A etiopatogenia da doença periodontal é complexa e exige estudos constantes em alternativas terapêuticas que possibilitem seu controle. Aggregatibacter actinomycetemcomitans está associado fortemente à periodontite crônica e à periodontite agressiva. A tecnologia do plasma frio sob pressão atmosférica tem potencial para uso na odontologia, mas sua aplicação na periodontia ainda é pouco explorada. Este estudo teve como objetivo avaliar os efeitos do plasma de baixa temperatura sob pressão atmosférica (PBTPA) como adjuvante no controle da doença periodontal experimental induzida por ligadura, inoculada com A. actinomycetemcomitans, na região cervical do primeiro molar inferior em modelo murino. Inicialmente, foram determinados os parâmetros físicos efetivos para inibição de biofilmes de A. actinomycetemcomitans utilizando cepa padrão American Type Culture Collection ATCC 29523. Foram realizados testes de screening inicial com a verificação da inibição do crescimento por exposição ao PBTPA de A. actinomycetemcomitans semeados em placas de Petri. Posteriormente, foram formados biofilmes em placas de microtitulação e foram determinados os parâmetros físicos efetivos do PBTPA. Foram realizados experimentos in vivo de indução de doença periodontal por A. actinomycetemcomitans em modelo utilizando ligaduras em ratos. A tomografia computadorizada de feixe cônico foi utilizada para avaliar o nível de perda óssea após período experimental. Os resultados mostraram que o PBTPA foi efetivo na eliminação do micro-organismo observado através da formação de halos de inibição nos tempos de 5 e 7, 5 minutos e sobre biofilmes de A. actinomycetemcomitans expostos ao PBTPA no tempo de 5 minutos. Os resultados exibiram diferença estatística significativa (p <0,001) na distância entre a junção esmalte-cemento e a crista óssea alveolar, do grupo tratado com PBTPA em relação ao grupo raspagem e ao grupo controle. Os resultados do presente estudo sugerem o potencial do PBTPA no controle da periodontite induzida, contudo análises adicionais são necessárias(AU)


The etiopathogenesis of periodontal disease is complex requiring constant studies of therapeutic alternatives that enable its control. Aggregatibacter actinomycetemcomitans is strongly associated with chronic periodontitis and aggressive periodontitis. Cold plasma technology under atmospheric pressure has potential for use in dentistry, but its application in periodontics is still not well explored. This study aimed to evaluate the effects of low temperature atmospheric pressure plasma (PBTPA) as an adjuvant in the control of experimental periodontal disease induced by ligature inoculated with A. actinomycetemcomitans in the cervical region of the lower 1st molar in a murine model. Initially, effective physical parameters for inhibition of A. actinomycetemcomitans biofilms were determined using the standard strain American Type Culture Collection ATCC 29523. Initial screening tests were performed to verify growth inhibition after exposure of A. actinomycetemcomitans seeded in Petri dishes to the PBTPA. Subsequently, biofilms were formed on microtiter plates and the effective physical parameters of the PBTPA were determined. In vivo experiments were carried out to induce A. actinomycetemcomitans periodontal disease induced by ligature in a rat model. Cone beam computed tomography was used to assess the level of bone loss after the experimental period. The results showed that PBTPA was effective in eliminating the microorganism observed through the formation of halos inhibition during 5 and 7.5 minutes and in A. actinomycetemcomitans biofilms exposed to PBTPA at 5 minutes. The previous study using PBTPA in 5 minutes did not show cytotoxicity in Vero cells. The results showed a statistically significant difference (p <0.0001) in the distance between the enamel-cement joint and the alveolar bone crest, of the group treated with PBTPA in relation to the scalling group and the control group, confirming the potential of the technique in dentistry(AU)

Abstract Objective Obesity is a chronic disease that negatively affects an individual's general and oral health. The present study aimed to compare the clinical and microbiological effects of non-surgical periodontal therapy with the full mouth disinfection (FMD) protocol on obese and non-obese individuals at 9 months post-therapy. Methodology This clinical study was first submitted and approved by the Ethics Committee. Fifty-five obese patients and 39 non-obese patients with periodontitis were evaluated. The full-mouth periodontal clinical parameters, clinical attachment level (CAL), probing depth (PD), gingival index (GI), and plaque index (PI), were monitored at baseline, 3, 6, and 9 months after periodontal treatment with full mouth disinfection (FMD) protocol. The mean count of Tannerella forsythia , Porphyromonas gingivalis , Treponema Denticola , and Aggregatibacter actinomycetemcomitans was determined by quantitative polymerase chain reaction on subgingival biofilm samples. Demographic data were assessed by Chi-square test. For clinical and microbiological parameters, two-factor repeated-measures ANOVA was used. Results In both groups, periodontal therapy using the one-stage full-mouth disinfection protocol significantly improved CAL, PD, GI, and PI (p<0.05). Obese and non-obese patients equally responded to non-surgical periodontal therapy (p>0.05). Microbial count found no major differences (p>0.05) between obese and non-obese individuals who had undergone non-surgical periodontal therapy. Conclusions Obesity did not affect the clinical and microbiological outcomes of non-surgical periodontal therapy.

Abstract There are several controversies regarding the efficacy of homeopathic substances; however, these remedies are used in many countries for the treatment of various pathological conditions. The purpose of this study was to evaluate the in vitro antibacterial activity of two homeopathic tinctures Arsenicum album (mineral extract) and Lycopodium clavatum (plant extract) on the periodontal bacteria Actinomyces israelii, Streptococcus sanguinis, Prevotella intermedia, Aggregatibacter actinomycetemcomitans and Phorphyromonas gingivalis (P. gingivalis). Materials and methods: Equal numbers of bacteria were seeded on agar plates containing enriched media with the homeopathic solutions at 1dH and 1cH dilutions. After 7 days of incubation under anaerobic conditions, colony forming units (CFUs) were counted. The antibacterial effect was calculated based on the total number of CFUs observed on non-tincture containing agar, and on the tincture containing plates. Results: No visible growth of any of the strains was observed on the plates containing Arsenicum album at any of the dilutions tested. In contrast, when Lycopodium clavatum at 1cH dilution was tested, only P. gingivalis was susceptible to this compound. Conclusions: The results suggest that the mineral extract tincture had a greater antibacterial activity than the plant extract tincture, also Lycopodium clavatum preparation could be an effective inhibitor of periodontal pathogens bacteria such as P. gingivalis.

