Purpose: Traumatic brain injury (TBI) remains a major public health problem and cause of death. Ulinastatin (UTI), a serine protease inhibitor, has been reported to have an anti-inflammatory effect and play a role in immunoregulation and organ protection by reducing reactive oxygen species (ROS) production, oxidative stress and inflammation. However, the neuroprotective of UTI in TBI has not been confirmed. Therefore, this study aimed to investigate the neuroprotection and potential molecular mechanisms of UTI in TBI-induced EBI in a C57BL/6 mouse model. Methods: The neurological score and brain water content were evaluated. Enzyme-linked immunosorbent assay was used to detect neuroinflammatory cytokine levels, ROS and malondialdehyde detection to evaluate oxidative stress levels, and TUNEL staining and western blotting to examine neuronal damages and their related mechanisms. Results: Treatment with UTI markedly increased the neurological score; alleviated brain oedema; decreased the inflammatory cytokine tumour necrosis factor a, interleukin-1ß (IL-1ß), IL-6 and nuclear factor kappa B (NF-kB) levels; inhibited oxidative stress; decreased caspase-3 and Bax protein expressions; and increased the Bcl-2 levels, indicating that UTI-mediated inhibition of neuroinflammation, oxidative stress and apoptosis ameliorated neuronal death after TBI. The neuroprotective capacity of UTI is partly dependent on the TLR4/NF-kB/p65 signalling pathway. Conclusions: Therefore, this study reveals that UTI improves neurological outcomes in mice and reduces neuronal death by protecting against neural neuroinflammation, oxidative stress and apoptosis.

Abstract Serine protease inhibitors (serpins), a superfamily of protease inhibitors, are known to be involved in several physiological processes, such as development, metamorphosis, and innate immunity. In our study, a full-length serpin cDNA, designated Haserpin1, was isolated from the cotton bollworm Helicoverpa armigera. The cDNA sequence of Haserpin1 is 1176 nt long, with an open reading frame encoding 391 amino acids; there is one exon and no intron. The predicted molecular weight of Haserpin1 is 43.53 kDa, with an isoelectric point of 4.98. InterProScan was employed for Haserpin1 functional characterization, which revealed that Haserpin1 contains highly conserved signature motifs, including a reactive center loop (RCL) with a hinge region (E341-N350), the serpin signature, (F367-F375) and a predicted P1-P1′ cleavage site (L357-S358), which are useful for identifying serpins. Transcripts of Haserpin1 were constitutively expressed in the fat body, suggesting that it is the major site for serpin synthesis. During the developmental stages, a fluctuation in the expression level of Haserpin1 was observed, with low expression detected at the 5th-instar larval stage. In contrast, relatively high expression was detected at the prepupal stage, suggesting that Haserpin1 might play a critical role at the H. armigera wandering stage. Although the detailed function of this serpin (Haserpin1) needs to be elucidated, our study provides a perspective for the functional investigation of serine protease inhibitor genes.


Resumo Sabe-se que os inibidores de serina protease (serpinas), uma superfamília de inibidores de protease, estão envolvidos em vários processos fisiológicos, como desenvolvimento, metamorfose e imunidade inata. Neste estudo, um cDNA de serpina de comprimento total, denominado Haserpin1, foi isolado da lagarta Helicoverpa armigera na cultura de algodão. A sequência de ADNc de Haserpin1 tem 1.176 nt de comprimento, com uma grelha de leitura aberta que codifica 391 aminoácidos; existe um éxon, mas nenhum íntron. O peso molecular previsto de Haserpin1 é de 43,53 kDa, com um ponto isoelétrico de 4,98. O InterProScan foi empregado para a caracterização funcional do Haserpin1, que revelou que o Haserpin1 contém motivos de assinatura altamente conservados, incluindo um loop central reativo (RCL) com uma região de dobradiça (E341-N350), a assinatura da serpina (F367-F375) e um local de clivagem previsto de P1-P1' (L357-S358), que são úteis para identificar serpinas. As transcrições de Haserpin1 foram expressas constitutivamente no corpo gordo, sugerindo que é o principal local para a síntese de serpinas. Durante os estágios de desenvolvimento, observou-se uma flutuação no nível de expressão de Haserpin1, com baixa expressão detectada no estágio larval do 5º ínstar. Embora a função detalhada dessa serpina (Haserpin1) precise ser elucidada, este estudo fornece uma perspectiva para a investigação funcional dos genes inibidores da serina protease.

