RESUMO

The recent introduction of the cultivation of Steviarebaudiana Bertoni in Mexico has gained interest for its potential use as a non-caloric sweetener, but some other properties of this plant require studies. Extracts from two varieties of S. rebaudiana Bertoni adapted to cultivation in Mexico were screened for their content of some phytochemicals and antioxidant properties. Total pigments, total phenolic and flavonoids contents of the extracts ranged between 17.7-24.3 mg/g, 28.7-28.4 mg/g, and 39.3-36.7 mg/g, respectively. The variety «Criolla» exhibited higher contents of pigments and flavonoids. Trolox equivalent antioxidant capacity ranged between618.5-623.7 mM/mg and DPPH decolorization assay ranged between 86.4-84.3%, no significant differences were observed between varieties. Inhibition of β-carotene bleaching ranged between 62.3-77.9%, with higher activity in the variety «Criolla». Reducing power ranged between 85.2-86% and the chelating activity ranged between57.3-59.4% for Cu2+ and between 52.2-54.4% forFe2+, no significant differences were observed between varieties. In conclusion, the results of this study showed that polar compounds obtained during the extraction like chlorophylls, carotenoids, phenolic compounds and flavonoids contribute to the antioxidative activity measured. The leaves of S. rebaudiana Bertoni could be used not only as a source of non-caloric sweeteners but also naturally occurring antioxidants (AU)


La reciente introducción del cultivo de Stevia rebaudiana Bertoni en México ha ganado interés debido a su potencial uso como fuente de edulcorantes no calóricos, pero otras propiedades de esta planta aun requieren de estudios. Extractos de hojas de dos variedades de S. rebaudiana Bertoni adaptadas al cultivo en México fueron evaluados en cuanto a su contenido de algunos fitoquímicos y sus propiedades antioxidantes. El contenido de pigmentos, fenoles totales y flavonoides en los extractos, osciló entre 17.7-24.3 mg/g, 28.7-28.4 mg/g, y 39.3-36.7 mg/g, respectivamente. La variedad «Criolla» exhibió los mayores contenidos de pigmentos y flavonoides. La capacidad antioxidante equivalente de Trolox osciló entre 618.5-623.7 mM/mg y el ensayo de decoloración del radical DPPH osciló entre 86.4-84.3%, no observándose diferencias significativas entre ambas variedades. La inhibición de la decoloración del β-caroteno osciló entre 62.3-77.9%, siendo mayor en la variedad «Criolla». El poder reductor osciló entre 85.2-86%, las capacidades quelantes de cobre y hierro oscilaron entre 57.3-59.4% y 52.2-54.4%, respectivamente, no observándose diferencias significativas entre ambas variedades. En conclusión, los resultados de este estudio demuestran que los compuestos de naturaleza polar obtenidos durante la extracción, tales como pigmentos clorofílicos, carotenoides, compuestos fenólicos y flavonoides contribuyen a la actividad antioxidante. Las hojas de S. rebaudiana Bertoni podrían ser empleadas no solo como fuente de edulcorantes no calóricos, sino también como fuente de antioxidantes de origen natural (AU)

Thylakoid membranes of the cryptophyte Chroomonas sp. strain LT were solubilized with dodecyl-beta-maltoside and subjected to sucrose density gradient centrifugation. The four pigment protein complexes obtained were subsequently characterized by absorption and fluorescence spectroscopy, SDS-PAGE, and Western immunoblotting using antisera against the chlorophyll a/c-binding proteins of the marine cryptophyte Cryptomonas maculata and the reaction-center protein D2 of photosystem II of maize. Band 1 consisted mainly of free pigments, phycobiliproteins, and chlorophyll-a/c-binding proteins. Band 2 represented a major chlorophyll a/c-binding protein fraction. A mixture of photosystem II and photosystem I proteins comprised band 3, whereas band 4 was enriched in proteins of photosystem I. Western immunoblotting demonstrated the presence of chlorophyll a/c-binding proteins and their association with photosystem I in band 4. Phosphorylation experiments showed that chlorophyll a/c-binding proteins became phosphorylated. Negative staining electron microscopy of band B4 revealed photosystem I particles with dimensions of 22 nm. Our work showed that PSI-LHCI complexes of cryptophytes are similar to those of Chlamydomonas rheinhardtii, the diatom Phaeodactylum tricornutum, and higher plants (AU)

