In Brazil, in view of the different soil and climatic conditions of the sugarcane-producing regions, a strategy of creating small breeding programs for the crop is being used. Since 2008, the sugarcane breeding program of the Federal University of Santa Maria (UFSM), has been developing new, better adapted and higher-yielding sugarcane cultivars for the climate conditions of the southern region of Brazil. Cultivar UFSM XIKA FW was developed at the Laboratories of Agroclimatology and Genetic Breeding and Plant Production of the UFSM, Campus Frederico Westphalen. It is the first mutation-induced sugarcane cultivar that was protected by the Ministry of Agriculture, Livestock and Food Supply, Brazil. The genotype used for mutation induction was cultivar IAC 87-3396, due to its good performance characteristics in the southern region. The mutation mechanism was induced in 2011, by immersing individual buds in a solution of the chemical mutagenic agent methyl methane sulfonate. The best genotypes were selected and tested in plant cane in 2013 and 2018 and in ratoon cane trials in 2014 and 2015. The new cultivar has a higher sugar production potential, and the mean yield of four years was 8.2 t ha-1 higher than that of the control, indicating a high yield potential.

Flavoring additives are of great technological importance for the food industry. However, there is little information regarding the toxicological properties of these micro-ingredients, especially at the cellular level. The present study used meristematic root cells of Allium cepa L. to evaluate the toxicity of a liquid, aroma and flavor synthetic chocolate additive, manufactured and widely marketed throughout Brazil and exported to other countries in South America. The flavoring concentrations evaluated were 100.00; 50.00; 25.00; 1.00; 0.50 and 0.25 µL/L, where the highest concentration established was one-hundred times lower than that commercially suggested for use. The concentration 100 µL/L substantially reduced cell division of meristems within 24- and 48-hours exposure. Concentrations from 100.00 to 0.50 µL/L resulted in a significant number of prophases to the detriment of the other phases of cell division, indicating an aneugenic activity, and induced a significant number of cellular changes, with emphasis on micronuclei, nuclear buds and chromosomal breaks. Under the established analysis conditions, with the exception of concentration 0.25 µL/L, the flavoring of chocolate caused cytotoxicity, genotoxicity and mutagenicity to root meristems.


Os aditivos aromatizantes têm grande importância tecnológica para a indústria de alimentos. Contudo, poucas são as informações quanto as propriedades toxicológicas desses microingredientes, especialmente, em nível celular. No presente estudo avaliou-se, sobre as células meristemáticas de raízes de Allium cepa L., a toxicidade de um aditivo sintético líquido de aroma e sabor de chocolate, fabricado e amplamente comercializado em todo Brasil, e exportado para outros países da América do Sul. As concentrações de aromatizante avaliadas foram 100,00; 50,00; 25,00; 1,00; 0,50 e 0,25 µL/L, onde a maior concentração estabelecida foi cem vezes menor que a sugerida comercialmente para uso. Com base na interpretação dos resultados, a concentração 100 µL/L reduziu substancialmente a divisão celular dos meristemas nas 24 e 48 horas de exposição. As concentrações 100,00 a 0,50 µL/L demonstraram número significativo de prófases em detrimento as outras fases da divisão celular, indicando ação aneugênica, e induziram número significativo de alterações celulares, com ênfase a micronúcleos, broto nucleares e quebras cromossômicas. Nas condições de análises estabelecidas, com exceção a concentração 0,25 µL/L, o aromatizante de chocolate causou citotoxicidade, genotoxicidade e mutagenicidade aos meristemas radiculares.

Amphibians are excellent bioindicators because they are sensitive to chemical pollution and can indicate ecosystem changes due to the presence of or exposure to chemical compounds. Here we report evidence of the impact of herbicides, including glyphosate, on amphibians in a locality in the central Brazilian Amazon and compare it with data from other biomes in Brazil. We observed malformations in three species of Leptodactylus and local extinctions of Scinax ruber and Rhinella marina from reproductive sites close to an area where herbicides had been applied. The observations in the Amazon are similar to reports from Brazils Atlantic forest on morphological anomalies and mortality in amphibians exposed to herbicides. We warn of the threat of expanding crops for the production of biofuels in the Amazon due to their cultivation being associated with agrochemicals, including glyphosate, posing a threat to the biodiversity of the Amazon biome.


Os anfíbios são ótimos bioindicadores porque são sensíveis à poluição química, indicando alterações ecossistêmicas pela presença de ou pela exposição a compostos químicos. Aqui nós relatamos evidências do impacto de herbicidas, incluindo glifosato, em anfíbios em uma localidade na Amazônia central brasileira e as comparamos com dados de outros biomas no Brasil. Nós observamos malformações em três espécies de Leptodactylus e extinção local de Scinax ruber e Rhinella marina em sítios reprodutivos próximos a uma área onde herbicidas haviam sido aplicados. Os resultados observados na Amazônia são semelhantes aos observados na Mata Atlântica do Brasil, com alterações morfológicas e mortalidade em anfíbios expostos a herbicidas. Alertamos para a ameaça de expansão de culturas para a produção de biocombustíveis na Amazônia, devido ao fato de o cultivo estar associado a agroquímicos, incluindo o glifosato, representando uma ameaça à biodiversidade do bioma Amazônia.

