RESUMO

Background: Mast cell (MC) degranulation via activation of the Mas-related G protein–coupled receptor X2 (MRGPRX2) plays a key role in immediate drug hypersensitivity (IDH). However, data in humans are limited to observations in specific cell lines. Objective: To study the usefulness of silencing MRGPRX2 in human MCs with the aim of further unveiling the MRGPRX2 pathway in IDH. Methods: MCs were cultured from CD34+ progenitor cells obtained from peripheral blood (PBCMCs) and incubated with substance P (as a positive control), rocuronium, moxifloxacin, morphine, or amoxicillin. Immunophenotyping of the cells included flow cytometry and microscopy analyses of the expression of CD117, CD203c, and MRGPRX2. Intracellular calcium was measured using Fluo-4. Degranulation was analyzed by quantifying CD63 expression. For MRGPRX2 silencing, MCs were electroporated with Dicer small interference RNAs. Results: Incubation of MCs with substance P, morphine, and moxifloxacin increased intracellular calcium levels and triggered MC degranulation, which, for the drugs, is almost completely abolished by selective MRGPRX2 silencing. Despite an increase in intracellular calcium in MRGPRX2+ cells, incubation with nontoxic concentrations of rocuronium does not result in degranulation of PBCMCs. Amoxicillin has no effect on PBCMCs. Conclusion: The use of MRGPRX2 silencing in human MCs can provide important insights into the role of MRGPRX2 in the pathogenesis of IDH. As induction of calcium signals does not necessarily translate into a secretory response, measurement of the degranulation reaction seems more meaningful in the context of drug testing (AU)


Antecedentes: La desgranulación de los mastocitos (MC) a través de la activación del receptor X2 acoplado a proteína G relacionada con Mas (MRGPRX2) se considera clave para la hipersensibilidad inmediata a fármacos. Sin embargo, los datos en humanos se limitan a observaciones en líneas celulares específicas. Objetivo: Estudiar la utilidad del silenciamiento de MRGPRX2 en MC humanos para conocer mejor la vía MRGPRX2 en la hipersensibilidad inmediata a fármacos. Métodos Los MC se cultivaron a partir de células progenitoras CD34+ obtenidas de sangre periférica (PBCMC) y se incubaron con la sustancia P como control positivo, rocuronio, moxifloxacina, morfina o amoxicilina. El inmunofenotipaje de las células incluyó análisis por citometría de flujo y microscopia de la expresión de CD117, CD203c y MRGPRX2. El calcio intracelular se midió usando Fluo-4. La desgranulación se analizó por cuantificación de la expresión de CD63. Para el silenciamiento de MRGPRX2, los MC se electroporaron con ARN silente del sustrato Dicer. Resultados: La incubación de MC con sustancia P, morfina y moxifloxacina provocó el aumento de los niveles de calcio intracelular y desencadenó la desgranulación de MC. En el caso de la desgranulación provocada por los fármacos, ésta se eliminó casi por completo mediante el silenciamiento selectivo de MRGPRX2. A pesar del aumento del calcio intracelular en las células MRGPRX2+, la incubación con concentraciones no tóxicas de rocuronio no produce la desgranulación de los PBCMC, mientras que la amoxicilina no tiene efecto sobre los PBCMC. Conclusión: El uso del silenciamiento de MRGPRX2 en MC humanos puede proporcionar información importante sobre el papel de MRGPRX2 en la patogénesis de la hipersensibilidad inmediata a fármacos. Como la inducción de señales de calcio no se traduce necesariamente en una respuesta secretora, parece más significativa la medición de la reacción de desgranulación en el contexto de las pruebas a fármacos (AU)

Tradicionalmente, la piel sensible se ha considerado como un problema cosmético o como una alteración puramente psicosomática con un fuerte componente subjetivo. Sin embargo, diversos estudios científicos sobre sus procesos fisiopatológicos y su etiopatogenia han demostrado que se trata de una entidad compleja que ya diversos autores consideran un síndrome neurodermatológico. Sus características hacen que su diagnóstico y tratamiento puedan resultar complicados en la práctica clínica habitual, siendo necesarias herramientas sencillas que se puedan usar de rutina, tanto para identificar esta entidad, que puede presentarse acompañada de otra patología, como para su manejo independiente. En este trabajo realizamos una revisión práctica de los avances científicos más recientes el campo de la piel sensible que justifican su consideración individual y ofrecen herramientas para identificarla y tratarla. Proponemos algoritmos de diagnóstico y de tratamiento basados en las evidencias de la literatura y en la opinión de los expertos que firman este artículo