Seminal plasma contains serine proteases and serine protease inhibitor, which are involved in mammalian fertilization, and the inhibitors can be applied to prevent cold-induced sperm capacitation. The effects of different concentrations of two serine protease inhibitors were analyzed, Plasminogen activator inhibitor 1 - PAI-1 (70ƞg, 140ƞg and 210 ƞg) and Antipain (10µg, 50µg and 100µg) as supplementation to bovine semen cryopreservation extender. The effects of the inhibitors on the sperm parameters (sperm kinetics - CASA, acrosome integrity, plasma membrane integrity, mitochondrial membrane potential, sperm defects and acrosome reaction rate) were evaluated in the post-thaw semen. Cryopreservation of sperm with Antipain decreased post-thaw kinetic parameters of MP, VSL, LIN, SRT and the percentage of hyper-activated sperm while PAI-1 (210 ƞg) decreased VSL and LIN. Antipain and PAI-1 had no effect on the integrity parameters of the plasma membrane, mitochondrial membrane potential and sperm defects. Sperm cryopreserved in the presence of Antipain and PAI-1 (70 and 140 ƞg) preserved acrosome integrity, as they were able to complete the in vitro acrosome reaction. In conclusion, the serine protease inhibitors, Antipain and PAI-1 (70 and 140ƞg) are able to preserve the acrosome integrity of cryopreserved bovine sperm.(AU)


A criopreservação é parcialmente prejudicial à fertilidade do sêmen de bovinos e induz mudanças semelhantes à capacitação em espermatozoides. O plasma seminal contém serina-proteases e inibidores de serina-proteases que estão envolvidos na fertilização de mamíferos, e os inibidores podem ser aplicados para evitar uma capacitação espermática induzida pelo frio. Analisaram-se os efeitos de diferentes concentrações de dois inibidores de serina-proteases, inibidor do ativador do plasminogênio 1 - PAI-1 (70ƞg, 140ƞg e 210ƞg) e antipaína (10µg, 50µg e 100µg) na suplementação ao diluidor de criopreservação de sêmen bovino. Trinta e seis ejaculados de quatro bovinos Curraleiro Pé-Duro foram usados para criopreservação. Os efeitos dos inibidores sobre os parâmetros dos espermatozoides (cinética espermática - CASA, integridade acrossomal, integridade da membrana plasmática, potencial de membrana mitocondrial, defeitos espermáticos e taxa de reação acrossomal) foram avaliados no sêmen pós-descongelamento. A criopreservação de espermatozoides com antipaína diminuiu os parâmetros cinéticos pós-descongelamento de MP, VSL, LIN, SRT e a porcentagem de espermatozoides hiperativados, PAI-1 (210ƞg) diminuiu VSL e LIN. Antipaína e PAI-1 não tiveram efeitos nos parâmetros de integridade da membrana plasmática, no potencial de membrana mitocondrial e nos defeitos espermáticos. Espermatozoides criopreservados na presença de antipaína e PAI-1 (70 e 140ƞg) preservaram a integridade acrossomal, assim como foram capazes de completar a reação acrossômica in vitro. Em conclusão, os inibidores de serina-proteases, antipaína e PAI-1 (70 e 140ƞg) são capazes de preservar a integridade acrossomal de espermatozoides criopreservados de bovinos.(AU)

Background Toad secretions are a source of molecules with potential biotechnological application on a wide spectrum of diseases. Toads from theRhinella family have two kinds of poisonous glands, namely granular and mucous glands. Rhinella schneideritoads produce granular secretions that comprise a great number of molecules, including serine proteases inhibitors. Serine proteases, such as trypsin, chymotrypsin and elastase, are enzymes that have a serine amino acid into its catalytic site and can be found in a large number of vertebrate species and pathogenic microorganisms. Therefore, the present work aims to purify a serine protease inhibitor from Rhinella schneiderigranular secretions.Findings This study presents the protocol used to purify a serine protease inhibitor from the Rhinella schneideri poison. The granular secretion was submitted to dialysis in order to separate the low molecular weight compounds, which were submitted to a reversed phase-fast protein liquid chromatography fractionation step in a C2C18 column. The major fractions were tested over trypsin, chymotrypsin and elastase through colorimetric assay. The inhibition tests were performed with the enzyme in absence (positive control) and presence of fractions, denatured enzyme (negative control) and the respective chromogenic substrate. Rs20 was the compound with the major inhibitory activity over chymotrypsin, inducing a delay in the formation of the chromogenic enzymatic product. The structure characterization of Rs20 was performed by high resolution electronspray ionization-mass spectrometry (HRESI-MS) and gas chromatography coupled with mass spectrometry (GC-MS). HRESI showed an intense signal suggesting the presence of bufadienolide with less than 10 ppm error. In addition, it was observed a low intense signal at m/z399 that could be lithocholic acid, a biosynthetic precursor of bufadienolide. Finally, GC-MS analysis applying NIST library identification reinforced this hypothesis.Conclusions The current study have isolated and partially characterized the function and structure of the first bufadienolide with inhibitory action over chymotrypsin.(AU)

