RESUMO

Los niveles plasmáticos de colesterol y triglicéridos son 2 veces más altos en los osos pardos (Ursus arctos) durante el periodo de hibernación que en los humanos sanos. Sin embargo, los osos no muestran signos de desarrollo de aterosclerosis. Para explorar esta aparente paradoja, analizamos lipoproteínas del plasmas de 10 osos recolectados durante el invierno (hibernación: febrero) y verano (activo: junio) en el mismo año. El plasma de 14 humanos sanos se analizó como comparador. Se utilizaron métodos estándar para el aislamiento de lipoproteínas, la composición y la investigación funcional. Los resultados muestran que en los osos pardos la ausencia de aterosclerosis a pesar del colesterol elevado probablemente se asocie con 2 propiedades ateroprotectoras principales de las lipoproteínas circulantes. En primer lugar, una afinidad significativamente, 10 veces menor, de las partículas de lipoproteínas de baja densidad (LDL) por los proteoglicanos arteriales y, en segundo lugar, una capacidad elevada de eflujo de colesterol en plasma comparado con humanos. ¿Qué nos dicen los datos del oso pardo? Ese colesterol total elevado y las lipoproteínas que contienen ApoB no siempre se asocian con la enfermedad de aterosclerosis. Necesitamos observar también las características bioquímicas y la funcionalidad de las lipoproteínas, ya que son relevantes para la fisiopatología arterial. ¿Cuál es la traducibilidad al humano de estos resultados? Los humanos necesitamos controlar nuestros niveles de colesterol total y LDL. ¡No somos osos pardos!.(AU)


Plasma cholesterol and triglyceride levels are twice as high in hibernating brown bears (Ursus arctos) than in healthy humans. Yet, bears display no sign of atherosclerosis development. To explore this apparent paradox, we analyzed lipoproteins from same ten individual bears plasma collected during winter (hibernation; February) and summer (active; June) in the same year. Plasma from fourteen healthy humans were analyzed as comparator. We used standard methods for lipoprotein isolation, composition and functional investigation. The results shows that in brown bears the absence of atherosclerosis despite elevated cholesterol is likely associated with two main athero-protective properties of circulating lipoproteins. First, a significant ten times lower affinity of low-density-lipoprotein (LDL) particles for arterial proteoglycans and secondly, an elevated plasma cholesterol efflux capacity. What does the brown bear data tell us? That elevated total cholesterol and ApoB-containing lipoproteins not always associates with atherosclerosis disease. We need to look also at the lipoprotein biochemical features and functionality as they are relevant for arterial pathophysiology. What is the translatability into human of these results? We humans need to control our total and LDL-cholesterol levels. We are not brown bears!.(AU)

INTRODUCCIÓN: La enfermedad de Alexander es una enfermedad rara causada por mutaciones en el gen que codifica la proteína glial ácida fibrilar (GFAP). En un estudio previo hemos observado que la diferenciación de neuroesferas transfectadas con estas mutaciones genera un tipo celular que comparte la expresión de GFAP y NG2. OBJETIVOS: Determinar el efecto de las mutaciones en marcadores moleculares en comparación con células de glioma diferenciados que expresan simultáneamente GFAP y NG2. MÉTODOS: Se utilizaron muestras de glioblastoma humana (GLM) y neuroesferas procedentes de rata transfectadas con mutaciones de GFAP para el análisis de la expresión tras diferenciación de GFAP y NG2, así como el análisis inmunocitoquímico de diferenciación de ambos tipos celulares y detección de ambas proteínas, junto a nestina, vimentina, Olig2 y caspasa 3 a los 3 y 7 días de diferenciación. RESULTADOS: Tanto las células transfectadas con mutaciones de GFAP como las células procedentes de GLM mostraron un incremento de NG2 y GFAP. Sin embargo, la expresión de células caspasa 3 positiva era marcadamente mayor entre las células transfectadas que entre las células procedentes de GLM. CONCLUSIÓN: Nuestros resultados parecen indicar que la expresión de GFAP no es el único factor que condiciona la muerte celular en la enfermedad de Alexander y que la expresión de caspasa 3 y el potencial papel de la NG2 en incrementar la resistencia a la apoptosis en las células que coexpresan GFAP y NG2 deben ser considerados en la búsqueda de acciones terapéuticas en esta enfermedad


