Introducción: los marcadores de repeticiones cortas en tándem del cromosoma Y (Y-STR) se ubican en la región no recombinante del cromosoma Y, su herencia es por vía paterna, no son detectables en el ADN femenino. Estas propiedades hacen de los Y-STR una herramienta útil en las investigaciones forenses, como las agresiones sexuales, paternidades y otros delitos violentos; asimismo son útiles en estudios genealógicos y evolutivos. El objetivo de la investigación es ampliar la evidencia científica de la distribución por regiones o país de los haplotipos del cromosoma sexual Y, estudios similares en poblaciones peruanas son escasas debido al número menor de polimorfismos Y-STR de uso frecuente en genética forense y de poblaciones. Material y métodos: en la investigación se analizaron 141 muestras de ADN de la selva del Perú, de las que 104 muestras corresponden a la región de Iquitos (Loreto), 29 muestras son Awajun (Amazonas) y 8 muestras de Tambopata (Madre de Dios). Las muestras fueron procesadas empleando PCR directa con el kit Yfiler™ Plus PCR Amplification para 27 STR, los productos amplificados fueron analizados por electroforesis capilar en el Applied Biosystem™ 3500XL Genetic Analyzer y los datos obtenidos se importaron al software GeneMapper® ID-X v1.5 para generar los perfiles genéticos. Con los resultados obtenidos se realizó el análisis estadístico y la estructura poblacional. Resultados: de las 141 muestras se obtuvieron 106 haplotipos únicos. La diversidad genética para cada marcador Y-STR estuvo entre 0,317 y 0,919. La diversidad haplotípica para la muestra total fue de 0,9906. El estudio registra que los haplotipos Y-STR estudiados presentaron elevado polimorfismo en la población analizada y, por lo tanto, son de gran utilidad en estudios forenses de identificación humana, así como en genética de poblaciones cuando se investigan grupos o individuos de América Latina. Y-chromosome-specific short tandem repeat (Y-STR) markers reside on the non-recombinant portion of the Y chromosome, their inheritance is paternal, they are not detectable in female DNA. These properties make Y-STRs a useful tool in forensic investigations such as sexual assault, parenting, and other violent crimes; likewise they are also useful in genealogical and evolutionary studies. The objective of the research is to expand the scientific evidence of the distribution by region or country of the Y sex chromosome haplotypes. Similar studies in Peruvian populations are scarce due to the smaller number of Y-STR polymorphisms frequently used in Forensic and Population Genetics. Material and method: In the investigation, 141 DNA samples from the jungle of Peru were analyzed, of which comprised of 104 samples from Iquitos region (Loreto), 29 samples from Awajun (Amazonas) and 8 samples from Tambopata (Madre de Dios). The samples were processed using direct PCR with the Yfiler™ Plus PCR Amplification kit for 27 STRs, the amplified products were analyzed by capillary electrophoresis on the Applied Biosystem™ 3500XL Genetic Analyzer, and the data obtained was imported into the GeneMapper® ID-X v1.5 software to generate the genetic profiles. With the results obtained, the statistical analysis and the population structure were carried out. Results: Of the 141 samples, 106 unique haplotypes were observed. Gene diversities for each Y-STR marker ranged from 0.317 to 0.919. The haplotype diversity for the total sample was 0.9906. This study supports that the Y-STR haplotypes in this population are highly polymorphic in the analyzed population and, therefore, are very useful in forensic studies of human identification, as well as in population genetics when investigating groups or individuals from Latin America. Introducción: el haplotipo GA de los polimorfismos rs1554483 y rs4864548 se ha asociado con componentes del síndrome metabólico como la hipertensión arterial y los niveles de triglicéridos. Sus portadores presentan un riesgo de obesidad 1,5 veces mayor que el resto de la población. Metodología: se obtuvieron los SNP rs1554483 y rs4864548 de 2504 individuos desde la base de datos “1000genomes phase 3”. Los datos se agruparon en cinco macropoblaciones (África, Asia Oriental, Asia Meridional, Europa y Latinoamérica) cubriendo un total de 26 poblaciones. Se analizaron las diferencias en la frecuencia del haplotipo entre las macropoblaciones y las poblaciones, para lo cual se utilizó el estadístico F de Fisher. Resultados: la macropoblación de África presentó la menor frecuencia (17,9 %) y la del Este de Asia la mayor (57,4 %). Dentro de las poblaciones existe una relativa homogeneidad en las frecuencias, excepto en el caso de las que componen la macropoblación de Latinoamérica, donde la población peruana de Lima y la puertorriqueña presentan frecuencias mucho mayores que el resto. Conclusiones: el haplotipo GA presenta heterogeneidad entre las macropoblaciones, lo que sugiere procesos microevolutivos altamente diferenciados entre los continentes. Se propone estudiar la asociación del haplotipo GA con otros polimorfismos, como rs3749474, rs11932595 y rs6859524, que también se han asociado con el riesgo de obesidad y factores asociados al síndrome metabólico. Introduction: the GA haplotype of polymorphisms rs1554483 and rs4864548 has been associated with components of the metabolic syndrome such as high blood pressure and triglyceride levels; its carriers have a risk of obesity, 1.5 times higher than the rest of the population. Methodology: SNP rs1554483 and rs4864548 were obtained from 2504 individuals from the "1000genomes phase 3" database. Data were grouped into five macro populations (Africa, East Asia, South Asia, Europe and Latin America) covering a total of 26 