A infecção endodôntica ocorre a após contaminação do tecido pulpar levando à uma colonização bacteriana dos canais radiculares resultando em uma resposta inflamatória dos tecidos periapicais e formação de uma lesão periapical, a periodontite apical (PA). O tabagismo, por sua vez, tem impactos prejudiciais à saúde oral e sistêmica sendo considerado um importante fator de risco para as doenças periodontais. Este trabalho tem por finalidade investigar a influência do tabagismo no desenvolvimento da periodontite apical em ratos. Trinta e dois ratos machos Wistar foram divididos em 4 grupos experimentais (n=8): Controle (sem periodontite apical induzida e sem inalação da fumaça do cigarro); FU (com inalação a fumaça do cigarro); PA (com periodontite apical induzida); FU+PA (com inalação a fumaça do cigarro e periodontite apical induzida). Para a inalar a fumaça do cigarro, grupo de cinco animais permaneceram em uma câmara de tabagismo inalando a fumaça de 10 cigarros por 8 minutos, três vezes ao dia, durante 50 dias. Decorridos 20 dias de inalação de fumaça do cigarro, foi realizada a cirurgia para indução da periodontite apical nos animais do grupo PA e FU+PA, no primeiro molar inferior direto, o qual permaneceu aberto na cavidade bucal por 30 dias. Nesses 30 dias subsequentes da indução da PA, os animais do grupo FU+PA e FU continuaram com a inalação a fumaça do cigarro. No 50º dia, os animais foram eutanasiados e coletados amostras de tecido hematológico para avalição dos níveis séricos de nicotina, cotinina, fosfatase alcalina, cálcio, fósforo, série vermelha e série branca. As hemimandibula removidas foram escaneadas em microtomógrafo para avaliar o volume da destruição óssea e processadas histologicamente para avaliação do perfil inflamatório e expressão das citocinas IL-6, IL-1ß, TNF-α e dos marcadores do metabolismo ósseo RANKL e OPG. Os dados foram tabulados e aplicados os testes estatísticos de Mann-Whitney para os dados não paramétricos e teste t para os dados paramétricos, a um nível de significância de P< 0.05. Com relação a análise histológica o grupo FU+PA apresentou infiltrado inflamatório mais intenso em relação aos demais grupos (P< 0,05). As citocinas pró-inflamatórias IL-6, IL-1ß, TNF-α apresentaram alto padrão de imunomarcação para o grupo FU+PA (P< 0,05). A análise histométrica mostrou maior área de reabsorção óssea no grupo FU+PA, assim como na análise microtomográfica (P< 0,05). As citocinas RANKL esteve mais expressa no grupo FU+PA (P< 0,05) e OPG teve maior expressão no grupo PA (P< 0,05). Na análise hematológica houve um aumento na concentração de hemácias, hemoglobina e leucócitos para o FU+PA (P< 0,05). Os níveis séricos de cálcio foram menores no FU+PA (P> 0,05), a fosfatase alcalina e o cálcio se manteve constante em todos os grupos experimentais (P> 0,05). A concentração sérica de nicotina e cotinina no grupo FU e FU+PA foram compatíveis com fumante humano(AU)


Endodontic infection occurs mainly after pulp tissue contamination by a carious process, leading to bacterial colonization of root canal system and inflammatory response of periapical tissues, followed by formation of apical periodontitis (AP). It is widely known that smoking has harmful impacts on oral and systemic health and is considered a risk factor for periodontal diseases. The objective of this research was to investigate the influence of smoking on the development of AP in rats. Thirty-two male Wistar rats were divided into 4 experimental groups (n = 8): C - Control (no induced AP and no cigarette smoke inhalation); CSI (cigarette smoke inhalation); AP (induced apical periodontitis); CSI + AP (cigarette smoke inhalation and induced apical periodontitis). To inhale cigarette smoke, group with five animals remained in a smoking chamber inhaling smoke from 10 cigarettes for 8 minutes, three times daily, for 50 days. After 20 days of cigarette smoke inhalation, pulp chamber access was performed to induce apical periodontitis in the first mandibular right molar, which remained with pulp chamber exposed to oral cavity for 30 days in animals in the AP and CSI + AP group. During these 30 days after the AP induction, animals in the CSI + AP and CSI group continued to inhale cigarette smoke. On the 50th day, animals were euthanized and blood sample collected to assess serum levels of nicotine, cotinine, alkaline phosphatase, calcium, phosphorus, red series and white series. The hemimandibles were removed and scanned in microtomograph to assess bone volume and histologically processed to assess inflammatory profile and expression of cytokines IL-6, IL-1ß, TNF-α and bone metabolism markers RANKL and OPG. Data were tabulated and statistically analyzed using Mann-Whitney tests for nonparametric data and analysis of variation test for parametric data, with a significance level of P< 0.05. Regarding histological analysis, CSI+AP group showed more intense inflammatory infiltrate compared to other groups (P < 0.05). Histometric analysis showed a larger area of bone resorption in the CSI + AP group, also observed in the microtomographic analysis (P< 0.05). Group CSI+AP had elevated pro-inflammatory cytokines IL-6, IL-1ß, TNF-α expression (P< 0.05). The RANKL cytokines were also more expressed in the CSI+AP group (P< 0.05), while OPG was more expressed in the AP group (P< 0.05). The hematological analysis revealed an increase in the concentration of red blood cells, hemoglobin, and leukocytes for CSI+AP (P< 0.05). Serum calcium levels were lower in CSI+AP (P> 0.05), and alkaline phosphatase and calcium remained constant in all experimental groups (P> 0.05). The serum concentrations of nicotine and cotinine in the CSI and CSI+AP group were compatible with human smokers(AU)

