Allogeneic hematopoietic stem cell transplantation (HSCT) is a treatment for many diseases; however, it can induce complications such as Oral Mucositis (OM) and Graft-versus- Host Disease (GVHD). The neutrophil-lymphocyte ratio (NLR) is a peripheral biomarker of systemic inflammation and an independent prognostic factor for several inflammatory diseases. Aim: This study aimed to evaluate the association of NLR with OM and GVHD in patients undergoing allogeneic HSCT. Methods: Patients who underwent allogeneic HSCT at the Bone Marrow Transplant Service of the Hospital de Clínicas Complex of the Federal University of Paraná were included in the study. Socio-demographic data and blood counts were collected from patients' medical records. The NLR was calculated and associated with OM and GVHD. Results: 45 patients were included in the study. Although NLR was higher in patients with OM and oral GVHD, no statistical difference was observed, and no relationship between OM and GVHD with NLR could be stated. Conclusion: Although both OM and GVHD are associated with an inflammatory response as well as the immune system, it was not associated with NLR. Further investigation considering other variables related to HSCT might find possible associations, as it could favor patient management and prevention

Introdução: As redes extracelulares de neutrófilos (NETs) são apontadas como um dos mecanismos relevantes na patogênese da periodontite e artrite reumatoide (AR). No entanto, permanece pouco compreendido a participação das NETs como mecanismo de ligação entre as duas doenças. Objetivos: 1) Investigar a concentração das NETs na saliva, no plasma e in vitro em indivíduos com AR e controles saudáveis e a associação com a periodontite e atividade da AR; 2) Avaliar o impacto do tratamento periodontal não cirúrgico na concentração das NETs na saliva e no plasma; 3) Investigar a associação entre a presença de polimorfismos de nucleotídeo único no gene codificador da enzima peptidil arginina deaminase 4 (PAD4) com a AR, a produção de NETs in vitro e a periodontite; 4) Sistematizar as evidências disponíveis na literatura sobre o efeito do tratamento periodontal não cirúrgico sobre os principais parâmetros clínicos e laboratoriais da AR e score de atividade da doença 28 (DAS28). Material e Métodos: Para atender aos objetivos 1 e 2, a concentração de NETs na saliva, no plasma e na cultura de neutrófilos isolados do sangue periférico foi determinada por meio da identificação do complexo mieloperoxidase (MPO)-DNA com o uso do kit PicoGreen®. Para atender ao objetivo 3 foi realizada a extração do DNA genômico das células mononucleares do sangue periférico de indivíduos com AR e controles e foi realizada a genotipagem para os polimorfismos de nucleotídeo único PADI4_89, PADI4_90, PADI4_92 e PADI_104. Para atender ao objetivo 4 foi realizada uma overview incluindo revisões sistemáticas que avaliaram o efeito do tratamento periodontal não cirúrgico sobre os parâmetros da AR. A busca foi realizada nas principais bases de dados, sem restrição de idioma ou data de publicação. Foi realizada ainda uma meta-análise incluindo dados dos estudos primários identificados nas revisões sistemáticas analisadas. Resultados: 1) e 2) Indivíduos com AR e com periodontite apresentaram maior concentração de NETs na saliva, no plasma e in vitro. O tratamento periodontal não cirúrgico reduziu a concentração de NETs na saliva e plasma de indivíduos com AR. 3) Não foi observada associação entre a presença de genótipos polimórficos e a AR. A presença de um haplótipo homozigoto para o polimorfismo foi associada a uma maior produção de NETs in vitro e piores parâmetros periodontais. 4) Foram incluídas na overview nove revisões sistemáticas. Os principais desfechos avaliados foram DAS28; proteína C-Reativa e/ou velocidade de hemossedimentação. A meta-análise mostrou que o tratamento periodontal não cirúrgico resultou em diminuição significativa do DAS28. Conclusão: A concentração das NETs na saliva, no plasma e na cultura de neutrófilos de sangue periférico está associada a AR e a periodontite, podendo representar o elo entre as duas doenças. O tratamento periodontal não cirúrgico leva à redução da atividade da AR. Polimorfismos no gene PADI4 estão associados a produção de NETs in vitro e à presença de periodontite.


