Background: The development and establishment of oral squamous cell carcinoma are confined to carcinogenesis, which involves oxidative stress via oxygen-free radical production as a hydroxyl radical (HO•), considered the most important cause of oxidative damage to basic biomolecules since it targets DNA strands. 8-Hydroxy-2´- deoxyguanosine (8-OHdG) is considered a free radical with a promutagenic capacity due to its ability to pair with adenosine instead of cytosine during replication. Material and Methods: We collected 30 paraffin-embedded tissue samples of OSCC from patients treated between 2013 and 2018. We recorded risk habits, disease stage, disease free survival and death with at least 3 years of followup. 8-Hydroxyguanosine was evaluated by immunohistochemistry and subsequently classified as weak-moderate or strong positive expression. Additionally, we noted whether it was expressed in the cytoplasm and/or nucleus. Results: Most of the cases expressed 8-OHdG with a strong intensity (80%). All neoplastic cells were preferentially stained in only the cytoplasm (70.0%), but nuclear positivity was found in 30%, independent of the intensity. Based on the location in the cytoplasm and/or nucleus, tumors >4 cm showed a high frequency (95.5%) of 8-OHdG expression in only the cytoplasm, with a significant difference (p value ≤ 0.001). Additionally, overall survival was affected when immunoexpression was present in the cytoplasm and nucleus because all deaths were in this group were statistically significant (p value = 0.001). Conclusions: All tumors showed DNA oxidative damage, and 8-OHdG was preferentially expressed in the cytoplasm. This finding was associated with tumor size and, when present in the nucleus, might also be related to death. (AU)

Introducción: En el mundo, alrededor de 50 millones de personas padecen demencia; la forma más común es la enfermedad de Alzheimer (EA), que representa el 60-70% de los casos. Dada su alta incidencia, se hace imperativo diseñar estudios que permitan ampliar el conocimiento sobre su aparición y desarrollo, para proponer diagnósticos tempranos y/o posibles tratamientos. Una de las estrategias metodológicas que se han desarrollado son los modelos transgénicos murinos para el estudio de los factores involucrados en su etiología, y entre ellos, el estrés oxidativo y la respuesta inmune.DesarrolloSe realizó una búsqueda de artículos originales y revisiones en PubMed, Scopus y Google Scholar (2013-2019). En esta revisión abordamos dos factores que han sido estudiados de forma independiente: el estrés oxidativo y la respuesta inmune en modelos transgénicos para la EA, y se discute la relación que existe entre ellos y que impacta en la pérdida de la plasticidad sináptica y estructural, produciendo como efecto final el deterioro cognitivo.ConclusiónEsta revisión describe posibles mecanismos en donde participan el estrés oxidativo y la respuesta inmune sobre los efectos moleculares, celulares y conductuales en la EA, observando una estrecha relación entre estos elementos que conducen hacia el deterioro cognitivo. Introduction: Worldwide, approximately 50 million people have dementia, with Alzheimer disease (AD) being the most common type, accounting for 60%-70% of cases. Given its high incidence, it is imperative to design studies to expand our knowledge about its onset and development, and to develop early diagnosis strategies and/or possible treatments. One methodological strategy is the use of transgenic mouse models for the study of the factors involved in AD aetiology, which include oxidative stress and the immune response.DevelopmentWe searched the PubMed, Scopus, and Google Scholar databases for original articles and reviews published between 2013 and 2019. In this review, we address two factors that have been studied independently, oxidative stress and the immune response, in transgenic models of AD, and discuss the relationship between these factors and their impact on the loss of synaptic and structural plasticity, resulting in cognitive impairment.ConclusionThis review describes possible mechanisms by which oxidative stress and the immune response participate in the molecular, cellular, and behavioural effects of AD, observing a close relationship between these factors, which lead to cognitive impairment. Introducción: Los estudios de capacidad antioxidante enbebidas de frutas se relacionan con el contenido de polifenoles, carotenoides y flavonoides. Objetivo: Evaluar la capacidad antioxidante del néctar deUngurahui (Oenocarpusbataua) en el proceso de la elaboración del néctar, en las etapas de selección, pre cocción, estandarización y néctar, y el análisis fitoquímico. Material y métodos: Los frutos fueron colectados delFundo “El bosque” del sector denominado “Loboyoc” del distrito de “Las Piedras”, Tambopata, Madre de Dios, Perú. Laformulación del néctar fue de 11° Brix y proporción aguapulpa (3:1), la determinación de la capacidad antioxidante fuemediante el método basado en la formación del complejo azuldel Fósforo Molibdeno y para el análisis fitoquímico cualitativose aplicó las reacciones generales de caracterización de metabolitos secundarios. Resultados: La capacidad antioxidante expresados enmg de ácido ascórbico en un gramo de muestra son: 4,61mg para la etapa selección, 1,12 mg; precocción, 0,99 mg;estandarizado, 0,91 mg y el néctar de frutas y para los metabolitos secundarios en la pulpa: la ausencia de alcaloides, baja proporción de aminoácidos, moderada presencia de taninos, compuestos fenólicos, flavonoides y azúcares reductores. Conclusión: El tratamiento térmico de pre cocción presenta una drástica reducción de la capacidad antioxidante debido a que algunos metabolitos secundarios que actúan como antioxidantes disminuyen después de ser sometidos a los procesos fisicoquímicos del néctar, de moderada a baja proporción.(AU)