O estudo avaliou o efeito da adição de cálcio (Ca) e fósforo (P), em quantidade correspondente ao ponto de saturação (PS) e ao coeficiente de solubilidade (CS), a géis clareadores contendo diferentes agentes espessantes na desmineralização do esmalte dentário. Foram manipulados géis clareadores contendo peróxido de hidrogênio (PH) a 35% e utilizados os espessantes carbopol 980 (CB); aerosil 200 (SP); poloxamer 407 (PX); goma guar (GG) e hidroxietilcelulose (HEC). Foram realizadas leituras iniciais da microdureza Knoop do esmalte empregando um microdurômetro, da rugosidade superficial em um perfilômetro de contato e da cor utilizando espectrofotômetro colorimétrico de reflectância. Os espécimes foram divididos em 5 grupos experimentais de acordo com o tipo de espessante. Cada grupo foi dividido em três subgrupos (n=20), de acordo com a suplementação mineral (0- nenhuma suplementação; PS ­ adição de Ca e P de acordo com o PS calculado para aquele espessante; CS ­ adição da quantidade máxima de Ca e P de acordo com o CS). Além disso, três grupos controles (n=20) foram preparados, sendo eles: CN (controle negativo ­água ultrapura); CP (controle positivo ­ PH a 35% sem a adição de qualquer mineral); CC (controle positivo comercial - gel comercial Whiteness HP). Sobre cada espécime foram aplicados 0,050 g dos respectivos géis clareadores, durante 45 min. Os dados dos grupos experimentais foram analisados estatisticamente com ANOVA a 2 fatores (TIPO DE ESPESSANTE x ADIÇÃO DE SAIS DE CA E P) e teste de Tukey. A comparação com os grupos controle foi realizada com o teste de Dunnett ( = 5%). Para todas as mensurações realizadas, a ANOVA mostrou diferenças significativas para os dois fatores (p<0,05). Para a alteração da microdureza, os resultados do teste de Tukey foram: TIPO DE ESPESSANTE (p=0,0001): HEC ­ 15,40(6,97)a, GG - 10,31(5,78)b, CB - 7,49(2,87)c, PX - 4,60(4,96)d, SP - 0,57(2,43)e; ADIÇÃO (p=0,0001): 0 - 12,34(7,22)a, S -10,31(5,76)b, CS -7,49(6,09)c. Para a alteração da rugosidade (p=0,0001), os resultados foram: TIPO DE ESPESSANTE (p=0,0001): SP - 5,19(23,36), GG - 9,70(17,75)ab, PX - 15,95(20,79)b, HEC - 53,58(71,82)c, CB - 55,49(63,69)c; ADIÇÃO (p=0,0001): 0 - 79,62(58,70)a, S - 3,23(12,08)a, CS - 1,09(9,44)b. Para a mudança de cor (ΔE), os resultados foram: TIPO DE ESPESSANTE (p=0,0001): SP - 5,40(2,92)a, GG - 3,13(1,83)a, PX - 3,18(2,57)ab, HEC - 3,82(2,05)bc, CB - 4,79(3,26)c. ADIÇÃO (p=0,2853): 0 - 4,37(2,60)a, S - 4,04(3,07)a, CS - 3,79(2,42)a. Os resultados para o teste de Dunnett para a microdureza: os grupos SP, SP-S e SP-CS não demonstraram diferenças com o grupo CN, e apenas os grupos GG e HEC-S não demostraram diferenças com o grupo CP e o grupo HEC-S para o CPC. Para a rugosidade, os grupos CB-CS, SP, PX-S, PX-CS, GG-S e GG-CS não demonstraram diferenças em relação ao grupo CN e todos os grupos apresentaram diferenças significativas para os grupos CP e CPC. Mediante os resultados desse estudo, podemos concluir que: A adição de cálcio e/ou fósforo em quantidades correspondente ao PS ou ao CS apenas do grupo SP impediu a queda da dureza e o aumento da rugosidade. Os demais grupos apresentaram alterações, porém sem afetar o tratamento clareador. (AU)


The aim of the study was to evaluate the effect of adding calcium (Ca) and phosphorus (P), in amounts corresponding to the saturation concentration (SC) and the solubility limit (SL), to bleaching gels containing different thickening agents on the demineralization of tooth enamel. Bleaching gels containing 35% hydrogen peroxide (PH) were manipulated and the thickeners were used: carbopol 980 (CB); aerosil 200 (SP); poloxamer 407 (PX); guar gum (GG) and hydroxyethylcellulose (HEC). The baseline readings of Knoop microhardness of the enamel using a microhardness, surface roughness using a contact profilometer and color using a colorimetric reflectance spectrophotometer were taken. The specimens were divided into 5 experimental groups according to the type of thickener. Each group was divided into three subgroups (n=20), according to mineral supplementation (0 - no supplementation; SC - addition of Ca and P according to the SC calculated for that thickener; SL - addition of the maximum amount of Ca and P according to SL). In addition, three control groups (n=20) were prepared, namely: NC (negative control ­ ultra pure water); PC (positive control ­ 35% pH without the addition of any mineral); CC (commercial positive control - commercial Whiteness HP gel). Were applied 0.050 g of the respective bleaching gels to each specimen for 45 min. The microhardness and surface roughness of the samples were measured immediately after bleaching. All samples were then immersed in artificial saliva for 7 days and the final color evaluated. Data from the experimental groups were statistically analyzed with 2-way ANOVA (THICKENER TYPE x MINERAL SUPPLEMENTATION) and Tukey test. Comparison with control groups was performed using Dunnett's test ( = 5%). For all measurements performed, ANOVA showed significant differences for the two factors (p<0.05). For microhardness change, the Tukey test results were: THICKENER TYPE (p=0.0001): HEC - 15.40(6.97)a, GG - 10.31(5.78)b, CB - 7.49(2.87)c, PX - 4 .60(4.96)d, SP - 0.57(2.43)e; ADDITION (p=0.0001): 0 - 12.34(7.22)a, SC -10.31(5.76)b, SL -7.49(6.09)c. The results for the Dunnett test for microhardness were: the SP, SP-S and SP-CS groups did not show differences with the CN group, and only the GG and HEC-S groups did not show differences with the CP group and the HEC-S for the CPC. For roughness, the CB-CS, SP, PX-S, PX-CS, GG-S and GG-CS groups did not show differences in relation to the CN group and all groups showed significant differences for the CP and CPC groups. By the results of this study, we can conclude that: The addition of calcium and/or phosphorus in amounts corresponding to the SC or to the SL of the aerosil group only prevented the drop in hardness and the increase in roughness. Abstract This study evaluated the physicochemical, biological, and antimicrobial properties of a new hydraulic calcium silicate-based modified material, and compared it with MTA Repair HP and MTA Angelus. The materials were assessed regarding color luminosity (L), color change, radiopacity, setting time, and ISO 6876:2012 linear flow. Volumetric filling and volume change were evaluated using microcomputed-tomography (µCT). Chemical characterization after 28 days in Hank's Balanced Salt Solution (HBSS) and pH analysis were also assessed. Biological characterization of cytotoxicity and microbiological assessment were also undertaken. Shapiro-Wilk, ANOVA, Levene and post hoc analyses with Bonferroni correction were performed, adopting a 5% significance level (p <0.05). Bio-C Pulpo exhibited the highest L values after 90 days. All tested materials demonstrated color change during the analyses, and had radiopacity above 5 mm Al. MTA Repair HP set faster than Bio-C Pulpo, whereas the latter had the highest linear flow. MTA Repair HP had the highest volumetric filling in µCT analysis. Bio-C Pulpo showed the highest alkalinity during all tested periods, and the highest volumetric loss (above 9%), in comparison with MTA Repair HP and MTA Angelus. Bio-C Pulpo did not form calcium hydroxide after hydration. MTA Repair HP demonstrated the highest cytocompatibility, and Bio-C Pulpo, the highest cytotoxicity. No inhibition halos were observed for any material, and similar higher turbidity values were seen after direct contact. Composition additives used in Bio-C Pulpo modified its properties, and both the absence of calcium hydroxide deposition after hydration, and the related cytotoxicity of this material are of particular concern.

