This study evaluated the numbers and determined the proportion of mutans streptococci and Lactobacillus spp., which are possible relevant cariogenic organisms, in biofilms recovered from lesions at root surfaces with active caries lesions (ARC), inactive caries lesions, and sound root surfaces (SRS). Samples were cultured in MSB agar for mutans streptococci counts, Rogosa agar for Lactobacillus spp. counts, and brain-heart infusion agar for total viable anaerobic counts. After incubation, the number of colony-forming units (CFUs) was determined and compared between groups by the Mann-Whitney U test with a significance level set at 95%. The proportion of counts of mutans streptococci and Lactobacillus spp. in the total viable microorganisms was also analyzed by Chi-square test. Ninety samples (30 from each surface) from 37 patients were cultured and analyzed. The CFU was similar between mutans streptococci and Lactobacillus spp.These species were present in at least half of the samples and no difference was found in the frequency of isolation of these species. Only 6 samples showed a proportion of more than 10% of mutans streptococci; 4 of the samples were from ARC. Most (93%) SRS samples did not contain viable Lactobacillus spp. The data indicate the low counts of mutans streptococci and Lactobacillus spp. in root sur-faces, regardless of the activity of caries lesions.


O estudo analisou contagens e proporções de mutans strep-tococci e Lactobacillus spp., que podem ser microorganismos importantes em lesões de cárie radicular com diferentes atividades. Biofilmes foram coletados em três locais: ARC ­ superfície radicular com lesão ativa de cárie; IRC ­ superfícies radiculares com lesão inativa de cárie; SRS ­ superfícies de raizes hígidas. As amostras foram cultivadas em agar MSB para contagens de mutans streptococci; agar Rogosa para Lactobacillus spp., e agar BHI para contagens de microrganis-mos viáveis anaeróbicos totais. Após a incubação, o número de unidades formadoras de colônias (UFCs) foi determinado e comparado entre os grupos pelo teste de Mann-Whitney U test. O nível de significância foi estabelecido em 95%. A pro-porção de contagem de mutans streptococci e Lactobacillusspp. no total de microrganismos viáveis também foi analisado através do teste de qui-quadrado. Um total de 90 amostras de 37 pacientes foram cultivadas e analisadas: 30 amostras de ARC, 30 de IRC e 30 de SRS. Números de UFC foram seme-lhantes entre os grupos para ambos, mutans streptococci e Lactobacillus spp. Estas espécies estavam presentes em pelo menos metade de todas as amostras e nenhuma diferença foi encontrada na frequência de isolamento dessas espécies dentro dos grupos. Apenas 6 amostras apresentaram mais de 10% de mutans streptococci e 4 foram de ARC. Em relação aos Lactobacillus spp., 93% das amostras não apresentaram proporção dessas bactérias nas SRS. Mutans streptococci e Lactobacillus spp. estão presentes em baixa proporção nas superfícies radiculares, independentemente da atividade das lesões de cárie.

Abstract Objective: The microbial composition of pericoronitis (Pc) is still controversial; it is not yet clear if the microbial profile of these lesions is similar to the profile observed in periodontitis (Pd). Therefore, the aim of the present study was to describe the microbial profile of Pc lesions and compare it directly with that of subjects with Pd. Methodology: Subjects with Pc and Pd were selected, and subgingival biofilm samples were collected from (i) third molars with symptomatic Pc (Pc-T), (ii) contralateral third molars without Pc (Pc-C) and (iii) teeth with a probing depth >3 mm from subjects with Pd. Counts and proportions of 40 bacterial species were evaluated using a checkerboard DNA-DNA hybridization technique. Results: Twenty-six patients with Pc and 18 with Pd were included in the study. In general, higher levels of microorganisms were observed in Pd. Only Actinomyces oris and Eubacterium nodatum were present in higher mean counts in the Pc-T group in comparison with the Pc-C and Pd-C groups (p<0.05). The microbiota associated with Pc-T was similar to that found in Pc-C. Sites with Pc lesions had lower proportions of red complex in comparison with the Pd sites. Conclusion: The microbiota of Pc is very diverse, but these lesions harbour lower levels of periodontal pathogens than Pd.

Abstract Objective: This clinical study sought to evaluate the effectiveness of passive ultrasonic activation (PUA) in eliminating microorganisms in primary endodontic infection (PEI) after instrumentation of root canals using microbiological culture and checkerboard DNA-DNA hybridization. Methodology: Twenty root canals with PEI and apical periodontitis were selected. The root canals were instrumented and then randomly divided into 2 groups, according to the irrigation method: PUA and conventional needle irrigation (CNI). Microbiological samples were collected before instrumentation (S1), after instrumentation (S2) and after irrigation with 17% EDTA (S3). The samples were subjected to anaerobic culture technique and checkerboard DNA-DNA hybridization analysis. Results: A statistically significant difference was found between CNI (23.56%) and PUA (98.37%) regarding the median percentage values for culturable bacteria reduction (p<0.05). In the initial samples, the most frequently detected species was S. constellatus (50%), and after root canal treatment was E. faecalis (50%). Conclusion: Both treatments significantly decreased the number of bacterial species compared with the initial sample. However, no statistical difference in the total microbial load between PUA and CNI groups was detected. The number of cultivable anaerobic bacteria reduced significantly using PUA, and the bacterial composition and number of bacterial species after using either CNI or PUA was similar.