A Periodontite Agressiva (PA), que atualmente pertence ao grupo das Periodontites estágios 3 e 4, distingue-se dos demais tipos de doença periodontal por seu início precoce, agregação familiar dos casos e por afetar pacientes sistemicamente saudáveis. Além disso, pode ser subclassificada em duas formas, localizada (PAL) e generalizada (PAG), em função de sua extensão. Muitas vezes, os depósitos de biofilme bacteriano são desproporcionais à quantidade de destruição óssea e perda de inserção que o paciente apresenta, independente da subclassificação. O microrganismo mais relacionado à etiopatogênese da doença é o Aggregatibacter actinomycetemcomitans (A. actinomycetemcomitans), incluindo os seus principais sorotipos a, b e c, amplamente estudados. Associado a estas condições, A. actinomycetemcomitans apresenta alguns fatores de virulência como uma leucotoxina, principalmente ligada ao sorotipo b - clone JP2 (que é altamente leucotóxico) e proteínas de membrana externa (OMPs), especialmente Omp29. A resposta de imunoglobulina G (IgG) sérica contra este patógeno foi anteriormente associada à ambas as formas de PA, porém, são escassos os estudos que avaliaram longitudinalmente a resposta sérica frente a variáveis como estas. Dessa maneira, o objetivo desse estudo foi avaliar a resposta sérica, de 27 pacientes com PA e 10 pacientes periodontalmente saudáveis, contra Omp29 e sorotipos de A. actinomycetemcomitans, através de um ensaio ELISA, correlacionando com o número de cópias de JP2 (obtidos por qPCR em tempo real) e parâmetros clínicos, a partir de dados anteriormente coletados por nosso grupo. Todos os dados foram obtidos antes do início do tratamento e um ano após seu término. O tratamento consistiu de orientações de higiene bucal, tratamento mecânico e antibioticoterapia. Os dados resultantes do estudo mostraram que em ambas as formas de PA houve uma redução significativa na profundidade clínica de sondagem (PCS)(p<0,001), nível clínico de inserção (NCI)(p<0,001) e na resposta sérica contra Omp29 e sorotipo c de A. actinomycetemcomitans(p>0,005). Após 1 ano, os valores de densidade óptica (D.O.) normalizados para Omp29 e sorotipos de A. actinomycetemcomitans, bem como o número de cópias do clone JP2 tornaram-se similares aos níveis encontrados nos controles. A redução no número de cópias do clone JP2 foi correlacionada com redução da PCS em PAL(r=0.80,p=0.0042) e valores de D.O. normalizados de Omp29 em PAG(r=0.66,p=0.005). O estudo concluiu que o tratamento periodontal foi eficaz em alterar a resposta sérica contra Omp29 e sorotipos de A. actinomycetemcomitans, além de reduzir o número de cópias do clone JP2 e melhorar os parâmetros clínicos.

ABSTRACT Objective: The aim of this double-blind, placebo-controlled and parallel- arm randomized clinical trial was to evaluate the effects of Lactobacillus rhamnosus SP1-containing probiotic sachet and azithromycin tablets as an adjunct to nonsurgical therapy in clinical parameters and in presence and levels of Tannerella forsythia, Porphyromonas gingivalis and Aggregatibacter actinomycetemcomitans. Material and Methods: Forty-seven systemically healthy volunteers with chronic periodontitis were recruited and monitored clinically and microbiologically at baseline for 3, 6 and 9 months after therapy. Subgingival plaque samples were collected from four periodontal sites with clinical attachment level ≥1 mm, probing pocket depth ≥4 mm and bleeding on probing, one site in each quadrant. Samples were cultivated and processed using the PCR technique. Patients received nonsurgical therapy including scaling and root planing (SRP) and were randomly assigned to a probiotic (n=16), antibiotic (n = 16) or placebo (n = 15) group. L. rhamnosus SP1 was taken once a day for 3 months. Azithromycin 500mg was taken once a day for 5 days. Results: All groups showed improvements in clinical and microbiological parameters at all time points evaluated. Probiotic and antibiotic groups showed greater reductions in cultivable microbiota compared with baseline. The placebo group showed greater reduction in number of subjects with P. gingivalis compared with baseline. However, there were no significant differences between groups. Conclusions: The adjunctive use of L. rhamnosus SP1 sachets and azithromycin during initial therapy resulted in similar clinical and microbiological improvements compared with the placebo group.

Objetivo: revisar quais seriam os diferentes fatores envolvidos na transmissão de periodontopatógenos entre membros de uma mesma família e quais as suas consequências. Material e métodos: uma revisão da literatura foi realizada na base de dados PubMed, utilizando os termos "vertical transmission", "periodontal pathogens", "oral colonization", e "periodontitis". Resultados: após a leitura do título e resumo, 30 artigos foram incluídos nesta revisão. A transmissão de patógenos periodontais entre indivíduos de uma mesma família está relacionada à passagem via salivar e ao compartilhamento alimentar e de higiene, aos cuidados dos filhos pelos pais ou cuidadores, e ao contato íntimo entre cônjuges. Estudos que avaliaram a transmissão do Aggregatibacter actinomycetemcomitans entre indivíduos de uma mesma família mostraram a ocorrência da transmissão vertical, embora também ocorra transmissão horizontal. Entretanto, resultados semelhantes não puderam ser observados para o Porphyromonas gingivalis. Enquanto alguns relatos indicam a ocorrência de transmissão horizontal desta bactéria, diversos outros estudos indicam características bacterianas que reduzem sua ocorrência. Conclusão: a colonização oral por microrganismos patogênicos está relacionada à transmissão vertical e horizontal de patógenos, embora a persistência dos microrganismos pareça estar relacionada a fatores individuais do hospedeiro e características dos patógenos. Além disso, atividades preventivas e terapêuticas devem ser realizadas de forma a alterar o processo de transmissão, colonização e o maior risco do desenvolvimento de problemas periodontais.


Objective: to review the different factors involved in the transmission of periodontopathogens between members of the same family and their consequences. Material and methods: an electronic literature review was conducted at the PubMed using the keywords "vertical transmission", "periodontal pathogens", "oral colonization", and "periodontitis". Results: after reading of title and abstract, 30 articles were included. The transmission of periodontal pathogens among individuals of the same family is related to the passage through salivary and food and hygiene sharing, the care of the children by parents or caregivers, and the intimate contact between individuals. Studies evaluating the transmission of Aggregatibacter actinomycetemcomitans among individuals from the same family showed the occurrence of vertical transmission and horizontal transmission. However, similar results could not be observed for Porphyromonas gingivalis. While some reports indicate the occurrence of horizontal transmission, several other studies indicate bacterial characteristics that reduce its occurrence. Conclusion: oral colonization by pathogenic microorganisms is related to its vertical and horizontal transmission, although the persistence of the microorganisms seems to be related to individual host factors and pathogen characteristics. In addition, preventive and therapeutic activities must be performed in a way that will alter the transmission, colonization and the greater risk of developing periodontal problems.