Introduction: Neutrophil extracellular traps (NETs) are recognized as one of the relevant mechanisms in the pathogenesis of periodontitis and rheumatoid arthritis (RA). However, the participation of NETs as a linking mechanism between the two diseases remains poorly understood. Objectives: 1) To investigate the concentration of NETs in saliva, plasma, and in vitro in individuals with RA and healthy controls and the association of NETs with periodontitis and RA activity; 2) To evaluate the impact of non-surgical periodontal treatment on the concentration of NETs in saliva and plasma; 3) To investigate the association between the presence of single nucleotide polymorphisms in the gene coding for the enzyme peptidyl arginine deaminase 4 (PAD4) with RA, in vitro production of NETs, and periodontitis; 4) To systematize the evidence available in the literature on the effect of non-surgical periodontal treatment on the main clinical and laboratory parameters of RA and disease activity score 28 (DAS28). Material and Methods: To accomplish Objectives 1 and 2, the concentration of NETs in saliva, plasma, and culture of neutrophils isolated from peripheral blood was determined by identifying the myeloperoxidase (MPO)-DNA complex using the PicoGreen kit®. To accomplish Objective 3, genomic DNA was extracted from peripheral blood mononuclear cells of individuals with RA and healthy controls, and genotyping was performed for single nucleotide polymorphisms PADI4_89, PADI4_90, PADI4_92, and PADI_104. To accomplish the Objective 4, an overview, including systematic reviews that evaluated the effect of non-surgical periodontal treatment on RA parameters, was performed. The search was carried out in the main databases, with no restriction on language or date of publication. A meta- analysis, including data from the primary studies identified in the analyzed systematic reviews, was also performed. Results: For Objectives 1 and 2, individuals with RA and periodontitis showed a higher concentration of NETs in saliva, plasma, and in vitro. Non-surgical periodontal treatment reduced the concentration of NETs in saliva and plasma of individuals with RA. For Objective 3, no association between the presence of polymorphic genotypes and RA was observed. The presence of a homozygous haplotype for the polymorphism was associated with a higher production of NETs in vitro and worse periodontal parameters. For Objective 4, nine systematic reviews were included in the overview. The main outcomes evaluated were DAS28, C-Reactive protein, and/or erythrocyte sedimentation rate. The meta- analysis showed that non-surgical periodontal treatment resulted in a significant decrease in DAS28. Conclusion: The concentration of NETs in saliva, plasma and culture of peripheral blood neutrophils is associated with RA and periodontitis and may represent the link between the two diseases. Non-surgical periodontal treatment leads to reduced RA activity. Polymorphisms in the PADI4 gene are associated with the in vitro production of NETs and with presence of periodontitis.

ABSTRACT Objective: To investigate the ability of a combination dental pulp mesenchymal stem cell secretome (DPMSCS), robusta green coffee bean extract (RGCBE), and Carboxymethylcellulose-Natrium (CMC-Na) in a Wistar rats model of traumatic ulcers. Material and Methods: Twenty-eight young, male, healthy Wistar rats (Rattus norvegicus) were divided into seven groups randomly: Group K0, group K1-3 (traumatic ulcer rats that received CMC-Na gel for three days), group K1-7 (traumatic ulcer rats that received CMC-Na gel for seven days), group K2-3 (traumatic ulcer rats that received RGCBE for three days), group K2-7 (traumatic ulcer rats that received RGCBE for seven days), group K3-3 (traumatic ulcer rats that received DPMSCS for three days), and group K3-7 (traumatic ulcer rats that received DPMSCS for seven days). An ulcer was made with an amalgam stopper on the right buccal mucosa of the rats. DPMSCS 50% gel was applied to the ulcer on the left buccal mucosa. The ulcer diameter was measured on day 3 and day 7. Results: There was a significant difference in the diameter of the ulcer, the number of neutrophils, and fibroblasts in the treatment group compared to the control group on day 7. Conclusion: A combination of DPMSCS and RGCBE 50% accelerates traumatic ulcer wound healing by lowering ulcer diameter, decreasing neutrophil counts, and increasing fibroblast proliferation in vivo.

Abstract Objective The aim of this study was to evaluate the effects of gliclazide on oxidative stress, inflammation, and bone loss in an experimental periodontal disease model. Material and Methods Male albino Wistar rats were divided into no ligature, ligature, and ligature with 1, 5, and 10 mg/kg gliclazide groups. Maxillae were fixed and scanned using micro-computed tomography to quantify linear and bone volume/tissue volume (BV/TV) and volumetric bone loss. Histopathological, immunohistochemical and immunofluorescence analyses were conducted to examine matrix metalloproteinase-2 (MMP-2), cyclooxygenase 2 (COX-2), cathepsin K, members of the receptor activator of the nuclear factor kappa-Β ligand (RANKL), receptor activator of nuclear factor kappa-Β (RANK), osteoprotegerin (OPG) pathway, macrophage migration inhibitory factor (MIF), superoxide dismutase-1 (SOD-1), glutathione peroxidase-1 (GPx-1), NFKB p 50 (Cytoplasm), NFKB p50 NLS (nuclear localization signal), PI3 kinase and AKT staining. Myeloperoxidase activity, malondialdehyde and glutathione levels, while interleukin-1 beta (IL-1β) and tumor necrosis factor-alpha (TNF-α) levels were evaluated by spectroscopic ultraviolet-visible analysis. A quantitative reverse transcription polymerase chain reaction was used to quantify the gene expression of the nuclear factor kappa B p50 subunit (NF-κB p50), phosphoinositide 3-kinase (PI3k), protein kinase B (AKT), and F4/80. Results Micro-computed tomography showed that the 1 mg/kg gliclazide treatment reduced linear bone loss compared to the ligature, 5 mg/kg gliclazide, and 10 mg/kg gliclazide treatments. All concentrations of gliclazide increased bone volume/tissue volume (BV/TV) compared to the ligature group. Treatment with 1 mg/kg gliclazide reduced myeloperoxidase activity, malondialdehyde, IL-1β, and TNF-α levels (p≤0.05), and resulted in weak staining for COX-2, cathepsin k, MMP-2, RANK, RANKL, SOD-1, GPx-1,MIF and PI3k. In addition, down-regulation of NF-κB p50, PI3k, AKT, and F4/80 were observed, and OPG staining was strong after the 1 mg/kg gliclazide treatment. Conclusions This treatment decreased neutrophil and macrophage migration, decreased the inflammatory response, and decreased bone loss in rats with ligature-induced periodontitis.

