide ketotic from healthy cow, d) correlation between Z-score and % of ketotic cow in pooled sample; and e) calculation of 95%CI of pooled sample Z-scores for each % of ketotic cow in pools. Z-score and all analysis were calculated for each metabolic parameter. The results of the study show that the mean values and Zscores of the pool and the calculated average value of the individual samples participating in that pool differ significantly in healthy cows and cows in ketosis, except for TPROT and Ca. A higher value and a higher Z-score were found for BHB, NEFA, UREA, TBIL and AST, and a lower value and a lower Z-score for TGC, CHOL, GLU, ALB and P in ketotic cows compared to healthy cows. The value of the Z-score of the pooled sample and the calculated mean values of individual samples participating in the pool are highly correlated with each other (coefficient of determination over 99%). Z-score of metabolites in the pooled sample can be used to distinguish healthy from ketotic cows (ROC AUC= 0.711 to 0.989), except for TPROT and Ca. The Z-score value of the pooled sample shows a linear correlation with the percentage of ketotic cows in the pool and the reference ranges of Z-scores change significantly as a function of the percentage of ketosis cows. Discussion: Modern research on the metabolic profile in cows requires obtaining a large amount of information from as few samples as possible. The advantages of using the Z-score are reflected in the following: this score does not depend on the absolute value of the metabolite, but on the position within the known population reference value, Z-score of sample and the arithmetic mean of individual samples included in the same pool are almost identical, the Z-score of these 2 groups of results is ideally correlated, the Z-score significantly correlates with the % of ketosis samples in the pooled sample. The use of pooled sample Z-score can be a useful in a herd level assessment of metabolic status and detection of ketosis as most important metabolic disease in dairy cows.

Crocodylus moreletii and Crocodylus acutus are two endangered crocodile species endemic to Mexico. In this descriptive cross-sectional study, a total of 58 crocodiles (30 females and 28 males) were examined to determine and evaluate 24 blood biochemical indicators associated with energy, lipid, protein, mineral and enzymatic metabolic processes. Most of the serum biochemistry parameters were similar among sexes by species. However, male crocodiles showed higher triacylglycerol concentration and alkaline phosphatase activity, and lower globulin concentration than females. There were some significant differences between species. Total bilirubin, triacylglycerol, carbon dioxide, and hydrogen carbonate were higher in Crocodylus moreletii, and urea nitrogen, creatinine, alanine aminotransferase, and aspartate aminotransferase were higher in Crocodylus acutus. These reference values are very important for the protection of crocodiles. The calculated confidence intervals could be used to detect alert situations when at least 5% of the sampled crocodiles would fall outside of the calculated reference interval for a given parameter.


Crocodylus moreletii e Crocodylus acutus são duas espécies de crocodilos ameaçadas de extinção endêmicas do México. Neste estudo transversal descritivo, um total de 58 crocodilos (30 fêmeas e 28 machos) foi examinado para determinar e avaliar 24 indicadores da bioquímica sanguínea associados a processos metabólicos energéticos, lipídicos, proteicos, minerais e enzimáticos. A maioria dos parâmetros bioquímicos séricos foram semelhantes entre os sexos da espécie. No entanto, os crocodilos machos apresentaram maior concentração de triacilgliceróis e atividade de fosfatase alcalina do que as fêmeas. Com uma concentração de globulina mais baixa do que as fêmeas, houve algumas diferenças significativas entre as espécies. A bilirrubina total, os triacilgliceróis, o dióxido de carbono e o bicarbonato de sódio foram maiores em Crocodylus moreletii, e o nitrogênio ureico, a creatinina, a alanina aminotransferase e o aspartato aminotransferase foram maiores em Crocodylus acutus. Esses valores de referência são muito importantes para a proteção dos crocodilos. Os intervalos de confiança calculados podem ser usados para detectar situações de alerta quando pelo menos 5% dos crocodilos amostrados estiverem fora do intervalo de referência calculado para um determinado parâmetro.