INTRODUCTION: Alexander disease is a rare disorder caused by mutations in the gene coding for glial fibrillary acidic protein (GFAP). In a previous study, differentiation of neurospheres transfected with these mutations resulted in a cell type that expresses both GFAP and NG2. OBJECTIVE: To determine the effect of molecular marker mutations in comparison to undifferentiated glioma cells simultaneously expressing GFAP and NG2. METHODS: We used samples of human glioblastoma (GBM) and rat neurospheres transfected with GFAP mutations to analyse GFAP and NG2 expression after differentiation. We also performed an immunocytochemical analysis of neuronal differentiation for both cell types and detection of GFAP, NG2, vimentin, Olig2, and aspase-3 at 3 and 7 days from differentiation. RESULTS. Both the cells transfected with GFAP mutations and GBM cells showed increased NG2 and GFAP expression. However, expression of caspase-3-positive cells was found to be considerably higher in transfected cells than in GBM cells. CONCLUSIONS: Our results suggest that GFAP expression is not the only factor associated with cell death in Alexander disease. Caspase-3 expression and the potential role of NG2 in increasing resistance to apoptosis in cells co-expressing GFAP and NG2 should be considered in the search for new therapeutic strategies for the disease

We appreciate the time and attention paid to our paper by Prof. Mestres-Ventura and similarly appreciate the opportunity to respond to his concerns. We would like to address what we believe are several fundamental misunderstandings in his commentary.1. Scale: The most significant misunderstanding is one of scale. The schematic (Fig. 1) provided by Prof. Mestres-Ventura is (per personal communication) at the nano scale, while in vivo microscopy of extrahepatic bile duct and dermis shows that the collagen bundles we report are at the micron scale, each containing many individual fibrils at the nanometer scale. Indeed, examining the tissues described in our paper - submucosae, dermis and subcutaneous fascia - fresh in resected specimens or intraoperatively, we find that the structures we describe are visible at the macroscopic level (if one leans in closely enough). In other words, they are macroscopic, not microscopic. Prof. Mestres-Ventura, in summarizing the prior pCLE work of Wallace and Fockens, which he notes is similar to ours, states "the 'holes' shown under intravital microscopy and in cryofixed samples are huge (over 20 µm)" This is exactly our point - we were surprised as well at the scale of these structures, as this has not been well appreciated in the past

Introduction: Endocan levels were found to be associated with severity and mortality of the respiratory system diseases. Objective: We aimed to figure out whether endocan was an important marker for the diagnosis, severity and follow-up of bronchopulmonary dysplasia (BPD). Materials and methods: Infants with moderate/severe BPD, and who required hydrocortisone treatment were included in the study group. Infants without BPD were allocated in the control group. Endocan levels were compared between the control group and the study group, and before and after the treatment in the study group. Results: A total of 148 infants, 74 infants in the control group and 74 infants in the BPD group, were included. The endocan level was higher in the BPD group than in the control group (P = .001). Endocan levels before treatment in the BPD group was found to be higher than endocan level after treatment (P = .021). Conclusion: Our study found that endocan levels increased in moderate/severe BPD. Serum endocan levels may be a safe and novel indicator for the follow-up of response to treatment and the prognosis of the severity of the disease


Introducción: Los niveles de endocan se han asociado con la mortalidad y la gravedad de enfermedades del aparato respiratorio. Objetivo: El objetivo fue averiguar si el endocan es un marcador útil para el diagnóstico, la gravedad y el seguimiento de la displasia broncopulmonar (DBP). Materiales y métodos: Se incluyeron en el estudio lactantes con DBP moderada/grave que requirieron tratamiento con hidrocortisona (grupo DBP). El grupo control lo constituyeron lactantes sin DBP. Los niveles de endocan se compararon entre el grupo control y el grupo de estudio y, en este último, tanto antes como después del tratamiento. Resultados: Se incluyeron un total de 148 lactantes; 74 en el grupo control y 74 en el grupo DBP. Los niveles de endocan fueron más elevados en el grupo DBP que en el grupo control (p = 0,001). Los niveles de endocan también resultaron superiores en el grupo DBP antes del tratamiento que después del mismo (p = 0,021). Conclusión: Nuestro estudio halló que los niveles de endocan se encuentran incrementados en la DBP moderada/grave. Los niveles séricos de endocan podrían utilizarse como un nuevo indicador seguro para el seguimiento de la respuesta al tratamiento y el pronóstico de gravedad de la enfermedad