Fundamentos: Se reconoce a la diabetes Mellitus tipo 2 como un tipo de enfermedad crónica no transmisible; entre los factores de riesgo a padecerla, destaca el estilo de vida no saludable, específicamente dietas altas en calorías y hábitos sedentarios. El presente estudio tuvo como objetivo, revisar los mecanismos etiológicos y o patológicos que conllevan al desarrollo de la diabetes Mellitus tipo 2 (DM2). Métodos: Se realizó una investigación de tipo documental bibliográfica, de estudios sobre los mecanismos y factores deriesgo, que contribuyen con la aparición de DM2, los cuales se encuentren publicados en revistas médicas en idioma inglés o español, indexadas en Scopus, Elservier, Medline o Scielo, entrelos años 2010 y 2020. Resultados: DM2 es una patología adquirida dependientedel síndrome metabólico, prevenible y tratable, en el que la resistencia a la insulina se vuelve un indicador de riesgo. Las evaluaciones sugieren que se debe corregir el estilo de vida y evitar los factores de riesgo, que son los generadores de ácidos grasos no esterificados y radicales libres exógenos.Conclusiones: La mejor estrategia de control, sigue siendo la aplicación de medicina preventiva e integral. Background: Diabetes Mellitus type 2 is recognized among the chronic non-communicable diseases and among the risk factors for developing it, unhealthy lifestyle is described, specifically high calorie diets and sedentary habits. The aim of the present study was to review the etiological and pathologicalmechanisms that lead to the development of type 2 diabetesmellitus (DM2). Methods: A bibliographic documentary research was conducted on the mechanisms and risk factors that contribute to the onset of DM2, which are published in medical journals in English or Spanish, indexed in Scopus, Elservier, Medline or Scielo, between 2010 and 2020. Results: DM2 is an acquired pathology dependent on the metabolic syndrome, preventable and treatable, in which insulin resistance becomes a risk indicator. Evaluations suggest that lifestyle should be corrected and risk factors, which arethe generators of non-esterified fatty acids and exogenous free radicals, should be avoided. Conclusions: The best control strategy remains the application of preventive and integrative medicine. (AU)

BACKGROUND: it is well known that moderate or vigorous physical exercise produces an increase in free radicals. AIM: the aims of this study were to observe changes in malondialdehyde and antioxidant vitamins after a maximum incremental test, and to relate malondialdehyde and antioxidant vitamin values to performance parameters. METHODS: eighty-four male athletes participated in this study. Participants performed a maximum incremental test until exhaustion on a treadmill. Malondialdehyde in plasma and antioxidant vitamins in plasma and erythrocytes were determined before and after the test. RESULTS: in plasma, there was a decrease in malondialdehyde after the test. In erythrocytes, results showed increases in vitamin C and decreases in vitamin E after the test. Maximal oxygen uptake values were associated positively with vitamin C and negatively with malondialdehyde levels before the test. On the other hand, maximal oxygen uptake, total test time, and total test distance were positively related to the malondialdehyde values obtained after the test. CONCLUSIONS: a maximum incremental test did not produce any changes in plasma vitamins in athletes. However, it increased the levels of vitamin C in erythrocytes and decreased malondialdehyde values in plasma and vitamin E in erythrocytes. The levels of malondialdehyde, vitamin C and vitamin E were related to performance parameters. These results may be linked to the adaptation of antioxidant systems due to regular training


INTRODUCCIÓN: es sabido que el ejercicio físico moderado o vigoroso produce un aumento de radicales libres. OBJETIVOS: los objetivos del estudio fueron: observar los cambios del malondialdehído y las vitaminas antioxidantes después de un test incremental máximo y relacionar los niveles de malondialdehído y vitaminas antioxidantes con parámetros de rendimiento. MÉTODOS: ochenta y cuatro atletas masculinos participaron en el estudio. Los participantes realizaron un test incremental hasta la extenuación en un tapiz rodante. El malondialdehído en plasma y las vitaminas antioxidantes en plasma y en eritrocitos se determinaron antes y después del test incremental. RESULTADOS: en el plasma hubo una disminución del malondialdehído después de la prueba incremental. En los eritrocitos, los resultados mostraron un aumento de la vitamina C y un descenso de la vitamina E después de la prueba. El consumo máximo de oxígeno se asoció positivamente con la vitamina C e inversamente con el malondialdehído antes del test. Por otro lado, el consumo máximo de oxígeno, el tiempo total del test y la distancia total durante el test se asociaron positivamente con los valores de malondialdehído obtenidos tras el test máximo. CONCLUSIÓN: un test incremental máximo hasta la extenuación no produjo cambios en las vitaminas antioxidantes del plasma. Sin embargo, sí aumentó los niveles de vitamina C en los eritrocitos y redujo los niveles de malondialdehído en el plasma y los de vitamina E en los eritrocitos. Los niveles de malondialdehído, vitamina C y vitamina E se relacionaron con los parámetros de rendimiento. Estos resultados podrían estar relacionados con la adaptación de los sistemas antioxidantes debido al entrenamiento regular

Objective: Cocktail honey is derived from a mixture of honey (trigona sp.), bee bread, and homogeneous royal jelly. The material has a phenolic content rich in antioxidants that are beneficial for women's reproductive health, especially for pre-conception, because it can suppress the content of free radicals in the body. Antioxidants are useful to overcome oxidative damage due to free radicals in the body that prevent various diseases from increasing fertility during pre-conception. Method: This study used the DPPH (2,2-diphenyl-1-picrylhydrazyl) test method using UV–vis spectrophotometry to express the value of free radical reduction activity as IC50 (inhibitory concentration) values. Results: The DPPH test on cocktail honey products obtained an average yield of 4577.7 μg/mL, which was included in the product category was very weak in the antioxidant activity content. Conclusion: The content contained in the honey cocktail contains weak bioactive content by assessing the antioxidant content using DPPH. The difference in the results of antioxidant activity tests using DPPH is caused by the test method and the conditions used in processing, homogeneous ingredients, solvent volume, extraction time, temperature, and pressure in product management. BACKGROUND: Drug-induced nephrotoxicity is a frequent adverse event that can lead to acute or chronic kidney disease and increase the healthcare expenditure. It has high morbidity and mortality incidence in 40-70% of renal injuries and accounts for 66% cases of renal failure in elderly population. OBJECTIVE: Amelioration of drug-induced nephrotoxicity has been long soughed to improve the effectiveness of therapeutic drugs. This study was conducted to review the melatonin potential to prevent the pathogenesis of nephrotoxicity induced by important nephrotoxic drugs. METHODS: We analyzed the relevant studies indexed in Pubmed, Medline, Scielo and Web of science to explain the molecular improvements following melatonin co-administration with special attention to oxidative stress, inflammation and apoptosis as key players of drug-induced nephrotoxicity. RESULTS: A robust consensus among researchers of these studies suggested that melatonin efficiently eradicate the chain reaction of free radical production and induced the endogenous antioxidant enzymes which attenuate the lipid peroxidation of cellular membranes and subcellular oxidative stress in drug-induced nephrotoxicity. This agreement was further supported by the melatonin role in disintegration of inflammatory process through inhibition of principle pro-inflammatory or apoptotic cytokines such as TNF-alfa and NF-κB. These studies highlighted that alleviation of drug-induced renal toxicity is a function of melatonin potential to down regulate the cellular inflammatory and oxidative injury process and to stimulate the cellular repair or defensive mechanisms. CONCLUSION: The comprehensive nephroprotection and safer profile suggests the melatonin to be a useful adjunct to improve the safety of nephrotoxic drugs


