Abstract The aim of the current study was to assess the physicochemical characteristics and wound healing activity of chitosan-polyvinyl alcohol (PVA) crosslinked hydrogel containing recombinant human epidermal growth factor (rh-EGF) or recombinant mouse epidermal growth factor (rm-EGF). The hydrogels were prepared and analyses were made of the morphological properties, viscosity, water absorption capacity, mechanical and bio-adhesive properties. The viscosity of the formulations varied between 14.400 - 48.500 cPs, with the greatest viscosity values determined in K2 formulation. F2 formulation showed the highest water absorption capacity. According to the studies of the mechanical properties, H2 formulation (0.153±0.018 N.mm) showed the greatest adhesiveness and E2 (0.245±0.001 mj/cm2) formulation, the highest bio-adhesion values. Hydrogels were cytocompatible considering in vitro cell viability values of over 76% on human keratinocyte cells (HaCaT, CVCL-0038) and of over 84% on human fibroblast cells (NIH 3T3, CRL-1658) used as a model cell line. According to the BrdU cell proliferation results, B1 (197.82±2.48%) formulation showed the greatest NIH 3T3 and C1 (167.43±5.89%) formulation exhibited the highest HaCaT cell proliferation ability. In addition, the scratch closure assay was performed to assess the wound healing efficiency of formulation and the results obtained in the study showed that F2 formulation including PEGylated rh-EGF had a highly effective role.

Abstract The aims of this work were to produce hyaluronic acid (HA) by Streptococcus zooepidemicus ATCC 39920 in a low cost sugarcane molasses fermentation medium and to employ the produced HA to obtain films blends based on polyvinyl alcohol (PVA). The films were produced using solution casting method and they were characterized according to their microstructure, mechanical and barrier properties. HA was added in different concentrations (0, 5, 10 and 15% (w/w)), and glycerol was used as a plasticizer (25 g/100 g solids). All formulations resulted in easily manipulated films with good appearance. The addition of HA on PVA films increased their thermal stability, solubility, swelling index, water vapor permeability and elongation. Microbial HA sample combined with PVA showed to be a promising material to biomedical application, and an addition between 5 and 10% (w/w) was sufficient to improve PVA films properties.

Composite membrane as a flexible materials have found diverse applications in industrial and biomedical simultaneously, the recent studies have shown intrinsic improvement for membrane properties by inclusion of nanoparticles as a fillers with high portion ratio in inorganic polymers, the combination between two parts polymer and filler is as a result of collection the advantage of two component systems parts together. In this work, samples of polyvinyl alcohol (PVA)-nanoHaydroxyapatite (nHAp) composites were prepared by using casting method. The effects of addition of (nHAp) with different concentration on the optical properties of (PVA- nHAp) composite membrane have been studied by using wavelength range (220-820) nm. The absorption spectra, transmittance spectra, absorption coefficient, energy gap, refractive index, optical conductivity and extinction coefficient have been determined. The results show that the optical constants change with the increase of nHPA concentrations.

Infecções cutâneas obtidas por cortes nas mãos de manipuladores podem gerar contaminação nos alimentos. O objetivo deste trabalho foi incorporar o bacteriófago UFV-AREG1 em hidrogel de álcool poli(vinílico) – PVA, aplicando-o futuramente como curativo adesivo. Foi adicionado 1,0 mL de solução do fago no PVA e seco em estufa por 48 h. Após, os hidrogeis foram submetidos a testes de Intumescimento, Espectroscopia no infravermelho (FTIR) e efeito antimicrobiano sobre E. coli O157:H7. O intumescimento do PVA-fago foi maior que o PVA-controle (p<0,05). A área de inibição do PVA-fago foi de 3,715 cm2 contra 2,916 cm2 do controle. As análises do FTIR mostraram um pico para o PVA-fago não encontrado no PVA-controle. Foi possível incorporar o bacteriófago em hidrogel de PVA e avaliar sua liberação para posterior aplicação como curativo adesivo.

Abstract Ammonia-oxidizing bacteria were immobilized by polyvinyl alcohol (PVA) and sodium alginate. The immobilization conditions and ammonia oxidation ability of the immobilized bacteria were investigated. The following immobilization conditions were observed to be optimal: PVA, 12%; sodium alginate, 1.1%; calcium chloride, 1.0%; inoculum concentration, 1.3 immobilized balls/mL of immobilized medium; pH, 10; and temperature, 30 °C. The immobilized ammonia-oxidizing bacteria exhibited strong ammonia oxidation ability even after being recycled four times. The ammonia nitrogen removal rate of the immobilized ammonia-oxidizing bacteria reached 90.30% under the optimal immobilization conditions. When compared with ammonia-oxidizing bacteria immobilized by sodium alginate alone, the bacteria immobilized by PVA and sodium alginate were superior with respect to pH resistance, the number of reuses, material cost, heat resistance, and ammonia oxidation ability.