populations. Differences in haplotype frequency between macro populations and populations were analyzed, for which Fisher's F statistic was used. Results: the macro population of Africa presented the lowest frequency (17.9 %) and that of East Asia the highest (57.4 %). Within the populations there is a relative homogeneity in the frequencies, except in the case of those that make up the macro population of Latin America where the Peruvian population of Lima and the Puerto Rican population present much higher frequencies than the rest. Conclusions: the GA haplotype presents heterogeneity between macro populations, which suggests highly differentiated micro evolutionary processes between continents. We propose to study the association of the GA haplotype with other polymorphisms such as rs3749474, rs11932595 and rs6859524 that have also been associated with risk of obesity and factors associated with metabolic syndrome. Introducción: Las variantes C1236T, G2677T/A y C3435T del gen ABCB1 alteran la función de la glicoproteína P y el transporte de sustancias endógenas y exógenas en la barrera hematoencefálica; además, actúan como factores de susceptibilidad para algunas enfermedades neurodegenerativas.El objetivo del estudio fue determinar la asociación de polimorfismos ABCB1 (C1236T, G2677T/A y C3435T), sus haplotipos y sus combinaciones de genotipos con la enfermedad desmielinizante.MétodoSe genotipificó a 199 pacientes con enfermedad desmielinizante y a 200 controles mestizos mexicanos mediante PCR-RFLP y secuenciación Sanger para comparar las frecuencias de alelos, genotipos, haplotipos y combinaciones de genotipos entre pacientes y controles. El análisis estadístico se realizó con regresión logística y χ2 de Pearson al 95% de confianza; se calculó la OR y se evaluó la asociación con enfermedad desmielinizante.ResultadosLos haplotipos TTT y CGC fueron los más frecuentes en pacientes y controles. El alelo G2677 (OR = 1,79; IC 95%: 1,12-2,86; p = 0,015) muestra asociación con enfermedad desmielinizante, así como los genotipos GG2677 (OR = 2,72; IC 95% = 1,11-6,68; p = 0,025) y CC3435 (OR = 1,82; IC 95%: 1,15-2,90; p = 0,010) y su combinación GG2677/CC3435 (OR = 2,02; IC 95%: 1,17-3,48; p = 0,010) y el haplotipo CAT (OR = 0,21; IC 95%: 0,05-0,66; p = 0,001).Los portadores TTTTTT presentaron la edad de inicio más temprana (23,0 ± 7,7 vs. 31,6 ± 10,7; p = 0,0001).ConclusionesLa combinación de genotipos GG2677/CC3435 está asociada al desarrollo de enfermedad desmielinizante en esta muestra, principalmente en el sexo masculino, en el cual puede darse acumulación tóxica de sustratos de glicoproteína P.En este estudio, la edad de inicio de la enfermedad desmielinizante podría ser modulada diferencialmente entre sexos por el alelo G2677 del gen ABCB1. Introduction: The C1236T, G2677T/A, and C3435T variants of the ABCB1 gene alter the functioning of P-glycoprotein and the transport of endogenous and exogenous substances across the blood-brain barrier, and act as risk factors for some neurodegenerative diseases.This study aimed to determine the association between demyelinating disease and the C1236T, G2677T/A, and C3435T variants of ABCB1 and its haplotypes and combinations of genotypes.MethodsPolymerase chain reaction with restriction fragment length polymorphism analysis (PCR-RFLP) and Sanger sequencing were used to genotype 199 patients with demyelinating disease and 200 controls, all Mexicans of mixed race; frequencies of alleles, genotypes, haplotypes, and genotype combinations were compared between patients and controls. We conducted a logistic regression analysis and calculated chi-square values and 95% confidence intervals (CI); odds ratios (OR) were calculated to evaluate the association with demyelinating disease.ResultsThe TTT and CGC haplotypes were most frequent in both patients and controls. The G2677 allele was associated with demyelinating disease (OR: 1.79; 95% CI: 1.12-2.86; P = .015), as were the genotypes GG2677 (OR: 2.72; 95% CI: 1.11-6.68; P = .025) and CC3435 (OR: 1.82; 95% CI: 1.15-2.90; P = .010), the combination GG2677/CC3435 (OR: 2.02; 95% CI, 1.17-3.48; P = .010), and the CAT haplotype (OR: 0.21; 95% CI: 0.05-0.66; P = .001).TTTTTT carriers presented the earliest age of onset (23.0 ± 7.7 years, vs. 31.6 ± 10.7; P = .0001).ConclusionsThe GG2677/CC3435 genotype combination is associated with demyelinating disease in this sample, particularly among men, who may present toxic accumulation of P-glycoprotein substrates.In our study, the G2677 allele of ABCB1 may differentially modulate age of onset of demyelinating disease in men and women. Introducción: Las células natural killer (NK) juegan un papel importante en la defensa contra las células tumorales. El desarrollo y la función de las células NK se rige por un equilibrio dinámico entre la inhibición y la activación de los receptores de la superficie celular, incluidos los receptores KIR. Pacientes y método: se realiza un estudio de casos y controles que compara a un grupo de 46 niños diagnosticados de enfermedades malignas, el grupo control está constituido por 82 niños sanos. Se determinaron y compararon entre grupos los genes, haplotipos y ligandos KIRs. Resultados: no existen diferencias en genes KIRs, haplotipos KIRs ni en ligandos de genes KIRs entre grupos. Sin embargo, al estudiar conjuntamente KIRs y ligandos, k2DS1_C2 fue significativamente superior en el grupo de niños oncológicos (p̊=̊0,016). Conclusiones: Nuestros resultados no proporcionan evidencia de una asociación entre enfermedades oncológicas pediátricas con genotipos y grupos de genes KIRs. El genotipo k2DS1_C2 podría predisponer a la susceptibilidad a procesos malignos en la población infantil. (AU)


