RESUMO

Objective: The present work aimed at developing novel hybrid molecules for targeting the prostate cancer. It is observed that two human shock proteins Hsp70 and Hsp90 are over-expressed in prostate cancer making them one of the important drug targets. We have designed and developed twelve new hybrid molecules 6a-j for targeting these proteins. Methods: The designed molecules were prepared following a four step reaction protocol and characterized on the basis of proton NMR and Mass spectrometry. These were subjected to in vitro studies by means of Oncotest and CCK-8 assays with two cell lines DU145 and 22Rv1. The selected molecules 6b and 6i were subjected to molecular docking and then for SPR based affinity assay. Results: Compounds 6b and 6i were found to be highly active anticancer compounds comparable to standard drug enzalutamide. They have significant IC50 and high dock score for the Hsp70 and Hsp90. These compounds are selective and have good binding affinity for the Hsp70 due to high Kd. Conclusion: Compound 6b and 6i can serve as lead molecules for the development of antiprostate cancer drugs with Hsp70 as target


Objetivo: El presente trabajo tuvo como objetivo desarrollar nuevas moléculas híbridas para atacar el cáncer de próstata. Se observa que dos proteínas de choque humano, Hsp70 y Hsp90, se sobreexpresan en el cáncer de próstata, lo que las convierte en uno de los objetivos farmacológicos importantes. Hemos diseñado y desarrollado doce nuevas moléculas híbridas 6a-j para dirigir estas proteínas. Métodos: Las moléculas diseñadas se prepararon siguiendo un protocolo de reacción de cuatro etapas y se caracterizaron sobre la base de RMN de protón y espectrometría de masas. Estos se sometieron a estudios in vitro por medio de ensayos Oncotest y CCK-8 con dos líneas celulares DU145 y 22Rv1. Las moléculas seleccionadas 6b y 6i se sometieron a acoplamiento molecular y luego a ensayo de afinidad basado en SPR. Resultados: Se descubrió que los Compuestos 6b y 6i son compuestos anticancerígenos muy activos comparables al fármaco estándar enzalutamida. Tienen un IC50 significativo y una puntuación alta para el muelle de Hsp70 y Hsp90. Estos compuestos son selectivos y tienen una buena afinidad de unión por la Hsp70 debido a la alta Kd. Conclusión: Los compuestos 6b y 6i pueden servir como moléculas principales para el desarrollo de fármacos antiprostáticos contra el cáncer con Hsp70 como objetivo

Chlamydia pneumoniae is a Gram-negative, obligate intracellular pathogen that causes community-acquired respiratory infections. After C. pneumoniae invades host cells, it disturbs the vesicle transport system to escape host lysosomal or autophagosomal degradation. By using a yeast mis-sorting assay, we found 10 C. pneumoniae candidate genes involved in aberrant vesicular trafficking in host cells. One of the candidate genes, CPj0783, was recognized by antibodies from C. pneumoniae-infected patients. The expression of CPj0783 was detected at mid to late-cycle time points and increased during the inclusion maturation. Two-hybrid screening in yeast cells revealed that CPj0783 interacted with Huntingtin-interacting protein 14 (HIP14). The specific interaction between CPj0783 and HIP14 could be demonstrated by an in vivo co-immunoprecipitation assay and an in vitro GST pull-down assay. It was also demonstrated that HIP14 was localized in the Golgi apparatus and colocalized with CPj0783. HIP14 has a palmitoyl transferase activity that is involved in the palmitoylation-dependent vesicular trafficking of several acylated proteins. These findings suggest that CPj0783 might cause abnormal vesicle-mediated transport by interacting with HIP14 (AU)

