RESUMO

Introduction: The exploitation of the sea cucumber (Holothuria (Panningothuria) forskali) in Ría de Vigo (NW Spain) is recent (2015) and it has been done until now with scarce or no information about some key biological aspects as reproduction, recruitment or growth. Objective: To describe the reproductive cycle of H. forskali in Ría de Vigo. Methods: We sampled fortnightly throughout 2018. We calculated gonadal condition indices (GCI) and gametogenic stages by classic histological methods. Results: The reproductive cycle of H. forskali in the Ría de Vigo is characterized by a sexual resting stage during spring, when temperature and daylight hours are lower; the beginning of gametogenesis during summer, when temperature is higher, daylight hours longer and the sea bottom is rich in nutrients; then, a period of spawns interspersed with a gonadal restoration during autumn and winter, when temperature is lower and food is scarce. Sex ratio is 1:1, however, the studied population is not synchronized, because females initiate maturation earlier. The comparison of the histological results with GCI indices suggest that GCI is a good indicator for gonadal stage. Fishery management strategies, such as a closure period, must be adapted to the reproductive stage. We recommend avoiding fishing between November and February to increase spawning potential ratio and, consequently, recruitment.

Abstract The population and reproductive biology of the commercial sea cucumber Holothuria mexicana (Ludwig, 1875) was studied in Southern Belize (Caribbean Sea), including the sex ratio, size at sexual maturity and reproductive cycle, as well as the size structure among habitats located inside and outside protected areas. Individuals found inside marine protected areas were consistently larger than individuals found outside these boundaries. The male:female sex ratio was 1.1:1.0 with a unimodal size distribution composed mainly of sexually mature individuals (87 %). Size at sexual maturity was established to be ~10 cm based on measures of gamogenetically mature individuals during the peak of the gametogenic maturity period. Juveniles and small adults measuring ≤ 15 cm (13 %), were collected exclusively in sandy and muddy areas of seagrass habitats associated with mangroves. Larger adults were commonly found on hard substrates in both seagrass-associated and coral-reef-associated habitats. Based on histology and gonad indices, the first sign of early gametogenesis was detected in August in both sexes. Gametes developed and matured throughout the colder months between November and February. Decreases in gonad index in both years suggest annual spawning, starting in MarchApril 2014 (single major event) and March-April and July-August 2015 (two smaller events) for both males and females. The spawning periods were correlated with warmer or increasing temperatures, low rainfall and increasing or high chlorophyll-a concentrations. These data will hopefully guide management and protection of the natural populations of H. mexicana, which are already suffering from overfishing in Belize and neighboring countries.(AU)

Abstract Holothuria grisea, despite not being considered a commercial resource has been the subject of illegal extractions in Colombia, a fact that becomes more serious due to the empty of biological information of these organisms. For this reason the density population and size structure of H. grisea were evaluated in three sectors south of the gulf of Morrosquillo from April to November 2015. To this end, three circular transects were located each month in each sampling site covering an area of 300 m2, where individuals were counted and measured in situ. The average density of H. grisea in the study area was 0.4 ind m-2, with the highest values in the La Ahumadera (1.09 ± 0.11 ind m-2) and the lowest values in Banco de Arena (0.02 ± 0.004 ind m-2). The analysis of variance showed a significance in the values of density and size of H. grisea in the studied sectors. The species presented a unimodal distribution of sizes with an average size of 13.5 ± 0.9 cm with a minimum size of 2.5 and a maximum of 30 cm, intermediate sizes generally showed a higher representativeness throughout this study (77.2 %). The biological aspects presented in this paper are of great importance for the conservation of this species and corresponds to an advance in the knowledge of the Holothuroidea class in the Colombian Caribbean. RESUMO

BACKGROUND: Sea cucumbers are highly prized seafood in Asia, where the demand for them has increased dramatically in recent years. However, nutritional information is only available for only a few of the commercially traded species. METHODS: In this study, the biochemical composition of the sea cucumber Australostichopus mollis was evaluated in terms of the major functional components: collagen, amino acid and fatty acid content using standard analytical procedures. RESULTS: The collagen, mostly type I, formed a homogeneously clustered network constituting 1.4% of the wet weight of adult A. mollis which is lower than for some other species of sea cucumber. The collagen consisted ofα1andα2 chains (around 116 kDa),αchain dimers,ßchains (around 212 kDa), and small amounts of γ components. The most abundant amino acids were glycine, alanine, threonine, serine and proline (lysine/arginine ratio of 0.1).Threonine was the most abundant essential amino acid, followed by methionine and valine, while glycine was the dominant non-essential amino acid. The sea cucumbers had a low lipid content (0.2 and 0.1% of wet weight forthe viscera and body wall, respectively) which is below the range reported for most other species of sea cucumber. The lipid contained high levels of PUFA (54%) compared to MUFA (23%) and SFA (24%). The dominant PUFA was arachidonic acid in both the body wall and viscera, followed by eicosapentaenoic acid. CONCLUSIONS: Overall, the Australasian sea cucumber has strong potential as a functional food due to its high levels of PUFA and essential amino acids, comparing favourably with the most commercially valuable sea cucumbers.