OBJETIVO: avaliar a utilização de uma nova partícula de polivinil álcool e polivinil acetato (PVA-PVAc) esférica, para embolização das artérias uterinas, em pacientes portadoras de mioma, com indicação cirúrgica. MÉTODOS: doze pacientes foram submetidas à embolização de miomas uterinos com partículas de PVA-PVAc. Três a nove meses depois, realizou-se uma laparotomia com miomectomia. Analisaram-se os seguintes parâmetros: volume do útero e do maior mioma; concentrações do hormônio folículo estimulante e de hemoglobina; sangramento menstrual (número de dias e de absorventes utilizados), sinais e sintomas antes do tratamento, após a embolização e após a miomectomia. RESULTADOS: a média de idade foi 37 anos e a média do volume uterino, previamente ao tratamento, de 939,3cc. Três anos após a embolização, observou-se diminuição do volume uterino (p=0,0005). Houve melhora na concentração de hemoglobina (p= 0,0004), com elevação após a embolização, sem variação subsequente à miomectomia. Não ocorreu variação significante do hormônio folículo estimulante, (p=0,17). Não foi constatado nenhum caso de falência ovariana, mas uma das pacientes apresentou atrofia de endométrio. Duas pacientes engravidaram, com bons indicadores obstétricos. Quanto aos sinais e sintomas, houve melhora após a embolização, que se manteve após a miomectomia. CONCLUSÃO: a embolização arterial com partículas de PVA-PVAc esférico mostrou-se promissora no preparo para uma intervenção cirúrgica com retirada dos miomas, pois, associou-se à redução do volume uterino, à diminuição do sangramento operatório e tornou possível a utilização de incisões menores, aumentando a chance de preservação do útero.


OBJECTIVE: To evaluate the use of a new spherical particle of polyvinyl alcohol and polyvinyl acetate (PVA-PVAc) for uterine artery embolization in patients with myoma with surgical indication. METHODS: twelve patients underwent uterine myoma embolization with PVA-PVAc particles. Three to nine months later, they were submitted to laparotomy with myomectomy. We analyzed the following parameters: volume of the uterus and of the bigger myoma; concentrations of follicle stimulating hormone and hemoglobin; menstrual bleeding (number of days and used absorbents), signs and symptoms before treatment, after embolization and after myomectomy. RESULTS: The mean age was 37 years and mean uterine volume prior to treatment, 939.3 cc. Three years after embolization, there was reduction in uterine volume (p = 0.0005), increase in hemoglobin concentration after embolization (p = 0.0004), without variation after the myomectomy. There was no significant variation of the follicle stimulating hormone (p = 0.17). There was no case of ovarian failure, but one of the patients had endometrial atrophy. Two patients became pregnant, with good obstetric indicators. Signs and symptoms improved after embolization, and remained after myomectomy. CONCLUSION: Arterial embolization with spherical PVA-PVAc particles is promising in the preparation for myoma surgery, since it was associated with a reduction in uterine volume, decrease in intraoperative bleeding and made possible the use of smaller incisions, increasing the chance of preserving the uterus.

Tissue engineering is a technique by which a live tissue can be re-constructed and one of its main goals is to associate cells with biomaterials. Electrospinning is a technique that facilitates the production of nanofibers and is commonly used to develop fibrous scaffolds to be used in tissue engineering. In the present study, a different approach for cell incorporation into fibrous scaffolds was tested. Mesenchymal stem cells were extracted from the wall of the umbilical cord and mononuclear cells from umbilical cord blood. Cells were re-suspended in a 10 percent polyvinyl alcohol solution and subjected to electrospinning for 30 min under a voltage of 21 kV. Cell viability was assessed before and after the procedure by exclusion of dead cells using trypan blue staining. Fiber diameter was observed by scanning electron microscopy and the presence of cells within the scaffolds was analyzed by confocal laser scanning microscopy. After electrospinning, the viability of mesenchymal stem cells was reduced from 88 to 19.6 percent and the viability of mononuclear cells from 99 to 8.38 percent. The loss of viability was possibly due to the high viscosity of the polymer solution, which reduced the access to nutrients associated with electric and mechanical stress during electrospinning. These results suggest that the incorporation of cells during fiber formation by electrospinning is a viable process that needs more investigation in order to find ways to protect cells from damage.

