Abstract The aim of this study was to evaluate the effect of acute sepsis in the periodontal ligament, alveolar and furcation bone in absence of periodontitis induction through histological and immunohistochemical analyses. A septic rat model was established by cecal ligation and puncture (CLP). Twelve rats were randomly divided into CLP (n=6) and Sham (n=6) groups. The animals were euthanized at 24 h and hemimandibles were submitted to histomorfometric (bone matrix, collagenous fibers, fibroblasts, osteocytes, inflammatory cells, and blood vessels) and immunohistochemical (BMP-2/4, RANKL and osteocalcin) evaluation in alveolar bone, furcation bone and periodontal ligament. Our results demonstrated that histomorphometric parameters were similar in alveolar bone, furcation bone and periodontal ligament of Sham and CLP rats. Regarding to immunohistochemical analyses, the number of BMP-2/4 and RANKL immunolabeled cells was also similar in both groups. Furthermore, it was detected a reduction in the osteocalcin immunolabeled cells in periodontal ligaments of CLP compared to Sham rats (p=0.0014). In conclusion, the acute sepsis induction resulted in reduced number of osteocalcin labelled cells in periodontal ligament region. Moreover, no significant histological differences were observed in the periodontium of rats under acute sepsis. Considering the role of osteocalcin in bone remodeling, the study contributes to revealing the importance of careful periodontal evaluation in the presence of sepsis.


Resumo O objetivo deste estudo foi avaliar o efeito da sepse aguda no ligamento periodontal, osso alveolar e osso da furca por meio de análise histológica e imunohistoquímica. O modelo de sepse em ratos foi estabelecido pelo procedimento de ligação e perfuração do ceco (CLP). Doze ratos foram divididos de forma randomizada em ratos sépticos (n=6) e controle - grupo Sham (n=6). Os animais foram eutanasiados após 24 horas e suas hemimandíbulas foram submetidas aos procedimentos histotécnicos para análise histomorfométricos (matriz óssea, fibras colágenas, fibroblastos, osteócitos, células inflamatórias e vasos sanguíneos) e imunohistoquímicos (BMP-2/4, RANKL e osteocalcina) no osso alveolar, osso de furca e ligamento periodontal. Nossos resultados demonstraram que os parâmetros histomorfométricos foram similares no osso alveolar, osso de furca e ligamento periodontal dos animais do grupo sepse e do grupo Sham. Em relação à análise por imunohistoquímica, o número de células imunomarcadas para BMP-2/4 e RANKL também foi similar em ambos os grupos. Entretanto, houve redução (p=0.0014) no número de células imunomarcadas para osteocalcina no ligamento periodontal de ratos sépticos em relação ao grupo Sham. Como conclusão, o estabelecimento de sepse aguda resultou em um número reduzido de células imunomarcadas para osteocalcina na região do ligamento periodontal (p=0,0014). Além disso, não foram observadas diferenças histológicas significativas no periodonto de ratos na presença de sepse aguda. Considerando o papel da osteocalcina na remodelação óssea, este estudo contribui para revelar a importância da avaliação periodontal cuidadosa na presença de sepse.

Abstract Objective Ameloblastoma is a representative odontogenic tumor comprising several characteristic invasive forms, and its pathophysiology has not been sufficiently elucidated. A stable animal experimental model using immortalized cell lines is crucial to explain the factors causing differences among the subtypes of ameloblastoma, but this model has not yet been disclosed. In this study, a novel animal experimental model has been established, using immortalized human ameloblastoma-derived cell lines. Methodology Ameloblastoma cells suspended in Matrigel were subcutaneously transplanted into the heads of immunodeficient mice. Two immortalized human ameloblastoma cell lines were used: AM-1 cells derived from the plexiform type and AM-3 cells derived from the follicular type. The tissues were evaluated histologically 30, 60, and 90 days after transplantation. Results Tumor masses formed in all transplanted mice. In addition, the tumors formed in each group transplanted with different ameloblastoma cells were histologically distinct: the tumors in the group transplanted with AM-1 cells were similar to the plexiform type, and those in the group transplanted with AM-3-cells were similar to the follicular type. Conclusions A novel, stable animal experimental model of ameloblastoma was established using two cell lines derived from different subtypes of the tumor. This model can help clarify its pathophysiology and hasten the development of new ameloblastoma treatment strategies.

Abstract Objective In this study, we aimed to histologically and immunologically evaluate the effect of diode laser treatment when applied adjunctive to scaling and root planing (SRP) in an experimental periodontitis model. Materials and methods We used Wistar-Albino rats (n=60) with average weight of 230 g. Experimental periodontitis was induced by ligature at the right and left first mandibular molar teeth in all rats. After 11 days, the ligature was removed and rats were divided into two groups. The control group (n=30) received only SRP treatment, while the laser group (n=30) received a diode laser (GaAlAs, 810 nm, 1 W, 10 J, 20 s) treatment adjunctive to SRP. Ten rats in each group were sacrificed after 7, 15, and 30 days. Histopathological examination was performed in the left mandible of rats. Myeloperoxidase (MPO) was evaluated by western blot in the gingival specimens from the right mandible. Results MPO levels in the laser group were statistically significantly lower compared with the control group (p≤0.05). There was no statistically significance at any time between MPO levels in the control group (p>0.05). MPO levels in the laser group at the 7th day were statistically significantly higher compared to the 15th (p≤0.05) and the 30th day (p≤0.05). Inflammatory cell infiltration decreased over time in both groups and was statistically significantly lower in the laser group than in the control group at all times (p≤0.01). Conclusions Within the limits of this study, we suggest that diode laser application is an adjunctive treatment because it reduced inflammation and MPO when applied in addition to SRP. On the other hand, more studies are needed for the assessment of the effects of diode laser application to periodontal tissues.