Abstract Objective: To compare colony forming unit (CFU) of oral bacterial from buccal mucosa and lingual buccal tongue among patients with a dental implant and normal oral hygiene individuals without a dental implant. Material and Methods: Twenty-six individuals with a dental implant and twenty-six individuals without dental implants were included in this study. The samples were sent to the laboratory to culture with Brain Heart Infusion Broth (BHI), prepared serial dilution and then spread to the blood agar. CFU was counted when a single layer of bacteria is formed on the blood agar at any dilution level. An independent-T test was used to compare the means different of CFU oral bacterial between control and test groups from buccal mucosa and lingual buccal mucosa, respectively. Results: Buccal mucosa control group (186.19 ± 5.61) and test group (186.65 ± 6.24) (p>0.05). The result from the lingual buccal tongue control group (198.38 ± 6.12) and test group (197.96 ± 6.50) (p>0.05). There was no significant difference between the control group and test group CFU bacterial load. Conclusion: The presence of implants in the oral cavity do not interfere or worsen the oral condition; nevertheless, the effect of implants surrounding oral flora is similar to natural teeth.

Abstract Acetaldehyde, associated with consumption of alcoholic beverages, is known to be a carcinogen and to be related to the tongue dorsum. Objective The aim of this study was to investigate the relationship between acetaldehyde concentration in mouth air and bacterial characteristics on the tongue dorsum. Methodology Thirty-nine healthy volunteers participated in the study. Acetaldehyde concentrations in mouth air were evaluated by a high-sensitivity semiconductor gas sensor. A 16S rRNA gene sequencing technique was used to compare microbiomes between two groups, focusing on the six samples with the highest acetaldehyde concentrations (HG) and the six samples with lowest acetaldehyde concentrations (LG). Results Acetaldehyde concentration increased in correlation with the increase in bacterial count (p=0.048). The number of species observed in the oral microbiome of the HG was higher than that in the oral microbiome of the LG (p=0.011). The relative abundances of Gemella sanguinis, Veillonella parvula and Neisseria flavescens in the oral microbiome of the HG were higher than those in the oral microbiome of the LG (p<0.05). Conclusion Acetaldehyde concentration in mouth air was associated with bacterial count, diversity of microbiome, and relative abundance of G. sanguinis, V. parvula, and N. flavescens.

Abstract Objectives Since most of the studies evaluates diabetics on multiple daily injections therapy and continuous subcutaneous insulin infusion may help gain better metabolic control and prevent complications, the objective of this study was to evaluate the prevalence of dental caries, the unstimulated salivary flow rate and the total bacteria load, Streptococcus spp. levels and Lactobacillus spp. levels in saliva and supragingival dental biofilm of type 1 diabetics on insulin pump. Material and Methods Sixty patients with type 1 diabetes on insulin pump and 60 nondiabetic individuals were included. The dental caries evaluation was performed using ICDAS and the oral hygiene was assessed according to Greene and Vermillion Simplified Oral Hygiene Index. Unstimulated saliva and supragingival dental biofilm were collected. Total bacteria, Streptococcus spp. and Lactobacillus spp. was quantified by qPCR. Results Patients with type 1 diabetes had a higher prevalence of dental caries and filled and missing teeth when compared with the control group. These patients were associated with more risk factors for the development of dental caries, namely a lower unstimulated salivary flow rate and a higher bacterial load in saliva and dental biofilm. Conclusion Some risk factors related to dental caries were associated with type 1 diabetics. An early diagnosis combined with the evaluation of the risk profile of the diabetic patient is imperative, allowing the dental caries to be analyzed through a perspective of prevention and the patient to be integrated into an individualized oral health program.