Introduction: Natural killer (NK) cells play an important role in defense against tumor cells. The development and function of NK cells is governed by a dynamic balance between inhibition and activation of cell surface receptors, including KIR receptors. Patients and method: A case-control study is carried out that compares a group of 46 children diagnosed with malignant diseases, the control group is made up of 82 healthy children. KIRs genes, haplotypes and ligands were determined and compared between groups. Results: There are no differences in KIRs genes, KIRs haplotypes or in KIRs gene ligands between groups. However, when KIRS and ligands were jointly studied, k2DS1_C2 was significantly higher in the group of cancer children (p̊=̊0.016). Conclusions: Our results do not provide evidence of an association between pediatric cancer disease with genotypes and groups of genes KIRs. The k2DS1_C2 genotype could predispose to susceptibility to malignant processes in children. (AU)

Hasta el momento, la mayor parte de los casos de hipercolesterolemia familiar (60-80%) se atribuyen a variantes patogénicas en el gen LDLR. Solo un 1-5% de los casos se produce por variantes en el gen APOB y un 0-3% por variantes en el gen PCSK9. Existen gran variedad en mutaciones patogénicas conocidas del gen LDLR mientras que, para las que afectan al gen APOB, la de mayor incidencia es p.Arg3527Gln, descrita predominantemente en poblaciones de Centroeuropa y América del Norte. En la Península Ibérica el gen predominante afectado es el del receptor de LDL, similar al resto del mundo, siendo la afectación del gen APOB descrita en individuos del noroeste y anecdótica en el resto del territorio. Analizamos genéticamente la población asistida en el primer año de una consulta de lípidos del suroeste de España con puntuación≥6 puntos de las clínicas de lípidos holandesas y describimos los hallazgos genéticos, bioquímicos y clínicos. Los primeros hallazgos muestran indicios de una posible mayor prevalencia de pacientes con mutación en el gen APOB respecto a otros territorios. Encontramos hechos históricos que darían una posible explicación a este hecho, apoyando así dicha presunción.(AU)


So far, most cases of hypercholesterolaemia (60-80%) are attributed to pathogenic variants in the LDLR gene. Only 1-5% of cases are caused by variants in the APOB gene, and 0-3% by variants in the PCSK9 gene. There is a large variety in known pathogenic mutations of the LDLR gene, while for those affecting the APOB gene, the highest incidence is p.Arg3527Gln, described predominantly in Central European and North American populations. In the Iberian Peninsula the predominant gene affected is that of the LDL receptor, similar to the rest of the world, with the involvement of the APOB gene being described in individuals from the northwest, and anecdotal in the rest of the territory. A genetics analysis was performed on the population attending the first year of a lipid clinic in southwestern Spain with a 6-point score from the Dutch lipid clinics. The genetic, biochemical and clinical findings are described. The first findings show indications of a possible higher prevalence of patients with mutation in the APOB gene compared to other territories. Historical evidence is presented that could give a possible explanation to this, thus supporting the assumption.(AU)

