Abstract Objectives: Ischemia-reperfusion injury is an important cause of multiple organ failure in cardiovascular surgery. Our aim is to investigate the effect of the probiotic Saccharomyces boulardii on oxidative stress, inflammatory response, and lung injury in an experimental model of aortic clamping. Methods: Twenty-one Wistar rats were randomized into three groups (n=7). Control group animals received saline gavage for a week before undergoing median laparotomy. In other groups, supraceliac aorta was clamped for 45 minutes to induce ischemia followed by reperfusion for 60 minutes. In the ischemia-reperfusion group, saline gavage was given preoperatively for one week. Ischemia-reperfusion+probiotic group rats received probiotic gavage for seven days before aortic clamping. The levels of oxidative stress markers and pro-inflammatory cytokines were determined in both serum and lung tissue samples. Ileum and lung tissues were harvested for histological examination. Results: Ischemia-reperfusion caused severe oxidative damage and inflammation evident by significant increases in malondialdehyde and cytokine levels (tumor necrosis factor alpha and interleukin-1 beta) and decreased glutathione levels in both serum and lung tissues. There was severe histological tissue damage to the lung and ileum in the ischemia-reperfusion group. Probiotic pretreatment before aortic clamping caused significant suppression of increases in serum and lung tissue malondialdehyde and tumor necrosis factor alpha levels. Histological damage scores in tissue samples decreased in the ischemia-reperfusion+probiotic group (P<0,005). Conclusions: Oral supplementation of probiotic S. boulardii before supraceliac aortic ischemia-reperfusion in rats alleviates lung injury by reducing oxidative stress, intestinal cellular damage, and modulation of inflammatory processes.

RESUMEN: La enfermedad diarreica aguda infantil (EDAI), constituye un problema de salud pública, representando la 2ª causa de morbimortalidad infantil en menores de 5 años, en el Ecuador. La hidratación oral y parenteral en los niños hospitalizados bajo normas de administración de conformidad con el grado de deshidratación y pérdida de peso, así como medidas preventivas como la vacunación obligatoria contra el rotavirus, han contribuido a disminuir, pero no a solucionar este problema de salud infantil. Múltiples factores contribuyen para que no se resuelva: socioeconómicos, educacionales, el destete temprano y malas prácticas alimenticias, entre otros. Últimos estudios han propuesto la utilización de probióticos que contribuyan a disminuir el problema sugieriendo el usode Saccharomyces boulardii (SB), asociado a un prebiótico; lo que permitiría acortar el tiempo de tratamiento de una EDAI; por lo que la simbiosis entre SB y un prebiótico denominado fructooligosacárido (FOS), podría ser una alternativa para reducir costos y complicaciones. Una alternativa para medir el curso clínico de una EDAI en infantes es la escala BITTS, de reciente creación y fácil aplicación por clínicos. El objetivo de este manuscrito fue resumir la evidencia existente respecto del rol de losprobióticos y prebióticos en la terapéutica de de la EDAI.


SUMMARY: In Ecuador childhood acute diarrheal disease (CADD) constitutes a serious public health problem, representing the 2nd cause of infant morbidity and mortality in children under 5 years of age. Oral and parenteral hydration in hospitalized children, with standard treatments according to their degree of dehydration and weight loss, as well as preventive measures such as mandatory vaccination against rotavirus, have contributed to a decrease. Nevertheless, this childhood disease has still not been resolved. There are multiple contributing factors involved that prevent complete eradication of the disease Among these are socio-economic problems, education, early weaning and poor feeding practices, all of which continue to affect infants. Recent studies have proposed the use of probiotics that help reduce the problem and it has been suggested that Saccharomyces boulardii (SB), associated with a prebiotic, would reduce the treatment time of an CADD. Therefore, the symbiosis between the SB probiotic and a prebiotic called fructo- oligosaccharide (FOS) could be an alternative to reduce complications and reduce costs. An alternative to measure the clinical course of an CADD in infants is the BITTS scale, which was recently created and can easily be applied by clinicians. The aim of this manuscript was to summarize the existing evidence regarding the role of PROBIOTICS and prebiotics in the treatment of CADD.