ANTECEDENTES: La nefrotoxicidad inducida por medicamentos es un acontecimiento adverso frecuente que puede conducir a una nefropatía aguda o crónica, e incrementar los costes sanitarios. Presenta una incidencia elevada de morbimortalidad en el 40-70% de las lesiones renales y es responsable del 66% de los casos de insuficiencia renal entre la población de edad avanzada. OBJETIVO: La mejora de la nefrotoxicidad inducida por medicamentos es un objetivo anhelado desde hace mucho tiempo, para mejorar la eficacia de los fármacos terapéuticos. Este estudio se llevó a cabo con el propósito de revisar el potencial de la melatonina para prevenir la patogenia de la nefrotoxicidad inducida por medicamentos nefrotóxicos importantes. MÉTODOS: Analizamos los estudios relevantes indexados en Pubmed, Medline, Scielo y Web of Science, para explicar las mejoras moleculares posteriores a la administración concomitante de melatonina; prestando especial atención al estrés oxidativo, la inflamación y la apoptosis como actores fundamentales de la nefrotoxicidad inducida por medicamentos. RESULTADOS: Un sólido consenso entre los investigadores de estos estudios sugirió que la melatonina erradica de forma eficiente la reacción en cadena de producción de radicales libres e induce las enzimas antioxidantes endógenas que atenúan la peroxidación lipídica de las membranas celulares y el estrés oxidativo subcelular en la nefrotoxicidad inducida por medicamentos. Este consenso se vio respaldado por el papel de la melatonina en la desintegración del proceso inflamatorio a través de la inhibición de las principales citocinas proinflamatorias o apoptóticas, como el TNF-α y el NF-κB. Estos estudios subrayan que la mitigación de la nefrotoxicidad inducida por medicamentos se deriva del potencial de la melatonina para regular a la baja el proceso celular de lesión inflamatoria y oxidativa, y estimular la reparación celular o los mecanismos defensivos de las células. CONCLUSIÓN: La nefroprotección exhaustiva y el perfil de seguridad más favorable sugieren que la melatonina es un complemento útil para mejorar la seguridad de los fármacos nefrotóxicos

Introducción: cualquier situación de estrés, entre las cuales se incluye la realización de ejercicio físico, implica la posibilidad de una excesiva producción de radicales libres y, por acción de estos, un estrés oxidativo en las células. Para combatir estos efectos, las células disponen de unos mecanismos de defensa denominados sistemas antioxidantes. Objetivos: el objetivo del presente estudio se basa en analizar los posibles cambios fisiológicos en relación a parámetros relacionados con el estrés oxidativo (MDA) y con la actividad antioxidante (vitamina A, vitamina C y vitamina E) en los eritrocitos de jugadores profesionales de fútbol, en comparación con sujetos no entrenados. Métodos: cuarenta y dos varones divididos en 21 jugadores de fútbol (SG) (24,95 ± 3,03 años) y 21 estudiantes sedentarios (CG) (23,71 ± 2,42 años) participaron en el estudio. Se les evaluaron los niveles basales de MDA, vitamina C, vitamina A y vitamina E en eritrocitos, así como las características antropométricas y el VO2max. Resultados: se observaron diferencias significativas en las características antropométricas (p < 0,05) y VO2max (p > 0,01). SG presentó niveles estadísticamente inferiores de MDA (p > 0,01), vitamina C (p > 0,05), vitamina E (p > 0,05) y vitamina A (p > 0,01). Conclusiones: existe un mayor estrés oxidativo en jugadores de fútbol que en sedentarios, por lo que puede ser necesaria una suplementación con antioxidantes en este grupo


Introduction: the situations of stress among which physical exercise is included imply the possibility of an excessive production of free radicals and, by their action, an oxidative stress in the cells. To combat these effects, cells have defense mechanisms called antioxidant systems. Objectives: the objective of this study is to analyze the possible physiological changes in relation to parameters related to oxidative stress (MDA) and antioxidant activity (vitamin A, vitamin C and vitamin E) in the erythrocytes of professional soccer players, in comparison with untrained subjects. Methods: forty-two men divided into 21 soccer players (SG) (24.95 ± 3.03 years) and 21 sedentary students (CG) (23.71 ± 2.42 years) participated in the study. Their basal levels of MDA, vitamin C, vitamin A and vitamin E in erythrocytes, as well as their anthropometric characteristics and VO2max, were evaluated. Results: significant differences were observed in the anthropometric characteristics (p < 0.05) and VO2max (p > 0.01). SG presented statistically lower levels of MDA (p > 0.01), vitamin C (p > 0.05), vitamin E (p > 0.05) and vitamin A (p > 0.01). Conclusions: there is higher oxidative stress in soccer players than in sedentary players and it may be necessary to supplement with antioxidants in this group