Abstract Objectives To evaluate the association between hypoxia during embryo development and oral clefts in an animal model, and to evaluate the association between polymorphisms in the HIF-1A gene with oral clefts in human families. Material and Methods The study with the animal model used zebrafish embryos at 8 hours post-fertilization submitted to 30% and 50% hypoxia for 24 hours. At 5 days post-fertilization, the larvae were fixed. The cartilage structures were stained to evaluate craniofacial phenotypes. The family-based association study included 148 Brazilian nuclear families with oral clefts. The association between the genetic polymorphisms rs2301113 and rs2057482 in HIF-1A with oral clefts was tested. We used real time PCR genotyping approach. ANOVA with Tukey's post-test was used to compare means. The transmission/disequilibrium test was used to analyze the distortion of the inheritance of alleles from parents to their affected offspring. Results For the hypoxic animal model, the anterior portion of the ethmoid plate presented a gap in the anterior edge, forming a cleft. The hypoxia level was associated with the severity of the phenotype (p<0.0001). For the families, there was no under-transmitted allele among the affected progeny (p>0.05). Conclusion Hypoxia is involved in the oral cleft etiology, however, polymorphisms in HIF-1A are not associated with oral clefts in humans.

ABSTRACT Among the many graft materials that have been used for the treatment of bone defects in oral and maxillofacial regions is xenograft. To improve osteoconductive effects of xenografts, they have been combined with various biocompatible materials, such as hyaluronic acid and bone morphogenetic protein. Objective: To determine bone-healing capacity of high molecular weight hyaluronic acid (HA) combined with xenograft in rabbit calvarial bone defects. Material and methods: Ten adult male New Zealand rabbits (mean weight 3 kg) were included in the study. Three 6-mm-diameter bicortical cranial defects were created on calvarial bone of all rabbits. These defects were filled as follows: a) xenograft; b) HA+xenograft; c) autograft. One month after the first operation, rabbits were sacrificed. Specimens were evaluated histomorphometrically. Results: Considering multiple comparisons, differences regarding new bone were statistically significant between all groups (p<0.05). The volume of residual graft was significantly decreased in HA group compared to xenograft group (p=0.035). Marrow space, trabecular thickness (TbTh), trabecular width (TbWi), trabecular separation (TbSp), and number of node: number of terminus (NNd:NTm) in the autograft group were significantly better than xenograft and HA groups (p<0.05). However, regarding marrow space, TbTh, TbWi, TbSp, and NNd:NTm values, xenograft and HA groups showed similar results and the difference were not significant (p>0.05). Conclusion: These results support that high molecular weight hyaluronic acid could contribute to the healing of xenograft by improving the percentage of new bone formation and reducing the percentage of residual graft. However, HA did not significantly affect the quality of newly formed bone assessed by microarchitectural parameters.

Abstract Local administration of toll-like receptor 9 (TLR9), agonist cytidine-phosphate-guanosine oligodeoxynucleotide (CpG ODNs), and CD40 ligand (CD40L) can decrease ligature-induced periodontal inflammation and bone loss in wild type (WT) mouse. Objective: This study aimed to explore whether such effect is dependent on TLR9 signaling. Material and Methods: Purified spleen B cells isolated from WT C57BL/6J mice and TLR9 knockout (KO) mice were cultured for 48 hours under the following conditions: CD40L, CpG+CD40L, CpG at low, medium and high doses. We determined B cell numbers using a hemocytometer at 24 h and 48 h. Percentages of CD1dhiCD5+ B cells were detected by flow cytometry. Interleukin-10 (IL-10) mRNA expression and protein secretion were measured by quantitative real-time polymerase chain reaction (qRT-PCR) and by ELISA, respectively. The silk ligature was tied around the maxillary second molars for 14 days, during which the CpG+CD40L mixture or PBS was injected into palatal gingiva on days 3, 6, and 9. Results: For both WT and TLR9 KO mice, CpG significantly induced B cell proliferation, increased IL-10 mRNA expression and protein secretion of IL-10 but reduced CD1dhiCD5+ B cells population; local injection of CpG+CD40L mixture significantly decreased alveolar bone loss and the number of TRAP-positive cells adjacent to the alveolar bone surface, and significantly increased the gingival mRNA expression of IL-10 and decreased RANKL and IFN-γ mRNA expression. Conclusions: These results indicated that CpG plus CD40L decreased periodontal inflammation and alveolar bone loss in a TLR9-independent manner in ligature-induced experimental periodontitis.