Background: The alga Laminaria japonica is the most economically important brown seaweed cultured in China, which is used as food and aquatic animal feedstuff. However, the use of L. japonica as a feedstuff in Apostichopus japonicasfarming is not ideal because A. japonicas does not produce enough enzyme activity for degrading the large amount of algin present in L. japonica. In this study, semi solid fermentation of the L. japonica feedstuff employing a Bacillus strain as the microbe was used to as a mean to degrade the algin content in L. japonica feedstuff. Results: The Bacillus strain, Bacillus amyloliquefaciens WB1, was isolated by virtue of its ability to utilize sodium alginate as the sole carbon source. Eight factors affecting growth and algin-degrading capacity of WB1 were investigated. The results of Plackett-Burman design indicated that fermentation time, beef extract, and solvent to solid ratio were the significant parameters. Furthermore, the mutual interaction between the solvent to solid ratio and beef extract concentration was more significant than the other pairs of parameters on algin degradation. Optimal values obtained from Central-Composite Design were 113.94 h for fermentation time, 0.3% (w/v) beef extract and 44.87 (v/w) ratio of solvent to feedstuff. Under optimal conditions, 56.88% of the algin was degraded when a 50-fold scale-up fermentation was carried out, using a 5-L fermenter. Conclusions: This study provides an alternative and economical way to reduce the algin content in L. japonicathrough degradation by WB1, making it a promising potential source of feed for cultured L japonica.

Abstract The fauna of the echinoderms of Mexico is represented by 643 species. Within these, 113 belong to the class Holothuroidea (sea cucumbers). The phyllophorids (Holothuroidea: Phyllophoridae) are characterized by the presence of more than 10 tentacles and a calcareous ring prolonged posteriorly with small pieces forming a mosaic-like appearance. Their cryptic habits have made them difficult to record during the many years of taxonomic studies regarding this group in Mexico. This paper aims to raise awareness of the state of knowledge on the diversity of the Phyllophoridae family in Mexican territorial waters. We present 16 species belonging to six genera, living in shallow (0 - 200 m) to deep (> 200 m) waters in the Mexican Pacific, Gulf of California, Mexican Caribbean and Gulf of Mexico. Within these, three new records are included. A taxonomic identification catalog was obtained for each species, providing information about external and internal morphology; the material was examined and recorded with information from type material and locality, all previous reports for Mexico, the geographic and bathymetric distribution as well as photographs taken with Scanning Electron Microscopy (SEM) of the shape and combination of the spicules. The work is complemented with a dichotomous key for identification. Rev. Biol. Trop. 63 (Suppl. 2): 77-85. Epub 2015 June 01.

RESUMO

Abstract The sea cucumber Holothuria (Halodeima) grisea is a widely distributed and economically important species that has been harvested in Brazil for several years, where there are no studies on its reproduction. We studied a population near Guaratuba Bay in Southern Brazil. Four sampling campaigns were made, in February, April, June and October 2008. We examined the macroscopic and microscopic features of the gonads to describe the gametogenesis. In addition, we analyzed the weight change of body organs. A total of 94 individuals were examined and no evidence of external sexual dimorphism was found. In April there were more males than females. Biometrically the body wall and gonads are the most important organs. The body wall increases in the June and the gonads during October and February; there might be an exchange of energy between these organs. Mature gonads are elongated and thick (red in females, cream in males). There were mature individuals year round, with an increase of sexual activity in February when oocyte mature diameter ranges from 70 to 160 µm. Spawning was in winter (June). Maturation period and body component variations were similar between sexes. Rev. Biol. Trop. 63 (Suppl. 2): 285-296. Epub 2015 June 01.