Introduction and objectives Chronic spontaneous urticaria (CSU) is thought to be an autoimmune disease in a subpopulation of patients. Protein tyrosine phosphatase-22 (PTPN22) polymorphisms are considered to be one of the strongest contributing factors to autoimmune diseases. In this study, we aimed to investigate the potential association of several PTPN22 single nucleotide polymorphisms (SNPs) with CSU in an Iranian population. Material and methods A total of 93 CSU patients and 100 healthy individuals were included in this study. Five SNPs within the PTPN22 gene were analyzed using TaqMan genotyping assays. The frequency of alleles, genotypes, and haplotypes of PTPN22 SNPs (rs12760457, rs2476601, rs1310182, rs1217414, and rs33996649) was investigated. Results A significantly higher prevalence of the rs1310182 T allele was observed among patients compared with controls [OR = 1.75 (95% CI: 1.17–2.63); P = 0.007]. In addition, the rs1310182 CC genotype and TT genotype were 0.47 and 2.06 times more common in patients, respectively (P = 0.03). Moreover, haplotype analysis demonstrated that CGCGC, CGTGC, and TGCGC (P < 0.001) were significantly associated with CSU. No significant differences were observed between the patients and controls in the other analyzed PTPN22 SNPs. Conclusions Polymorphisms of the PTPN22 gene are associated with an increased susceptibility to CSU in the studied Iranian population (AU)

INTRODUCCIÓN: en la enfermedad celiaca (EC), la activación de la respuesta inmune lleva a una alteración de la red local de citoquinas. La IL-10 es una citoquina antiinflamatoria esencial en la prevención de patologías inflamatorias. OBJETIVO: analizar la asociación de polimorfismos de nucleótido simple ubicados en el promotor del gen de interleuquina 10 con la EC, en una población de la provincia de Misiones, Argentina. PACIENTES Y MÉTODOS: se extrajo ADN de sangre entera de 40 pacientes con EC y de 80 controles y se amplificó una región en el promotor del gen de IL10 que contiene los polimorfismos rs1800896A/G, rs1800871T/C y rs1800872A/C. Se estableció el riesgo mediante el cálculo de odds ratios (OR), considerando estadísticamente significativo un p < 0,05. RESULTADOS: entre pacientes celiacos y controles no se observaron diferencias significativas en la distribución de los genotipos del polimorfismo rs1800896. La frecuencia de los genotipos CC de rs1800871T/C y rs1800872A/C fue menor en los celiacos (35 % vs. 65 %; p = 0,002). Presentaron riesgo de EC los portadores del alelo menos frecuente T del rs1800871T/C y del alelo menos frecuente A del rs1800872A/C con un modelo dominante (OR = 2,79; IC 95 %: 1,27-6,09; p = 0,01). Se encontró un efecto de riesgo del haplotipo ATA (OR = 3,05; IC 95 %: 1,25-7,46; p = 0,01). CONCLUSIÓN: los portadores del alelo menos frecuente T del rs1800871T/C y del alelo menos frecuente A del rs1800872A/C en el promotor del gen IL10 presentan riesgo elevado de EC con un modelo dominante, sin mostrar riesgo para el rs1800896A/G. El haplotipo ATA muestra asociación para el desarrollo de EC

La distrofia miotónica tipo 1 (DM1) es la forma más común de distrofia muscular en adultos causada por la expansión de una repetición de trinucleótidos CTG en el gen DMPK (Distrofia miotónica proteína quinasa) y su herencia es autosómica dominante. Hasta el momento, la técnica de Diagnóstico Genético Preimplantacional parece ser la aproximación reproductiva más eficaz para aquellas parejas en las que alguno de sus progenitores es afecto de DM1. En este estudio se evaluó la técnica de Diagnóstico Genético Preimplantacional en función del sexo del progenitor afecto y el número de repeticiones CTG y su influencia sobre el resultado reproductivo de la misma. Un total de 13 parejas, del programa de Diagnóstico Genético Preimplantacional de la Unidad de Reproducción Asistida del Hospital Universitari i Politècnic La Fe, fueron incluidas en el estudio. En 8 de éstas la afecta es la mujer, mientras que en las 5 restantes es el hombre el portador de la enfermedad. En todas ellas se llevaron a cabo todos los pasos del programa. Los embriones fueron analizados mediante mTP-PCR y PCR multiplex. El efecto de ambas variables se analizó mediante análisis bivariado (prueba T, Chi-cuadrado, regresión lineal y test Anova) y análisis multivariante utilizando análisis de correlación (Rho de Spearman). Los resultados obtenidos revelaron una transmisión preferencial de los alelos expandidos en DM1, independiente del sexo del progenitor afecto. A pesar de ello, no mostraron ninguna relación estadísticamente significativa entre el sexo del progenitor afecto ni el número de repeticiones CTG y el resultado reproductivo de la técnica. De modo que, atendiendo a nuestros resultados, el Diagnóstico Genético Preimplantacional sería una buena aproximación reproductiva en casos de DM1 con una tasa de gestación de 53,8 % y de nacidos sanos de 38,5 %, independientemente del sexo del progenitor afecto y del número de repeticiones CTG