O objetivo deste estudo foi caracterizar as respostas das células mesenquimais indiferenciadas derivadas do ligamento periodontal (PDLSCs) ao extrato proteico total de Porphyromonas gingivalis (PgPE) e avaliar seu impacto nas propriedades biológicas das células leucêmicas promielocíticas humanas HL-60. PDLSCs enriquecidas com CD105 foram semeadas em placas de 6 poços durante 24h. Em seguida, as células foram desafiadas com PgPE (0 e 2 mg/ml) durante 3h (período de exposição). Os sobrenadantes foram então descartados; PDLSCs foram lavadas com PBS e cultivadas por 18h adicionais. Por fim, os sobrenadantes foram coletados. Os níveis de citocinas e quimiocinas nos sobrenadantes foram avaliados por ensaios multiplex. Na sequência, o efeito dos sobrenadantes derivados de PDLSCs (tratadas ou não com PgPE) sobre a ativação, o recrutamento e a resposta inflamatória de HL-60 foi avaliado. PDLSCs responderam ao tratamento com PgPE. RANTES, eotaxina e IP-10 (proteína produzida por IFN-?) foram detectados apenas em sobrenadantes de PDLSCs/PgPE. Além disso, PgPE induziu maior secreção de proteína quimiotática de neutrófilos (MCP)-1, intérferon (IFN)-?, interleucina (IL)-6, IL-8 e IL-1ra (p> 0,05). O recrutamento de HL-60D aumentou 4,7 vezes quando estas células foram expostas a sobrenadantes PDLSCs/PgPE, enquanto que os sobrenadantes de PDLSCs não afetaram a quimiotaxia de HL-60D. Sobrenadantes PDLSCs promoveram uma redução de 16% na produção de espécies de oxigênio reativo (ROS) por HL-60D estimuladas por PMA (p=0,013). Em contraste, sobrenadantes PDLSCs/PgPE promoveram um aumento de 1,78±1,04 vezes (p=0,046) na produção de ROS. Finalmente, tanto sobrenadantes PDLSCs, como sobrenadantes PDLSCs/PgPE, não influenciaram a produção de fator de necrose tumoral (TNF)-? e IL-1? pelas HL-60D em resposta ao lipopolissacarídeo (LPS). Esses achados sugerem um importante papel das PDLSCs no reconhecimento de P. gingivalis, recrutamento de células imunes inatas e ativação de mecanismos antimicrobianos.

O objetivo deste trabalho foi avaliar a utilização do LPRF após extração de terceiros molares inferiores. Neste estudo prospectivo, duplo cego, boca-dividida, 20 pacientes foram avaliados nos seguintes parâmetros: regeneração óssea, dor e cicatrização de tecido mole. A avaliação da densidade óssea, foi realizada através de tomografias computadorizadas de feixe cônico, realizadas no período pós-operatório imediato e, no pós-operatório, após 3 meses. O programa de computador ITK-SNAP foi utilizado para segmentação e avaliação das imagens através da intensidade de cinza de cada voxel. Na avaliação dos desfechos secundários, foram realizadas as seguintes análises: dor, através de escala analógica visual e numérica, e tecido mole, através da escala de Landry et al. e sondagem pré e pós-operatória da região distal aos segundos molares inferiores. O resultado encontrado nos Grupos Teste e Controle foram então confrontados na busca de diferenças entre as duas técnicas. Observou-se diferença estatística tendo o grupo teste mostrado maior densidade óssea após 3 meses, p<0.05. Os valores obtidos através da escala analógica visual e numérica, para avaliação de dor, foram submetidos ao Teste t, onde p>0.05 e os parâmetros para avaliação de tecido mole e sondagem periodontal foram submetidos ao teste de Mann-Whitney, onde p>0.05. Para os parâmetros dor e cicatrização tecidual não houve diferença estatística entre os grupos.


The purpose of this study was to assess the effect of LPRF after mandibular third molar extraction. In this prospective, double bind, split-mouth study, 20 patients were evaluated. The patients were assessed for post-operative bone regeneration, pain and soft tissue healing. The evaluation of bone healing, primary outcome, was performed through tomographic evaluation in the immediate postoperative period and 3 months after the procedure. The ITK-SNAP software was used for image evaluation by the intensity of gray of each voxel. In the evaluation of secondary outcomes, the variables assessed were: pain, through visual and numerical analogue scale and soft tissue, using Landry et al. and pre and postoperative probing of the distal region of lower second molar. The evaluation of bone healing, primary outcome, showed a significant difference, with test group showing higher bone density after 3 months, p<0.05. The results found in the Test and Control Groups were then confronted searching differences between the two techniques. The values obtained through the visual and numerical analogue scale for pain evaluation were submitted to the t test, where p> 0.05. The parameters for evaluation of soft tissue and periodontal probing were submitted to the Mann-Whitney test, where p> 0.05. For the parameters pain and tissue healing there was no statistical difference between the groups.