Galectina-3, uma proteína que se liga a -galactosídeos, é expressa por neutrófilos e inúmeras evidências indicam que esta molécula atua como uma possível reguladora da resposta imune. Sabe-se que galectina-3 ao ligar com LPS pode levar a formação de oligômeros, que podem alterar o limiar de ativação de células da resposta imune inata. Apesar de existirem diversos estudos que mostram a influência de galectina-3 na resposta de neutrófilos frente a componentes bacterianos, os resultados são em sua maioria contraditórios e inconclusivos. Para elucidar a influência da galectina-3 na reposta imune inata a patógenos periodontais, o presente trabalho avaliou a atividade antimicrobiana in vitro de neutrófilos, isolados de camundongos selvagens (WT) ou geneticamente deficientes de galectina-3 (Gal-3KO), previamente estimulados com LPS de Aa e Pg. Os resultados não evidenciaram diferenças significativas no número de unidades formadoras de colônia (UFC) recuperadas das culturas de neutrófilos provenientes de animais deficientes de galectina-3 e do grupo controle (WT). Contudo, a estimulação de neutrófilos com LPS por 18 horas levou a redução no número de UFC recuperadas das culturas, quando comparado com as culturas estimuladas com LPS por apenas 3 horas.


Galectin-3, a protein that binds -galactosides, is expressed by neutrophils and numerous evidences indicate that this molecule acts as a possible regulator of the immune response. It is known that galectin-3 binding to LPS can lead to the formation of oligomers and thus changing the activation threshold of cells of the innate immune response. Although there are several studies that show the influence of galectin-3 in neutrophil response against bacterial components, the results are conflicting and inconclusive in their majority. To elucidate the influence of galectin-3 in the innate immune response to periodontal pathogens, the present study evaluated the in vitro antimicrobial activity of neutrophils, isolated from wild-type or galectin-3 deficient mice, previously stimulated with LPS of Aa and Pg. The results showed no significant differences in the number of colony forming units (CFU) recovered from cultured galectin-3 deficient neutrophils or control group. However, in a 18 hours time course of LPS stimulation, we observed reduction in the number of CFU, when compared to 3 hours of LPS stimulation.

O tratamento da doença periodontal consiste na remoção mecânica do biofilme,sendo que terapias complementares como a terapia fotodinâmica antimicrobiana(aPDT) podem melhorar os resultados obtidos. Este trabalho avaliou in vitro o efeitodos corantes azul de toluidina e azul de metileno com distintos parâmetros de laser(70; 100mW) e LED em Aggregatibacter actinomycetemcomitans (A.a) emsuspensão. O primeiro experimento avaliou efeito bactericida de diferentesconcentrações dos dois corantes sozinhos (0,05; 0,1; 0,5, 1,0; 2,0; 5,0; 10 mg/ml). Osegundo foi dividido nos seguintes grupos: controles positivo e negativo(gentamicina), grupos somente com luz aplicada, grupos com três concentrações(0,05; 0,1; 10 mg/ml) de corante sozinhos ou associados ao laser de baixaintensidade (660 nm; 2,91 e 4,16 W/cm2; 70 e 100mW; 45 J/cm2; 0,024 cm2; 12 e18s) e LED (627 ± 10 nm; 150mW/cm2; 10,5 mW; 20 J/cm2; 0,07cm2; 123s). Osdados foram analisados pelo teste ANOVA complementado por Tukey (p<0,05). Osresultados demonstraram que ambos os corantes na concentração de 10 mg/mlsozinhos ou associados com laser (ambas as potências) ou LED causaram 100 %de morte bacteriana semelhante ao controle negativo (p>0,05). Com isso, pode-seconcluir que aPDT pode eliminar o A.a. de forma dependente da concentração docorante.(AU(


The main treatment of periodontal disease is the mechanical removal of biofilm.Adjuvant therapies as antimicrobial photodynamic therapy (aPDT) may offer betterresults. The aim of this in vitro study was to evaluate the effect of toluidine andmethylene blue dyes, associated to red laser (70; 100mW) and LED on elimination ofa suspension of Aggregatibacter actinomycetemcomitans (A.a). A first test evaluatedthe bactericidal effect of various concentrations of both dyes (0.05; 0.1; 0.5, 1.0; 2.0;5.0, 10 mg/ml) in absence of light. In the second test, the experimental groupsconsisted of positive and negative (gentamicin) controls, groups with threeconcentrations (0.05; 0.1,10 mg/ml) of dyes alone of associated to low level laser(660 nm; 2.91 and 4.16 W/cm2; 70 and 100mW; 45 J/cm2; 0.024 cm2; 12 and 18s)and LED (627 ± 10 nm; 150mW/cm2; 10.5 mW; 20 J/cm2; 0.07cm2; 123s). Data wereanalyzed by ANOVA complemented by Tukey’s test (p<0.05). The results showedthat both dyes, with 10 mg/ml, alone or associated to laser and LED caused 100% ofdeath as the negative control (p>0.05). It can be concluded that aPDT is capable ofeliminate A.a depending of the dye concentration.

O uso de implantes osseointegrados vem crescendo nas ultimas decadas e, juntamente com eles, suas complicacoes. A periimplantite se apresenta como uma infeccao bacteriana que afeta os tecidos moles e duros ao redor do implante, promovendo perda da osseointegracao. Assim, o objetivo primario deste estudo foi analisar a efetividade da remocao de bacterias, atraves do software ImageJ, aderidas as superficies de titanio por diferentes agentes quimicos condicionantes, por meio de analise em microscopia eletronica de varredura (MEV). Juntamente foi analisado a alteracao da rugosidade de superficie apos a utilizacao dos agentes. Foi realizado estudo in vitro, no qual 70 covers (prototipos de implantes) passaram por preparo das superficies para adequacao do meio a cultura bacteriana, fixacao das bacterias; e em seguida, foram divididos em 7 grupos (n=10), de acordo com o tratamento: AF180– aplicacao de acido fosforico (AF) por 180 segundos; AF90- AF por 90 segundos; EDTA180 – EDTA por 180 segundos; EDTA90 – EDTA por 90 segundos; AC180 – acido citrico por 180 segundos; AC90 –AC por 90 segundos; Controle – RAR. A analise comparativa do grau de contaminacao bacteriana observado antes e depois do tratamento entre os diferentes grupos foi realizada por meio do teste nao parametrico de Kruskal-Wallis; e as alteracoes da rugosidade superficial foram analisadas por meio do teste ANOVA a dois criterios, pos-teste de Dunnett. Atraves desta metodologia, este trabalho sugere que o tratamento de superficies de titanio contaminadas por meio do emprego de solucao em gel de EDTA a 24% por 90 e 180 segundos e acido citrico a 50% por 180 segundos e efetiva para remocao de A. atinomycetencomitans. Alem disso, o tratamento por meio de EDTA por 90 e 180 segundos promove alteracao significativa dos parametros de rugosidade superficial, especialmente quando comparado aos grupos controle e AF180, indicando que este tratamento pode resultar em subtracao acida adicional.


