This review aims to present the mechanisms of protein interactions with titanium dental implant surfaces. Methods: the analyses were based on searches of scientific articles available in English and Portuguese in PubMed (MEDLINE), Bireme (LILACS), Scielo, Web of Science and Google Scholar. Results: titanium dental implant treatments success rates (95-98%) are mainly due to the biocompatibility of titanium oxide on the implant surface, surgical techniques adopted, good implants manufacturing processes and biomechanical knowledge of the systems. Studies in past decades has empirically developed implant surfaces with significant changes in morphologies, roughness, wettability, surface energy, chemical composition, and chemical groups density or deposited molecules. These changes promoted better protein adsorption, osteoblast adhesion, and changes in the mechanisms involved in osseointegration. Thus, the time to put the implant in function has been reduced and the success rates have increased. In the osseointegration process, at the nanoscale, there is no contact between the bone and the implant surface, but there is the formation of a protein anchorage between the periosteum and the implant with an interface formed by proteins. In all the reactions between the body and the implant surface, the activities of fibronectin and integrin are essential, since they are responsible for transmitting information to the cell for its differentiation, adhesion and mobility. Conclusion: thus, the analyses of protein-implant interactions are indispensable for a better understanding of the performance of osseointegrated dental implants

The peri-implant ligament is formed from the interface of bone tissue, through the anchoring of proteins and the surface of the dental implant. In this sense, it is relevant to understand the extent to which this ligament is structured and biomimics the periodontal ligament functions. Aim: The goal of this scoping review is to present and analyze the peri-implant ligament composition and compare the extent to which this ligament is structured and biomimics the periodontal ligament functions. Methods: This scoping review was performed according to the Joanna Briggs Institute methodology for scoping reviews and following the Preferred Reporting Items for Systematic Reviews and Meta-analyses extension for scoping review. Two independent researchers searched Pubmed, Cochrane, Embase, Virtual Health Library, Scielo, Scopus, Web of Science, Brazilian Bibliography of Dentistry, Latin American and Caribbean Literature in Health Sciences, Digital Library of Theses and Dissertations from the University of São Paulo and Portal Capes. Studies published in English, Portuguese and Spanish, over the last 21 years (2000-2021). Results: A total of 330 titles were identified and after applying inclusion and exclusion factors, 27 studies were included in this review. All proteins were identified regarding their tissue function and classified into 6 major protein groups. After that this new protein ligament was compared with the periodontal ligament regarding its function and composition. The main proteins associated with osseointegration, and thus, with the peri-implant ligament are recognized as belonging to the periodontal ligament. Conclusion: This scoping review results suggest evidence of the composition and function of the periimplant ligament. However, variations may still exist due to the existence of several modulants of the osseointegration process

Abstract Objective: To determine if protein profiles identified in saliva could be used to determine risk and severity of erosive tooth wear. Material and Methods: Three types of saliva sampling were performed to obtain saliva from 34 18-year old individuals that received regular dental check-ups, along with clinical status of the dentition and risk factor related to erosive tooth wear using the VEDE scale. Protein profiles in saliva were determined using electrophoresis and the calculation of the percentage of a specific band at a specific molecular weight in relationship to the total protein in that sample (% of total) using molecular weight standards. This quantification was repeated for each protein band across a range of molecular weights for each sample to test for association with erosive tooth wear status. Results: There were no differences in the number of detectable proteins sourced from the parotid gland, nor the unstimulated and stimulated whole saliva. Five out of the 34 individuals had no signs of erosive tooth wear despite an acidic diet and were more likely to have proteins with molecular weight smaller than 1 KDa (p=0.03). Conclusion: There is potential for the use of protein profiling to determine risks for erosive tooth wear.

Objective: The resistance of fungi and bacteria to the available antimicrobials has increased and the development of alternative products to control them has become a very requirement. The use of plant products could be a viable option due to the efficacy, viability, and availability they present. Thereby, this study evaluated the effect of R. officinalis L. extract on C. albicans and S. mutans biofilms, by the total protein level analysis presented by the microorganisms. Material and Methods: For this purpose, monomicrobial biofilms were formed for 48 h and exposed to the R. officinalis L. extract for 5 min. Then, total protein quantification was performed by Lowry method. Results: The analysis showed significant total protein reduction of the biofilms after exposure to the plant extract, with 39 ± 11%, for C. albicans, and 32 ± 11%, for S. mutans. Conclusion: R. officinalis L. extract decreased the total protein level in both biofilms. Thus, C. albicans and S. mutans protein composition could be a target for action of antimicrobial agents. Objetivo: A resistência de fungos e bactérias aos antimicrobianos disponíveis tem se elevado e o desenvolvimento de produtos alternativos para controlar micróbios tem se tornado uma necessidade real. A utilização de produtos de origem vegetal poderia ser uma opção viável, devido à eficácia, viabilidade e disponibilidade que apresentam. Sendo assim, este estudo avaliou o efeito do extrato de R. officinalis L. (alecrim) sobre biofilmes de C. albicans and S. mutans, analisando o nível de proteína total apresentada pelos micro-organismos. Material e Métodos: Para tanto, biofilmes monomicrobianos foram formados por 48 h e expostos ao extrato de R. officinalis L. por 5 min. Então, a quantificação de proteína total foi realizada por método de Lowry. Resultados: A análise demonstrou reduções significativas de proteína total de cada biofilme após exposição ao extrato, sendo 39 ± 11% no biofilme de C. albicans e 32 ± 11%, no caso de S. mutans. Conclusão: O extrato de R. officinalis L. diminuiu o nível de proteína total em ambos os biofilmes. Com isso, a composição proteica de C. albicans e S. mutans poderia ser um alvo para ação de agentes antimicrobianos. (AU)

Abstract The acquired enamel pellicle (AEP) is an organic film, bacteria-free, formed in vivo as a result of the selective adsorption of salivary proteins and glycoproteins to the solid surfaces exposed to the oral environment. Objective: This study aimed to compare the proteomic profile of AEP formed in situ on human and bovine enamel using a new intraoral device (Bauru in situ pellicle model - BISPM). Material and Methods: One hundred and eight samples of human and bovine enamel were prepared (4×4 mm). Nine subjects with good oral conditions wore a removable jaw appliance (BISPM) with 6 slabs of each substrate randomly allocated. The AEP was formed during the morning, for 120 minutes, and collected with an electrode filter paper soaked in 3% citric acid. This procedure was conducted in triplicate and the pellicle collected was processed for analysis by LC-ESI-MS/MS. The obtained mass spectrometry MS/MS spectra were searched against human protein database (SWISS-PROT). Results: The use of BISPM allowed the collection of enough proteins amount for proper analysis. A total of 51 proteins were found in the AEP collected from the substrates. Among them, 15 were common to both groups, 14 were exclusive of the bovine enamel, and 22 were exclusive of the human enamel. Proteins typically found in the AEP were identified, such as Histatin-1, Ig alpha-1, Ig alpha 2, Lysozyme C, Statherin and Submaxillary gland androgen-regulated protein 3B. Proteins not previously described in the AEP, such as metabolism, cell signaling, cell adhesion, cell division, transport, protein synthesis and degradation were also identified. Conclusion: These results demonstrate that the proteins typically found in the AEP appeared in both groups, regardless the substrate. The BISPM revealed to be a good device to be used in studies involving proteomic analysis of the AEP.