Além disso, PgPE induziu maior secreção de proteína quimiotática de neutrófilos (MCP)-1, intérferon (IFN)-?, interleucina (IL)-6, IL-8 e IL-1ra (p> 0,05). O recrutamento de HL-60D aumentou 4,7 vezes quando estas células foram expostas a sobrenadantes PDLSCs/PgPE, enquanto que os sobrenadantes de PDLSCs não afetaram a quimiotaxia de HL-60D. Sobrenadantes PDLSCs promoveram uma redução de 16% na produção de espécies de oxigênio reativo (ROS) por HL-60D estimuladas por PMA (p=0,013). Em contraste, sobrenadantes PDLSCs/PgPE promoveram um aumento de 1,78±1,04 vezes (p=0,046) na produção de ROS. Finalmente, tanto sobrenadantes PDLSCs, como sobrenadantes PDLSCs/PgPE, não influenciaram a produção de fator de necrose tumoral (TNF)-? e IL-1? pelas HL-60D em resposta ao lipopolissacarídeo (LPS). ABSTRACT Sesquiterpene lactones (SLs) are the active constituents of a variety of medicinal plants used in traditional medicine for the treatment of inflammatory diseases and other ailments. Objective In this study, we evaluated whether budlein A modulates the activation of innate and adaptive immune cells such as neutrophils and lymphocytes. Material and Methods Our research group has investigated several plant species and several compounds have been isolated, identified, and their medical potential evaluated. Budlein A is a SL isolated from the species Aldama buddlejiformis and A. robusta (Asteraceae) and shows anti-inflammatory and anti-nociceptive activities. Advances in understanding how plant-derived substances modulate the activation of innate and adaptive immune cells have led to the development of new therapies for human diseases. Results Budlein A inhibited MPO activity, IL-6, CXCL8, IL-10, and IL-12 production and induces neutrophil apoptosis. In contrast, budlein A inhibited lymphocyte proliferation and IL-2, IL-10, TGF-β, and IFN-γ production, but it did not lead to cell death. Conclusions Collectively, our results indicate that budlein A shows distinct immunomodulatory effects on immune cells.

Galectina-3, uma proteína que se liga a -galactosídeos, é expressa por neutrófilos e inúmeras evidências indicam que esta molécula atua como uma possível reguladora da resposta imune. Sabe-se que galectina-3 ao ligar com LPS pode levar a formação de oligômeros, que podem alterar o limiar de ativação de células da resposta imune inata. Apesar de existirem diversos estudos que mostram a influência de galectina-3 na resposta de neutrófilos frente a componentes bacterianos, os resultados são em sua maioria contraditórios e inconclusivos. Para elucidar a influência da galectina-3 na reposta imune inata a patógenos periodontais, o presente trabalho avaliou a atividade antimicrobiana in vitro de neutrófilos, isolados de camundongos selvagens (WT) ou geneticamente deficientes de galectina-3 (Gal-3KO), previamente estimulados com LPS de Aa e Pg. Os resultados não evidenciaram diferenças significativas no número de unidades formadoras de colônia (UFC) recuperadas das culturas de neutrófilos provenientes de animais deficientes de galectina-3 e do grupo controle (WT). Contudo, a estimulação de neutrófilos com LPS por 18 horas levou a redução no número de UFC recuperadas das culturas, quando comparado com as culturas estimuladas com LPS por apenas 3 horas.


Galectin-3, a protein that binds -galactosides, is expressed by neutrophils and numerous evidences indicate that this molecule acts as a possible regulator of the immune response. It is known that galectin-3 binding to LPS can lead to the formation of oligomers and thus changing the activation threshold of cells of the innate immune response. Although there are several studies that show the influence of galectin-3 in neutrophil response against bacterial components, the results are conflicting and inconclusive in their majority. To elucidate the influence of galectin-3 in the innate immune response to periodontal pathogens, the present study evaluated the in vitro antimicrobial activity of neutrophils, isolated from wild-type or galectin-3 deficient mice, previously stimulated with LPS of Aa and Pg. The results showed no significant differences in the number of colony forming units (CFU) recovered from cultured galectin-3 deficient neutrophils or control group. However, in a 18 hours time course of LPS stimulation, we observed reduction in the number of CFU, when compared to 3 hours of LPS stimulation.

O objetivo deste trabalho foi investigar a influência da extensão da doença periodontal no perfil sanguíneo de ratos Wistar. Foram utilizados 30 ratos divididos em 3 grupos de 10 animais: C- ratos controle; DP1- ratos com doença periodontal em 3 molares superiores; DP2 ­ ratos com doença periodontal em 3 molares superiores e 3 inferiores. A doença periodontal foi induzida por meio da confecção de ligadura em torno do colo dentário dos molares selecionados de acordo com cada grupo. Após 30 dias, os animais foram anestesiados e, por meio de uma punção cardíaca, foi coletado 5ml de sangue para as análises dos parâmetros do hemograma, creatinina, triglicérides e colesterol. Em seguida, os animais foram sacrificados e as maxilas removidas e processadas para análise histopatológica em coloração de H.E. para caracterizar o perfil da doença periodontal em cada animal. Os resultados obtidos a partir do tecido hematológico foram analisados estaticamente por meio dos testes de ANOVA e Tukey, com nível de significância de 5%. Pode-se observar nos grupos DP1 e DP2 presença de infiltrado inflamatório crônico, perda de inserção conjuntiva, perda de estrutura óssea cortical e alveolar, presença de reabsorções dentárias, presença de biofilme e sequestro ósseo. Quanto ao perfil sanguíneo, pode-se observar diferença estatisticamente significante entre o grupo controle e os grupos DP1 e DP2 em relação à quantidade de leucócitos e linfócitos (p<0,05). Ainda, o grupo DP2 apresentou maior quantidade de leucócitos e neutrófilos em relação ao grupo DP1 e controle (p<0,05). Pode-se concluir que a presença da doença periodontal eleva a quantidade de leucócitos, neutrófilo e linfócitos no sangue de ratos Wistar, e que, a extensão da doença periodontal influencia na quantidade de leucócitos e neutrófilos(AU)