Abstract Raloxifene is an antiresorptive drug, selective estrogen receptor modulator (SERM) used in the treatment of osteoporosis. Objective To evaluate proteins related to bone repair at the peri-implant bone in a rat model of osteoporosis treated with raloxifene. Material and Methods 72 rats were divided into three groups: SHAM (healthy animals), OVX (ovariectomized animals), and RLX (ovariectomized animals treated with raloxifene). Raloxifene was administered by gavage (1 mg/kg/day). Tibial implantation was performed 30 days after ovariectomy, and animals were euthanized at 14, 42, and 60 days postoperatively. Samples were collected and analyzed by immunohistochemical reactions, molecular analysis, and microtomographic parameters. Results RLX showed intense staining of all investigated proteins at both time points except for RUNX2. These results were similar to SHAM and opposite to OVX, showing mild staining. The PCR gene expression of OC and ALP values for RLX (P<0.05) followed by SHAM and OVX groups. For BSP data, the highest expression was observed in the RLX groups and the lowest expression was observed in the OVX groups (P<0.05). For RUNX2 data, RLX and SHAM groups showed greater values compared to OVX (P<0.05). At 60 days postoperatively, microtomography parameters, related to closed porosity, showed higher values for (Po.N), (Po.V), and (Po) in RLX and SHAM groups, whereas OVX groups showed lower results (P<0.05); (BV) values (P=0.009); regarding total porosity (Po.tot), RLX group had statistically significant lower values than OVX and SHAM groups (P=0.009). Regarding the open porosity (Po.V and Po), the SHAM group presented the highest values, followed by OVX and RLX groups (P<0.05). The Structural Model Index (SMI), RLX group showed a value closer to zero than SHAM group (P<0.05). Conclusions Raloxifene had a positive effect on the expression of osteoblastogenesis/mineralization-related proteins and on micro-CT parameters related to peri-implant bone healing.

Abstract Periodontal research involves the use of animal models to better understand the biological processes of periodontal diseases and the potential of new or existing therapies. Currently, ligature-induced periodontitis in rats is the main model used in periodontal research, in this model, alveolar bone loss (ABL) is the main parameter evaluated by radiographic, morphometric, and histological techniques. Interestingly, although these methodologies are widely used, it is not totally clarified neither the kinetics of ABL over the induction time nor the agreement degree (repeatability and reproducibility) of these techniques. Objective: To characterize ABL kinetics at 0, 3, 7, 15, 30, and 60 days after ABL induction by ligature and to evaluate the intra- (repeatability) and inter-examiner (reproducibility) agreement and the correlation among the radiographic, morphometric, and histological methodologies. Material and Methods: 60 male Wistar rats with induced ABL were randomly divided into 6 experimental groups (n = 10 animals/group). After 0, 3, 7, 15, 30, and 60 days, the animals were euthanized and their hemimandibles were removed for ABL determination using radiographic, morphometric and histological techniques. Results: Radiographic and morphometric/linear techniques allowed the detection of statistically significant ABL on the third day, while histological and morphometric/area techniques could only detect ABL after the seventh day (ANOVA/Tukey, p<0.05). After the fifteenth day, except for histological analysis, the ABL was stabilized. Concerning the agreement of the methodologies, Bland Altman's test (intra and inter-examiner evaluations) showed no difference among the measurements (p>0.05). In addition, high correlations (Pearson's test, r2>0.9, p<0.05) were observed. Conclusion: The results indicated that the minimum time for ABL induction could vary from 3 to 7 days, according to the chosen analysis methodology. Agreement and correlation data support the comparison of results between studies with same induction time.

Abstract Objective: This study aimed to evaluate the effects of continual intermittent administration of parathyroid hormone (PTH) on implant stability in the presence of osteoporosis, using rabbit models. Material and Methods: Fifteen female New Zealand white rabbits underwent ovariectomy and were administered glucocorticoids to induce osteoporosis, following which they were divided into three groups. The first group received intermittent subcutaneous PTH for 4 weeks until implant placement (PTH1), while the second and third groups received PTH (PTH2) and saline (control), respectively, for 4 weeks before and after implant placement. After intermittent administration of PTH or saline, titanium implants were inserted into the left femoral epiphyses of all animals, and the implant stability quotient (ISQ) was measured immediately after placement to assess the primary stability and at 2 and 4 weeks after implant placement to assess osseointegration. At 4 weeks after implant placement, histological and histomorphometric evaluations were conducted and the bone area around the implant socket was measured as a ratio of the total bone area to the total tissue area. Results: Regarding primary stability, the ISQ values for the PTH1 and PTH2 groups were significantly higher than those for the control group (p<0.05). Concerning osseointegration, the ISQ values at 2 and 4 weeks were significantly higher for the PTH2 group than for the PTH1 and control (p<0.05) groups. Histological assessments showed a thicker and more trabecular bone around the implant sockets in the PTH2 specimens than in the PTH1 and control specimens. The bone area around the implant socket was significantly greater in the PTH2 group than in the PTH1 and control groups (p<0.05). Conclusions: Our results suggest that continual intermittent PTH administration before and after dental implant placement is effective for the achievement of favorable stability and osseointegration in the presence of osteoporosis.