Abstract Raloxifene is an antiresorptive drug, selective estrogen receptor modulator (SERM) used in the treatment of osteoporosis. Objective To evaluate proteins related to bone repair at the peri-implant bone in a rat model of osteoporosis treated with raloxifene. Material and Methods 72 rats were divided into three groups: SHAM (healthy animals), OVX (ovariectomized animals), and RLX (ovariectomized animals treated with raloxifene). Raloxifene was administered by gavage (1 mg/kg/day). Tibial implantation was performed 30 days after ovariectomy, and animals were euthanized at 14, 42, and 60 days postoperatively. Samples were collected and analyzed by immunohistochemical reactions, molecular analysis, and microtomographic parameters. Results RLX showed intense staining of all investigated proteins at both time points except for RUNX2. These results were similar to SHAM and opposite to OVX, showing mild staining. The PCR gene expression of OC and ALP values for RLX (P<0.05) followed by SHAM and OVX groups. For BSP data, the highest expression was observed in the RLX groups and the lowest expression was observed in the OVX groups (P<0.05). For RUNX2 data, RLX and SHAM groups showed greater values compared to OVX (P<0.05). At 60 days postoperatively, microtomography parameters, related to closed porosity, showed higher values for (Po.N), (Po.V), and (Po) in RLX and SHAM groups, whereas OVX groups showed lower results (P<0.05); (BV) values (P=0.009); regarding total porosity (Po.tot), RLX group had statistically significant lower values than OVX and SHAM groups (P=0.009). Regarding the open porosity (Po.V and Po), the SHAM group presented the highest values, followed by OVX and RLX groups (P<0.05). The Structural Model Index (SMI), RLX group showed a value closer to zero than SHAM group (P<0.05). Conclusions Raloxifene had a positive effect on the expression of osteoblastogenesis/mineralization-related proteins and on micro-CT parameters related to peri-implant bone healing.

Fluoride (F) is a potent anti-cariogenic element, but only an appropriate dose is effective to have therapeutic action, else systemic toxicity may be observed. Additionally, two factors, amount of F and time of exposure, drive its action. Surprisingly, the susceptibility to toxic effects of F is genetically determined. The present study identified the effects of F on the liver proteome of mice susceptible (A/J) or resistant (129P3/J) to the effects of F. Weanling male A/J (n=6) and 129P3/J mice (n=6) were housed in pairs and assigned to three groups given low-F diet and drinking water containing 0, 15 or 50 ppm F for 7 weeks. Liver proteome profiles were examined using nano-LC-ESI-MS/MS. Protein function was classified by GO biological process (Cluego v2.0.7 + Clupedia v1.0.8). Difference in expression among the groups was determined using the PLGS software. In the control group (0 ppm F), most proteins with fold change were increased in A/J mice. Precisely the proteins related to energy flux and oxidative stress were quite significant in this context, suggesting the high susceptibility of these mice to the effects of F, since the exposure also induces oxidative stress. Treatment with the lower F concentration provoked more pronounced alterations in fold change in liver proteins in comparison to the treatment with the higher F concentration. Strikingly, most of the proteins with fold change upon following 15 ppm F treatment, were increased in the A/J mice compared with their 129P3/J counterparts, thus suggesting attempt of the former to fight against the toxic effects of F. With respect to 50 ppm F, most proteins with fold change were decreased in the A/J mice compared with their 129P3/J counterparts, especially proteins related to oxidative stress and protein folding, which might be related to the higher susceptibility of the A/J animals to the deleterious effects of F. Our findings can provide new insights into the molecular mechanisms underlying genetic susceptibility to fluorosis by indicating key protein players which need to be better addressed in future experimental studies.(AU)


O Fluoreto (F) é um potente elemento anti-cariogênico, mas é somente efetivo terapeuticamente em uma dose apropriada. Por outro lado, doses acima das recomendadas levam a toxicidade sistêmica. Em adição, dois fatores decidem sua efetividade de ação: quantidade de F e tempo de exposição. A suscetibilidade aos efeitos tóxicos do F é determinada geneticamente. O presente estudo avaliou os efeitos do F no proteoma do fígado de camundongos suscetíveis (A/J) ou resistentes (129P3/J) aos efeitos do F. Camundongos machos desmamados A/J (n=6) e 129P3/J (n=6) foram alojados em pares e divididos em três grupos tratados com ração com baixo teor de F e água contendo 0, 15, ou 50 ppm de F por 7 semanas. Perfis proteômicos do fígado foram examinados usando nano-LC-ESI-MS/MS. A função de proteínas foi classificada pelo processamento biológico GO (Cluego v2.0.7 + Clupedia v1.0.8). A diferença de expressão entre os grupos foi determinada usando o software PLGS. No grupo controle (0 ppm F), a expressão da maioria das proteínas foi aumentada nos camundongos A/J e precisamente as proteínas relacionadas ao fluxo de energia e estresse oxidativo foram significativas neste contexto, sugerindo portanto, a alta sucetibilidade destes camundongos aos efeitos do F, já que a exposição também induz o estresse oxidativo. O tratamento com baixa concentração de F provocou alterações mais pronunciadas em proteínas do fígado comparado ao tratamento com alta concentração de F. Notadamente, a maioria das proteínas encontradas no fígado dos animais tratados com 15 ppm de F foi aumentada em camundongos A/J comparados aos camundongos 129P3/J, demonstrando portanto, uma tentativa dos A/J de neutralizar os efeitos tóxicos do F. Já nos animais tratados com 50 ppm de F, a maioria das proteínas foi diminuída nos camundongos comparados aos seus pares 129P3/J, especialmente proteínas relacionadas ao estresse oxidativo e enovelamento de proteínas, o que pode estar relacionado à alta suscetibilidade dos animais A/J aos efeitos deletérios do F. Nossos achados podem fornecer novos insights que podem contribuir para a interpretação os mecanismos moleculares relacionados à suscetibilidade genética à fluorose, indicando proteínas chaves que precisam ser melhor estudadas em estudos futuros.(AU)