Ticks are rich sources of serine protease inhibitors, particularly those that prevent blood clotting and inflammatory responses during blood feeding. The tick Rhipicephalus (Boophlus) annulatus is an important ectoparasite of cattle. The aims of this study were to characterize and purify the serine protease inhibitors present in R. (B.) annulatus larval extract. The inhibitors were characterized by means of one and two-dimensional reverse zymography, and purified using affinity chromatography on a trypsin-Sepharose column. The analysis on one and two-dimensional reverse zymography of the larval extract showed trypsin inhibitory activity at between 13 and 40 kDa. Through non-reducing SDS-PAGE and reverse zymography for proteins purified by trypsin-Sepharose affinity chromatography, some protein bands with molecular weights between 13 and 34 kDa were detected. Western blotting showed that five protein bands at 48, 70, 110, 130 and 250 kDa reacted positively with immune serum, whereas there was no positive reaction in the range of 13-40 kDa. Serine protease inhibitors from R. (B.) annulatus have anti-trypsin activity similar to inhibitors belonging to several other hard tick species, thus suggesting that these proteins may be useful as targets in anti-tick vaccines.


Carrapatos são uma rica fonte de inibidores da serina protease, particularmente aqueles que previnem coagulação e respostas inflamatórias durante a alimentação com sangue. O carrapato Rhipicephalus (B.) annulatus é um ectoparasita importante de bovinos. O objetivo deste estudo foi caracterizar e purificar os inibidores da serina protease presentes no extrato de larva do R. (B.) annulatus. Os inibidores foram caracterizados através de zimografia reversa uni e bidimensional e purificados com cromatografia de afinidade em uma coluna de sepharose-tripsina. A análise do extrato de larva pela zimografia reversa uni e bidimensional mostrou atividade inibitória de tripsina entre 13 e 40 kDa. Através de SDS-PAGE e zimografia reversa para proteínas purificadas pela cromatografia por sepharose-tripsina, algumas bandas de proteínas com pesos moleculares entre 13 e 34 kDa foram detectadas. Western blotting mostrou que cinco bandas de proteínas a 48, 70, 110, 130 e 250 kDa reagiram positivamente com o soro imune, enquanto não houve reação positiva nas bandas 13-40 kDa. Inibidores da serina protease do R. (B.) annulatus têm atividade antitripsina semelhante àquelas dos inibidores de outras espécies de carrapatos duros, sugerindo, assim, que essas proteínas podem ser úteis como alvo de vacinas contra carrapatos.

Diabetic retinopathy is one of the most important causes of blindness. The underlying mechanisms of this disease include inflammatory changes and remodeling processes of the extracellular-matrix (ECM) leading to pericyte and vascular endothelial cell damage that affects the retinal circulation. In turn, this causes hypoxia leading to release of vascular endothelial growth factor (VEGF) to induce the angiogenesis process. Alpha-1 antitrypsin (AAT) is the most important circulating inhibitor of serine proteases (SERPIN). Its targets include elastase, plasmin, thrombin, trypsin, chymotrypsin, proteinase 3 (PR-3) and plasminogen activator (PAI). AAT modulates the effect of protease-activated receptors (PARs) during inflammatory responses. Plasma levels of AAT can increase 4-fold during acute inflammation then is so-called acute phase protein (APPs). Individuals with low serum levels of AAT could develop disease in lung, liver and pancreas. AAT is involved in extracellular matrix remodeling and inflammation, particularly migration and chemotaxis of neutrophils. It can also suppress nitric oxide (NO) by nitric oxide sintase (NOS) inhibition. AAT binds their targets in an irreversible way resulting in product degradation. The aim of this review is to focus on the points of contact between multiple factors involved in diabetic retinopathy and AAT resembling pleiotropic effects that might be beneficial.