Sensitive skin has traditionally been viewed as a cosmetic problem or as a purely psychosomatic alteration with a major subjective component. Different studies of its pathophysiologic etiology, however, have shown it to be a complex entity that several authors now consider to be a neurodermatological syndrome. Because of this complexity, skin sensitivity can be difficult to diagnose and treat, particularly considering that it may present with another disease. Simple tools applicable to clinical practice are thus necessary to identify and manage this disease as an independent entity. In this study, we perform a practical review of the most recent scientific advances in the area of sensitive skin that justify it being considered an individual entity, and provide tools for its identification and treatment. We propose diagnostic and treatment algorithms based on evidence from the literature and our experience and expertise

La ingesta de alcohol está facilitada por la relación con la conducta alimentaria, y ambas conductas están altamente influidas por situaciones de estrés y ansiedad. La desregulación de estos procesos puede llegar a situaciones patológicas, como la anorexia, la bulimia o la obesidad. Los elementos neurobiológicos que subyacen a este control no están completamente esclarecidos. El núcleo incertus (NI) en el tegmento pontino constituye un elemento común a la ingesta y a la adicción al alcohol. Las neuronas del NI utilizan como señalización el neuropéptido relaxina-3 (RLN3) y su receptor RXFP3. En esta revisión se analiza la participación del sistema NI-RLN3-RXFP3 en estas conductas bajo condiciones de ansiedad o estrés en modelos animales. La activación del NI tiene un efecto positivo sobre la ingesta (orexígeno) y desarrolla una respuesta amplia en la amígdala, donde se modulan los estados de ansiedad. La actividad de RLN3-RXFP3 en la amígdala podría afectar a la adicción al alcohol, ya que la aplicación del antagonista de RXFP3 en la amígdala extendida atenúa la recaída al alcohol inducida por el estrés. Los datos neuroanatómicos indican que el sistema NI-RLN3-RXFP3 actúa sobre la conducta de ingesta y adicción al alcohol mediante proyecciones paralelas a las vías canónicas mesolímbicas. Con ello, los datos en modelos animales indican que el sistema NI-RLN3-RXFP3 debería tenerse en cuenta como diana en el tratamiento futuro de trastornos de las conductas alimentarias y adictivas


Alcohol intake is facilitated by its relationship with eating behavior and both processes are highly influenced by situations of stress and anxiety. The dysregulation of these processes can reach pathological situations such as anorexia, bulimia or obesity. The neurobiological elements which underlie this control are not completely clarified. The nucleus incertus (NI) in the pontine tegmentum is a common element in the food intake and alcoholism. NI is characterized by using the neuropeptide relaxin-3 (RLN3) as transmitter and its receptor RXFP3. In the present review, we will analyze the participation of the NI-RLN3-RXFP3 system in these behaviors under conditions of anxiety or stress in animal models. The activation of NI has a positive effect on intake (orexigenic) and generates a wide response in the amygdala modulating anxiety states. The activity of RLN3-RXFP3 in the amygdala could affect alcohol addiction since the application of the RXFP3 antagonist in extended amygdala attenuates the relapse to alcohol induced by stress. The neuroanatomical data indicate that the NI-RLN3-RXFP3 system acts on the feeding behavior and alcohol intake by means of projections parallel to the canonical mesolimbic pathways. Thus, data in animal models indicate that the NI-RLN3-RXFP3 system should be taken into account as a target in the future treatment of disorders of eating and alcohol addictive behaviors

Las bacterias han colonizado al Homo sapiens como parásitos (provocando la enfermedad y la muerte) y también como mutualistas (microbioma). Como mutualistas el microbioma se utiliza como marcador de enfermedades específicas, y se propone la existencia de un eje microbiota-intestino-cerebro cuyo funcionamiento incorrecto es responsable de trastornos cerebrales: comportamiento, ansiedad, depresión, esclerosis múltiple, enfermedad de Alzheimer… Entre los posibles intermediarios que conectan el intestino con el cerebro se encuentran el sistema inmunitario, neuropéptidos y metabolitos microbianos. La conexión con el cerebro puede canalizarse por el nervio vago pero también por el sistema nervioso autónomo. Se han obtenido imágenes cerebrales en humanos que relacionan la composición de la microbiota con la emoción y actualmente necesitamos que los estudios realizados con roedores se amplíen a humanos para poder confirmar que los probioticos tienen efecto sobre la salud mental. El microbioma forma parte de un ecosistema mutualista necesario al Homo sapiens para que pueda sobrevivir como especie (AU)

As a mutualist the microbioma is used as a marker of specific diseases and it is proposed the existence of a microbiota-intestine-brain axis whose incorrect functioning is responsible for brain disorders: behavior, anxiety, depression, multiple sclerosis, Alzheimer's disease… Among the possible intermediates that connect the intestine with the brain are the immune system, neuropeptides and microbial metabolites. The connection to the brain can be channeled through the vagus nerve but also through the autonomous nervous system. Human brain images have been obtained that relate the composition of the microbiota to emotion, and we now need the studies done with rodents to be extended to humans in order to confirm that probiotics have an effect on mental health. The microbiome is part of a mutualistic ecosystem necessary to Homo sapiens so that it can survive as a species (AU)

