Introducción y objetivos Nuestro objetivo es evaluar los resultados de la fitoterapia a largo plazo, centrándonos en el intervalo entre la fitoterapia y los tratamientos farmacológicos y los factores de riesgo que predisponen a dicho cambio en un seguimiento de 10 años. Material y métodos Se revisaron retrospectivamente los datos de los pacientes varones que tomaban fitoterapia para los síntomas del tracto urinario inferior (STUI) de leves a moderados entre enero y diciembre de 2010, a partir de una base de datos mantenida prospectivamente. Se realizó un seguimiento de los pacientes durante 10 años mediante consultas médicas presenciales y telefónicas. Resultados Ciento dos pacientes se sometieron al menos a un ciclo de fitoterapia para los STUI. Veinte (19,6%) pacientes resolvieron sus síntomas tras un ciclo fitoterápico y abandonaron el tratamiento, 27 (26,4%) continuaron con la fitoterapia y 52 (51%) cambiaron a bloqueadores alfa y/o inhibidores de la 5a-reductasa tras un intervalo medio de 24 meses. El motivo del cambio de tratamiento fue la sintomatología (n=45) o la progresión clínica (aumento del volumen residual n=15; retención urinaria, n=5). Los pacientes que cambiaron a fármacos sintéticos tenían una mediana de edad más alta (60 frente a 49), mayor volumen prostático (40 frente a 26cc) y antígeno prostático específico (1,9 frente a 0,9ng/ml), volumen residual más elevado (40 frente a 0cc) y una tasa de flujo máximo (Qmáx) más baja (12 frente a 15ml/s) en el momento de la presentación. Conclusiones El 46% de los pacientes con STUI leves o moderados sometidos a fitoterapia estarán libres de tratamiento o seguirán con la fitoterapia a los 10 años de la presentación de la enfermedad. Los pacientes de mayor edad, con próstatas más grandes, con volumen residual y antígeno protático específico más alto, deben ser informados sobre un mayor riesgo de progresión sintomática o clínica (AU)


Introduction and objectives Our objective is to assess the long-term results of phytotherapy, focusing on the interval between phytotherapy and pharmacological treatment and the predisposing risk factors to such switch on a 10-year follow-up. Material and methods The data of patients taking phytotherapy for mild to moderate male lower urinary tract symptoms (LUTS) from January to December 2010 were retrospectively reviewed from a prospectively maintained database. Patients were followed for 10 years through medical visits and telephone consultations. Results 102 patients underwent at least one cycle of phytotherapy for LUTS. Twenty (19.6%) patients resolved their symptoms after one phytotherapy cycle and stopped any treatment, 27 (26.4%) continued phytotherapy, and 52 (51%) switched to alpha-blockers and/or 5a-reductase inhibitors after a median interval of 24 months. The reasons for treatment switch were symptoms (n=45) or clinical progression (increased residual volume n=15; urinary retention, n=5). Patients switching to synthetic drugs had median higher age (60 vs 49), prostate volume (40 vs 26cc), prostate specific antigen (PSA) (1.9 vs 0.9ng/ml), residual volume (40 vs 0cc), and a lower maximum flow rate (Qmax) (12 vs 15ml/sec) at presentation. Conclusions 46% patients with mild to moderate LUTS undergoing phytotherapy will be either free of treatment or still on phytotherapy at 10 years from disease presentation. Older patients with larger prostates, increased residual volume and PSA, should be informed regarding their higher risk of symptomatic or clinical progression: the risk of a treatment switch to alpha-blockers or 5a-reductase inhibitors becomes an actual fact after an average span of 2 years (AU)

Background: Topiroxostat, an inhibitor of xanthine oxidoreductase (XOR) was shown to reduce urinary albumin excretion of hyperuricemic patients with chronic kidney disease. However, its pharmacological mechanism is not well understood. In this study, we examined the effects of topiroxostat on glomerular podocytes. Podocyte is characterized by foot process and a unique cell-cell junction slit diaphragm functioning as a final barrier to prevent proteinuria. Methods: The effects of topiroxostat on the expressions of podocyte functional molecules were analysed in db/db mice, a diabetic nephropathy model, anti-nephrin antibody-induced rat podocyte injury model and cultured podocytes treated with adriamycin. Results: Topiroxostat treatment ameliorated albuminuria in db/db mice. The expression of desmin, a podocyte injury marker was increased, and nephrin and podocin, key molecules of slit diaphragm, and podoplanin, an essential molecule in maintaining foot process were downregulated in db/db mice. Topiroxostat treatment prevented the alterations in the expressions of these molecules in db/db mice. XOR activity in kidney was increased in rats with anti-nephrin antibody-induced podocyte injury. Topiroxostat treatment reduced XOR activity and restored the decreased expression of nephrin, podocin and podoplanin in the podocyte injury. Furthermore, topiroxostat enhanced the expression of podoplanin in injured human cultured podocytes. (AU)