Objetivos. Dada la importancia en la actualidad que tiene el envejecimiento cutáneo, en este trabajo se realiza una revisión del mismo, resaltando las diferentes teorías que lo intentan explicar así como las causas que pueden inducir este envejecimiento; se revisaran los distintos compuestos utilizados en su tratamiento, tanto desde el punto de vista de la cosmética preventiva como productos utilizados como reparadores. Métodos. Se utilizó la base de datos de la National Library of Medicine, Washington, DC (MEDLINE: PubMed) así como informaciones disponibles online. Resultados. Se analizan los distintos compuestos que desde un punto de vista preventivo y reparador son utilizados, se observa como la utilización de muchos de ellos puede ejercer una acción doble, y como pueden ser utilizados tanto con una finalidad hidratante como antioxidante por ejemplo, es decir las acciones ejercidas pueden ser muy variadas y con distinta intensidad. Bajo este prisma se estudian los proteoglicanos, ácido hialurónico, péptidos, y dos hormonas de empleo mas reciente como son melatonina y DHEA. Entre los antioxidantes analizados, la vitamina E y los polifenoles parecen ser los que más evidencias a favor de su efecto beneficioso presentan. Conclusiones. El envejecimiento cutáneo debe tratarse asociando varios activos para conseguir una mayor efectividad. Por otro lado, el futuro de este tratamiento puede estar en el estudio de los genes, que nos indicarán el preparado más adecuado a cada tipo de piel. Son necesarias más investigaciones para poder avalar el concepto de genocosmética


Objectives. Currently the skin aging is very important in the cosmetic field. For this reason, in this work a review about the skin aging is made. Therefore, our work focuses on the different theories that explain the skin aging as well as the causes that can induce this aging. In addition the compounds used in its treatment are studies, both from the point of view of preventive cosmetics as products used as repairmen. Methods. To carry out this work were used different database as the National Library of Medicine, Washington, DC (MEDLINE PubMed) and a lot of interesting information available online. Results. Different compounds are analyzed from a preventive and reparative used. It is observed how the use of many of them can have a dual action, and these compounds can be used with a hydrating and antioxidant purpose moreover the effects taken may be varied and with different intensity. Therefore proteoglycans, hyaluronic acid, peptides, melatonin and DHEA, two hormones recently used, are studied. Among the analyzed antioxidants, vitamin E and polyphenols seem to be more evidence that supporting their beneficial effect Conclusions. The combination of several cosmetic compounds is a factor to be taken into account. In this sense, the future of antiaging treatment may depend of a previous genetic study and the development of appropriate formulation for each type of skin. However more studies are needed to substantiate the concept of genocosmetic

Objetivo: Obtención mediante electrohilado de fibras micro- y submicrométricas de poliésteres funcionalizadas con glicósidos que constituyen elementos estructurales de proteoglicanos, para su uso en la reparación del tejido medular. Material y métodos: Las fibras se prepararon a partir de disoluciones de poli(3-hidroxibutirato-co-3-hidroxihexanoato) con glicósidos sintéticos mediante electrohilado variando sistemáticamente las condiciones del proceso. La morfología de las fibras fue analizada mediante microscopía electrónica de barrido. Asimismo, se evaluó la estabilidad de la interacción entre el glicósido y la fibra en medio acuoso, y su toxicidad en cultivos de células neurales. Resultados: La morfología de las fibras obtenidas depende principalmente de los parámetros de la disolución. En medio acuoso, el glicósido sulfatado se liberó de las fibras más lentamente que el que no tenía dicho grupo funcional. La viabilidad de las células neurales no se vio afectada por los glicósidos. Conclusión: La preparación de microfibras alineadas de poliéster funcionalizadas con glicósidos es posible. La mayor parte del glicósido permanece retenido en las fibras sumergidas en agua después de varios días. El electrohilado es una técnica muy accesible y versátil para la fabricación de soportes en estrategias de terapia celular de lesiones medulares (AU)