Background The sea cucumber lysozyme belongs to the family of invertebrate lysozymes and is thought to be a key defense factor in protecting aquaculture animals against bacterial infection. Recently, evidence was found that the sea cucumber lysozyme exerts broad spectrum antimicrobial action in vitro against Gram-negative and Gram-positive bacteria, and it also has more potent antimicrobial activity independent of its enzymatic activity. To explore the antimicrobial role of this non-enzymatic lysozyme and model its structure to novel antimicrobial peptides, the peptide from the C-terminal amino acid residues 70-146 of the sea cucumber lysozyme in Stichopus japonicus (SjLys-C) was heterologously expressed in Escherichia coli Rosetta(DE3)pLysS. Results The fusion protein system led to over-expression of the soluble and highly stable product, an approximate 26 kDa recombinant SjLys-C protein (rSjLys-C). The present study showed that rSjLys-C displayed strong antimicrobial activity against the tested Gram-positive and Gram-negative bacteria. In particular, the heat-treated rSjLys-C exhibited more inhibitive activity than the native rSjLys-C. The structural analysis of SjLys-C showed that it is a typical hydrophilic peptide and contains a helix-loop-helix motif. The modeling of SjLys-C molecular structures at different temperatures revealed that the tertiary structure of SjLys-C at 100°C underwent a conformational change which is favorable for enhancing antimicrobial activity. Conclusion These results indicate that the expressed rSjLys-C is a highly soluble product and has a strong antimicrobial activity. Therefore, gaining a large quantity of biologically active rSjLys-C will be used for further biochemical and structural studies and provide a potential use in aquaculture and medicine.

RESUMO

The brown sea cucumber fishery is active in the Galapagos Islands since the year 1991 after its collapse in mainland Ecuador. This paper analyzes the Galapagos Sea cucumber fishery over the past decade and the reasons for its management pitfalls and chronic over fishing, and proposes an improved strategy for estimating stock size and harvest potential. Based on the historical distribution of the fishing fleet and past fishery surveys, 15 macrozones were defined; their areas were estimated from the coastline to the 30m isobaths and the numbers of sample replicates per macrozone were calculated for a density estimate precision of ±25%. Overall stock size was calculated by summing over all macrozones and was multiplied by 0.122 to obtain the annual quota. This multiplier was derived by inserting an exploitation rate of E=0.3 and a published natural mortality value of M=0.17 into Cadimas formula, thereby obtaining a more conservative precautionary quota estimate. Pre-fishery stock densities in 2009 were below the legal threshold value and the fishery remained closed. Mean densities were significantly lower in the deeper (>15m) than in the shallower (<15m) stratum, contrary to fishermen expectations. Through an empirical regression of (log) pre-fishery density versus subsequent annual catch for the period 1998-2008 we found that catches of most years greatly exceeded the here proposed quota explaining the collapsed nature of the stock.

Distribution and density of two holothurian species in Cubagua Island, Venezuela. Holothurian populations in the Caribbean have been significantly depleted in some areas, and others are still in danger to satisfy international markets. In Venezuela, this marine resource has been illegally exploited since the early 1990s. To contribute with their management and protection, this work analyzed the distribution, abundance and population structure of two commercial holothurians Isostichopus badionotus and Holothuria mexicana in Cubagua Island shores. A total of 52 stations were studied around the island during the year 2008. Surveys included visits to each station, for which a band transect of 50m² with four replicates were carried out. The results show that I. badionotus has a higher distribution on the East coast of the island, and is found over Thalassia testudinum beds or bivalve molluscs aggregations, with an average density of 0.011ind/m², C.I. 95%=0.005-0.017, n=122, and a general abundance estimated in 117ind/ha. H. mexicana has been practically depleted (0.001 ind/m2 C.I. 95%=0.0002-0.0013, 7.7 ind/ha, n=8). We recommend the maintenance of the total ban adopted in 1997 to assure the protection of these species, mainly because no previous data supported the start of the fishery, and also, because the current densities are still very low when compared to other areas in Venezuela and the Caribbean. These are animals easy to catch, susceptible to overfishing, and with a very low recovery rate; thus, more control is required from the local authorities. Rev. Biol. Trop. 59 (2): 843-852.

The Isostichopus fuscus fishery in Mexico was heavily exploited until 1994, when it was closed due to overfishing. However, no information existed on the status of the populations. The fishery was evaluated through an age structured simulation model, and according to our analysis of the stock, the fishery can be feasible and sustainable as long as fishing mortality and age of first catch are optimized. In order to evaluate exploitation strategies, several scenarios were simulated considering different combinations of fishing intensities and ages of first catch. Input data for the model included population parameters, commercial catch and costs and benefits of the fishing operations. Yield production was strongly influenced by the fishing pressure and by the age of first capture. When the first one increased, significant decreases in yield and profits occurred. The best exploitation strategy was these parameters: fishing mortality level F = 0.15, age at first capture t(c) = 4 years, and yielding of approximately 430 tons. However, since the species reproduces for the first time at 5 years, extracting younger specimens would collapse the population. The critical value of fishing mortality was detected at Fc = 0.25. If exceeded, the population tends to exhaustion and the fishery is no longer profitable. In conclusion, I. fuscus fishery is highly vulnerable to overfishing and age of catch. It must be taken into account that the management policies should be considered as pilot and used on a regional basis. Continuous monitoring of the stock, control of the number of fishing licenses and extracting only specimens 5 yeasr-old and older (around 20 cm and >400 g), will allow the populations to recover from fishing activities
