Discovered in 1865 by Jules Bernard Luys, the subthalamic nucleus is a set of small nuclei located in the diencephalon, inferior to the thalamus and superior to the substantia nigra, that can be visualized in a posterior coronal section. Histologically, it consists of neurons compactly distributed and filled with a large number of blood vessels and sparse myelinated fibers. This review presents an analysis of this anatomical region, considering what is most recent in the literature. Subthalamic neurons are excitatory and use glutamate as the neurotransmitter. In healthy individuals, these neurons are inhibited by nerve cells located in the side globus pallidus. However, if the fibers that make up the afferent circuit are damaged, the neurons become highly excitable, thus causing motor disturbances that can be classified as hyperkinetic, for example ballism and chorea, or hypokinetic, for example Parkinson disease (PD). The advent of deep brain stimulation has given the subthalamic nucleus great visibility. Studies reveal that the stimulation of this nucleus improves themotor symptoms of PD.

Objective: Sleep apnea has been associated with anxiety, but the mechanisms of the sleep apnea-anxiety relationship are unresolved. Sleep apnea causes oxidative stress, which might enhance anxiety-like behavior in rodents. To clarify the apnea-anxiety connection, we tested the effect of intermittent hypoxia, a model of sleep apnea, on the anxiety behavior of mice. Methods: The rodents were exposed daily to 480 one-minute cycles of intermittent hypoxia to a nadir of 7±1% inspiratory oxygen fraction or to a sham procedure with room air. After 7 days, the mice from both groups were placed in an elevated plus maze and were video recorded for 10 min to allow analysis of latency, frequency, and duration in open and closed arms. Glyoxalase-1 (Glo1) and glutathione reductase-1 (GR1) were measured in the cerebral cortex, hippocampus, and striatum by Western blotting. Results: Compared to controls, the intermittent hypoxia group displayed less anxiety-like behavior, perceived by a statistically significant increase in the number of entries and total time spent in open arms. A higher expression of GR1 in the cortex was also observed. Conclusion: The lack of a clear anxiety response as an outcome of intermittent hypoxia exposure suggests the existence of additional layers in the anxiety mechanism in sleep apnea, possibly represented by sleepiness and irreversible neuronal damage.

Em diversos estudos, as mulheres são definidas como mais propensas a atitudes pró-sociais, em comparação aos homens. Especula-se que essas diferenças de comportamento se devem às influências sociais. Entretanto, pesquisadores da Universidade de Zurich creditam essa diferença a questões de ordem biológica. O presente artigo consiste em síntese e comentário desse estudo, intitulado "The dopaminergic reward system underpins gender differences in social preferences", o qual verificou que o sistema de recompensa dopaminérgico das mulheres reage mais fortemente a comportamentos pró-sociais e o dos homens a comportamentos não sociais. Estudos anteriores já haviam demonstrado diferenças nas preferências entre os gêneros de bebês recém-nascidos e a influência no nível de testosterona fetal na tendência de crianças para sistematizar (analisar e construir sistemas) ou "empatizar" (perceber informações não verbais no comportamento). Diferenças de gênero entre habilidades e tendências comportamentais podem ter uma base biológica subjacente para além de influências sociais. Esta base incluiria a atuação hormonal no período gestacional e uma consequente diferenciação de funcionamento das estruturas cerebrais, em especial do sistema de recompensa e do sistema límbico.

