RESUMO

ABSTRACT Objective: To assess the efficacy of bioactive glass, self-assembling peptide, and ozone-remineralizing agents on the artificial carious lesion. Material and Methods: On the extracted 60 premolar teeth, an artificial carious lesion/demineralization was created. Later, the remineralization of demineralized teeth was done with respective remineralizing agents (Group A: Calcium sodium phosphosilicate (bioactive glass), Group B: Self-assembling peptide, Group C: Ozone remineralizing agents and Group D (Control): De ionized water. The degree of demineralization and remineralization were evaluated using the Vickers Hardness Number. Results: There was a decrease in microhardness from baseline to demineralization in all the groups, and this reduction was found to be statistically considerable. After the remineralization of demineralized samples with respective remineralizing agents, there was an increase in microhardness of 312.38, 276.67, and 254.42 in groups A, B, and C, respectively. In contrast, in Group D, there were no changes. Conclusion: Bioactive glass and self-assembling peptides had higher remineralizing capacities, which can be used to treat early carious lesions.

RESUMO

Abstract Objective: This study evaluated the effects of local vitamin C treatment on tissue advanced glycation end products (AGE), interleukin (IL)-6, 8-hydroxy-2-deoxyguanosine (8-OHdG), and matrix metalloproteinases (MMP)-8 in tissues; serum C-terminal telopeptide fragments (CTX); and alveolar bone loss (ABL) in rats. Methodology: 35 male Sprague Dawley rats were divided equally into five groups: 1) control (C), 2) experimental periodontitis (P), 3) experimental diabetes (D), 4) experimental diabetes and experimental periodontitis (D + P), and 5) experimental diabetes-experimental periodontitis-locally applied vitamin C (D + P + LvitC). Diabetes was induced in rats with alloxan monohydrate, after which periodontitis was induced by ligature placement in the right mandibular first molar teeth for 11 days. In the treatment group, vitamin C was administered locally three times with two-days interval after ligature removal. The animals were sacrificed, and the samples were analyzed histometrically and immunohistochemically. Results: CTX, 8-OHdG, and AGE values significantly decreased in the treatment group compared to the D + P group. IL-6 and MMP-8 values decreased in the treatment group compared to the D + P group, but this is not significant. ABL was significantly reduced by the local delivery of vitamin C. Conclusion: This study reveals that vitamin C treatment may be beneficial to reduce serum CTX and gingival MMP-8 levels, oxidative stress, inflammation, and AGE accumulation in periodontal tissue. Vitamin C may be an immunomodulator and antioxidant locally applied in the treatment of periodontitis to reduce the adverse effects of diabetes in periodontal tissues.

RESUMO

Abstract Mineralization-promoting peptides are attractive candidates for new remineralization systems. In previous studies, peptides have been applied as aqueous solutions, which is not a clinically relevant form. Objective This study aims to investigate the efficiency of a mineralization-promoting peptide, applied in varnish, on remineralizing artificial caries on primary teeth. Methodology 55 primary molars were collected. Specimens were immersed in a demineralizing solution for 7 days and then, divided into 7 groups: Baseline: No-remineralization, Placebo: Blank colophony, F: Colophony 5% fluoride, P: Colophony 10% peptide, P+F: Colophony 5% fluoride and 10% peptide, Embrace: Embrace™ varnish, Durashield: Durashield™ varnish. A mixture of 35% w/v colophony varnishes were prepared in ethanol and applied accordingly. Specimens were immersed in a remineralization solution for 4 weeks and it was evaluated using PLM and SEM. Lesion depth reduction was examined by one-way ANOVA. Results There was no significant difference in mean lesion depths between baseline (147.04 ± 10.18 µm) and placebo groups (139.73 ± 14.92 µm), between F (120.95 ± 12.23 µm) and Durashield (113.47 ± 14.36 µm) groups and between P (81.79 ± 23.15 µm) and Embrace (90.26 ± 17.72 µm) groups. Lesion depth for the P+F group (66.95±10.59 µm) was significantly higher compared to all other groups. All groups contained samples with subsurface demineralized regions. Number of subsurface demineralized regions were higher in fluoride-containing groups. Conclusions We conclude that the mineralization-promoting peptide (MPP3) is effective in this in vitro study and the peptide shows benefits over fluoride as it yields less subsurface demineralized regions.