Objective: Preparation of functionalized micro- and submicrofibers by electrospinning of polyesters with glycosides which are structural elements of proteoglycans, for application to the repair of spinal cord lesions. Material and methods: Solutions of poly(3-hydroxybutyrate-co-3-hydroxyhexanoate) with synthetic glycosides were prepared varying systematically the processing conditions. Fiber morphology assessed by scanning electron microscopy. The stability of the interaction between the glycoside and the polymer fiber was evaluated in aqueous medium, and their toxicity in cultures of neural cells. Results: The fiber morphology was altered mainly by the solution parameters. In aqueous medium, the glycoside with a sulfate group was released from fibers at slower rate than the non-sulfated glycoside. The viability of neural cells was not affected by the glycosides. Conclusion: It is possible to fabricate aligned polyester micro fibers with glycosides. Most of the glycoside present in the fibers remains in the substrate after extraction in water for several days. Electrospinning is a very accessible and versatile technique for application to strategies of cellular therapy in spinal cord injuries (AU)

Objectives: To study the preventive effects of chlorhexidine against root caries under oral biofilm in an artificial mouth. Study Design: Sixteen human tooth-root disks were inoculated with a salivary sample that was produced by mixing the unstimulated saliva of three adults who had no untreated caries. The disks were incubated in an artificial mouth fed with a 5% sucrose solution three times daily for one week. Eight disks received a twice daily rinse of 0.12% chlorhexidine (test group). The other eight disks were rinsed in distilled water (control). The biofilm was then studied with three techniques: colony forming unit (CFU) counting, scanning electron microscopy (SEM) and confocal laser scanning microscopy (CLSM). The changes in the chemical structure of the root surface were studied using Fourier transform infra-Red spectroscopy. Type-I collagen and proteoglycans on the root surface were quantified using immunocytochemical staining. Results: The log CFU for the test and control groups were 4.21 and 8.27, respectively (p<0.001). The CFU count of Streptococci and Lactobacilli were negligible. Both the SEM and the CLSM showed suppressed bacteria growth in the test group. The log [amide-I: HPO42-] of the test and control groups were 1.11 and 1.93, respectively (p=0.02). The mean counts of sound type-I collagen in the test and control groups were 16.8/μm2 and 13.0/μm2, respectively (p<0.001), whereas the mean counts of intact proteoglycans were 5.6/μm2 and 3.5/μm2, respectively (P<0.001). Conclusions: Chlorhexidine suppressed the growth of selected cariogenic bacteria in oral biofilm on the root surface and thus protected tooth-root from cariogenic challenge (AU)

La apolipoproteína CIII (apoCIII) inhibe la hidrólisis de triglicéridos de la VLDL y quilomicrones por la lipasa lipoproteica. Se ha pensado que la hipertrigliceridemia resultante explica la asociación entre el aumento de la apoCIII con exageración del riesgo cardiovascular. Sin embargo, estudios clínicos indican que un aumento de la apoCIII asociada a las LDL es un factor de riesgo independiente de la hipertrigliceridemia. Experimentos recientes indican que la apoCIII incrementa la adhesión de monocitos al endotelio, un requisito para la acumulación de macrófagos en la aterogénesis. Además, LDL ricas en apoCIII tienen una alta afinidad por los proteoglicanos de la íntima, lo cual podría aumentar su deposición subendotelial. Estas dos propiedades y efectos de la LDL rica en apoCIII pueden ser la base de los hallazgos indicativos de que la apoCIII es un agente causal de la aterogénesis y un factor de riesgo cardiovascular independiente (AU)