Abstract Objective The objective of this study was to clarify differences in bacterial accumulation between gastrointestinal cancer patients who underwent severely invasive surgery and those who underwent minimally invasive surgery. Material and Methods We performed a preliminary investigation of gastrointestinal cancer patients who were treated at the Department of Surgery, Takarazuka Municipal Hospital, from 2015 to 2017 (n=71; 42 laparoscopic surgery, 29 open surgery) to determine changes in bacterial numbers at different sites of the oral cavity (tongue dorsum, gingiva of upper anterior teeth, palatoglossal arch), as well as mouth dryness and tongue coating indices. Specifically, patients received professional tooth cleaning (PTC), scaling, tongue cleaning, and self-care instruction regarding tooth brushing from a dental hygienist a day before the operation. Professional oral health care was also performed by a dental hygienist two and seven days after surgery. Oral bacteria numbers were determined using a bacterial counter with a dielectrophoretic impedance measurement method. Results The number of bacteria at all three examined sites were significantly higher in the open surgery group when compared to the laparoscopic surgery group on the second postoperative day. Relevantly, bacterial count in samples from the gingiva of the upper anterior teeth remained greater seven days after the operation in patients who underwent open surgery. Furthermore, the dry mouth index level was higher in the open surgery group when compared to the laparoscopic surgery group on postoperative days 2 and 7. Conclusions Even with regular oral health care, bacterial numbers remained high in the upper incisor tooth gingiva in gastrointestinal cancer patients who received open surgery. Additional procedures are likely needed to effectively reduce the number of bacteria in the gingival area associated with the upper anterior teeth.

Abstract New technical and scientific developments have been advocated to promote the success of the endodontic treatment. In addition to rotary and reciprocating systems, irrigating solution agitation has been suggested and passive ultrasonic irrigation (PUI) is the most used. Objective: To evaluate, in vitro, the effect of ultrasound streaming (US) in the disinfection of flattened root canal systems prepared by the ProTaper, BioRaCe and Reciproc systems, utilizing the microbiological culture. Methodology: Extracted human mandibular incisors (n=84) were used. Suspensions of Enterococcus faecalis (ATCC 29212) were standardized and inserted along with the teeth immersed in brain-heart infusion (BHI) broth. The contamination was made following a protocol during 5 days. The teeth were randomly divided into six groups: G1, ProTaper Universal; G2, ProTaper Universal with US; G3, BioRaCe; G4, BioRaCe with US; G5, Reciproc; and G6, Reciproc with US. Irrigation was performed with saline solution. After biomechanical preparation, microbiological samples were performed with sterilized paper points, which were diluted and spread on BHI agar; after 48 h, the colony forming units (CFU/mL) were counted for each sample. Results: Groups using ultrasonic agitation presented a greater antibacterial effect than the other ones, even using saline solution as irrigant. The ProTaper Universal system showed the best antibacterial activity of the tested systems (median of 0 CFU/mL with and without surfactant or ultrasonic activation [PUI]). Even with PUI, Reciproc (median of 2.5 CFU/mL with PUI and 5 without it) could not reduce as many colonies as ProTaper Universal without US. The BioRaCe system had greater bacterial reduction when using US (median of 0 CFU/mL with PUI and 30 without it). Conclusions: US promoted greater reduction in the number of bacteria in the flattened root canals prepared with nickel-titanium mechanized systems. Regarding the instruments used, the ProTaper Universal system was the most effective in reducing the bacterial number.

Abstract Streptococcus mutans and Streptococcus sobrinus are strongly associated with dental caries. However, the relationship between oral streptococci and dental caries in children with Down syndrome is not well characterized. Objective To assess and compare dental caries experience and salivary S. mutans, S. sobrinus, and streptococci counts between groups of Down syndrome and non-Down syndrome children and adolescents. Material and Methods This study included a sample of 30 Down syndrome children and adolescents (G-DS) and 30 age- and sex-matched non-Down syndrome subjects (G-ND). Dental caries experience was estimated by the number of decayed, missing, and filled teeth in the primary dentition and the permanent dentition. Unstimulated whole saliva samples were collected from all participants. The fluorescence in situ hybridization technique was used to identify the presence and counts of the bacteria. The statistical analysis included chi-square, Student's t-test and Spearman's correlation. Results The G-DS exhibited a significantly higher caries-free rate (p<0.001) and a lower S. mutans salivary density (p<0.001). No significant differences were found in the salivary densities of S. sobrinus or streptococci between the groups (p=0.09 and p=0.21, respectively). The salivary S. mutans or S. sobrinus densities were not associated with dental caries experience in neither group. Conclusion The reduced dental caries experience observed in this group of Down syndrome children and adolescents cannot be attributed to lower salivary S. mutans densities, as determined with the fluorescence in situ hybridization technique.