El exceso de radicales libres en el entorno de una herida contribuye de forma significativa al estancamiento de la misma en la fase inflamatoria, favoreciendo la aparición de una herida de difícil cicatrización. Se genera una situación de estrés oxidativo, puesto que las enfermedades de base que suelen presentar los pacientes con heridas crónicas dificultan en gran medida el retorno a una situación de balance oxidativo. Esto incide directamente sobre la capacidad de respuesta del tejido, ya que se afectan directamente estructuras celulares y la matriz extracelular del entorno. En este trabajo se ha aplicado un tratamiento antioxidante para el control del exceso de radicales libres en el entorno de la herida, con el fin de conseguir la activación en el caso de úlceras crónicas de etiología vascular venosa. Para ello, se ha aplicado un apósito antioxidante con componentes de origen natural, una matriz absorbente de algarrobo y una solución antioxidante con cúrcuma y acetilcisteína, indicado para la fase inflamatoria de las heridas y la adecuada terapia compresiva. Para la evaluación de la evolución de las úlceras se utilizó la escala RESVECH 2.0. Transcurridas 2-3 semanas, el apósito antioxidante consiguió la activación de la herida, lo que se reflejó en el mantenimiento del lecho de la úlceras libre de tejido desvitalizado, la inducción de la formación de tejido de granulación y la activación de los bordes perilesionales. A partir de ese momento, se aplicaron apósitos de cura húmeda convencionales, llegando hasta la cicatrización total de las heridas en las semanas 5-6 desde el inicio del tratamiento


The excess of free radicals in the wound environment significantly contributes to the arrest of the wound in the inflammatory phase, favoring the appearance of a hard-to-heal wound. An oxidative stress condition is generated, as the comorbidities that patients with chronic wounds frequently present hinder the return to an oxidative balance. This fact affects the ability of the tissue to respond to an injury, since cellular structures and extracellular matrix of the environment are affected. In this work, an antioxidant treatment has been applied, to control the excess of free radicals in the wound environment, in order to achieve the activation of chronic ulcers from venous vascular etiology. This antioxidant dressing has two components from natural origin, an absorbent matrix from carob tree and an antioxidant solution containing curcumin and acetylcysteine, and is indicated for the inflammatory phase of the healing process of wounds. The appropriate compressive therapy was also applied. RESVECH 2.0 scale was used as wound evolution assessment. The results showed that after 2-3 weeks of treatment, the antioxidant dressing achieved the activation of the wound, which was reflected in the maintenance of a wound bed free from devitalized tissue, the induction of the formation of granulation tissue and the activation of the perilesional edges. After that, conventional moist wound healing dressings were applied, reaching wound closure after 5-6 weeks from the beginning of the treatment

Objetivos: El conocimiento acerca de los mecanismos subyacentes al envejecimiento de la piel es, en la actualidad, una estrategia clave en la prevención de sus manifestaciones cutáneas. A través del estudio de la principal vía responsable de la senescencia celular, el estrés oxidativo, se revisan activos cosméticos capaces de neutralizar su efecto y la relevancia del mismo. Métodos: Se utilizó la base de datos PubMed para la revisión de artículos científicos, así como diversos textos de Cosmética y Dermofarmacia. Resultados: En el transcurso del envejecimiento cutáneo se solapan dos procesos, uno intrínseco, con alteración del propio sistema redox antioxidante, y otro extrínseco derivado de factores externos, en concreto la radiación solar. El resultado es un grave daño celular a través de especies reactivas de oxígeno y otros mecanismos de oxidación. La revisión de activos cosméticos antioxidantes revela la eficacia de dichas sustancias en el curso de la senescencia celular, gracias a su capacidad de neutralizar los productos dañinos que son generados. Destaca la acción conjunta de ácido ascórbico y vitamina E. Otros, como los derivados del retinol, carotenoides, vitamina B3 y flavonoides han demostrado tener un efecto beneficioso. Conclusiones: En el objetivo de tratar y prevenir el envejecimiento cutáneo es esencial la aplicación de activos antioxidantes. La importancia de su inclusión en productos cosméticos antienvejecimiento o anti-age, en la actualidad, ha alcanzado su auge con el planteamiento de aprovechar su acción antioxidante para reducir al mínimo el efecto del estrés oxidativo y retrasar la muerte celular que se oculta tras el proceso de envejecimiento de la piel


Objectives: Knowledge about skin aging underlying process, nowadays, is the key strategy in prevention of its main cutaneous expression. Through the study of the major mechanism responsible of cellular senescence, oxidative stress, it is reviewed the relevance of various cosmetic actives with the potential to neutralise the oxidative damaging effect. Methods: To carry out the review of related scientific articles we used the databases PubMed, as well as various texts about Cosmetic and Dermopharmacy. Results: During the course of skin aging two mechanisms overlap, an intrinsic process with disorder of the natural anti-oxidative redox system, and an extrinsic process resulting of external factors, specifically solar radiation. A serious cellular damage is the result of reactive oxygen species (ROS) and other oxidative mechanisms. The review of several antioxidant cosmetic actives reveals the efficacy of these substances in the cellular senescence development, because of its action neutralizing harmful products generated. Therefore, combination of ascorbic acid and vitamin E stands out. Others, as retinoids, carotenoids, vitamin B3 or flavonoids have beneficial effects demonstrated. Conclusions: In order to treat and prevent effectively skin aging, we conclude it results essential the use of antioxidant actives. The relevance of including them in anti-aging cosmetic products has reach a new height, approaching their antioxidant property to reduce to a minimum stress oxidative effect and slow down cellular death disguised by skin aging