Antecedentes: El topiroxostat, un inhibidor de la xantina oxidorreductasa (XOR), mostró reducir la excreción de albúmina en la orina de pacientes hiperuricémicos con enfermedad renal crónica. Sin embargo, su mecanismo farmacológico no se conoce con exactitud. En este estudio examinamos los efectos del topiroxostat en los podocitos glomerulares. El podocito se caracteriza por unas prolongaciones en forma de pie y un diafragma de hendidura de unión célula-célula único que funciona como barrera final en la prevención de la proteinuria. Métodos: Se analizaron los efectos del topiroxostat en las expresiones de las moléculas funcionales de los podocitos en ratones db/db, en un modelo de nefropatía diabética, en un modelo de lesión podocitaria inducida por anticuerpos antinefrina en ratas y en podocitos cultivados tratados con adriamicina. Resultados: El tratamiento con topiroxostat mejoró la albuminuria en ratones db/db. La expresión de la desmina, un marcador de lesión podocitaria, estaba aumentada, y la nefrina y la podocina, moléculas clave del diafragma de hendidura, y la podoplanina, una molécula esencial en el mantenimiento de las prolongaciones en forma de pie, estaban atenuadas en los ratones db/db. El tratamiento con topiroxostat evitó alteraciones en las expresiones de estas moléculas en los ratones db/db. La actividad de la XOR en el riñón se incrementó en ratas con lesión podocitaria inducida por anticuerpos antinefrina. El tratamiento con topiroxostat redujo la actividad de la XOR y restauró la disminución de la expresión de nefrina, podocina y podoplanina en la lesión podocitaria. Además, el topiroxostat aumentó la expresión de podoplanina en podocitos humanos cultivados lesionados. (AU)

Purpose Molecular mechanisms of uveal melanoma development in association with high pigmentation are unclear. Tyrosinase Related Protein (TYRP1) is not only one of the important melanogenesis marker that contributes to melanin synthesis, but can also prevents the melanocyte death. The induction of melanogenesis leads to induction of HIF-1α which can affect the behavior of melanoma cells and its surrounding environment. The aim of our study was to determine the expression of TYRP1 and HIF-1α at the protein and RNA level and determine its prognostic significance. Methods In the present study, the expression of TYRP1 and HIF-1α was investigated on 61 formalin-fixed paraffin-embedded choroidal melanoma samples by immunohistochemistry. Fresh 50 samples were validated by real-time PCR. Results were correlated with clinicopathological parameters and Kaplan–Meier was performed to determine the prognostic significance. Results High immunoexpression of TYRP1 and HIF-1α was present in 61 and 54% of patients, respectively. Both TYRP1 and HIF-1α correlated well with high pigmentation and BAP1 (BRCA1 Associated Protein-1) loss (p < 0.05) at IHC level as well as transcriptional level. There was reduced metastatic free survival in patients with necrosis and this was statistically significant (p = 0.010). Conclusion Our findings indicate that TYRP1 can be used as a potential biomarker in the development of targeted therapy in UM. Further studies on melanogenesis markers associated with TYRP1 could provide us a better understanding in this field (AU)

The present study attempts to identify the novel inhibitors of shikimate dehydrogenase (SD), the enzyme that catalyzes the fourth reaction in the shikimate pathway, through virtual screening and toxicity studies. Crystal structure of SD was obtained from Protein Data Bank (PDB ID 4P4G, 1.7 Å) and subjected to energy minimization and structure optimization. A total of 13,803 compounds retrieved from two public databases and used for the virtual screening based on physicochemical properties (Lipinski rule of five) and molecular docking analyses. A total of 26 compounds with good AutoDock binding energies values ranging between −12.03 and −8.33 kcal/mol was selected and further filtered for absorption distribution metabolism excretion and toxicity analyses (ADMET). In this, eight compounds were selected, which satisfied all the ADME and toxicity analysis properties. Three compounds with better AutoDock binding energies values (ZINC12135132, −12.03 kcal/mol; ZINC08951370, −10.04 kcal/mol; and ZINC14733847, 9.82 kcal/mol) were considered for molecular dynamic (MD) simulation and molecular generalized born surface area (MM-GBSA) analyses. The results of the analyses revealed that the two ligands (ZINC12135132 and ZINC08951370) had better inhibitory activities within their complexes, after the 50-ns MD simulation, which suggested that the complexes formed stable conformation. It is noteworthy that compounds identified by docking, MD simulation, and MM-GBSA methods could be a drug for tuberculosis which required further experimental validation