Sandflies (Diptera: Psychodidae) are important disease vectors of parasites of the genus Leishmania, as well as bacteria and viruses. Following studies of the midgut transcriptome of Phlebotomus papatasi, the principal vector of Leishmania major, two non-classical Kazal-type serine proteinase inhibitors were identified (PpKzl1 and PpKzl2). Analyses of expression profiles indicated that PpKzl1 and PpKzl2 transcripts are both regulated by blood-feeding in the midgut of P. papatasi and are also expressed in males, larva and pupa. We expressed a recombinant PpKzl2 in a mammalian expression system (CHO-S free style cells) that was applied to in vitro studies to assess serine proteinase inhibition. Recombinant PpKzl2 inhibited α-chymotrypsin to 9.4% residual activity and also inhibited α-thrombin and trypsin to 33.5% and 63.9% residual activity, suggesting that native PpKzl2 is an active serine proteinase inhibitor and likely involved in regulating digestive enzymes in the midgut. Early stages of Leishmania are susceptible to killing by digestive proteinases in the sandfly midgut. Thus, characterising serine proteinase inhibitors may provide new targets and strategies to prevent transmission of Leishmania.

O Tripanosoma cruzi, agente etiológico da doença de Chagas, constitui um grave problema desaúde pública que afeta 18 milhões de pessoas na América latina. Para produzir a infecçãoformas tripomastigotas metacíclica de T. cruzi , liberadas durante a picada do inseto vetor,precisam evadir o sistema imune inato do hospedeiro vertebrado para infectar as células. Osistema complemento, devido à sua capacidade para rapidamente reconhecer e eliminarmicrorganismos constitui a principal linha de defesa do sistema imune inato. O sistema docomplemento é composto por vßrias proteínas, ativadas em cascata por serino proteases, queculmina com a formação do complexo de ataque a membrana e lise do patógeno. Todavia,muitos organismos patogênicos desenvolveram formas de escapar do ataque do sistema docomplemento por meio de diversos mecanismos. Visando à melhor caracterização dos fatoresde virulência do T. cruzi, o grupo foi verificar se os sobrenadantes de formas metacíclicasapresentam atividade inibitória de complemento. Devido às vias do complemento seremativadas por serino proteases, foi realizada uma busca no banco genômico de T. cruzi atrßs demotivos de SERPINAS (inibidores de serino protease) que o parasito poderia estar liberandono primeiro contato parasito-hospedeiro. Foram realizados experimentos de lise mediada pelocomplemento com sobrenadantes das formas metacíclicas de T. cruzi. Observou-se, ainda, queos sobrenadantes das formas metacíclicas do T. cruzi são capazes de inibir a lise mediada pelocomplemento de formas epimastigotas...


The Trypanosoma cruzi, etiologic agent of Chagas disease, is a serious public health concernthat affects 18 million people in Latin America. To produce the infection, metacyclictrypomastigotes de T cruzi released during the bite of the insect vector must evade the innateimmune system of the vertebrate host to infect cells. The complement system, due to itsability to rapidly recognize and eliminate microorganisms is the major line of innate immunesystemdefense. The complement system is composed of various proteins, activated by serineproteases cascade that culminates in the formation of the membrane attack complex and lysisof the pathogen. However, many pathogens have developed ways to evade the attack of thecomplement system through several mechanisms. Aiming to better characterize the virulencefactors of T. cruzi, the teamwas to verify whether the supernatants of metacyclic forms exhibitinhibitory activity of complement. Due to the complement pathways are activated by serineproteases, we seek in genomic library of T. cruzi motifs related to serpin (serine proteaseinhibitors), a protein that the parasite could be releasing at the first parasite-host contact.Experiments were carried out to complement-mediated lysis with supernatants of metacyclicforms of T. cruzi. Furthermore, it was observed, that the supernatants of metacyclic forms ofT. cruzi are capable of inhibiting the complement mediated lysis of epimastigotes...

La hiperfiltración glomerular y el aumento de la reabsorción de sodio son factores fundamentales para el desarrollo de la unidad feto placentaria. Dichos factores resultan de adaptaciones hemodinámicas y renales en las que participan sistemas vasoactivos. Se pudo demostrar en ratas que la activación del sistema kallicreína kinina (SKK) precede a la instalación de la hiperfiltración glomerular, dado que su inhibición por aprotinina previene el aumento del filtrado glomerular. Además, la inhibición individual o asociada de los efectores específicos del SKK, las prostaglandinas (PGs) y el óxido nítrico (ON), confirman la dependencia del filtrado glomerular del SKK durante la preñez. Encontramos también que el sistema renina angiotensina (SRA) participa en la generación de la hiperfiltración glomerular dado que ésta es afectada por la administración de bloqueantes del SRA. La inhibición máxima sobre el pico de hiperfiltración se obtuvo con el bloqueo de ambos sistemas (SKK y SRA). Además, estrategias para alterar la hiperfiltración glomerular y la reabsorción de sodio de la preñez evidenciaron alteraciones en el desarrollo de la unidad feto placentaria, menor número de crías, mayor cantidad de reabsorciones intrauterinas y retardo en el crecimiento. El tratamiento combinado de inhibidores del SKK asociados a bloqueantes del SRA o de óxido nítrico mostraron los mayores efectos. En consecuencia, demostramos que el SKK juega un rol central en los fenómenos de adaptación que acompañan la preñez normal. La interrelación del SKK con varios sistemas vasoactivos parecería formar una red que participa en las adaptaciones hemodinámicas para un adecuado desarrollo de la gestación y de la unidad feto-placentaria.