ABSTRACT The most recently identified serotonin (5-HT) receptor is the 5-HT7 receptor. The antinociceptive effects of a 5-HT7 receptor agonist have been shown in neuropathic and inflammatory animal models of pain. A recent study demonstrated the functional expression of 5-HT7 receptors in the substantia gelatinosa (SG) of the trigeminal subnucleus caudalis, which receives and processes orofacial nociceptive inputs. Objective To investigate the antinociceptive effects of pharmacological activation of 5-HT7 receptors on orofacial pain in mice. Material and Methods Nociception was evaluated by using an orofacial formalin test in male Balb-C mice. Selective 5-HT7 receptor agonists, LP 44 and LP 211 (1, 5, and 10 mg/kg), were given intraperitoneally 30 min prior to a formalin injection. A bolus of 10 µl of 4% subcutaneous formalin was injected into the upper lip of mice and facial grooming behaviors were monitored. The behavioral responses consisted of two distinct periods, the early phase corresponding to acute pain (Phase I: 0–12 min) and the late phase (Phase II: 12–30 min). Results LP 44 and LP 211 (1, 5, and 10 mg/kg) produced an analgesic effect with reductions in face rubbing time in both Phase I and Phase II of the formalin test. Conclusion Our results suggest that 5-HT7 receptor agonists may be promising analgesic drugs in the treatment of orofacial pain.

Objectives To compare three methods of intratubular contamination that simulate endodontic infections using confocal laser scanning microscopy (CLSM). Material and Methods Two pre-existing models of dentinal contamination were used to induce intratubular infection (groups A and B). These methods were modified in an attempt to improve the model (group C). Among the modifications it may be included: specimen contamination for five days, ultrasonic bath with BHI broth after specimen sterilization, use of E. faecalisduring the exponential growth phase, greater concentration of inoculum, and two cycles of centrifugation on alternate days with changes of culture media. All specimens were longitudinally sectioned and stained with of LIVE/DEAD® for 20 min. Specimens were assessed using CLSM, which provided images of the depth of viable bacterial proliferation inside the dentinal tubules. Additionally, three examiners used scores to classify the CLSM images according to the following parameters: homogeneity, density, and depth of the bacterial contamination inside the dentinal tubules. Kruskal-Wallis and Dunn’s tests were used to evaluate the live and dead cells rates, and the scores obtained. Results The contamination scores revealed higher contamination levels in group C when compared with groups A and B (p<0.05). No differences were observed between group A and B (p>0.05). The volume of live cells in group C was higher than in groups A and B (p<0.05). Conclusion The new protocol for intratubular infection resulted in high and uniform patterns of bacterial contamination and higher cell viability in all specimens when compared with the current methods.

ABSTRACT The ability of hemostatic agents to promote bone repair has been investigated using in vitro and in vivo models but, up to now, the results are inconclusive. Objective In this context, the aim of this study was to compare the potential of bone repair of collagen sponge with fibrin glue in a rat calvarial defect model. Material and Methods Defects of 5 mm in diameter were created in rat calvariae and treated with either collagen sponge or fibrin glue; untreated defects were used as control. At 4 and 8 weeks, histological analysis and micro-CT-based histomorphometry were carried out and data were compared by two-way ANOVA followed by Student-Newman-Keuls test when appropriated (p≤0.05). Results Three-dimensional reconstructions showed increased bone formation in defects treated with either collagen sponge or fibrin glue compared with untreated defects, which was confirmed by the histological analysis. Morphometric parameters indicated the progression of bone formation from 4 to 8 weeks. Additionally, fibrin glue displayed slightly higher bone formation rate when compared with collagen sponge. Conclusion Our results have shown the benefits of using collagen sponge and fibrin glue to promote new bone formation in rat calvarial bone defects, the latter being discreetly more advantageous.

Objective : To investigate the influence of a three-dimensional cell culture model on the expression of osteoblastic phenotype in human periodontal ligament fibroblast (hPDLF) cultures. Material and Methods : hPDLF were seeded on bi-dimensional (2D) and three-dimensional (3D) collagen type I (experimental groups) and and on a plastic coverslip (control) for up to 14 days. Cell viability and alkaline phosphatase (ALP) activity were performed. Also, cell morphology and immunolabeling for alkaline phosphatase (ALP) and osteopontin (OPN) were assessed by epifluorescence and confocal microscopy. The expression of osteogenic markers, including alkaline phosphatase, osteopontin, osteocalcin (OC), collagen I (COL I) and runt-related transcription factor 2 (RUNX2), were analyzed using real-time polymerase chain reaction (RT-PCR). Mineralized bone-like nodule formation was visualized by microscopy and calcium content was assessed quantitatively by alizarin red assay. Results : Experimental cultures produced an increase in cell proliferation. Immunolabeling for OPN and ALP in hPDLF were increased and ALP activity was inhibited by three-dimensional conditions. OPN and RUNX2 gene expression was significantly higher on 3D culture when compared with control surface. Moreover, ALP and COL I gene expression were significantly higher in three-dimensional collagen than in 2D cultures at 7 days. However, at 14 days, 3D cultures exhibited ALP and COL I gene expression significantly lower than the control, and the COL I gene expression was also significantly lower in 3D than in 2D cultures. Significant calcium mineralization was detected and quantified by alizarin red assay, and calcified nodule formation was not affected by tridimensionality. Conclusion : This study suggests that the 3D cultures are able to support hPDLF proliferation and favor the differentiation and mineralized matrix formation, which may be a potential periodontal regenerative therapy. .