Abstract Food supplements have been increasingly investigated. Probiotics have several benefits for human and animal health and selenium (Se) is widely recommended against oxidative stress. In this context, the aim of this study was to develop a low-cost bioprocess to produce a functional food product comprising both probiotic and Se accumulation. Yeast cells of Saccharomyces boulardii CCT 4308 were cultivated using sugarcane molasses as substrate. Optimization studies were performed to evaluate the best medium composition for biomass production and Se-accumulation in batch and fed-batch systems. Optimized conditions were defined with a medium composed of 150 g L-1 sugarcane molasses and 12 g L-1 yeast extract, with feeding of 100 g L-1 sugarcane molasses and 100 μg mL-1 of Se incorporation after 4 h and 10 h of fermentation, respectively, during 48 h in STR (stirred tank reactor). Best biomass production reached 14.52 g L-1 with 3.20 mg Se g-1 biomass at 12 h. Process optimization led to 4.82-fold increase in biomass production compared to initial condition. A final Se-enriched S. boulardii CCT 4308 biomass was obtained, which is comparable to commercial products. An alternative probiotic yeast biomass was efficiently produced as a new food-form of Se supplement in a sustainable process using an inexpensive agro-industrial residue.

Cholera is endemic in over 50 countries with an estimated mortality of 100,000-120,000. Vaccination is considered the complementary key to prevent and control cholera; therefore, alternative vaccine preparations are needed. Toxin Co-regulated Pilus is part of the toxR virulence regulon, which is necessary for colonization in the intestinal mucosa. In order to express Vibrio cholerae TcpA protein in Saccharomyces boulardii, the expression plasmid pYES2 was constructed by inserting tcpA gene isolated from local Vibrio cholerae Eltor Inaba isolates. The new construct was transferred into Saccharomyces boulardii cells and the expression of tcpA gene was induced from the GAL1 promoter by adding galactose to the medium. The SDS-PAGE and Western blot analysis showed the presence of TcpA in yeast. These results showed that Saccharomyces boulardii is a promising host to express Vibrio cholerae toxin TcpA as the first step in attempt to produce an oral Vibrio cholerae vaccine(AU)


El cólera es endémico en más de 50 países. Se estima una mortalidad entre 100.000 - 120.000 debido a esta enfermedad. La vacunación se considera una medida complementaria para prevenir y controlar el cólera, por lo tanto, se necesitan preparaciones vacunales alternativas a las existentes. El Pili corregulado con la toxina, es parte del regulón de virulencia toxR, y es necesario para la colonización en la mucosa intestinal. Para expresar la proteína tcpA de Vibrio cholerae en Saccharomyces boulardii, se construyó el plásmido de expresión pYES2 insertando el gen tcpA obtenido a partir de aislamientos locales de Vibrio cholerae El Tor Inaba. La nueva construcción se transfirió a las células de Saccharomyces boulardii y se indujo la expresión del gen tcpA a partir del promotor GAL1 mediante la adición de galactosa al medio. El análisis mediante SDS-PAGE y Western blot demostró la presencia de TcpA en levaduras. Los resultados demostraron que Saccharomyces boulardii es un hospedero prometedor para expresar el gen tcpA de Vibrio cholerae como el primer paso en el intento de producir una vacuna oral contra Vibrio cholerae(AU)