Myotonic dystrophy type 1 (DM1) is the most common form of muscular dystrophy in adults caused by the expansion of a CTG trinucleotide repeat in the DMPK gene (dystrophia myotonica-protein kinase gene) and its inheritance is autosomal dominant. So far, the technique of Preimplantation Genetic Diagnosis (PGD) seems to be the most effective reproductive approach for those couples in which one of their parents is affected by DM1. In this study, the Preimplantation Genetic Diagnosis technique was evaluated according to the sex of the affected progenitor and the number of CTG repetitions and their influence on the reproductive result of itself. A total of 13 couples from the Preimplantation Genetic Diagnosis program of the Assisted Reproduction Unit of the Hospital Universitario i Politècnic La Fe were included in the study were included in the study. In 8 of them the affected parent is the woman, whereas the 5 remaining is the man who is the disease carrier. In all of them, all the steps of the program were followed. The embryos were analyzed by mTP-PCR and multiplex PCR. The variables effect was analyzed by bivariate analysis (T-test, Chi-square, linear regression and Anova test) and multivariate analysis using correlation analysis (Spearman's rho). The results obtained revealed a preferential transmission of the expanded alleles in DM1, regardless of the sex of the affected parent. In spite of, the results obtained do not show any statistically significant relation between the sex of the progenitor and the number of repetitions and the reproductive result of the technique. Thus, according to our results, Preimplantation Genetic Diagnosis would be a good reproductive approach in cases of DM1 with a gestation rate of 53.8% and healthy births of 38.5%, regardless of the sex of the affected parent and the number of CTG repetitions

Introducción y objetivo: Cambios moleculares en el gen CTLA-4 pueden modificar la habilidad para controlar la proliferación de los linfocitos T, y promover la persistencia o eliminación del virus de la hepatitis C (VHC). Nuestro objetivo fue investigar la frecuencia y asociación de los polimorfismos −319 C/T y +49 A/G del gen CTLA-4, en pacientes con infección por VHC. Métodos: Los polimorfismos del gen CTLA-4 (−319 C/T en la región promotora y +49 A/G en el exón 1) fueron analizados por T-ARMS-PCR en 420 individuos, incluidos 205 pacientes con infección crónica por VHC y 215 sujetos sanos. Resultados: Se encontró una asociación positiva del alelo +49G con la infección por VHC (OR 1,48; IC 95% 1,09-2,02; p=0,02), y con el sexo masculino (OR 1,80; IC 95% 1,16-2,79; p=0,02), ambos en enfermedad crónica (sin cirrosis). Se observaron diferencias significativas en la distribución de los genotipos del polimorfismo +49 A/G, entre los pacientes con infección por VHC y los sujetos sanos en un modelo genético dominante (GG+GA frente a AA; OR1,57; IC 95% 1,05-2,33; p=0,04). No se observaron diferencias en las frecuencias del polimorfismo −319 C/T, entre pacientes con VHC y sujetos sanos. El haplotipo -319C/+49G confiere susceptibilidad a la infección por el genotipo 3 del VHC (OR 10,68; IC 95% 1,17-96,97; p=0,04). Conclusiones: El alelo +49G confiere susceptibilidad a infección por VHC y a infección en el sexo masculino, ambos en enfermedad crónica. Además, el haplotipo -319C/+49G confiere susceptibilidad a la infección por el genotipo 3 del VHC. Nuestros resultados evidencian una implicación importante de los polimorfismos −319 C/T y +49 A/G en la infección por VHC


Introduction and objective: Molecular changes in the CTLA-4 gene can modify the ability to control T lymphocyte proliferation, and promote the persistence or elimination of the hepatitis C virus (HCV). We aimed to investigate the frequency and association of -319 C/T and +49 A/G polymorphism in the CTLA-4 gene in patients infected with HCV. Methods: The CTLA-4 gene polymorphisms (-319 C/T in the promoter region, and +49 A/G in exon 1) were analysed by T-ARMS-PCR in 420 individuals, including 205 chronic HCV infected patients and 215 healthy subjects. Results: We found a positive association of +49G allele with HCV infection (OR 1.48; 95% CI 1.09-2.02; p=.02), and with males (OR 1.80; 95% CI 1.16-2.79; p=.02), both in chronic disease (without cirrhosis). Also, significant differences in +49 A/G genotypes distribution between HCV infected patients and healthy subjects were shown in a dominant genetic model (GG+GA versus AA; OR 1.57; 95% CI 1.05-2.33; p=.04). No significant differences were observed in the -319 C/T polymorphism between HCV infected patients and healthy subjects. Moreover, -319C/+49G haplotype confers susceptibility to HCV genotype 3 infection (OR 10.68; 95% CI 1.17-96.97; p=.04). Conclusions: The +49G allele confers susceptibility to HCV infection and with male gender, both in chronic disease. In addition, the −319C/+49G haplotype confers susceptibility to HCV genotype 3 infection. Our results support an important role of the −319 C/T and +49 A/G polymorphisms in HCV infection