The aim of this study was to evaluate the performance of young bulls from three genetic groups, ½ Brangus x ½ Nellore (BRN), Nellore (NEL) and ½ Canchim x ½ Nellore (CAN), reared on pasture and supplemented with mineral (MS) or energy-mineral (MES) supplement. Eighty-one bulls, with a mean age of 12 months and mean body weight of 252 ± 33 kg were used. The experiment was conducted in a 3x2 factorial completely randomized design. Each genetic group was subdivided into six experimental plots, three received MS and three received MES. Animals were managed in a rotational stocking system in a Tifton 85 grass pasture. The consumption of MS was similar between the genetic groups with an average of 0.073 kg animal-1 day-1, whereas the consumption of MES was higher for BRN, 2.10 kg animal-1 day-1, followed by CAN, with 1.57 kg animal-1 day-1, and lower for NEL, with 1.28 kg animal-1 day-1. The average daily weight gain (ADG) was greater for animals that received MES compared to those that were given MS. For animals that received MS, the BRN group had ADG of 0.64 kg animal-1, while the NEL and CAN groups were similar with a mean of 0.46 kg animal-1. For animals that received MES, the CAN group had higher ADG, 0.97 kg animal-1, while the NEL and BRN groups were similar, with an average of 0.86 kg animal-1. Blood levels of total protein, albumin, creatinine, glucose and cholesterol did not change depending on the types of supplements used or between genetic groups. Higher serum urea levels were observed in NEL and CAN animals that received MS. Serum aspartate aminotransferase levels were higher in BRN and CAN animals that received MES. Gains in rump height, height at the withers, body length, rump width and chest perimeter were greater in animals that received MES. Mostly, the gains in morphometric measurements were greater for crossbred animals than for the NEL group. The supply of mineral-energy supplement in Tifton 85 grass pasture during the rainy season is recommended only for Nellore and ½ Canchim x ½ Nellore young bulls. Crossbred young bulls show greater gains in morphometric measurements than Nellore young bulls during rearing.(AU)


Objetivou-se com este estudo avaliar o desempenho de tourinhos de três grupos genéticos, ½ Brangus x ½ Nelore (BRN), Nelore (NEL) e ½ Canchim x ½ Nelore (CAN), recriados à pasto e suplementados com mineral (SM) ou energético-mineral (SEM). Foram utilizados 81 tourinhos, com idade média de 12 meses e peso corporal médio de 252 ± 33 kg. O experimento foi conduzido em delineamento inteiramente ao acaso em esquema fatorial 3x2. Cada grupo genético foi subdividido em seis parcelas experimentais, em que três delas receberam SM e três receberam SEM. Os animais foram manejados em sistema de lotação rotacionada em pasto de capim Tifton 85. O consumo de SM foi semelhante entre os grupos genéticos com média de 0,073 kg animal-1 dia-1, já o consumo de SEM foi maior para o BRN com 2,10 kg animal-1 dia-1, seguido pelo CAN com 1,57 kg animal-1 dia-1 e menor para o NEL com 1,28 kg animal-1 dia-1. O ganho de peso médio diário (GMD) foi maior para os animais que receberam SEM em relação aos que receberam SM. Para os animais que receberam SM, o grupo BRN apresentou GMD de 0,64 kg animal-1, enquanto os grupos NEL e CAN foram semelhantes com média de 0,46 kg animal-1. Para os animais que receberam SEM o grupo CAN apresentou maior GMD com 0,97 kg animal-1, enquanto os grupos NEL e BRN foram semelhantes com média de 0,86 kg animal-1. Os níveis sanguíneos de proteína total, albumina, creatinina, glicose e colesterol não foram alterados em função dos tipos de suplementos utilizados ou entre os grupos genéticos. Maiores níveis séricos de ureia foram observados nos animais NEL e CAN que receberam SM. Os níveis séricos de aspartato aminotransferase foram maiores em animais BRN e CAN que receberam SEM. Os ganhos de altura de garupa, altura de cernelha, comprimento, largura de garupa, perímetro torácico e largura de peito foram maiores em animais que receberam SEM. Em sua maioria, os ganhos de medidas morfométricas foram maiores para os animais mestiços do que para o NEL. O fornecimento de suplemento energético-mineral em pasto de capim Tifton 85 durante o período das águas é recomendado apenas para tourinhos Nelore e ½ Canchim x ½ Nelore. Tourinhos mestiços apresentam maiores ganhos em medidas morfométricas que tourinhos Nelore durante a recria.(AU)

The study aimed to determine the biochemical reference ranges for lambs from birth to 1 year of age in the tropics. Data were obtained from experiments performed in several institutions and commercial farms using Santa Inês, Dorper, Lacaune, Morada Nova, Bergamacia and Suffolk lambs reared under different conditions (grazing, feedlot, semi-feedlot) from 2006 to 2017. Serum energy-, protein-, mineral-, and enzyme-related metabolites were evaluated. The metabolic energy profile included data from glucose, cholesterol, triglycerides, fructosamine, high-density lipoprotein cholesterol, low-density lipoprotein cholesterol, and very-low-density lipoprotein cholesterol; the metabolic protein profile included total protein, uric acid, urea, albumin, and creatinine; the metabolic mineral profile included data on calcium, phosphorus, and magnesium; and the metabolic enzymatic profile included the enzymes aspartate aminotransferase, gamma glutamyl transferase, alkaline phosphatase, and creatine kinase. The reference ranges were estimated using confidence intervals with a 95% confidence level, and percentiles were estimated or bootstrapped nonparametrically when the data were not normally distributed. The serum biochemical reference ranges determined for lambs are strongly divergent from those established by one of the most cited books on the topic, especially considering the high serum urea and cholesterol concentrations and low levels of blood glucose observed. Therefore, the serum biochemical reference ranges for lambs from birth to 1 year of age in the tropics differ from international data, which consider adult sheep in temperate climate zones.