Abstract This research aimed to conduct a systematic review and metanalysis to compare the frequency of cell damage in crack users and nonusers, through Micronucleous (MN) test in buccal mucosa cells. A comprehensive search was carried out on MEDLINE via PubMeb, Web of Science, LILACS and the grey literature without restrictions. It was included case-control studies that report the frequency of micronuclei in the oral mucosa of adult crack users and nonusers. A review protocol was registered with PROSPERO (CRD42018115672), and conducted in accordance with the PRISMA guidelines for the report of this systematic review. Furthermore, study quality was evaluated using an adapted Newcastle-Ottawa Scale for cross-sectional studies.The original search yielded 27 references, after eligibility criteriaonly five articles were included. The number of micronuclei was higher in crack users compared to nonusers. Also, secondary outcomes: binucleated cells, nuclear buds, pyknosis, karyorrhexis and karyolysis had higher prevalence in crack users.Crack use is associated with genotoxic and mutagenic effects because there is a higher frequency of micronuclei in exfoliated cells of crack users. In addition, MN test proved to be a goodbiomarker to assess the mutagenic impact of crack use in oral epithelium.


Resumen Esta investigación tuvo como objetivo realizar una revisión sistemática y un meta-análisis para comparar la frecuencia de daño celular en usuarios de crack y sin crack, a través de la prueba de micronúcleos (MN) en células de la mucosa bucal. Se realizó una búsqueda exhaustiva en MEDLINE a través de PubMeb, Web of Science, LILACS y la literatura gris sin restricciones. Se incluyeron estudios de casos y controles que informaron la frecuencia de micronúcleos en la mucosa oral de usuarios adultos de crack y sin crack. Se registró un protocolo de revisión con PROSPERO (CRD42018115672), y se realizó de acuerdo con las pautas de PRISMA para el informe de esta revisión sistemática. Además, la calidad del estudio se evaluó mediante una escala Newcastle-Ottawa adaptada para estudios transversales. La búsqueda original arrojó 27 referencias, después de los criterios de elegibilidad se incluyeron un total de cinco artículos. El número de micronúcleos fue mayor en los usuarios de crack en compa ración con los usuarios sin crack. Además, los resultados secundarios de células binucleadas, yemas nucleares, picnosis, cario- rrexis y cariólisis tuvieron una mayor prevalencia en los usuarios de crack. El uso de crack se asocia con efectos genotóxicos y mutagénicos porque hay una mayor frecuencia de micronúcleos en las células exfoliadas de los usuarios de crack. Además, la prueba de MN demostró ser un buen biomarcador para evaluar el impacto mutagénico del uso de crack en el epitelio oral.

Resumo O agrotóxico malathion vem sendo amplamente utilizado no mundo em programas de controle de arboviroses e em 2015 foi classificado pela Agência Internacional para Pesquisas em Câncer (IARC) como provável agente carcinogênico para seres humanos. Este trabalho objetivou a sistematização das evidências dos efeitos carcinogênicos e mutagênicos associados à exposição do malathion e seus análogos, malaoxon e isomalathion. A busca foi realizada nas bases de dados TOXLINE, PUBMED e SCOPUS por artigos originais publicados de 1983 a 2015. Do total de 273 artigos elegíveis, foram selecionados 73. Os resultados dos estudos in vitro e in vivo evidenciaram danos genéticos e cromossômicos provocados pelo malathion; os estudos epidemiológicos evidenciaram associações significativamente positivas para cânceres de tireóide, de mama, e ovariano em mulheres na menopausa. Estas evidências do efeito carcinogênico do malathion devem ser considerados diante de sua utilização em programas de controle de arboviroses.


Abstract Malathion has been widely used worldwide in arbovirus control programs. In 2015, it was classified by the International Agency for Research on Cancer (IARC) as a probable carcinogen to humans. This work aimed to systematize the evidence of the carcinogenic and mutagenic effects associated with the exposure of malathion and its analogs, malaoxon and isomalathion. The search was carried out in Toxline, PubMed and Scopus databases for original papers published from 1983 to 2015. In all, 73 papers were selected from a total of 273 eligible papers. The results of in vitro and in vivo studies showed mainly genetic and chromosomal damages caused by malathion. The epidemiological studies evidenced significant positive associations for thyroid, breast, and ovarian cancers in menopausal women. This evidence of the carcinogenic effect of malathion should be considered before its use in arbovirus control programs.