Na cavidade oral, qualquer superfície exposta é propensa à formação da película adquirida (PA), sendo considerada um filme orgânico, livre de bactérias que se forma in vivo como resultado da adsorção seletiva de proteínas e glicoproteínas salivares às superfícies solidas que estão expostas ao meio bucal. O objetivo deste trabalho será avaliar a influência da adição ou não de carga (vidro de bário alumina silicato e sílica) e/ou de inibidores de proteases (EGCG ou CHX) a resinas compostas experimentais no perfil proteico da PA formada sobre estes espécimes, utilizando estratégias proteômicas quantitativas livres de marcadores. Foram preparadas 324 amostras de esmalte bovino (6x6x2mm), foi feita uma cavidade no centro de 4x4mm, a qual foi preenchida com resinas experimentais. As amostras foram divididas em 6 grupos de 54 espécimes cada, de acordo com os grupos experimentais: Sem carga, sem inibidor (NF-NI); carga, sem inibidor (F-NI); sem carga e CHX (NF-CHX); carga e CHX (F-CHX); sem carga e EGCG (NF-EGCG); carga e EGCG (F-EGCG). Nove adultos jovens de ambos os gêneros paticiparam, usando um aparelho mandibular removível (BISPM - Bauru in situpellicle model) com duas amostras de cada grupo. O experimento foi conduzido por 9 dias consecutivos, durante a manhã por 120 minutos. A PA foi obtida através da ajuda do papel filtro de eletrotodo, umidecido em 3% de ácido cítrico. A película coletada, foi processada por LC-ESI-MS/MS. Os espectros MS/MS obtidos foram confrontados com bases de dados de proteínas humanas (SWISS-PROT). A quantificação livre de marcadores foi feita utilizando o software PLGS. A diferença de expressão entre os grupos foi expressa como p<0.05 para as proteínas down-regulated e 1-p>0.95 para as proteínas up-regulated. Um total de 140 proteínas foram identificadas na PA. Destas, 16 foram encontradas em comum em todos os grupos, dentre elas muitas proteínas típicas da PA, tais como, duas isoformas de Basic salivary proline-rich protein, Cystatin-S, Cystatin-AS, Cystatin-SN, Histatin-1, Ig alpha-1 chain C region, Lysozyme C, Mucin-7, Proline-rich protein 4, Protein S100- A9, Salivary acidic proline-rich phosphoprotein ½, Statherin e Submaxillary gland androgen-regulated protein 3B. O número total de proteínas identificadas em cada grupo foi 31, 51, 18, 38, 106 and 54 para NF-NI, F-NI, NF-CHX, F-CHX, NF-EGCG e F-EGCG, respectivamente. A respectiva quantidade de proteínas exclusivas de cada grupo foi 6, 14, 1, 6, 51 e 5. A maioria das proteínas que não são comumente descritas na PA e que tem funções distintas no organismos, estando envolvidas no metabolismo, sinalização celular, adesão celular, divisão celular, transporte, síntese proteica e degradação foram encontradas no grupo NF-EGCG. Estes resultados demonstram que houve uma diferença no perfil preteico da PA, devido à composição das resinas experimentais, além de oferecer informações importantes sobre o desenvolvimento de materiais restauradores com componentes que podem aumentar a proteção na cavidade oral.(AU)


In the oral cavity, any exposed surface is prone to the formation of the acquired pellicle (AP), an organic film, free of bacteria, which is formed in vivo as a result of the selective adsorption of salivary proteins and glycoproteins to the solid surfaces exposed to the oral environment. The objective of this work was to evaluate the influence of the addition or not of filler (Barium glass alumina silicate and silica) and/or protease inhibitors [epigallocatechin-3-gallate (EGCG) or chlorhexidine (CHX)] to experimental composite resins in the protein profile of the AP formed on these specimens, using quantitative label-free proteomic analysis. Three-hundred and twenty-four samples of bovine enamel (6x6x2mm) were prepared. A cavity (4x4mm) was made, filled with experimental resins and divided into 6 groups of 54 specimens each, according to the experimental groups: no filler, no inhibitor (NF-NI); filler, no inhibitor (F-NI); no filler plus CHX (NF-CHX); filler plus CHX (F-CHX); no filler plus EGCG (NF-EGCG); filler plus EGCG (F-EGCG). Nine young adults of both genders participated using a removable jaw appliance (BISPM - Bauru in situ pellicle model)) with 2 slabs of each group. The experiment was carried out in 9 consecutive days, during the morning for 120 minutes. The pellicle was obtained through the aid of electrodes filter paper moistened in 3% citric acid. The pellicles collected were processed for analysis by LC-ESI-MS/MS. The obtained MS/MS spectra were searched against human protein database (SWISS­PROT). The proteomic data related to protein quantification were analyzed using the PLGS software. Difference in expression among the groups was expressed as p<0.05 for down-regulated proteins and 1-p>0.95 for up-regulated proteins. A total of 140 proteins were identified in the AP. From these, 16 were found in all the groups, among which are many proteins typically found in the AP, such as two isoforms of Basic salivary proline-rich protein, Cystatin-S, Cystatin-AS, Cystatin-SN, Histatin-1, Ig alpha-1 chain C region, Lysozyme C, Mucin-7, Proline-rich protein 4, Protein S100-A9, Salivary acidic proline-rich phosphoprotein ½, Statherin and Submaxillary gland androgen-regulated protein 3B. The total number of proteins identified in each group was 31, 51, 18, 38, 106 and 54 for NF-NI, F-NI, NF-CHX, F-CHX, NF-EGCG and F-EGCG, respectively. The respective amount of proteins exclusively in each group was 6, 14, 1, 6, 51 and 5. Most of the proteins that are not commonly described in the AP that have distinct functions in the organism, being involved in metabolism, cell signaling, cell adhesion, cell division, transport, protein synthesis and degradation were found most prominently in the NF-EGCG group. These results demonstrate that there was a difference in the protein profile of the AP due to the composition of the experimental resins, beyond offering important information on the development of restorative materials with components that can increase the protection in the oral cavity.(AU)