Abstract The yeast Saccharomyces cerevisiae var. boulardii is a biotherapeutic agent used for the prevention and treatment of several gastrointestinal diseases. We report a case of fungemia in a patient suffering from Clostridium difficile-associated diarrhea and treated with metronidazole and a probiotic containing S. cerevisiae var. boulardii. The yeasts isolated from the blood culture and capsules were identified by MALDI-TOF MS and API ID 32 C as S. cerevisiae, and showed the same appearance and color on CHROMAgar Candida. Treatment with fluconazole 400mg/day was initiated and the probiotic was stopped. The patient was discharged from hospital in good condition and was referred to a rehabilitation center. We suggest that the potential benefit of S. cerevisiae var. boulardii should be accurately evaluated, especially in elderly patients. Moreover, all physicians should be trained in the use of probiotic agents and enquire whether the use probiotics was included in the patients'medical histories. © 2019 Asociación Argentina de Microbiología. Resumen Saccharomyces cerevisiae var. boulardii es un agente bioterapéutico usado en la prevención y el tratamiento de varias enfermedades gastrointestinales. Informamos de un caso de fungemia en una paciente con diarrea asociada a Clostridium difficile, y tratada con metron-idazol y un probiótico que contenía S. cerevisiae var. boulardii. Las levaduras aisladas a partir del hemocultivo y del contenido de las cápsulas tomadas por la paciente se identificaron como S. cerevisiae mediante MALDI-TOF MS y API® ID 32C, las colonias mostraron el mismo color y aspecto en el medio CHROMAgar™ Candida. Se instauró un tratamiento con fluconazol 400mg/día y se suspendió el probiótico. La paciente fue dada de alta del hospital en buenas condiciones, y remitida a un centro de rehabilitación. Sugerimos que el beneficio potencial del uso de S. cerevisiae var. boulardii debe ser evaluado en cada paciente, especialmente en personas añosas. El uso de probióticos debería incluirse en los interrogatorios orientados al diagnóstico y formar parte de la historia clínica. © 2019 Asociación Argentina de Microbiología. O objetivo do presente estudo foiavaliar os efeitos do probiótico Saccharomyces boulardii na modulação da resposta imune humoral de animais expostos a antígenos de Leishmania infantum. Para isso, 16 camundongos BALB/c foram imunizados com antígeno particulado de Leishmania infantum e divididos em dois grupos experimentais, um composto por animais suplementados e outro por animais não suplementados com o probiótico. Amostras de sangue dos animais foram colhidas semanalmente durante o período experimental e submetidas ao Ensaio da Imunoabsorbância Ligado à Enzima indireto para avaliação dos títulos de IgG totais e o perfil dos isotipos de IgG produzidos (IgG1 e IgG2a). A suplementação com o probiótico não exacerbou a produção de IgG total em comparação ao grupo controle, não havendo diferenças significativas entre os dois grupos. Porém, as soroconversões de IgG2a foram mais elevadas no grupo suplementado, no qual registrou-se um aumento de 1,46 vezes no final do experimento. Assim,a suplementação com S. boulardii foi capaz de modular a resposta de IgG2a/IgG1 nos animais expostos aos antígenos de Leishmania infantum.


The aim of the present study was to evaluate the effects of the Saccharomyces boulardii probiotic on the modulation of humoral immune response in animals exposed to Leishmania infantum antigens. For this, 16 BALB/c mice were immunized with Leishmania infantum particulate antigen and divided into two experimental groups, one consisting of supplemented animals and the other not probiotic supplemented animals. Blood samples from the animals were taken weekly during the experimental period and subjected to the Indirect Enzyme-Linked Immunosorbance Assay for evaluation of total IgG titers and the profile of the produced IgG isotypes (IgG1 and IgG2a). Probiotic supplementation did not exacerbate total IgG production compared to the control group, with no significant differences between the two groups. However, IgG2a seroconversions were higher in the supplemented group, which showed a 1.46-fold increase at the end of the experiment. Thus, supplementation with S. boulardii was able to modulate the IgG2a/IgG1 response in animals exposed to Leishmania infantum antigens.

There have been significant efforts towards the development of more efficient vaccines for animal health. A strategy that may be used to improve vaccine efficacy is the use of probiotics to enhance the immune response of the host, leading to increased immunogenicity of antigen preparations. Bovine herpesvirus 5 (BoHV-5) is an example of an important animal pathogen for which vaccines have provided only limited protection. In this study, we examined the use of the probiotic Saccharomyces boulardii (Sb) as a potential adjuvant to improve vaccine efficiency. We found that the supplemented animals exhibited an enhanced systemic IgG antibody response toward a Th1 response in favor of IgG2a and increased mRNA expression levels of the cytokines IFN-y, IL-12, IL-17 and IL-10 in the spleen. These results suggest that Sb supplementation may provide a promising means for improving the efficiency of vaccines, particularly those that rely on a cell-mediated immune response.(AU)


Esforços significativos têm sido realizados para o desenvolvimento de vacinas mais eficientes em saúde animal. Uma estratégia que pode ser usado para melhorar a eficácia da vacina é o uso de probióticos para melhorar a resposta imune do hospedeiro, conduzindo ao aumento da imunogenicidade de preparações de antígenos. Herpesvírus bovino 5 (BoHV-5) é um exemplo de um importante patógeno animal para os quais vacinas têm fornecido apenas uma protecção limitada. Neste estudo, examinou-se o uso do probiótico Saccharomyces boulardii (Sb) como um adjuvante potencial para melhorar a eficiência da vacina. Verificou-se que os animais suplementados apresentaram uma produção de anticorpos IgG superior e com desvio para Th1 em favor de IgG2, além do aumento dos níveis de expressão de mRNA para as citocinas IFN-γ, IL-12, IL-17 e IL-10. Esses resultados sugerem que a suplementação de Sb pode fornecer um meio promissor para melhorar a eficiência de vacinas, particularmente aquelas que dependem de uma resposta imune mediada por células.(AU)