The aim of this study was to evaluate the influence of the periodontal disease extension on the blood profile of Wistar rats. Thirty rats were divided into 3 groups of 10 animals each: C control rats; DP1- rats with periodontal disease in 3 upper molars; DP2 - rats with periodontal disease in 3 upper and 3 lower molars. Periodontal disease was induced by ligature around the tooth according to each group. After 30 days, the animals were killed and 5ml of blood was collected for analyzes of blood count parameters, creatinine, triglycerides and cholesterol. Then the animals were sacrificed and the jaws removed and processed for histopathological analysis to characterize, in H.E. staining, the profile of periodontal disease in each animal. The results from the hematologic tissue were analyzed statistically by ANOVA and Tukey test, with 5% significance level. It can be observed in groups DP1 and DP2 presence of chronic inflammatory infiltrates, connective tissue attachment loss and alveolar bone cortical structure loss, presence of resorptions, presence of biofilm and bone sequestration. As for the blood profile, it is can observe a statistically significant difference between the control group and the groups DP1 and DP2 to the amount of leukocytes and ymphocytes (p <0.05). Further, DP2 group had a higher amount of leukocytes and neutrophils compared to DP1 and control group (p <0.05). It can be concluded that the presence of periodontal disease increases the amount of leukocytes, neutrophils, and lymphocytes in the blood of Wistar rats, and the extent of periodontal disease affects the quantification of leukocytes and neutrophils(AU)

A hiperglicemia, bem como o diabetes, é reconhecida como fator de risco para infecções pós-cirúrgicas, assim manter o controle glicêmico perioperatório tem sido padrão de cuidado em saúde. Entretanto há poucos estudos sobre o impacto do controle glicêmico no processo de reparação de cirurgias de extração dentária. Apesar da escassez de estudos e de evidências clínicas e científicas que investiguem o risco de infecções pós-exodônticas em pessoas com diabetes, existem livros, artigos e guias que recomendam aos dentistas o uso de antibióticos profiláticos nesses indivíduos, especialmente para aqueles descompensados. O objetivo deste estudo foi avaliar clinicamente a cicatrização pós-exodôntica, em relação à cronologia dos eventos reparacionais e em relação à ocorrência de complicações pós-operatórias, em indivíduos com diabetes tipo 2 comparados a um grupo controle. Além disso, procuramos relacionar os eventos pós-cirúrgicos com o controle metabólico e com o perfil imunológico dos pacientes. Este estudo prospectivo longitudinal caso controle incluiu 53 indivíduos com diabetes tipo 2 (grupo de estudo=GE) e 29 indivíduos sem diabetes (grupo controle=GC). Foi aplicado questionário sobre a história médica, realizados exames clínicos intraoral e extraoral e exames laboratoriais para conhecimento do controle glicêmico e do perfil imunológico do indivíduo, tais como, hemograma completo, hemoglobina glicada (A1C), glicemia de jejum, IgA, IgG, IgM, C3, C4, linfócitos T CD3+, CD4+, CD8+, quimiotaxia de neutrófilos, oxidação de neutrófilos, fagocitose de neutrófilos e monócitos. Hyperglycemia and diabetes are recognized as a risk factor for postoperative infections. Thus, maintaining perioperative glucose control has become the standard of care. However, there are scarce data on the appropriate glucose control during minor dental surgery. Despite the paucity of studies investigating the risk of postsurgical oral infections in persons with diabetes, there are text books, papers and guidelines recommending to dentists the use of prophylactic antibiotics for patients with poorly controlled diabetes undergoing invasive oral procedures. The aim of this study was to clinically evaluate the post extraction healing regarding to the chronology and to the occurrence of postoperative complications in patients with type 2 diabetes compared to a control group. Additionally, we associated the postoperative events with metabolic control and the immunological profile of the participants. This prospective case-control study included 53 subjects with type 2 diabetes (SG) and 29 controls without diabetes (CG). Um material obturador deve apresentar boas propriedades biológicas e físico-químicas, uma vez que ficará em íntimo contato com os tecidos periapicais. Sendo assim, não deve ser irritante aos tecidos adjacentes, possibilitando, ou mesmo favorecendo, o reparo da região periapical, para isso não devem alterar o processo inflamatório. O presente estudo avaliou a citotoxicidade dos cimentos AH Plus, Sealapex, MTA Fillapex e Sealepox em neutrófilos humanos. Neutrófilos humanos cultivados na presença ou ausência de LPS foram tratados com os cimentos em diferentes diluições (1:1, 1:4, 1:8, 1:16 e 1:32) e tempo de presa (24 e 48 horas) durante 24 horas. A viabilidade celular foi analisada por citometria de fluxo, a dosagem de CXCL8 pelo método de ELISA, e Óxido Nítrico na absorvância de 540nm. Os dados foram analisados com o auxílio do programa GraphPad Prism5 por ANOVA a 1 critério seguido pelo teste de Tukey, e os valores considerados significantes quando p<0,05. O cimento AH Plus interferiu apenas na síntese de NO, estimulando-a, tendo a diluição 1:16 melhor comportamento biológico, em ambos períodos experimentais. O Sealapex diminuiu a produção de NO, sendo significante para 1:32 em 48 horas. O MTA Fillapex induziu apoptose, a produção de CXCL8 (1:4 e 1:8 em 48 horas) e diminuiu a síntese de NO (1:32 em 48 horas). Sealepox diminuiu a apoptose (1:16 e 1:32 em 24 horas) e interferiu na produção de CXCL8, diminuindo-a (1:8 em 48 horas, e 1:16 em ambos os períodos). A citotoxicidade em ordem crescente dos cimentos foram: AH Plus, Sealapex, MTA Fillapex e Sealepox. Nosso estudo concluiu que os cimentos AH Plus e Sealapex foram os menos citotóxicos, que menos interferiram na viabilidade celular e na sua função (não indução de CXCL8 e na produção de NO), tanto em 24 horas como em 48 horas. MTA Fillapex e o Sealepox, apesar de causarem mais morte celular e interferirem na produção de NO e CXCL8, seus efeitos podem ser aceitáveis, uma vez que os níveis...