A saliva é um importante meio de proteção contra danos ao esmalte e dentina, e é quando ela entra em contato com a superfície dentária, que ocorre uma adsorção seletiva de proteínas salivares, glicoproteínas e lipídeos. Esta adsorção forma um filme orgânico, que é isenta de bactérias, que quando formada sobre esmalte dentário é denominada de película adquirida do esmalte (PAE). A presença destas proteínas recobrindo os tecidos dentários auxilia na lubrificação, tem capacidades de tamponamento e de remineralização, tornando-se um importante fator de proteção contra erosão dentária. O objetivo deste trabalho foi detectar as alterações no perfil protéico na película adquirida do esmalte (PAE) formada in vivo, após a exposição ao ácido clorídrico. Os experimentos foram realizados em 12 dias consecutivos. Em cada dia, os voluntários (n=9), com idade entre 18 a 35 anos, não fumantes, e com um bom estado de saúde geral e bucal, eram submetidos a uma profilaxia dentária com pedra pomes. Depois de 3 min ou 120 min e após a formação PAE, os dentes eram isolados com rolos de algodão e submetidos a 3 procedimentos distintos, sendo um deles realizado a cada dia: aplicação de 50µL de ácido clorídrico (0,1 M, pH 1), ácido clorídrico (0,01 M, pH 2) ou água deionizada por 10 segundos. A aplicação foi feita, em todos os dentes dos arcos superiores e inferiores na face vestibular. Na sequência, a película foi removida com um papel de filtro umedecido em ácido cítrico a 3%. Este procedimento foi repetido por mais uma vez e foi feito um "pool' com os papeis de filtro obtidos dos 9 voluntários, para cada procedimento e tempo de formação (Água-3min, Água-2h, pH2-3min, pH2-2h, pH1-3min e pH1-2h). Após extração das proteínas, as mesmas foram submetidas à cromatografia líquida de fase reversa interligada a um espectrômetro de massas (nLC-ESI-MS/MS). Quantificação proteômica livre de marcadores foi feita utilizando o software (Protein Lynx Global Service software). Um total de 180 proteínas foram encontradas nas amostras de PAE. E o número de proteínas identificadas crescia conforme aumentou-se o seu tempo de formação da película. Somente 4 proteínas foram presentes em todos os grupos sendo estas isoforms de IgA, Serum albumin e Statherin. Um grande número proteínas foram identificadas como sendo únicas dos grupos tratados com HCl, depois de 2h de formação de película (~ 50 proteínas). Em conclusão as proteínas são resistentes a remoção por HCl, e tanto que Serum Albumin e Statherin, foram identificadas em películas formadas em tempos precoces. Para películas formadas no tempo de 120-min foram encontradas muitas proteínas que são resistentes a remoção por HCl. Este fato sugere um aumento da proteção contra ácidos intrínsecos conforme o tempo de formação de película, o que deverá ser avaliada em estudos futuros.(AU)


Saliva it is an important factor against enamel and dentin damages. When the saliva enter in contact with the dental surface, results in a selective adsorption of salivary proteins, glycoproteins and lipids. This adsorption formed an organic free-bacterial film, which when formed in the enamel, is denominated acquired enamel pellicle. The presence of this proteins covering the enamel tissues, has the function of lubrication, buffering and remineralization capabilities, making it an important factor against dental erosion. The objective of this study was detected changes in the protein profile of acquired enamel pellicles (AEP) formed in vivo for different times, after application of hydrochloric acid (HCl). The experimental was realized in 12 consecutive days. On each day, nine subjects, (aged 18 to 35 years, non-smokers, with good general and oral health) were submitted to dental prophylaxis with pumice. After 3 or 120 min, time of formation of the acquired pellicle, the teeth were isolated with cotton rolls and, submitted for a 3 different procedures, one procedure of each day, 50 µL of 0.1 M HCl (pH = 1.0), 0.01 M HCl (pH = 2.0) or deionized water were applied on the buccal surface of the teeth for 10 s. The application of HCl was in all teethes from the superior and lower arch, in vestibular surface. In sequence the AEP was collected using an electrode filter paper pre-soaked in 3% citric acid. This procedures was repeted for one more day. After protein extraction, the samples were submitted to reverse phase liquid chromatography coupled to mass spectrometry (nLC-ESI-MS/MS). Label-free quantification was performed (Protein Lynx Global Service software). A total of 180 proteins were successfully identified in the AEP samples. The number of identified proteins increased with the time of pellicle formation. Only 4 proteins were present in all the groups (isoforms of IgA, Serum albumin and Statherin). The greatest number of proteins identified uniquely in one of the groups was obtained for the groups treated with HCl after 2 h of pellicle formation (~ 50 proteins). Conclusion: Proteins resistant to removal by HCl, such as Serum Albumin and Statherin, were identified even in the short-term AEP. In addition, 120-min pellicle present many proteins that are resistant to removal by HCl. This suggests an increase in the protection against intrinsic acids along the time of pellicle formation, which should be evaluated in future studies.(AU)

This study aimed to answer the following questions: 1) does whole fluoridated milk protect more against enamel and dentin erosion than fat-free fluoridated milk? 2) does the protective effect of fluoridated milk against erosion follow a dose-response relationship? 3) is the treatment with whole or fat-free fluoridated milk before the first erosive challenge more protective against enamel and dentin erosion? 4) does the fat content of milk change the proteomic profile of the acquired enamel pellicle (AEP)? This study was divided into 2 parts. The first part analyzed in vitro the effect of milk against dental erosion, considering three factors: type of bovine milk (whole/fat-free), presence of different fluoride concentrations (0- 10.0 ppm) and time of application (before/after erosive challenge). Bovine enamel (n=15/group) and root dentin (n=12/group) specimens were submitted to the following treatments: 0.9% NaCl solution (negative control)( after first erosive challenge); whole milk with 0, 2.5, 5.0, 10.0 ppm F; fat-free milk with 0, 2.5, 5.0, 10.0 ppm F; 0.05% NaF solution (positive control) (before or after first erosive challenge). Specimens were submitted to demineralization - remineralization regimes, 4 times/ day, for 5 days. The response variables were enamel and dentin loss, evaluated by profilometry (µm). Data were analyzed using Kruskal­Wallis/Dunn's test (p<0.05). The presence of fluoride, especially at 10 ppm, was the most important factor in reducing dental erosion. The second part detected changes in protein profile of AEP formed in vivo after rinsing with whole milk, fat-free milk or water. Nine subjects with good oral conditions participated. The AEP was formed in the morning, for 120 min, after prophylaxis with pumice. In sequence, the volunteers rinsed with 10 mL of whole milk, fat-free milk or deionized water for 30 s, following a blind, crossover protocol. After 60 min, the AEP was collected with filter paper soaked in 3% citric acid and processed for analysis by liquid chromatography-electrospray ionization tandem mass spectrometry (LCESI- MS/MS). The obtained MS/MS spectra were searched against human protein database (SWISS­PROT). The proteomic data related to protein quantification were analyzed using the PLGS software. A total of 260 proteins were successfully identified in the AEP samples collected in all groups. Forty-nine were common to the 3 groups, while 72, 62 and 49 were specific for groups treated with whole milk, fat-free milk and water, respectively. Some were typical components of the AEP, such as Cystatin-B, Lysozyme C, Histatin-1, Statherin and Lactotransferrin. Other proteins are not commonly described as part of the AEP but could act in the defense of the organism against pathogens. Distinct proteomic profiles were found in the AEP after rinsing with whole or fat-free milk, which could have an impact in bacterial adhesion and tooth dissolution. The use of fat-free milk could favorably modulate the adhesion of bacteria in the AEP and the biofilm formation in comparison to whole milk.(AU)