A acne é uma doença bem comum, que vem a causar transtornos estéticos e psicológicos nas pessoas acometidas por esta patologia, que atinge principalmente o público jovem, deixando cicatrizes como sequelas. Visando uma atenuação dessas cicatrizes, muitos métodos propõem uma associação de procedimentos, sendo bastante eficazes, porem uma das fontes mais utilizadas atualmente na estética é a fototerapia, que vem sendo aplicada em vários tratamentos como: estrias, manchas celulite, acne, alopecia, rejuvenescimento, cicatrização da pele entre outros, e que nada mais é que a interação da luz com a pele com o âmbito de potencializar e acelerar o processo de reparação da epiderme. O advento Lasers e LEDs mensuram entre diferentes comprimentos de onda ou cores, permitindo realizar de forma controlada com efetividade e segurança do tratamento almejado e associando-se à técnica de microagulhamento também promissora nesses casos, promovendo micro lesões no tecido através de um "roller", estimulando a produção de colágeno, que é fundamental no processo de cicatrização, sendo que a aplicação dessa técnica é dividida em sessões, que variam de acordo com a necessidade de cada indivíduo. Dessa forma, o objetivo foi constatar a eficácia da aplicação da fototerapia e a técnica de microagulhamento, aperfeiçoando os resultados de cicatriz de acne, esperando potencializar os efeitos, tendo uma melhora nítida no quadro da cicatriz de acne. Acne is a very common disease, which causes aesthetic and psychological disorders in people affected by this pathology, which mainly affects the young public, leaving scars as sequels. Aiming at attenuating these scars, many methods propose an association of procedures, being quite effective, but one of the most commonly used sources in esthetics today is phototherapy, which has been applied in several treatments such as stretch marks, cellulite spots, acne, alopecia, rejuvenation , skin healing among others, and that is nothing more than the interaction of light with skin with the scope to potentiate and accelerate the process of repairing the epidermis, the advent Lasers and LEDs measure between different wavelengths or colors, allowing to perform in a controlled manner with the effectiveness and safety of the targeted treatment and associated with the microaggregation technique also promising in these cases, promoting micro-lesions in the tissue through a roller, stimulating the production of collagen, which is fundamental in the cicatrization process, being that the application of this technique is divided into sessions, which vary according to the of each individual. Thus, the objective was to verify the effectiveness of the application of phototherapy and the technique of microneedle, improving the results of acne scar, hoping to potentiate the effects, having a clear improvement in the framework of the acne scar. ABSTRACT Studies have shown that schizophrenic patients seem to have nutritional deficiencies. Ascorbic acid (AA) has an important antioxidant effect and neuromodulatory properties. The aim of this study was to evaluate the effects of AA on locomotor activity and the acetylcholinesterase activity (AChE) in an animal model of schizophrenia (SZ). Rats were supplemented with AA (0.1, 1, or 10 mg/kg), or water for 14 days (gavage). Between the 9th and 15th days, the animals received Ketamine (Ket) (25 mg/kg) or saline (i.p). After the last administration (30 min) rats were subjected to the behavioral test. Brain structures were dissected for biochemical analysis. There was a significant increase in the locomotor activity in Ket treated. AA prevented the hyperlocomotion induced by ket. Ket also showed an increase of AChE activity within the prefrontal cortex and striatum prevented by AA. Our data indicates an effect for AA in preventing alterations induced by Ket in an animal model of SZ, suggesting that it may be an adjuvant approach for the development of new therapeutic strategies within this psychiatric disorder.

BACKGROUND: Chitosan, the N-deacetylated derivative of chitin, is a cationic polyelectrolyte due to the presence of amino groups, one of the few occurring in nature. The use of chitosan in protein and drug delivery systems is being actively researched and reported in the literature RESULTS: In this study, we used chitosan-coated levodopa liposomes to investigate the behavioral character and the expression of phosphorylated extracellular signal-regulated kinase (ERK1/2), dopamine- and cAMP-regulated phos-phoprotein of 32 kDa (DARPP-32) and FosB/AFosB in striatum of rat model of levodopa-induced dyskinesia (LID). We found that scores of abnormal involuntary movement (AIM) decreased significantly in liposome group (P < 0.05), compared with levodopa group. Levels of phospho-ERK1/2, phospho-Thr34 DARPP-32 and FosB/AFosB in striatum decreased significantly in liposome group lesion side compared with levodopa group (P < 0.05). However, both of two groups above have significantly differences compared with the control group (P < 0.05). CONCLUSION: Chitosan-coated levodopa liposomes may be useful in reducing dyskinesias inducing for Parkinson disease. The mechanism might be involved the pathway of signaling molecular phospho-ERK1/2, phospho-Thr34 DARPP-32 and AFosB in striatum

Optical coherence tomography (OCT) is a promising medical imaging technique that uses light to capture real-time cross-sectional images from biological tissues in micrometer resolution. Commercially available optical coherence tomography systems are employed in diverse applications, including art conservation and diagnostic medicine, notably in cardiology and ophthalmology. Application of this technology in the brain may enable distinction between white matter and gray matter, and obtainment of detailed images from within the encephalon. We present, herein, the in vivo implementation of OCT imaging in the rat brain striatum. For this, two male 60-day-old rats (Rattus norvegicus, Albinus variation, Wistar) were stereotactically implanted with guide cannulas into the striatum to guide a 2.7-French diameter high-definition OCT imaging catheter (Dragonfly™, St. Jude Medical, USA). Obtained images were compared with corresponding histologically stained sections to collect imaging samples. A brief analysis of OCT technology and its current applications is also reported, as well as intra-cerebral OCT feasibility on brain mapping during neurosurgical procedures.