O objetivo do presente estudo foi sintetizar e caracterizar ciclotrifosfato de sódio (NaTMP) contendo cálcio, e verificar seu efeito utilizando modelo de lesões iniciais de erosão do esmalte. Os ciclotrifosfatos contendo cálcio (CaNaTMP) foram sintetizados utilizando coluna para cromatografia e adição de sobrenadante de solução contendo hidróxido de cálcio e analisados por meio de microscopia eletrônica de varredura e espectroscopia de raios-X por dispersão de energia. Para determinar o efeito sobre lesões erosivas iniciais, blocos de esmalte bovino sadios (n=96) foram selecionados por dureza de superfície inicial e divididos em 8 grupos experimentais (12 blocos/grupo): controle (água deionizada), 0,24% NaF (1100 ppm F), 0,25%, 0,5% e 1% de NaTMP e CaNaTMP nas mesmas concentrações. Os blocos de esmalte foram imersos em 4 mL das soluções experimentais durante 2 minutos, seguidos por 4 desafios erosivos (ácido cítrico, 0,75%, pH 3,5, por 1 minuto, sob agitação). A porcentagem de perda da dureza de superfície (%SH) foi calculada após cada desafio ácido. Os dados foram submetidos à análise de variância de medidas repetidas a dois critérios, seguida pelo teste de Tukey (p< 0,05). O processo de síntese levou a substituição de átomos de Na por átomos de Ca e as partículas apresentaram tamanhos homogêneos. As soluções contendo 0,25%, 0,5% e 1% CaNaTMP apresentaram menor %SH quando comparadas as suas contrapartes sem cálcio (p< 0,001), após os quatro desafios erosivos. Quando comparado a solução contendo 1100 ppm F, as soluções 0,5% e 1% CaNaTMP promoveram redução na perda de dureza (p< 0,001). Concluiu-se que soluções contendo CaNaTMP promoveram efeitos protetores superiores em comparação ao grupo 1100 ppm F sobre lesões iniciais do esmalte(AU)


The objective of the present study was to synthesize and characterize sodium cyclotriphosphate (NaTMP) containing calcium and verify its effect using an initial enamel erosion model. Cyclotriphosphate containing calcium (CaNaTMP) was synthesized using column chromatography, and addition of a solution with calcio hydroxide supernatant and analyzed by scanning electron microscopy and energydispersive X-ray spectroscopy. To determine the effect on enamel initial erosion, sound bovine enamel blocks (n=96) were selected by initial surface hardness and divided into to 8 experimental groups (12 blocks/group): control (deionized water), 0.24% NaF (1100 ppm F), 0.25%, 0.5% and 1% NaTMP and CaNaTMP at the same concentrations. The enamel blocks were immersed in 4 mL of the experimental solutions for 2 minutes followed by 4 erosive challenges (citric acid, 0.75%, pH 3.5, for 1 minute, under stirring). The percentage of surface hardness variation (%SH) was calculated after each acid challenge. Data were subjected to two-way repeated measures analysis of variance, followed by Tukey's test (p< 0.05). The synthesis process led to the replacement atoms of Na by atoms of Ca with particles having, homogeneous sizes. The solutions containing 0.25%, 0.5% and 1% CaNaTMP promoted lower %SH when compared to their counterparts without calcium (p< 0.001), after the four erosive challenges. When compared to the solution containing 1100 ppm F, the 0.5% and 1% CaNaTMP solutions were superior in reducing hardness loss (p< 0.001). It was concluded that solutions containing CaNaTMP led to superior protective effects compared to the 1100 ppm F group on initial enamel erosion(AU)

ABSTRACT Objective: To compare salivary and serum biochemical levels in patients with chronic renal failure undergoing hemodialysis. Material and Methods: The sample was composed of 57 patients treated in Hemodialysis Reference Centers, from a state of Northeastern Brazilian, with age ≥21 years old with at least 3 months of hemodialysis treatment time. Serum data were obtained from records. Unstimulated and stimulated saliva were collected. Flow rate (mL/min) was measured. Spectrophotometry was performed for the measurement of salivary levels of calcium (570 nm), urea (340 nm), and creatinine (510 nm). Statistical analysis used Mann Whitney and Kruskal-Wallis tests (p<0.05). Results: Unstimulated and stimulated salivary flow rates were 0.43 mL/min and 1.69 mL/min, respectively. There was significant difference (p<0.001) of levels of calcium (5.41 mg/dL and 9.70 mg/dL), urea (118.03 mg/dL and 183.22 mg/dL) and creatinine (0.59 mg/dL and 9.20 mg/dL) between saliva and serum, respectively. Concerning the time of hemodialysis, salivary and serum calcium not exhibited significant association; however, serum urea (p=0.012) and serum creatinine (p=0.025) showed significant association to the time of hemodialysis. Conclusion: Salivary biochemical levels of urea, creatinine and calcium can indicate the presence of a possible chronic renal failure and the saliva demonstrated to be a potential auxiliary biofluid for clinical monitoring renal alterations.

Abstract Homeostasis between salivary calcium and phosphorus is important for maintaining oral health. The aim of this study was to evaluate if polymorphisms in ESR1 (Estrogen Receptor Alpha), ESR2 (Estrogen Receptor Beta) and miRNA17 (microRNA17) are associated with calcium and phosphorus levels in saliva. Saliva from 276 12-year-old children were collected by masticatory stimulation and calcium and phosphorus levels were determined by Mass Spectrometry. Genomic DNA was extracted from remaining saliva and genetic polymorphisms in ESR1 (rs12154178, rs1884051, rs9340799 and rs2234693), in ESR2 (rs4986938 and rs1256049) and in miRNA17 (rs4284505) were genotyped using TaqMan chemistry and a real-time PCR equipment. Statistical differences in genotype and allele distributions between 'low' and 'high' calcium and phosphorus levels were determined using chi-square or Fisher´s exact tests. The analysis was also adjusted by sex (alpha of 5%). ESR1 rs9340799 had the less common genotype associated with higher calcium levels (p=0.03). The less common allele of ESR1 rs1884051 was associated with lower phosphorus levels (p=0.005) and there was an excess of heterozygotes for miRNA17 rs4284505 among individuals with lower calcium levels (p=0.002), both adjusted by sex. This study provides evidence that genetic polymorphisms in ESR1 and miRNA17 are involved in determining salivary calcium and phosphorus levels.


Resumo A homeostasia entre cálcio e fósforo salivares é importante para a manutenção da saúde bucal. O objetivo deste estudo foi avaliar se polimorfismos genéticos no receptor de estrógeno alfa (ESR1), receptor de estrógeno beta (ESR2) e no microRNA17 (microRNA17) estão associados com os níveis salivares de cálcio e fósforo. Saliva de 276 crianças com 12 anos de idade foi coletada com estímulo mastigatório e os níveis de cálcio e fósforo foram determinados por Espectrofotometria de Massa. O DNA genômico foi extraído da saliva remanescente e os polimorfismos genéticos em ESR1 (rs12154178, rs1884051, rs9340799 e rs2234693), em ESR2 (rs4986938 e rs1256049) e no miRNA17 (rs4284505) foram genotipados pela reação em cadeia da polimerase em tempo real utilizando sondas TaqMan. As diferenças estatísticas nas distribuições alélicas e genotípicas entre "baixo" e "alto" níveis de cálcio e fósforo foram determinadas usando os testes qui-quadrado e teste exato de Fisher. As análises foram ajustadas por sexo (alfa de 5%). O polimorfismo rs9340799 em ESR1 foi o genótipo menos comum associado com altos níveis de cálcio (p=0,03). O alelo menos comum em ESR1 rs1884051 foi associado com baixos níveis de fósforo (p=0,005) e houve um excesso de heterozigotos para miRNA17 rs4284505 entre os indivíduos com baixos níveis de cálcio salivar (p=0,002), ambos ajustados pelo sexo. Este estudo fornece evidências de que polimorfismos genéticos em ESR1 e miRNA17 estão envolvidos na determinação dos níveis de cálcio e fósforo salivares.