El arco aórtico izquierdo con arteria subclavia derecha aberrante constituye la anomalía vascular congénita más común del arco aórtico. En el 60% de casos se origina de un segmento dilatado, denominado divertículo de Kommerell. El aneurisma de la arteria subclavia derecha aberrante es raro, puede ser clínicamente silente o cursar con síntomas inespecíficos y su rotura se asocia a una elevada mortalidad. No hay criterios precisos para el tratamiento, pero se recomienda su reparación cuando se producen síntomas por compresión o cuando alcanza 30-50mm. La radiografía puede hacer sospechar la malformación, pero la resonancia magnética (RM) o la tomografía computarizada (TC) son las pruebas de elección para realizar el diagnóstico y planificar el tratamiento. Presentamos un caso de una arteria subclavia derecha aberrante con un pequeño aneurisma calcificado en un divertículo de Kommerell que provocó dolor torácico y disfagia y fue tratado mediante un procedimiento combinado endovascular y quirúrgico (AU)


Left aortic arch with aberrant right subclavian artery is the most common congenital vascular anomaly involving the aortic arch. In 60% of cases, the aberrant right subclavian artery arises from a dilated segment of the aortic arch called Kommerell's diverticulum. Aneurysm of the aberrant right subclavian artery is rare; this condition could remain clinically silent or it could originate nonspecific symptoms. Rupture of an aneurysm of the aberrant right subclavian artery is associated with high mortality. Although there are no exact criteria to indicate the treatment of this condition, repair of the aneurysm is recommended when symptoms occur or when it reaches a size of 30mm to 50mm. The malformation can be suspected at plain-film X-ray examination, but magnetic resonance imaging (MRI) or computed tomography (CT) are the imaging tests of choice for the diagnosis and for planning treatment. We present the case of a patient with an aberrant right subclavian artery with a small calcified aneurysm in a Kommerell's diverticulum that caused chest pain and dysphagia; the patient underwent a procedure combining endovascular and surgical treatment (AU)

Antecedentes. La determinación del virus del papiloma humano (VPH) en muestras ginecológicas es en la actualidad un elemento clave para la toma de decisiones en patología cervical. El método más contrastado clínicamente es la captura de híbridos de segunda generación (HC2). Métodos. Estudio comparativo de los resultados obtenidos, mediante HC2 y Cervista™, para la detección de VPH de alto riesgo (VPH-AR) en muestras ginecológicas. Resultado. La comparación de resultados muestra una concordancia del 96,4%, con una sensibilidad de Cervista™ para CIN2+ del 98,3% y una especificidad del 98,4%. Conclusiones. Creemos que Cervista™ es un buen método para la detección de VPH-AR, aplicable tanto en el cribado como en la clínica(AU)


Introduction. HPV detection in gynaecological samples currently provides key data in cervical pathology for diagnostic and management decisions. At present HC2 is the most clinically proven method for this purpose. Methods. Comparative study of HPV-HR detection between HC2 and Cervista™. Results. The concordance between the two methods is 96.4%. The sensitivity of Cervista™ for CIN2+ is 98.3% and specificity exceeds 98.4%. Conclusions. We conclude that Cervista™ is an appropriate method for the detection of HPV-HR in gynecological samples for screening and clinical purposes(AU)

Objetivo. Estudiar el valor de la carga viral para predecir la persistencia y/o la progresión en LSIL. Diseño del estudio. Estudio prospectivo. Todos los casos diagnosticados como LSIL en el año 2006, y seguidos a los 6, 12 y 24 meses después del diagnóstico. Método. Citología líquida (ThinPrep®, Hologic, Malborough, MA, EE. UU.) para realizar el estudio citológico. Test de captura de híbridos (HCII-HR, Quiagen, Hilden, Alemania), utilizando el test para determinar la presencia de VPH de alto riesgo. Resultados. De los 102 casos, 87 fueron HPV positivos (85,3%). La persistencia o progresión se observó en el 54% de los casos en el primer control, en el 42,4% en el segundo control y en el 33,3% en el tercer control. No se demostró lesión en el seguimiento en el 46% de los casos en el primer control, en el 57% de los casos en el segundo control y en el 66,7% de los casos en el tercer control. Diecinueve casos progresaron a HSIL: 11 detectados en el primer control, 6 en el segundo y 2 en el tercero. La sensibilidad para HSIL nunca superó el 89%, con una especificidad siempre menor del 40%. Conclusiones. La determinación de la carga viral mediante HC2 en LSIL no es útil para predecir su comportamiento(AU)