Objetivou-se determinar as faixas de referência dos parâmetros bioquímicos para cordeiros desde o nascimento até a idade de um ano nos trópicos. Os dados foram obtidos a partir de experimentos realizados em diversas instituições e fazendas comerciais, utilizando cordeiros Santa Inês, Dorper, Lacaune, Morada Nova, Bergamacia e Suffolk criados em diferentes condições (pastejo, confinamento, semi-confinamento) de 2006 a 2017. Foram avaliados os metabólitos séricos relacionados à energia, proteína, minerais e vitaminas. O perfil metabólico incluiu dados de glicose, colesterol, triglicerídeos, frutosamina, lipoproteína de alta densidade, lipoproteína de baixa densidade e lipoproteína de densidade muito baixa; o perfil metabólico proteico incluiu proteína total, ácido úrico, ureia, albumina e creatinina; o perfil mineral os dados sobre cálcio, fósforo e magnésio; e o perfil enzimático as enzimas aspartato aminotransferase, gama glutamil transferase, fosfatase alcalina e creatina quinase. As faixas de referência foram estimadas usando intervalos de confiança com nível de confiança de 95%, e os percentis foram estimados por métodos não paramétricos de reamostragem quando não apresentavam distribuição normal. Os intervalos bioquímicos séricos de referência para cordeiros determinados são fortemente divergentes dos estabelecidos por um dos livros mais citados sobre o tema, principalmente considerando as altas concentrações séricas de ureia e colesterol e baixos níveis de glicose observados no sangue. The study aimed to determine the biochemical reference ranges for lambs from birth to 1 year of age in the tropics. Data were obtained from experiments performed in several institutions and commercial farms using Santa Inês, Dorper, Lacaune, Morada Nova, Bergamacia and Suffolk lambs reared under different conditions (grazing, feedlot, semi-feedlot) from 2006 to 2017. Serum energy-, protein-, mineral-, and enzyme-related metabolites were evaluated. The metabolic energy profile included data from glucose, cholesterol, triglycerides, fructosamine, high-density lipoprotein cholesterol, low-density lipoprotein cholesterol, and very-low-density lipoprotein cholesterol; the metabolic protein profile included total protein, uric acid, urea, albumin, and creatinine; the metabolic mineral profile included data on calcium, phosphorus, and magnesium; and the metabolic enzymatic profile included the enzymes aspartate aminotransferase, gamma glutamyl transferase, alkaline phosphatase, and creatine kinase. The reference ranges were estimated using confidence intervals with a 95% confidence level, and percentiles were estimated or bootstrapped nonparametrically when the data were not normally distributed. The serum biochemical reference ranges determined for lambs are strongly divergent from those established by one of the most cited books on the topic, especially considering the high serum urea and cholesterol concentrations and low levels of blood glucose observed. Therefore, the serum biochemical reference ranges for lambs from birth to 1 year of age in the tropics differ from international data, which consider adult sheep in temperate climate zones.(AU)


Objetivou-se determinar as faixas de referência dos parâmetros bioquímicos para cordeiros desde o nascimento até a idade de um ano nos trópicos. Os dados foram obtidos a partir de experimentos realizados em diversas instituições e fazendas comerciais, utilizando cordeiros Santa Inês, Dorper, Lacaune, Morada Nova, Bergamacia e Suffolk criados em diferentes condições (pastejo, confinamento, semi-confinamento) de 2006 a 2017. Foram avaliados os metabólitos séricos relacionados à energia, proteína, minerais e vitaminas. O perfil metabólico incluiu dados de glicose, colesterol, triglicerídeos, frutosamina, lipoproteína de alta densidade, lipoproteína de baixa densidade e lipoproteína de densidade muito baixa; o perfil metabólico proteico incluiu proteína total, ácido úrico, ureia, albumina e creatinina; o perfil mineral os dados sobre cálcio, fósforo e magnésio; e o perfil enzimático as enzimas aspartato aminotransferase, gama glutamil transferase, fosfatase alcalina e creatina quinase. As faixas de referência foram estimadas usando intervalos de confiança com nível de confiança de 95%, e os percentis foram estimados por métodos não paramétricos de reamostragem quando não apresentavam distribuição normal. Os intervalos bioquímicos séricos de referência para cordeiros determinados são fortemente divergentes dos estabelecidos por um dos livros mais citados sobre o tema, principalmente considerando as altas concentrações séricas de ureia e colesterol e baixos níveis de glicose observados no sangue. Portanto, as faixas bioquímicas séricas de referência para cordeiros do nascimento a um ano nos trópicos diferem dos dados internacionais, que contemplam ovelhas adultas em países temperados.(AU)