El trauma ocular pediátrico es frecuente y es la principal causa de ceguera unilateral no congénita. La información en países en vías de desarrollo es escasa. El objetivo de esta serie de casos es describir las características clínicas y epidemiológicas del trauma ocular en niños menores de 14 años que consultaron al Hospital Dr. Rodolfo Robles Val verde en la Ciudad de Guatemala durante el año 2010. Se incluyeron 119 pacientes en el estudio. El género masculino en edad escolar (7-9 años) fue el más comprometido. El trauma más común fue el de globo cerrado. Los objetos más frecuentes causantes de la lesión fueron madera, juguetes y químicos. La vivienda fue el lugar donde más ocurrió el trauma. Se intervinieron 21 pacientes. Son necesarios programas de educación y prevención.


Pediatric ocular trauma is common and the leading cause of non congenital unilateral blindness. The information in developing countries is scarce. The objective of this case series is to describe clinical and epidemiological characteristics of ocular trauma in children under 14 years of age who visited Hospital Dr. Rodolfo Robles Valverde in Guatemala City in 2010. In this study 119 patients were included. School-aged (7-9 years) male gender was the most affected. Closed globe injury was the commonest. The most frequent objects causing the lesions were: wooden objects, toys and chemicals. Trauma occurred most frequently at home. Twenty one of the patients were surgically intervened. Education and prevention programs for pediatric ocular trauma are necessary.

Stroke is the third most common cause of death worldwide, and most stroke survivors present some functional impairment. We assessed the striatal oxidative balance and motor alterations resulting from stroke in a rat model to investigate the neuroprotective role of physical exercise. Forty male Wistar rats were assigned to 4 groups: a) control, b) ischemia, c) physical exercise, and d) physical exercise and ischemia. Physical exercise was conducted using a treadmill for 8 weeks. Ischemia-reperfusion surgery involved transient bilateral occlusion of the common carotid arteries for 30 min. Neuromotor performance (open-field and rotarod performance tests) and pain sensitivity were evaluated beginning at 24 h after the surgery. Rats were euthanized and the corpora striata was removed for assay of reactive oxygen species, lipoperoxidation activity, and antioxidant markers. Ischemia-reperfusion caused changes in motor activity. The ischemia-induced alterations observed in the open-field test were fully reversed, and those observed in the rotarod test were partially reversed, by physical exercise. Pain sensitivity was similar among all groups. Levels of reactive oxygen species and lipoperoxidation increased after ischemia; physical exercise decreased reactive oxygen species levels. None of the treatments altered the levels of antioxidant markers. In summary, ischemia-reperfusion resulted in motor impairment and altered striatal oxidative balance in this animal model, but those changes were moderated by physical exercise.

Nitric oxide (NO) is a major neurotransmitter associated with motor control in basal ganglia. Movement disorders, as essential tremor and Parkinson’s disease, are more prevalent on aged individuals. We investigated the effects of aging on neuronal density and diameter/area of nitrergic neurons in samples of striatum (caudate and putamen) and subthalamic nucleus of 20 human brains from normal subjects, stained by histochemistry for NADPH-diaphorase and immunohistochemistry for neuronal NO synthase. Our data showed aging does not modify the neuronal density and size of nitrergic neurons in striatum and subthalamic nucleus. These findings suggest a lack of association between aging and morphologic changes on nitrergic neurons.


O óxido nítrico (NO) é um importante neurotransmissor associado ao controle motor nos núcleos da base. Os distúrbios de movimento, como tremor essencial e a doença de Parkinson, são mais prevalentes em indivíduos idosos. Nós investigamos os efeitos do envelhecimento sobre a densidade neuronal e diâmetro/área dos neurônios nitrérgicos em amostras de estriado (caudado e putâmen) e núcleo subtalâmico de 20 encéfalos humanos de indivíduos normais, corados pela técnica histoquímica da NADPH-diaforase e imunohistoquímica para a sintase do NO neuronal. Nossos resultados mostraram que o envelhecimento não modifica a densidade neuronal e as dimensões dos neurônios nitrérgicos no estriado e núcleo subtalâmico. Estes achados sugerem uma falta de associação entre envelhecimento e mudanças morfológicas nos neurônios nitrérgicos.