Resumen Introducción. La exposición a solventes orgánicos y pinturas se ha asociado con efectos genotóxicos y mayor riesgo de neoplasias. Sin embargo, aún no se ha caracterizado bien el tipo de daño que esta exposición induce en el ADN humano, ni los mecanismos por los cuales se genera. Uno de los grupos con mayor exposición a dichos solventes y pinturas son los pintores de automóviles del sector informal que trabajan sin adecuadas prácticas de seguridad ocupacional. Objetivo. Determinar el daño oxidativo y por metilación del ADN de linfocitos de pintores de automóviles expuestos a solventes orgánicos y pinturas. Materiales y métodos. Se analizaron linfocitos aislados de sangre periférica de 62 pintores y 62 sujetos no expuestos mediante el ensayo cometa de gran eficiencia acoplado a las enzimas Fpg y AlkA. Las categorías de daño en el ADN evaluadas fueron el daño basal (sin enzimas), el daño oxidativo y el daño por metilación, y el parámetro de medición, el porcentaje de ADN en la cola. Resultados. El porcentaje de ADN en la cola fue mayor en el grupo expuesto con respecto al no expuesto (p<0,05). En el grupo expuesto, dicho porcentaje fue mayor en la categoría de daño oxidativo comparado con la del basal (16,50 Vs. 12,87; p<0,001), en tanto que en el daño por metilación no se encontraron diferencias significativas (14,00 Vs. 12,87; p>0,05). Conclusión. La exposición a solventes orgánicos y pinturas se asoció con el aumento de las lesiones oxidativas del ADN de los linfocitos de pintores de automóviles, tales como la producción de 8-oxo-2'-desoxiguanosina (8-oxodG) y otros productos formamidopirimidina, los cuales se consideran considerablemente mutagénicos.


Abstract Introduction: The exposure to organic solvents and paints has been associated with genotoxicity and a greater risk of neoplasms. However, the type of DNA damage induced in humans by the exposure to these compounds, which would help explain the mechanisms of their genotoxicity, is still not fully characterized. Due to inadequate practices of occupational safety, car painters in the informal sector are a highly exposed group to organic solvents and paints. Objective: To identify the oxidative and methylating damage in the DNA of lymphocytes of car painters exposed to organic solvents and paints. Materials and methods: Isolated peripheral blood lymphocytes from 62 painters and 62 unexposed subjects were analyzed by the modified high-throughput comet assay with the Fpg and AlkA enzymes. The categories used for the evaluation of the DNA damage were basal damage (without enzymes), oxidative and methylating damage. The measurement parameter used to establish the damage was the percentage of DNA in the tail. Results: The percentage of DNA in the tail was higher in the exposed group compared to the unexposed group (p<0.05). In the exposed group, this percentage was higher in the oxidative damage category than the baseline (16.50 vs. 12.87; p<0.001), whereas methylating damage did not show significant differences (14.00 vs. 12.87; p>0.05). Conclusion: In this study, exposure to organic solvents and paints was associated with an increase in oxidative lesions in the DNA of car painters' lymphocytes, such as the production of 8-oxodG and other formamidopyrimidine products which are considered highly mutagenic.

Pyrostegia venusta is usually found in the secondary growth of the Atlantic forests, and in the Brazilian Savanna. Flowers and leaves of this plant are used in folk remedies for treating a wide variety of healthy conditions, this way is important evaluate its safety and antioxidant potential for this applications. For this, was made a ethanolic extract from its flowers and analyzed with toxicological,genotoxicity and antioxidant tests, the toxicological analysis was made by reproductive toxicity in rats and clatogenicity/aneugenicity in human lymphocytes. The genotoxicity was studied by micronucleus test mice bone marrow. The antimutagenic test in root cells of Allium cepa, the antioxidant assays used was DPPH, FRAP, Lipid Perxidation and REM, beyond of that the extract was analyzed in HPLC showing the profile of its compounds. The toxicological analysis showed that P. venusta has no negative significant effect on reproductive and cellular level. The micronucleus test in mouse bone marrow, the extract protected cells from cyclophosphamide, mutagenic compound, in a similar way. The A. cepa test showed that the extract reduced chromosomal disorders formations. The antioxidant activity of extract was significant, except in REM test. The phytochemical analysis showed the presence of flavonoids compounds. P. venusta extract does not present reproductive toxicity and genotoxic effects. However, the extract of this species showed antigenotoxic and antioxidant potential, possibly due to the different flavonoid compounds present in its extract.


Pyrostegia venusta é geralmente encontrada no crescimento secundário das florestas atlânticas e na savana brasileira. Flores e folhas desta planta são utilizadas em remédios populares para tratar uma grande variedade de doenças, desta forma é importante avaliar a segurança e o potencial antioxidante para estas aplicações. Para tanto, o extrato etanólico das flores foi avaliado com testes toxicológicos, genotóxicos e antioxidants. A análise toxicológica foi realizada por meio da toxicidade reprodutiva em ratos e a clatogenicidade/aneugenicidade em linfócitos humanos, a genotoxicidade foi estudada por teste de micronúcleo em medula óssea de camundongo. A antimutagenicidade em células da raiz de Allium cepa. Os ensaios antioxidantes utilizados foram DPPH, FRAP, TARBS e MRE. O extrato foi analisado em HPLC. A análise toxicológica reprodutiva mostrou que P. venusta não tem efeito negativo sobre o nível reprodutivo e cellular. No teste do micronúcleo o extrato protegeu as células da ciclofosfamida, um composto mutagênico. O teste de A. cepa mostrou que o extrato reduziu as formações dos distúrbios cromossômicos. A atividade antioxidante do extrato foi significativa, exceto no teste REM. A análise fitoquímica mostrou a presença de compostos flavonoídicos. O extrato de P. venusta não apresenta toxicidade reprodutiva e efeitos genotóxicos. No entanto, o extrato desta espécie apresentou potencial antigenotóxico e antioxidante, possivelmente devido aos diferentes compostos flavonoídicos presentes em seu extrato.