In the search for an early biomarker of renal injury, this study aimed to determine the urinary protein profile of dogs with leishmaniasis without treatment and treated as determined by Brazilian legislation. The identification of proteinuria, its classification and the circumstances in which it takes place instigated this study. For this, 30 dogs from an outpatient clinic at a Veterinary Hospital in Belo Horizonte were evaluated. All animals underwent clinical and laboratory tests, which included renal biomarkers. The proteins were characterized using the SDS-page electrophoresis technique, and thus, a urinary protein profile was developed comparing patients considered clinically healthy with dogs infected with leishmaniasis that were under treatment and with untreated infected dogs. The results showed that the hematological and biochemical parameters showed similar behavior between the groups of healthy dogs and dogs with leishmaniasis treated, however a very heterogeneous pattern of urinary proteins can be observed and differed between healthy animals and animals with leishmaniasis, as well as between treated and untreated animals. The results suggest that the classification of proteinuria can be a tool that helps in the staging of animals infected with L. infantum and can differentiate them as to the severity of existing kidney injuries.(AU)


Na busca por um biomarcador precoce de injúria renal, este trabalho teve como objetivo determinar o perfil proteico urinário de cães infectados com leishmaniose sem tratamento e tratados conforme determina a legislação brasileira. A identificação da proteinúria, sua classificação e as circunstâncias em que ocorrem instigaram este estudo. Para tanto, foram avaliados 30 cães oriundos do atendimento clínico ambulatorial de um Hospital Veterinário em Belo Horizonte. Todos os animais passaram por exame clínico e laboratorial, que incluíram biomarcadores renais. As proteínas foram caracterizadas através da técnica de eletroforese por SDS-PAGE, e assim, foi elaborado um perfil proteico urinário comparando pacientes considerados clinicamente hígidos, com cães infectados por Leishmania (L.) infantum e que estavam sob tratamento e cães infectados não tratados. Os resultados demonstraram que os parâmetros hematológicos e bioquímicos apresentaram comportamento semelhante entre os grupos de cães hígidos e de cães infectados com L. infantum tratados, entretanto um padrão muito heterogêneo de proteínas urinárias pode ser observado e diferiu entre animais hígidos e animais com leishmaniose, assim como entre os animais tratados e não tratados. Os resultados sugerem que a classificação da proteinúria pode ser uma ferramenta que auxilia no estadiamento de animais infectados por L. infantum podendo diferenciá-los quanto à gravidade de lesões renais existentes.(AU)

Diagnosing Canine Visceral Leishmaniosis is challenging for veterinarians given that its hematological and biochemical abnormalities greatly resemble those of other illnesses of dogs, such as canine ehrlichiosis, which is caused by Ehrlichia canis and is highly prevalent with high pathogenicity. The aim of this study was to determine occurrences of serological positivity for the antigens of Ehrlichia canis and Leishmania infantum and its relationship to globulin concentrations in samples from dogs. Out of 93 samples tested, 12.9% were negative for the antigens of both L. infantum and E. canis; 33.3% were seropositive for both antigens. Discordant results were found from 18.3% that were positive only for L. infantum, while 35.5% were positive only for E. canis. Hyperglobulinemia was observed in 88.2% and the statistical analysis showed that there was a significant relationship between the high levels of globulins and seropositivity for the antigen of E. canis. However, the relationship between positivity for the antigen of L. infantum and hyperglobulinemia, showed that there was no statistically significant relationship between the two laboratory findings. From these results, it can be concluded that concomitant infections occur frequently and that hyperglobulinemia is more closely related to ehrlichiosis than to leishmaniosis, in dogs.


O diagnóstico da leishmaniose canina é um desafio ao Médico Veterinário visto que anormalidades hematológicas e bioquímicas muito se assemelham à outras enfermidades dos cães, como a erliquiose canina, causada pela Ehrlichia canis, muito prevalente e de alta patogenicidade. O objetivo deste estudo foi determinar a positividade sorológica frente aos antígenos de Ehrlichia canis e Leishmania infantum e a sua relação com as concentrações de globulinas em amostras de cães. Das 93 amostras testadas, 12,9% foram negativas frente aos antígenos de E. canis e L. infantum e 33% foram soropositivas. Resultados discordantes foram encontradas em 18,3% positivas apenas para L. infantum, enquanto que 35,5% reagiram apenas frente a E. canis. Hiperglobulinemia foi observada em 88,2% das amostras e a análise estatística mostrou haver relação significante entre a soropositividade frente aos antígenos de E. canis e a hiperglobulinemia, e em contrapartida, a relação entre esse aumento de globulinas e a positividade para L. infantum foi insignificante. Pelos resultados apresentados, pode-se concluir que as infecções concomitantes são frequentes e que a hiperglobulinemia está mais intimamente relacionada a erliquiose quando comparada à leishmaniose em cães.