Background: Atopic dermatitis is an inflammatory skin disease in which both genetic and environmental factors interact to determine the susceptibility and severity of the disease. Objective: The aim of this study was to determine the association between atopic dermatitis and IL-10 and TGF-Beta1 gene polymorphisms. Methods: The allele and genotype frequencies of genes encoding for IL-10 and TGF-Beta1 were investigated in 89 patients with atopic dermatitis in comparison with 138 in the control group using the PCR-SSP method. Results: A significant increase was found in the frequency of the TGF-Beta1 codon 10/C allele among patients (p < 0.001, OR = 6.77), whereas a significant decrease was observed in the frequency of the T allele at the same position (p < 0.001, OR = 0.14). The frequency of the TGF-Beta1 codon 25/G allele in the control group was significantly higher than among patients (p < 0.001, OR = 0.08). A significant positive correlation was seen between CC (p < 0.001, OR = 15.10) and CG (p < 0.001) genotypes and AD at codons 10 and 25, respectively. The most frequent haplotypes among patients was TGF-Beta1 CG which was significantly higher than in the control subjects (50% in patients vs. 39.9% in controls, p = 0.042). A significant increase was found in the frequency of TGF-Beta CC (36% in patients vs. 7.6% in controls, p < 0.001) and TC (14% in patients vs. 0% in controls, p < 0.001) haplotypes among patients compared to controls. By contrast, the TGF-Beta1 TG haplotype was significantly lower in patients than controls (0% in patients vs. 52.5% in controls, p < 0.001). There were no significant differences in the frequency of alleles, genotypes and haplotypes of the IL-10 gene. Conclusions: We found a strong association between the polymorphisms of the TGF-Beta1 gene at codon 10 and codon 25 positions and atopic dermatitis (AU)

Introducción: Se ha sugerido que una degradación disminuida de histamina puede contribuir en la patogénesis de migraña y alergia. Este trabajo investiga una posible asociación entre 2 polimorfismos de un solo nucleótido (SNP) de 2 enzimas que degradan histamina, C314T para la histamina N-metil-transferasa (HNMT) y C2029G para diaminoxidasa (DAO), con la presencia, discapacidad y severidad de la migraña. Material y métodos: Se reclutó a 162 madres de niños alérgicos (80 con migraña y 82 sin migraña) determinando las variantes alélicas por PRC tiempo real usando un modelo de casos y controles. Mediante regresión logística se determinaron las OR para los genotipos y haplotipos. Resultados: El alelo mutado G para DAO fue significativamente más frecuente en el grupo de mujeres migrañosas que en los controles (OR = 1,6; IC del 95% = 1,1-2,1). No encontramos diferencias significativas para el alelo mutado T de la HNMT. Ambos alelos mutados estuvieron asociados a la discapacidad causada por la migraña. La coexistencia de ambas mutaciones (haplotipos) mostró una fuerte asociación con migraña. Los haplotipos que tenían ambos alelos mutados (ya sea como homocigotos o heterocigotos) estuvieron fuertemente asociados a la discapacidad por migraña grado iv (OR = 45,0, IC del 95% = 5,2-358). Esto sugiere que los alelos mutados T para HNMT y G para DAO pueden interactuar incrementando el riesgo y el impacto de la migraña. Conclusiones: Se sugiere una asociación sinérgica de polimorfismos de HNMT y DAO con migraña el cual debe ser confirmado en futuros estudios. La interpretación debe tomar en cuenta las características étnicas de la población estudiada (AU)