Iron is a trace element and a structural part of antioxidant enzymes, and its requirements vary according to age and gender. We hypothesized that iron deficiency (ID) leads to an increase in free radicals which mainly affect the brain, and the severity of damage would therefore be dependent on age and gender. Two groups of Wistar rats were evaluated evolutionarily: 100 rats (50 males; 50 females) with ID diet and 100 rats (50 males; 50 females) with standard diet. Both groups were offspring from mothers who were previously under the same dietary intervention. The ages studied roughly correspond to stages of human development: birth (0 postnatal day "PND" in rats), childhood (21 PND), early adolescence (42 PND), late adolescence (56 PND), and adulthood (70 PND). The following biomarkers in the brain, blood, and liver were analyzed: lipid peroxidation products (LPO), protein carbonyl content and activity of the antioxidant enzymes, superoxide dismutase, catalase, and glutathione peroxidase. It was demonstrated that ID subjects are born with high levels of LPO in the brain and low antioxidant activity, the damage being more severe in males. After birth, antioxidant defense focuses on the central level (brain) in ID females and on the peripheral level (blood and liver) in ID males. In two critical stages of development, birth and late adolescence, antioxidant protection is insufficient to counteract oxidative damage in ID subjects. Moreover, we observed that the variability of results in the literature on oxidative stress and ID comes from gender and age of the subjects under study. With this, we can establish patterns and exact moments to carry out studies or treatments (AU)

Introduction. The aim of this study was to determine the usefulness of oxidase test and time-to-positivity (TTP) in aerobic and anaerobic blood culture vials to detect the presence of Pseudomonas aeruginosa in patients with Gram-negative bacilli (GNB) bacteraemia. Material and methods. TTP was recorded for each aerobic and anaerobic blood culture vial of monomicrobial bacteraemia due to GNB. Oxidase test was performed in a pellet of the centrifuged content of the positive blood culture. An algorithm was developed in order to perform the oxidase test efficiently taking into account TTP and type of vial. Results. A total of 341 episodes of GNB bacteraemia were analysed. Sensitivity, specificity, positive predictive value and negative predictive value of the oxidase test performed on positive vials with GNB to predict P. aeruginosa were 95%, 99%, 91%, and 99%, respectively. When growth was first or exclusively detected in anaerobic vials, P. aeruginosa was never identified hence the performance of the oxidase test could be avoided. When growth was only or first detected in aerobic vials, a TTP≥8h predicted P. aeruginosa in 37% or cases (63 of 169), therefore oxidase test is highly recommended. Conclusions. Oxidase test performed onto positive blood culture vials previously selected by TTP and type of vials is an easy and inexpensive way to predict P. aeruginosa. In most cases, this can lead to optimization of treatment in less than 24 hours (AU)


Introducción. El objetivo del estudio fue determinar la utilidad de la prueba de oxidasa y del tiempo de positividad del hemocultivo (TPH) para detectar la presencia de Pseudomonas aeruginosa en pacientes con bacteriemia por bacilos gramnegativos (BGN). Material y métodos. Se registró el TPH de cada vial aerobio y anaerobio en todos los episodios de bacteriemia monomicrobiana por BGN. La prueba de oxidasa se realizó sobre el contenido centrifugado del hemocultivo positivo. Se diseñó un algoritmo para optimizar la realización de la prueba de oxidasa según el TPH y el tipo de vial. Resultados. Se analizaron 341 episodios de bacteriemia por BGN. La sensibilidad, especificidad, valor predictivo positivo y valor predictivo negativo de la prueba de oxidasa para predecir P. aeruginosa fueron del 95%, 99%, 91% y 99%, respectivamente. Cuando el crecimiento se detectó primero o exclusivamente en viales anaerobios, nunca se identificó P. aeruginosa pudiendo evitar la realización de la prueba de oxidasa. Cuando el crecimiento se detectó antes o exclusivamente en viales aerobios un TPH ≥8h predijo la presencia de P. aeruginosa en el 37% de los casos (63 de 169), por lo que es recomendable la realización de la prueba de oxidasa. Conclusiones. La prueba de oxidasa realizada a viales de hemocultivos positivos previamente seleccionados por el TPH y el tipo de medio es una forma fácil y económica de predecir P. aeruginosa. En la mayoría de los casos, esto puede contribuir a la optimización del tratamiento antibiótico en menos de 24h (AU)