Este estudo objetivou responder as seguintes questões: 1) o leite integral fluoretado protege mais contra a erosão do esmalte e dentina do que o leite fluoretado desnatado? 2) o efeito protetor do leite fluoretado segue um padrão dose-resposta? 3) o tratamento com leite integral ou leite desnatado fluoretado antes do primeiro desafio erosivo protege mais contra a erosão do esmalte e dentina? 4) o leite contendo gordura altera o perfil proteico da película adquirida do esmalte (PAE)? O estudo foi dividido em 2 partes. Na primeira parte foi realizado um estudo in vitro, considerando três fatores: tipo de leite bovino (integral/ desnatado), diferentes concentrações de fluoreto e tempo de aplicação (antes/após desafio erosivo). Os espécimes de esmalte bovino (n=15 /grupo) e dentina radicular (n=12 /grupo) foram submetidos aos seguintes tratamentos: solução de NaCl a 0,9% (controle negativo)(após o desafio erosivo); Leite integral com 0, 2,5, 5,0, 10,0 ppm F Leite desnatado com 0, 2,5, 5,0, 10,0 ppm F 0,05% de solução de NaF (controle positivo) (antes ou após o primeiro desafio erosivo). Os espécimes foram submetidos a regimes de desmineralização e remineralização, 4 vezes/dia, durante 5 dias. As variáveis de resposta foram perda de esmalte e dentina, avaliadas por perfilometria (µm). Os dados foram analisados usando o teste de Kruskal-Wallis / Dunn (p <0,05). A presença de fluoreto, especialmente na concentração de 10 ppm, demonstrou ser o fator mais importante na redução da erosão dentária. A parte II do estudo detectou alterações no perfil proteico da PAE formada in vivo após bochecho com leite integral, leite desnatado ou água. Nove indivíduos com boas condições de saúde bucal participaram. A PAE foi formada pela manhã, durante 120 minutos, após profilaxia com pedra-pomes. Em seguida, os voluntários bochecharam com 10 mL de leite integral, leite desnatado ou água deionizada durante 30 s, seguindo um protocolo cego e cruzado. Após 60 min, a película foi coletada com papel de filtro embebido em ácido cítrico a 3% e processada para análise por cromatografia líquida acoplada à espectrometria de massas com ionização por eletrospray (LC-ESI-MS / MS). Os espectros MS/MS obtidos foram confrontados com bases de dados de proteínas humanas (SWISSPROT). Os dados proteômicos relacionados à quantificação de proteínas foram analisados usando o software PLGS. Um total de 260 proteínas foi identificado nas amostras de PAE coletadas em todos os grupos. Quarenta e nove eram comuns aos 3 grupos, enquanto 72, 62 e 49 eram específicas para grupos tratados com leite integral, leite desnatado e água, respectivamente. Algumas proteínas encontradas são típicas da PAE, como Cistatina-B, Lisozima C, Histatina-1, Estaterina e Lactotransferrina. Outras proteínas não são comumente descritas como parte da PAE, mas podem atuar na defesa do organismo contra patógenos. Perfis proteômicos distintos foram encontrados na PAE após o bochecho com leite integral ou desnatado, o que poderia ter um impacto na adesão bacteriana e na dissolução dentária. O uso de leite desnatado pode modular favoravelmente a adesão de bactérias na PAE e a formação do biofilme em comparação com o leite integral.(AU)

ABSTRACT A/J and 129P3/J mice strains have been widely studied over the last few years because they respond quite differently to fluoride (F) exposure. 129P3/J mice are remarkably resistant to the development of dental fluorosis, despite excreting less F in urine and having higher circulating F levels. These two strains also present different characteristics regardless of F exposure. Objective In this study, we investigated the differential pattern of protein expression in the liver of these mice to provide insights on why they have different responses to F. Material and Methods Weanling male A/J and 129P3/J mice (n=10 from each strain) were pared and housed in metabolic cages with ad libitum access to low-F food and deionized water for 42 days. Liver proteome profiles were examined using nLC-MS/MS. Protein function was classified by GO biological process (Cluego v2.0.7 + Clupedia v1.0.8) and protein-protein interaction network was constructed (PSICQUIC, Cytoscape). Results Most proteins with fold change were increased in A/J mice. The functional category with the highest percentage of altered genes was oxidation-reduction process (20%). Subnetwork analysis revealed that proteins with fold change interacted with Disks large homolog 4 and Calcium-activated potassium channel subunit alpha-1. A/J mice had an increase in proteins related to energy flux and oxidative stress. Conclusion This could be a possible explanation for the high susceptibility of these mice to the effects of F, since the exposure also induces oxidative stress.

Objective The identification of stem cells (SC) remains challenging. In the human oral mucosal epithelium, these cells are believed to be in the basal layer (stem cell niche), but their exact location is unclear. The aim of this study was to examine the dysplastic oral epithelium for these SC-like proteins in order to assess their diagnostic value as biomarkers complementing the histological grading of dysplasia. Material and Methods Thirty oral epithelial dysplasia (OED), 25 oral lichen planus (OLP), 10 oral hyperkeratosis and 5 normal oral epithelium (OE) were immunohistochemically examined for four SC markers [integrin β1, neuron-glial-2 (NG2), notch 1 (N1) and keratin 15 (K15)]. Results Three of four SC markers were heterogeneously detected in all samples. K15 overexpression in the lower two-thirds of severe OED suggests an expanded SC niche. Integrin β1 distribution pattern was not measurably different between OEDs and control. NG2 was almost negative to absent in all samples examined. N1 expression was weak and highly variable in normal and dysplastic epithelium, making it an unreliable epithelial stem cell marker. Conclusions Present findings suggest that these markers were unable to identify individual epithelial stem cells. Instead, subpopulations of cells, most probably stem cells and transit amplifying cells with stem cell-like properties were identified in the dysplastic oral epithelium. The characteristic expressions of K15 might be of diagnostic value for oral dysplasia and should be investigated further. .