The treatment of diseases affecting the posterior segment of the eye is limited by the difficulty in transporting effective doses of drugs to the vitreous, retina, and choroid. Topically applied drugs are poorly absorbed due to the low permeability of the external ocular tissues and tearing. The blood-retina barrier limits drug diffusion from the systemic blood to the posterior segment, thus high doses of drug are needed to maintain therapeutic levels. In addition, systemic side effects are common. Intraocular injections could be an alternative, but the fast flowing blood supply in this region, associated with rapid clearance rates, causes drug concentration to quickly fall below therapeutic levels. To obtain therapeutic levels over longer time periods, polymeric sustained-drug release systems implanted within the vitreous are being studied for the treatment of vitreoretinal disorders. These systems are prepared using different kinds of biodegradable or non-biodegradable polymers. This review aims to demonstrate the main characteristics of these drug delivery implants and their potential for clinical application.


O tratamento de doenças do segmento posterior do olho é limitado pela dificuldade no transporte de doses efetivas de fármacos para o vítreo, retina e coróide. Os fármacos aplicados topicamente são pouco absorvidos por causa da baixa permeabilidade dos tecidos oculares externos e ao lacrimejamento. Embora a administração sistêmica seja capaz de transportar fármacos para o segmento posterior do olho, as barreiras hemato-aquosa e hematorretiniana dificultam a absorção e, normalmente, são necessárias doses elevadas, as quais estão geralmente associadas a potenciais efeitos adversos. Injeções intravitreais são capazes de transportar fármacos para o segmento posterior do olho, mas é uma técnica invasiva, pouco tolerada pelos pacientes e apresenta riscos de infecções oculares e danos aos tecidos. Visando a obtenção de níveis terapêuticos adequados de fármacos no segmento posterior do bulbo do olho por longos períodos, sistemas de liberação poliméricos implantados diretamente no vítreo estão sendo investigados para o tratamento de várias doenças vítreo-retinianas. Esses implantes podem ser preparados a partir de diferentes polímeros biocompatíveis, biodegradáveis ou não-biodegradáveis. Nesta revisão, as principais características destes implantes transportadores de fármacos são descritas, expondo suas potencialidades de aplicação clínica.

OBJECTIVES: Evaluate the degree of vascular occlusion, vascular recanalization, and necrosis of the vascular wall caused by polyvinyl alcohol-covered polyvinyl acetate (PVAc) particles compared to trisacryl particles after renal embolization. METHODS: Seventy-nine female albino New Zealand rabbits underwent arterial catheterization of the right kidney. Thirty-three animals were embolized with trisacryl particles, thirty-one with PVAc particles, and fifteen were kept as controls. Four animals were excluded (three trisacryl and one PVAc) due to early death. Five subgroups of six animals were created. The animals in the different groups were sacrificed either 48 hours, 5 days, 10 days, 30 days, or 90 days after embolization. The control group was divided into subgroups of three animals each and kept for the same periods of time. The kidneys were dyed with hematoxylin-eosin and Masson's trichrome and then examined using optical microscopy. RESULTS: There were significant differences in the degree of vascular occlusion caused by the trisacryl and the PVAc particles between the five-day and the ten-day groups. Additional differences were noted between the five-day and 48-hour groups in regard to the amount of necrosis. For both findings, the PVAc group members showed adequate tissue reaction (ischemia and volumetric reduction) and less recanalization than those treated with trisacryl. CONCLUSION: The use of PVAc as an embolization material exhibited an adequate tissue reaction (ischemia and volumetric reduction), more expressive vascular occlusion and necrosis, and less recanalization than the trisacryl material.

Cultures of adipose tissue explants are a valuable tool for studying the intracellular mechanisms involving hormones and nutrients. However, testing how fatty acids affect cells requires a carrier molecule; bovine serum albumin (BSA) has been used for this purpose. However, contaminants can alter the cellular response. Our objectives were to: 1) test BSA as a fatty acid carrier and 2) evaluate polyvinyl alcohol (PVA) as a replacement for BSA. Adipose tissue explants from nine pigs were cultured in medium 199 for 4, 12, 24, and 48 h, with the following treatments: control, PVA (100 mM PVA added) and PVA + pGH (100 mM PVA plus 0.1 mg/mL porcine growth hormone). After each culture period, explants were collected and assayed for lipogenesis. After 48 h in culture, explants were assayed for lipolysis. A preliminary study with different commercial sources and high concentrations showed that BSA affected lipogenic rates. On the other hand, there were no effects of PVA on lipid synthesis, while pGH (positive control) reduced glucose incorporation into lipids (P < 0.01) when compared to both control and PVA (P < 0.05). There was no difference between control and PVA for lipolysis rates. However, pGH increased lipolysis when compared to control (P < 0.01) and PVA (P < 0.05). We demonstrated that BSA can alter lipogenesis, which precludes its use as a carrier molecule. On the other hand, addition of PVA had no effect on lipolysis or lipogenesis. We suggest the use of PVA instead of BSA for adding bioactive fatty acids to cultures of adipose tissue