La oxitocina (Oxt) es un neuropéptido especialmente relevante por su implicación en las conductas sociales y de afiliación, incluyendo la regulación de la conducta agresiva. En este artículo presentamos una revisión actualizada que examina la relación entre la Oxt y la agresión en modelos animales. La recopilación de artículos se realizó a través de una búsqueda combinada en las bases de datos Web of Knowledge, Scopus y Pubmed (1990-septiembre 2016). Los resultados de estos estudios incluyen evidencias que proceden de la administración de Oxt intranasal, intracerebral, intracerebroventricular, o sus antagonistas, de estudios de lesión, microdiálisis, así como de trabajos con animales modificados genéticamente. En conjunto, los resultados indican que tanto en roedores machos como en hembras la Oxt modula la agresión, aunque no siempre lo hace en el mismo sentido. Se concluye que en la agresión ofensiva la Oxt ejerce generalmente un papel inhibitorio sobre la agresión, mientras que en la agresión maternal se ha observado tanto un papel facilitador como inhibidor, dependiendo de diferentes factores. Se sugiere la necesidad de analizar las variables contextuales e individuales que podrían estar modulando la acción de la Oxt sobre la agresión (AU)


Oxytocin (Oxt) is a neuropeptide particularly relevant for its involvement in social and affiliation behaviours, including the regulation of aggressive behaviours. An updated review is presented, examining the relationships between oxytocin and aggression in animal models. The articles for review were gathered using a combined search on Web of Knowledge, Scopus, and PubMed databases (from 1990 to September 2016). The results of these studies include evidence derived from intranasal, intraventricular and intra-cerebro-ventricular Oxt administration, or from antagonist drugs, lesion studies, microdialysis, as well as works with genetically modified animals. Overall, the results indicate that Oxt modulates aggression both in male and female rodents, although it does not always act in the same direction. It is concluded that Oxt generally exerts an inhibitory role in offensive aggression, whereas in maternal aggression both a facilitating as well as an inhibitor role has been observed, depending on different factors. It is suggested that there is a need to analyse the contextual and individual variables that might be modulating the Oxt action in aggression (AU)

La obesidad es la enfermedad metabólica más prevalente y un factor de riesgo para la aparición de enfermedades cardiovasculares razón por la que debe ser tratada. El tratamiento farmacológico debe entenderse como una medida coadyuvante de los cambios en el estilo de vida que constituyen el primer escalón del tratamiento, estando indicado tan sólo en pacientes obesos y en pacientes con sobrepeso que presentan patologías asociadas (p.ej., hipertensión arterial, diabetes tipo 2 o dislipemias) en los que la dieta y el ejercicio durante los 2-3 meses anteriores han sido inefectivas. Debemos tener presente que los fármacos no curan la obesidad, su efecto a largo plazo es modesto, pueden producir reacciones adversas (a veces graves) y que la interrupción del tratamiento conduce a un rápido aumento del peso corporal En este capítulo analizaremos el tratamiento farmacológico de la obesidad, analizando los objetivos del mismo, las barreras que lo dificultan y la evidencia que avala los fármacos que actualmente han sido aceptados tanto por la Food and Drug Administration de los Estados Unidos y la European Medicines Agency (AU)


Obesity is the most prevalent metabolic disease and a risk factor for cardiovascular diseases and, therefore, it should be treated. Drug treatment should be understood as an adjunctive measure of changes in lifestyle that are the first step of treatment, being indicated only in obese patients and in overweight patients with associated cardiovascular diseases (eg, hypertension, type 2 diabetes or dyslipidemia) when diet and exercise during the previous 2-3 months have been found ineffective. We must remember that drugs do not cure obesity, its long-term effect is modest, they can produce adverse reactions (sometimes severe) and discontinuation of treatment leads to a rapid gain in body weight This chapter discusses the pharmacological treatment obesity, analyses the main objectives of drug therapy, the barriers that hinder the treatment and the evidence supporting the drugs at the present time accepted by the Food and drug Administration and the European medicines Agency (AU)

OBJETIVO: Revisar y actualizar la fisiología de la regulación de fertilidad y reproducción, poniendo especial énfasis en los factores neuroendocrinos que controlan la secreción pulsátil de GnRH. MATERIAL Y MÉTODOS: Revisión bibliográfica, utilizando como palabras clave «GnRH», «Kisspeptin», «GnIH», «RF-amide» y RPRF-3, entre otras. RESULTADOS: Siendo la secreción pulsátil de GnRH el elemento clave para el control de la secreción de gonadotrofinas, se describen los mecanismos neuroendocrinos (Kisspeptina, GnIH) que regulan la actividad de las neuronas GnRH