The use of dental implants has grown in recent decades and, with them, their complications. The periimplantitis is presented as a bacterial infection that affects the soft and hard tissue around the implant, promoting loss of osseointegration. Thus, the primary objective of this study was to analyze the effectiveness of removal of bacteria tby means of the ImageJ software, adhered to the titanium surfaces by different chemical conditions, through analysis in scanning electron microscopy (SEM). The change of surface roughness was also analyzed after using the chemical agents. An in vitro study in which 70 covers (implant prototypes) was prepared for the bacteria culture, fixation of the bacterias; and then they were divided into 7 groups (n = 10), according to the surface treatment: AF180- application of phosphoric acid (FA) for 180 seconds; AF90- AF for 90 seconds; EDTA180 - EDTA for 180 seconds; EDTA90 - EDTA for 90 seconds; AC180 - citric acid for 180 seconds; AC90 -AC for 90 seconds; Control - RAR. The comparative analysis of the degree of bacterial contamination was performed using Kruskal-Wallis non parametric test; and changes of surface roughness were analyzed by ANOVA two criteria, post-test Dunnett. Through this method, this work suggests that treatment of titanium contaminated surfaces by means of EDTA gel solution employing 24% for 90 and 180 seconds and citric acid 50% for 180 seconds is effective for removing A. atinomycetencomitans. Moreover, treatment using EDTA for 90 to 180 seconds promotes significant change of surface roughness parameters, especially when compared to control groups and AF180, indicating that this treatment can result in additional acidic subtraction.

O uso de implantes osseointegrados vem crescendo nas ultimas decadas e, juntamente com eles, suas complicacoes. A periimplantite se apresenta como uma infeccao bacteriana que afeta os tecidos moles e duros ao redor do implante, promovendo perda da osseointegracao. Assim, o objetivo primario deste estudo foi analisar a efetividade da remocao de bacterias, atraves do software ImageJ, aderidas as superficies de titanio por diferentes agentes quimicos condicionantes, por meio de analise em microscopia eletronica de varredura (MEV). Juntamente foi analisado a alteracao da rugosidade de superficie apos a utilizacao dos agentes. Foi realizado estudo in vitro, no qual 70 covers (prototipos de implantes) passaram por preparo das superficies para adequacao do meio a cultura bacteriana, fixacao das bacterias; e em seguida, foram divididos em 7 grupos (n=10), de acordo com o tratamento: AF180– aplicacao de acido fosforico (AF) por 180 segundos; AF90- AF por 90 segundos; EDTA180 – EDTA por 180 segundos; EDTA90 – EDTA por 90 segundos; AC180 – acido citrico por 180 segundos; AC90 –AC por 90 segundos; Controle – RAR. A analise comparativa do grau de contaminacao bacteriana observado antes e depois do tratamento entre os diferentes grupos foi realizada por meio do teste nao parametrico de Kruskal-Wallis; e as alteracoes da rugosidade superficial foram analisadas por meio do teste ANOVA a dois criterios, pos-teste de Dunnett. In Aggregatibacter actinomycetemcomitans, different serotypes have been described based on LPS antigenicity. Recently, our research group has reported a differential immunogenicity when T lymphocytes were stimulated with these different serotypes. In particular, it was demonstrated that the serotype b of A. actinomycetemcomitans has a stronger capacity to trigger Th1- and Th17-type cytokine production.Objective This study aimed to quantify the expression of different CC chemokines (CCLs) and receptors (CCRs) in T lymphocytes stimulated with the differentA. actinomycetemcomitans serotypes. In addition, the expression of the transcription factors T-bet, GATA-3, RORC2, and Foxp3, master-switch genes implied in the Th1, Th2, Th17, and T-regulatory differentiation, respectively, was analysed in order to determine T-cell phenotype-specific patterns of CCL and CCR expression upon A. actinomycetemcomitans stimulation.Material and Methods Human naïve CD4+ T lymphocytes were obtained from healthy subjects and stimulated with autologous dendritic cells primed with the differentA. actinomycetemcomitans serotypes. The expression levels for the chemokines CCL1, CCL2, CCL3, CCL5, CCL11, CCL17, CCL20, CCL21, CCL25, and CCL28, as well as the chemokine receptors CCR1, CCR2, CCR3, CCR4, CCR5, CCR6, CCR7, CCR8, CCR9, and CCR10 were quantified by qPCR. Similarly, the expression levels for the transcription factors T-bet, GATA-3, RORC2, and Foxp3 were quantified and correlated with the CCL and CCR expression levels.Results Higher expression levels of CCL2, CCL3, CCL5, CCL20, CCL21, CCL28, CCR1, CCR2, CCR5, CCR6, CCR7, and CCR9 were detected in T lymphocytes stimulated with the serotype b of A. actinomycetemcomitans compared with the other serotypes. In addition, these higher expression levels of CCLs and CCRs positively correlated with the increased levels of T-bet and RORC2 when T lymphocytes were stimulated with the serotype b.Conclusion A T-lymphocyte response biased towards a Th1- and Th17-pattern of CCL and CCR expression was detected under stimulation with the serotype b ofA. actinomycetemcomitans.