Objective. To study the HPV viral load (VL) in the prediction of the persistence and/or progression in LSIL cases. Study design. Prospective study. 102 consecutive cases diagnosed as LSIL in 2006 with follow-ups at 6, 12 and 24 months subsequent to diagnosis. Method. Liquid based cytology (ThinPrep®, Hologic, Malborough, MA, USA), for the cytological study. Hybrid Captute II Test (HCII-HR, Quiagen, Hilden, Germany), for the determination of the presence or absence of High-Risk HPV. Results. 87 of the 102 cases studied were HR-HPV positive (85.3%). The presence or progression was detected in 54%, 42% and 33.35% of the cases in the first, second and third controls respectively. No lesion was demonstrated in 46%, 57% and 66.7% of the cases at first, second and third controls respectively. 19 cases progressed to HSIL; 11 of which were detected on the first control, 6 on the second and 2 on the third. The sensitivity of the VL for detecting HSIL never exceeded 89%, with a specificity always inferior to 40%. Conclusion. The VL determined by the HC2 method is not a useful indicator of prognosis in the follow-up of LSIL cases(AU)

Introducción. La disección aórtica y los aneurismas de aorta torácica se consideran procesos graves que afectana una población importante. La reparación quirúrgica tradicional supone el empleo de técnicas complejas con unaalta morbimortalidad, mayor aún cuando la enfermedad afecta retrógradamente al arco aórtico. Es posible realizar unprocedimiento endovascular sobre el arco aórtico mediante la asociación de técnicas de revascularización de los troncossupraaórticos con una tasa de éxito aceptable en pacientes que, por su edad o comorbilidad, no son candidatos a una reparaciónquirúrgica convencional. Caso clínico. Presentamos un caso de tratamiento híbrido, quirúrgico y endovascularen un varón con un aneurisma de aorta torácico secundario a disección crónica tipo B, con afectación retrógrada del arcoaórtico. Se consigue la exclusión completa del aneurisma, con permeabilidad de ramas viscerales y reducción significativadel diámetro mayor al año del tratamiento. No se objetivan complicaciones mayores durante el seguimiento y el pacienterealiza una vida activa en la actualidad. Conclusiones. El tratamiento híbrido puede ser una alternativa eficaz con menormorbilidad que la cirugía convencional en pacientes de alto riesgo con patología del cayado aórtico. Se precisan estudiosmás amplios para demostrar la viabilidad, tasas de morbilidad y mortalidad, complicaciones y duración de estas técnicas,ya que los existentes corresponden a series muy cortas de casos seleccionados


Introduction. Aortic dissection and thoracic aortic aneurysms are considered to be serious conditions thataffect a large number of patients. Traditional surgical repair entails the use of complex techniques with high morbidityand mortality rates, which become even higher when the disease affects the aortic arch retrogradely. An endovascularprocedure can be performed on the aortic arch by associating supra-aortic trunk revascularisation techniques, withan acceptable rate of success in patients who, owing to their age or comorbidity, are not suitable candidates forconventional surgical repair. Case report. We report a case of hybrid (surgical and endovascular) treatment in a malepatient with thoracic aortic aneurysm secondary to type B chronic dissection, with retrograde compromise of the aorticarch. Complete exclusion of the aneurysm was achieved, with patency of the visceral branches and a significantreduction in the major diameter at one year after treatment. No important complications were observed during thefollow-up and the patient currently leads an active life. Conclusions. Hybrid treatment can be an effective alternative,with a lower rate of morbidity, than conventional surgery in high risk patients suffering from a pathology affecting theaortic arch. Further studies are needed to show the feasibility, morbidity and mortality rates, complications andduration of these techniques, since those that have been conducted to date included only very short series of selected cases