The aim of this study was to identify demographic and modifiable lifestyle factors that may be related to endogenous oxidant and antioxidant activity measured in blood specimens from putatively healthy women recruited at the Roswell Park Cancer Institute (Buffalo, NY, USA). Total glutathione (TGSH), catalase (CAT), CuZn-superoxide dismutase (CuZn-SOD), glutathione peroxidase (GPx), glutathione reductase (GR), and myeloperoxidase (MPO) activity, and total antioxidant capacity (TAC) were measured in 124 healthy women, and associations with epidemiological factors were tested using general linear models. There were significant differences in oxidant and antioxidant enzyme activities according to lifestyle factors, after adjusting for duration of blood storage and season of blood draw. Compared to women who consumed ≤2.8 servings of fruits and vegetables daily, those consuming >5.3 servings had on average 31 % lower MPO activity (p-trend = 0.02), as a marker of oxidative stress, 16 % higher antioxidant GPx activity (p-trend = 0.08), and 9 % higher TAC (p-trend = 0.05). Obese women (body mass index, BMI ≥ 30) in contrast showed 17 % lower antioxidant GPx activity, 44 % higher MPO activity (p-trend = 0.03), and 10 % higher TAC (p-trend = 0.03) compared to women with normal BMI < 25. Smoking was associated with higher TGSH activity (p-trend = 0.01) and lower TAC (p-trend = 0.05). Higher TAC levels were most strongly associated with increasing age (standardized β = 0.40, p < 0.0001), BMI (standardized β = 0.17, p = 0.03), and GPx activity (standardized β = 0.23, p = 0.005), and inversely associated with CuZn-SOD activity (standardized β = −0.14, p = 0.07). Physical activity levels, multivitamin use, and alcohol intake were not associated with TAC. Our data indicate that endogenous oxidant and antioxidant enzyme activities are associated with lifestyle factors and, therefore, may be potentially modifiable, with implications for risk reduction of chronic conditions related to oxidative stress (AU)

Mass spectrometry has become a reference resource for identifying microorganisms in clinical microbiology services. One hundred and fifty one clinical isolates were selected from respiratory specimens routinely identified as Streptococcus pneumoniae (43), Haemophilus influenzae (64) and Moraxella catarrhalis (44). These identifications were compared with other phenotypical methods and mass spectrometry (MALDI -TOF -MS Vitek). Result discrepancies were assessed by 16S rRNA sequencing. Thirty-eight of the 43 strains of S. pneumoniae (86%) were identified as such using phenotypical methods and spectrometry. In 5 cases, MALDI-TOF identified 4 of them as Streptococcus pseudopneumoniae and 1 as S. mitis/oralis. Forty-eight of the 64 strains were identified as H. influenzae (75%) using biochemical identification systems and automated identification systems, whereas MALDI-TOF-MS Vitek identified 51 strains (79%) as such. Conventional methods and spectrometry identified all the 40 strains tested (100%) as M. catarrhalis. All strains with discrepant results were sequenced, and in all cases, the identification obtained by spectrometry was confirmed. The results obtained in this study show that mass spectrometry provides identification of these bacteria faster and in a more reliable way than those based on conventional phenotypical methods (AU)


La espectrometría de masas se ha convertido en un recurso de referencia para la identificación de microorganismos en los servicios de Microbiología Clínica. Se estudiaron 151 aislamientos clínicos procedentes de muestras respiratorias que se identificaron rutinariamente como Streptococcus pneumoniae (43), Haemophilus influenzae (64) y Moraxella catarrhalis (44). Estos resultados se compararon con otros métodos fenotípicos y espectrometría de masas (MALDI-TOF Vitek-MS). Las discrepancias en los resultados se valoraron mediante secuenciación del ARNr 16S. Treinta y ocho de las 43 cepas de S. pneumoniae (86%) fueron identificadas como tales tanto por métodos bioquímicos como por espectrometría. En 5 casos MALDI-TOF identificó 4 como Streptococcus pseudopneumoniae y 1 como S. mitis/oralis. Cuarenta y ocho de las 64 cepas fueron identificadas como H. influenzae (75%) al utilizar galerías comerciales y sistemas automáticos, mientras que MALDI-TOF identifica como tales a 51 cepas (79%). Los métodos convencionales y la espectrometría identificaron como M. catarrhalis las 40 cepas estudiadas (100%). Todas las cepas con resultados discrepantes fueron secuenciadas, confirmándose en todos los casos la identificación obtenida por espectrometría. Los resultados obtenidos en este estudio demuestran que la espectrometría de masas ofrece una identificación de estas bacterias más rápida y fiable que la basada en métodos convencionales (AU)