Introduction: Cerebral ischemia is the third leading cause of death and the primary cause of permanent disability worldwide. Atorvastatin is a promising drug with neuroprotective effects that may be useful for the treatment of stroke. However, the effects of atorvastatin on specific neuronal populations within the nigrostriatal system following cerebral ischemia are unknown. Objective: To evaluate the effects of atorvastatin on dopaminergic and GABAergic neuronal populations in exofocal brain regions in a model of transient occlusion of the middle cerebral artery. Materials and methods: Twenty-eight male eight-week-old Wistar rats were used in this study. Both sham and ischemic rats were treated with atorvastatin (10 mg/kg) or carboxymethylcellulose (placebo) by gavage at 6, 24, 48 and 72 hours post-reperfusion. We analyzed the immunoreactivity of glutamic acid decarboxylase and tyrosine hydroxylase in the globus pallidus, caudate putamen and substantia nigra. Results: We observed neurological damage and cell loss in the caudate putamen following ischemia. We also found an increase in tyrosine hydroxylase immunoreactivity in the medial globus pallidus and substantia nigra reticulata, as well as a decrease in glutamic acid decarboxylase immunoreactivity in the lateral globus pallidus in ischemic animals treated with a placebo. However, atorvastatin treatment was able to reverse these effects, significantly decreasing tyrosine hydroxylase levels in the medial globus pallidus and substantia nigra reticulata and significantly increasing glutamic acid decarboxylase levels in the lateral globus pallidus. Conclusion: Our data suggest that post-ischemia treatment with atorvastatin can have neuro-protective effects in exofocal regions far from the ischemic core by modulating the GABAergic and dopaminergic neuronal populations in the nigrostriatal system, which could be useful for preventing neurological disorders.


Introducción. La isquemia cerebral es la tercera causa de muerte y la primera de discapacidad permanente en el mundo. La atorvastatina es un fármaco neuroprotector prometedor para el tratamiento de la apoplejía; sin embargo, su acción sobre las poblaciones neuronales del sistema nigroestriatal después de la isquemia aún se desconoce. Objetivo. Evaluar el efecto de la atorvastatina sobre poblaciones gabérgicas y dopaminérgicas en regiones exofocales en un modelo de oclusión transitoria de la arteria cerebral media. Materiales y métodos. Se utilizaron 28 ratas Wistar macho de ocho semanas de edad. Los ejemplares con isquemia simulada y los ejemplares sometidos a isquemia fueron tratados con atorvastatina (10 mg/kg) y carboximetilcelulosa (placebo) administrados por medio de sonda a las 6, 24, 48 y 72 horas después de la reperfusión. Se analizó la inmunorreacción de la descarboxilasa del ácido glutámico y de la tirosina hidroxilasa en el globo pálido, el putamen caudado y la sustancia negra. Resultados. Los datos confirmaron el daño neurológico y la pérdida celular en el putamen caudado. Se incrementó la inmunorreacción de la tirosina hidroxilasa en el globo pálido medial y la sustancia negra pars reticulata , disminuyendo la inmunorreacción de la descarboxilasa del ácido glutámico en el globo pálido lateral de los animales isquémicos tratados con placebo; sin embargo, el tratamiento con atorvastatina pudo revertirla, lo que logró una disminución significativa de la tirosina hidroxilasa en el globo pálido medial y la sustancia negra pars reticulata y aumentando los niveles de descarboxilasa del ácido glutámico en el globo pálido lateral. Conclusión. Nuestros datos sugieren que la atorvastatina en el tratamiento posterior a la isquemia ejerce neuroprotección en las zonas exofocales, modulando las poblaciones neuronales gabérgicas y dopaminérgicas del sistema nigroestriatal, lo que podría prevenir trastornos neurológicos.

Objective: To evaluate behavioral changes and brain-derived neurotrophic factor (BDNF) levels in rats subjected to ketamine administration (25 mg/kg) for 7 days. Method: Behavioral evaluation was undertaken at 1 and 6 hours after the last injection. Results: We observed hyperlocomotion 1 hour after the last injection and a decrease in locomotion after 6 hours. Immobility time was decreased and climbing time was increased 6 hours after the last injection. BDNF levels were decreased in the prefrontal cortex and amygdala when rats were killed 6 hours after the last injection, compared to the saline group and to rats killed 1 hour after the last injection. BDNF levels in the striatum were decreased in rats killed 6 hours after the last ketamine injection, and BDNF levels in the hippocampus were decreased in the groups that were killed 1 and 6 hours after the last injection. Conclusion: These results suggest that the effects of ketamine on behavior and BDNF levels are related to the time at which they were evaluated after administration of the drug. .