O chumbo (Pb) é um metal pesado que pode ocasionar alterações em todos os sistemas. Porém os maiores danos à saúde ocorrem quando este acomete o sistema nervoso central (SNC). Muitos estudos demonstram as alterações clinicas/comportamentais causadas pela ação do Pb no SNC. Entretanto, ainda são necessários estudos que demonstrem as alterações bioquímicas causadas pelo Pb neste sistema. Por outro lado, tem sido relatado que o ferro (Fe) parece ter um efeito protetor na toxicidade cerebral causada pelo Pb. Assim, o objetivo deste estudo foi analisar a concentração de Pb no tecido cerebral, bem como realizar análise proteômica em cérebro de ratos intoxicados por Pb, submetidos à suplementação com Fe ou não. O experimento foi realizado com 30 ratos recém-desmamados (Rattus norvegicus, variedade Wistar) divididos em 6 grupos (n=5/grupo), de acordo com o tratamento recebido por 6 semanas, a saber: Controle (não exposto ao Pb ou Fe), Controle Fe (exposto à administração de 20 mg/Kg p.c. de FeSO4 a cada 2 dias, por gavagem gástrica), Pb 100 (exposto à água contendo 100 mg/L de Pb), Pb 400 (exposto à água contendo 400 mg/L de Pb) Pb100 + Fe (exposto à água contendo 100 mg/L de Pb e à gavagem com FeSO4) e Pb400 + Fe (exposto à água contendo 400 mg/L de Pb e à gavagem com FeSO4). Decorrido o período experimental, os animais foram eutanasiados e o cérebro dos animais foi removido, sendo descartados o cerebelo e o tronco encefálico. O restante foi submetido à concentração de Pb e à análise proteômica. Foi observada uma dose-resposta em relação à concentração de Pb no cérebro. A administração de FeSO4 reduziu os níveis de Pb no cérebro, embora sem significância estatística. A análise dos géis com os spots proteicos demonstrou uma redução na quantidade destes de acordo com o tratamento recebido pelos grupos. Lead (Pb) is a heavy metal that may yield changes in all body systems, yet the greatest health damages occur when it affects the central nervous system (CNS). Many studies demonstrate the clinical/behavioral changes caused by the action of Pb on the CNS. However, studies are necessary to demonstrate the biochemical changes caused by Pb in this system. Conversely, it has been reported that iron (Fe) seems to play a protective role on the brain toxicity caused by Pb. Therefore, this study analyzed the concentration of Pb in the brain tissue, and conducted proteomic analysis in the brain of rats intoxicated by Pb, submitted or not to Fe supplementation. The study was conducted on 30 weaning rats (Rattus norvegicus, Wistar type) divided in 6 groups (n=5/group), according to the treatment established for 6 weeks, as follows: Control (not exposed to Pb or Fe), Control Fe (exposed to administration of 20 mg/Kg p.c. of FeSO4 at every 2 days, by gastric gavage), Pb 100 exposed to water containing 100 mg/L of Pb), Pb 400 (exposed to water containing 400 mg/L of Pb) Pb100 + Fe (exposed to water containing 100 mg/L of Pb and gavage with FeSO4) and Pb400 + Fe (exposed to water containing 400 mg/L of Pb and gavage with FeSO4). After the experimental period, the animals were killed and the brains of animals were removed, discarding the cerebellum and brainstem. The remaining structure was submitted to analysis of Pb concentration and proteomic analysis. A dose-response relationship was observed in Pb concentration in the brain. O trato gastrointestinal (TGI) é a principal rota de exposição ao fluoreto (F) e o seu mais importante sítio de absorção. Acredita-se que a toxicidade do F comprometa a fisiologia do intestino, devido à relevante sintomatologia gastrointestinal relatada em consequência da exposição excessiva ao F. A função intestinal é controlada por uma complexa rede neuronal interligada e incorporada à parede deste órgão, denominada Sistema Nervoso Entérico (SNE). Embora os efeitos tóxicos do F sobre o Sistema Nervoso Central sejam descritos na literatura, não há estudos relacionados à sua toxicidade sobre o SNE. Neste estudo realizado em ratos, foi avaliado o efeito da exposição aguda ou crônica ao F, sobre a população geral de neurônios entéricos e sobre as subpopulações que expressam os principais neurotransmissores entéricos: Acetilcolina (ACh), Óxido Nítrico (NO), Peptídeo Vasoativo Intestinal (VIP), Peptídeo Relacionado ao Gene da Calcitonina (CGRP) e Substância P (SP). Os animais foram divididos em 5 grupos: 3 destinados à exposição crônica (0 ppm, 10 ppm ou 50 ppm de F na água de beber) e 2 à exposição aguda (0 ou 25 mgF/Kg por gavagem gástrica). Foram coletados os 3 segmentos do intestino delgado (duodeno, jejuno e íleo) e processados para a detecção da HuC/D, ChAT, nNOS, VIP, CGRP e SP, através de técnicas de imunofluorescência, no plexo mioentérico. Foram obtidas imagens para a realização da análise quantitativa dos neurônios da população geral (HuC/D) e nitrérgicos (imunorreativos à nNOS); e morfométrica dos neurônios imunorreativos à HuC/D ou nNOS; e das varicosidades imunorreativas à ChAT, VIP, CGRP ou SP. Amostras dos 3 segmentos intestinais foram preparadas e coradas em Hematoxilina e Eosina para análise histológica da morfologia básica. The gastrointestinal tract (GIT) is the main route of fluoride (F) exposure, and the most important site of its absorption. It is believed that F toxicity compromises the intestine physiology, due to the relevant gastrointestinal symptomatology reported in consequence to excessive exposure. The intestinal function is controlled by a complex neuronal net, which is interconnected and embedded in the wall of this organ, named Enteric Nervous System (ENS). Although the toxic effects of F on the Central Nervous system are described in the literature, there are no studies related to its toxicity on the ENS. Therefore, in this study performed in rats, the effects of chronic and acute F exposure were evaluated, on the general population of enteric neurons and on the subpopulations that express the main enteric neurotransmitters: Acetylcholine (Ach), Nitric Oxide (NO), Vasoactive Intestinal Peptide (VIP), Calcitonin gene related peptide (CGRP), and Substance P (SP). The animals were divided into 5 groups: 3 designed to the chronic exposure (0 ppm, 10 ppm ou 50 ppm de F in the drinking water) and 2 to the acute exposure (0 ou 25 mgF/Kg - gastric gavage). Three intestinal segments were collected (duodenum, jejunum, and ileum) and processed for the immunofluorescence techniques to detect HuC/D, ChAT, nNOS, VIP, CGRP and SP, on the myenteric plexus. Images were obtained for the quantitative analysis of the general population of neurons (HuC/D immunoreactive) and the nitrergic neurons (nNOS immunoreactive), for the morphometric analysis of the general population and nitrergic neurons and also for the immunoreactive varicosities to ChAT, VIP, CGRP or SP. Lead (Pb) is a heavy metal that may yield changes in all body systems, yet the greatest health damages occur when it affects the central nervous system (CNS). Many