Metastatic breast carcinoma has a great tendency to spread to the mandible. It is concomitantly associated with bone destruction, food intake disorder, and a poorer prognosis. Appropriate animal models need to be developed for a better understanding of the mechanisms underlying the metastatic process of breast cancer cells to mandible and to test the effects of potential lead compounds. Here, we assessed the metastasis model of intracardiac injection using luciferase-transfected metastatic breast cancer cells (MDA-MB-231Luc+) by determining the incidences of metastasis, mCT images, and histopathological results. A high bioluminescence signal mainly detected mandibular lesions with less frequent distal femora and proximal tibiae lesions. Extensive mandibular bone destruction occurred in nude mice grafted with metastatic breast cancer cells. This type of animal model might be a useful tool in assessing therapeutic implications and the efficacy of anti-cancer drugs for osteolytic cancers.

A osteonecrose dos maxilares associada ao uso dos Bisfosfonatos é uma exposição óssea que persiste por mais de 8 semanas na cavidade oral em pacientes sob tratamento com bisfosfonatos e que não foram submetidos a radioterapia de cabeça e pescoço. Explicar o motivo do desenvolvimento destas lesões ósseas principalmente nos maxilares e desvendar a sua patofisiologia ainda é necessário. Por isso, o reparo ósseo em um modelo animal de osteonecrose dos maxilares associada ao uso de bisfosfonatos em ratos Wistar (Rattus norvegicus, albinus) foi avaliado através de análise microscópica e molecular. A amostra foi composta por 48 ratos machos, com 12 semanas de vida e peso aproximado de 300 gramas, que foram submetidos a administração de Ácido Zoledrônico, 0,6 mg/kg a cada 28 dias com um total de 5 doses. Os animais foram dividos em quatro grupos, cada um composto por 12 animais; 2 grupos de tratamento AZ e AZ-experimental (AZ-exp) e 2 grupos controles CO e CO-experimental (CO-exp) com administração de cloreto de sódio 0,9% no mesmo volume e frequencia do Ácido Zoledrônico. As soluções foram administradas por via intraperitoneal. O grupo AZ-exp e o CO-exp foram submetidos a exodontias dos molares superiores direitos e a realização de defeito ósseo no fêmur esquerdo 45 dias após a primeira aplicação das soluções. A eutanásia dos animais ocorreu após 150 dias do início do experimento. A avaliação histológica foi realizada através de análises qualitativa e quantitativa, verificou a presença de sequestros ósseos, áreas de osteonecrose e áreas de osso total por meio de estudos de microscopia óptica pela coloração Hematoxilina e Eosina. Análise quantitativa da expressão do RNAm de proteínas envolvidas no processo de reparo ósseo pelo método de reação em cadeia da polimerase em tempo real (RealTimePCR) também foi realizada para avaliação dos osteoclastos (RANK, RANKL e OPG), osteoblastos (ALPL e OCN) osteócitos (DMP-1 e PHEX) e vascularização (VEGF).