Abstract The goal of this study was to analyze cytotoxicity, genotoxicity and mutagenicity to bone marrow cells of mice of nature identical synthetic flavorings, passion fruit and strawberry, and artificial synthetic flavorings, vanilla, chocolate, tutti-frutti and cookie, at doses 0.5; 1.0; 2.0; 5.0 and 10.0 mL/kg. The additives were given to the animals by gavage in a single daily application for seven days. Data were subjected to analysis of variance (ANOVA) followed by post Tukey's post hoc test, p <0.05. Animals treated with 2.0; 5.0 and 10.0 mL/Kg of flavorings chocolate, strawberry and cookie, and 5.0 and 10.0 mL/Kg of flavorings vanilla and passion fruit died on the fifth and sixth day of the experiment, respectively. The doses 0.5 and 1.0 mL/Kg of the six additives significantly reduced erythropoiesis in the examined tissue. Also, treatments 0.5 and 1.0 mL/Kg of chocolate, and 1.0 mL/Kg of strawberry and biscuit induced the formation of micronuclei in the bone marrow erythrocytes, at a significant frequency. Therefore, under the study conditions, the six microingredients analyzed were cytotoxic and genotoxic, and additives strawberry, chocolate and cookie were also mutagenic in at least one of the evaluated doses.


Resumo Os aromatizantes são essenciais para a indústria na confecção de alimentos industrializados. Porém, pouco se sabe sobre o potencial tóxico desses microingredientes alimentares. Dessa forma, objetivou-se neste trabalho analisar, em células de medula óssea de camundongos, a citotoxicidade, genotoxicidade e mutagenicidade de aromatizantes alimentares sintéticos idênticos ao natural, de maracujá e morango, e artificiais, de baunilha, chocolate, tutti-frutti e biscoito, nas doses 0,5; 1,0; 2,0; 5,0 e 10,0 mL/Kg. Os aditivos foram administrados aos animais via gavagem em aplicação diária única durante sete dias. Os dados obtidos foram submetidos ao procedimento estatístico ANOVA com pós teste de Tukey, com p < 0.05. Os animais tratados com 2,0; 5,0 e 10,0 mL/Kg dos aromatizantes de chocolate, morango e biscoito, e 5,0 e 10,0 mL/Kg dos aromatizantes de baunilha e maracujá vieram a óbito no quinto e sexto dia de experimento, respectivamente. As doses 0,5 e 1,0 mL/Kg dos seis aditivos reduziram significativamente a eritropoiese do tecido analisado. Ainda, os tratamentos 0,5 e 1,0 mL/kg de chocolate, e 1,0 mL/Kg de morango e biscoito induziram a formação de micronúcleos aos eritrócitos de medula em frequência significante. Portanto, nas condições de estudo estabelecidas, os seis microingredientes analisados foram citotóxico e genotóxicos, e os aditivos de morango, chocolate e biscoito também foram mutagênicos em pelo menos uma das doses avaliadas.

Abstract Salacia crassifolia (Mart. Ex. Schult.) G. Don. is a bush which belongs to Celastraceae family and occurs specially in Brazilian Cerrado. Its leaves, stem, seeds and fruits are popularly used for several medicinal purposes, such as antitumoral, antirheumatic, anti-inflammatory and antimicrobial. In this study, the mutagenic and antimutagenic activities of S. crassifolia stem bark fractions (hexane, ethyl acetate and hydroalcoholic) were evaluated by the Ames mutagenicity assay in Salmonella typhimurium TA98 and TA100 strains. By the obtained results, all S. crassifolia fractions did not significantly increase the number of prototrophic revertants for histidine (His+) in both S. typhimurium strains tested (p > 0.05), suggesting absence of mutagenicity. Regarding antimutagenicity, the fractions ethyl acetate and hydroalcoholic significantly decreased the number of His+ revertants colonies induced by positive control for strain TA98 (p < 0.05), demonstrating protection against mutagenicity induced by 4-nitroquinolile1-oxide, whereas the hexane fraction did not show antimutagenic effect in this strain. In the TA100 strain, all fractions of S. crassifolia protected DNA against the harmful action of sodium azide, and the hexane fraction exhibited the greatest protection in this work. Thus, it's possible conclude that the fractions of S. crassifolia tested in this study could be used in chemoprevention.