Os mastócitos (MCs) estão presentes tanto no periodonto normal quanto inflamado, em diferentes quantidades e em vários locais. Nos últimos anos, a eficácia e a contribuição dos MCs em eliminar bactérias, através de sua atividade microbicida intracelular, estão se tornando cada vez mais reconhecidas. Assim, a partir de MCs murinos desafiados in vitro com o periodontopatógeno Aggregatibacter actinomycetemcomitans (ATCC 29523) por 3, 5, 10 e 24 horas, o presente estudo teve como objetivo investigar a capacidade microbicida intracelular de MCs, e comparar com a capacidade microbicida de macrófagos peritoneais murinos (MPs), considerados fagócitos profissionais, por meio da contagem das unidades formadoras de colônias. Além disso, avaliou-se a produção e liberação de mediadores microbicidas, óxido nítrico (NO) e peróxido de hidrogênio (H2O2), por meio do método colorimétrico de Griess e pela degradação de substratos fluorescentes, respectivamente. Para a análise estatística, foram utilizados os testes estatísticos ANOVA Fatorial seguido do teste de Tukey e teste de correlação de Pearson (p<0.05). Nossos resultados revelaram que os MCs foram capazes de eliminar eficientemente o periodontopatógeno, principalmente após 10h de desafio intracelular. Comparando-se a atividade microbicida dos dois tipos celulares, verificou-se, nos períodos de 3h e 5h de desafio, um menor percentual de colônias viáveis no interior de MPs, em comparação aos MCs. Inversamente, nos períodos de 10h e 24h, observaram-se menores valores percentuais de colônias intracelulares nos MCs em relação aos MPs. Além disso, a produção/liberação de NO bem como, em menor proporção, de H2O2 pelos MCs foram concordantes com a sua capacidade microbicida. Este é o primeiro estudo que demonstra a eficiente ação microbicida intracelular de MCs murinos contra Aggregatibacter actinomycetemcomitans, com produção e liberação de substâncias potencialmente bactericidas, e de forma mais eficaz que os macrófagos...


Mast cells (MCs) are present in both normal and inflamed periodontal tissues, in varying amounts and locations. Recently, MCs contribution in eliminating bacteria and its effectiveness, through its intracellular microbicidal activity, have been increasingly recognized. Thus, this study aimed to investigate the intracellular microbicide capacity of MCs, and compare it with the microbicide capacity of murine peritoneal macrophages (MPs), considered professional phagocytes, by counting the colony forming units. Both cell types were challenged in vitro with periodontopathogen Aggregatibacter actinomycetemcomitans (ATCC 29523) by 3, 5, 10 and 24 hours. Additionally, the production and release of microbicidal agents, nitric oxide (NO) and hydrogen peroxide (H2O2) were evaluated by means of colorimetric Griess method and by the degradation of fluorescent substrates, respectively. Statistical analysis was performed by ANOVA Factorial test followed by Tukey and Pearson's correlation test (p <0.05). Our results revealed that MCs are able to efficiently eliminate periodontopathogen, mainly after 10 hours of intracellular challenge. The microbicidal activity of both cell types, in 3 and 5 hours of challenge showed a lower percentage of viable colonies inside MPs, compared to MCs. Contradictorily, in 10 and 24 hours a lower percentage of intracellular colonies in MCs was observed in relation to MPs. Moreover, the production/release of NO and, in minor proportion, of H2O2 by MCs was in agreement with its microbicidal capacity. Therefore, this is the first report to describe the intracellular microicidal activity of murine MCs against Aggregatibacter actinomycetemcomitans, concerning production and release of potentially bactericidal substances, which is more effective than macrophages. These results suggest the importance of these cells in pathogenesis and defense mechanisms of biofilm-associated periodontal disease.

Objectivo In this study, the gingival conditions and the quantitative detection for Aggregatibacter actinomycetemcomitans, Fusobacterium nucleatum, Porphyromonas gingivalis and Prevotella intermedia in pregnant women were determined. Material and Methods Quantitative determinations of periodontal bacteria by using a SyBr green system in women during pregnancy were performed. Women at the 2nd and 3rd trimesters of pregnancy and non-pregnant women were included in this study. A. actinomycetemcomitans was observed in high numbers in women at the 2nd and 3rd trimesters of pregnancy with a significant difference (p<0.05). F. nucleatum and P. intermedia were also observed in high levels. Results and Conclusion Our results show that pregnant women are more susceptible to gingivitis, and the presence of A. actinomycetemcomitans in subgingival biofilm might be taken into account for the treatment of periodontal disease. .

OBJECTIVE: The purpose of this study was to examine the leukotoxin promoter types of Aggregatibacter actinomycetemcomitans clones in subjects with generalized aggressive periodontitis (GAgP) and in their family members (FM). MATERIAL AND METHODS: Thirty-five patients with GAgP (33.9±7.1 years), 33 of their FM (22.8±11.4 years), and 41 patients with chronic periodontitis (CP) (44.1±9.4 years) were clinically analyzed using the plaque index, gingival index, probing depth (PD), and clinical attachment level (CAL). Subgingival biofilm samples were collected from four interproximal periodontal sites (>PD and >CAL) of each patient. The presence of A. actinomycetemcomitans and its leukotoxic clone was confirmed by polymerase chain reaction (PCR). RESULTS: A. actinomycetemcomitans was observed in 23 (51.1%) GAgP patients and 16 (30.1%) CP patients. Thirty-seven (94.8%) patients showed minimally leukotoxic strains and 2 (5.1%) showed highly leukotoxic strains. In the FM group, 10 (30.3%) had aggressive periodontitis (AgP), 12 (36.3%) had CP, 11 (33.3%) were periodontally healthy or had gingivitis, and 12.2% were A. actinomycetemcomitans positive. Greater full mouth PD and CAL were observed in GAgP patients positive for the bacteria than those negative for it (p<;0.05), and the presence of A. actinomycetemcomitans positively correlated with GAgP (Odds ratio, 3.1; confidence interval, 1.4-7.0; p=0.009). CONCLUSIONS: The presence of A. actinomycetemcomitans was associated with the clinical condition of GAgP, with most patients exhibiting a generalized form of the disease and minimally leukotoxic clones. Most of the relatives of GAgP patients presented either CP or AgP. .