Introducción: El progresivo envejecimiento de la población y la elevada prevalencia de la hiperplasia benigna prostática (HBP), unidos a la existencia de tratamientos médicos efectivos para esta afección, produce que cada vez más indiquemos la cirugía en pacientes tratados a largo plazo con inhibidores de la 5-alfa-reductasa (5ARI). En un principio, los pacientes tratados con 5ARI no se consideraban buenos candidatos para la fotovaporización prostática con láser verde. Objetivo: En el siguiente trabajo, mediante un estudio retrospectivo basado en nuestra experiencia, pretendemos dilucidar si el tratamiento a largo plazo (mas de 6 meses) con inhibidores de la 5ARI reduce la eficacia del tratamiento con láser verde en pacientes afectados de HBP. Material y métodos: Entre septiembre de 2005 y enero de 2008 hemos tratado a 102 pacientes mediante fotovaporización prostática con láser verde, de los que 25 (24,5%) se encontraban en tratamiento con 5ARI durante al menos 6 meses; los otros 77 (75,5%) pacientes formaron el grupo control. Realizamos un estudio retrospectivo en el que comparamos los parámetros clínicos y funcionales pre y postoperatorios de los pacientes tratados con 5ARI respecto a los que no lo estaban. Resultados: No encontramos diferencias estadísticamente significativas en cuanto a volumen prostático (50 cc y 49 cc), IPSS (17,6 y 17,8), residuo posmiccional (16% y 18%) o antígeno prostático específico (PSA) (1,4 ng/ml y 2,2 ng/ml) preoperatorios entre los grupos tratamiento y control, respectivamente. Del mismo modo, si bien existieron diferenciasen cuanto a la energía usada (180 kJ y 175 kJ) y el tiempo quirúrgico empleado (63 min y 57 min), ambas fueron más elevadas en el grupo tratado con 5ARI, estas diferencias no resultaron significativas estadísticamente. Tampoco encontramos diferencias en los parámetros clínicos (IPSS 13,8 y 14 al primer mes) ni flujométricos (Qmáx 13,9 ml/s y 14,5ml/s en la revisión al primer mes) entre pacientes tratados y no tratados. Subjetivamente, se apreció una mejor visualización del campo endoscópico atribuida a un menor sangrado durante la intervención. Conclusiones: Nuestros resultados no demuestran diferencias estadísticamente significativas entre pacientes con y sin tratamiento con inhibidores de la 5ARI en cuanto a parámetros intra y preoperatorios, por lo que pensamos que este tratamiento no disminuye la eficacia de la fotovaporización prostática con láser verde (AU)


Introduction: High prevalence of BPH and gradual ageing of the population, combined with the existence of effective medical treatments for this condition, has led to an increasing use of surgery in patients on long-term treatment with 5-alpha reductase inhibitors (5-ARIs). Initially, patients treated with 5-ARIs were not considered good candidates for photoselective vaporization of the prostate with green laser. Objective: To assess, using a retrospective study based on our experience, wheter long-term treatment (longer than 6 months) with 5-alpha reductase inhibitors decreases effectiveness of photoselective vaporization of the prostate with green light laser in BPH. Materials and methods: From September 2005 to January 2008, 102 patients underwent photoselective vaporization of the prostate with green light laser. Of these, 25 patients (24.5%) had been treated with 5-ARIs for at least 6 months, and the remaining 77 patients (75.5%) were used as controls. A retrospective study was conducted to compare the pre and postoperative clinical and functional parameters of patients with and without prior 5-RAI therapy. Results: No statistically significant differences were found between the treated and control groups in preoperative prostatic volume (50 mL vs 49 mL), IPSS (17.6 vs 17.8), postvoiding residue (16% vs 18%), or PSA (1.4 ng/mL vs 2.2 ng/mL). Similarly, while differences were seen in energy spread (180 kJ vs 175 kJ for the treated and control groups respectively) and operating time (63 min vs 57 min), these were not statistically significant. No between group differences were found either in clinical or flow rate parameters one month after surgery (IPSS 13.8 vs 14 and Qmax 13.9mL/s vs 14.5 nL/s in the treated and control groups respectively). Surgeons reported a better visualization of the endoscopic field that was attributed to less bleeding during the procedure. Conclusions: Our results show no statistically significant differences in peroperative and preoperative parameters between patients with and without treatment with 5-alphareductase inhibitors. We therefore think that photoselective vaporization of the prostate with green light laser is a safe and effective technique in patients treated with 5-ARIs (AU)