RESUMO

Abstract The acquired enamel pellicle (AEP) is an organic film, bacteria-free, formed in vivo as a result of the selective adsorption of salivary proteins and glycoproteins to the solid surfaces exposed to the oral environment. Objective: This study aimed to compare the proteomic profile of AEP formed in situ on human and bovine enamel using a new intraoral device (Bauru in situ pellicle model - BISPM). Material and Methods: One hundred and eight samples of human and bovine enamel were prepared (4×4 mm). Nine subjects with good oral conditions wore a removable jaw appliance (BISPM) with 6 slabs of each substrate randomly allocated. The AEP was formed during the morning, for 120 minutes, and collected with an electrode filter paper soaked in 3% citric acid. This procedure was conducted in triplicate and the pellicle collected was processed for analysis by LC-ESI-MS/MS. The obtained mass spectrometry MS/MS spectra were searched against human protein database (SWISS-PROT). Results: The use of BISPM allowed the collection of enough proteins amount for proper analysis. A total of 51 proteins were found in the AEP collected from the substrates. Among them, 15 were common to both groups, 14 were exclusive of the bovine enamel, and 22 were exclusive of the human enamel. Proteins typically found in the AEP were identified, such as Histatin-1, Ig alpha-1, Ig alpha 2, Lysozyme C, Statherin and Submaxillary gland androgen-regulated protein 3B. Proteins not previously described in the AEP, such as metabolism, cell signaling, cell adhesion, cell division, transport, protein synthesis and degradation were also identified. Conclusion: These results demonstrate that the proteins typically found in the AEP appeared in both groups, regardless the substrate. The BISPM revealed to be a good device to be used in studies involving proteomic analysis of the AEP.

The immune system has several forms of defense against pathogenic microorganisms. The mucous membranes are a source of potent antimicrobial cationic peptides against a broad range of bacteria, fungi and enveloped viruses. The aim of study was evaluated the cytotoxicity and antimicrobial activity under planktonic and biofilm conditions of fragments derived from cationic peptides (CP): LL-37 (from hCAP-18), D6-17 and D1-23 (from ß-defensin-3 derivative) against cariogenic bacteria. For cytotoxicity analysis, two lines of epithelial cells were exposed to serial dilutions of the CP fragments. MTT assays and DAPI staining were performed to evaluate cell metabolism and morphology, respectively. Minimum inhibitory concentration (MIC) and minimal bactericidal concentration (MBC) were determined for the CP fragments and control (Chlorhexidine digluconate-CHX) against Streptococcus mutans (Sm), S. mitis, S. oralis, S. salivarius, S. sanguinis, Lactobacillus acidophilus, L. casei, L. rhamnosus, L. brevis, L. fermentum and Actinomyces israelii. Fractional inhibitory concentration (FIC) was obtained for the combinations of CP fragments on S. mutans. Biofilm assays were conducted with CHX and the best antimicrobial CP fragment against S. mutans strains. Confocal Laser Scanning Microscopy (CLSM) was used to analyze the Live/Dead cells and biofilm thickness. The results indicated that D6-17 did not affect the metabolism of either cell line. D1-23, LL-37 and CHX were not toxic for both cells, in concentrations below 0.2, 0.02 and 0.01mM, respectively. DAPI-staining cells demonstrated nuclei fragmentation for LL-37 group and cells with the aspect of apoptosis in the CHX group. Cells treated with D1-23 and D6-17 presented morphology similar to the control group (culture medium). D1-23 presented the best bactericidal activity against S. mutans, S. mitis and S. salivarius. LL-37 had a better effect against Lactobacillus and Actinomyces species. D6-17 showed bactericidal activity only against S. mutans, L. brevis and L. fermentum. Combinations of CP fragments did not show a synergic effect against S. mutans. D1-23 (10x MBC) presented a higher effect against S. mutans biofilm compared to CHX. CLSM analysis showed that D1-23 and CHX groups presented higher quantification of dead cells when compared to control (culture medium). The biofilm thickness were lower in the D1-23 group compared to CHX and control groups. We concluded that D1-23 showed a remarkable antimicrobial/anti-biofilm effect against cariogenic bacteria and low toxicity for epithelial cells(AU)