Background: Low histamine metabolism has been suggested to play a role in the pathogenesis of allergy and migraine. We investigated the possible association between 2 single-nucleotide polymorphisms (SNP), C314 T HNMT and C2029G DAO, and the presence and severity of migraine and migraine-related disability. Materials and methods: We studied the frequency of C314 T HNMT and C2029G DAO allelic variants in 162 mothers of children with allergies (80 with migraine and 82 without) using a TaqMan-based qPCR Assay and a case-control model. We conducted a logistic regression analysis to examine the association between migraine and the allelic and haplotype variants. Results: Mutant C2029G DAO SNP was found significantly more frequently in the group of women with migraine than in controls (OR, 1.6; 95% CI, 1.1–2.1). No significant differences were found in frequencies of genotypes or alleles in the case of C314T HNMT SNP. Both mutated alleles were associated with migraine-related disability. Coexistence of alleles for both SNPs (haplotypes) showed a strong association with migraine. Haplotypes containing both mutated alleles (either heterozygous or homozygous) were very strongly associated with MIDAS grade iv migraine (OR, 45.0; 95% CI, 5.2-358). This suggests that mutant alleles of C314 T for HNMT and C2029G for DAO polymorphisms may interact in a way that increases the risk and impact of migraine. Conclusions: We suggest a synergistic association between HNMT and DAO functional polymorphisms and migraine; this hypothesis must be further confirmed by larger studies. However, the characteristics and ethnic differences between analysed populations should be considered when interpreting the results (AU)

El carcinoma nasofaríngeo es el tumor más frecuente que surge en la nasofaringe. Su etología es multifactorial, considerándose como factores de riesgo la distribución racial y geográfica, la infección por el virus de Epstein-Barr (VEB), así como la exposición ambiental a determinas sustancias. Esta afección es endémica en algunas zonas asiáticas, donde se han encontrado predisposición genética en su oncogénesis. Existe una fuerte susceptibilidad entre el carcinoma de nasofaríngeo y el HLA, donde se han encontrado haplotipos relacionados específicos. En zonas donde la incidencia es baja, existen pocos casos publicados sobre familias afectadas. Reportamos 3 casos de familias con carcinoma de nasofaringe entre hermanos, en población no asiática, probablemente relacionada con la infección por el VEB (AU)


Nasopharyngeal carcinoma is the predominant tumour type arising in the nasopharynx. Its aetiology is multifactorial; racial and geographical distribution, EBV infection and environmental exposure to specific substances are considered risk factors. This condition is endemic in some Asian areas, where a genetic predisposition in its oncogenesis has been established. There is a strong susceptibility between nasopharyngeal carcinoma and HLA, where related specific haplotypes have been found. In areas where the incidence is low, there are few reported cases of families affected. We report 3 cases of families with nasopharyngeal carcinoma among siblings, in the non-Asian population, probably related to EBV infection (AU)

Introduction. Hereditary hemochromatosis is a disease responsible for excess blood iron. The hemochromatosis gene has two predominant variants, H63D and C282Y single nucleotide polymorphisms. Our study aims to analyze the diagnostic utility of genotyping the 63 and 282 loci, and examine the geographic distribution of these mutations in Spain. Methods and materials. Genotyping was performed on 94 healthy control individuals and 324 patients suspected of hereditary hemochromatosis, and also biochemical test to 313 individuals in the patients group. Results. The comparison of allelic frequencies between East and West of Spain, as well as other countries located at a similar longitude, evidenced a west-east distribution gradient of the C282Y allele. In addition, heterogeneous distribution of the H63D mutation in Spain was observed. Patients who carried the 282YY genotype showed significantly higher biochemical parameters (ferritin>300μg/L, Fe>180μg/L, IST>60%, UIBC>355μg/L and CTFH>370μg/dL), which confirmed the correlation between the mutated homozygous genotype and the associated hemochromatosis phenotype. Conclusion. Our results strengthen the importance of executing genetic tests to increase the efficiency of hereditary hemochromatosis diagnosis, which reveal an interesting variability among geographical regions (AU)


Introducción. La hemocromatosis hereditaria es una enfermedad responsable del exceso de hierro en sangre. El gen de la hemocromatosis tiene dos variantes predominantes, los polimorfismos de un solo nucleótido H63D y C282Y. Nuestro estudio trata de analizar la utilidad diagnóstica del genotipado de los loci 282 y 63, y examinar la distribución geográfica de estas mutaciones en España. Material y métodos. Se realizó genotipado en 94 controles sanos y 324 pacientes con sospecha de hemocromatosis hereditaria y además, test bioquímico a 313 individuos del grupo de pacientes. Resultados. La comparación de frecuencias alélicas entre poblaciones del este y el oeste de España, así como de otros países localizados en longitudes similares, evidenció una distribución en gradiente del alelo C282Y. Además se observó una distribución heterogénea de la mutación H63D en España. Los pacientes portadores del genotipo 282YY mostraron parámetros bioquímicos significativamente más elevados (ferritina >300μg/L, Fe > 180μg/L, IST > 60%, UIBC > 355μg/L y CTFH>370μg/dL), confirmando la correlación entre el genotipo homozigoto mutado y el fenotipo característico de hemocromatosis. Conclusión. Nuestros resultados refuerzan la importancia de realizar pruebas genéticas para confirmar el diagnóstico de la hemocromatosis hereditaria, teniendo en cuenta la variabilidad de los datos entre ámbitos geográficos (AU)