Abstract Decalcification of enamel during fixed orthodontic appliance treatment remains a problem. White spot lesions are observed in nearly 50% of patients undergoing orthodontic treatment. The use of fluoride-containing orthodontic materials has shown inconclusive results on their ability to reduce decalcification. The aims of this investigation were to compare the levels of Streptococcus mutans (SM) in saliva and biofilm adjacent to orthodontic brackets retained with a resin-modified glass ionomer cement (RMGIC) (Fuji ORTHO LC) and a light cured composite resin (Transbond XT), and to analyze the influence of topical application of the 1.23% acidulated phosphate fluoride (APF) on SM counts. In a parallel study design, two groups (n=14/15) were used with random allocation and high salivary SM counts before treatment. Biofilm was collected from areas adjacent to the brackets on teeth 13, 22, 33, and 41. Both saliva and biofilm were collected on the 7th, 21st, 35th, and 49th days after appliance placement. Topical fluoride application was carried out on the 35th day. Bonding with RMGIC did not alter SM counts in saliva or biofilm adjacent to the brackets. On the other hand, the biofilm adjacent to brackets retained with composite resin showed a significant increase in SM counts along the trial period. Topical application of 1.23% APF did not reduce salivary or biofilm SM counts regardless of the bonding material. In conclusion, fluoride topical application did not show efficacy in reducing SM. The use of RMGIC as bonding materials allowed a better control of SM cfu counts in dental biofilm hindering the significant increase of these microorganisms along the trial period, which was observed in the biofilm adjacent to the composite material.

Abstract Objective This study evaluated the influence of glycemic control on the levels and frequency of subgingival periodontal pathogens in patients with type 2 diabetes mellitus (DM) and generalized chronic periodontitis (ChP). Material and Methods Fifty-six patients with generalized ChP and type 2 DM were assigned according to the levels of glycated hemoglobin (HbA1c) into one of the following groups: HbA1c<8% (n=28) or HbA1c≥8% (n=28). Three subgingival biofilm samples from sites with probing depth (PD)<5 mm and three samples from sites with PD≥5 mm were analyzed by quantitative Polymerase Chain Reaction (PCR) for the presence and levels of Porphyromonas gingivalis, Tannerella forsythia, Treponema denticola, Eubacterium nodatum, Parvimona micra, Fusobacterium nucleatum ssp. and Prevotella intermedia. Results The mean counts of F. nucleatum ssp. were statistically significantly higher in the sites with PD≥5 mm of the HbA1c≥8% group (p<0.05). Frequencies of detection of T. forsythia, E. nodatum, P. micra and F. nucleatum ssp. were all higher in the sites with PD≥5 mm of the patients with HbA1c≥8%, compared with those of patients with HbA1c<8% (p<0.05). Frequency of detection of P. intermedia was higher in the sites with PD<5 mm of the patients with HbA1c≥8% than those of the patients with HbA1c<8% (p<0.05). Conclusions Poor glycemic control, as indicated by HbA1c≥8%, is associated with increased levels and frequencies of periodontal pathogens in the subgingival biofilm of subjects with type 2 DM and ChP.

ABSTRACT Objective Halitosis can be caused by microorganisms that produce volatile sulphur compounds (VSCs), which colonize the surface of the tongue and subgingival sites. Studies have reported that the use of natural products can reduce the bacterial load and, consequently, the development of halitosis. The aim of this study was to evaluate the antimicrobial activity of the essential oil of Melaleuca alternifolia on the growth and volatile sulphur compound (VSC) production of oral bacteria compared with chlorhexidine. Material and Methods The effects of these substances were evaluated by the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC) in planktonic cultures of Porphyromonas gingivalis and Porphyromonas endodontalis. In addition, gas chromatography analyses were performed to measure the concentration of VSCs from bacterial cultures and to characterize M. alternifolia oil components. Results The MIC and MBC values were as follows: M. alternifolia - P. gingivalis (MIC and MBC=0.007%), P. endodontalis (MIC and MBC=0.007%=0.5%); chlorhexidine - P. gingivalis and P. endodontalis (MIC and MBC=1.5 mg/mL). M. alternifolia significantly reduced the growth and production of hydrogen sulfide (H2S) by P. gingivalis (p<0.05, ANOVA-Dunnet) and the H2S and methyl mercaptan (CH3SH) levels of P. endodontalis (p<0.05, ANOVA-Dunnet). Chlorhexidine reduced the growth of both microorganisms without altering the production of VSC in P. endodontalis. For P. gingivalis, the production of H2S and CH3SH decreased (p<0.05, ANOVA-Dunnet). Conclusion M. alternifolia can reduce bacterial growth and VSCs production and could be used as an alternative to chlorhexidine.

Objectivo In this study, the gingival conditions and the quantitative detection for Aggregatibacter actinomycetemcomitans, Fusobacterium nucleatum, Porphyromonas gingivalis and Prevotella intermedia in pregnant women were determined. Material and Methods Quantitative determinations of periodontal bacteria by using a SyBr green system in women during pregnancy were performed. Women at the 2nd and 3rd trimesters of pregnancy and non-pregnant women were included in this study. A. actinomycetemcomitans was observed in high numbers in women at the 2nd and 3rd trimesters of pregnancy with a significant difference (p<0.05). F. nucleatum and P. intermedia were also observed in high levels. Results and Conclusion Our results show that pregnant women are more susceptible to gingivitis, and the presence of A. actinomycetemcomitans in subgingival biofilm might be taken into account for the treatment of periodontal disease. .