Summary The apolipoprotein CIII (apoCIII) is an inhibitor of the hydrolysis of triglycerides inchylomicrons and VLDL by lipoprotein lipase. It has been accepted that the hypertriglyceridemia induced by high apoCIII can explain its association with increased cardiovascular risk.However, several studies indicate that increased apoCIII is a risk factor independent of hypertriglyceridemia. Recent experimental evidence in vitro and in vivo indicates that apoCIII byitself, or associated with lipoproteins, increase expression of cell adhesion molecules in theendothelium and monocytes. Thus causing associations between these cells. In addition, it hasbeen shown that LDL rich in apoCIII have a high affinity for proteoglycans of the arterial intima.A property that could increase the rate of LDL entrapment in the subendothelial intima. Thesetwo properties of LDL rich in apoCIII indicate that apoCIII is a significant risk factor because itcan have a causal role in atherogenesis (AU)

Objetivo: Evaluar el efecto de una preparación con ribosomas bacterianos y proteoglicanos de Klebsiella pneumoniae «R» sobre la expresión in vitro de las moléculas CD11c y CD103 en neutrófilos de sangre periférica. Métodos: Aislamiento de neutrófilos de sangre periférica con Ficoll-Paque, incubación con R y detección de CD11c y CD103 mediante citometría de flujo. Resultados: A las 6h postincubación, la expresión de CD11c aumentó de manera significativa respecto al control con 125 y 500 microgramos/ml de R (p=0,017 y p=0,006, respectivamente). La expresión de CD103 inducida con 125 microgramos/ml de R a las 6h fue significativamente mayor que la observada a las 4h a la misma concentración (p=0,014) y que la encontrada con 62,5 microgramos/ml (p=0,017). Conclusiones: El aumento en la expresión de CD11c y CD103 inducido por R en neutrófilos podría contribuir al mecanismo de acción de R contra patógenos respiratorios(AU)


Aim: To evaluate the effect of a preparation with bacterial ribosomes and proteoglycans from Klebsiella pneumoniae «R» on the in vitro expression of CD11c and CD103 molecules in neutrophils from peripheral blood. Methods: Isolation of neutrophils from peripheral blood with Ficoll-Paque, incubation with R and detection of CD11c and CD103 through flow cytometry. Results: Six hours after the incubation period, CD11c expression increased significantly compared with the control with 125 and 500 microgramos/ml of R (P=0,017 and P=0,006, respectively). CD103 expression induced with 125 microgramos/ml of R after 6hours was significantly higher than that observed after 4hours at the same concentration (P=0,014) and that found with 62.5 microgramos/ml (P=.017) of R. Conclusions: The increased expression of CD11c and CD103 induced by R in the neutrophils could contribute to the R mechanism against respiratory pathogens(AU)

RESUMO

Considerando todas las investigaciones realizadas hasta la fecha sobre las causas que contribuyen al deterioro de la membrana peritoneal y su fisiopatología, se puede concluir que es de gran interés investigar si la administración de heparinaintraperitoneal puede aportar un beneficio sobre el mantenimiento de la función peritoneal en los enfermos tratados con diálisis peritoneal (DP). Las acciones descritas a favor de esta idea son:1) La inflamación crónica del peritoneo es una causa de alteración de la función peritoneal y la heparina tiene acciónantiinflamatoria.2) La fibrosis peritoneal debida a diálisis peritoneal o a traumatismo puede ser evitada o mejorada con heparina ip.3) La heparina (HNF y HBPM, incluida bemiparina) indúcela síntesis de tPA por las células mesoteliales, lo que supone una acción fibrinolítica.4) La heparina, más la HBPM que la HNF, inhibe la angiogénesis.5) La heparina intraperitone al favorece la eliminación delos AGE en la DP.6) Modelos animales y estudios clínicos de corta extensión han demostrado una mejora de la función peritoneal con heparina.7) Por el momento, no se han encontrado problemas de seguridad en su administración intraperitoneal. Es por tanto una hipótesis verosímil que el uso de heparinaintraperitoneal puede modificar favorablemente la función peritoneal de pacientes en diálisis peritoneal (AU)