The striatum, the largest component of the basal ganglia, is usually subdivided into associative, motor and limbic components. However, the electrophysiological interactions between these three subsystems during behavior remain largely unknown. We hypothesized that the striatum might be particularly active during exploratory behavior, which is presumably associated with increased attention. We investigated the modulation of local field potentials (LFPs) in the striatum during attentive wakefulness in freely moving rats. To this end, we implanted microelectrodes into different parts of the striatum of Wistar rats, as well as into the motor, associative and limbic cortices. We then used electromyograms to identify motor activity and analyzed the instantaneous frequency, power spectra and partial directed coherence during exploratory behavior. We observed fine modulation in the theta frequency range of striatal LFPs in 92.5 ± 2.5% of all epochs of exploratory behavior. Concomitantly, the theta power spectrum increased in all striatal channels (P < 0.001), and coherence analysis revealed strong connectivity (coefficients >0.7) between the primary motor cortex and the rostral part of the caudatoputamen nucleus, as well as among all striatal channels (P < 0.001). Conclusively, we observed a pattern of strong theta band activation in the entire striatum during attentive wakefulness, as well as a strong coherence between the motor cortex and the entire striatum. We suggest that this activation reflects the integration of motor, cognitive and limbic systems during attentive wakefulness.

Anxiolytic and anxiogenic-like behavioral outcomes have been reported for methylenedioxymethamphetamine (MDMA or ecstasy) in rodents. In the present experiment, we attempted to identify behavioral, hormonal and neurochemical outcomes of MDMA treatment to clarify its effects on anxiety-related responses in 2-month-old Balb/c male mice (25-35 g; N = 7-10 mice/group). The behavioral tests used were open field, elevated plus maze, hole board, and defensive behavior against predator odor. Moreover, we also determined striatal dopamine and dopamine turnover, and serum corticosterone levels. MDMA was injected ip at 0.2, 1.0, 5.0, 8.0, 10, or 20 mg/kg. MDMA at 10 mg/kg induced the following significant (P < 0.05) effects: a) a dose-dependent increase in the distance traveled and in the time spent moving in the open field; b) decreased exploratory activity in the hole board as measured by number of head dips and time spent in head dipping; c) increased number of open arm entries and increased time spent in open arm exploration in the elevated plus maze; d) increased time spent away from an aversive stimulus and decreased number of risk assessments in an aversive odor chamber; e) increased serum corticosterone levels, and f) increased striatal dopamine level and turnover. Taken together, these data suggest an anxiogenic-like effect of acute MDMA treatment, despite the fact that behavioral anxiety expression was impaired in some of the behavioral tests used as a consequence of the motor stimulating effects of MDMA.

The seizures can produce neuronal damage in several brain structures. The aim of this study was to investigate the potential neuroprotective effect of essential oil of Citrus limon (EOCL) on the histopathological changes observed in the hippocampus and striatum of mice after seizures induced by pilocarpine. Adult Swiss mice were 2 months old. The animals were divided into four groups. The first group was treated with 0.05 percent Tween 80 (control group) and the second with pilocarpine (400 mg/kg group P400). The third and fourth group were treated with EOCL (150 mg/kg) and 30 min after received P400 (P400 +/- EOCL group) or 0.05 percent Tween 80, respectively. After treatment, all groups were observed for 24 h, then sacrificed and their brains removed for histopathological analysis. The group P400, presented with seizures that progressed to status epilepticus in 75 percent of animals. Pretreatment with OECL produced a 25 percent reduction in this index. Groups P400 and P400 + EOCL showed 83.33 percent and 25 percent of animals with brain damage in the hippocampus, respectively. In the striatum of group P400 was a compromise of 75 percent. In turn, in the striatal region of group EOCL P400 + was seen a decrease of 58.34 percent in this neuronal damage. The seizures induced by pilocarpine are installed by the cholinergic system and produce brain damage. According to our results we suggest that the EOCL may modulate epileptogenesis and promote neuroprotective effects during the seizures in the model investigated.