Introduction: The relationship between diet and health has aroused scientific interest, especially the consumption of antioxidant nutrients naturally present in foods, because of its action against the deleterious effects of free radicals in the body. Objective: This study aimed to estimate the intake of antioxidant nutrients and its relationship with lipid profile and oxidative stress in student users of a university restaurant in comparison with non-users. Methods: This cross-sectional study involved 145 university students divided into two groups: users of the university restaurant (group 1, n = 73) and non-users (group 2, n = 72). We measured body mass index and waist circumference, and estimated the intake of antioxidant micronutrients. Serum concentrations of total cholesterol, high-density lipoprotein cholesterol, low-density lipoprotein cholesterol, and triglyceride levels, and plasma concentrations of malondialdehyde were determined. Results and conclusion: Intake of copper, zinc, selenium, and vitamin C were within the recommendations in both groups, and vitamins A and E were below the reference values. There was a correlation between the intake of vitamin C and triglycerides. Group 1 members had better dietary patterns in relation to antioxidant micronutrients, reflecting less atherogenic lipid profile and lower exposure to oxidative stress compared to group 2 (AU)


Introducción: la relación entre la dieta y la salud ha despertado interés científico, especialmente en lo referente al consumo de nutrientes antioxidantes presentes de forma natural en los alimentos, debido a su acción contra los efectos nocivos de los radicales libres en el cuerpo. Objetivo: el objetivo de este estudio fue estimar la ingesta de nutrientes antioxidantes y su relación con el perfil lipídico y el estrés oxidativo en estudiantes usuarios de un restaurante universitario, en comparación con los no usuarios. Métodos: este es un estudio transversal que incluye 145 estudiantes universitarios, divididos en dos grupos: los usuarios del restaurante universitario (grupo 1, n = 73) y no usuarios (grupo 2, n = 72). Fueron medidos el índice de masa corporal y la circunferencia de la cintura, y se estimó la ingesta de micronutrientes antioxidantes. Se determinaron las concentraciones séricas de colesterol total, colesterol unido a lipoproteínas de alta densidad, colesterol unido a lipoproteínas de baja densidad, los niveles de triglicéridos y las concentraciones plasmáticas de malondialdehído. Resultados y conclusiones: la ingesta de cobre, zinc, selenio y la vitamina C se encontraba dentro de las recomendaciones de los dos grupos, y la de vitaminas A y E estaba por debajo de los valores de referencia. Hubo una correlación entre el consumo de vitamina C y los triglicéridos. Miembros del grupo 1 mostraron mejores patrones dietéticos en relación con micronutrientes antioxidantes, lo que refleja menos perfil lipídico aterogénico y una menor exposición al estrés oxidativo en comparación con el grupo 2 (AU)

Introducción: la influencia de los antioxidantes en los procesos oculares ha tomado en los últimos años una relevancia importante por sus efectos en la salud visual. El estrés oxidativo es un factor implicado en el desarrollo de las principales patologías tales como cataratas, glaucoma y degeneración macular asociada a la edad (DMAE). Objetivo: establecer la evidencia científica que existe sobre los distintos tipos de antioxidantes y sus efectos en procesos patológicos oculares mediante una revisión sistemática. Métodos: búsqueda bibliográfica en MEDLINE, Scielo y Cochrane de estudios que evalúan la utilización de antioxidantes en la prevención y/o tratamiento de las enfermedades oculares. Se seleccionaron ensayos clínicos controlados y aleatorizados publicados en los últimos 7 años. Resultados: la relación entre distintos tipos de antioxidantes, vitamina E, C, betacaroteno, zinc, luteína, antocianinas y carotenoides, sugiere una relación positiva ante el riesgo y progresión de DMAE y parámetros del glaucoma, indicando un menor riesgo de la enfermedad ante un mayor consumo de antioxidantes en la dieta. Informes iniciales sugieren un papel potencial para la modificación de la dieta en el tratamiento de la degeneración macular asociada a la edad y glaucoma principalmente, no evidenciándose para la prevención de la catarata. Conclusiones: promover el consumo adecuado de antioxidantes en la dieta puede prevenir y proteger frente a patologías oculares de gran prevalencia. Los antioxidantes del grupo de vitaminas son los más estudiados hasta el momento en las patologías oculares. Es necesario llevar a cabo más ensayos clínicos para establecer de forma más precisa estas relaciones (AU)


Introduction: The influence of antioxidants on ocular processes has taken on a significant importance in recent years for its effects on visual health. Oxidative stress is a factor involved in the development of major pathologies such as cataracts, glaucoma and age-related macular degeneration (AMD). Objective: To establish the scientific evidence that exists about the different types of antioxidants and their effects on ocular pathological processes through a systematic review. Methods: Literature search in MEDLINE, Scielo and Cochrane for studies evaluating the use of antioxidants in the prevention and/or treatment of eye diseases. Selected randomized controlled clinical trials over the past 7 years were selected. Results: The relationship between different types of antioxidants, vitamin E, C, beta carotene, zinc, lutein, anthocyanins and carotenoids, suggests a positive relationship with the risk and progression of AMD and glaucoma parameters, indicating a lower risk of the disease due to an increased consumption of antioxidants in the diet. Initial reports suggest a potential role for diet modification in the treatment of age-related macular degeneration and glaucoma primarily, not evidencing for the prevention of cataract. Conclusions: Promoting adequate consumption of antioxidants in the diet can prevent and protect against highly prevalent eye diseases. The antioxidants of the group of vitamins are the most studied so far in the ocular pathologies. More clinical trials are needed to establish these relationships more precisely (AU)

The updated mitochondrial free radical theory of aging (MFRTA) is reviewed as part of the cell aging regulatory system (CARS). Any valid theory of aging should explain why different animal species age at so different rates. Only two known parameters correlate with species longevity in the right sense: the mitochondrial rate of reactive oxygen species production (mitROSp) and the degree of fatty acid unsaturation of tissue membranes calculated as the double bond index (DBI). Both are low in long-lived animals. Dietary restriction (DR), which increases longevity, also decreases mitROSp and % free radical leak (FRL) at complex I and oxidative damage to mtDNA. This can increase longevity by decreasing mtDNA fragments accumulation inside nuclear DNA which revitalizes MFRTA. Lowered mitROSp and FRL at complex I also occurs during protein or methionine restriction, and rapamycin treatment (which also increases longevity). The decrease in mitROSp during DR (dietary restriction) is due to restriction of a single substance, methionine, and occurs at the matrix domain of complex I. This updated MFRTA focuses on low mitROSp and low sensitivity of membranes to oxidation in long-lived animals. The three best known aging effectors of the genetic Aging Program of aerobic tissues are mitROSp, membrane fatty acid unsaturation, and autophagy. This program reacts to cytoplasmic signaling proteins, influenced by nutrients, drugs and hormones, varying the activity of the mitROSp and macroautophagy aging effectors. An analogous program, although with additional gene clusters of aging involved, and different output activity, can determine longevity in different animal species (AU)