Abstract The present study evaluated the odontogenic potential of human dental pulp cells (HDPCs) exposed to chitosan scaffolds containing calcium aluminate (CHAlCa) associated or not with low doses of simvastatin (SV). Chitosan scaffolds received a suspension of calcium aluminate (AlCa) and were then immersed into solutions containing SV. The following groups were established: chitosan-calcium-aluminate scaffolds (CHAlCa - Control), chitosan calcium-aluminate with 0.5 µM SV (CHAlCa-SV0.5), and chitosan calcium-aluminate with 1.0 µM SV (CHAlCa-SV1.0). The morphology and composition of the scaffolds were evaluated by SEM and EDS, respectively. After 14 days of HDPCs culture on scaffolds, cell viability, adhesion and spread, mineralized matrix deposition as well as gene expression of odontogenic markers were assessed. Calcium aluminate particles were incorporated into the chitosan matrix, which exhibited regular pores homogeneously distributed throughout its structure. The selected SV dosages were biocompatible with HDPCs. Chitosan-calcium-aluminate scaffolds with 1 µM SV induced the odontoblastic phenotype in the HDPCs, which showed enhanced mineralized matrix deposition and up-regulated ALP, Col1A1, and DMP-1 expression. Therefore, one can conclude that the incorporation of calcium aluminate and simvastatin in chitosan scaffolds had a synergistic effect on HDPCs, favoring odontogenic cell differentiation and mineralized matrix deposition.


Resumo O presente estudo avaliou o potencial odontogênico de células da polpa dental humana (HDPCs) em contato com scaffolds de quitosana contendo aluminato de cálcio (CHAlCa) associado ou não à baixas dosagens de sinvastatina (SV). Scaffolds de quitosana receberam uma suspensão de aluminato de cálcio e foram imersos em soluções contendo a droga. Foram estabelecidos três grupos experimentais: scaffolds de quitosana e aluminato de cálcio (CHAlCa - controle), scaffolds de quitosana-aluminato de cálcio com 0.5 µM SV (CHAlCa-SV0.5), e quitosana-aluminato de cálcio com 1.0 µM SV (CHAlCa-SV1.0). A morfologia e composição foram avaliados por MEV e EDS, respectivamente. Após 14 dias do cultivo das HDPCs sobre os scaffolds, foram avaliados a viabilidade celular, adesão e espalhamento, deposição de matriz mineralizada e expressão gênica de marcadores odontogênicos. Observou-se que as partículas de aluminato de cálcio foram incorporadas à matriz de quitosana, a qual exibiu poros regulares distribuídos por toda sua estrutura. As dosagens selecionadas de sinvastatina foram biocompatíveis com as HDPCs. A concentração de 1 µM de SV induziu intensa expressão de fenótipo odontoblástico pelas HDPCs, demonstrando aumento da deposição de matriz mineralizada e maior expressão de ALP, Col1A1 e DMP-1. Portanto, podemos concluir que a incorporação de aluminato de cálcio e sinvastatina em scaffolds de quitosana apresentou um efeito sinérgico nas HDPCs, favorecendo a diferenciação celular e deposição de matriz mineralizada.

Abstract This study aimed to evaluate penetrability on dentinal tubule of a new bioceramic sealer through confocal laser scanning microscopy (CLSM). A specific fluorophore (Fluo-3) was mixed with the sealer. Forty distobuccal roots from maxillary molars were selected, and root canal preparation was carried out with Wave One Gold # 35.06 instruments. Roots were randomly assigned to 4 groups according to the filling procedures: Bioceramic/Lentulo (Sealer Plus BC); Bioceramic/EasyClean group, three activation of the sealer (3x20 s) with Easy Clean instrument; Bioceramic/Irrisonic: ultrasonic activation for 30 s; and AHplus/Lentulo: epoxy resin based sealer (AH Plus) was utilized with the same protocol as the BC/LE group. After 72 h, specimens were transversally sectioned at 2 and 7 mm from root apex and then analyzed through CLSM. Sealer penetration area on dentinal tubule was measured by Adobe Photoshop CC2018. Kruskal Wallis and Wilcoxon T tests were carried out. Penetrability results were similar for both sealers regardless of which technique was performed to activate them inside the root canal (p>0.05). It is reasonable to conclude that penetration of bioceramic and epoxy resin based sealers occurred unimpressively. The type of instrument used to activate bioceramic sealer did not affect penetrability. Fluo 3 should be recommended as the fluorophore to evaluate dentinal tubule penetration of bioceramic sealers.


Resumo Este estudo objetivou avaliar a penetração nos túbulos dentinários de um novo cimento biocerâmico utilizando microscopia confocal de varredura a laser (MCVL). Um fluoróforo específico (Fluo-3) foi misturado com o cimento. Quarenta raízes distovestibulares de molares superiores foram selecionados e o preparo do canal radicular foi realizado com instrumentos Wave One Gold #35.06. As raízes foram divididas randomicamente em quatro grupos de acordo com os procedimentos obturadores: Bioceramic/Lentulo: cimento biocerâmico (Sealer Plus BC); Bioceramic/EasyClean: três ativações do cimento com instrumento Easy Clean (3 x 20 s); Bioceramic/Irrisonic: ativação ultrassônica do cimento por 30 s e AHplus/Lentulo: cimento à base de resina epóxica (AH Plus) foi utilizado com o mesmo protocolo que o grupo Bioceramic/Lentulo. Após 72h, os espécimes foram seccionados transversalmente em 2 e 7 mm do ápice radicular e analisados com MCVL. A área de penetração nos túbulos dentinários foi mensurada com Adobe Photoshop CC2018. O teste de Kruskal Wallis e T de Wilcoxon foram realizados. Os resultados de penetração foram similares para ambos os cimentos independentemente de qual técnica foi utilizada para ativá-los no interior do canal radicular (p>0,05). É razoável concluir que a penetração de cimentos biocerâmicos e à base de resina epóxica ocorreram de forma pouco expressiva. O tipo de ativação do cimento biocerâmcio não afeta sua penetrabilidade nos túbulos dentinários. Fluo 3 deve ser recomendado como o fluoróforo para avaliar a penetração intratubular de cimentos biocerâmicos.

Abstract The aim of this study was to compare the effects of a single application of a new calcium mesoporous silica nanoparticle (Ca2+-MSN) versus other calcium and/or fluoride products against dental erosion. Enamel blocks were half-covered and assigned to six groups (n = 10): Ca2+-MSNs; casein phosphopeptide-amorphous calcium phosphate mousse (CPP-ACP); CPP-ACP/F− (900 ppm F−); titanium tetrafluoride (TiF4 1%); sodium fluoride (NaF 1.36%); and Milli-Q® water (negative control). A single application for each product was completed on the exposed areas of the blocks and were submitted to an erosive challenge. Differences in volumetric roughness (Sa), and tooth structure loss (TSL) by use of three-dimensional noncontact optical profilometry were evaluate. Scanning electron microscopy (SEM) was performed. Analysis of variance and Tukey's test for Sa and the Kruskal-Wallis and Mann-Whitney U tests (p< 0.05) for TSL, respectively. Results: When evaluating Sa, all products presented differences in roughness when compared with the control group (p< 0.05) but not with each other (p > 0.05). However, when analyzing the TSL, it was observed that Ca2+-MSNs, TiF4, and NaF were more effective in preventing dental erosion versus CPP-ACP, CPP-ACP/F−, and Milli-Q® water (p< 0.05). In the SEM images, the negative control presented the worst loss of dental structure, with more porous enamel. Ca2+-MSNs were as effective as TiF4 and NaF to reduce the tooth structure loss.