Glomerular hyperfiltration and increased sodium reabsorption are key factors for the development of the fetus and placenta in pregnancy. These adjustments result from hemodynamic and renal factors involving vasoactive systems. It was demonstrated in rats that activation of KKS precedes the installation of glomerular hyperfiltration as aprotinin prevents the increase in glomerular filtration. In addition, individual or associated inhibition of specific kallikrein kinin system effectors, prostaglandins (PGs) and nitric oxide (NO), confirm the glomerular filtration rate dependence of KKS during pregnancy. It was also found that the renin-angiotensin system (RAS) contributes to glomerular hyperfiltration as this is affected by the administration of RAS blockers. The peak of hyperfiltration maximum inhibition was obtained by the blockade of both systems (KKS and RAS). In addition, strategies used to alter the glomerular hyperfiltration and increased sodium reabsorption during pregnancy, showed abnormalities in the development of the fetus and placenta, fewer offspring, more fetus resorptions and intrauterine growth retardation. KKS inhibitors associated with RAS or nitric oxide blockers showed the greatest impact. As a consequence, it was demonstrated that KKS plays a central role in the adaptation phenomenom that accompanies normal pregnancy. The interplay of KKS with several vasoactive systems, seem to arrange a network involved in the hemodynamic adaptations to allow the proper development of pregnancy and the fetus and placenta.

RESUMO

Serpins are a structurally conserved family of macromolecular inhibitors found in numerous biological systems. The completion and annotation of the genomes of Schistosoma mansoni and Schistosoma japonicum has enabled the identification by phylogenetic analysis of two major serpin clades. S. mansoni shows a greater multiplicity of serpin genes, perhaps reflecting adaptation to infection of a human host. Putative targets of schistosome serpins can be predicted from the sequence of the reactive center loop (RCL). Schistosome serpins may play important roles in both post-translational regulation of schistosome-derived proteases, as well as parasite defense mechanisms against the action of host proteases.


Serpinas são uma família de inibidores macromoleculares estruturalmente conservados encontrados em inúmeros sistemas biológicos. O término e a anotação dos genomas de Schistosoma mansoni e de Schistosoma japonicum permitiram a identificação por análise filogenética de dois principais clados de serpinas. S. mansoni mostra uma multiplicidade maior de genes de serpinas, talvez refletindo uma adaptação à infecção de um hospedeiro humano. Alvos putativos das serpinas de esquistossomos podem ser preditos a partir da sequência do "loop" do centro reativo. Serpinas de esquistossomos podem ter importantes papeis tanto na regulação pós-traducional de proteases derivadas do esquistossoma, quanto nos mecanismos de defesa contra a ação de proteases do hospedeiro.

OBJETIVO: Avaliar se a aprotinina em altas doses hemostáticas pode reduzir o processo inflamatório após circulação extracorpórea (CEC) em crianças. MÉTODOS: Estudo prospectivo randomizado em crianças de 30 dias a 4 anos de idade, submetidas à correção de cardiopatia congênita acianogênica, com CEC e divididas em dois grupos, um denominado Controle (n=9) e o outro, Aprotinina (n=10). Neste, o fármaco foi administrado antes e durante a CEC. A resposta inflamatória sistêmica e disfunções hemostática e multiorgânicas foram analisadas por marcadores clínicos e bioquímicos. Foram consideradas significantes as diferenças com P<0,05. RESULTADOS: Os grupos foram semelhantes quanto às variáveis demográficas e intra-operatórias, exceto por maior hemodiluição no Grupo Aprotinina. Não houve benefício quanto aos tempos de ventilação pulmonar mecânica, permanência no CTIP e hospitalar, nem quanto ao uso de inotrópicos e função renal. A relação PaO2/FiO2 (pressão parcial de oxigênio arterial/fração inspirada de oxigênio) apresentou queda significativa com 24 h pós-operatório, no Grupo Controle. As perdas sanguíneas foram semelhantes nos dois grupos. No grupo Aprotinina surgiu leucopenia significativa, em CEC, seguida de leucocitose. Fator de necrose tumoral alfa (TNF-α), Interleucinas (IL)-6, IL-8, IL-10, proporção IL-6/IL-10 não apresentaram diferenças marcantes intergrupos. A proporção IL-6/IL-10 PO aumentou no grupo Controle. Não houve complicações com o uso da aprotinina. CONCLUSÃO: Nesta casuística, a Aprotinina em altas doses hemostáticas não minimizou as manifestações clínicas e os marcadores séricos de resposta inflamatória sistêmica.