Se revisa la teoría del envejecimiento por radicales libres de origen mitocondrial (MFRTA) como parte del Sistema de Regulación Celular del Envejecimiento (CARS). Cualquier teoría del envejecimiento debe explicar porqué las especies animales envejecen a velocidades tan diferentes. Solo dos parámetros conocidos correlacionan con la longevidad de las especies en el sentido correcto: la producción mitocondrial de radicales de oxígeno (mitROSp) y el grado de insaturación de los ácidos grasos de las membranas celulares. Ambos están disminuidos en las especies longevas. La restricción calórica, de proteínas, o de metionina, y la rapamicina, que aumentan la longevidad, también disminuyen la mitROSp y la fuga % de radicales libres en el complejo I y el daño oxidativo al ADNmt. Esto puede aumentar la longevidad disminuyendo la acumulación de fragmentos del ADNmt dentro del ADN nuclear con la edad, lo cual revitaliza la MFRTA. El descenso en mitROSp durante la restricción calórica se debe sólo a la restricción de metionina, y ocurre en el dominio de membrana del complejo I. La mitROSp, el grado de insaturación de los ácidos grasos, y la autofagocitosis son los tres efectores de envejecimiento conocidos dependientes del programa genético pro-envejecimiento (PAP, que es parte del CARS). El PAP responde a proteínas de señalización celular en función de la disponibilidad de nutrientes y hormonas en los medios ambiente e interno. Este programa, aunque con más clusters génicos del envejecimiento implicados, y con diferente intensidad efectora, podría ser responsable de la regulación de la longevidad de las distintas especies animales (AU)

Objetivos. Dada la importancia en la actualidad que tiene el envejecimiento cutáneo, en este trabajo se realiza una revisión del mismo, resaltando las diferentes teorías que lo intentan explicar así como las causas que pueden inducir este envejecimiento; se revisaran los distintos compuestos utilizados en su tratamiento, tanto desde el punto de vista de la cosmética preventiva como productos utilizados como reparadores. Métodos. Se utilizó la base de datos de la National Library of Medicine, Washington, DC (MEDLINE: PubMed) así como informaciones disponibles online. Resultados. Se analizan los distintos compuestos que desde un punto de vista preventivo y reparador son utilizados, se observa como la utilización de muchos de ellos puede ejercer una acción doble, y como pueden ser utilizados tanto con una finalidad hidratante como antioxidante por ejemplo, es decir las acciones ejercidas pueden ser muy variadas y con distinta intensidad. Bajo este prisma se estudian los proteoglicanos, ácido hialurónico, péptidos, y dos hormonas de empleo mas reciente como son melatonina y DHEA. Entre los antioxidantes analizados, la vitamina E y los polifenoles parecen ser los que más evidencias a favor de su efecto beneficioso presentan. Conclusiones. El envejecimiento cutáneo debe tratarse asociando varios activos para conseguir una mayor efectividad. Por otro lado, el futuro de este tratamiento puede estar en el estudio de los genes, que nos indicarán el preparado más adecuado a cada tipo de piel. Son necesarias más investigaciones para poder avalar el concepto de genocosmética


Objectives. Currently the skin aging is very important in the cosmetic field. For this reason, in this work a review about the skin aging is made. Therefore, our work focuses on the different theories that explain the skin aging as well as the causes that can induce this aging. In addition the compounds used in its treatment are studies, both from the point of view of preventive cosmetics as products used as repairmen. Methods. To carry out this work were used different database as the National Library of Medicine, Washington, DC (MEDLINE PubMed) and a lot of interesting information available online. Results. Different compounds are analyzed from a preventive and reparative used. It is observed how the use of many of them can have a dual action, and these compounds can be used with a hydrating and antioxidant purpose moreover the effects taken may be varied and with different intensity. Therefore proteoglycans, hyaluronic acid, peptides, melatonin and DHEA, two hormones recently used, are studied. Among the analyzed antioxidants, vitamin E and polyphenols seem to be more evidence that supporting their beneficial effect Conclusions. The combination of several cosmetic compounds is a factor to be taken into account. In this sense, the future of antiaging treatment may depend of a previous genetic study and the development of appropriate formulation for each type of skin. However more studies are needed to substantiate the concept of genocosmetic

The aim of the study was to compare the innate immune system of severely malnourished children admitted to the Instituto de Medicina Integral Professor Fernando Figueira and treated according to the protocol of the World Health Organization (WHO) at admission and discharge. An experimental study was conducted with 20 children under two years of age. Ten of them had severe malnutrition and ten were a control group. The malnourished group consisted of hospitalized infants and it was submitted to WHO’s protocol. Children with HIV and re-admitted during the study period were excluded. A blood sample was taken at admission and at discharge. Later, an analysis of blood leukocytes, adherence index, phagocytic capacity, production of free radicals superoxide and nitric oxide was performed. Patients with severe malnutrition at hospital discharge showed improved phagocytic function, release of oxygen radicals and reduction of the number of lymphocytes when compared to the time of admission. When compared to the control group, patients at hospital discharge had lower lymphocyte values and lower production of free radicals. Thus, it can be concluded that the duration of hospitalization was insufficient to restore cell-mediated immunity and microbicide activity (AU)