Bisphosphonate-related osteonecrosis of the jaws is a bone exposure persisting for more than 8 weeks in the oral cavity in patients receiving bisphosphonates and who did not undergo radiation therapy for head and neck. Explain the reason for these development bone lesions mainly in the jaws and unveil its pathophysiology is still needed. Therefore, the bone repair in an animal model of Bisphosphonate-related osteonecrosis of the jaws in Wistar rats (Rattus norvegicus, Albinus) was evaluated by microscopic and molecular analysis. The sample was composed of 48 male rats, with 12 weeks old and weighing approximately of 300 grams, who underwent the administration of zoledronic acid, 0.6 mg / kg every 28 days with a total of 5 doses. The animals were divided into four groups, each consisting of 12 animals; 2 treatment groups AZ and AZ-experimental (AZ-exp) and 2 control groups CO and CO-experimental (CO-exp) with sodium chloride administration 0.9% in the same volume and frequency of zoledronic acid. All solutions were administered intraperitoneally. The AZ-exp group and the CO-exp group underwent extractions rights upper molars and the bone defect was performed in the left femur 45 days after the first application of the solutions. Euthanasia of animals occurred after 150 days from the beginning of the experiment. Histological evaluation was performed through quantitative and qualitative analysis checked the presence of bone sequestration, osteonecrosis area and whole bone areas by means of optical microscopy by hematoxylin-eosin staining. O reparo ósseo de alvéolos após exodontia dos molares superiores em um modelo animal em ratos Wistar (Rattus norvegicus, albinus) de osteonecrose dos maxilares associada ao uso de bisfosfonatos foi avaliado através de analise microscópica e molecular. Foram utilizados 48 ratos (Rattus norvegicus, albinus, Wistar) machos, com 12 semanas de vida e peso aproximado de 300 gramas, que foram dividos em 4 grupos. Cada grupo era composto por 12 animais, sendo 2 grupos experimentais AZ e AZ-Cirúrgico (AZ-C), que foram submetidos a administração de ácido zoledrônico, 0,6 mg/kg a cada 28 dias com um total de 5 doses e 2 grupos controles CO e CO-Cirúrgico (CO-C) com administração de cloreto de sódio 0,9% no mesmo volume e frequencia do ácido zoledrônico. Todas as soluções foram administradas por via intraperitoneal. O grupo AZ-C e o grupo CO-C foram submetidos a exodontia do primeiro, segundo e terceiro molares superiores 45 dias após a primeira aplicação das soluções. Todos os animais foram eutanasiados após 150 dias do início do experimento (105 dias após as exodontias). As maxilas dos animais foram avaliadas macroscopicamente para presença de lesões espontâneas e com uma sonda clinica número 5 as regiões das exodontias dos molares foram avaliadas para presença ou ausência de solução continua do epitélio. Após feita a avaliação macroscópica as regiões das exodontias dos molares superior esquerdo e do lado contralateral de cada animal foram submetidas a análises qualitativa e quantitativa para presença de sequestros ósseos, restos radiculares, área de osteonecrose, área de espaço trabecular, área de reação periosteal, através de estudos por microscopia óptica pela coloração Hematoxilina e Eosina. The alveolar bone repair following extraction of maxillary molars in an animal model of bisphosphonate related osteonecrosis of the jaws in Wistar rats (Rattus norvegicus, Albinus) was assessed through microscopic and molecular analysis. A total of 48 rats (Rattus norvegicus, Albinus, Wistar rats) with 12 weeks old and weighing approximately 300 grams were used, they were divided into 4 groups. Each group consisted of 12 animals, with 2 experimental groups AZ and AZ-Cirúrgico (AZ-C), who underwent the administration of zoledronic acid, 0.6 mg / kg every 28 days with a total of 5 doses. After macroscopic evaluation, the upper left molar and contralateral side of the extraction regions of each animal were submitted to qualitative and quantitative analyzes for the presence of bone sequestrum, root fragments, osteonecrosis area, trabecular space area, area of periosteal reaction, through optical microscopic studies by hematoxylin and eosin staining. And quantitative analysis of mRNA expression of proteins involved in bone repair (RANK, RANKL, OPG and VEGF), by the method of RealTimePCR were carried out. Considerando que os dados epidemiológicos apontam uma elevação alarmante na incidência de diabetes na população mundial, muitos estudos vêm sendo realizados buscando estabelecer propostas preventivas e terapêuticas para esta patologia e suas complicações, principalmente em relação a cicatrização de feridas. A maneira convencional de unir os tecidos e as margens de feridas, utilizando suturas pode causar problemas como fístula e formação de granulomas, devido a uma incompatibilidade do tecido com os materiais de sutura, deiscência quando os materiais de sutura absorvível mostram desintegração precoce e isquemia tecidual com conseqüente necrose de borda da ferida quando uma sutura é muito apertada. Dentre os novos meios de adesão tecidual que têm sido idealizados, um deles é o adesivo biológico, tendo como objetivo selar tecidos e feridas sem produzir qualquer tipo de trauma O adesivo cirúrgico derivado do veneno de serpente é um produto biológico e biodegradável que não produz reações adversas, não contém produtos derivados do sangue humano, portanto não favorece transmissão de doenças infecciosas, tem uma capacidade de bom adesivo, e pode ser usado como coadjuvante nos procedimentos de sutura convencional. Este trabalho analisou, portanto, em feridas cirúrgicas feitas no dorso de ratos Wistar machos de três meses de idade diabéticos e não diabéticos três materiais utilizados como coadjuvantes na cicatrização de feridas cirúrgicas: sutura de nylon 5.0, a cola de fibrina comercial Tissucol® e o adesivo cirúrgico do veneno de serpente. Os animais foram eutanasiados nos períodos de três e sete dias do pós-operatório e as áreas das feridas foram analisadas microscopicamente através das colorações de Hematoxilina & Eosina e Tricrômio de Mallory. No experimento realizado constatamos que a reepitelização no grupo controle foi favorecida pelo material Tissucol®. Whereas epidemiological data point to an alarming increase in the incidence of diabetes in the world population, many studies have been conducted to establish preventive and therapeutic proposals for this disease and its complications, especially in relation to wound healing. The conventional way of joining tissue and wound edges, using sutures may cause problems such as fistulas and granulomas due to incompatibility with the materials of the fabric of suture dehiscence when the absorbable suture materials show premature disintegration and tissue ischemia with subsequent necrosis of the wound edge when a suture is too tight. Among the new means of tissue adhesion that have been devised, one of them is the biological adhesive, aiming to "paste" and injured tissues without producing any kind of trauma. The surgical adhesive derived from snake venom is a biological product that is not biodegradable and produce adverse reactions, it does not contain human blood therefore does not favor the transmission of infectious diseases, have a good adhesive ability , and can be used as an adjunct to conventional suturing procedures. This study therefore considered in surgical wounds made in the back of male Wistar rats three months old diabetic and non-diabetic three materials used as adjuvants in the healing of surgical wounds: 5.0 nylon suture, the commercial fibrin seletante Tissucol® and surgical adhesive snake venom. The animals were sacrificed at three and seven days after surgery and wound areas were analyzed microscopically by staining with hematoxylin & eosin and Mallory trichrome . In the experiment we found that re-epithelialization in the control group was favored by the material Tissucol®. The presence of mononuclear cell infiltration was prominent significantly by the use of surgical adhesive derived from snake venom. O presente trabalho teve como objetivo associar modelos indutivos de diabetes e hipertensão, analisar e comparar o efeito de agentes vasoconstritores presentes nas soluções anestésicas locais, injetados por via intravenosa nas doses de 80, 160, 320, 640 e 1280ng (adrenalina) ou 0,125; 0,25; 0,5; 1; 2 e 3 x10-3UI (felipressina), sobre a pressão arterial de ratos normotensos, diabéticos, hipertensos renais um-rim, um clip (1R-1C) e hipertensos1R-1C-diabéticos, além de verificar a glicemia após injeção de doses correspondentes a 20 e 80% da resposta pressora máxima. Ratos Wistar machos pesando 110-160g, foram anestesiados com mistura de quetamina e xilazina (50+10mg/ml/kg de peso), tiveram seu abdômen aberto e receberam um clip de prata com abertura 0,25mm na artéria renal esquerda, removendo-se cirurgicamente o rim direito (ratos 1R-1C). Após 14 dias, receberam injeção subcutânea de estreptozotocina (50 e 60mg/kg de peso) para indução do diabetes mellitus sendo a glicemia testada pela veia caudal previamente aos experimentos (diabéticos). Após 28 dias da implantação do clip, todos os grupos foram novamente anestesiados e implantaram-se cânulas de polietileno (PE-50) na artéria carótida esquerda e veia jugular direita, para registro direto da pressão arterial e injeção de drogas, respectivamente. Animais controle sofreram cirurgia sem implantação do clip (normotensos). O cateter arterial foi conectado ao sistema de registro computadorizado (PowerLab®) utilizando software específico (Chart 5Pro®). A pressão arterial registrada durante os primeiros 5 minutos foi considerada como valor basal. Analisaram-se: a integral da resposta, menor resposta hipotensora, maior resposta hipertensora, duração e intervalo de tempo para atingir respostas máxima e mínima, em todos os grupos, para cada dose injetada dos dois vasoconstritores. No dia seguinte, foi medida a glicemia inicial e após a injeção das doses de 160 e 640ng (adrenalina) ou 0,25 e 2 x10-3UI (felipressina)...