Measuring metabolic parameters in the blood has been an indispensable tool for assessing the productive and health status of dairy cows for more than 100 years. The values of laboratory parameters depend on various preanalytical, analytical and postanalytical factors. The most important preanalytical factors are sample transport time and temperature, hemolysis, anticoagulant type, and sample volume. Preanalytical factors can lead to reduced stability of the analyte in the sample, which changes their concentration. Loss of stability changes the time of storage and manipulation of the sample, which determines the criteria for its acceptance or rejection. The two stability indicators are stability limit and maximum permissible instability. A stability limit (SL) is defined as the period of time in which a property variation does not exceed a maximum permissible instability (MPI). The aim of this study was to determine the SL and MPI for each analyte in the blood serum of cows and to determine whether SL differs in the function of the presence of preanalytical errors in the blood sample. Three hundred samples of dairy cow origin in different periods of lactation participated in this research. They were classified into 6 groups of 50 samples: according to the time from sampling to processing in the laboratory (0-4 h, 4-8 h and over 8 h; all transported on dry ice, protected from environmental factors, without preanalytical errors) and according to the presence of preanalytical errors (group with hemolysis, a group transported at ambient temperature and a group with a small sample volume). Each sample was aliquoted in two portions. One portion was left at +4°C and tested once a day for 6 days of sample storage, and the second portion, placed at -20°C, was tested once a month for 6 months. The MPI had a value ranging from 1.51 to 8.4. Metabolic profile analytes with lower MPI values (1.51-3.22) were albumin (ALB), total protein (TPROT), UREA, glucose (GLU), calcium (Ca), and phosphorus (P). Higher MPI values (5.1-8.4) were found for nonesterified fatty acids (NEFA), beta-hydroxybutirate (BHB), cholesterol (CHOL), triglycerides (TGC), total bilirubin (TBIL) and aspartat aminotransferase (AST). For most parameters, we can conclude that their PD% changed faster in storage conditions at +4°C compared to the regime of -20°C. The largest number of biochemical analytes in bovine blood serum shows preserved stability in the first 6 days at +4°C or 6 months at -20°C if transported to the laboratory within 8 h after sampling in ideal conditions and without the action of preanalytical errors. Prolonged transport under ideal conditions or the existence of preanalytical errors such as transport at room temperature, hemolysis or small sample volume shorten the stability of the ALB, NEFA, GLU, UREA and P. Concentration of all analytes decreases during the stability test except for UREA, NEFA, BHB and for CHOL and TGC in some groups. Variations in parameters such as BHB, NEFA, TBIL, AST, and Ca have shown potential clinical significance. At storage conditions at +4°C, clinically significant variations at at least one measurement point were found for AST (7.5% of samples), BHB (6.1% of samples), NEFA (9.9% of samples) and for TBIL (in 7% of samples). This study can help define acceptable delay times and storage conditions for bovine blood samples, which is of great importance because in working with farm animals it is often not possible to take samples in a short time and deliver them to the laboratory, and samples are often burdened with certain preanalytical errors with limited possibilities of re-sampling.(AU)

Malnutrition occurs when the supply of available nutrients is less than organic demand. It is an underdiagnosed problem in veterinary medicine and can result in several negative metabolic consequences, with greater morbidity and mortality. Currently, the classification of nutritional status (NS) is performed subjectively in veterinary medicine, so studies and discoveries about laboratory markers (objectives) of malnutrition are desirable. This study evaluated the correlations between several laboratory variables (practical and lowcost measurements) and the nutritional status (NS) of 246 dogs from a veterinary school hospital in southern Brazil. In this way, the laboratory profile of malnutrition in this population is evident. NS was classified by body condition score (BCS) and muscle mass score (MMS). A patient was considered to be malnourished if the BCS was less than 3 (values from 1 to 9). The laboratory variables analyzed were hemogram, cholesterol, total protein (TP), albumin, C-reactive protein (CRP), CRP/albumin ratio, total iron-binding capacity (TIBC), transferrin (TF), transferrin saturation index (TSI), and transthyretin. The means and standard deviations of the variables, correlation analyses, and comparative analyses (Kruskal-Wallis in α = 5%) were calculated. This research aimed to identify objective and practical malnutrition markers that help in the elaboration of a protocol for nutritional evaluation in diseased dogs. Low values of TF, TIBC, TP, hemogram, and TSI elevation were indicative of malnutrition. Additionally, cholesterol and albuminemia are not good markers of malnutrition in dogs with systemic diseases. For the graduation of the inflammatory state (important to differentiate inflammatory hypoproteinemia from protein malnutrition), it is more reliable to measure the CRP/albumin ratio. Canine transthyretin cannot be measured with reagents designed for humans, such as those used in this study. It was possible to conclude that laboratory indicators of malnutrition in sick dogs include low values of TF, TIBC, and TP, anemia, and elevations of TSI. The joint changes in these markers gradually reinforced the diagnosis.(AU)