El objetivo del estudio fue comparar el sistema inmune innato de niños con malnutrición grave ingresados en el Instituto de Medicina Integral Professor Fernando Figueira, tratados de acuerdo con el protocolo de la Organización Mundial de la Salud (OMS), al ingreso y al alta hospitalaria. Se llevó a cabo un estudio experimental con 20 niños menores de dos años de edad, 10 con malnutrición grave y 10 niños del grupo de control. El grupo de malnutridos se compuso de lactantes hospitalizados y sometidos al protocolo de la OMS. Se excluyeron los niños afectados por el HIV y los readmitidos durante el período del estudio. Se recogió una muestra de sangre al ingreso y otra al alta, y posterioriormente se realizó el análisis del perfil leucocitario, y el índice de adherencia, la capacidad fagocítica y la producción de los radicales libres superóxido y óxido nítrico. Los pacientes con malnutrición grave en el alta hospitalaria mostraron mejoría de la función fagocítica, la liberación de radicales oxidantes y la reducción del número de linfocitos en comparación con el ingreso hospitalario. En comparación con el grupo de control, los pacientes en el alta hospitalario presentaron valores más bajos de linfocitos y de producción de radicales libres. Por lo tanto, se puede concluir que el tiempo de hospitalización fue insuficiente para restablecer la inmunidad mediada por células, así como para restaurar la actividad microbicida (AU)

Un desequilibrio entre la producción de radicales libres y defensas antioxidantes puede conducir a un estado de estrés oxidativo que podría causar un cambio en el equilibrio redox intracelular generando un entorno más oxidante, con la posibilidad de causar enfermedad. El síndrome de Down (SD) es el primer síndrome cromosómico clínicamente definido y está causado por la presencia de tres copias del cromosoma 21. Es uno de los defectos congénitos humanos más importantes, con una incidencia de 1 de cada 700-1000 nacimientos. Se sabe que la actividad física, si se practica regularmente, es un factor importante para la salud; sin embargo, practicada de forma exhaustiva e intensa incrementa la formación de radicales libres y de especies reactivas de oxígeno y nitrógeno (ERON). Para la contracción muscular es necesaria una cantidad mínima de ERON. Por otra parte, el estrés oxidativo que resulta de la concentración de ERON en músculo está asociado con fatiga muscular durante la contracción y daño muscular después del ejercicio. Las principales fuentes de ERON durante y después del ejercicio son: cadena respiratoria mitocondrial, isquemia reperfusión y reacción inflamatoria. Sin embargo, los resultados recogidos en la bibliografía muestran efectos beneficiosos del ejercicio en el SD, aunque los efectos del ejercicio en parámetros de estrés oxidativo han sido muy poco estudiados en el SD (AU)


An imbalance between free radical production and antioxidant defence leads to an oxidative stress state that can potentially result in a change in the intracellular redox balance towards a more oxidizing environment, with the possibility of causing ill-health and disease. Down’s syndrome (DS) is the first clinically defined chromosomal syndrome, and is caused by the presence of three copies of chromosome 21. It is one of the most significant human congenital defects, with an incidence of 1 in every 700-1000 births. Regular physical activity is known as an important factor for health. However, exhaustive and/or intense physical activity increases free radical and reactive oxygen/nitrogen species (RONS) production. A minimal amount of RONS is necessary for muscular contraction. Nevertheless, oxidative stress, which results in muscle-increased RONS concentration, is associated with muscular fatigue during contraction and in post-exercise muscular damage suffering. The mitochondrial respiratory chain, the ischemia reperfusion phenomenon and the inflammatory reaction have been identified as the main sources of RONS during and after exercise. However, results reported in literature seem to show a beneficial effect of exercise on DS subjects, although the effect of exercise on oxidative stress parameters has been poorly studied in this condition (AU)

Excess fat accumulation renders the liver more vulnerable to ethanol, but it is still unclear how alcohol enhances lipid dysmetabolism and oxidative stress in a pre-existing steatosis condition. The effects produced by binge ethanol consumption in the liver of male Wistar rats fed a standard (Ctrl) or a high-fat diet HFD were compared. The liver status was checked through tissue histology and standard serum parameters. Alteration of hepatic lipid homeostasis and consequent oxidative unbalance were assessed by quantifying the mRNA expression of the lipid-regulated peroxisome proliferator-activated receptors (PPARs), of the cytochromes CYP2E1 and CYP4A1, and of some antioxidant molecules such as the metallothionein isoforms MT1 and MT2 and the enzymes catalase and superoxide dismutase. The number of adipose differentiation-related protein (ADRP)-positive lipid droplets (LDs) was evaluated by immunohistochemical staining. As a response to the double insult of diet and ethanol the rat liver showed: (1) a larger increase in fat accumulation within ADRP-positive LDs; (2) stimulation of lipid oxidation in the attempt to limit excess fat accumulation; (3) induction of antioxidant proteins (MT2, in particular) to protect the liver from the ethanol-induced overproduction of oxygen radicals. The data indicate an increased susceptibility of fatty liver to ethanol and suggest that the synergistic effect of diet and ethanol on lipid dysmetabolism might be mediated, at least in part, by PPARs and cytochromes CYP4A1 and CYP2E1 (AU)

Introducción: La diabetes se asocia a un incremento en la peroxidación de lípidos, cuantificada a partir del nivel de sustancias reactivas al ácido tiobarbitúrico (TBARS). En paralelo, se activa el sistema de defensa antioxidante (SDA) para delimitar el daño. Objetivo: Determinar el grado de peroxidación de lípidos en individuos obesos diabéticos tipo 2 (DM2) y la respuesta del SDA en comparación con individuos con DM2 sin obesidad. Método: Se evaluó el daño a lípidos a través de la medición de las TBARS en dos grupos de 30 individuos. Se evaluó la respuesta del SDA por medio de la medición de la actividad de las enzimas catalasa (CAT), superóxido dismutasa (SOD) y glutatión peroxidasa (GPx). Resultados: El grupo de DM2 obesos presentó un índice de masa corporal (IMC) promedio de 38,6 ± 3,5 kg m2 en comparación con el grupo control 24,7 ± 3,6 kg m2 (p < 0,01). Los niveles de TBARS en el grupo en estudio fueron más altos en comparación al grupo control (p < 0.01). En un análisis de regresión lineal múltiple la actividad de SOD y CAT explicó los niveles de TBARS en el obeso con DM2. Conclusión: Los niveles de TBARS sugieren mayor daño por estrés oxidativo en DM2 obesos por un exceso en la producción de radicales libres (RL), así como incapacidad del SDA para delimitar el daño (AU)