The present study was designed to induce arterial hypertension associated with diabetes mellitus, analyze and compare the effects of vasoconstrictors drugs, presents in anesthetic cartridge, injected by intravenous route (IV), in doses of 80, 160, 320, 640 and 1280ng (epinephrine) or 0,125; 0,25; 0,5; 1; 2 e 3x10-3UI (felypressin) on arterial pressure (AP) of normotensive, diabetic, one-kidney,one-clip (1K-1C) hypertensive and 1K-1C hypertensive-diabetic rats. The blood glucose levels were determined after IV injection of both drugs in doses that produced 20 and 80% of maximal pressure response. Male Wistar rats weighing 110-160g were anesthetized with a ketamine and xylazine mixture (50+10mg/ml/kg de peso). After abdominal incision, a silver clip with 0.25-mm gap was implanted in the main left renal artery and right kidney was removed (1K-1C rats). Fourteen days after, some animals received subcutaneous injection of streptozotocin (50 and 60mg/kg weight) to induce diabetes mellitus. Tail blood glucose was tested before experiments (diabetics rats). Four weeks after clip implantation all rats were anesthetized again and a catheter (PE50) was inserted into the left carotid artery and right jugular vein, respectively to obtain a direct arterial pressure register and to inject drugs. Control groups were submitted to surgery procedures without clip installation (normotensive). The arterial catheter was then connected to the transducer and to the computer register system (PowerLab®) using a Chart 5 Pro® software. The AP registered in the first five minutes was considered as a basal value. The following parameters were registered: integral of complete response, minimal hypotensive response, maximal hypertensive response, response length (duration) and duration of time to attain maximum and minimum responses, in all animals groups, to every injected dose of both vasoconstrictor drugs. A Estomatite protética associada a Candida (EPC) acomete a mucosa bucal em contato com próteses removíveis e, clinicamente, caracteriza-se por eritema com diferentes graus de inflamação. Esta doença é considerada de etiologia multifatorial, isto é, uma associação de fatores como trauma, falta de higienização, uso contínuo da prótese, hipersensibilidade ao material usado na dentadura, diabetes, terapia imunossupressora e infecção por fungo, como diferentes espécies de Candida. Os principais fatores de virulência deste fungo são a formação de hifas, dimorfismo, alterações fenotípicas, aderência, persistência, sinergismo com as bactérias, interferências com o sistema de defesa do hospedeiro e a produção de enzimas hidrolíticas. Dentre as enzimas hidrolíticas, a proteinase aspartil secretada (SAP) é uma das mais importantes para a patogenia de C. albicans, sendo nociva para o tecido epitelial e para o sistema imune do hospedeiro. Não está totalmente compreendida a real penetração do fungo nos tecidos e sua correlação com a presença da SAP, na doença estomatite protética. Essa dificuldade de avaliação pode ser justificada pelas divergências intrínsecas e extrínsecas observadas em muitos aspectos, como diferentes costumes, hábitos sociais, estado emocional e fisiológico. A utilização de um modelo experimental em animais poderá minimizar essas divergências e fornecer condições mais padronizadas para o experimento. Neste trabalho, foram avaliadas, quantitativamente, a expressão gênica das enzimas SAP-2, SAP-5 e SAP-9, presentes no biofilme formado na superfície interna das placas acrílicas superiores de ratos e, microscopicamente, a invasão do fungo no tecido epitelial do palato. Denture stomatitis (D.S.) affects the oral mucosa in contact with removable dentures, and clinically characterized by erythema with varying degrees of inflammation. This disease is considered a multifactorial etiology, with a combination of factors such as trauma, lack of hygiene, continuous use of stents, hypersensitivity to the material used for dentures, diabetes, immunosuppressive therapy and fungal infection, such as different species of Candida. The main virulence factors of the fungus are the formation of hyphae, dimorphism, phenotypic changes, adherence, persistence, synergism with bacteria, interference with the host defense system and the production of hydrolytic proteins. Among the hydrolytic proteins, the secreted aspartyl proteinase (SAP) is one of the most important in the pathogenesis of C. albicans. SAP is harmful to both the epithelial tissue and to the host's immune system. It is not fully understood the real penetration of the fungus in the epithelium tissue and its correlation with the presence of SAP, in denture stomatitis. This difficulty in evaluation can be justified by the intrinsic and extrinsic differences observed in many aspects, different customs, social`s habits, emotional and physiological state. Using an experimental animal model may minimize these differences and provide more standardized conditions for the experiment. A Camellia sinensis, vem sendo amplamente estudada, em função das suas propriedades medicinais, devido aos polifenóis com funções antioxidantes, tendo seu uso propostos como terapêutica para algumas doenças metabólicas como o Diabetes mellitus. A ingestão diária de antioxidantes, principalmente com compostos fenólicos pode retardar o aparecimento destas doenças. O atual trabalho o avaliou possível efeito terapêutico do chá verde (Camellia Sinensis) em ratos diabéticos induzido pela estreptozotocina, parâmetros clínicos (peso, ingestão hídrica) e laboratoriais (glicemia de jejum, colesterol total, triglicérides, proteínas totais, albumina e uréia) esse efeito na glândula submandibular pelo método morfométrico. Cento e sessenta ratos machos com 2 meses de idade foram distribuídos inicialmente em controle (n = 80) e diabéticos (n = 80). Os animais do grupo controle água (CA) e diabético água (DA) consumiram água como dieta líquida e os animais dos grupos controle chá (CC) e diabético chá (DC) receberam apenas dieta líquida de chá verde. Os parâmetros bioquímicos e morfométricos foram avaliados nos períodos experimentais de 15, 30, 60 e 90 dias (n = 5), após a indução. Os resultados mostraram que: a) a ingestão hídrica dos animais dos grupos DA e DC foram em média 6 vezes maior em relação aos controles; b) aos 15 dias, todos os animais dos grupos CA, CC e DA apresentaram aumento nos índices glicêmicos final de, respectivamente, 45 mg/dL, 65 mg/dL e 88mg/dL em relação ao período inicial, enquanto que todos os animais do grupo DC tiveram seus índices glicêmicos reduzidos em média de 119 mg/dL; c) nos ratos diabéticos o perfil bioquímico das proteínas totais e albumina foram menores em relação aos controles e o da uréia maior, enquanto que, o colesterol total, triglicérides não apresentaram diferenças; d) os ácinos das glândulas submandibulares dos animais dos grupos DA e DC exibiram aumento da eosinofilia citoplasmática e...