Nineteen plant extracts obtained from plants from the Brazilian Amazon showed activity against planktonic Streptococcus mutans, an important bacterium involved in the first steps of biofilm formation and the subsequent initiation of several oral diseases. Objective: Our goal was to verify whether plant extracts that showed activity against planktonic S. mutans could prevent the organization of or even disrupt a single-species biofilm made by the same bacteria. Material and Methods: Plant extracts were tested on a single-bacteria biofilm prepared using the Zürich method. Each plant extract was tested at a concentration 5 times higher than its minimum inhibitory concentration (MIC). Discs of hydroxyapatite were submersed overnight in brain-heart infusion broth enriched with saccharose 5%, which provided sufficient time for biofilm formation. The discs were then submersed in extract solutions for one minute, three times per day, for two subsequent days. The discs were then washed with saline three times, at ten seconds each, after each treatment. Supports were allowed to remain in the enriched medium for one additional night. At the end of the process, the bacteria were removed from the discs by vortexing and were counted. Results: Only two of 19 plant extracts showed activity in the present assay: EB1779, obtained from Dioscorea altissima, and EB1673, obtained from Annona hypoglauca. Although the antibacterial activity of the plant extracts was first observed against planktonic S. mutans, influence over biofilm formation was not necessarily observed in the biofilm model. The present results motivate us to find new natural products to be used in dentistry. .

Objectives: The intraoral transmission of cariogenic and periodontopathogenic species seems to be facilitated by contaminated toothbrushes and other oral hygiene devices. The aim of this investigation was to analyze the in vitro retention and survival rate of Streptococcus mutans and Streptococcus sanguinis on different toothbrushes. The impacts of human saliva and antimicrobial toothpaste on these parameters were further evaluated. Material and Methods: Part I: Four toothbrushes (Colgate 360°, Curaprox CS5460 ultra soft, elmex InterX, Trisa Flexible Head3) were contaminated by S. mutans DSM 20523 or S. sanguinis DSM 20068 suspensions for three minutes. Bacteria were removed from the toothbrushes after either three minutes (T0) or 24 hours (T24) of dry storage and grown on Columbia blood agar plates for the quantification of colony-forming units (CFUs). Part II: The effects of saliva from a caries-active or a caries-inactive person and of toothpaste containing 0.12% chlorhexidine digluconate were also tested. Results: Part I: After three minutes of dry storage, approximately one percent of the bacteria were still detectable on the toothbrushes. After 24 hours, S. sanguinis exhibited a more pronounced decrease in viable cell numbers compared with S. mutans but the differences were not significant (Kruskal-Wallis test, p>0.05). Part II: The addition of human saliva from a caries-active or caries-inactive person slightly increased the retention of both streptococcal species at T0. The use of toothpaste had no influence on the amount of viable streptococci at T0, but it reduced the microbial load after 24 hours of storage. There were only slight nonsignificant differences (p>0.05) between the four toothbrushes. Conclusions: In vitro bacterial retention and survival of S. sanguinis and S. mutans on different toothbrushes occurred. A total descontaminação do sistema de canais radiculares e da massa dentinária é uma constante preocupação clínica. Diante disso, esta pesquisa teve o objetivo de avaliar o nível de descontaminação dentinária alcançada após irrigação com soluções de hipoclorito de sódio em diferentes concentrações valores de pH. Oitenta dentes bovinos unirradiculados foram divididos em 9 grupos experimentais diferentes. As coroas foram seccionadas e despregadas das raízes. Foram obtidos segmentos de 12mm e os canais instrumentados até a lima K120, preenchidos com EDTA 17%, durante 10 minutos. As raízes foram impermeabilizadas externamente com duas camadas de esmalte. Suspensões de Enterococcus faecalis (ATCC 29212) foram padronizadas em espectrofotômetro (3x108 UFC/mL) e depositadas em microtubos com caldo BHI e um espécime. O protocolo de contaminação seguiu a metodologia de MA et al. (2011) com adaptações. Após 5 dias, os espécimes foram fixados em um dispositivo de apoio esterilizado e irrigados durante 5 minutos com as soluções-teste estabilizadas com tampões: (G1) NaOCl 1% - pH5; (G2) NaOCl 1% - pH7; (G3) NaOCl 1% - pH10; (G4) NaOCl 2,5% - pH5; (G5) NaOCl 2,5% - pH 7; (G6) NaOCl 2,5% - pH10; (G7) NaOCl 5% - pH5; (G8) NaOCl 5% - pH7; (G9) NaOCl 5% - pH10, contendo 8 espécimes cada, além dos grupos controles. Após o tratamento com as soluções irrigadoras, metade dos espécimes é avaliada por cultura microbiológica (contagem de unidades formadoras de colônias UFC/mL das raspas de dentina retiradas das paredes dos canais) e a outra metade do mesmo grupo avaliada por microscopia confocal de varredura a laser (MCVL) e corante Live & Dead. Houve diferença estatística entre diversos grupos, analisados pelos testes de Kruskal-Wallis e Dunn. (p<0,05) Concluiu-se que a solução de hipoclorito de sódio acidificada provocou uma redução significante no número de bactérias. Essa redução foi ainda maior quando a concentração da solução era elevada.