studies demonstrate the clinical/behavioral changes caused by the action of Pb on the CNS. However, studies are necessary to demonstrate the biochemical changes caused by Pb in this system. Conversely, it has been reported that iron (Fe) seems to play a protective role on the brain toxicity caused by Pb. Therefore, this study analyzed the concentration of Pb in the brain tissue, and conducted proteomic analysis in the brain of rats intoxicated by Pb, submitted or not to Fe supplementation. The study was conducted on 30 weaning rats (Rattus norvegicus, Wistar type) divided in 6 groups (n=5/group), according to the treatment established for 6 weeks, as follows: Control (not exposed to Pb or Fe), Control Fe (exposed to administration of 20 mg/Kg p.c. of FeSO4 at every 2 days, by gastric gavage), Pb 100 exposed to water containing 100 mg/L of Pb), Pb 400 (exposed to water containing 400 mg/L of Pb) Pb100 + Fe (exposed to water containing 100 mg/L of Pb and gavage with FeSO4) and Pb400 + Fe (exposed to water containing 400 mg/L of Pb and gavage with FeSO4). O trato gastrointestinal (TGI) é a principal rota de exposição ao fluoreto (F) e o seu mais importante sítio de absorção. Acredita-se que a toxicidade do F comprometa a fisiologia do intestino, devido à relevante sintomatologia gastrointestinal relatada em consequência da exposição excessiva ao F. A função intestinal é controlada por uma complexa rede neuronal interligada e incorporada à parede deste órgão, denominada Sistema Nervoso Entérico (SNE). Embora os efeitos tóxicos do F sobre o Sistema Nervoso Central sejam descritos na literatura, não há estudos relacionados à sua toxicidade sobre o SNE. Neste estudo realizado em ratos, foi avaliado o efeito da exposição aguda ou crônica ao F, sobre a população geral de neurônios entéricos e sobre as subpopulações que expressam os principais neurotransmissores entéricos: Acetilcolina (ACh), Óxido Nítrico (NO), Peptídeo Vasoativo Intestinal (VIP), Peptídeo Relacionado ao Gene da Calcitonina (CGRP) e Substância P (SP). Os animais foram divididos em 5 grupos: 3 destinados à exposição crônica (0 ppm, 10 ppm ou 50 ppm de F na água de beber) e 2 à exposição aguda (0 ou 25 mgF/Kg por gavagem gástrica). Foram coletados os 3 segmentos do intestino delgado (duodeno, jejuno e íleo) e processados para a detecção da HuC/D, ChAT, nNOS, VIP, CGRP e SP, através de técnicas de imunofluorescência, no plexo mioentérico. Foram obtidas imagens para a realização da análise quantitativa dos neurônios da população geral (HuC/D) e nitrérgicos (imunorreativos à nNOS); e morfométrica dos neurônios imunorreativos à HuC/D ou nNOS; e das varicosidades imunorreativas à ChAT, VIP, CGRP ou SP. The gastrointestinal tract (GIT) is the main route of fluoride (F) exposure, and the most important site of its absorption. It is believed that F toxicity compromises the intestine physiology, due to the relevant gastrointestinal symptomatology reported in consequence to excessive exposure. The intestinal function is controlled by a complex neuronal net, which is interconnected and embedded in the wall of this organ, named Enteric Nervous System (ENS). Although the toxic effects of F on the Central Nervous system are described in the literature, there are no studies related to its toxicity on the ENS. Therefore, in this study performed in rats, the effects of chronic and acute F exposure were evaluated, on the general population of enteric neurons and on the subpopulations that express the main enteric neurotransmitters: Acetylcholine (Ach), Nitric Oxide (NO), Vasoactive Intestinal Peptide (VIP), Calcitonin gene related peptide (CGRP), and Substance P (SP). The animals were divided into 5 groups: 3 designed to the chronic exposure (0 ppm, 10 ppm ou 50 ppm de F in the drinking water) and 2 to the acute exposure (0 ou 25 mgF/Kg - gastric gavage). Three intestinal segments were collected (duodenum, jejunum, and ileum) and processed for the immunofluorescence techniques to detect HuC/D, ChAT, nNOS, VIP, CGRP and SP, on the myenteric plexus. A previous study demonstrated that the amount of Candida spp. in saliva is higher in children with sickle-cell disease. The results from a recent study demonstrate its participation in the etiology of dental caries. Objective This study assessed caries-associated virulence (production of acid, extracellular polysaccharides, proteins and metabolic activity) of biofilms from Candida albicans isolated from saliva of patients with sickle-cell anemia in comparison to isolates obtained from matched healthy children. Material and Methods The isolates were previously obtained from 25 children (4-6 years) and their matched controls (healthy children). One isolate of C. albicans per children was used, totaling 25 isolates per group. The C. albicans biofilms were grown for five days and analyzed regarding the production of lactic acid, extracellular polysaccharides, proteins and metabolic activity. The production of lactic acid was determined by the enzymatic method. The concentration of extracellular polysaccharides was determined by the phenol-sulphuric acid method, and the concentration of the protein was analyzed using the QuantiPro BCA kit. The XTT reduction was used to verify the metabolic activity. The data were analyzed with GraphPad Prism at 5%. Results The Mean±standard deviation for acid production, extracellular polysaccharides, proteins and metabolic activity of isolates from sickle-cell group was, respectively: 7.1±5.0 mmol/L; 15.6±2.5 μg glucose/mg biofilm; 7,503±3,097 μg/mL; A490 3.5±0.7. For isolates from control group the values obtained were: 3.5±3.3 mmol/L; 12.8±3.4 μg glucose/mg biofilm; 4,995±682 μg/mL; A490 3.4±0.5. Os ameloblastomas são tumores odontogênicos, de origem epitelial, que demonstra crescimento lento e comportamento invasivo e altamente recidivante. No entanto os mecanismos moleculares e as vias de sinalização celulares envolvidas na progressão desse tumor não estão claros. Estudos relatados na literatura indicam que aberrações nas vias de sinalização similares as encontradas durante o desenvolvimento dentário, incluindo a via Wnt1 e as proteínas GSK3 ß, APC e Axina1, que formam o complexo de degradação da ß-catenina podem estar alteradas nos ameloblastomas, levando a proliferação e progressão tumoral. O objetivo deste trabalho foi analisar a relação da expressão das proteínas Wnt1, GSK3 ß, APC e Axina1 com a proliferação tumoral dos ameloblastomas por meio de reações de imuno-histoquímicas. Para este estudo foram selecionados 40 casos com o diagnóstico de ameloblastoma, pertencentes aos arquivos do Serviço de Patologia da Disciplina de Patologia Bucal da FOUSP e utilizada a técnica imunohistoquimica da estreptavidina-biotina em blocos parafinados destes casos selecionados. Nos resultados foram observados marcações positivas para a proteína Wnt1 em 25 (62,5%) casos, Axina1 em 23(57,5%), APC de 26(65%) casos e GSK3 ß em 34(85%) casos.