Aggregatibacter actinomycetemcomitans (Aa) é uma bactéria associada à Periodontite Agressiva (PA). Ela invade tecidos moles, com ocorrência de lisogenia e bacteriófagos presentes em até 69% das subespécies. Estudos in vitro sugerem que a indução do bacteriófago (Aa17) ocorre numa co-cultura de Aa lisogênico com fibroblastos humanos. Se esta interação ocorre in vivo, com liberação do vírus, uma reação imunológica contra o Aa17 aconteceria. O objetivo deste estudo é constatar se anticorpos (AC) contra proteínas do Aa17 existem e estão associados à doença periodontal. Um objetivo adicional foi testar a resposta de AC contra os sorotipos do Aa. 52 indivíduos participaram: 31 com PA, 5 com Periodontite Crônica (PC) e 16 com Periodonto Saudável (PS). Soro foi coletado após a classificação clínica. As proteínas do Aa17 foram obtidas de preparações purificadas. As subespécies do Aa utilizadas para amostras de proteínas através de sonicação foram: 43717(American Tissue Culture Collection - ATCC) – sorotipo A, 43718 (ATCC) – sorotipo B, 33384 (ATCC) – sorotipo C, IDH781 – sorotipo D, NJ9500 – sorotipo E and CU1000 – sorotipo F. As proteínas foram separadas em géis de poliacrilamida e transferidas para membranas de nitrocelulose. As reações de Western-blotting ocorreram com o AC primário sendo o soro de cada indivíduo. Todas as membranas foram lidas pelo sistema Odyssey que captura sinais no AC secundário (antihumano). A resposta de AC contra ao menos uma proteína do Aa17, assim como pelo menos um sorotipo do Aa foi observado em todos, com exceção de dois indivíduos (com PS), participantes. Um indivíduo do grupo PC e três do PA tiveram resposta de AC contra alguns, mas não todos os sorotipos do Aa. A resposta de AC contra todos os sorotipos foi o achado mais comum nos grupos PA (28/31), PS (14/16) e PC (4/5). Aggregatibacter actinomycetemcomitans (Aa) is a bacteria associated with Aggressive Periodontitis (AP). It invades soft tissues, with occurrence of lysogeny and bacteriophage presence up to 69% of Aa subspecies. In vitro studies suggested that bacteriophage (Aa17) induction occurs upon co-culture of Aa lysogens subspecies with human fibroblasts. If such an in vivo interaction resulted in Aa17 induction and release of virions, an immunologic reaction to Aa17 proteins could ensue. The purpose of this investigation was to learn whether serum antibodies (AB) to Aa17 proteins are found in human sera, and whether they are associated with periodontal disease. An additional purpose was to test the AB response against known Aa serotypes.52 individuals took part: 31 with AP, 5 with Chronic Periodontitis (CP) and 16 with a Healthy Periodontium (HP). Serum was collected after clinical classification. Aa17 proteins were obtained from purified Aa17 preparations. The Aa strains used for protein sampling through sonication were: 43717(American Tissue Culture Collection - ATCC) – serotype A, 43718 (ATCC) – serotype B, 33384 (ATCC) – serotype C, IDH781 – serotype D, NJ9500 – serotype E and CU1000 – serotype F. Proteins were separated by SDS-PAGE gels and then transferred to nitrocellulose membranes. Western-blotting reactions were carried out with the primary AB being each subject’s serum. All membranes were read through the Odyssey system which captures signals from a dye in a secondary (antihuman) AB. AB response against at least one Aa17 protein, as well as a response to at least one Aa serotype, was observed in all but two individuals (with HP) who participated in the study. Serum from one individual from the CP group and three from the AP group had AB response to some, but not all Aa serotypes. Introdução: O conhecimento das inter-relações bacterianas e sua associação com a condição clínica periodontal podem auxiliar o manejo terapêutico. Objetivos: Avaliar a distribuição de Aggregatibacter actinomycetemcomitans em relação a duas espécies bacterianas membros dos complexos Vermelho e Laranja, e analisar a relação entre a combinação de cepas leucotóxicas de A.actinomycetemcomitans, Porphyromonas gingivalis e Prevotella intermedia com a manifestação clínica da doença. Material e método: Incluíram-se 50 indivíduos (38,38 ± 10,15 anos), de ambos os gêneros, com periodontite crônica (n = 25) ou agressiva (n = 25). De cada indivíduo, amostras subgengivais foram coletadas de quatro dentes em quadrantes diferentes, com profundidade de sondagem (PS) maior ou igual a 6 mm e nível clínico de inserção (NCI) maior ou igual a 4 mm. Os parâmetros avaliados foram: clínicos - PS, NCI, Índice de Placa (IP-0/1) e Índice Gengival (IG-0/1) - e microbiológicos, com detecção de A. actinomycetemcomitans (cepas de máxima e mínima leucotoxicidades), P. gingivalis e P. intermedia por reação em cadeia da polimerase. Os dados foram estatisticamente analisados usando-se testes t-Student e Qui-quadrado (p <0,05). Resultado: Apresentaram P. gingivalis 90% dos indivíduos, P. intermedia, 82%, e A. actinomycetemcomitans, 28%. A. actinomycetemcomitans foi mais frequente em indivíduos jovens ou com periodontite agressiva. Apenas IG apresentou maiores frequências médias (p = 0,019) quando da presença simultânea de cepas A. actinomycetemcomitans de máxima leucotoxicidade, P. gingivalis e P. intermedia. Conclusão: Indivíduos com periodontite agressiva exibiram maiores frequências de A. actinomycetemcomitans. A presença de P. gingivalis e/ou P. intermedia não potencializou a patogenicidade das cepas de A. actinomycetemcomitans de máxima leucotoxicidade, pois tal combinação foi somente acompanhada de maior inflamação.

Objective: To evaluate the distribution of Aggregatibacter actinomycetemcomitans in relation to 2 bacterial species members of Red and Orange Complexes; also to exam the relationship between the combined presence of A. actinomycetemcomitans, Porphyromonas gingivalis and Prevotella intermedia with clinical pattern of disease. For each subject subgingival samples were collected from 4 teeth in different quadrants showing probing depth (PD) is greater than or equal to 6 mm and clinical attachment level (CAL) is greater than or equal to 4 mm. The parameters evaluated were: clinical - PD, CAL, Plaque Index (PI-0/1) and Gingival Index (GI-0/1); microbiological - detection of A. actinomycetemcomitans (highly and minimally leukotoxic strains), P. gingivalis and P. intermedia by polymerase chain reaction. Data were statistically analyzed using Student t and Chi-square tests (p <0.05). Result: 90% of the subjects harbored P. gingivalis, 82% P. intermedia and 28% A. actinomycetemcomitans. A. actinomycetemcomitans was most frequent in young or aggressive periodontitis subjects. Only GI showed higher mean frequency (p = 0.019) in the combined presence of highly leukotoxic A. actinomycetemcomitans strains,P. gingivalis and P. intermedia. Conclusion: Subjects with aggressive periodontitis exhibited higher frequency of A. actinomycetemcomitans. The presence of P. gingivalis and/or P. intermedia did not improve the pathogenicity of highly leukotoxic A. actinomycetemcomitans strains due to the fact that this combination was only followed by increased inflammation.