Resumo Salacia crassifolia (Mart. Ex. Schult.) G. Don. é uma árvore que pertence à família Celastraceae e ocorre especialmente no Cerrado Brasileiro. Suas folhas, caule, sementes e frutos são popularmente utilizados para vários fins medicinais, tais como antitumoral, antirreumático, anti-inflamatório e antimicrobiano. Neste estudo, nós avaliamos as atividades mutagênica e antimutagênica de frações da casca do caule de S. crassifolia (hexânica, acetato de etila e hidroalcoólica) pelo ensaio de mutagenicidade de Ames em Salmonella typhimurium, cepas TA98 e TA100. Pelos resultados obtidos todas as frações de S. crassifolia não aumentaram significativamente o número de revertentes prototróficas para histidina (His+) em ambas as cepas de S. typhimurium testadas (p > 0.05), sugerindo ausência de mutagenicidade. Em relação à antimutagenicidade, as frações acetate de etila e hidroalcoólica reduziram significativamente o número de colônias revertentes His+ induzidas pelo controle positive para a cepa TA98 (p < 0.05), demonstrando sua ação protetora contra a mutagenicidade induzida por 4-nitroquinolile1-oxide, enquanto a fração hexânica não demonstrou efeito antimutagênico nesta cepa. Na cepa TA100, todas as frações de S. crassifolia protegeram o DNA contra a ação lesiva de azida sódica, e a fração hexânica exibiu a maior proteção desse trabalho. Assim, concluímos que as frações de S. crassifolia testadas neste estudo poderiam ser utilizadas em quimioprevenção.

Solanum lycocarpum St.- Hil popularly known as 'fruta-de-lobo' or 'lobeira' is native to the Brazilian Cerrado, and used in folk medicine due to its phytotherapic properties. The action of S. lycocarpum on the cell cycle and chromosomes in order to demonstrate whether there are aneugenic and/or clastogenic effects is unknown. Thus, this study aimed at investigating the cytotoxic and genotoxic potential of methanol and hexane extracts of S. lycocarpum on growth and cell cycle of Lactuca sativa and Allium cepa. Roots from both species were exposed for 72 hours to methanol and hexane extracts with 50, 100, and 200 µg mL-1 of S. lycocarpum. Slides were prepared by the squash technique and then analyzed to determine the mitotic index and the total of chromosomal and nuclear abnormalities. The frequencies of chromosomal and nuclear abnormalities were high and significant with a dose-dependent effect, indicating that S. lycocarpum has a cytotoxic and genotoxic action depending on the dose used on meristem cells of A. cepa and L. sativa.


Solanum lycocarpum St.-Hil (Solanaceae) conhecida popularmente como fruta-de-lobo ou lobeira é uma planta nativa do Cerrado brasileiro, utilizada na medicina popular nos tratamentos de diabetes, obesidade e redução do colesterol. Ainda não é conhecida a ação de S. lycocarpum sobre o ciclo celular e os cromossomos, demonstrando se possuem efeitos aneugênicos e/ou clastogênicos. O objetivo desse estudo foi investigar o potencial citotóxico e genotóxico dos extratos metanólico e hexânico de S. lycocarpum sob o crescimento e ciclo celular de Lactuca sativa e Allium cepa. As raízes de ambas as espécies foram expostas por 72h aos extratos metanólico e hexânico com 50, 100 e 200 µg mL-1 de S. lycocarpum. As lâminas foram montadas pela técnica de esmagamento e em seguida foram analisadas, afim de determinar o índice mitótico e anormalidades cromossômicas e nucleares. As frequências de anormalidades cromossômicas e nucleares foram altas e significativas, com efeito dose-dependente e confirmando que S. lycocarpum tem ação citotóxica e genotóxica de acordo com a dose utilizada sobre as células meristemáticas de A. cepa e L. sativa.

Abstract Dyes are recalcitrant compounds that resist conventional biological treatments. The degradation of three textile dyes (Indigo, RBBR and Sulphur Black), and the dye-containing liquid effluent and solid waste from the Municipal Treatment Station, Americana, São Paulo, Brazil, by the cyanobacteria Anabaena flos-aquae UTCC64, Phormidium autumnale UTEX1580 and Synechococcus sp. PCC7942 was evaluated. The dye degradation efficiency of the cyanobacteria was compared with anaerobic and anaerobic-aerobic systems in terms of discolouration and toxicity evaluations. The discoloration was evaluated by absorption spectroscopy. Toxicity was measured using the organisms Hydra attenuata, the alga Selenastrum capricornutum and lettuce seeds. The three cyanobacteria showed the potential to remediate textile effluent by removing the colour and reducing the toxicity. However, the growth of cyanobacteria on sludge was slow and discoloration was not efficient. The cyanobacteria P. autumnale UTEX1580 was the only strain that completely degraded the indigo dye. An evaluation of the mutagenicity potential was performed by use of the micronucleus assay using Allium sp. No mutagenicity was observed after the treatment. Two metabolites were produced during the degradation, anthranilic acid and isatin, but toxicity did not increase after the treatment. The cyanobacteria showed the ability to degrade the dyes present in a textile effluent; therefore, they can be used in a tertiary treatment of effluents with recalcitrant compounds.

Abstract Vernonanthura polyanthes (Spreng.) A.J. Vega & Dematt. (Asteraceae), known as “assa-peixe”, has been used in ethnomedicine for the treatment of various diseases such as bronchitis, pneumonia, hemoptysis, persistent cough, internal abscesses, gastric and kidney stone pain. Moreover, some studies demonstrated that species of Genus Vernonia present antifungal activity. Due to the biological relevance of this species, the aim of this study was to investigate the toxic, genotoxic, antigenotoxic and antifungal potential of V. polyanthes leaves aqueous extract in somatic cells of Drosophila melanogaster or against Candida spp. The aqueous extract of the plant showed no toxic, genotoxic and antigenotoxic activity in the experimental conditions tested using the wing somatic mutation and recombination test (SMART/wing). However, when the extract was associated with doxorubicin, used in this work as a positive control, the mutagenic potential of doxorubicin was enhanced, increasing the number of mutations in D. melanogaster somatic cells. In the other hand, no inhibitory activity against Candida spp. was observed for V. polyanthes leaves aqueous extract using agar-well diffusion assay. More studies are necessary to reveal the components present in the V. polyanthes leaves aqueous extract that could contribute to potentiate the doxorubicin genotoxicity.