Abstract Objective: The use of probiotics is increasingly popular in preterm neonates, as they may prevent necrotizing enterocolitis sepsis and improve growth and feeding tolerance. There is only limited literature on Saccharomyces boulardii CNCM I-745 (S. boulardii) in preterm infants. Method: A prospective, randomized, case-controlled trial with the probiotic S. boulardii (50 mg/kg twice daily) was conducted in newborns with a gestational age of 30-37 weeks and a birth weight between 1500 and 2500 g. Results: 125 neonates were enrolled; 63 in the treatment and 62 in the control group. Weight gain (16.14 ± 1.96 vs. 10.73 ± 1.77 g/kg/day, p < 0.05) and formula intake at maximal enteral feeding (128.4 ± 6.7 vs. 112.3 ± 7.2 mL/kg/day, p < 0.05) were significantly higher in the intervention group. Once enteral feeding was started, the time needed to reach full enteral feeding was significantly shorter in the probiotic group (0.4 ± 0.1 vs. 1.7 ± 0.5 days, p < 0.05). There was no significant difference in sepsis. Necrotizing enterocolitis did not occur. No adverse effects related to S. boulardii were observed. Conclusion: Prophylactic supplementation of S. boulardii at a dose of 50 mg/kg twice a day improved weight gain, improved feeding tolerance, and had no adverse effects in preterm infants >30 weeks old.


Resumo Objetivo: O uso de probióticos está cada vez mais popular em neonatos prematuros, já que podem prevenir a enterocolite necrosante (ECN) e a sepse e aumentar o crescimento e a tolerância de alimentação. Há apenas uma literatura limitada sobre a Saccharomyces boulardii CNCM I-745 (S. boulardii) em neonatos prematuros. Método: Um ensaio de caso-controle prospectivo randomizado com o probiótico S. boulardii (50 mg/kg duas vezes por dia) foi feito com recém-nascidos com idade gestacional de 30 a 37 semanas e peso ao nascer entre 1.500 e 2.500 g. Resultados: Foram incluídos 125 neonatos, 63 no grupo de tratamento e 62 no de controle. O ganho de peso (16,14 ± 1,96 em comparação com 10,73 ± 1,77 g/kg/dia, p < 0,05) e a ingestão de fórmula com nutrição enteral máxima (128,4 ± 6,7 em comparação com 112,3 ± 7,2 mL/kg/dia, p < 0,05) foram significativamente maiores no grupo de intervenção. Assim que a nutrição enteral foi iniciada, o tempo necessário para atingir a nutrição enteral completa foi significativamente menor no grupo probiótico (0,4 ± 0,1 em comparação com 1,7 ± 0,5 dia, p < 0,05). Não houve diferença significativa em sepse. Não ocorreu ECN. Não foi observado efeito colateral relacionado à S. boulardii. Conclusão: A suplementação profilática de S. boulardii em uma dose de 50 mg/kg duas vezes por dia melhorou o ganho de peso, aumentou a tolerância de alimentação e não teve efeito colateral em neonatos prematuros > 30 semanas de idade.

Saccharomyces boulardii é uma levedura não patogênica, termotolerante e resistente à ação dos sucos gástrico, entérico e pancreático. Esta levedura atua no intestino por três mecanismos: efeito antagonista direto, imunoestimulação e efeito trófico na mucosa intestinal. O objetivo deste trabalho de revisão é contribuir para uma atualização dos profissionais de saúde sobre as indicações terapêuticas e as formas de uso da levedura S. boulardii como agente probiótico. S. boulardii é utilizada sob a forma de medicamentos como agente terapêutico na diarreia, com eficácia comprovada em muitas condições patológicas que cursam com diarreias agudas e crônicas. Além de não alterar a morfologia intestinal nem a microbiota colônica residente como os antibióticos, esta levedura tem resultados equivalentes aos medicamentos padrões utilizados. Devido a esses fatores, é vantajosa a utilização de medicamentos à base deste probiótico.