As doenças periodontais afetam os tecidos de suporte dos dentes e são desencadeadas por microrganismos que possuem a capacidade de invadir os tecidos periodontais. A evolução desta doença é influenciada pela resposta inflamatória e imunológica do hospedeiro e envolve a participação de diversos tipos celulares. Atualmente, existem evidências de que os mastócitos, além de outras funções, possuem a capacidade de eliminar bactérias, através da fagocitose. Assim sendo, este estudo teve por objetivo avaliar a capacidade fagocítica dos mastócitos frente ao periodontopatógeno A. actinomycetemcomitans, além de comparar sua capacidade fagocítica com a dos macrófagos, considerados fagócitos profissionais. Para este fim, foram realizados ensaios fagocíticos in vitro utilizando mastócitos e macrófagos murinos, desafiados ora com A. actinomycetemcomitans ora com Escherichia coli, opsonizados ou não, sob diferentes proporções célula: bactérias. Após 1 hora de desafio, as células foram coradas com laranja de acridina e avaliadas qualitativamente utilizando-se microscópio de varredura confocal a laser e quantitativamente através do microscópio de fluorescência convencional. Nossos resultados demonstraram que os mastócitos murinos se mostraram eficientes quanto a sua capacidade fagocítica frente a A. actinomycetemcomitans. Os valores percentuais de mastócitos com A. actinomycetemcomitans internalizados, na ausência de opsonização com complemento, foram maiores que aqueles na presença da opsonização, sugerindo a participação de receptores opsoninas-independentes no reconhecimento deste patógeno pelos mastócitos, além do receptor de complemento tipo 3 (CR3). Periodontal diseases affect the supporting tissues of the teeth and are triggered by microorganisms which are capable of invading periodontal tissues. The evolution of this disease is influenced by inflammatory and immune response of the host and involves the participation of different cell types. Currently, there is evidence that mast cells, among other functions, have the ability to eliminate bacteria by phagocytosis. Thus, this study aimed to evaluate the phagocytic ability of mast cells against the periodontopathogens A. actinomycetemcomitans, and compare with the phagocytic capacity of macrophages, which are considered professional phagocytes. Therefore, in vitro phagocytic assays were conducted using murine mast cells and macrophages, challenged with A. actinomycetemcomitans or Escherichia coli, at the same time, opsonized or not, under different proportions cell: bacteria. After 1 hour of challenge, cells were stained with acridine orange and qualitatively assessed by using a confocal laser scanning electron microscope and quantitatively by the conventional scanning fluorescence microscope. The results demonstrated that phagocytic ability of murine mast cells was effective against A. actinomycetemcomitans. The percentages of mast cells with A. actinomycetemcomitans internalized in the absence of opsonization with complement, were higher than those in the presence of opsonization, suggesting the involvement of opsonin-independent receptors in recognition of this pathogen by mast cells, as well as complement receptor type 3 (CR3). OBJETIVE: This preliminary study aimed to evaluate clinically and microbiologically the treatment of patients with light to moderate periodontal disease by using an ultrasonic device. MATERIAL AND METHODS: Sixteen adults with moderate periodontitis were studied. A Real-time PCR protocol was optimised using specific primers for the three microorganisms. Clinical and microbiological testing was performed prior and following treatment. RESULTS: Despite clinical improvement, bacteria quantification didn’t change. All the patients were positive for P. gingivalis and P. intermedia and nine for A. actinomycetemcomitans. CONCLUSION: VectorTM-system treatment showed clinical improvement. A Real-time PCR protocol is now available for periodontal diagnostics and monitorization. However, further studies with larger populations are indicated.


OBJETIVOS: Este estudo preliminar teve a finalidade de avaliar clinicamente microbiologicamente o tratamento de pacientes portadores de periodontite leve a moderada, utilizando-se um novo aparelho piezo-elétrico disponível no mercado (VectorTM system). MATERIAL E MÉTODO: Dezesseis pacientes adultos, portadores de periodontite leve a moderada, foram submetidos ao tratamento. Utilizou-se um protocolo de PCR em tempo real utilizando primers específicos para três microrganismos. Realizaram-se avaliação clínica e um estudo microbiológico simultâneo antes e após os tratamentos. RESULTADOS: A avaliação clínica foi positiva, embora não tendo ocorrido alterações na quantidade de bactérias. Todos os pacientes apresentaram amplificação específica para P. intermedia e P. gingivalis e nove pacientes para A. Actinomycetemcomitans. CONCLUSÃO: Considerando-se os resultados deste estudo preliminar, tratamento com o aparelho testado mostrou eficácia clínica. O protocolo utilizado neste estudo está agora disponível para diagnóstico periodontal e monitoração de resultados de tratamento. Entretanto, indica-se estudos clínicos e microbiológicos mais aprofundados, com amostras maiores, para conclusões mais consistentes.

O desenvolvimento da doença periodontal (DP) é influenciado pela resposta imunológica do hospedeiro frente ao desafio bacteriano. Apesar da função protetora de tal resposta frente à infecção, a mesma leva à destruição dos tecidos periodontais. Neste estudo, analisamos o papel de MIP-1a na imunomodulação da DP experimental em camundongos. Camundongos C57Bl/6 (WT) infectados com A. actinomycetemcomitans desenvolveram uma intensa reação inflamatória e severa reabsorção óssea alveolar, associada a uma alta expressão de MIP-1a e uma intensa migração de células CCR5+ e CCR1+ para os tecidos periodontais. Além disso, verificamos que uma intensa expressão de citocinas inflamatórias (TNF-'alfa') e do tipo Th1 (IFN-'gama'), RANKL e MMPs estava associada à progressão da doença. Entretanto, camundongos geneticamente modificados para não expressar MIP-1'alfa'(MIP-1'alfa'KO), quando infectados com A. actinomycetemcomitans, apresentaram um padrão de resposta muito semelhante aos animais WT. De fato, a ausência de MIP-1'alfa' não interferiu na migração de células inflamatórias para os tecidos periodontais e na reabsorção óssea alveolar nos camundongos infectados. Semelhantemente, a expressão de citocinas (TNF-'alfa', IFN-'gama' e IL-10), dos fatores osteoclastogênicos (RANKL e OPG) e das MMPs e seus inibidores (MMP-1, MMP-2, MMP-3, TIMP-1 e TIMP-3) foram similares entre os camundogos das linhagens WT e MIP-1'alfa'KO.


Além disso, a ausência de MIP-1a não interferiu no controle da infecção por A. actinomycetemcomitans como demonstrado pela similaridade na quantidade de carga bacteriana presente nos tecidos e nos níveis de MPO e iNOS. Os resultados obtidos podem ser explicados pela relativa redundância no sistema quimiocinas/receptores de quimiocinas, onde mais de uma quimiocina pode ligar-se ao mesmo receptor. De fato quimiocinas como MIP-1ß e RANTES, que utilizam os mesmos receptores de MIP-1'alfa' (CCR5 e CCR1), presentaram-se intensamente expressas nos tecidos periodontais dos camundongos infectados com A. actinomycetemcomitans, independentemente da ausência de MIP-1'alfa'. De fato, camundongos tratados com Met-RANTES, um antagonista específico dos CCR5 e CCR1, resultou em uma redução significativa no influxo de células inflamatórias e perda óssea alveolar quando comparados com os camundongos não tratados. Nossos resultados demonstraram que a ausência de MIP-1'alfa' não afeta o desenvolvimento da doença periodontal experimental em camundongos, provavelmente pela presença de quimiocinas homólogas que suprem sua ausência.