The nrdDG promoter regulates transcriptional expression of the anaerobic ribonucleotide reductase of Escherichia coli, an essential enzyme required to supply the building blocks for DNA synthesis. In this work, binding of the pleiotropic FNR (fumarate and nitrate reduction) transcriptional regulator to the nrdDG promoter region and the effects of binding on transcription were investigated. Gel retardation analysis with purified FNR demonstrated FNR interaction at two FNR sites, termed FNR-2 and FNR-1, while studies with altered FNR boxes indicated that the upstream FNR-2 site was essential for anaerobic activation of the nrdDG promoter. Although the FNR-1 site was not absolutely required, it allowed maximal expression of this promoter. These results suggest that the two sites have an additive effect in coordinating nrdDG expression in response to shifting oxygen concentrations (AU)

The taxonomy of the penicillia is unstable particularly in the important antibiotic and mycotoxin-producing subgenus Penicillium. There are difficulties relating identifications to mycotoxin production. Also, the validity of dual nomenclature for pleomorphic fungi is under discussion increasingly. Patulin is an important mycotoxin produced by various fungi and has strict limits in the European Union. The mycotoxin and/or the isoepoxydon dehydrogenase (IDH) gene of the metabolic pathway have been assessed in 318 strains predominately of subgenus Penicillium. These data were used to classify the isolates. Subgenus Penicillium contained most of the IDH and patulin positives. The species and varieties in subgenus Penicillium which were associated with patulin detection can be reduced to one name, viz. Penicillium Pen p+ (p = patulin). This has been extended to other mycotoxin producing penicillia to indicate the scope of the scheme. The classification will lead to the number of taxa being reduced, while avoiding species names and hence dual nomenclature. Culture independent analysis of environmental samples is mentioned. The scheme could be used with advantage for other fungi(AU)

Wood is the main renewable material on Earth and is largely used as building material and in paper-pulp manufacturing. This review describes the composition of lignocellulosic materials, the different processes by which fungi are able to alter wood, including decay patterns caused by white, brown, and soft-rot fungi, and fungal staining of wood. The chemical, enzymatic, and molecular aspects of the fungal attack of lignin, which represents the key step in wood decay, are also discussed. Modern analytical techniques to investigate fungal degradation and modification of the lignin polymer are reviewed, as are the different oxidative enzymes (oxidoreductases) involved in lignin degradation. These include laccases, high redox potential ligninolytic peroxidases (lignin peroxidase, manganese peroxidase, and versatile peroxidase), and oxidases. Special emphasis is given to the reactions catalyzed, their synergistic action on lignin, and the structural bases for their unique catalytic properties. Broadening our knowledge of lignocellulose biodegradation processes should contribute to better control of wood-decaying fungi, as well as to the development of new biocatalysts of industrial interest based on these organisms and their enzymes (AU)


La madera es el principal material renovable en la Tierra y es utilizada en gran parte como material de construcción y en la fabricación de celulosa. Esta revisión describe la composición de los materiales lignocelulósicos y diversos procesos de alteración de la madera por parte de hongos, como el deterioro causado por los llamados hongos de podredumbre blanca, de podredumbre parda y de podredumbre blanda y por los hongos cromógenos o manchadores de la madera. También se tratan los aspectos químicos, enzimáticos y moleculares del ataque a la lignina por los hongos, que es clave en el deterioro de la madera. Se describen las técnicas analíticas modernas para investigar la degradación y la modificación del polímero de la lignina causadas por hongos, así como las diversas enzimas oxidativas (oxidoreductasas) que intervienen en la degradación de la lignina. Entre éstas se encuentran lacasas, peroxidasas ligninolíticas de alto potencial redox (lignina-peroxidasa, manganeso-peroxidasa y peroxidasas versátiles) y las oxidasas. Se destacan las reacciones catalizadas, su acción sinérgica sobre la lignina, y las bases estructurales de sus exclusivas propiedades catalíticas. Un mejor conocimiento de los procesos de biodegradación de la lignocelulosa debería contribuir a un mejor control de los hongos descomponedores de la madera, así como al desarrollo de nuevos biocatalizadores de interés industrial basados en estos organismos y sus enzimas (AU)