The Camellia sinensis, has been widely studied because of their medicinal properties due to the polyphenols with antioxidant functions, and proposed its use as treatment for various metabolic disorders such as diabetes mellitus. The daily intake of antioxidants, mainly phenolic compounds may prevent or delay onset of these diseases. In this study, the possible therapeutic action of green tea (Camellia Sinensis) in diabetic rat induced chemically by streptozotocin was evaluated by clinical (weight, water intake) and laboratory parameters (fasting glucose, total cholesterol, triglycerides, total protein, albumin and urea) and its effect in the submandibular glands by morphometric methods. One hundred and sixty male rats with 2 months of age were distributed initially in control (n = 80) and diabetic (n = 80). The animals of the water control (WC) and water diabetic (WD) groups received only regular drinking water and green tea control (GTC) and green tea diabetic (GTD) groups received green tea solution ad libitum. Morphometric and biochemical parameters were evaluated after 15, 30, 60 and 90 days (n = 5) after induction. The results showed that: a) the water intake of animals from group WD and GTD were six times higher on average than control groups; b) at 15 days, all animals of the WC, WD and GTC groups showed an increase of the glycemic index of, respectively, 45 mg/dL, 65 mg/dL and 88 mg/dL compared to the initial period, while that all animals in CTD group, showed a decrease of 119mg/dL; c) in the diabetic rats of WD and GTD groups the total protein and albumin levels were lower than control groups and urea level higher, whereas, the total cholesterol and triglyceride did not show significant differences, d) the acini of submandibular glands of animals in groups WD and GTD showed increase of the cytoplasmic eosinophilia and absence of the basal basophilia, which may be indicative of changes in protein content and/or structure, and pyknotic...