Resumo Vernonanthura polyanthes (Spreng.) A.J. Vega & Dematt. (Asteraceae), conhecida como “assa-peixe”, tem sido utilizada na medicina popular para o tratamento de várias doenças, como bronquite, pneumonia, hemoptise, tosse persistente, abcessos internos, afecções gástricas e cálculo renal. Além disso, alguns estudos já demonstraram que espécies do Gênero Vernonia apresentam atividade antifúngica. Devido à relevância biológica dessa espécie, o objetivo deste estudo foi investigar os efeitos citotóxico, genotóxico, antigenotóxico e antifúngico do extrato aquoso das folhas de V. polyanthes em células somáticas de Drosophila melanogaster ou contra Candida spp. O extrato aquoso da planta não apresentou atividade citotóxica, genotóxica e antigenotóxica nas condições experimentais testadas usando o teste de recombinação e mutação somática em asa (SMART-asa). No entanto, quando o extrato foi associado com a doxorrubicina, utilizada neste trabalho como controle positivo, o potencial mutagênico da doxorrubicina foi potencializado, aumentando o número de mutações em células somáticas de D. melanogaster. Por outro lado, nenhuma atividade inibitória contra Candida spp. foi observada utilizando o extrato aquoso das folhas de V. polyanthes por meio do método de difusão em ágar. Mais estudos são necessários para desvendar os componentes presentes no extrato aquoso das folhas de V. polyanthes que possam contribuir para potencializar a genotoxicidade da doxorrubicina.

Abstract The objective of this study was to evaluate the action of Hymenaea stigonocarpa bark hydroalcoholic extract against a mutagenic compound using A. cepa meristematic root cells as a test system. The treatment groups were: Negative Control (NC) – distilled water; Positive Control (PC) – paracetamol at a concentration of 0.008 mg/mL, Jatoba Control (JC) – aqueous fraction jatobá-do-cerrado at 0.5 or 1.0 or 1.5 mg/mL, and Simultaneous Treatment (ST) - jatobá-do-cerrado aqueous fraction at a concentration of 0.5 or 1.0 or 1.5 mg/mL associated with paracetamol solution at a concentration of 0.008 mg/mL. All groups were analyzed at 24 and 48 h. Five onion bulbs (five replications) were used for each treatment group. The root tips were fixed in Carnoy and slides prepared by the crush technique. Cells were analyzed throughout the cell cycle, totaling 5,000 for each treatment group at each exposure time. Mitotic indices were subjected to statistical analysis using the chi-square test (p<0.05). From the results it was found that the ST group, at the three concentrations, significantly potentiated the antiproliferative effect of the test system cells when compared to PC, NC and TJ at the three concentrations. Furthermore, the three ST concentrations significantly reduced the number of cell aberrations when compared to the number of aberrant cells obtained for the PC, demonstrating antimutagenic action on the A. cepa test system cells.


Resumo O objetivo do presente trabalho foi avaliar a ação do extrato hidroalcólico do ritidoma de Hymenaea stigonocarpa frente a um composto mutagênico, utilizando como sistema teste as células meristemáticas de raízes de A. cepa. Os grupos tratamentos avaliados foram: Controle Negativo (CN) – água destilada; Controle Positivo (CP) – paracetamol na concentração de 0,008 mg/mL, Controle Jatobá (CJ) – fração aquosa de jatobá-do-cerrado na concentração de 0,5 ou 1,0 ou 1,5 mg/mL, e Tratamento Simultâneo (TS) – fração aquosa de jatobá-do-cerrado na concentração de 0,5 ou 1,0 ou 1,5 mg/mL associada a solução de paracetamol na concentração de 0,008 mg/mL. Todos os grupos foram analisados nos tempos de 24 e 48 h. Para cada grupo tratamento cinco bulbos de cebolas (cinco repetições) foram utilizados. As radículas foram fixadas em Carnoy e as lâminas preparadas pela técnica de esmagamento. Analisaram-se células em todo ciclo celular, totalizando 5.000 para cada grupo tratamento em cada tempo de exposição. Os índices mitóticos obtidos foram submetidos à análise estatística do Qui-quadrado (p<0,05). A partir dos resultados verificou-se que o grupo TS, nas três concentrações, potencializou o efeito antiproliferativo significativo as células do sistema teste quando comparado ao CP, CN e TJ nas três concentrações. Ainda, o TS nas três concentrações reduziu de forma significativa o número de aberrações celulares quando comparado com o número de células aberrantes obtidas para o CP, demonstrando ação antimutagênica as células do sistema teste A. cepa.