This study was designed to evaluate the protective effects of zinc on the liver activities of antioxidant enzymes in protein-deficient rats. Zinc sulfate at a dose level of 227 mg/l in drinking water was administrated to Sprague Dawley normal control as well as to protein-deficient rats for a total duration of eight weeks. The effects of zinc treatment and protein deficiency alone as well as combined were studied on rat liver antioxidant enzymes which included catalase, glutathione peroxidase (GPX), glutathione reductase (GR), superoxide dismutase (SOD), and glutathione S-transferase (GST). Protein deficiency in normal rats resulted in a significant increase in hepatic lipid peroxidation and in catalase, Gpx, GR and GST activity. A significant inhibition in the levels of SOD activity and reduced glutathione (GSH) was observed following protein deficiency in normal rats. Zn treatment to protein deficient animals lowered lipid peroxidation and catalase, Gpx and GST activities, and also resulted in a significant elevation in the levels of GSH and SOD activity. The concentration of zinc decreased significantly in protein deficient animals but returned to normal levels when zinc was administered (AU)


Este estudio fue diseñado para estudiar los efectos protectores del zinc sobre la actividad de los enzimas antioxidantes del hígado en ratas con déficit proteico. Se administró sulfato de zinc en una dosis de 227 mg por litro en agua a ratas Sprage Dawley control y a un grupo de ratas con déficit proteico durante un período de ocho semanas. Los efectos del tratamiento de zinc y de la deficiencia proteica per se, así como su combinación, fueron estudiados sobre los encimas antioxidantes de¡ hígado de la rata, incluyendo catalasa, glutation peroxidasa (GPX), glutation reductasa (GR), superóxido dismutasa (SOD) y glutation s-transferasa (GST). La deficiencia protéica en ratas normales produjo un aumento significativo en la peroxidación lipídica del hígado, así como en la actividad de catalasa GPX, GR y GST. Hubo una inhibición significativa de los niveles de actividad SOD y una reducción del glutation en las ratas con déficit proteico. El tratamiento con zinc a los animales con déficit proteico disminuyó la peroxidación lipídica y la actividad de catalasa GPX y GST, y también produjo una elevación significativa en los niveles de actividad de GSH y SOD. La aciduria glutárica tipo I es una enfermedad neurometabólica, de herencia autosómico recesiva (1 caso/ 30.000), caracterizada por discinesia y distonía progresiva en niños, patológicamente por degeneración estriatal, en particular de los núcleos caudado y putamen, y bioquímicamente por deficiencia en tejidos de glutaril-CoA deshidrogenasa, con cifras usualmente elevadas de ácidos glutárico y betahidroxiglutárico en orina. Actualmente no se considera una enfermedad tratable, puesto que al diagnóstico del paciente suele haber lesiones irreversibles en sistema nervioso central, beneficiándose del tratamiento los niños presintomáticos y, en general, los hermanos de otros pacientes ya diagnosticados. Se presenta un niño de 23 meses con macrocefalia y mínimas manifestaciones neurológicas al diagnóstico, atribuidas a que el niño seguía una dieta semivegetariana. El establecimiento dietético y suplementos vitamínicos ha detenido e incluso mejorado la progresión sintomática de la enfermedad. Se concluye que a todo niño con macrocefalia progresiva de etiología no filiada se le debe realizar estudio de aminoácidos y ácidos orgánicos en orina para descartar aciduria glutárica tipo I (AU)