A filling material must have good biological and physicochemical, since you'll be in close contact with the periapical tissues. Thus it should not be irritating to the surrounding tissues, allowing, or even encouraging, the repair of the periapical region, for it must not alter the inflammatory process. The present study evaluated the cytotoxicity of AH Plus, Sealapex, MTA and Fillapex Sealepox in human neutrophils. Human neutrophils cultured in the presence or absence of LPS cements were treated with different dilutions (1:1, 1:4, 1:8 , 1:16, 1:32) and setting time (24 and 48 hours) for 24 hours. Cell viability was analyzed by flow cytometry, the dosage of CXCL8 by ELISA and nitric oxide in absorbance at 540 nm. Data were analyzed with the aid of GraphPad Prism5 ANOVA for the first criterion followed by Tukey test, and values considered significant when p < 0.05. The AH Plus sealer interfered only in NO synthesis , stimulating it , having a 1:16 dilution better biological behavior , in both experimental periods . The Sealapex decreased production of NO, with 1:32 significant to within 48 hours. The MTA Fillapex induced apoptosis, production of CXCL8 (1:4 to 1:8 within 48 hours) and reduced NO synthesis (1:32 in 48 hours). Sealepox decreased apoptosis (1:16 and 1:32 in 24 hours) and interfere with the production of CXCL8, reducing it (1:8 in 48 hours, and 1:16 in both periods). The cytotoxicity of the cements in ascending order were: AH Plus, Sealapex, MTA and Fillapex Sealepox. We conclude that AH Plus and Sealapex cements were less cytotoxic than less interfered with cell viability and function ( CXCL8 and noninduced NO production ) both in 24 hours and in 48 hours. MTA and Fillapex Sealepox, although most causing cell death and interferes the production of NO and CXCL8, their effects may be acceptable, since the levels of these changes are so mild, non-aggressive.

Periodontal disease (PD) is characterized as an inflammatory process that compromises the support and protection of the periodontium. Patients with Down’s syndrome (DS) are prone to develop PD. Neutrophils (NE) are the first line of defense against infection and their absence sets the stage for disease. Aim: To compare the activity and function of NE in the peripheral blood from DS patients with and without PD, assisted at the Center for Dental Assistance to Patients with Special Needs affiliated with the School of Dentistry of Araçatuba, Brazil. Methods: Purified NE were collected from peripheral blood of 22 DS patients. NE were used to detect the 5-lypoxigenase (5-LO) expression by RT-PCR. Plasma from peripheral blood was collected to measure tumor necrosis factor-a (TNF-a) and interleukin-8 (IL-8) by ELISA and nitrite (NO3) using a Griess assay. Results: Data analysis demonstrated that DS patients with PD present high levels of TNF-a and IL-8 when compared with DS patients without PD. However, there was no statistically significant difference in the levels of NO3 production between the groups. The levels of the inflammatory mediator 5-LO expression increased in DS patients with PD. Conclusions: According with these results, it was concluded that TNF-a and IL-8 are produced by DS patients with PD. Furthermore, DS patients with PD presented high levels of 5-LO expression, suggesting the presence of leukotriene B4 (LTB4) in PD, thus demonstrating that the changes in NE function due to the elevation of inflammatory mediators contribute to PD.

O exsudato purulento (pus) indica presença de bactérias no local. Podemos afirmar que convivemos, compartilhamos e nos equilibramos biologicamente com a grande quantidade de bactérias que temos em nossas superfícies externas e internas. Entretanto, o organismo tem que se preparar para a eventualidade de bactérias entrarem no meio interno. No dia a dia, a possibilidade de entrada bacteriana no meio interno é muito grande e ela ocorre praticamente todos os dias. Dessa forma, quando bactérias entram nos tecidos conjuntivos, a inflamação é imediatamente desencadeada. Os vasos sanguíneos deixarão passar para a região muitas substâncias plasmáticas, ou exsudato, e também muitas células leucocitárias que, no conjunto, recebem o nome de infiltrado.