Abstract The kinetics of an extracellular β-D-fructofuranosidase fructohydrolase production by Saccharomyces cerevisiae in a chemically defined medium, i.e., sucrose peptone agar yeast extract at pH 6, was investigated. The wild-type was treated with a chemical mutagen, methyl methane sulfonate. Among the six mutants isolated, methyl methane sulfonate-V was found to be a better enzyme producing strain (52 ± 2.4a U/mL). The maximum production (74 ± 3.1a U/mL) was accomplished after at 48 h (68 ± 2.7a mg/mL protein). The mutants were stabilized at low levels of 5-fluoro-cytocine and the viable ones were further processed for optimization of cultural conditions and nutritional requirements. The sucrose concentration, incubation period and pH were optimized to be 30 g/L, 28 °C, and 6.5, respectively. The methyl methane sulfonate-V exhibited an improvement of over 10 folds in enzyme production when 5 g/L ammonium sulfate was used as a nitrogen source. Thin layer chromatography and high-performance liquid chromatography analysis illustrated the optimal enzyme activity supported by the higher rate of hydrolysis of sucrose into monosaccharides, particularly α-D-glucose and β-D-fructose. The values for Qp (2 ± 0.12c U/mL/h) and Yp/s (4 ± 1.24b U/g) of the mutant were considerably increased in comparison with other yeast strains (both isolates and viable mutants). The mutant could be exploited for enzyme production over a wider temperature range (26–34 °C), with significantly high enzyme activity (LSD 0.048, HS) at the optimal temperature.

Abstract The antagonistic potential of Trichoderma strains was assayed by studying the effect of their culture filtrate on the radial growth of Sclerotium rolfsii, the causal agent of chickpea collar rot. Trichoderma harzianum-1432 (42.2%) and Trichoderma atroviride (40.3%) were found to be strong antagonists. To enhance their antagonistic potential, mutagenesis of these two selected strains was performed. Two mutants, Th-m1 and T. atroviride m1, were found to be more effective than their parent strains. The enzymatic activities of the selected parent and mutant strains were assayed, and although both mutants were found to have enhanced enzymatic activities compared to their respective parent strains, Th-m1 possessed the maximum cellulase (5.69 U/mL) and β-1,3-glucanase activity (61.9 U/mL). Th-m1 also showed high competitive saprophytic ability (CSA) among all of the selected parent and mutant strains, and during field experiments, Th-m1 was found to successfully possess enhanced disease control (82.9%).

Maytenus macrocarpa (MM) (Ruiz & Pav.) Briq. es un árbol utilizado por poblaciones amazónicas de forma medicinal, para el control de la fertilidad. El objetivo de este trabajo fue evaluar el efecto abortivo potencial de diferentes dosis del extracto acuoso de M. macrocarpa "chuchuwasi" (25, 50 y 100 mg/kg) sobre embriones preimplantacionales de ratón de la cepa albina Swiss Rockfeller. El extracto fue administrado a los ratones vía intraperitoneal entre el primer y cuarto día de gestación. Se evaluó la calidad y desarrollo de los embriones. Se realizó el test de Micronucleo (MN) a fin de conocer si el extracto de MM era genotoxico. Ninguno de los tratamientos mostro significancia en el número de embriones degradados o inviables y en la tasa de MN. Los resultados sugieren que el extracto acuoso de M. macrocarpa no tiene un efecto sobre el desarrollo normal de los embriones.

Abstract The present study evaluated the genotoxic effects of the atmospheric air on Tradescantia pallida var. purpurea in urban areas with different intensities of vehicular traffic and in riparian forest fragments in the Sinos River Basin (Rio Grande do Sul, Brazil), considering the influence of climatic conditions prevailing in these environments. Bimonthly, from May 2012 to March 2013, cuttings with flower buds were exposed for 8 h in urban and riparian forest environments in the municipalities of Caraá, Taquara and Campo Bom in the upper, middle and lower sections, respectively, of the Sinos River Basin. Simultaneously, negative controls were made and climatic data were recorded. Micronuclei (MCN) frequencies were determined in young tetrads of pollen mother cells and expressed as MCN/100 tetrads. Significantly higher MCN frequencies were observed in buds exposed in urban and riparian forest environments in Taquara (up to 7.23 and 4.80, respectively) and Campo Bom (up to 4.90 and 4.23, respectively) than in buds exposed in Caraá (up to 2.90 and 2.50, respectively), in the majority of samplings, and in relation to the negative control (up to 1.93) in all months. Over the course of the period monitored, there were significant variations in MCN frequencies at all sampling points, with the exception of the urban environment in Caraá. For the urban environments, relation between the MCN frequency, vehicular traffic and mean temperature was observed. For the riparian forest fragments, there was no association between MCN frequency and climatic factors. Tradescantia pallida var. purpurea can be considered a useful tool to point out areas with increased atmospheric pollution, since the exposure of plants under severe climatic conditions is avoided to minimize their negative influence on the formation of micronuclei.