Saccharomyces boulardii is a nonpathogenic yeast which is thermotolerant and resistant to gastric, pancreatic and enteric secretions. This yeast works in the bowel by three ways: direct antagonist effect, immunostimulation and trophic effect upon the intestinal mucosa. The objective of this revision is to contribute to an upgrade of health professionals about the therapeutic indications and ways of using the yeast S.boulardii as a probiotic. S. boulardii is employed in the form of medicines as therapeutic agent on the treatment of diarrhea with proven efficiency in pathological conditions that develop with acute and chronic diarrhea. Neither interfering in intestinal morphology nor in the colonic microbiota like antibiotics, this yeast has shown efficacy equivalentto standard medicines employed. Due to these factors it is very profitable the use of this probiotic-based medicines.

Neste trabalho objetivou-se a indução de fitoalexinas em mesocótilos de sorgo e cotilédones de soja pela levedura Saccharomyces boulardii na forma do produto comercial Floratil (Merck) (com 2 x 106 células/mg produto comercial - pc) e massa de células obtidas de meio líquido YEPG (primeiramente com 14 dias de cultivo e, posteriormente, com 7, 14, 21, 28 e 35 dias) ambos em concentrações de 0,005; 0,05; 0,5; 5; 15 e 25 mg/mL, além de filtrado desse meio nas concentrações de 0,01; 0,1; 1; 5; 10 e 20%. Como tratamentos controle utilizou-se água e S. cerevisiae (25 mg/mL de pc) para soja e água e acibenzolar-S-metil (ASM) (125 mg i.a./L) para sorgo. Em soja os três produtos apresentaram efeito dose-dependente, com ajustes de equações de 1° grau e R2 de 0,64; 0,94 e 0,98 não tendo efeito do tempo de cultivo da levedura na indução de fitoalexinas. Em sorgo apenas o filtrado e Floratil tiveram efeito dose-dependente com equação de 1° grau e R2 de 0,63 e 0,94 respectivamente e obteve-se nos diferentes dias de cultivo R2 de 0,62 com a massa de células somente. Portanto, pode-se evidenciar o potencial indutor de fitoalexinas dos produtos a base de S. boulardii para ensaios com indução de resistência em patossistemas envolvendo sorgo e soja.


Saccharomyces yeast compounds have been used as a resistance elicitor or for biological control in many pathosystems. Thus, the aim of this research was to verify the induction of phytoalexins in sorghum mesocotyls and soybean cotyledons by using Saccharomyces boulardii in the form of the commercial product Floratil (Merck) (with 2 x 106 cells/mg) and yeast-cell mass obtained from liquid culture in YEPG medium (with 7, 14, 21, 28 and 35 days old), both at concentrations of 0.005, 0.05, 0.5, 5, 15 and 25 mg/mL, as well as the filtrate of this medium in concentrations of 0.01, 0.1, 1, 5, 10 and 20%. The control treatments consisted of distilled water and S. cerevisiae (25 mg of commercial product per mL) for the soybean tests and distilled water and acibenzolar-S-methyl (125 mg of active ingredient per L) for the sorghum tests. In soybeans the three tested S. boulardii products presented a dose-dependent effect with R2 of 0.64, 0.94 and 0.98 for the culture filtrate, cell suspension and commercial product of S. boulardii, respectively, with no effect of culture time of yeasts on phytoalexin induction. In sorghum, only the culture filtrate and Floratil presented a dose-dependent effect, with R2 of 0.63 and 0.94, respectively, and the cell suspension of S. boulardii showed dependence of culture time with R2 of 0.62. Thus, S. boulardii and its derivates induce phytoalexins and have potential to be used as an elicitor for assays with induction resistance in pathosystems involving sorghum and soybean plants.