Multiple investigations performed on peritoneal pathophysiology during peritoneal dialysis (PD) suggest that intraperitonealheparin might modify most of the causes of membrane deterioration. The actions described favouring this idea are:1) Peritoneal Chronic inflammation alters peritoneal function and heprain has anti-inflammatory properties.2) Peritoneal fibrosis related to peritoneal dialysis or traumaticinjury may be avoided or limited with heparin.3) Heparine induces tPA síntesis by mesothelial cells, which represents a potentiation of fibrinolytic action.4) Heparine, sècifically low-molecular weight heparin, inhibitsangiogenesis.5) Intraperitoneal heparin favors the removal of advanced glycosilation end products in PD.6) Animal models and clinical studies with small series of patients have demonstrated an improvement of peritoneal function with intraperitoneal heparine use.7) Until now, no adverse effects of the intraperitoneal heparinuse have been found. In consequence, it is a plausible hipothesis to consider that intraperitonealheparin may favourably modify peritoneal function inpatients under peritoneal dialysis (AU)

Los ácidos siálicos están entre las moléculas de mayor importancia del reinoanimal encontrándose también en algunos microorganismos. Son cetoácidos con unesqueleto glucídico de nueve carbonos, están cargados negativamente y se descubrieronen mamíferos aunque se encuentran en la mayor parte de los celomados, enprotostomados (por ejemplo, artrópodos), y deutorostomados (por ejemplo, cordadosy equinodermos). La ruta biosintética del Neu5Ac tiene lugar a través de las siguientesreacciones: a) síntesis de ManAc-6-P, b) síntesis de ManNAc, c) síntesis deNeu5Ac, d) activación del monómero to CMP-Beta-Neu5Ac, y e) transferencia delNeu5Ac a un aceptor. En el catabolismo de estos compuestos las enzimas neuraminidasay N-acetil neuraminato liasa tienen un papel importante. Sialic acids are among the most important molecules in the animal kingdomand also occur in some microorganisms. They are alpha-ketoacids with a nine-carbonglycid backbone. They are negatively charged and they were first discovered inmammals, although it appears that they are present in most Coelomata in bothprotostomes (e.g. Arthropoda) and deutorostomes (e.g. Chordata or Echinodermata).The biosynthetic pathway of Neu5Ac proceeds through the following reactions:a) synthesis of ManAc-6-P, b) synthesis of ManNAc, c) synthesis of Neu5Ac, d)activation of the monomer to CMP-Beta-Neu5Ac, and e) transfer of Neu5Ac to theacceptor structure. En la reacción de hipersensibilidad inmediata, el alérgenose une a su anticuerpo específico tipo IgE, unido a su vez a los receptores de alta afinidad para la IgE (FccRI) de las células efectoras, fundamentalmente mastocitos y basófilos. El entrecruzamiento de estas moléculas de F ccRI tras la unión de antígenos polivalentes a la IgE, activa a estas células produciéndóse tres respuestas biológicas: exocitosis del contenido preformado de sus gránulos, sintetización de mediadores lipídicos y secreción de citoquinas. Los mediadores de la inflamación son en último termino, las sustancias responsables de la sintomatología clínica. Pueden dividirse en general en mediadores preformados (aminas biógenas y macromoléculas de los gránulos) y mediadores de nueva síntesis (mediadores lípidicos y citoquinas)

El ácido hialurónico (AH) es un glucosaminoglucano compuesto de disacáridos de ácido glucurónico y Nacetilglucosamina, presente a altas concentraciones en el tejido conectivo de los mamíferos. En la artrosis (AO), la longitud y la concentración del AH se hallan disminuidas. A principios de los años setenta empezaron a utilizarse las inyecciones intraarticulares de AH con el fin de restaurar las propiedades reológicas del líquido sinovial. Sin embargo, a pesar de su amplia utilización en clínica, numerosas cuestiones sobre su mecanismo de acción y sus propiedades como fármaco continúan sin esclarecerse. La revisión de la literatura médica realizada en este artículo pone de manifiesto las propiedades del AH como modificador lento de los síntomas en la atrosis (SYSADOA) y, a su vez, evidencia la no existencia de suficientes estudios que permitan calificarlo como fármaco modificador de la estructura (DMOAD). El AH posee una amplia variedad de mecanismos de acción que van desde su interacción con los mecanismos nociceptivos del dolor hasta su capacidad para modular la homeostasis de la matriz extracelular. A pesar de que el AH de bajo peso molecular presenta un mejor perfil de seguridad y se muestra ligeramente superior en los estudios in vitro y de experimentación animal, los ensayos clínicos que valoran la eficacia del fármaco no aportan la suficiente evidencia para aconsejar el uso del AH de un determinado peso molecular en detrimento del resto(AU)