A desnutrição ocorre quando a oferta de nutrientes disponíveis é inferior à demanda orgânica. É um problema subdiagnosticado em medicina veterinária e pode resultar em diversas consequências metabólicas negativas, com maior morbidade e mortalidade. Atualmente, a classificação do estado nutricional (EN) é feita subjetivamente na medicina veterinária, de maneira que estudos e descobertas sobre marcadores laboratoriais objetivos de desnutrição são desejáveis. O propósito da pesquisa foi encontrar marcadores de desnutrição objetivos e práticos que ajudem na elaboração de protocolos de avaliação nutricional em cães doentes. Este estudo avaliou as correlações entre diversas variáveis laboratoriais (de mensurações práticas e de baixo custo) com o estado nutricional (EN) de 246 cães de um hospital veterinário escola do sul do Brasil, evidenciando o perfil laboratorial da desnutrição desta população. O EN foi classificado por meio do escore de condição corporal (ECC) e escore de massa muscular (EMM). Foram considerados desnutridos os animais com ECC ≤ 3 (Laflamme, 1997) ou EMM ≤ 2 (World Small Animal Veterinary Association [WSAVA], 2013). As variáveis laboratoriais analisadas foram: hemograma, colesterol, proteína total (PT), albumina, proteína C reativa (PCR), relação PCR/albumina, capacidade total de ligação com ferro (CTLF), transferrina (TF), índice de saturação da transferrina (IST) e transtirretina. Foram calculadas as médias e desvios-padrões das variáveis, análises de correlação e análises comparativas (Kruskal-Wallis em α = 5%). Foram indicativos de desnutrição: baixos valores de TF, CTLF e PT, anemia e elevações da IST. Ademais, o colesterol e a albumina não são bons marcadores de desnutrição em cães com doenças sistêmicas. Para a graduação do estado inflamatório (importante para diferenciar hipoproteinemia inflamatória da desnutrição proteica), é mais confiável a mensuração da relação PCR/albumina. A transtirretina canina não pode ser mensurada com reagentes elaborados para humanos, como os utilizados nesta pesquisa. Foi possível concluir que são indicadores laboratoriais da desnutrição em cães doentes: baixos valores de TF, CTLF e PT, anemia e elevações da IST. As alterações conjuntas desses marcadores reforçam gradativamente o diagnóstico.(AU)

This study aimed to evaluate the hematologic response and the serum and peritoneal fluid (PF) proteinogram of cattle affected by digestive diseases. Twenty-seven animals were distributed in two groups: GI (intestinal diseases) and GII (traumatic reticuloperitonitis, TRP). The animals were previously submitted to a physical exam. Subsequently, blood samples were collected to perform the complete blood count, determine the plasma protein and fibrinogen, and obtain the serum for proteinogram in polyacrylamide gel (SDS-PAGE). Simultaneously, PF was collected to perform physical and chemical evaluation and the electrophoretic profile (SDS-PAGE). ANOVA at the 5% probability level was used to compare the groups. The animals showed signs of apathy, dehydration, and gastrointestinal hypomotility in both groups. However, GI animals showed more significant clinical changes. The blood count of both groups (P > 0.05) showed leukocytosis due to neutrophilia and a regenerative left shift with hyperfibrinogenemia. The proteinogram of both body fluids allowed the identification of proteins albumin (ALB), transferrin (TRF), ceruloplasmin, haptoglobin, α1-acid glycoprotein (α1-AGP), MW 23000 Da, α1-antitrypsin, IgA, and IgG. The [PT] PF/[PT] blood serum ratio of each of the identified proteins increased, showing statistical differences between groups (P < 0.05) regarding PT, ALB, TRF, α1-AGP, and IgG values, with GI animals showing the highest ratio. Intestinal diseases and TRP triggered a systemic and local response characterized by clinical, hematological, and serum and PF proteinogram alterations. The proteins α1-GPA, haptoglobin, and TRF measured in PF were good inflammation biomarkers and useful as an auxiliary tool for the diagnosis and prognosis of digestive diseases in cattle.(AU)