Introduction: Diabetes is associated with increased lipid peroxidation, quantified as the levels of thiobarbituric acid reactive substances (TBARS). In parallel, the antioxidant defense system (ADS) reacts to diminish the oxidative damage. Objective: To determine the levels of lipid peroxidation and the activity of antioxidant enzymes in obese type 2 diabetic (DM2) individuals compared to non-obese DM2 individuals. Methods: Lipid peroxidation was quantified by measuring TBARS and the ADS response by measuring the activity of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx). Results: Two groups of 30 subjects were studied. The obese DM2 group had a mean body mass index (BMI) 38.6 ± 3.5 kg m-2 compared to the control group 24.7 ± 3.6 kg m-2 (p<0.01). TBARS levels in the study group were higher compared to the control group (p <0.01). Multiple linear regression analysis suggested that activities of SOD and CAT adjusted to lipid peroxidation (TBARS) in the obese DM2 individuals. Conclusion: TBARS levels suggest greater oxidative damage in obese DM2 subjects with a diminished response of ADS (AU)

Objetivo: Determinar el grado de Estrés Oxidativo en pacientes obesos mórbidos comparando los resultados con los de una población normal. Material y método: Hemos estudiado los metabolitos más representativos del EO, tanto en sangre (MDA, 8-oxo-dG, GSSG y la relación GSSG/GSH) como en orina (8-oxo-dG), así como el antioxidante GSH. Realizamos un análisis descriptivo de la muestra. Se realizó la prueba de Kolmogorv-Smirnov para evaluar si la distribución de los distintos metabolitos seguía un modelo de normalidad. En los casos de distribución normal, se empleó la T de Student para comparar medias, utilizando la U de Mann-Whitney para los datos no paramétricos, utilizando en los contrastes de hipótesis el nivel de significación p < 0,05. Resultados: Los pacientes fueron 28 en cada grupo, sin diferencias estadísticamente significativas en cuanto a edad y sexo. El grupo de pacientes con obesidad mórbida presentó un IMC medio de 50,1 ± 4 y de 23,9 ± 6 el grupo normopeso. Un 67,8% de los pacientes obesos mórbidos presentaron comorbilidades. No había patología asociada en el grupo control. Todos los valores de los distintos metabolitos de EO fueron más elevados en el grupo de obesos mórbidos que en el grupo control mientras que la actividad de los sistemas antioxidantes (GSH) fue menor en el grupo de obesos mórbidos. Conclusión: Los valores de los metabolitos de EO obtenidos en el grupo de obesos mórbidos confirma la presencia de EO en la obesidad, de un modo que se puede considerar patológico dadas las diferencias obtenidas en el grupo de población normal (AU)


OBJECTIVE: To determine the level of oxidative stress in morbid obese patients by comparing their results to those of a normal population. MATERIAL AND METHODS: We have studied the metabolites most representative of OS, both in the blood (MDA, 8-oxo-dG, GSSG and the ratio GSSG/GSH) and in the urine (8-oxo-dG), as well as the GSH antioxidant. A descriptive analysis of the sample was performed. The Kolmogorv-Smirnov test was used to assess whether the distribution of the different metabolites was normal. In the case of normal distribution, the Student's t test was used to compare the means, the Mann-Whitney U test was used for non-parametric data, with a significance level of p < 0.05 for hypothesis contrast. RESULTS: There were 28 patients in each group, without statistically significant differences regarding age and gender. The group of patients with morbid obesity presented an average BMI of 50.1 ± 4 and 23.9 ± 6 in the group with normal weight. 67.8% of the patients with morbid obesity had other comorbidities. There were no associated pathologies in the control group. All the values for the different OS metabolites were higher in the group of patients with morbid obesity than in the control group, whereas the activity of the antioxidant systems (GSH) was lower in the group with morbid obesity. CONCLUSION: The figures of OS metabolites obtained in the group of patients with morbid obesity confirm the presence of OS in obesity at a pathological level given the differences obtained in the group of normal population (AU)

RESUMO

This mini-review tries to summarize the main interdependences between the free radicals of oxygen, nitrogen, and carbon. Also, the main metabolic pathways for these radical species are described, as well as how these affect their interaction and functional implications. Emphasis is made on the metabolic disturbances induced by stressing aggressions that produce radical species. In this way, cellular oxidative imbalances created by the superiority of reactive oxygen species over the antioxidant systems produce both activation of nitroxide synthases and the oxidation of terminal nitrogen from L-arginine, as well as the metabolization of heme until carbon monoxide by nitric oxide-activated hemoxygenase. Also, multiple cellular protein and nucleoprotein alterations determined by these three kinds of radical species are completed by the involvement of hydrogen sulfide, which results from the degradation of L-cysteine by cistationine-ã-lyase. In this way, sufficient experimental data tend to demonstrate the involvement of hydrogen sulfide and other thiol derivatives in the interrelations between oxygen, nitrogen, and carbon, which results in a true radical cascade. Thus, oxidative stress, together with nitrosative and carbonilic stress, may constitute a central point where other factors of vulnerability meet, and their interactions could have an important impact in many modern diseases. Considering that the actions of reactive species can be most of the time corrected, future studies need to establish the therapeutical importance of various agents which modulate oxidative, nitrosative, or carbonilic stress (AU)