OBJECTIVE: To evaluate if the hemostatic high-dose aprotinin seems to reduce the inflammatory process after extracorporeal circulation (ECC) in children. METHODS: A prospective randomized study was conducted on children aged 30 days to 4 years submitted to correction of acyanogenic congenital heart disease with ECC and divided into two groups: Control (n=9) and Aprotinin (n=10). In the Aprotinin Group the drug was administered before and during ECC and the systemic inflammatory response and hemostatic and multiorgan dysfunctions were analyzed on the basis of clinical and biochemical markers. Differences were considered to be significant when P<0.05. RESULTS: The groups were similar regarding demographic and intraoperative variables, except for a greater hemodilution in the Aprotinin Group. The drug had no benefit regarding time of mechanical pulmonary ventilation, permanence in the postoperative ICU and length of CONCLUSION: In this series, hemostatic high-dose aprotinin did not minimize the clinical manifestations or serum markers of the inflammatory systemic response.

The objective of the present study was to determine the efficacy of prophylactic administration of gabexate for the prevention of post-endoscopic retrograde cholangiopancreatography (ERCP) pancreatitis, hyperamylasemia and pancreatic pain. Patients scheduled for ERCP were randomized into two groups in a double-blind manner: the patients in the gabexate group were treated with continuous intravenous infusion of 300 mg gabexate dissolved in 500 mL Ringer's solution at 111 mL/h, starting 30 min before the endoscopic maneuvers and continuing up to 4 h after them; placebo group patients were treated only with Ringer's solution also starting 30 min before the endoscopic maneuvers and continuing up to 4 h. Data for 193 patients were analyzed. The incidence of post-ERCP pancreatitis was 3 patients (3.1 percent) in the gabexate group and 10 (10.5 percent) in the placebo group (P = 0.040). The incidence of hyperamylasemia was 33 patients (33.7 percent) in the gabexate group and 42 (43.7 percent) in the placebo group (P = 0.133). The incidence of pancreatic pain was 15 patients (15.3 percent) in the gabexate group and 28 (29.5 percent) in the placebo group (P = 0.018). The results suggest that a 4.5-h infusion of gabexate (for a total of 300 mg) could prevent post-ERCP pancreatitis and pancreatic pain.

Como primeiro objetivo desse projeto decidimos realizar a expressão do inibidor recombinante BmTIsint, inibidor sintético construído durante o período de mestrado. BmTIsint tem como base a seqüência do primeiro domínio, Kunitz-BPTI, do inibidor BmTI¬ A. BmTIsint foi expresso em sistema Pichia pastoris, apresentou inibição sobre tripsina e HuPK, Ki de 3,3 nM e 11 nM, respectivamente. Os dados cinéticos sugerem que a atividade inibitória para HuPK está presente no primeiro domínio do inibidor BmTI-A. BmTIsint apresentou uma outra atividade inibitória interessante, a inibição de catepsina L humana (Ki= 100 nM), atividade que foi determinada pela primeira vez para BPTI (Ki= 500 nM). Neste projeto realizamos também a clonagem e expressão do inibidor rBmTI-6 (inibidor de tripsina contendo 3 domínios Kunitz-BPTI). A expressão do gene BmTI6 foi identificada em ovário e corpos gordurosos de B. microplus. O inibidor rBmTI-6 expresso em Pichia pastoris apresentou constante de dissociação (Ki) sobre tripsina igual a 1,7 nM. rBrnTI-6 foi glicosilado em pelo menos 5 sítios de N-glicosilação da proteína e sofreu processamento dividindo-se em duas cadeias protéicas, uma em tomo de 8 kDa e a outra em tomo de 26kDa. Em experimentos de western-blotting, anti-soros de bovinos de regiões com alta infestação de carrapatos reconheceram bandas rBmTI-6, anti-soros de animais imunizados com o "pool" de inibidores de tripsina presentes em larvas de B. microplus (BmTIs) reconheceram intensamente a mesma amostra de rBmTI-6. O silenciamento gênico por interferência de RNA utilizando dsRNAs do gene BmTI6, inoculado em teleóginas ingurgitadas, mostrou redução de 16 por cento a 25 por cento em relação às controles, assim como a atividade inibitória sobre tripsina dos extratos de larvas e ovos de teleóginas silenciadas. Paralelamente ao trabalho com os BmTIs, neste projeto objetivamos identificar outros inibidores presentes em tecidos do carrapato B. microplus...