The complete decontamination of root canal system and dentinal mass is a constant clinical concern. Therefore, this study aimed to evaluate the level of dentin decontamination achieved after irrigation with sodium hypochlorite solutions at different concentrations and pH values. Eighty single rooted bovine teeth were divided into 9 different groups. The crowns were sectioned and separated from the roots. Then, there was obtained segments of 12mm and the root canals were instrumented until size #120 K-file and filled with 17% EDTA during 10 minutes. The roots were externally sealed with two layers of nail polish. Suspensions of Enteroccus faecalis (ATCC 29212) were standardized using a spectrophotometer (3x108 CFU/mL) and placed in microtubes containing BHI broth and one sample. The protocol of contamination followed the methodology of MA et al. (2011) with some adjustments. After five days, the samples were fixed in a sterilized device and irrigated during 5 minutes with the tested solutions stabilized by buffers substances: (G1) 1% NaOCl - pH5; (G2) 1% NaOCl - pH7; (G3) NaOCl 1% - pH10; (G4) 2.5% NaOCl - pH5, (G5) 2.5% NaOCl - pH 7, (G6) 2.5% NaOCl - pH10; (G7) 5% NaOCl - pH5; (G8 ) NaOCl 5% - pH 7 (G9) 5% NaOCl - pH10, containing 8 specimens each, and the control groups. After the treatment with the irrigating solutions, half of the specimens were evaluated by microbiological cultures (counting colony forming units CFU/mL of dentine chips removed from the wall of root canals) and the other half of the same group were evaluated by confocal laser scanning microscopy (CLSM) with fluorescent Live & Dead stain. There was statistical difference among various groups (p<0.05) by Kruskal-Wallis and Dunns test. It was concluded that the acidified sodium hypochlorite solution resulted in a significant reduction in the number of bacteria.

O objetivo foi avaliar a influência da agitação ultrassônica (U) na atividade antimicrobiana e penetrabilidade de pastas de hidróxido de cálcio (HC) em propilenoglicol, com ou sem extrato etanólico de própolis (EEP) em propilenoglicol a 10%, através da Microscopia Confocal de Varredura a Laser (MCVL) e cultura microbiológica (CM). Para isso setenta e dois tubos de dentina bovina padronizados foram infectados com Enterococcus faecalis em caldo BHI (Brain Heart Infusion) utilizando um novo protocolo de contaminação com dois ciclos de centrifugação por 5 dias. Durante o período experimental os espécimes foram divididos em 4 grupos e preenchidos com as medicações a seguir: Grupo1- HC, Grupo 2 HC + EEP, Grupo 3 HC + U e Grupo 4 HC + EEP + U. A agitação ultrassônica foi realizada durante 1 minuto nas direções vestíbulo-lingual e mésio-distal, com o auxílio de um inserto liso. As medicações permaneceram no interior dos espécimes por 15 dias. A MCVL analisou as bactérias viáveis (verde) e mortas (vermelho), com o auxílio do corante Live and Dead® nos tubos de dentina após o período de medicação. A contagem de Unidades Formadoras de Colônia (UFCs) foi realizada a partir da cultura microbiológica (CM). As raspas de dentina foram coletadas e diluídas para semeadura em placas de Petri com ágar BHI. Para a penetração foi utilizado o corante Rodamina B durante a manipulação das pastas de HC e as análises feitas por MCVL. O grupo 1 (HC) mostrou a maior viabilidade bacteriana nos tubos de dentina contaminados e o grupo 4 (HC + EEP +U) a menor. A agitação ultrassônica (U) reduziu significativamente a viabilidade bacteriana para todos os grupos. A pasta de HC + EEP mostrou-se mais eficaz que aquela somente com HC (p<0,05%). Concluiu-se que a agitação ultrassônica das pastas de HC aumentou sua atividade antimicrobiana e possibilitou maior penetração intratubular, com preenchimento de túbulos dentinários.