As convulsões podem produzir danos neuronais em diversas estruturas cerebrais. O objetivo desse estudo foi investigar o potencial efeito neuroprotetor do óleo essencial de Citrus limon (OECL) nas alterações histopatológicas observadas no hipocampo e corpo estriado de camundongos após convulsão induzida por pilocarpina. Foram utilizados camundongos Swiss adultos com 2 meses de idade. Os animais foram divididos em 4 grupos. O primeiro grupo foi tratado com Tween 80 0,05 por cento (grupo controle) e o segundo com pilocarpina (400 mg/kg, grupo P400). Já o terceiro e quarto grupo foram tratados com OECL (150 mg/kg), e 30 min depois receberam P400 (grupo OECL + P400) ou Tween 80 0,05 por cento 0.9 por cento (grupo OECL), respectivamente. Após os tratamentos, todos os grupos foram observados durante 24 h e em seguida sacrificados e seus cérebros removidos para as análises histopatológicas. O grupo P400, apresentou convulsões que progrediram para o estado epiléptico em 75 por cento dos animais. O pré-tratamento com OECL produziu uma redução de 25 por cento nesse índice. Os grupos P400 e OECL + P400 apresentaram 83,33 por cento e 25 por cento de animais com lesão cerebral no hipocampo, respectivamente. No corpo estriado dos animais do grupo P400 houve um comprometimento de 75 por cento. Por sua vez, na região estriatal dos animais do grupo OECL + P400 foi visto uma redução de 58,34 por cento nesse comprometimento. As convulsões induzidas pela pilocarpina são instaladas pelo sistema colinérgico e produzem dano cerebral. De acordo com nossos resultados podemos sugerir que o OECL pode modular a epileptogênese e promover ação neuroprotetora durante as convulsões no modelo investigado.

7-Nitroindazole (7-NI) inhibits neuronal nitric oxide synthase in vivo and reduces l-DOPA-induced dyskinesias in a rat model of parkinsonism. The aim of the present study was to determine if the anti-dyskinetic effect of 7-NI was subject to tolerance after repeated treatment and if this drug could interfere with the priming effect of l-DOPA. Adult male Wistar rats (200-250 g) with unilateral depletion of dopamine in the substantia nigra compacta were treated with l-DOPA (30 mg/kg) for 34 days. On the 1st day, 6 rats received ip saline and 6 received ip 7-NI (30 mg/kg) before l-DOPA. From the 2nd to the 26th day, all rats received l-DOPA daily and, from the 27th to the 34th day, they also received 7-NI before l-DOPA. Animals were evaluated before the drug and 1 h after l-DOPA using an abnormal involuntary movement scale and a stepping test. All rats had a similar initial motor deficit. 7-NI decreased abnormal involuntary movement induced by l-DOPA and the effect was maintained during the experiment before 7-NI, median (interquartile interval), day 26: 16.75 (15.88-17.00); day 28: 0.00 (0.00-9.63); day 29: 13.75 (2.25-15.50); day 30: 0.5 (0.00-6.25); day 31: 4.00 (0.00-7.13), and day 34: 0.5 (0.00-14.63), Friedman followed by Wilcoxon test,vs day 26, P < 0.05;. The response to l-DOPA alone was not modified by the use of 7-NI before the first administration of the drug (l-DOPA vs time interaction, F1,10 = 1.5, NS). The data suggest that tolerance to the anti-dyskinetic effects of a neuronal nitric oxide synthase inhibitor does not develop over a short-term period of repeated administration. These observations open a possible new therapeutic approach to motor complications of chronic l-DOPA therapy in patients with Parkinson’s disease.

CONTEXTO: Tem sido sugerido que durante as crises convulsivas induzidas pela pilocarpina pode ser observado aumento no estresse oxidativo cerebral. Estudos sugerem que compostos com atividade antioxidante podem fornecer certo grau de proteção contra a neurotoxicidade induzida pelas crises convulsivas. OBJETIVOS: O presente estudo investigou as ações farmacológicas da vigabatrina nas alterações comportamentais e na atividade enzimática da superóxido dismutase (SOD) no corpo estriado de ratos adultos. MÉTODOS: Ratos Wistar adultos (2 meses de idade) foram usados nos experimentos e divididos em quatro grupos. O primeiro foi tratado com solução salina 0,9 por cento (grupo controle). O segundo grupo foi tratado com pilocarpina (400 mg/kg, i.p., grupo P400). O terceiro grupo foi tratado com vigabatrina (500 mg/kg, i.p., grupo VGB) e o quarto grupo foi tratado com vigabatrina (500 mg/kg, i.p.) e 30 minutos depois com pilocarpina (400 mg/kg, i.p., grupo VGB + P400). Os animais que apresentaram crises convulsivas, estado de mal epiléptico e não morreram durante o período de 24 horas de observação foram sacrificados para dissecação do corpo estriado para realização da determinação da atividade da SOD. RESULTADOS: Os estudos comportamentais revelaram que, após administração de pilocarpina, todos os animais apresentaram sinais colinérgicos periféricos, movimentos estereotipados e tremores. No mesmo grupo, foram observados, em 75 por cento dos animais, crises convulsivas e o estado de mal epiléptico. Por sua vez, o pré-tratamento com vigabatrina produziu redução significativa de 50 por cento nas crises convulsivas. Com relação aos estudos neuroquímicos, não foram observadas alterações na atividade da SOD no corpo estriado do grupo P400, em comparação aos valores do grupo controle. No entanto, no grupo VGB + P400 foi visto aumento significativo na atividade da SOD de 34 por cento e 35 por cento, quando comparado aos grupos controle e P400, respectivamente. ...