The diatom Cyclotella cryptica was grown under low- and high-intensity white light of 50 and 500 µmol photons m–2 s–1, respectively. Western immunoblotting showed that the diatom adapted its light-harvesting apparatus, giving rise to different amounts of distinct fucoxanthin chlorophyll a/c binding polypeptides (Fcp). The amount of Fcp2 was approximately two-fold higher under low-light than under high-light conditions, whereas the amount of Fcp6 increased four- to five-fold under high-light conditions. For Fcp4, no significant differences were detected in response to either light regime. Cells of Cyclotella grown under high- and low-light intensity were subjected to immunoelectron microscopy. Quantification of the gold label, expressed as gold particles per µm2, confirmed the results obtained by Western immunoblotting. Exposure to low light resulted in the detection of approximately six times more Fcp2-bound gold particles per µm2 in thylakoid membranes, whereas in cells grown under high light the number of Fcp6-bound gold particles increased ten-fold. For Fcp4, similar amounts of gold particles per µm2 were counted under the two light regimes. These immunocytochemical results confirmed molecular data derived from phylogenetic analyses of the sequences of genes encoding fucoxanthin chlorophyll a/c binding polypeptides (fcp genes) and from measurements of steady-state fcp mRNA concentrations. The results show that Fcp2 and Fcp6 accumulate under low- and high-light intensity, respectively, whereas Fcp4 seems to be constitutively synthesized (AU)


La diatomea Cyclotella cryptica se cultivó a intensidades de luz blanca baja y alta (50 y 500 fotones m–2 s–1 µmol, respectivamente). La inmunotransferencia mostró que la diatomea adaptó su aparato captador de luz, produciendo cantidades diferentes de polipéptidos de unión a la fucoxantina clorofila a/c (Fcp). A intensidad baja de luz, la cantidad de Fcp2 era aproximadamente el doble que a intensidad elevada, mientras que a intensidad elevada la cantidad de Fcp6 era de cuatro a cinco veces la cantidad producida a baja intensidad. No se detectaron diferencias significativas para Fcp4 como respuesta a ninguno de los dos tipos de luz. Las células de Cyclotella cultivadas a intensidades alta y baja de luz se observaron por microscopia immunoelectrónica. La cuantificación del marcado de oro, expresada como partículas de oro por µm2 confirmó los resultados obtenidos por inmunotransferencia. Tras la exposición a luz de baja intensidad se detectaron aproximadamente seis veces más partículas oro unidas a Fcp2 por µm2 en las membranas de los tilacoides, mientras que en las células cultivadas a intensidad elevada, el número de partícula de oro unidas a Fcp6 aumentó diez veces. Para Fcp4, se contaron cantidades similares de partículas oro por µm2 en los dos regímenes de luz. Estos resultados immunocitoquímicos confirmaron los datos moleculares derivados de los análisis filogenéticos de las secuencias de los genes que codifican los polipéptidos de unión de la fucoxantina clorofila a/c (genes fcp) y de la medida de las concentraciones de mRNA fcp en el estado estacionario. Los resultados demuestran que Fcp2 y Fcp6 se acumulan, respectivamente, a intensidades de luz alta y baja, mientras que Fcp4 parece sintetizado de manera constitutiva (AU)