Os autores relatam um caso de tumor de corpo carotídeo (paraganglioma) em um paciente de 74 anos de idade, submetido a embolização intra-arterial com micropartículas esféricas, de polivinil acetato, com casca de polivinil álcool (PVAc + PVA), previamente à ressecção do tumor. O estudo angiográfico demonstrou massa altamente vascularizada na bifurcação carotídea esquerda, sendo a embolização pré-operatória utilizada no intuito de diminuir a vascularização e reduzir a perda sangüínea, aumentando a segurança do tratamento cirúrgico. O estudo histopatológico confirmou a presença de trombose e isquemia tecidual.


The authors report a case of carotid body tumor (paraganglioma) in a 74-year-old male patient, submitted to intraarterial embolization with spherical core/shield polyvinyl acetate and polyvinyl alcohol (PVAc + PVA) microparticles prior to surgical excision. Angiography has demonstrated a highly vascularized mass in the left carotid bifurcation, and preoperative embolization was used in order to decrease vascularity, reduce blood loss and improve safety of surgical treatment. Microscopic study confirmed presence of thrombosis and tissue ischemia.

Los reportes de mortalidad en pacientes con hemoptisis fluctúan aproximadamente en un 50 a 85 por ciento, (según caso clínico), para ello, es importante la evaluación cuidadosa e iniciar un tratamiento para de tener la hemorragia y permitir un adecuado tratamiento de la patología de fondo con un paciente estabilizado hemodinßmicamente en un manejo multidisciplinario, entre el neumólogo, el radiólogo intervencionista, el anestesiólogo. El siguiente trabajo, muestra la experiencia del Instituto Brazzini Radiólogos Asociados, en la embolización de arterias bronquiales, en pacientes con hemoptisis; procedimiento que se realiza desde 1974. Objetivos: Determinar el éxito del manejo en pacientes con hemoptisis, mediante la embolización de las arterias bronquiales. Materiales y métodos: El trabajo de investigación es de tipo longitudinal, prospectivo de tipo descriptivo; el estudio abarcó un total de 11 pacientes con hemoptisis severa, masiva, rebelde al tratamiento médico, con una edad media de 36.6 a±os de edad (rango: 23 - 62 a±os). Se realizaron estudios angiogrßficos y embolización en todos los pacientes (2 de ellos con hemoptisis recidivante), así como la revisión de las arterias sistémicas del tórax (arterias mamaria y torßcico laterales), y una embolización de las mismas si fuera necesario. El material de embolización que se utilizó, fueron partículas de polivinil - alcohol (PVA), émbolos metßlicos espirales entretejidos con fibras de dacrón y émbolos de gelfoam. Resultados: Se consiguió selectivizar las arterias responsables del sangrado, obteniéndo un éxito técnico para la embolización del 100 por ciento. Se presentó recidiva en un 18,1 por ciento , en los que se realizó una nueva embolización. No se observaron complicaciones. Conclusión: La embolización de arterias bronquiales, constituye un tratamiento eficaz de la hemoptisis severa, masiva y recidivante.

Objetivo: dar una nueva opción, sencilla en su metodología, con materiales al alcance de todos, que permita evaluar y comparar los efectos de las técnicas de instrumentación en la geometría de los conductos radiculares de dientes extraídos, previo y posterior a la instrumentación. Hipótesis: con esta mufla no existe pérdida de estructura dental al corte de las muestras y se observan claramente los límites entre la pared de las raíces y la mufla. Materiales: resina acrílica isotérmica autopolimerizable (polímero), monómero de metilmetacrilato, resina poliéster insaturada cristal (MC40), alcohol polivinilo, peróxido de metil-etil-cetona (MEC), hule de silicón o caucho (poliórgano silicato, etil silicato, dibutilin dilaurate, etanol). Método: las superficies de las muestras fueron cubiertas con acrílico autopolimerizable transparente, se les colocó una película separadora a base de alcohol polivinilo o vaselina, se realizaron impresiones a cubos de acrílico prefabricados, se mezcló la resina cristal con catalizador de peróxido de metil-etil-cetona al 1-2 por ciento, se vacía la mezcla en los moldes impresionados, se introduce la raíz cubierta por el acrílico en la resina cristal con previo separador, retirando la raíz cubierta por acrílico y somete a secciones en la longitud radicular. Resultados: permitió hacer las mediciones en cada pared y en cada tercio. Conclusiones: las ventajas de este dispositivo es que no existe pérdida de estructura dental y se observan claramente los límites entre la pared de las raíces y la mufla y a un bajo costo