Ameloblastomas is an odontogenic tumors of epithelial origin, which shows slow-growing, highly invasive and recurrent behavior. Nevertheles the molecular mechanisms and cellular signaling pathways involved in the progression of this tumour are not yet clear. Reported studies in leterature indicates that aberrations in signaling pathways similar to those found during tooth development, including Wnt1 and GSK3 ß pathways, APC and Axin1 proteins which form the ß-catenin degradation complex may be altered in ameloblastoma, leading to proliferation and tumour progression. The aim of this study was to analyze the relationship between the expression of Wnt1, GSK3 ß, APC and Axin1 proteins with the tumour proliferation in ameloblastomas by immunohistochemical reactions. For this study, 40 cases were selected with the diagnosis of ameloblastoma, from the Oral Pathology department archives, Dental School, University of São Paulo and the immunohistochemistry technique of streptavidin-biotin in paraffin blocks of these selected cases was perfomed. Positive results to the Wnt protein were observed in 25 (62.5%) cases, Axina1 in 23 (57.5%), APC of 26 (65%) cases and GSK3 ß in 34 (85%) cases.

Neste trabalho foram analisados 30 casos de linfomas de células B da região oral, fixados em solução de formaldeído e incluídos em parafina, através da técnica de imuno-histoquímica para as proteínas c-Myc, Bcl-2, Bcl-6 e ciclina D1. Dos casos analisados 40% foram positivos para a marcação para c-Myc, 33,3% para a marcação para ciclina D1, 83,3% para a marcação para Bcl-2 e 53,3% para a marcação para Bcl-6. Todos os casos foram diagnosticados como linfomas difusos de grandes células B, o subtipo de linfoma com a maior casuística. A análise destas proteínas é de fundamental importância para o diagnóstico e direcionamento do tratamento de doenças hematopoiéticas, pois estão envolvidas em vários processos de controle da transcrição gênica, do ciclo celular e dos processos apoptóticos e o aumento do conhecimento sobre sua ação em diferentes subtipos de linfomas pode corroborar outros estudos


In this study, 30 cases of formalin-fixed and paraffin-embedded B-cell lymphomas of the oral region were submitted to immunohistochemistry for the detection of proteins c-Myc, Bcl-2, Bcl-6 and cyclin D1. Fourty percent (40%) of the studied cases were positive for c-Myc, 10% for cyclin D1, 83.3% for Bcl-2 and 53.3% for Bcl-6. The analysis of these proteins has fundamental importance for the diagnosis and treatment course of hematopoietic diseases, because they are involved in various processes controlling gene transcription and cell cycle. All cases were diffuse large B-cell lymphomas, the subtype with the highest incidence. O objetivo deste trabalho foi analisar as mudanças no perfil protéico em películas adquiridas formadas in situ, em diferentes tempos, sobre o esmalte e sobre a dentina humana após a exposição a dois tipos de ácido: lático e cítrico. Foram utilizados 162 blocos de esmalte e 162 blocos de dentina humana (3X3 mm). Os experimentos foram realizados em três dias consecutivos. Em cada dia, os voluntários (n=9) utilizavam um dispositivo mandibular contendo 6 blocos de esmalte e 6 blocos de dentina. Na sequência, os voluntários permaneceram com o dispositivo na cavidade bucal por 10 ou 120 minutos para formação de película adquirida. Os blocos foram então imersos em ácido cítrico (1%, pH 2,5) ou ácido lático (0,1 M pH 4,8) ou água deionizada por 20 segundos. Na sequência, a película foi removida com um papel de filtro umedecido em ácido cítrico a 3%. Este procedimento foi repetido por mais dois dias adicionais e foi confeccionado um pool com os papeis de filtro obtidos dos 9 voluntários, para cada tipo de substrato, tempo de coleta e meio de imersão. Após extração das proteínas, as mesmas foram submetidas à cromatografia líquida de fase reversa interligada a um espectrômetro de massas (nLC-ESI-MS/MS). Os dados obtidos de MS/MS foram processados e submetidos conjuntamente ao programa SEQUEST [Proteome Discoverer 1.3 (Thermo Scientific)]. As buscas foram feitas utilizando-se os bancos de dados SWISS-PROT e TrEMBL. Para o esmalte, a taxa de identificação de proteínas foi baixa (13 proteínas no total). Já para a dentina, a taxa de identificação de proteínas foi maior (223 proteínas no total), sendo que a exposição ao ácido cítrico reduziu dramaticamente o número de proteínas identificado, o que não aconteceu para o ácido lático. Proteínas ácido-resistentes foram identificadas tanto para o esmalte quanto para a dentina, destacando-se as queratinas e as mucinas, respectivamente. The purpose of this work was to analyze changes in protein profile in the acquired pellicle formed on enamel and dentine at different times in situ after exposure to lactic and citric acids. Enamel (n=162) and dentin (n=162) blocks (3X3 mm) were be used. The experiments were conducted on three consecutive days. Each day, volunteers (n = 9) used a mandibular device containing 6 blocks of enamel and 6 of human dentin. After the volunteers remained with the device in the oral cavity for 10 or 120 minutes in order to allow the formation of the acquired pellicle, the blocks were immersed in citric acid (1%, pH 2.5) or lactic acid (0.1 M, pH 4,8) or deionized water for 20 seconds. Following, the pellicle was collected with an electrode filter paper soaked in 3% citric acid. This procedure was repeated for two additional days and 'pools' with the filter papers obtained from the 9 volunteers for each type of substrate, sampling time and type of acid were made. After extraction, proteins were subjected to reverse phase liquid chromatography coupled to mass spectrometry (nLC-ESI-MS/MS). MS/MS data were processed and submitted to SEQUEST software [Proteome Discoverer 1.3 (Thermo Scientific)]. Searches were done using SWISS-PROT and TrEMBL databases. The rate of protein identification was low for enamel (13 proteins in total). As for dentin, the rte of protein identification was higher (223 proteins in total). Exposure to citric acid dramatically reduced the number of identified proteins, what did not occur for lactic acid. Acid-resistant proteins, especially keratins and mucins were identified for enamel and dentin, respectively. These proteins, or the peptides originated from them that are responsible for the protective effect, are candidates to be used for the enrichment of dental products, aiming to prevent dental caries and erosion.