OBJECTIVE: To Review and update the physiology regulating fertility and reproduction, with particular emphasis on neuroendocrine factors controlling the pulsatile secretion of GnRH. Exercise is recognized as an effective method of weight management and short-term appetite regulation tool. The effect of different exercise intensities on appetite regulation hormones in healthy overweight participants has not been intensively studied. The aim of this study was to examine the influence of exercise at individual anaerobic threshold (IAT) and maximal fat oxidation (Fatmax) intensities on the nesfatin-1 response and metabolic health biomarkers in overweight men. Nine healthy overweight males (age, 23.1 ± 1.1 years) volunteered in this study in a counterbalanced order. Blood samples were obtained before, immediately after, and following the first 45 min of recovery for measuring plasma variables. There was significant decrease in plasma levels of nesfatin-1 and leptin after exercise at the IAT intensity which remained lower than baseline following 45 min of recovery. However, nesfatin-1 and leptin levels did not change significantly in any time courses of Fatmax intensity (P > 0.09). Plasma interleukin-6 (IL-6) concentration increased during exercise in both intensities (P < 0.05), whereas changes in free fatty acids (FFAs) and epinephrine concentrations were significant only at the IAT. In addition, a significant correlation was found among nesfatin-1 levels with insulin (r = 0.39, P < 0.05) and glucose (r = 0.41, P < 0.05) at basal and in response to exercise. These results indicate that IAT has a greater exercise-induced appetite regulation effect compared with Fatmax. Based on these data, the intensity of exercise may have an important role in changes of nesfatin-1, leptin, FFA, and epinephrine concentrations even though this was not the case for IL-6 and insulin resistance

Introducción. Se ha descrito recientemente una nueva clase de neuropéptidos, las orexinas, también llamadas hipocretinas, producidos por un reducido grupo de neuronas hipotalámicas y cuyas acciones son mediadas por dos tipos de receptores, OX1R y OX2R. En concreto, las neuronas orexinérgicas se han localizado en exclusiva en células de áreas del hipotálamo lateral, dorsomedial y perifornical. A pesar de este origen anatómico tan localizado, las neuronas orexinérgicas se proyectan ampliamente a numerosas regiones troncoencefálicas, corticales y límbicas. Desarrollo. Este patrón difuso de distribución de las fibras orexinérgicas estaría indicando la intervención de este sistema peptídico en una amplia variedad de funciones y, de hecho, se ha relacionado con los mecanismos que permiten la regulación del ciclo sueño-vigilia, la ingesta de comida y de bebida y determinados aprendizajes como el aprendizaje de preferencias gustativas. Se ha sugerido también que la alteración en el funcionamiento del sistema orexinérgico explicaría la aparición de determinados trastornos clínicos como la narcolepsia, la obesidad o la adicción a drogas de abuso. Conclusiones. Nuevas investigaciones ayudarán a conocer el funcionamiento de las neuronas orexinérgicas y la interacción entre los sistemas que regulan la emoción, la homeostasis energética y los mecanismos de recompensa con los sistemas que regulan el ciclo de sueño-vigilia. Se confía en que ese conocimiento permita desarrollar nuevos fármacos que, actuando sobre el sistema orexinérgico, sean eficaces en el tratamiento de las alteraciones del sueño como el insomnio o la narcolepsia, de los trastornos de la alimentación o de la drogadicción (AU)


Introduction. Recent research has reported the existence of a new class of neuropeptides, called orexins or hypocretins, which are produced by a small group of neurons in the hypothalamus and whose actions are mediated by two types of receptors: OX1R and OX2R. More specifically, the orexinergic neurons have been located exclusively in cells in the lateral, dorsomedial and perifornical areas of the hypothalamus. Despite this highly specific anatomical origin, the orexinergic neurons are projected widely into a number of brainstem, cortical and limbic regions. Development. This fuzzy pattern of distribution of the orexinergic fibres would be indicating the involvement of this peptidic system in a wide range of functions; indeed, it has been related with the mechanisms that enable regulation of the sleep-wake cycle, the ingestion of food and drink, and some particular types of learning, such as learning certain preferences regarding tastes. It has also been suggested that upsets in the functioning of the orexinergic system would explain the appearance of certain clinical disorders like narcolepsy, obesity or addiction to drug of abuse. Conclusions. Further research will help to determine the functioning of orexinergic neurons and the interaction between the systems that regulate emotion, energetic homeostasis and the reward mechanisms, on the one hand, and the systems that regulate the sleep-wake cycle on the other. That knowledge would almost certainly make it possible to develop new drugs that, by acting upon the orexinergic system, would be effective in the treatment of sleep disorders such as insomnia or narcolepsy, eating disorders or drug addiction (AU)