O objetivo deste estudo foi avaliar a resposta hematológica e o proteinograma sérico e do líquido peritoneal (LP) de bovinos acometidos com doenças digestivas. Foram avaliados 27 bovinos distribuídos em dois grupos, GI (enfermidades intestinais) e GII (reticuloperitonite traumática-RPT). Os animais foram previamente submetidos ao exame físico. Posteriormente foram colhidas amostras de sangue para realização do hemograma, determinação plasmática da proteína e do fibrinogênio e obtenção do soro para realização do proteinograma em gel de poliacrilamida (SDS-PAGE). Simultaneamente foi colhido o LP para avaliação física e química, assim como a realização do perfil eletroforético (SDS-PAGE). Empregou-se a análise de variância ao nível de 5% de probabilidade visando à comparação entre os grupos. Em ambos os grupos os animais demonstraram sinais de apatia, desidratação e hipomotilidade gastrointestinal, no entanto, os animais do GI apresentaram alterações clínicas mais expressivas. No hemograma observou-se em ambos os grupos (P > 0,05) leucocitose por neutrofilia e desvio à esquerda regenerativo com hiperfibrinogenia. O proteinograma de ambos os fluidos corpóreos permitiu a identificação das proteínas albumina (ALB), transferrina (TRF), ceruloplasmina, haptoglobina, α-1 glicoproteína ácida (α1-GPA), PM 23.000 Da, α-1 anti-tripsina, IgA e IgG. Os valores da relação [PT] LP / [PT] soro sanguíneo de cada uma das proteínas identificadas demonstrou elevação dos mesmos, bem como diferença estatística entre grupos (P < 0,05) nos valores da PT, ALB, TRF, α1-GPA e IgG, nos quais a relação foi mais elevada nos animais do GI. As enfermidades intestinais e a RPT desencadearam resposta sistêmica e local caracterizada pelas alterações clínicas, hematológicas, e do proteinograma sérico e do LP. A α1-GPA, a haptoglobina e a TRF, mensuradas no LP se mostraram bons biomarcadores de inflamação, sendo úteis como recurso auxiliar de diagnóstico e prognóstico das doenças digestivas dos bovinos.(AU)

Canine Leishmaniasis (CanL) is a multisystemic and chronic inflammatory disease characterized by nonspecific clinical manifestations. In CanL, inflammatory cells and chemical mediators released in response to the parasite play a role in disease development and progression. Alterations on hematological parameters have been documented in CanL. These changes can also be assessed in relation to systemic inflammation caused by this disease. The circulating leukocyte counting, such as neutrophils, as well as the albumin level, are considered direct indicators of an inflammatory host environment. Several studies point to the use of biomarkers on the assistance in diagnosis and prognosis of several canine pathologies. The present study investigated the Neutrophils to Lymphocyte Ratio (NLR), Albumin to Globulin Ratio (AGR), and Neutrophils to Albumin Ratio (NAR) on systemic inflammatory response induced by Canine Leishmaniasis (CanL). For this purpose, adult dogs with confirmed diagnosis to CanL were divided into symptomatic (SD, n = 33) and asymptomatic (AD, n = 20) dogs for L. infantum and control dogs (CD, n = 20). Routine hematological and biochemical parameters were determined in blood samples using a veterinary automatic hematology and biochemical analyzers. Asymptomatic dogs (AD) had a higher number of white blood cells and neutrophils (16.48 ± 4.93; 13.41 ± 3.60, respectively) in relation to symptomatic dogs (SD) (13.54 ± 5.13; 10.42 ± 3.69, respectively) (P = 0.015 and P < 0.0001, respectively). Neutrophils to Lymphocyte Ratio (NLR) was higher in dogs with leishmaniasis (9.45 ± 3.76) than in healthy dogs (3.39 ± 1.19) (P < 0.0001). Serum total proteins (STP) and globulins increased in CanL, while albumin and AGR decreased in CanL, when compared to CD and references values to canine species. Neutrophils to Albumin Ratio (NAR) was higher in AD and SD (5.02 ± 1.14; 4.79 ± 1.07, respectively) when compared to CD (2.36 ± 0.55) (P < 0.0001). As reported in scientific researches, dogs with Leishmaniasis present alterations in circulating cell counts. Based on these data, we decided to expand this information using the NLR as a parameter in an attempt to better clarify the changes in these cells in CanL. We observed that NLR was increased on CanL in relation to healthy dogs, which could be a consequence of relative neutrophilia rather than lymphopenia. Neutrophils to Lymphocyte Ratio (NLR) is a biomarker that conveys information about inflammatory conditions. An elevated NLR can reflect an upregulated innate immune response, since neutrophils are effector cells of innate immunity and are involved in several acute and chronic inflammatory processes. Albumin is an acute phase protein that is considered an immune-inflammatory biomarker, which can be found reduced systemically in progressive inflammatory response. Serum total proteins (STP) and globulins were increased in CanL. These data are already well documented in CanL, which serum globulins are mainly associated with the increase of acute phase proteins, cytokines, and increase of specific antibodies to Leishmania infantum. Our results showed neutrophilia with hypoalbuminemia in CanL. So, in an attempt to assess the relationship of these two available markers, we used NAR calculation in order to evaluate the changes induced by CanL. In this study NAR was higher in CanL when compared to control dogs. Thus, our data indicate that NLR and NAR could be used as biomarkers in veterinary medical clinics in order to assess inflammatory profile in CanL, mainly in asymptomatic dogs. These parameters obtained from routine blood tests might be useful as cost-effective, easily accessible and helpful markers in order to distinguish the inflammatory response intensity in CanL.(AU)