The antimicrobial peptides LL-37, hBD-1, hBD-2 and hBD-3 are considered an endogenous antibiotic, with important role in the prevention of periodontal diseases due to their ability to regulate the immune response. However those peptides could be degraded by periodontal pathogens. Therefore, therapies able to up regulate the secretion of those peptides by human cells, and the association of antimicrobial peptides with natural compounds, which may act in synergism to modulate the immune response, may be a novel approach for effectively controlling periodontal diseases. The aim of this in vitro study were: i) investigate the ability of green tea extract and EGCG to induce hBD-1 and hBD-2 secretion and gene expression by gingival epithelial cells (B11) and to protect hBDs from degradation by P. gingivalis, ii) A 3D co-culture model of gingival epithelial cells and fibroblasts stimulated with A. actinomycetemcomitans LPS (1 µg/ml) were used to investigated the anti-inflammatory properties of the hBD-3, LL-37, ACPACs and EGCG and to determine whether LL-37 acts in synergy with AC-PACs, EGCG and hBD-3. Gingival epithelial cells were stimulated with green tea extract or EGCG in the presence and absence of specific inhibitors. The secretion and gene expression of hBD-1 and hBD-2 was respectively measured by ELISA and qPCR. The ability of green tea extract and EGCG to prevent hBDs degradation by P. gingivalis present in a bacterial culture supernatant was evaluated by ELISA. A 3D co-culture model composed of gingival fibroblasts embedded in a collagen matrix overlaid with gingival epithelial cells had a synergistic effect with respect to the secretion of IL-6 and IL-8 in response to A. actinomycetemcomitans LPS stimulation compared to fibroblasts and epithelial cells individually. The 3D co-culture model was stimulated with noncytotoxic concentrations of: i) hBD-3 (10 and 20 µM) and LL -37 (0.1 and 0.2 µM), ii ) EGCG (1 and 5 µg/ml) and LL -37 (0.1 and 0.2 µM), iii) AC- PACs (25 and 50 µg/ml) and LL-37 (0.1 and 0.2 µM) alone and in association in the presence of A. actinomycetemcomitans LPS. A multiplex ELISA assay was used to quantify the secretion of 41 different cytokines, chemokines, growth factors and 13 different MMPs and TIMPs. Ours results showed that: i) the secretion and gene expression of hBD-1 and hBD-2 was up-regulated in a dose-dependent manner by green tea extract and EGCG. Green tea extract and EGCG were also able to prevent the degradation of recombinant hBD-1 and hBD-2 by a culture supernatant of P. gingivalis, ii) hBD-3 in association with LL-37 showed a synergistic effect to reduce the secretion of GRO- α, G-CSF, IP- 10, IL -6 and MCP -1, however had only additive effect on reducing the secretion of IL-8 in response to A. actinomycetemcomitans LPS stimulation, iii) the association of the peptide LL- 37 with EGCG and with AC- PACs showed a synergistic effect to reduce the secretion of GRO-α, G-CSF and IL-6 in response to A. actinomycetemcomitans LPS stimulation. Considering that the antimicrobial peptides hBD-3 and LL-37, as well as green tea extract, EGCG and AC- PACs, demonstrated the ability to regulate the secretion of inflammatory cytokines, up regulated the secretion of defensins by the cells and even to protect the defensins degradation by P. gingivalis, emerged as promising alternative adjunct therapy to conventional periodontal treatment. However future clinical studies are necessary to evaluate the role of peptides and natural compounds in the oral cavity and periodontal tissues