Resumo O objetivo deste estudo foi comparar os efeitos de uma única aplicação de uma nova nanopartícula de sílica mesoporosa de cálcio (Ca2+ -MSN) versus outros produtos à base de cálcio e / ou fluoreto contra a erosão dentária. Blocos de esmalte foram parcialmente cobertos e distribuídos em seis grupos (n = 10): Ca2+ -MSNs; fosfopeptídeos de caseína/fosfato de cálcio amorfo (CPP-ACP); CPP-ACP / F- (900 ppm F-); tetrafluoreto de titânio (TiF4 1%); fluoreto de sódio (NaF 1,36%); e água Milli-Q® (controle negativo). Uma única aplicação para cada produto foi realizada nas áreas expostas dos blocos e submetida a desafio erosivo. Diferenças na rugosidade volumétrica (Sa) e na perda de estrutura dentária (TSL) por meio de perfilometria tridimensional de não contato foram avaliadas. Microscopia eletrônica de varredura (MEV) foi realizada. Foram realizadas análise de variância e teste de Tukey para os testes Sa e Kruskal-Wallis e Mann-Whitney (p<0,05) para TSL, respectivamente. Na avaliação de Sa, todos os produtos apresentaram diferenças de rugosidade quando comparados ao grupo controle (p<0,05), mas não entre si (p> 0,05). No entanto, ao analisar o TSL, observou-se que Ca2+ -MSNs, TiF4 e NaF foram mais eficazes na prevenção da erosão dental versus CPP-ACP, CPP-ACP / F- e Milli-Q® (p<0,05). Nas imagens de MEV, o controle negativo apresentou a pior perda de estrutura dentária, com o esmalte mais poroso. A Ca2+ -MSNs foi tão eficaz quanto o TiF4 e o NaF para reduzir a perda da estrutura dentária.

Introdução: Os distúrbios alimentares são transtornos psiquiátricos graves, com altos níveis de mortalidade, incapacidade e morbidade física e psicológica; o que provoca redução na qualidade de vida. Objetivo: O presente trabalho teve como objetivos realizar uma revisão sistemática com base na análise de publicações nacionais e internacionais presentes na literatura científica sobre a ocorrência de manifestações bucais em pacientes com anorexia e bulimia nervosa; comparar a ocorrência de cárie, erosão dentária e o perfil bioquímico salivar de mulheres com e sem transtornos alimentares; além de relatar a condição de saúde bucal de uma paciente com anorexia nervosa internada em um hospital de psiquiatria pertencente à Faculdade de Medicina de Marília (FAMEMA), São Paulo, Brasil. Metodologia: A tese foi dividida em três partes. No primeiro momento da pesquisa, realizou-se vasto levantamento bibliográfico para a elaboração do primeiro capítulo, que consistiu em uma revisão sistemática sobre o assunto. O segundo capítulo foi composto por um estudo observacional do tipo casocontrole. Realizou-se a coleta de dados com os pacientes diagnosticados com bulimia e anorexia nervosa e que estavam sob tratamento no ambulatório. O grupo controle foi composto por acompanhantes dos doentes, que possuíam características similares. O terceiro capítulo foi composto por um caso clínico de uma paciente que estava hospitalizada com anorexia nervosa no Hospital das Clínicas da FAMEMA (Unidade I), onde foi realizada toda a descrição de suas condições de saúde bucal. Resultados: No capítulo 1, foi obtido um número final de seis trabalhos, onde todos respondiam a questão sobre quais manifestações bucais são encontradas em pacientes com anorexia e bulimia nervosa, e apresentavam validade e poder em sua estrutura metodológica e resultados. No segundo capítulo, podemos observar que, o grupo das pacientes com anorexia e bulimia nervosa apresenta maiores índices de cárie e erosão dentária, assim como alterações nos parâmetros salivares, em especial o fluxo salivar e os marcadores bioquímicos fósforo, cálcio, FRAP e ácido úrico; quando comparados ao grupo controle. E por fim, na terceira parte da presente tese, observamos que a paciente apresentava 11 dentes atacados pela cárie dentária. No exame periodontal, todos os dentes sondados apresentaram sangramento, além de cálculo nos incisivos inferiores e molares superiores pela face lingual; e o fluxo salivar da paciente encontrava-se extremamente reduzido. Quanto aos parâmetros salivares, o valor encontrado para o cálcio e fósforo estavam aumentados, 13,36mg/dL e 6,31mg/dL, respectivamente. Conclusão: Dessa forma, conclui-se que pacientes diagnosticados com anorexia e bulimia nervosa normalmente apresentam lesões cariosas e erosivas nos dentes e gengivas, além de alterações nos parâmetros bioquímicos salivares(AU)


Introduction: Eating disorders are serious psychiatric disorders, with high levels of mortality, disability and physical and psychological morbidity; which causes a reduction in quality of life. Objective: The present study aimed to carry out a systematic review based on the analysis of national and international publications present in the scientific literature on the occurrence of oral manifestations in patients with anorexia and bulimia nervosa; to compare the occurrence of caries, dental erosion and the biochemical salivary profile of women with and without eating disorders; in addition to reporting the oral health condition of a patient with anorexia nervosa admitted to a psychiatric hospital belonging to the Faculty of Medicine of Marília (FAMEMA), São Paulo, Brazil. Methodology: The thesis was divided into three parts. In the first moment of the research, a vast bibliographic survey was carried out for the elaboration of the first chapter, which consisted of a systematic review on the subject. The second chapter consisted of an observational case-control study. Data collection was performed with patients diagnosed with bulimia and anorexia nervosa and who were under treatment at the outpatient clinic. The control group was composed of patients' companions, who had similar characteristics. The third chapter consisted of a clinical case of a patient who was hospitalized with anorexia nervosa at Hospital das Clínicas da FAMEMA (Unit I), where the entire description of her oral health conditions was carried out. Results: In chapter 1, a final number of six studies was obtained, in which all answered the question about which oral manifestations are found in patients with anorexia and bulimia nervosa, and presented validity and power in their methodological structure and results. In the second chapter, we can see that the group of patients with anorexia and bulimia nervosa has higher rates of caries and dental erosion, as well as changes in salivary parameters, especially the salivary flow and the biochemical markers phosphorus, calcium, FRAP and uric acid ; when compared to the control group. Finally, in the third part of this thesis, we observed that the patient had 11 teeth attacked by dental caries. In the periodontal examination, all probed teeth showed bleeding, in addition to calculus in the lower incisors and upper molars through the lingual surface; and the patient's salivary flow was extremely reduced. As for salivary parameters, the values found for calcium and phosphorus were increased, 13.36mg/dL and 6.31mg/dL, respectively. Conclusion: Thus, it is concluded that patients diagnosed with anorexia and bulimia nervosa usually have carious and erosive lesions on the teeth and gums, in addition to changes in the salivary biochemical parameters(AU)

Abstract The aim of the present transversal study was to evaluate the clinical and biochemical salivary parameters of children with and without erosive tooth wear (ETW). The study population was children aged 4 to 9 years. A trained and calibrated examiner (kappa value for intraexaminer reliability = 0.89) classified the children into ETW (n = 24) and control groups (n = 24), and applied the O'Brien index. The salivary flow rate was initially evaluated by stimulated sialometry (paraffin chewing). Afterwards, the collected saliva was submitted to biochemical analyses of pH, uric acid, total buffering capacity, ferric-reducing antioxidant power, reduced glutathione, calcium, and phosphorus. Among the ETW children, 20 (83%) had dental lesions restricted to enamel, and 4 (17%) presented lesions affecting both enamel and dentin. A statistically significant difference between the groups was obtained only for the pH values (t-test; p = 0.004), with averages of 7.31 and 7.56 for the control and the ETW groups, respectively. Considering the parameters evaluated in general, it is suggested that the salivary profile of children with ETW does not differ considerably from that of children without ETW. However, the pH mean value seems to be slightly higher in ETW children, but is still within the normal physiological range.