Animais hematófagos são vetores responsáveis pela transmissão de vários patógenos e parasitas para animais vertebrados. A mosca-dos-chifres Haematobia irritans irritans é um dos ectoparasitas responsáveis por grandes perdas na produção de bovinos no Brasil e em outras partes do mundo, devido ao seu hábito hematofágico. Este trabalho tem como objetivos: a purificação e caracterização do inibidor de serinoproteases presentes em cabeça e tórax da H. i. irritans. O inibidor HiTI (Haematobia irritans irritans trypsin inhibitor) purificado por cromatografia de afinidade em tripsina . Sepharose apresentou atividade inibitória para tripsina bovina, elastase de neutrófilos humanos e a enzima tipo-tripsina do abdome da mosca H. i. irritans com constante de dissociação (Ki) de 0,57 nM, 1,3 nM e 0,20 nM, respectivamente. O HiTI purificado apresentou massa molecular de 7029 Da por espectrometria de massa (MALDI-TOF) e seqüência N-terminal que sugeria a sua inclusão na família de inibidores do tipo Kunitz. O fragmento de cDNA de HiTI foi clonado e seqüenciado. A seqüência de aminoácidos traduzida da seqüência de nucleotídeos confirmou a classificação do HiTI na família tipo Kunitz e mostrou alta similaridade com um inibidor do tipo Kunitz isolado de hemolinfa de mosca Sarcophaga bullata. O HiTI recombinante (rHiTI) produzido em sistema de expressão utilizando levedura Pichia pastoris apresentou níveis de expressão de 4,2 mg de proteína ativa por litro de cultura. O rHiTI purificado em cromatografia de afinidade tripsina-Sepharose apresentou massa molecular de 6532 Da, determinada por espectrometria de massa LC-ESI e constantes de dissociação (Ki) similares às obtidas para HiTI nativo para tripsina bovina e elastase de neutrófilos humanos de 0,4 nM e 1,0 nM, ...(au)

Here we report the subcellular localization of an intracellular serine protease of 68 kDa in axenic promastigotes of Leishmania (Leishmania) amazonensis, using subcellular fractionation, enzymatic assays, immunoblotting, and immunocytochemistry. All fractions were evaluated by transmission electron microscopy and the serine protease activity was measured during the cell fractionation procedure using a-N-r-tosyl-L-arginine methyl ester (L-TAME) as substrate, phenylmethylsulphone fluoride (PMSF) and L-1-tosylamino-2-phenylethylchloromethylketone (TPCK) as specific inhibitors. The enzymatic activity was detected mainly in a membranous vesicular fraction (6.5-fold enrichment relative to the whole homogenate), but also in a crude plasma membrane fraction (2.0-fold). Analysis by SDS-PAGE gelatin under reducing conditions demonstrated that the major proteolytic activity was found in a 68 kDa protein in all fractions studied. A protein with identical molecular weight was also recognized in immunoblots by a polyclonal antibody against serine protease (anti-SP), with higher immunoreactivity in the vesicular fraction. Electron microscopic immunolocalization using the same polyclonal antibody showed the enzyme present at the cell surface, as well as in cytoplasmic membranous compartments of the parasite. Our findings indicate that the internal location of this serine protease in L. amazonensis is mainly restricted to the membranes of intracellular compartments resembling endocytic/exocytic elements.