The synthesis of carotenoids begins with the formation of a phytoene from geranylgeranyl pyrophosphate, a well conserved step in all carotenogenic organisms and catalyzed by a phytoene synthase, an enzyme encoded by the crtB ( spy) genes. The next step is the dehydrogenation of the phytoene, which is carried out by phytoene dehydrogenase. In organisms with oxygenic photosynthesis, this enzyme, which accomplishes two dehydrogenations, is encoded by the crtP genes. In organisms that lack oxygenic photosynthesis, dehydrogenation is carried out by an enzyme completely unrelated to the former one, which carries out four dehydrogenations and is encoded by the crtI genes. In organisms with oxygenic photosynthesis, dehydrogenation of the phytoene is accomplished by a zeta-carotene dehydrogenase encoded by the crtQ ( zds) genes. In many carotenogenic organisms, the process is completed with the cyclization of lycopene. In organisms exhibiting oxygenic photosynthesis, this step is performed by a lycopene cyclase encoded by the crtL genes. In contrast, anoxygenic photosynthetic and non-photosynthetic organisms use a different lycopene cyclase, encoded by the crtY ( lyc) genes. A third and unrelated type of lycopene beta-cyclase has been described in certain bacteria and archaea. Fungi differ from the rest of non-photosynthetic organisms in that they have a bifunctional enzyme that displays both phytoene synthase and lycopene cyclase activity. Carotenoids can be modified by oxygen-containing functional groups, thus originating xanthophylls. Only two enzymes are necessary for the conversion of beta-carotene into astaxanthin, using several ketocarotenoids as intermediates, in both prokaryotes and eukaryotes. These enzymes are a beta-carotene hydroxylase ( crtZ genes) and a beta-carotene ketolase, encoded by the crtW (bacteria) or bkt (algae) genes (AU)

Microbial activity is responsible for the transformation of at least one third of the elements in the periodic table. These transformations are the result of assimilatory, dissimilatory, or detoxification processes and form the cornerstones of many biogeochemical cycles. Arsenic and selenium are two elements whose roles in microbial ecology have only recently been recognized. Known as «essential toxins», they are required in trace amounts for growth and metabolism but are toxic at elevated concentrations. Arsenic is used as an osmolite in some marine organisms while selenium is required as selenocysteine (i.e. the twenty-first amino acid) or as a ligand to metal in some enzymes (e.g. FeNiSe hydrogenase). Arsenic resistance involves a small-molecular-weight arsenate reductase (ArsC). The use of arsenic and selenium oxyanions for energy is widespread in prokaryotes with representative organisms from the Crenarchaeota, thermophilic bacteria, low and high G+C gram-positive bacteria, and Proteobacteria. Recent studies have shown that both elements are actively cycled and play a significant role in carbon mineralization in certain environments. The occurrence of multiple mechanisms involving different enzymes for arsenic and selenium transformation indicates several different evolutionary pathways (e.g. convergence and lateral gene transfer) and underscores the environmental significance and selective impact in microbial evolution of these two elements (AU)

El arsénico y el selenio son dos elementos cuyo papel en la ecología microbiana ha sido reconocido sólo recientemente. La enfermedad cardiovascular y cerebrovascular continúa siendo una de las causas principales de mortalidad en los países occidentales. La modificación de todos y cada uno de los factores de riesgo de una forma integrada resulta esencial para conseguir una reducción óptima de los episodios cardiovasculares. De hecho, el beneficio del tratamiento antihipertensivo quedaba significativamente limitado cuando no se modificaban o empeoraban los niveles de colesterol. En la población general se sabe que existe una asociación entre la presencia de niveles elevados de colesterol y de la presión arterial (PA), tanto en pacientes hipertensos como en la hipertensión (HTA) límite, y también se ha observado que la prevalencia de hipercolesterolemia por encima de 240 mg/dl es el doble en hipertensos que en normotensos y que existe además una agregación familiar entre la HTA, hiperlipidemia y la resistencia a la insulina. La importancia de estas asociaciones de factores de riesgo radica en que la elevación tanto de cifras de PA como de niveles de colesterol provoca no una suma del riesgo del accidente cardiovascular, sino que se observa un incremento casi exponencial de este riesgo. Pese a la frecuente asociación entre HTA e hiperlipidemia se conoce relativamente poco sobre posibles mecanismos patogénicos comunes o sobre el efecto que la terapia provoca en ambos y en la protección de los órganos diana. Todos los datos orientan a que debe existir algún mecanismo que directa o indirectamente sea responsable tanto de la elevación de los niveles de colesterol como de la elevación de la PA (AU)

La actividad física, principalmente la que se desarrolla de forma intensa hasta la extenuación, lleva asociada una situación de estrés oxidativo. El entrenamiento y una correcta alimentación permiten aumentar la capacidad del organismo de hacer frente a la elevada producción de especies reactivas de oxígeno y de radicales libres que tiene lugar durante el ejercicio físico. En este artículo se presentan las características principales de las especies reactivas de oxígeno, su síntesis y su potencial capacidad de generar estrés oxidativo en el deportista. Se describen los principales sistemas de defensa antioxidantes celulares, sus interacciones y como se ven afectados por el ejercicio físico intenso y por la suplementación de la dieta con antioxidantes naturales como la vitamina E, vitamina C y b-caroteno.