The purpose was to evaluate the influence of ultrasonic agitation (U) on the antimicrobial potential and penetrability of calcium hydroxide (CH) pastes with or without the ethanolic extract of propolis (EEP) in propylene glycol (10%) using Confocal Laser Scanning Microscopy (CLSM) and microbiological cultures (MC) of infected dentin. Seventy two cylindrical dentin specimens were infected with Enterococcus faecalis in BHI broth using a new contamination protocol for 5 days with centrifugations. During the experimental period, the specimens were divided into 4 groups and dressed with the following: Group1- CH paste, Group 2 - CH + EEP, Group 3 CH + U and Group 4 CH + EEP + U. The ultrasonic activation was made during 1 minute in both directions (buccal-lingual / mesio-distal), with the aid of plain point insertion. The medications remained in the root canals for 15 days. The CLSM analyzed the viable (green) and dead (red) bacteria in the infected dentinal tubules, with Live and Dead dye. The colony forming units (CFUs) count was made possible with the MC method. The dentinal wall debris were collected and diluted to be seeded on Petri BHI-agar plates. For the penetration test, the dye Rodamine B was added to CH pastes during the manipulation and analysed by CLSM. Group 1 (CH) showed the greatest bacteria viability in the infected dentinal tubes and Group 4 (CH + EEP+ U) the lowest. Ultrasonic activation created a significant reduction of bacteria viability for all groups. The CH + EEP paste was more bactericidal than the CH (p< 0.05%). It was possible to conclude that ultrasonic activation of CH pastes increased the antimicrobial action and was responsible for better intradentinal penetration with fulfilling of the dentinal tubules.

Este estudo longitudinal teve como objetivo avaliar o impacto da cirurgia bariátrica na condição periodontal e quantificar bactérias periodontopatogênicas em pacientes submetidos a esta cirurgia. Trata-se de um estudo prospectivo, composto por 50 pacientes submetidos à cirurgia bariátrica por derivação gastrojejunal em Y de Roux, em 3 hospitais do estado de São Paulo. A coleta de dados foi realizada em três períodos: pré-operatório, pós-operatório de 6 e de 12 meses e consistiu de: avaliação do fluxo salivar; exame clínico bucal para avaliar desgaste dentário e doença periodontal; coleta de amostras do fluido gengival para quantificação das bactérias periodontopatogênicas Porphyromonas gingivalis, Tannerella forsythia, Treponema denticola ePrevotella intermédia por meio de Reação em Cadeia da Polimerase em Tempo Real (q-PCR); aplicação de questionário relacionado aos fatores para ocorrência de desgaste dentário e autopercepção da saúde bucal; avaliação do peso e altura para obtenção do índice de massa corporal (IMC) e coleta de dados complementares relacionados à saúde do indivíduo, por meio do prontuário médico. No baseline, 51 pacientes com IMC normal foram analisados e comparados aos obesos. ANOVA, teste de Tukey, correlação de Pearson e teste t de Student foram utilizados na análise estatística (p<0,050). No baseline, foi observado maior índice de sangramento gengival (ISG), presença de cálculo e sítios com bolsa 4-5 mm em obesos comparados ao grupo controle (p<0,050), que mostrou uma melhor autopercepção da saúde bucal do que os obesos (p=0,008). A média do IMC nos pacientes submetidos à cirurgia bariátrica diminuiu de 49,69±8,76 kg/m2 para 36,16±8,12 kg/m2 e para 32,26 kg/m2 após 6 e 12 meses da cirurgia (p<0,000), respectivamente. Antes da cirurgia, 67% dos pacientes apresentavam altos níveis séricos de proteína C-reativa (PCR) e 38% glicemia alta. The aim of this longitudinal study was to evaluate the impact of bariatric surgery on periodontal condition and quantify periodontopathogenic bacteria in patients undergoing this surgery. This was a prospective study composed of 50 patients who underwent bariatric surgery for Roux-en-Y gastric bypass in three hospitals in the state of São Paulo. Data collection was performed in three periods: preoperative, postoperative of 6 and 12 months and consisted of: salivary flow evaluation; oral clinical examination to assess tooth wear and periodontal disease; gingival fluid sample collection for quantification of periodontopathogenic bacteria Porphyromonas gingivalis, Tannerella forsythia, Treponema denticola and Prevotella intermedia using Real-time Polymerase Chain Reaction (q-PCR); questionnaire applied as regards factors related to occurrence of tooth wear and self-perceived oral health; weight and height evaluation to determine the body mass index (BMI), in addition to collection of individual's health-related data from medical files. At baseline, 51 patients with normal BMI were analyzed and compared with obese subjects. ANOVA, Tukey test, Pearson correlation and Students t-test were used for statistical analysis (p<0.050). At baseline, greater gingival bleeding index (GBI), presence of calculus and sites with 4-5 mm pocket depth were observed in obese compared with the control group (p<0.050), which showed better self-perceived oral health than the obese (p=0.008). Mean BMI decreased from 49.69±8.76 kg/m2 to 36.16±8.12 kg/m2 and to 32.26 kg/m2 after 6 and 12 months postoperatively (p <0.000), respectively. Before surgery 67% of patients had high C-reactive protein (CRP) serum levels and 38% high glucose level. There was significant reduction in serum CRP and glucose after surgery. Estudos recentes sugeriram associação entre doença periodontal e doenças cardiovasculares e respiratórias. O objetivo deste estudo foi investigar a presença de bactérias periodontopatogênicas em placas ateromatosas humanas. Foram incluídos no estudo 24 pacientes de ambos os sexos, 40-60 anos de idade, internados na UTI do Hospital Nossa Senhora da Conceição em Tubarão-SC com indicação para endarterectomia. Após a coleta das placas ateromatosas, procedeu-se a identificação de sete diferentes espécies bacterianas periodontopatógenos por meio da reação em cadeia da polimerase em tempo real (RT-PCR). Para realização da análise estatistica foi verificada a associação entre as duas variáveis qualitativas nominais (presença ou ausência de bácterias X tipo de bactérias) por meio do teste Qui-quadrado, com nível designificância de 5 %. Os resultados desse trabalho mostraram haver associação entre presença ou ausência de bactérias e dos diferentes tipos de bactérias nos ateromas analisados (p = 0001). Dos ateromas avaliados 75% apresentaram Porphyromonas gingivalis., 58,3% Treponema denticola., 37,5% apresentaram Fusobacteriumnucleatum., 33,3% Prevotella intermedia., 20,8% Agreggatibacter actinomycetencomitanse. 0,4% de Prevotella nigrescens. Esses achados permitem concluir que bactérias periodontopatogênicas apresentam a capacidade de invadir a parede das artérias, podendo ter papel significativo no desenvolvimento ou ruptura das placas ateroescleróticas.