Probióticos são definidos como microrganismos viáveis que exibem um efeito benéfico na saúde do hospedeiro, quando ingeridos em quantidades suficientes. Muitas espécies de bactérias são usadas para esse fim e a única levedura utilizada como probióticos em seres humanos é Saccharomyces boulardii. Resultados anteriores obtidos em nosso laboratório mostraram que a levedura Saccharomyces cerevisiae linhagem UFMG 905, isolada da produção de cachaça, foi capaz de colonizar e sobreviver no trato gastrintestinal de camundongos, isentos de germes e convencionais, respectivamente, além de proteger esses animais contra um desafio oral com Salmonella enterica subsp. enterica sorovar Typhimurium e Clostridium difficile. Na primeira parte desse trabalho, os efeitos de S. cerevisiae UFMG 905 na translocação de Salm. Typhimurium para os linfonodos mesentéricos, placas de Peyer, baço e fígado, assim como o efeito no sistema imune, pela contagem de células de Küpffer, produção de imunoglobulinas e clareamento de Escherichia coli B41 da corrente sanguínea, foram avaliados em camundongos gnotobióticos e/ou convencionais. O tratamento com a levedura reduziu significantemente a translocação de Salm. Typhimurium para o fígado de animais gnotobióticos e, para todos os órgãos testados, em animais convencionais. O número de células de Küpffer (por 100 hepatócitos) foi significantemente maior (P < 0,05) em camundongos monoassociados com a levedura (52,9 ± 15,7) que em camundongos isentos de germes (38,1 ± 9,0). Provavelmente, como uma consequência do aumento do número de células de Küpffer, o clareamento de E. coli B41 da corrente sanguínea foi mais eficiente nos animais monoassociados com a levedura, quando comparado com os animais controle isentos de germes. Foram observados maiores níveis de sIgA no conteúdo intestinal e de IgA e IgM no soro (P < 0,05) nos camundongos monoassociados com a levedura que no grupo isento de germes. Concluindo, a proteção observada contra a bactéria enteropatogênica em nosso estudo anterior foi, provavelmente, devida à modulação local e sistêmica do sistema imune de camundongos tratados com S. cerevisiae UFMG 905. Na segunda parte desse trabalho, estudou-se os efeitos de S. boulardii e S. cerevisiae UFMG 905 na inflamação e na sinalização celular induzidos pela Salm. Typhimurium 14028, em células T84. Observou-se que os dois probióticos mantiveram a resistência elétrica transepitelial e diminuiram, significativamente, a secreção de IL-8 em células T84 infectadas por Salm. Typhimurium 14028 (P < 0,05). Saccharomyces cerevisiae UFMG 905 e S. boulardii diminuiram, significativamente, os níveis de invasão pela Salm. Typhimurium 14028 (P < 0,05), mas nenhum efeito das leveduras no xxxii crescimento ou adesão bacteriana em células T84 foi observado. Diferentemente de S. boulardii, S. cerevisiae UFMG 905 não foi capaz de diminuir a ativação de Rac1 e Cdc42 em células infectadas por Salm. Typhimurium 14028. Essas duas proteínas são Rho-GTPases ativadas pela bactéria e estão envolvidas na internalização de bactérias invasivas. A ligação de Salm. Typhimurium 14028 à superfície de S. cerevisiae UFMG 905 e S. boulardii, e não às células epiteliais do intestino, pode ser responsável pela diminuição da invasão e ativação de MAPKs (mitogen-activated protein kinases) e, conseqüentemente, pela diminuição dos níveis de IL-8, uma vez que esse processo diminui o número de bactérias ligadas às células T84. Entretanto, a diminuição dos níveis de IL-8 também pode ser explicada por uma imunomodulação, via secreção de citocinas anti-inflamatórias, como a IL-10, mas essa hipótese não foi testada nesse trabalho. A presença das leveduras em células infectadas com Salm. Typhimurium 14028 também reduziu e/ou inibiu a fosforilação das MAPKs ERK1/2, p38 e JNK, mas não diminuiu a ligação do fator de transcrição para IL-8 (NF-κB) ao DNA, sugerindo que a diminuição dos níveis de IL-8 não foi devido à inibição do fator de transcrição para IL-8, mas, provavelmente, ao efeito das leveduras na inibição da MAPK p38, que é a proteína responsável pela estabilização do mRNA para IL-8. Uma outra explicação é o fato de as duas leveduras inibirem a MAPK JNK, uma vez que AP-1 (outro fator de transcrição para IL-8) é dependente da fosforilação dessa proteína. Finalmente, S. cerevisiae UFMG 905 e S. boulardii inibem, também, a ativação de mecanismos antiinflamatórios (a via PI3K/Akt) induzidos por Salm. Typhimurium 14028 em células T84. Concluindo, S. cerevisiae UFMG 905 e S. boulardii apresentaram um efeito protetor e modulador na função de barreira e na sinalização celular induzida em células T84 pela Salm. Typhimurium 14028.