Resumo

Understanding the relation between the environmental stress factors and the hypothalamus-pituitary-thyroid (HPT) axis efficiency can reduce the susceptibility to thyroid diseases. In our study, thyroid dysfunction was induced in female rats by administration of 40 mg Na F/kg.bd.wt/day for a month. Co-administration of the water extract of Arca noae (300 mg/kg. bw) was tested as a treatment for Na F induced thyroid dysfunction. A group of rats injected Arca noae extract only (300 mg/kg.bd.wt) was performed to observe the impact of the extract on the (HPT) axis in addition to the normal control group. Results showed that there was a significant decrease in serum triglycerides, total protein and albumin levels in the fluoride supplemented group in addition to abnormal levels of TSH, (T4) and (T3) compared to the control group. In the treated group there was an improvement in the proteins level and lipid profile but pseudo-corrected serum (T4) and (T3) levels were observed in addition to a continuous increase in TSH level. Histological findings confirmed the harmful effect of fluoride on both the non treated and the treated groups. Consequently, fluoride supplementation must be considered as a harmful stress that may affect permanently the HPT axis.(AU)


Compreender a relação entre os fatores de estresse ambiental e o eixo hipotálamo-hipófise-tireoide (HPT) pode reduzir a suscetibilidade a doenças da tireoide. Em nosso estudo, a disfunção tireoidiana foi induzida em ratos fêmeas pela administração de 40 mg Na F/kg.bw/dia durante um mês. A administração concomitante do extrato aquoso de Arca noae (300 mg/kg.Pc) foi testada como tratamento para a disfunção tireoidiana induzida por Na F. Um grupo de ratos injetados apenas com extrato de Arca noae (300 mg/kg. Pc) foi pré-formado com o intuito de observar o impacto do extrato no eixo (HPT), além do grupo controle normal. Os resultados mostraram que houve uma diminuição significativa nos níveis séricos de triglicerídeos, proteína total e albumina no grupo suplementado com fluoreto, além de níveis anormais de TSH, (T4) e (T3) em comparação ao grupo controle. No grupo tratado, houve uma melhora no nível de proteínas e perfil lipídico. Os níveis séricos pseudocorrigidos (T4) e (T3) foram observados, além de um aumento contínuo no nível de TSH. Os achados histológicos confirmaram o efeito prejudicial do flúor nos grupos não tratado e tratado. Consequentemente, a suplementação de flúor é considerada um estresse prejudicial que pode afetar permanentemente o eixo HPT.(AU)

To investigate the effect of feeding heat-treated colostrum at different duration on the health and performance, 48 Holstein calves were enrolled randomly into four treatment groups before first feeding and consumed untreated colostrum (H0, n = 12), heat-treated colostrum at 60ºC for 30 min. (H30, n = 12), heat-treated colostrum at 60ºC for 60 min. (H60, n = 12) and heat-treated colostrum at 60ºC for 90 min. (H90, n = 12). Blood samples were collected for analyses of IgG and protein profile at 0, 6, and 24h of age. The colostrum sample from treated and untreated batches and feces sample from each calf also were taken. The results showed heat-treatment of colostrum at 60ºC for 60 (p = 0.03) and 90 min. (p = 0.01) reduced total bacterial count, while colostral IgG concentration maintained up to 60 min. Serum total protein (p = 0.02), IgG concentrations (p = 0.03), and apparent efficiency of IgG absorption (p = 0.02) were significantly greater at 6 and 24h in calves that were fed heat-treated colostrum (H90) compared to calves fed unheated colostrum (H0). General health status of calves that were received heat-treated colostrum was better and the prevalence of diarrhea-induced pathogens was lower than calves were fed unheated colostrum. In conclusion, the consumption of heat-treated colostrum had a positive effect on health, growth characteristics, and performance of calves during the suckling period.(AU)