Os tecidos periodontais estão em confronto continuo com microorganismos capazes de disparar mecanismos da resposta imune inata, dando origem ao infiltrado inflamatório. Estudos recentes mostraram a importancia dos receptores do tipo Toll (TLRs) na fase inicial de reconhecimento de diferentes patogenos. A participação de receptores tipo Toll (TLRs) na resposta de neutrófilos e macrófagos frente a periodontopatógenos precisa ser determinada. Nesse estudo procuramos caracterizar o infiltrado inflamatório presente no peritônio de animais deficientes de TLR4-/-, avaliar a atividade fagocítica, bem como a produção de óxido nítrico (NO) e a atividade de mieloperoxidase (MPO) no curso da infecção por Aggregatibacter actinomycetemcomitans. A ausência de TLR4 não influenciou a quimiotaxia de neutrófilos e macrófagos para o local da infecção, a produção de óxido nítrico, a atividade de MPO e a viabilidade celular. No entanto, neutrófilos e macrófagos de animais TLR4-/- apresentaram menor atividade fagocítica quando comparado ao grupo controle (camundongos WT). Em relação a doença periodontal induzida experimentalmente com Aggregatibacter actinomycetemcomitans em camundongos deficientes de TLR4, os resultados mostraram que 100% dos animais deficientes de TLR4 sobreviveram a infecção durante o período de observação. Em relação a análise de perda óssea, os dados revelaram uma menor perda progressiva de osso alveolar na região dos molares de animais deficientes de TLR4. A ausência do receptor interferiu na disseminação da bactéria, uma vez que se observou um grande número de bacilos no linfonodo e baco dos animais que não expressaram TLR4, diferente do observado para os animais selvagens (WT). Os resultados indicam a importância da sinalização via TLR4 durante a resposta imune contra Aggregatibacter actinomycetemcomitans (AU).


Aggregatibacter actinomycetemcomitans is an oral gram negative bacteria that contributes to periodontitis progression. Isolated antigens from A. actinomycetemcomitans could be activating innate immune cells through Toll-like receptors (TLRs), molecules that recognize structural components conserved among microorganisms. In this study, we evaluated the role of TLR4 in the recognition of Aggregatibacter actinomycetemcomitans. Neutrophils and macrophage from TLR4 deficient mice and WT mice were collected and used for the subsequent assays. The phagocytosis of leukocytes against A. actinomycetemcomitans and the presence of apoptotic cells were determined by flow cytometry. The in vivo and in vitro production of NO and MPO was evaluated 24h after A. actinomycetemcomitans challenge. In addition, we examined the mechanisms that modulate the outcome of A. actinomycetemcomitans-induced periodontal disease in TLR4-/- mice. The results show that inflammatory cells influx in peritoneal cavity of TLR4-/- mice was similar to that observed into their littermate controls. The phagocytic activity was diminished by cells from TLR4-/- mice. In addition, we did not observe difference in NO and MPO production and the frequency of apoptotic cells between cells from TLR4-/- and WT mice. The results showed that TLR4-deficient mice developed less severe periodontitis after A. actinomycetemcomitans infection, characterized by significantly lower bone loss and inflammatory cell migration to periodontal tissues. Together, these data demonstrate the role TLR4 signals for neutrophils activation afterA. actinomycetemcomitans infection and development of periodontal disease.

A destruição dos tecidos periodontais está associada a liberação de enzimas proteolíticas e espécies reativas de oxigênio, predominantemente de neutrófilos ativados por receptores Fcy. O objetivo do estudo foi revisar a literatura sobre a importância do receptor Fcy na hiperativação de neutrófilos e, consequentemente, na suscetibilidade do hospedeiro a periodontite. A ativação do neutrófilo requer a ligação dos receptores Fcy por moléculas de Ig que, através de mediadores intracelulares, resulta em eventos de transdução de sinal, incluindo a transcrição de genes codificadores de citocinas, enzimas microbicidas e mobilização do citoesqueleto, levando a fagocitose, exocitose de grânulos e migração celular. Na hiperatividade do neutrófilo, suas funções estão exacerbadas. Uma explicação plausível para essa hiperatividade vem da ativação do receptor Fcy visto que os mesmos respondem de forma distinta a diferentes subclasses de IgG, dependendo: (1) de sua afinidade a estes e da atividade específica de cada categoria do receptor Fcy; (2) pela expressão do receptor Fcy pelas células; e/ou (3) da presença de polimorfismo do receptor Fcy. Sendo assim, podemos concluir que a destruição tecidual no indivíduo portador de periodontite pode estar relacionada a uma hiperatividade de neutrófilos quando ativados por IgG 1 e 2 através dos seus receptores FcαRII e III. Isto se dá principalmente nos casos de polimorfismo para FcαR II a 131, e em meio às altas concentrações de elastase, possibilitando ao FcαR maior afinidade ao ligante, independente dos patógenos periodontais Aa e Pg e do diagnóstico para periodontite ser agressiva ou crônica.