The high glycemic index diet was an independent predictor to explain changes in agouti-related protein in obese adolescents. Background & Aims: The role of diet glycemic index (GI) in the control of orexigenic and anorexigenic factors of the energy balance is still not clear. The present study aimed to assess whether the habitual diet, according to different GI foods, exerts influence on regulation of energy balance markers and the effects of interdisciplinary intervention in obese adolescents. Methods: A total of 55 obese adolescents, aged from 14 to 19 years, were submited to one year of interdisciplinary therapy and were divided in two groups, according to the predominant dietary pattern of food intake: high-GI group (H-GI; n = 29) and moderate/low-GI group (M/L-GI; n = 26). Results: The concentration of orexigenic factor AgRP (p < 0.01), visceral fat (p=0.04) and visceral/subcutaneous ratio (p = 0.03) were higher in the group of H-GI when compared with M/L-GI group. Moreover, the habitual consumption of H-GI foods was an independent predictor to explain changes in AgRP concentrations. After one year of interdisciplinary therapy, the adolescents presented significant reductions in body weight, total body fat (%), visceral and subcutaneous fat and HOMA-IR, as well as a significant increase of fat free mass (%). Conclusions: Our results may suggest that habitual H-GI diet could upregulate orexigenic pathways, contributing to vicious cycle between undesirable diets, deregulates energy balance and predispose to obesity. One the other hand, one year of interdisciplinary therapy can significant improves metabolic profile and central obesity in adolescents (AU)


La Dieta de alto índice glucémico es un predictor independiente para explicar los cambios en la proteína relacionada al agouti en adolescentes obesos. Introducción y objetivos: El papel de la dieta de índice glucémico (GI) en el control de los factores orexigénicos y anorexígenos del balance de energía todavía no está claro. El presente estudio tuvo como objetivo evaluar si la dieta habitual, de acuerdo con diferentes alimentos con IG, ejerce influencia sobre la regulación de los marcadores del balance de energía y los efectos de la intervención interdisciplinaria en adolescentes obesos. Métodos: Un total de 55 adolescentes obesos, con edades de 14 a 19 años, han sido sometidos a un año de tratamiento interdisciplinario y se dividieron en dos grupos, de acuerdo al patrón de dieta predominante de la ingesta de alimentos: el grupo IG alto (H-GI; n = 29) y GI moderada/bajo grupo (M/L-GI, n = 26). Resultados: La concentración de orexigenic factor de AgRP (p < 0,01), la grasa visceral (p = 0,04) y la relación visceral/subcutánea (p = 0,03) fueron mayores en el grupo de H-GI en comparación con el grupo M/L-GI. Por otra parte, el consumo habitual de alimentos H-GI fue un predictor independiente para explicar los cambios en las concentraciones de AgRP. Después de un año de tratamiento interdisciplinario, los adolescentes presentan una reducción significativa en el peso corporal, la grasa corporal total (%), visceral y la grasa subcutánea y el HOMA-IR, así como un aumento significativo de la masa libre de grasa (%). Conclusiones: Nuestros resultados pueden sugerir que la dieta H-GI habitual podría upregulate vías orexigénicos, contribuyendo al círculo vicioso entre las dietas indeseables, desregula el equilibrio energético y predisponen a la obesidad. Uno por otro lado, un año de tratamiento interdisciplinario puede perfil metabólico mejora significativa y la obesidad central en los adolescentes (AU)

Purpose: The localization and distribution of neuropetideexpression in the cat visual pathway can provideinformation about the function of that pathway.Method: Study of optic pathway in eight cats.Following extraction of the brain, slices were preparedusing a microkeratome. The slices were examinedby indirect immunocytochemistry using antimetenkephalinas antibody to determine the presenceor absence of this pentapeptide in the visualpathway.Results: Met-enkephalin receptors in both corticaland subcortical regions of the brain were detected.This suggests that met-enkephalin could be involvedin the visual mechanism.Conclusions: The presence of met-enkephalinreceptors in both cortical and subcortical regions ofthe brain suggests that this pentapeptide could beinvolved in the visual mechanism(AU)

Debido al rápido aumento de la incidencia de obesidad en los países desarrollados con el consiguiente riesgo para la salud, también han aumentado los fondos dedicados a la investigación biomédica y al desarrollo de medicinas en el área de metabolismo y el apetito. Esto ha aumentado dramáticamente el conocimiento del control neuroendocrino de estos mecanismos. Los primeros estudios que aparecieron en la literatura describiendo al hipotálamo como un órgano fundamental en el control del peso corporal y el apetito son de hace 60 años. Actualmente conocemos algunas de las poblaciones neuronales y las conexiones implicadas en este proceso. La presente revisión describe brevemente las poblaciones neuronales más importantes relacionadas con el control metabólico, como son las del neuropéptido. Y las del propio melanocortina y sus interconexiones. Asimismo, se discute algunos de los factores centrales que recientemente han sido descritos como necesarios para un ajuste fino del sistema metabólico como son los endocanabinoides. Finalmente, se discute cómo los cambios en el ambiente local durante el desarrollo prenatal o neonatal podrían influir posteriormente en el sistema neuroendocrino del adulto haciéndolo más susceptible de padecer obesidad (AU)