Abstract Objective There is increasingly common the consumption more times a day of foods and acidic drinks in the diet of the population. The present study aimed to evaluate and compare the effects of a calcium mesoporous silica nanoparticle single application of other calcium and/or fluoride products in reducing the progression of dental erosion. Methodology Half of the eroded area was covered of 60 blocks of enamel, after which the block was submitted to the following treatments: (Ca2+-MSN), casein phosphopeptide-amorphous calcium phosphate (CPP-ACP); CPP-ACP/F-(900 ppm F−); titanium tetrafluoride (TiF4 1%) (positive control); sodium fluoride (NaF 1.36%) (positive control); and Milli-Q® water (negative control) before being submitted to a second erosive challenge. A surface analysis was performed via a three-dimensional (3D) noncontact optical profilometry to assess the volumetric roughness (Sa) and tooth structure loss (TSL) and and through scanning electron microscopy (MEV). An analysis of variance (ANOVA) and Tukey's test were performed. Results Regarding Sa, all experimental groups exhibited less roughness than the control (p<0.05). The TSL analysis revealed that the Ca2+-MSN and NaF groups were similar (p>0.05) and more effective in minimizing tooth loss compared with the other groups (p<0.05). Conclusions The Ca2+-MSN and NaF treatments were superior compared with the others and the negative control.

Abstract Objective To investigate the effects of intro-oral injection of parathyroid hormone (PTH) on tooth extraction wound healing in hyperglycemic rats. Methodology 60 male Sprague-Dawley rats were randomly divided into the normal group (n=30) and DM group (n=30). Type 1 diabetes mellitus (DM) was induced by streptozotocin. After extracting the left first molar of all rats, each group was further divided into 3 subgroups (n=10 per subgroup), receiving the administration of intermittent PTH, continuous PTH and saline (control), respectively. The intermittent-PTH group received intra-oral injection of PTH three times per week for two weeks. A thermosensitive controlled-release hydrogel was synthesized for continuous-PTH administration. The serum chemistry was determined to evaluate the systemic condition. All animals were sacrificed after 14 days. Micro-computed tomography (Micro-CT) and histological analyses were used to evaluate the healing of extraction sockets. Results The level of serum glucose in the DM groups was significantly higher than that in the non-DM groups (p<0.05); the level of serum calcium was similar in all groups (p>0.05). Micro-CT analysis showed that the DM group had a significantly lower alveolar bone trabecular number (Tb.N) and higher trabecular separation (Tb.Sp) than the normal group (p<0.05). The histological analyses showed that no significant difference in the amount of new bone (hard tissue) formation was found between the PTH and non-PTH groups (p>0.05). Conclusions Bone formation in the extraction socket of the type 1 diabetic rats was reduced. PTH did not improve the healing of hard and soft tissues. The different PTH administration regimes (continuous vs. intermittent) had similar effect on tissue healing. These results demonstrated that the metabolic characteristics of the hyperglycemic rats produced a condition that was unable to respond to PTH treatment.

O objetivo deste estudo foi avaliar a influência das suplementações de ômega-3 e melatonina sobre a resposta tecidual de cimentos endodônticos. Foram utilizados 72 ratos Wistar alocados em 3 grupos principais: ratos controle tratados com água (TA), ratos tratados com ômega-3 (TO) e ratos tratados com melatonina (TM). Em cada rato foram implantados 4 tubos de polietileno, sendo 1 tubo vazio para controle, e os demais contendo um dos cimentos endodônticos: Sealapex®, AH Plus® e Endofil®. Os períodos de avaliação foram de 5, 15 e 30 dias (n=8/tempo). Após eutanásia, os tubos foram removidos e processados para análise microscópica. A inflamação foi avaliada em coloração de H.E. e técnica imunohistoquímica para IL-6 e TNF-α, o colágeno pela coloração de Picrosírios red e a deposição de cálcio, pela técnica de von Kossa e luz polarizada. A estatística foi realizada de acordo com cada tipo de análise, com nível de significância de 5% (p <0,05) e considerando cada tipo de suplementação comparado ao controle. Podese observar infiltrado inflamatório reduzido com o tempo e de menor intensidade nos grupos TO ou TM comparado a TA, assim como a imunomarcação para IL-6 e TNF-α. O cimento Sealapex induziu menor inflamação aos 30 dias, tanto em TO quanto em TM. Em relação ao colágeno, foi observado maior expressão de fibras imaturas no grupo TO aos 15 e 30 dias, entretanto, no grupo TM, foi observado maior quantidade de fibras maduras aos 5 dias, ambos comparados a TA. O cimento Sealapex foi único a apresentar marcação positiva para cálcio e cristais de calcita birrefringentes à luz polarizada, independente das suplementações. Pode-se concluir que a suplementação com ômega-3 ou melatonina modularam a inflamação, diminuindo o infiltrado inflamatório e a imunomarcação de IL-6 e TNF- α, estimularam o reparo tecidual e não influenciaram no processo de deposição de cálcio(AU)


The aim of this study was to evaluate the influence of omega-3 and melatonin supplements on the tissue response of endodontic sealers. 72 Wistar rats were allocated in 3 main groups: control rats treated with water (TW), rats treated with omega-3 (TO) and rats treated with melatonin (TM). In each rat, 4 polyethylene tubes were implanted, 1 empty tube for control, and the others containing one of the endodontic sealers: Sealapex®, AH Plus® and Endofil®. The evaluation periods were of 5, 15 and 30 days (n = 8 / time). After euthanasia, the tubes were removed and processed for microscopic analysis. Inflammation was evaluated in H.E. staining and immunohistochemical technique for IL-6 and TNF-α, collagen by Picrosírios red staining and calcium deposition by von Kossa and polarized light technique. Statistics were performed according to each type of analysis, with a significance level of 5% (p <0.05) and considering each type of supplementation compared to the control. It is possible to observe a reduced inflammatory infiltrate over time and less intensity in the TO or TM groups compared to TW, as well as the immunolabeling for IL-6 and TNF-α. Sealapex sealer induced less inflammation at 30 days, both in TO and TM. Regarding collagen, a greater expression of immature fibers was observed in the TO group at 15 and 30 days, however, in the TM group, a greater amount of mature fibers was observed at 5 days, both compared to TW. Sealapex sealer was the only one to present positive marking for calcium and birefringent calcite crystals under polarized light, regardless of supplementation. It can be concluded that supplementation with omega-3 or melatonin modulated the inflammation, reducing the inflammatory infiltrate and the immunolabeling of IL-6 and TNF-α, they stimulated tissue repair and did not influence the calcium deposition process(AU)