RESUMO

Immunodetection of human IgG anti-Toxocara canis was developed based on ELISA and on the use of polysiloxane/polyvinyl alcohol (POS/PVA) beads. A recombinant antigen was covalently immobilized, via glutaraldehyde, onto this hybrid inorganic-organic composite, which was prepared by the sol-gel technique. Using only 31.2 ng antigen per bead, a peroxidase conjugate dilution of 1:10,000 and a serum dilution of 1:200 were adequate for the establishment of the procedure. This procedure is comparable to that which utilizes the adsorption of the antigen to conventional PVC plates. However, the difference between positive and negative sera mean absorbances was larger for this new glass based assay. In addition to the performance of the POS/PVA bead as a matrix for immunodetection, its easy synthesis and low cost are additional advantages for commercial application

We developed an efficient method to prepare a hybrid inorganic-organic composite based on polyvinyl alcohol (PVA) and polysiloxane using the sol-gel disc technique. Antigen obtained from Yersinia pestis was covalently immobilized onto these discs with glutaraldehyde and used as solid phase in ELISA for antibody detection in serum of rabbits experimentally immunized with plague. Using 1.25 æg antigen per disc, a peroxidase conjugate dilution of 1:4,000 and a serum dilution of 1:200 were adequate for the establishment of the procedure. These values are similar to those used for PVA-glutaraldehyde discs, plasticized filter paper discs and the polyaniline-Dacron composite discs. This procedure is comparable to that which utilizes the adsorption of the antigen to conventional PVC plates, with the amount of antigen being one fourth that employed in conventional PVC plates (5 æg/well). In addition to the performance of the polysiloxane/PVA-glutaraldehyde disc as a matrix for immunodetection, its easy synthesis and low cost are additional advantages for commercial application

F1-antigen purified from Yersinia pestis was covalently linked to 5-mm diameter filter paper discs plasticized with polyvinyl alcohol-glutaraldehyde. These discs were used both for ELISA and dot-ELISA for the detection of anti-F1 IgG in rabbits. The best conditions were achieved using 1.25 µg of F1 antigen/disc, 3 percent w/v skim milk in PBS as blocking agent, anti-IgG peroxidase conjugate diluted 12,000 times, and serum from rabbits immunized or not against Y. pestis, diluted 6,400 times. The absorbance values obtained from the comparative study between this procedure and conventional ELISA were not significantly different but the low cost of the reagents employed in ELISA using the filter paper discs plasticized with polyvinyl alcohol-glutaraldehyde makes this method economically attractive.

Preparaçöes antigenicas soluveis de Schistosoma mansoni adulto (SWAP) fixadas covalentemente em discos de polivinil alcool-glutaraldeido foram usadas em teste de ELISA. As melhores condiçöes para o ensaio foram estabelecidas e foi encontrado que uma quantidade mínima de antigeno, em torno de 1,5 ug e necessária. Comparaçäo entre este procedimento e o de ELISA convencional foi estabelecida. Um metodo seguro de imobilizaçäo do antigeno foi alcançado e os baixos preços dos reagentes empregados säo economicamente atraentes...

Discs of polyvinyl alcohol cross-linked with glutaraldehyde were synthesized under acid catalysis (H2SO4). Then, the antigen F1 purified from Yersinia pestis was covalently linked to this modified polymer. Afterwards, an enzyme-linked immunosorbent assay (ELISA) was established for the diagnosis of plague in rabbit and human. The best conditions for the method were achieved by using 1.3µg of F1 prepared in 0.067 M phosphate buffer, pH 7.2, containing 1 M NaCl (PBS); anti-IgG peroxidase conjugate diluted 6,000 times and as a blocking agent 3 per cent w/v skim milk in PBS. The titration of positive rabbit serum according to this procedure detected antibody concentrations up to 1:12,800 times. The present method, the conventional ELISA and passive haemagglutination assay are compared.