BACKGROUND: Pilocarpine-induced seizures have been suggested to be mediated by increases in oxidative stress. Current studies have suggested that antioxidant compounds may afford some level of neuroprotection against the neurotoxicity of seizures. OBJECTIVES: This study investigated the pharmacological actions of vigabatrin on behavioral changes and superoxide dismutase (SOD) activity in striatum of adult rats. METHODS: Adult rats (2 months old) were used in the experiments and divided into four groups. The first was treated with 0.9 percent saline (control group). The second group was treated with pilocarpine (400 mg/kg, i.p., P400 group). The third group received vigabatrin alone (500 mg/kg, i.p., VGB group) and the fourth group was treated with vigabatrin (500 mg/kg, i.p.) and 30 minutes later received pilocarpine (400 mg/kg, i.p., VGB + P400 group). The animals which had seizures and status epilepticus (SE) and did not die within 24 hours of observation were sacrificed to perform the neurochemical studies. RESULTS: Behavioral studies showed that the administration of pilocarpine produces peripheral cholinergic signs, tremors and stereotyped movements in all animals. An amount of 75 percent of those rats developed to seizures and SE. In turn, the pre-treatment with vigabatrin produced a 50 percent reduction in the rate of seizures and SE. Regarding the neurochemical studies, there were no changes in the striatal SOD activity in P400 group as compared to the control group. However, in the VGB + P400 group it was verified significant increases in SOD activity of 34 percent and 35 percent as compared to control and P400 group, respectively. DISCUSSION: Our results indicate that behavioral changes occur during seizures, but SOD activity remained unaltered during the acute phase of the convulsive crisis. CONTEXTO: As convulsões podem produzir danos neuronais em diversas áreas e, especialmente, nas estruturas límbicas. OBJETIVOS: O objetivo do presente trabalho foi estudar os efeitos neuroprotetores da vitamina C nas alterações histopatológicas observadas no corpo estriado de ratos convulsivos. MATERIAL E MÉTODOS: Foram utilizados ratos Wistar adultos. Os animais foram divididos em quatro grupos. O primeiro grupo foi tratado com salina 0,9 por cento (grupo controle) e o segundo, com pilocarpina (400 mg/kg, grupo P400). Já o terceiro e o quarto grupo foram tratados com vitamina C (250 mg/kg), e, 30 minutos depois, receberam P400 (grupo VIT C + P400) ou solução salina 0,9 por cento (grupo VIT C), respectivamente. Após os tratamentos, todos os grupos foram observados por 24 horas e, em seguida, sacrificados e seus cérebros removidos para as análises histopatológicas. RESULTADOS: O grupo P400 apresentou convulsões que progrediram para o estado epiléptico em 75 por cento dos animais. O pré-tratamento com vitamina C produziu uma redução de 35 por cento nesse índice. Os grupos P400 e VIT C + P400 apresentaram 80 por cento e 20 por cento de animais com lesão cerebral, respectivamente. No corpo estriado dos animais do grupo P400, houve um comprometimento de 50 por cento. Por sua vez, na região estriatal dos animais do grupo VIT C + P400 foi vista uma redução de 40 por cento nesse comprometimento. DISCUSSÃO: As convulsões induzidas pela pilocarpina são instaladas pelo sistema colinérgico e propagadas pela produção de radicais livres e pelo sistema glutamatérgico, resultado no desenvolvimento de dano cerebral. As drogas antioxidantes podem apresentar um potencial terapêutico para pacientes epilépticos na proteção contra as lesões cerebrais por meio da remoção desses radicais livres formados. Acredita-se, assim, que a vitamina C pode influenciar a epileptogênese e promover ações neuroprotetoras durante as convulsões.