The destruction of periodontal tissue is associated with the release of proteolytic enzymes and reactive oxygenspecies, predominantly by Fcγ activated neutrophils. The aim was to review the literature about the importance of Fcy receptors in the hyperactivation of neutrophils and thus, in the susceptibility of the host in periodontitis. The activation of neutrophil requires Fcy receptor (FcγR) binding to Ig molecules that, through intracellular mediators, results in signal transduction events, including the transcription of genes for cytokines, microbicides enzymes and mobilization of the cytoskeleton, leading to phagocytosis, exocytosis of granules and cell migration. During neutrophilhyperactivity, such functions are exacerbated. A plausible explanation this hyperactivity comes from the FcγR, since they respond differently to different subclasses of IgG. It depends on: (1) its affinity to the receptors and tothe specific activity of each category of FcγR, (2) the expression of the FcγR in cells, and/or (3) polymorphisms forFcγR. Therefore, we concluded that tissue destruction in individuals with periodontitis may be related to a hyperactivity of neutrophils when activated by IgG 1 and 2 through its receptors FcγR II and III. This happens especially in cases of polymorphism for FcγR IIa 131, but also the presence of high concentrations of elastase, which allows a greatest affinity to the FcγR ligand, regardless the presence of periodontal pathogens Aa and Pg, and the specificity of the diagnostics to aggressive or chronic periodontitis.

Os neutrófilos são as primeiras células a migrar para sítios de infecção e desempenham importante papel na defesa contra vários patógenos, especialmente Candida albicans. A função dos neutrófilos envolve fagocitose e destruição de leveduras e formas filamentosas do fungo, além de produção de citocinas e quimiocinas, capazes de ativar outras células envolvidas na eliminação de C. albicans. Recentemente, tem sido mostrado que estas células apresentam prejuízos com o avanço da idade e estes fenômenos fazem parte de uma área da Imunologia conhecida como Imunossenescência. A estomatite protética é a lesão oral mais verificada em idosos e a sua persistência é diretamente relacionada com a presença de espécies de Candida, principalmente C. albicans, nas lesões e na superfície interna de próteses, especialmente totais superiores (PTS). Com o objetivo de avaliar possíveis alterações em neutrófilos que poderiam predispor usuários de prótese total superior à estomatite protética, foram analisados neutrófilos de sangue e de saliva de idosos e jovens com estomatite protética associada à Candida (EPC), bem como de indivíduos controles. No primeiro capítulo foi avaliada a positividade de Candida e as diferentes espécies deste patógeno em lesões de estomatite protética e superfície interna de PTS. C. albicans foi a espécie mais detectada em lesões de EP e na PTS. No segundo capítulo, os neutrófilos do sangue dos indivíduos da pesquisa foram caracterizados quanto ao fenótipo ex vivo, função fagocítica e produção de citocinas e quimiocinas após desafio com C. albicans. Neutrófilos do sangue de indivíduos com EPC, bem como de idosos sem EPC, apresentaram características relacionadas com suscetibilidade às doenças causadas por C. albicans. Neutrophils are the first line of immune cells to migrate into infection sites, playing an important role against pathogens, especially Candida albicans. These cells phagocytose and destroy yeasts or filamentous forms of this fungus, beyond secrete cytokines and chemokines that activate other immune cells to eliminate C. albicans. Nowadays, it has been shown that the ageing damages neutrophil function and such events are described as part of Imunosenescence. Denture stomatitis is the most oral lesion present in elderly, and its persistence is related to Candida species, especially C. albicans, in the lesions and prosthesis fitting surface, particularly maxillary prosthesis (MP). In order to evaluate possible neutrophil alterations predisposing denture wearers to Candida- related denture stomatitis (DS), blood and salivary neutrophils from elderly and younger individuals with DS, as well control ones, were analyzed. Thus, the first chapter addresses the presence of Candida species on the denture stomatitis lesions and MP fitting surface. C. albicans was the most species detected in DS lesions and MP. On the second chapter, blood neutrophils from the individuals were characterized about ex vivo phenotype, phagocytic function, and cytokines and chemokines production after challenged with C. albicans. RESUMO

O Objetivo do presente estudo foi avaliar a quantidade de elastase livre no fluido gengival de pacientes com periodontite crônica. Uma população não tratada com inflamação gengival foi utilizada. Amostras do fluido gengival foram coletadas, de 19 pacientes do Grupo controle (GC) que apresentavam gengivite, e de 24 pacientes do Grupo experimental (GE) portadores de periodontite crônica. Os sítios foram divididos em sítios sem destruição tecidual em pacientes com gengivite (sítios GG); sítios sem destruição tecidual em pacientes com periodontite (sítios GP) e sítios com destruição tecidual em pacientes com periodontite (sítios PP). Os parâmetros clínicos utilizados foram : Índice de placa (IP), Índice Gengival (IG), Profundidade de Bolsa a sondagem (PBS) e nível de inserção a sondagem (NIS). Os resultados demonstraram que a atividade de elastase estava significativamente aumentada em sítios PP, enquanto a lactoferina estava aumentada nos sítios GG. Em todos os sítios analisados, a maior parte da elastase estava presente na forma de elastase livre. A taxa entre atividade enzimática de elastase e lactoferina foi significativamente mais elevada nos sítios PP quando comparados aos sítios GP e GG, o que confirma a hiperatividade de neutrófilos em sítios com destruição tecidual em pacientes portadores de periodontite do adulto.