Recent studies suggested an association between periodontal disease and cardiovascular and respiratory diseases. The aim of this study was to investigate the presence of periodontopathogens in human atherosclerotic plaque. It were included in this study 24 non-smoking patients, both genders, 40-60 years, admitted at intensive unit care with indication of endarterectomy. After collection of carotid samples containing the lesions, the identification of seven different bacterial species was performed by real time polymerase chain reaction (RT-PCR). For statistical analysis, the association between two nominal qualitative variables (presence or absence of bactérias vs. bacterial specieis) was verified by Chi-squared test, with a 5% significance level. The results obtained showed an association between the presence or absence and bacterial species found in atheroma (p= .0001). Among the atheroma examined, 75% showed the presence of Porphyromonas gingivalis, 58.3% showed Treponema denticola, 37.5% showed Fusobacteriumnucleatum, 33.3% showed Prevotella intermedia, 20.8% Agreggatibacter actinomycetencomitans and 0.4% Prevotella nigrescens. These findings suggest that periodontopatoghens are capable of invading artery wall and could play a significant role in the development or rupture of atherosclerotic lesions.

Estudos recentes sugeriram associação entre doença periodontal e doenças cardiovasculares e respiratórias. O objetivo deste estudo foi investigar a presença de bactérias periodontopatogênicas em placas ateromatosas humanas. Foram incluídos no estudo 24 pacientes de ambos os sexos, 40-60 anos de idade, internados na UTI do Hospital Nossa Senhora da Conceição em Tubarão-SC com indicação para endarterectomia. Após a coleta das placas ateromatosas, procedeu-se a identificação de sete diferentes espécies bacterianas periodontopatógenos por meio da reação em cadeia da polimerase em tempo real (RT-PCR). Para realização da análise estatistica foi verificada a associação entre as duas variáveis qualitativas nominais (presença ou ausência de bácterias X tipo de bactérias) por meio do teste Qui-quadrado, com nível designificância de 5 %. Os resultados desse trabalho mostraram haver associação entre presença ou ausência de bactérias e dos diferentes tipos de bactérias nos ateromas analisados (p = 0001). Dos ateromas avaliados 75% apresentaram Porphyromonas gingivalis., 58,3% Treponema denticola., 37,5% apresentaram Fusobacteriumnucleatum., 33,3% Prevotella intermedia., 20,8% Agreggatibacter actinomycetencomitanse. 0,4% de Prevotella nigrescens. Esses achados permitem concluir que bactérias periodontopatogênicas apresentam a capacidade de invadir a parede das artérias, podendo ter papel significativo no desenvolvimento ou ruptura das placas ateroescleróticas.


Recent studies suggested an association between periodontal disease and cardiovascular and respiratory diseases. The aim of this study was to investigate the presence of periodontopathogens in human atherosclerotic plaque. It were included in this study 24 non-smoking patients, both genders, 40-60 years, admitted at intensive unit care with indication of endarterectomy. After collection of carotid samples containing the lesions, the identification of seven different bacterial species was performed by real time polymerase chain reaction (RT-PCR). For statistical analysis, the association between two nominal qualitative variables (presence or absence of bactérias vs. bacterial specieis) was verified by Chi-squared test, with a 5% significance level. The results obtained showed an association between the presence or absence and bacterial species found in atheroma (p= .0001).