Abstract Sources of calcium and phosphate have been added to dental restorative materials to improve their anticaries effect. Objective This study evaluated the effect of adding calcium glycerophosphate (CaGP) to resin-modified glass ionomer cement (RMGIC) on the physico-mechanical properties, ion release, and enamel demineralization. Material and Methods: Specimens were fabricated for each experimental group: RMGIC without CaGP (Control), RMGIC with 1, 3 and 9% CaGP. To determine the release of fluoride (F), calcium (Ca) and phosphorus (P), six specimens were immersed in demineralization and remineralization solutions for 15 days. In another experimental trial, the following physico-mechanical properties were evaluated at time intervals of 1 and 7 days after fabrication: compressive strength (n=12), diametral tensile strength (n=12), surface hardness of material (n=6) and the degree of conversion of monomers (n=8). To study enamel demineralization, specimens (n=12) were attached to enamel blocks and submitted to pH-cycling. Subsequently, surface and cross-sectional hardness and the concentration of F, Ca and P in enamel were determined. Results The addition of CaGP to RMGIC led to higher mean release of F, Ca and P when compared with control (p<0.001). Mechanical properties were within the range of those of the ionomer cements after addition of 1% and 3% CaGP. The degree of conversion did not differ between groups at the 1st and the 7th day (p>0.439). The addition of 3% and 9% CaGP reduced mineral loss and increased F, Ca and P in the enamel when compared with control (p<0.05). Conclusion The addition of 3% CaGP in RMGIC increased the release of F, P and Ca, reduced enamel demineralization, and maintained the physico-mechanical properties within the parameters for this material.

Objective: To examine the effect of aloe vera that containing bioactive materials on the levels of calcium (Ca) and phosphorus (P) minerals and their ratios around the immediate implanted in alveolar bone. Material and Methods: Research method by conducting experimental test on experimental animals: 9 male mongrel dogs are divided into 3 groups each 3 tails. In each animal was pulled the second premolar teeth on right side and left side, and immediately inserted titanium implant (3mm x 10mm diameter) after the socket filled with 10% aloevera extract on the right side and control on the left side. Analysis of calcium and phosphorus minerals content formed by examination of Energy Dispersive Spectroscopy (EDS) on Scanning Electron Microscope on implant and around bone tissue was done on days 14, 28 and 56. The result of statistical analysis using repeated ANOVA with independent t test. The level of significance was set at 5%. Results: There was a significant difference on calcium level between control and treatment groups on days 14, 28 and 56 (p<0.05). There was no significant difference on phosphorus level between control and treatment groups (p>0.05). The ratio of Ca / P in both control and treatment groups was also significantly different in every observation day (p<0.05). Conclusion: The addition of aloe vera extract that containing bioactive materials has an effect on increasing levels of mineral elements calcium and calciumphosphorus ratio after immediate implant insertion.

O objetivo deste estudo foi avaliar in vitro a energia livre de superfície (Æ´S) da dentina após tratamento com trimetafosfato de sódio (TMP) associado ou não ao fluoreto (F), exposto ou não ao cálcio (Ca), bem como, a adsorção de TMP, F e Ca na dentina. Blocos de dentina bovino (4 mm x 4 mm, n=12 blocos/grupo) foram tratados por meio da imersão em soluções contendo TMP nas concentrações: 0%; 1%; 3% e 9% seguido da aplicação ou não de Ca. Estas concentrações de TMP foram associadas ou não ao F na concentração de 1100 ppm F. Foram determinadas as concentrações de F, Ca e TMP nas soluções antes e após o tratamento para calcular a adsorção destes a dentina. Para analisar a energia livre de superfície (Æ´S) da dentina, os componentes apolar (Æ´S LW) e polar (Æ´S AB) eram determinados após os tratamentos por meio da medida do ângulo de contato. Os dados foram submetidos à análise de variância a dois critérios seguidos pelo teste Student-Newman-Keuls (p<0,05). O TMP reduziu a Æ´S da dentina e aumentou os sítios doadores de elétrons (Æ´S- ). Houve correlação positiva entre a adsorção de TMP e os valores de Æ´S- (Pearson's r=0,801; p<0,001). Maiores valores de Æ´S- levou a maior adsorção de Ca (p< 0,001). A associação F/TMP não alterou a Æ´S e Æ´S LW e reduziu os valores de Æ´S-, entretanto houve maior adsorção de Ca. Houve correlação positiva entre a adsorção de TMP e F (Pearson's r=0,871; p< 0,001). Concluiu-se que o TMP aumentou a Æ´S- e adsorção de Ca, e reduziu a Æ´S. A associação com o F aumentou a adsorção de TMP sem aumentar a Æ´S-, porém houve maior adsorção de Ca(AU)


The objective of this study was to evaluate in vitro the free surface energy (Æ´S) of dentin after treatment with sodium trimetaphosphate (TMP) associated or not to fluoride (F), exposed or not to calcium (Ca), as well as the adsorption of TMP, F and Ca in dentin. Bovine dentin blocks (4 mm x 4 mm, n=12 blocks/group) were treated by immersion in solutions containing TMP at concentrations: 0%; 1%; 3% and 9% followed or not by the application of Ca. These concentrations of TMP were or not associated with F at the concentration of 1100 ppm F. Concentrations of F, Ca and TMP were determined in the solutions before and after the treatment to calculate the adsorption to the dentin. To analyze the free surface energy (Æ´S) of the dentin, the apolar (Æ´S LW) and polar (Æ´S AB) components were determined after the treatments by means of the contact angle measurement. Data were submitted to analysis of variance at two criteria followed by the Student-Newman-Keuls test (p< 0.05). The results showed that TMP reduces Æ´S of dentin and increases electron donor sites (Æ´S- ). There was a positive correlation between the adsorption of TMP and the values of Æ´S- (Pearson's r=0.801; p< 0.001). Higher values of Æ´S- led to higher adsorption of Ca (p< 0.001). The F/TMP association did not change Æ´S and Æ´S LW and reduced the values of Æ´S- , but the adsorption of calcium was higher. There was a positive correlation between the adsorption of TMP and F (Pearson's r=0.871, p< 0.001). TMP increased Æ´S- and Ca adsorption, and reduced Æ´S. The association with F increased the adsorption of TMP without increasing Æ´S- , however there was higher adsorption of Ca(AU)