P400 and P400 + VIT C groups revealed 80 percent and 20 percent of animals with brain injury, respectively. In P400 group, lesion severity of the striatum was 50 percent. In turn, in striatal region of animals treated with P400 + VIT C group, we detected a reduction of 40 percent in the severity degree. DISCUSSION: Pilocarpine-induced seizures are installed by the cholinergic system and propagated by free radicals and by glutamatergic system, leading to brain damage. The antioxidant drugs may have therapeutic potential for epileptic patients to protect against brain injure through removing free radicals produced, suggesting that vitamin C may influence epileptogenesis and promote neuroprotective actions during seizures.

Female rats are intensely affected by cocaine, with estrogen probably playing an important role in this effect. Progesterone modulates the GABA system and attenuates the effects of cocaine; however, there is no information about its relevance in changing GABA synthesis pathways after cocaine administration to female rats. Our objective was to investigate the influence of progesterone on the effects of repeated cocaine administration on the isoenzymes of glutamic acid decarboxylase (GAD65 and GAD67) mRNA in brain areas involved in the addiction circuitry. Ovariectomized, intact and progesterone replacement-treated female rats received saline or cocaine (30 mg/kg, ip) acutely or repeatedly. GAD isoenzyme mRNA levels were determined in the dorsolateral striatum (dSTR) and prefrontal cortex (PFC) by RT-PCR, showing that repeated, but not acute, cocaine decreased GADs/β-actin mRNA ratio in the dSTR irrespective of the hormonal condition (GAD65: P < 0.001; and GAD67: P = 0.004). In the PFC, repeated cocaine decreased GAD65 and increased GAD67 mRNA ratio (P < 0.05). Progesterone replacement decreased both GAD isoenzymes mRNA ratio after acute cocaine in the PFC (P < 0.001) and repeated cocaine treatment reversed this decrease (P < 0.001). These results suggest that cocaine does not immediately affect GAD mRNA expression, while repeated cocaine decreases both GAD65 and GAD67 mRNA in the dSTR of female rats, independently of their hormonal conditions. In the PFC, repeated cocaine increases the expression of GAD isoenzymes, which were decreased due to progesterone replacement.

As estrias que são uma atrofia de pele adquirida devido ao rompimento de fibras elásticas e colágenas. Um dos métodos de tratamento para as estrias é a galvanopuntura. Seus efeitos estão relacionados ao estímulo elétrico, baseado na compilação dos efeitos intrínsecos da corrente contínua (galvânica) que promovem uma inflamação aguda localizada, extremamente eficiente na regeneração do tecido conjuntivo rompido. Este estudo utilizou a corrente galvânica associada a uma máquina de tatuar (protótipo). Foram aplicadas sete sessões, em uma paciente, uma vez por semana, tratando três estrias, uma com galvanopuntura tradicional e as outras duas com a máquina de tatuar associada ou não a corrente. A avaliação foi realizada por meio de ficha de avaliação e biópsias antes e após as sete sessões. O objetivo foi verificar os efeitos da aplicação da corrente galvânica associada a um aparelho de tatuagem na obtenção de uma resposta inflamatória. Concluiu-se que a maior eficácia desse tratamento foi utilizando a corrente galvânica, porém, associada com a máquina de tatuar, os resultados foram superiores, obtendo maior tempo inflamatório, menor tempo para efetuar a aplicação e, sobretudo, promovendo menor dor e desconforto à paciente. Acreditamos que este protótipo seja útil na prática clínica da fisioterapia dermato-funcional.


The striae are skin atrophy due to the disruption of elastic and collagen fibers. One treatment method for the striae is the galvanopuncture. Its effect is related to electrical stimulation, that promote an acute located inflammation, without any systemic effect, but extremely efficient in the regeneration of broken tissue. This study used the galvanic current associated with a tattoo machine (prototype). It is a study with one patient, applying seven sessions once a week, treating 3 striae with a traditional galvanopuncture and other 2 striae with the tattoo machine associated or not to electric current. The evaluation was conducted through an evaluation form and biopsies before and after the seven sessions. The objective was to verify the effects of the application of galvanic current equipment associated with tattoo machine on achieving an inflammatory response. It was concluded that such treatment was more effective using the galvanic current, but with the tattoo machine associated, the results were better, gaining more inflammatory time, less time to the application and, above all, promoting minor pain and discomfort to the patient. We concluded that this prototype could be useful in clinical practice of physical therapy dermato-functional.