Probiotics are defined as viable microorganisms that exhibit a beneficial effect on the host health when ingested in adequate amounts. Many species of bacteria are used for this purpose and the only one yeast used as probiotic in humans is Saccharomyces boulardii. Previous results in our laboratory showed that Saccharomyces cerevisiae strain UFMG 905, isolated from "cachaça" production, was able to colonize and survive in the gastrointestinal tract of germ-free and conventional mice, respectively, and to protect these animals against oral challenge with Salmonella enterica subsp. enterica serovar Typhimurium and Clostridium difficile. In the first part of this work, the effects of S. cerevisiae UFMG 905 on the translocation of Salm. Typhimurium to mesenteric lymph nodes, Peyer's patches, spleen and liver, as well as on the immune system by number of Küpffer cells, immunoglobulin production and clearance of Escherichia coli B41, were evaluated in gnotobiotic and/or conventional mice. The treatment with the yeast reduced significantly the translocation of Salm. Typhimurium to liver in gnotobiotic animals and to all the organs tested in conventional mice. The number of Küpffer cells per 100 hepatocytes in liver was significantly higher (P < 0.05) in yeast mono-associated mice (52.9 ± 15.7) than in germ-free controls (38.1 ± 9.0). Probably, as a consequence, clearance of E. coli B41 from the bloodstream was more efficient in yeast mono-associated animals when compared to germ-free mice. Higher levels (P < 0.05) of sIgA in intestinal content and of IgA and IgM in serum were observed in yeast mono-associated mice when compared to germ-free group. Concluding, the protection against pathogenic bacteria observed in a previous study was probably due to a modulation of both local and systemic immunity of mice treated with S. cerevisiae UFMG 905. In a second part of this work, we have studied the effects of S. boulardii and S. cerevisiae UFMG 905 on the inflammation and signal transduction induced by Salm. Typhimurium ATCC 14028 in T84 cells. We have observed that both probiotics maintained the transmonolayer electrical resistance and significantly diminished IL-8 secretion in Salm. Typhimurium 14028-infected T84 cells (P < 0.05). Saccharomyces cerevisiae UFMG 905 and S. boulardii also decreased significantly the levels of Salm. Typhimurium 14028 invasion (P < 0.05), but had no effect on Salm. Typhimurium 14028 growth or adhesion to T84 cells. Differently from S. boulardii, S. cerevisiae UFMG 905 was not implicated in the diminution of the activation of Rac1 and Cdc42 in Salm. Typhimurium 14028-infected cells, which are Rho-GTPases activated by the bacteria involved in the internalization of invasive bacteria. The binding of Salm. Xxxv Typhimurium 14028 to S. cerevisiae UFMG 905 and S. boulardii surface instead of T84 cells may be responsible for the diminution of invasion and activation of MAPKs (mitogen-activated protein kinases) and consequently by the diminution of IL-8 levels, since this process diminishes the number of bacteria bound to T84 cells. However, the diminution of IL-8 may also be explained by an immunomodulation through secretion of anti-inflammatory cytokines, such as IL-10, but this hypothesis was not tested in this work. The presence of the yeasts in Salm. Typhimurium 14028-infected cells also reduced and/or inhibited the phosphorylation of ERK1/2, p38 and JNK MAPKs, but did not inhibit NF-κB DNA binding activity, suggesting that the diminution of IL-8 levels was not due to inhibition of the IL-8 transcription factor but more probably to the effects of the yeasts on the p38 MAPK inhibition, which is responsible for the stabilization of IL-8 mRNA. The inhibition of JNK by the yeasts may be another explanation, once AP-1, another IL-8 transcription factor, is dependent on the phosphorylation of this protein. Finally, S. cerevisiae UFMG 905 and S. boulardii inhibited the activation of anti-inflammatory mechanism (PI3K/Akt pathway) induced by Salm. Typhimurium 14028 in T84 cells. Concluding, S. cerevisiae UFMG 905 and S. boulardii showed a protective and modulating effect on barrier function and signal transduction pathway in T84 cells when infected by Salm. Typhimurium 14028.