Animais hematófagos possuem uma necessidade vital em manter o sangue fluido, durante a aquisição, o armazenamento e digestão do alimento. Para tal, esses animais desenvolveram ao longo de seus processos evolutivos mecanismos para interferir na hemostasia dos seus hospedeiros. Foi identificado anteriormente no barbeiro Triatoma infestans vetor da doença de Chagas, um potente inibidor de trombina denominado infestina 1-2 esta proteína é codificada por um gene composto por sete domínios tipo Kazal, que possuem isoladamente diferentes atividades inibitórias para as serinoproteases plasmáticas. Esta proteína é provavelmente processada em inibidores menores compostos por 1 ou 2 domínios após a sua tradução por um mecanismo ainda desconhecido. Neste trabalho, identificou-se a presença de um potente inibidor de fator XIIa no estômago do barbeiro T. infestans correspondente aos domínios 3 e 4 do gene da infestina, confirmando a atividade inibitória sobre fator XIIa caracterizada na infestina 34 recombinante. Determinou-se ainda a estrutura tridimensional do domínio 1 de infestina em complexo com tripsina bovina a uma resolução de 2,5A. Foram analisadas as interações intramoleculares da infestina 1 responsáveis pela manutenção da estrutura do inibidor e também as interações entre as moléculas de infestina 1 e a tripsina. Adicionalmente, a infestina 4 foi clonada, expressa e a mesma apresentou características cinéticas semelhantes ao inibidor de fator XIIa nativo, inibindo não apenas fator XIIa (128 pM) mas também fator Xa (53 nM), plasmina (2,1 nM) e tripsina (11 nM). Finalmente, determinou-se a estrutura do domínio 4 da infestina a uma resolução de 1,8A ,1,73A e 1,4A. E a mesma apresentou as mesmas características estruturais de outros inibidores da família Kazal

Inibidores do tipo Kunitz de plantas são proteínas encontradas principalmente na família de leguminosas. Os inibidores de Bauhinia foram caracterizados de diversas espécies. Esses inibidores de Bauhinia apresentam uma diferença principal quando comparados aos outros inibidores tipo Kunitz pois estes são destituídos de pontes dissulfeto (Oliveira et a1.,2001; Oliva et al., 2001). Dois inibidores foram isolados das sementes de Bauhinia bauhinioides: o Inibidor de Calicreína Plasmática de B. bauhinioides (BbKI) e Inibidor de Cruzaína de B. bauhinioides (BbCI). 0 BbKI inibe tripsina (Ki = 20 nM), calicreína plasmática (Ki = 0.39 nM), calicreína tecidual (Ki = 200 nM), plasmina (Ki = 33,17 nM) e quimotripsina (Ki = 3.90 pM). BbCI inibe elastase pancreática porcina (Ki 2.5 nM), elastase de neutrófilos humano (Ki 5.3 nM), catepsina G (Ki 160 nM) exceto enzimas anticoagulantes, tripsina e quimotripsina. 0 sítio reativo de BbCI é VRFETPLAIAIITESFFL sendo Ala o aminoácido da posição P1, ao passo que, o sítio reativo de BbKI é VRFESPLRINIIKSYFL tendo Arg na posição P1. BbCI inibe elastase de neutrófilo (Ki = 5.3 nM) e catepsina G (Ki = 160 nM) e PPE (40 nM). A ação desses inibidores foi estudada em modelos de inflamação como edema de pata e pleurisia. Ambos inibidores reduzem o edema da pata de rato induzido por carragenina. 0 BbKI reduziu o edema de pata na primeira (20 por cento) e segunda hora (31 por cento) após sua administração, indicando um bloqueio da liberação de cinina, enquanto a redução do edema pelo BbCI indica interferência sobre os mediadores inflamatórios (cininas e prostaglandinas) da segunda a quarta horas (24, 44 e 40 por cento, respectivamente). No modelo de pleurisia, tanto BbKI como BbCI promovem uma diminuição significante da migração celular (BbKI 35 por cento e BbCl 45 por cento), confirmado por microscopia intravital. Deste modo, inibidores de B. bauhinioides podem ser ferramentas úteis nos estudos dos processos inflamatórios

There are few reports concerning the biological role and the mechanisms of interaction between proteinases and carbohydrates other than those involved in clotting. It has been shown that the interplay of enzymes and glycosaminoglycans is able to modulate the activity of different proteases and also to affect their structures. From the large number of proteases belonging to the well-known protease families and also the variety of carbohydrates described as widely distributed, only few events have been analyzed more deeply. The term "family" is used to describe a group of proteases in which every member shows an evolutionary relationship to at least one other protease. This relationship may be evident throughout the entire sequence, or at least in that part of the sequence responsible for catalytic activity. The majority of proteases belong to the serine, cysteine, aspartic or metalloprotease families. By considering the existing limited proteolysis process, in addition to the initial idea that the proteinases participate only in digestive processes, it is possible to conclude that the function of the enzymes is strictly limited to the cleavage of intended substrates since the destruction of functional proteins would result in normal tissue damage. In addition, the location as well as the eventual regulation of protease activity promoted by glycosaminoglycans can play an essential role in the development of several physiopathological conditions