Due to the rapidly increasing incidence of obesity in developing countries and the associated health risks, funding for biomedical research and drug development in the area of metabolism and appetite has also increased considerably. This has added dramatically to our understanding of the neuroendocrine control of these mechanisms. It has been over 60 years since the first descriptions of body weight and appetite appeared in the scientific literature. We now know some of the important neuronal populations and connections involved in this process. This review will briefly describe some of the most important neuronal populations involved in metabolic control, such as hypothalamic neuropeptide Y and proopiomelanocortin producing neurons, and their interconnections. We will also discuss briefly some of the central factors that have more recently been drawn to the forefront as being important for fine-tuning the metabolic system, such as the endogenous cannabinoids. Finally, we will briefly discuss how during prenatal or neonatal development changes in the local environment could have lasting influences on the neuroendocrien system in the adult animal, making it more susceptible to becoming overweight (AU)

Periglomerular cells are interneurones thatmodulate the primary sensory information inthe olfactory bulb. It was originally assumedthat periglomerular cells constituted a homogeneousGABAergic population in the rat olfactory bulb, but in other species studies addressing this are scarce. However, several authors have shown that this neuronal type exhibits extraordinarily heterogeneous neurochemicalfeatures. The aim of this review is to compile and describe in detail the expression patterns of neuronal markers in the rat olfactory bulb, in particular in periglomerular cells,and to compare such information with previous data on other macrosmatic and microsmaticanimals. Interspecies differences in the neurochemical composition of periglomerular cells could indicate different modes in the modulation of olfactory information (AU)

El aparato digestivo, más allá de la digestión y absorción de nutrientes, es un órgano endocrino modulador del apetito, la saciedad y el metabolismo hidrocarbonado mediante la producción, por parte de diversas células enteroendocrinas, de numerosos péptidos codificadores de señales orexígenas (ghrelina, orexinas) o anorexígenas (polipéptido pancreático, péptido YY, colecistoquinina, amilina, homólogos de bombesina, apolipoproteína A-IV, polipéptido insulinotrópico dependiente de la glucosa, péptido similar al glucagón tipo 1, oxintomodulina). El conocimiento de las acciones reguladoras del apetito y el metabolismo energético de estos péptidos, a través de su compleja interacción con otras señales periféricas (leptina, adiponectina) y con distintos neuropéptidos centrales, así como la síntesis de análogos, en especial los derivados de la amilina e incretinas, proporciona nuevos enfoques terapéuticos para 2 trastornos íntimamente relacionados: la obesidad y la diabetes mellitus tipo 2


The gastrointestinal tract, besides digesting and processing nutrients, is now regarded as an endocrine organ able to modulate appetite, satiety, and carbohydrate metabolism. Several enteroendocrine cells produce numerous peptides codifying either orexigenic (ghrelin, orexins) or anorexigenic signals (pancreatic polypeptide, peptide YY, cholecystokinin, amylin, bombesin homologs, apolipoprotein A-IV, glucose-dependent insulinotropic polypeptide, glucagon-like peptide 1, oxyntomodulin), which interact in a complex network with other peripheral signals of energy balance and with different neuropeptides involved in the central control of appetite and energy homeostasis. The growing knowledge of the actions of these gastrointestinal peptides on appetite regulation and carbohydrate metabolism, and subsequent synthesis of analogs, particularly those derived from amylin and incretins, herald a new era in the therapy of 2 closely related diseases, obesity and type 2 diabetes mellitus

Introducción. De los aproximadamente 200 péptidos que se sabe que existen en el organismo, 80 realizan funciones neurotransmisoras y unos 20 se han relacionado con la enfermedad de Alzheimer (EA). Desarrollo. En este artículo elaboramos una revisión de los principales estudios que se han realizado con neuropéptidos, tales como la hormona liberadora de corticotropina, la hormona liberadora de tirotropina, la somatostatina, el neuropéptido Y, la oxitocina, la arginina-vasopresina, la galanina, la insulina y los factores de crecimiento similares a insulina, los péptidos similares a glucagón, el péptido intestinal vasoactivo, la colecistoquinina, la sustancia P, los péptidos opiodes y el neuropéptido NAP. Aunque intentamos encontrar una asociación causal con la EA, hallamos en muchos casos resultados contradictorios o poco concluyentes. Conclusión. Lo más destacable podría ser la disminución de la sustancia P en corteza cerebral, hipocampo, ganglios basales y líquido cefalorraquídeo, la somatostatina disminuida en las mismas estructuras salvo en ganglios basales, la disminución de vasopresina salvo en el lóbulo temporal y el aumento en la corteza cerebral de la dinorfina y la leucoencefalina (AU)