O presente estudo teve por objetivo verificar o efeito do trimetafosfato de sódio (TMP), associado ou não ao fluoreto (F), sobre células cultiváveis, biomassa total, atividade metabólica e composição da matriz extracelular de biofilmes mistos de S. mutans e C. albicans, bem como sobre as concentrações de F, cálcio (Ca) e fósforo (P) (biofilme total e fluido do biofilme) e no pH destes biofilmes formados in vitro. Para ambos os estudos, os biofilmes foram formados em poços de placas de microtitulação, colocando uma suspenção (1x107 células/mL C. albicans + 1x108 células/mL S. mutans) em saliva artificial suplementada com sacarose (0,4%), a qual tinha metade de seu conteúdo renovada a cada 24 horas. Os biofilmes foram tratados três vezes (72, 78 e 96 horas de formação), por um minuto, com soluções contendo TMP (0,25, 0,5 ou 1%) com ou sem 500 ppm F, além de soluções contendo 500 e 1100 ppm F, adotadas como controles positivos. A saliva artificial foi utilizada como tratamento e considerada como controle negativo. Para o estudo mircrobiológico, após o terceiro tratamento foram realizados os testes de quantificação de células cultiváveis (CFU), biomassa total (teste colorimétrico de cristal violeta ­ CV), atividade metabólica (redução de XTT) e quantificação dos componentes da matriz extracelular (proteína, carboidrato e DNA). Todos os ensaios foram realizados em triplicata, em três ocasiões diferentes. Os resultados foram submetidos à análise de variância a um critério, seguida pelo teste Fisher LSD (p<0,05). O TMP apresentou efeito redutor principalmente no metabolismo e nos0020 componentes da matriz extracelular do biofilme. Para o estudo da concentração de F, Ca, e P, após o período de tratamento, estes foram analisados no biofilme total e no fluido do biofilme após a mensuração do pH do biofilme. Em outro conjunto de experimentos, após o terceiro tratamento (96 h de formação de biofilme) o biofilme foi exposto, por 3 minutos, à solução de sacarose a 20%. Esta foi removida e, após 1 minuto, analisou-se o pH do meio e as concentrações de F, Ca, e P tanto na biomassa como no fluido do biofilme. Os dados foram submetidos a análise de variância a dois critérios, seguida pelo teste de Fisher LSD (p<0,05). O tratamento com TMP aumentou a concentração de F e P no fluido do biofilme, além de manter o pH do meio mais próximo do neutro, mesmo após a exposição do biofilme à sacarose. Assim, é possível concluir que o TMP interfere no metabolismo, composição orgânica e inorgânica, bem como no pH do biofilme testado(AU)


The aim of the present study was to verify the effect of sodium trimetaphosphate (TMP), associated or not to fluoride (F), on cultivable cells, total biomass, metabolic activity and composition of the extracellular matrix of mixed biofilms of S. mutans and C. albicans, as well as on the concentrations of F, calcium (Ca) and phosphorus (P) (total biofilm and biofilm fluid) and pH of these biofilms formed in vitro. For both studies, the biofilms were formed in wells of microtiter plates by placing a suspension (1 x 107 cells/mL C. albicans + 1x108 cells/mL S. mutans) in artificial saliva supplemented with sucrose (0,4%), which had half of its content renewed every 24 hours. Biofilms were treated three times (72, 78 and 96 hours of formation), for one minute, with solutions containing TMP (0.25, 0.5 or 1%) with or without 500 ppm F, as well as solutions containing 500 and 1100 ppm F, adopted as positive controls. Artificial saliva was used as treatment and considered as the negative control. For the microbiological study, the following tests were performed: quantification of cultivable cells (CFU), total biomass (colorimetric crystal violet test - CV), metabolic activity (XTT reduction) and quantification of matrix components (protein, carbohydrate and DNA). All assays were performed in triplicate on three different occasions. The results were submitted to one-way analysis of variance, followed by the Fisher LSD?s test (p<0.05). TMP showed a reducing effect mainly on the metabolism and components of the extracellular matrix of the biofilm. For the study of the concentrations of F, Ca, and P, these ions were analyzed in the total biofilm and in the biofilm fluid after treatment with the test solutions and after the pH measurement of the biofilm. In another set of experiments, after the third treatment (96 h of biofilm formation), the biofilms were exposed for 3 minutes to a 20% sucrose solution. This was removed and after 1 minute the biofilms were collected, and the pH of the medium and F, Ca, and P concentrations were determined both in the biomass and in the biofilm fluid. The data were submitted by two-way analysis of variance, followed by Fisher LSD's test (p<0.05). Treatment with TMP increased F and P concentration of the biofilm fluid, and maintained the pH of the medium close to neutral values even after exposure of the biofilm to sucrose. Thus, it is possible to conclude that TMP interferes with the metabolism, organic and inorganic composition and the pH of the biofilm tested(AU)

Abstract Objectives To analyze the association between periodontal conditions and inflammation, nutritional status and calcium-phosphate metabolism disorders in hemodialysis (HD) patients. Material and Methods We analyzed 128 HD patients divided into two groups: dentate (n = 103) and edentulous (n=25). The following items were assessed: baseline characteristics, age at the start and duration of HD, biochemical data: C-reactive protein (CRP), serum albumin, calcium, phosphorus, alkaline phosphatase, parathormone. A single dentist performed a complete dental/periodontal examination, including parameters of oral hygiene and gingival bleeding. Results One person had healthy periodontium, 62.14% of the patients had gingivitis, and 36.9% had moderate or severe periodontitis. The age at HD onset had a positive impact on periodontal status and negatively correlated with the number of teeth. A positive correlation between age and CRP level and negative correlations between age and serum albumin and phosphorus were found. Pocket depth (PD) was negatively correlated with serum albumin. The number of teeth was negatively correlated with serum CRP. Conclusions High prevalence and severity of periodontal disease are observed in hemodialysis patients. There is a high probability that periodontal disease may be present at the early stages of chronic kidney disease (CKD) before the hemodialysis onset.

Abstract High concentrations of hydrogen peroxide can cause adverse effects on composition and structure of teeth. However, the addition of calcium and fluoride in bleaching agents may reduce enamel demineralization. Objective: To evaluate chemical changes of sound and demineralized enamels submitted to high concentrations of hydrogen peroxide containing fluoride (F) or calcium (Ca). Material and Methods: Enamel blocks of bovine incisors with standard dimensions were obtained and half of them were submitted to pH-cycling to promote initial enamel caries lesions. Sound and demineralized enamel samples were divided into (n=10): (C) Control (no whitening treatment); (HP) 35% hydrogen peroxide; and two experimental groups: (HPF) 35% HP+0.2% F and (HPC) 35% HP+0.2% Ca. Experimental groups were submitted to two in-office bleaching sessions and agents were applied 3 times for 15 min to each session. The control group was kept in remineralizing solution at 37°C during the bleaching treatment. The surface mineral content of sound and demineralized enamels was determined through Fourier Transform Raman spectroscopy (FT-Raman), Energy dispersive Micro X-ray fluorescence spectroscopy (μ-EDXRF); and the subsurface, through cross-sectional microhardness (CSMH). In addition, polarized light microscopy (PLM) images of enamel subsurface were observed. Results: According to three-way (FT-Raman and μ-EDXRF analyses) or two-way analysis of variance (ANOVA) (CSMH) and Tukey test (α=5%), the calcium or fluoride added to high-concentrated bleaching agents increased phosphate and carbonate concentrations on sound and demineralized enamels (p<0.05). However, HPC and HPF were unable to completely reverse the subsurface mineral loss promoted by bleaching on sound and demineralized enamels. The calcium/ phosphate (Ca/P) ratio of sound enamel decreased after HP treatment (p<0.001). Conclusion: Even though experimental bleaching agents with Ca or F reduced mineral loss for both sound and demineralized enamel surfaces, these agents were unable to reverse the enamel subsurface demineralization.