Hyaluronic acid is a glycosaminoglycan composed of disaccharides of glucuronic acid and N-acetylglucosamine. It is present at high concentrations in mammalian connective tissues. Since the length and concentration of HA decrease in osteoarthritis (AO), intraarticular HA injections began to be used in the early 1970s to restore the rheological properties of synovial fluid. Despite the widespread use of HA, many questions about its mechanism of action and properties remain to be clarified. A literature review presented in this article reveals that HA is a symptomatic slow-acting drug in osteoarthritis (SYSADOA) and that there is insufficient evidence to qualify it as a disease modifying osteoarthritis drug (DMOAD). HA is involved in many mechanisms of action ranging from interaction with mechanosensitive articular pain receptors to its ability to modulate extracellular matrix homeostasis. Although low molecular weight HA has a better safety profile and is slightly superior in in vitro studies and animal experimentation, clinical trials evaluating the efficacy of the drug show insufficient evidence to enable one type of HA to be recommended in preference to any other(AU)

Introducción. El crecimiento óseo precisa de una intensa actividad anabólica que se centra, sobre todo, en la síntesis proteica. Cualquier alteración que afecte a la multiplicación celular y su diferenciación, la síntesis del colágeno o la formación de proteoglicanos puede producir un cambio patológico. Revisión de la bibliografía. Los autores han llevado a cabo una profunda revisión bibliográfica referente al cartílago de crecimiento. Conclusiones. Las hormonas actúan según diferentes patrones sobre el desarrollo esquelético, cambiando el grosor de las fisis y el índice y magnitud de su crecimiento. Hay factores locales en y alrededor de las epífisis unidos a los factores sistémicos (hormona de crecimiento, hormona tiroidea, estrógenos y andrógenos, glucocorticoides y vitamina D) que influyen sobre la función fisaria, sin olvidar que la modificación de los factores mecánicos puede producir importantes alteraciones en la magnitud del crecimiento y en su orientación


Introduction. Bone growth requires intense anabolic activity centering mainly on protein synthesis. Any disturbance affecting cell multiplication and differentiation, collagen synthesis, or proteoglycan formation can produce a pathologic disorder. Literature review. The literature on the growth cartilage has been reviewed in depth. Conclusions. Hormones act in different ways on skeletal development, changing the thickness of the growth cartilage and the index and magnitude of growth. Local factors act in and around the growth cartilage, together with systemic factors (growth hormone, thyroid hormone, estrogens and androgens, glucocorticoids and vitamin D) that influence growth cartilage function. In addition, modifications in mechanical factors can produce important disturbances in the magnitude and direction of growth

Objetivo. En este artículo realizamos un estudio de las características de la matriz extracelular (MEC) del sistema nervioso central (SNC). Desarrollo. Los proteoglicanos (PG) son glicoproteínas, un tipo de molécula muy abundante en la MEC. Dentro de este grupo de PG destacan los hialectanos o lecticanos. Los hialectanos se caracterizan por poseer dos componentes: la proteína eje y la fracción glucídica, que está formada por los glucosaminoglicanos (GAG). Los GAG están formados por la polimerización de dímeros de azúcares sencillos. Los GAG de los hialectanos son del tipo condroitinsulfato (CS). Por esta razón, estos PG se denominan PG del tipo CS (PG-CS). Los PG-CS se unen al ácido hialurónico y a otras moléculas de la MEC para formar una red tridimensional de naturaleza proteica, que cumple funciones muy importantes en la homeostasis del tejido nervioso. Conclusiones. Diversas hipótesis se han planteado para tratar de explicar la falta de regeneración del SNC tras la lesión neural o en ciertas patologías. Se ha propuesto que los PG-CS, cuya expresión se incrementa tras las lesiones del SNC, pudieran desempeñar algún papel en este proceso de inhibición de la capacidad regenerativa neural. En este sentido, realizamos finalmente un análisis de las aproximaciones terapéuticas, experimentales, que se realizan para tratar de inducir la regeneración del SNC tras la actuación sobre los PG-CS de la MEC (AU)