Introduction. Of approximately 200 peptides that are known to exist in the body, 80 carry out functions as neurotransmitters and about 20 have been linked to Alzheimer’s disease (AD). Development. In this article we review the most salient studies that have been conducted on neuropeptides such as corticotropin-releasing hormone, thyrotropin-releasing hormone, somatostatin, neuropeptide Y, oxytocin, arginine vasopressin, galanin and insulin, as well as the insulin-like growth factors, the glucagon-like peptides, vasoactive intestinal peptide, cholecystokinin, substance P, opioid peptides and the neuropeptide NAP. Although attempts are made to find a causal association with AD, in many cases the findings are contradictory or not very conclusive. Conclusions. The most notable points could be the reduction in substance P in the cerebral cortex, hippocampus, basal ganglia and cephalospinal fluid; the diminished levels of somatostatin in the same structures except for the basal ganglia; the reduction in the amount of vasopressin except in the temporal lobe; and the increased levels of dynorphin and leucine enkephalin (AU)

La psoriasis es un trastorno dermatológico inflamatoriocrónico. Se cree que es un proceso auto inmune. Diferentesinvestigaciones recientes sitúan a la inflamaciónneurogénica, un proceso proliferativo de fibras nerviosasno mielinizadas, como uno de los factores etiopatogénicoscomunes en la proliferación y mantenimiento de la placapsoriasica. El NGF (Factor de crecimiento Nervioso), unamolécula de la familia de las neurotrofinas, podría ser eliniciador, y perpetuar la señalización celular, mediantesegundos mensajeros, como los neuropéptidos(Substáncia P, Péptido del gen relacionado con lacalcitonina y Péptido Intestinal Vasoacitivo), de dichoproceso inflamatorio, lo que explicaría fenómenos comoel de Koebner o la simetría de las lesiones, en la Psoriasis.Por dicha razón, se comienza a observar a la enfermedad,como un proceso psicosomático. El cerebro generaríaseñalizaciones neuroquímicas, que modularían elproceso periférico, y viceversa. Se explicaría porqué elestrés es el factor de riesgo principal en la psoriasis. Laenfermedad mental, que produce estrés neuronal, pormedio de mecanismos neurobiológicos, podría generar,en el nervio periférico, mecanismos anti-apoptoticos, enpersonas genéticamente vulnerables. Se explicaría tambiénque la misma psoriasis produjera estrés psicosocial,por lo que se cerraría el círculo fisiopatologico


Psoriasis is a cronic inflamatory dermatologic disease.Scientific works believe that is an auto-inmune disease.Novel investigations describe Neurogenic Inflammation,a proliferative process into the unmielinized nervous fibers,like one of the most important common etiopathogenicfactor in the proliferation and maintenance of the psoriaticplaque. The Neural Grow Factor, a molecule of theneurotrophine familily, probably can act like aninicialization and maintenance of the molecular cellularsignaling, with the contribution of other second messengers(Substance P, Calcitonin-gene related peptide andVasoactive Intestinal Peptide), of the inflammatory process,that explain the Koebener phenomenon or the simmetryof the lesions of Psoriasis. For this reason, the disease is actually observed like a psicosomatic illnes. The brain,probably generates neurochemical signaling, thatmodulate the periferical process, and viceversa. Thisexplains the stress like the most important risk factor inthe development of Psoriasis. The mental disease, thatproduce neuronal stress, with neurobiologicalmechanims, probably generates, in the periferal nerve,antiapoptotic mechanims, in genetically vulnerablepatients. This explains, in the other hand, that psoriasisgenerates psicosocial stress, and close the vicious circleof the etiopathogenic mechanisms of Psoriasis

Vasoactive intestinal peptide (VIP) is a neuropeptide produced by the lymphoid cells, which exerts a wide spectrum of immunological functions controlling the homeostasis of immune system through different receptors expressed in various immunocompetent cells. During the last decade, VIP has been clearly identified as a potent anti-inflammatory factor, both in innate and adaptive immunity. In innate immunity, VIP inhibits the production of pro-inflammatory cytokines and chemokines from macrophages, microglia and dendritic cells. In addition, VIP reduces the expression of costimulatory molecules on the antigen-presenting cells, and therefore reduces the activation of antigen-specific CD4+ T cells. In terms of adaptive immunity, VIP promotes Th2-type responses, and reduces the pro-inflammatory Th1-type responses. Several of the molecular mechanisms involved in the inhibition of cytokine and chemokine expression, and in the preferential development and/or survival of Th2 effectors, are perfectly known. Therefore, VIP and its analogues have been recently proposed as very promising candidates, alternative to other existing treatments, for treating acute and chronic inflammatory and autoimmune diseases, such as septic shock, rheumatoid arthritis, multiple sclerosis, Parkinson’s disease, Crohn’s disease, or autoimmune diabetes. The aim of this review is firstly to update our knowledge of the cellular and molecular events relevant to VIP function on the immune system; and secondly to gather together recent data that support its role as a type 2 cytokine. Recognition of the central functions that VIP plays in cellular processes is focusing our attention on this «very important peptide» as exciting new candidates for therapeutic intervention and drug development (AU)