Periodontal disease (PD) development is highly influenced by the host immune response to the bacterial challenge. Despite the protective role of this response against infectious agents it leads to periodontal tissues destruction. In this study, we analyzed the role of chemokine MIP-1'alfa' on the immunomodulation of experimental PD in mice. C57Bl/6 (WT) mice infected with A. actinomycetemcomitans developed an intense inflammatory reaction and severe alveolar bone resorption, associated with a high expression of MIP-1'alfa' and the migration of CCR5+ and CCR1+ cells to the periodontal tissues. In addition, an intense expression of Th1 (IFN-'gama') and inflammatory (TNF-'alfa') cytokines, RANKL and MMPs are associated with the disease progression. However, mice genetically deficient of MIP-1'alfa' (MIP-1'alfa'KO), when infected with A. actinomycetemcomitans, developed a very similar response pattern to that observed for WT strain. Indeed, the absence of MIP-1'alfa' does not interfere in inflammatory cells migration to periodontal tissues and alveolar bone resorption in response to A. actinomycetemcomitans infection. Similarly, the expression of cytokines (TNF- 'alfa', IFN-'gama' and IL-10), osteoclastogenic factors (RANKL and OPG) and MMPs and its inhibitors (MMP-1, MMP-2, MMP-3, TIMP-1 and TIMP-3) was similar between WT and MIP-1'alfa'KO strains.


Furthermore, the lack of MIP-1'alfa' does not interfere in the control of A. actinomycetemcomitans infection as demonstrated by the similar bacterial load, and similar levels of the antimicrobial MPO and iNOS. Such results can be explained by the relative redundancy of chemokine system, where more than one chemokine can bind to the same receptor. Indeed, chemokines, such as MIP-1ß and RANTES, which use the same receptors used by MIP-1a (CCR1 and CCR5), are intensely expressed in periodontal tissues of mice infected with A. actinomycetemcomitans, regardless of the absence of MIP-1'alfa'. In accordance, the treatment of mice with Met-RANTES, a specific antagonist of the receptors CCR5 and CCR1, resulted in a significant reduction in the influx of inflammatory cells and alveolar bone loss when compared with untreated mice. Our results demonstrate that the absence of MIP-1a does not affect the development of experimental periodontitis in mice, probably due to the presence of homologous chemokines that overcome the absence of this chemokine.

As doenças periodontais (DP) afetam os tecidos de suporte dos dentes e são desencadeadas por micro-organismos gram-negativos anaeróbios presentes no biofilme periodontal. A evolução da doença é influenciada pela resposta inflamatória e imunológica do hospedeiro e envolve a participação de diversos tipos celulares, que atuam no micro ambiente local modulando a resposta do hospedeiro em busca do controle da infecção. Acredita-se que citocinas inflamatórias, quimiocinas e seus receptores estão envolvidos na migração celular para os tecidos periodontais, contudo, pouco se sabe sobre os mecanismos de determinação de resistência ou susceptibilidade às DP; ou no desencadeamento do dano tecidual decorrente da resposta. Neste projeto, avaliou-se o papel das células CCR5+ na DP experimental induzida pela inoculação oral de Aggregatibacter actinomycetemcomitans em camundongos C57BL/6 wild type e camundongos CCR5-knockout. Os resultados mostram que a maioria das células CCR5+ possuem fenótipo compatível com células T do subtipo Th1, devido a co-expressão de CD3 e CXCR3; além de co-expressarem RANKL. Na ausência das células CCR5+, houve uma significativa diminuição da migração de células inflamatórias totais e RANKL+ para os tecidos periodontais, diminuição da reabsorção óssea alveolar, diminuição dos níveis de expressão de citocinas pró-inflamatórias TNFα-, IL-1β e IFN-γ, assim como diminuição na expressão de MMP-1, MMP-2 e MMP-13. Sua ausência não interferiu no controle da infecção periodontal apesar da diminuição dos níveis de iNOS.


Estes resultados conduzem à conclusão de que a maioria das células CCR5+ são células T do subtipo Th1, que atuam como importantes moduladoras das citocinas TNFα-, IL-1β e IFN-γ, das metaloproteinases de matriz MMP-1, MMP-2 e MMP-13, e que também expressam e modulam a expressão de RANKL, tendo participação importante na imunopatogenese da DP experimental, sem interferir no controle da infecção periodontal. Estes fatos tornam as células CCR5+ potenciais alvos para intervenção terapêutica visando ao controle das doenças periodontais.


The periodontal diseases (PD) affect the supportive tissues of the teeth and are triggered by periodontopathogens present in the dental biofilm. The clinical outcome is highly influenced by the host inflammatory and immune response with participation of many cellular types, that act in the local microenvironment modulating the host response to control the infection. Inflammatory cytokines, chemokines and its receptors are thought to be involved in the cellular migration to the periodontal tissues, but there is little knowledge about the mechanisms of determination of resistance or susceptibility to the PD and in the triggering of tissue damage by immune response components. This study evaluated the role of CCR5+ cells in the experimental PD induced by oral inoculation of Aggregatibacter actinomycetemcomitans in C57BL/6 wild type mice and CCR5-knockout mice. The phenotypic analysis of inflammatory infiltrate demonstrated that the most of CCR5+ cells coexpress CD3 and CXCR3, suggesting a phenotype compatible with Th1-type cells, and also co-express RANKL. In the absence of CCR5+ cells there was a significant overall reduction of inflammatory cells and RANKL+ cells influx to the periodontal tissues, reduction in the alveolar bone resorption, reduction in the levels of pro-inflammatory cytokines TNFα-, IL-1β and IFN-γ expression, as a reduction in the expression of MMP-1, MMP-2 and MMP-13.


The absence of CCR5+ cells did not impair the control of periodontal infection, despite the reduction of iNOS levels. In conclusion, these data demonstrate that the most of CCR5+ cells are Th1 cells, which act as important modulators of TNFα-, IL-1β and IFN-γ, MMP-1, MMP- 2 and MMP-13 levels, and which also express and modulate the expression of RANKL, playing an important role in the immunopathogenesis of experimental PD, without impairing the control of periodontal infection. These facts point to CCR5+ cells as potentials targets to therapeutic interventions aimed to control periodontal diseases.