The steady-state mRNA concentrations of two fcp genes encoding fucoxanthin chlorophyll a/c light-harvesting polypeptides of the centric diatom Cyclotella cryptica were investigated over a 4-day period by RNA dot-blotting experiments. Before and during the first day of the experiment, the cultures were grown under a 12-h light/12-h dark regime. On the following 3 days, the algae were kept in darkness. On the first day, the steady-state mRNA concentration of fcp2 followed a diurnal pattern, with a maximum occurring around noon, approximately 6h after the onset of light. The gene fcp6 also had a diurnal pattern on the first day. Its maximum, however, occurred immediately after the onset of light. During the subsequent incubation period in darkness, the diurnal pattern of expression of both fcp genes continued, thus demonstrating that their steady-state mRNA concentrations oscillated in a circadian manner (AU)


A lo largo de períodos de cuatro días, se estudiaron las concentraciones de mRNA en el estado estacionario correspondientes a dos genes fcp que codifican los polipéptidos captadores de luz de las clorofilas a y c y de la fucoxantina de la diatomea céntrica Cyclotella cryptica. Se hicieron experimentos de hibridación puntual de RNA. Antes del experimento y durante el primer día del mismo, el cultivo se realizó en régimen de alternancia luz-oscuridad de 12 horas y los tres días siguientes las algas se mantuvieron en la oscuridad. El primer día, la concentración de mRNA del gen fcp2 en el estado estacionario seguía un modelo diurno, con un máximo alrededor del mediodía, unas seis horas después de iniciarse la fase iluminada. El gen fcp6también siguió un modelo diurno el primer día, si bien alcanzaba el valor máximo justo al principio de la fase iluminada. Durante el período subsiguiente de incubación en la oscuridad se mantuvo el modelo diurno de ambos genes, lo que demuestra que sus concentraciones de mRNA en el estado estacionario oscilaban de manera circadiana (AU)

RESUMO

Microorganisms are but a few micrometers in diameter and are not visible to the naked eye. Yet, the large numbers of microorganisms present in the oceans and the global impact of their activities make it possible to observe them from space. Here a few examples of how microorganisms can be studied from satellites are presented. The first case is the best known: the main pigment used in photosynthesis (chlorophyll a) can be determined from satellites. These kinds of studies have contributed a tremendous amount of understanding about the distribution and dynamics of primary production in the oceans. Two other examples will concern analysis of heterotrophic prokaryotic production and estimates of dimethyl sulfide (DMS) concentration and flux to the atmosphere. These three processes are of fundamental importance for the functioning of the biosphere. Marine microbes carry out about half of the total primary production in the planet. A substantial fraction of the respiration in the oceans is due to the activity of heterotrophic prokaryotes. Finally, the flux of DMS to the atmosphere is believed to constitute one of the mechanisms by which the biota can regulate climate. The global implications of microbial processes in the oceans can only be addressed with the help of satellites (AU)


Los microorganismos no miden más que unos micrómetros de diámetro y no son visibles a simple vista. Sin embargo, el gran número de microorganismos presentes en los océanos y el impacto global de sus actividades hacen posible su observación desde el espacio. Este estudio presenta algunos ejemplos de cómo pueden estudiarse los microorganismos desde los satélites. El primer caso es el más conocido: el principal pigmento utilizado en la fotosíntesis (la clorofila a) puede determinarse desde un satélite. Este tipo de estudios han contribuido de forma espectacular a la comprensión de la distribución y dinámica de la producción primaria en los océanos. Otros dos ejemplos tienen que ver con la producción de los procariotas heterótrofos y con las estimaciones de la concentración de sulfuro de dimetilo (DMS) y de su flujo hacia la atmósfera. Estos tres procesos son de enorme importancia para el funcionamiento de la biosfera. Los microorganismos marinos llevan a cabo aproximadamente la mitad de la producción primaria total del planeta. Una parte importante de la respiración en los océanos se debe a la actividad de los procariotas heterótrofos. Por último, el flujo de DMS hacia la atmósfera constituye uno de los mecanismos por los cuales la biota puede regular el clima. La implicación global de los procesos microbianos en los océanos sólo puede estudiarse con ayuda de los satélites (AU)