El Péptido Intestinal Vasoactivo (VIP) es un neuropéptido producido por diferentes células inmunitarias, con un amplio abanico de funciones implicadas en el control homeostático del sistema inmunitario. Estos efectos son llevados a cabo a través de varios receptores específicos expresados en distintas células inmunocompetentes. En la última década, se ha identificado a VIP como un potente factor anti-inflamatorio, tanto en la inmunidad innata como en la adquirida. En la inmunidad innata, VIP inhibe la producción de citocinas y quimiocinas proinflamatorias por macró- fagos, microglía y células dendríticas. Además, VIP reduce la expresión de moléculas coestimuladoras en las células presentadoras de antígeno, reduciendo así la estimulación antígeno-específica de células T. En cuanto a la inmunidad adquirida, VIP favorece las respuestas tipo Th2 frente a las proinflamatorias tipo Th1. Actualmente se conocen perfectamente muchos de los mecanismos moleculares y celulares implicados en esta regulación. Por lo tanto, se ha propuesto recientemente a VIP y análogos como candidatos muy prometedores, alternativos a terapias existentes, en el tratamiento de varias enfermedades autoinmunitarias e inflamatorias crónicas, tales como shock endotóxico, artritis reumatoide, esclerosis múltiple, enfermedad de Parkinson y de Crohn, y diabetes tipo I. El principal objetivo de esta revisión es primeramente actualizar nuestro conocimiento sobre los procesos celulares y moleculares relevantes en la función inmunitaria de VIP, y en segundo lugar, reunir todos los datos recientes que sustentan la hipótesis de que este «very important peptide» podría ser considerado una citocina tipo 2 candidato en el diseño futuro de nuevas terapias inmunomoduladoras (AU)

Objetivos. El reciente descubrimiento de las hipocretinas (orexinas) y su vinculación en la fisiopatología de la narcolepsia y el papel de la adenosina como parte de la integración entre metabolismo cerebral y sueño. Desarrollo. El funcionamiento del hipotálamo en la regulación del sueño y la vigilia se ha consolidado ha raíz de los descubrimientos de las hipocretinas y del papel de la adenosina cerebral. Las hipocretinas son dos péptidos de 33 y 28 aminoácidos, cuyas neuronas se localizan de manera predominante en hipotálamo lateral y regiones aledañas. En el modelo de narcolepsia del perro dóberman, en el que presenta esta enfermedad con una patrón autosómico recesivo, se detectó una mutación de uno de los receptores del sistema de las hipocretinas, el receptor hipocretina-2. Los fallos en el sistema de las hipocretinas como factor clave en la narcolepsia, se han consolidado por otros hallazgos en animales de laboratorio y humanos. En el caso de la adenosina, ésta se acumula durante la vigilia como resultado del metabolismo neuronal y esto se relaciona con la somnolencia. Los episodios de sueño disminuyen los niveles cerebrales de esta sustancia. Los antagonistas de receptores a adenosina aumentan la vigilia (p. ej., cafeína), mientras que los agonistas promueven el sueño de onda lentas. Conclusiones. Hipocretinas y adenosina ejercen funciones de regulación sobre el sueño y la vigilia, desde el hipotálamo. El entendimiento de estos dos sistemas puede tener un impacto en problemas clínicos como el insomnio, hipersomnia y otras alteraciones neuropsiquiátricas (AU)


Aims. To review the recent discovery of hypocretins (orexins) and their link to the pathophysiology of narcolepsy and the role of adenosine in the integration of brain metabolism and sleep. Development. The importance of the functions carried out by the hypothalamus in the regulation of sleep and the waking state has been consolidated by the discovery of hypocretins and the role played by cerebral adenosine. Hypocretins are two peptides made up of 33 and 28 amino acids whose neurons are located predominantly in the lateral hypothalamus and surrounding regions. In the Doberman canine narcolepsy model, in which this disease is presented with an autosomal recessive pattern, a mutation was detected in one of the receptors involved in the hypocretin system, namely the hypocretin-2 receptor. Failures in the hypocretin system have been confirmed as a key factor in narcolepsy by other findings in laboratory animals and humans. Adenosine, on the other hand, is accumulated during the waking state as a result of neuronal metabolism and this in turn is related to drowsiness. Sleep episodes lower the levels of this substance in the brain. Adenosine receptor antagonists increase wakefulness (e.g. caffeine), while the agonists promote slow-wave sleep. Conclusions. Hypocretins and adenosine from the hypothalamus perform functions involving the regulation of sleep and wakefulness.