Aim. In this paper we present a review of the main features of the extracellular matrix (ECM) of the central nervous system (CNS). Development. Proteoglycans (PG) are glycoproteins, a very common type of protein in the ECM. Among the PG, the most abundant type is the hyalectan or lectican family. The PG is formed by two main components; a protein and a sugar chain, which that is termed glycosaminoglycan (GAG). These GAGs are polymers of two simple sugars. The hyalectan family has GAGs of the Chondroitin sulphate type (CS), and for this reason they are termed PG-CS. PG-CS are linked to the hyaluronic acid (HA) and other molecules of the ECM in order to form a three-dimensional network. This network has several important roles in the maintenance of the homeostasis of the CNS. Conclusions. Several hypotheses have been proposed to elucidate the failure of the CNS regeneration after an injury or in several pathologies. It has been suggested that PG-CS, which expression is up-regulated after CNS injury, may play some role in this process of inhibition of the CNS regeneration. Furthermore, we present an approach to the therapeutic potential of the CNS regeneration after the inactivation of the PG-CS up-regulation (AU)

El cartílago articular está formado por células (condrocitos) y una compleja matriz extracelular, sintetizada por ellas, que sufre una remodelación continua regulada por factores anabólicos (factores de crecimiento) y catabólicos (interleucinas). Este artículo es una actualización sobre la composición, metabolismo, bioquímica y biología del cartílago articular. Se realiza una descripción detallada de la composición de la matriz extracelular, con especial énfasis en los colágenos, proteoglicanos, glicoproteínas y proteasas. Además, se exponen detalladamente las principales familias de factores de crecimiento e interleucinas que regulan la biología del cartílago articular en condiciones normales. (AU)

RESUMO

The expansive force generated by the positive pressure of the neural tube fluid confined inside brain vesicles has been shown to be a key factor during the earliest stages of brain morphogenesis and development of chick and rat embryos. In previous studies, we demonstrated the existence in these species of an intracavity extracellular matrix rich in proteoglycans that could be involved in the regulation of the expansive process. Our results show that the enzymatic digestion of the intracavity proteoglycans by testicular hyaluronidase selectively disrupts the expansive process of brain vesicles, significantly reduces the rate of mitosis in the brain neuroepithelium, and increases the number of the apoptotic cells, leading to a decrease in neuroepithelial volume. These results support the hypothesis that intracerebral proteoglycans play a relevant role in the regulation of the expansive process of the brain primordium in rat embryos that they could be involved in regulating the survival and replication of neuroblasts (AU)


Se ha demostrado que la fuerza expansiva generada por la presión positiva del fluido del tubo neural confinado en el interior de las vesículas cerebrales es un factor clave durante las primeras etapas de la morfogénesis y desarrollo del cerebro de embriones de pollo y de rata. En estudios anteriores demostramos en estas especies la existencia de una matriz extracellular intracavitaria rica en proteoglicanos que podía estar implicada en la regulación del proceso expansivo. Los presentes resultados demuestran que la digestión enzimática de los proteoglicanos intracavitarios por la hialuronidasa testicular interrumpe selectivamente el proceso expansivo de las vesículas cerebrales, reduce significativamente la proporción de mitosis en el neuroepitelio cerebral y aumenta el número de células apoptóticas, conduciendo a una disminución del volumen neuroepitelial. Estos resultados mantienen la hipótesis de que los proteoglicanos intracerebrales juegan un papel relevante en la regulación del proceso expansivo del primordio cerebral en embriones de rata y que podrían estar implicados en la regulación de la supervivencia y replicación de los neuroblastos (AU)