Introduction: Incidence of childhood-onset type 1 diabetes mellitus in the Canary Islands is the highest reported so far in Spain, and among the highest worldwide. The HLA region accounts for approximately half the genetic risk of type 1 diabetes. Our aim was to assess distribution of high-risk and protective HLA haplotypes in the Canarian families included in the T1DGC, as compared to the rest of Spain. Methods: The T1DGC study, an international project to study the genetics and pathogenesis of type 1 diabetes, enrolled more than 3000 families with type 1 diabetes worldwide. Spain provided 149 of these families, of whom 42 were from Tenerife and Gran Canaria. HLA was genotyped centrally using a PCR-based, sequence-specific oligonucleotide probe system. Haplotypes were reconstructed using the deterministic algorithm alleHap in the R programming environment. Based on prior T1DGC results in Caucasian population, haplotypes DRB1*0405-DQA1*0301-DQB1*0302, DRB1*0401-DQA1*0301-DQB1*0302, DRB1*0301-DQA1*0501-DQB1*0201, DRB1*0402-DQA1*0301-DQB1*0302 and DRB1*0404-DQA1*0301-DQB1*0302 were considered high-risk. DRB1*0701-DQA1*0201-DQB1*0303, DRB1*1401-DQA1*0101-DQB1*0503, DRB1*1501-DQA1*0102-DQB1*0602, DRB1*1101-DQA1*0501-DQB1*0301, DRB1*1104-DQA1*0501-DQB1*0301, DRB1*1303-DQA1*0501-DQB1*0301, DRB1*1301-DQA1*0103-DQB1*0603 and DRB1*0403-DQA1*0301-DQB1*0302 were considered protective. The distribution of protective, high-risk, and other haplotypes in the (first two) affected siblings and unaffected parents from Canarian and non-Canarian Spanish families was compared (Chi-square test). Results: No significant differences were found between the regions in distribution of the HLA haplotypes in the affected siblings or in the non-affected parents. Conclusions: The high incidence of childhood-onset type 1 diabetes in the Canarian population does not appear to be explained by a greater prevalence of high-risk class II HLA haplotypes in families with the disease. However, sample size limits the differences that can be detected in this study (AU)


Introducción: La incidencia de diabetes tipo 1 infantil en Canarias es la más alta descrita hasta el momento en España y una de las mayores a nivel mundial. La región HLA explica aproximadamente el 50% del riesgo genético de la diabetes tipo 1. Nuestro objetivo fue comparar la frecuencia de haplotipos de HLA de riesgo y protectores en familias españolas canarias y peninsulares incluidas en el T1DGC. Métodos: El T1DGC es un proyecto internacional que estudia la genética y patogenia de la diabetes tipo 1, para el que fueron inluidas más de 3000 familias con la enfermedad. Un total de 149 familias provenían de España, y 42 de ellas, de Tenerife y Gran Canaria. El HLA fue genotipado en un laboratorio central, utilizando un método basado en PCR y sondas específicas de secuencia. Los haplotipos fueron reconstruidos utilizando el algoritmo determinista alleHap en el entorno de programación R. En base a los resultados previos del T1DGC en población caucásica, los haplotipos DRB1*0405-DQA1*0301-DQB1*0302, DRB1*0401-DQA1*0301-DQB1*0302, DRB1*0301-DQA1*0501-DQB1*0201, DRB1*0402-DQA1*0301-DQB1*0302 y DRB1*0404-DQA1*0301-DQB1*0302 fueron definidos como de alto riesgo. DRB1*0701-DQA1*0201-DQB1*0303, DRB1*1401-DQA1*0101-DQB1*0503, DRB1*1501-DQA1*0102-DQB1*0602, DRB1*1101-DQA1*0501-DQB1*0301, DRB1*1104-DQA1*0501-DQB1*0301, DRB1*1303-DQA1*0501-DQB1*0301, DRB1*1301-DQA1*0103-DQB1*0603 y DRB1*0403-DQA1*0301-DQB1*0302 fueron considerados protectores. La distribución de haplotipos de riesgo, protectores y otros en los (dos primeros) hermanos afectos y en los padres no afectos fue comparada entre las familias canarias y no canarias (chi cuadrado). Resultados: No se encontraron diferencias significativas en la distribución de haplotipos HLA entre las regiones estudiadas, ni en los hermanos afectos ni en los padres no afectos. Conclusiones: La alta incidencia de la enfermedad en la población canaria no parece ser explicada por una mayor prevalencia de haplotipos de HLA de clase II de riesgo en los casos con agregación familiar, aunque el tamaño de la muestra limita las diferencias detectables en este estudio (AU)