Resumo O presente estudo avaliou os efeitos genotóxicos do ar atmosférico sobre Tradescantia pallida var. purpurea em áreas urbanas com diferentes intensidades de tráfego veicular e em fragmentos de mata ciliar na Bacia do Rio dos Sinos (Rio Grande do Sul, Brasil), considerando a influência de condições climáticas prevalecentes nesses ambientes. Bimensalmente, de maio de 2012 a março de 2013, ramos com botões florais foram expostos por 8 h em ambientes urbanos e de mata ciliar nos municípios de Caraá, Taquara e Campo Bom nos trechos superior, médio e inferior, respectivamente, da Bacia do Rio dos Sinos. Simultaneamente, foram realizados controles negativos e levantados dados climáticos. Frequências de micronúcleos (MCN) foram determinadas em tétrades jovens de células-mãe de grãos de pólen e expressas como MCN/100 tétrades. Frequências de MCN significativamente superiores foram observadas em botões expostos nos ambientes urbanos e de matas ciliares em Taquara (até 7,23 e 4,80, respectivamente) e Campo Bom (até 4,90 e 4,23, respectivamente) do que em botões expostos em Caraá (até 2,90 e 2,50, respectivamente), na maioria das amostragens, e em relação ao controle negativo (até 1,93) em todos os meses. Ao longo do período monitorado, ocorreram variações significativas nas frequências de MCN em todos os pontos amostrais, com exceção do ambiente urbano de Caraá. Para os ambientes urbanos, foi observada relação entre a frequência de MCN, tráfego veicular e temperatura média. Para os fragmentos de mata ciliar, não houve associação entre a frequência de MCN e fatores climáticos. Tradescantia pallida var. purpurea pode ser considerada uma ferramenta útil para apontar áreas com poluição atmosférica aumentada, desde que, sob condições climáticas severas, a exposição das plantas seja evitada, para minimizar o efeito destas sobre a formação de micronúcleos.

Introducción. En Colombia, la minería es una actividad económica importante; sin embargo, genera grandes cantidades de residuos que contienen elementos potencialmente tóxicos, como los metales pesados, que contaminan los ecosistemas y ponen en riesgo la salud humana. La región de La Mojana es una de las zonas más ricas en biodiversidad del planeta y se ha visto sometida a procesos de contaminación muy relacionados con la minería de oro que se desarrolla en sus alrededores. Objetivo. Evaluar la genotoxicidad en una población expuesta a residuos de metales pesados en la región de La Mojana. Materiales y métodos. Se evaluaron los efectos genotóxicos y su relación con la concentración de metales pesados (mercurio, cadmio y plomo) en muestras de sangre de la población expuesta y el grupo de control. El grupo expuesto lo conformaron habitantes de los municipios de Guaranda, Sucre, Majagual y San Marcos; en el grupo de control se incluyó a habitantes del municipio de Montería. Se determinó el daño en el ADN mediante el ensayo cometa en condiciones alcalinas. Las concentraciones de mercurio se establecieron mediante espectrometría de absorción atómica con vapor frío, en tanto que las de cadmio y plomo se determinaron por espectrometría de absorción atómica en horno de grafito. Resultados. Las concentraciones de los metales sobrepasaron los límites permitidos por la Organización Mundial de la Salud. Se evidenciaron efectos genotóxicos posiblemente asociados a la presencia de los metales en la sangre. Se encontraron asociaciones significativas (p<0,05) entre la presencia de mercurio y de cadmio, y el daño en el ADN. Conclusión. Estos resultados sugieren que el daño genético registrado en pobladores de la región de La Mojana, Colombia, puede estar asociado a la presencia de los metales estudiados en las muestras de sangre.


Introduction: Mining is an economically important activity in Colombia which generates large quantities of residues containing potentially toxic elements such as heavy metals. These contaminate ecosystems and place human health at risk. La Mojana lies within one of the most biodiversity-rich zones on Earth and has been subjected to processes of contamination closely related to gold mining activities in the surrounding areas. Objective: To evaluate genotoxicity in the population of La Mojana region exposed to heavy metals. Materials and methods: Genotoxic effects and their relationship with concentrations of heavy metals (mercury, cadmium and lead) in blood were evaluated among an exposed population and a control group. The exposed group comprised inhabitants of the municipalities of Guaranda, Sucre, Majagual and San Marcos; inhabitants of the municipality of Montería were chosen as a control group. DNA damage was determined using the alkaline comet assay. Concentrations of mercury were determined by cold vapor atomic absorption spectrometry, and those of cadmium and lead by graphite furnace atomic absorption spectrometry. Results: Concentrations of the heavy metals exceeded the limits permitted by the World Health Organization. Genotoxic effects were found in the exposed population, possibly associated with the presence of these metals in blood. Significant associations (p<0.05) were found between mercury and cadmium levels and damage to DNA. Conclusion: These results suggest that the genetic damage recorded among inhabitants of the region of La Mojana, Colombia, may be associated with the presence of heavy metals in the blood.

The species Citrus sinensis (L.) Osbeck (orange) is used in folk medicine as an infusion (tea) for the treatment of nerve irritation, insomnia, spasms, whooping cough, flue, and gastrointestinal problems. The aim of this study was to evaluate the antiproliferative and genotoxic potential of aqueous extracts of C. sinensis testing by Allium cepa L., as well as to determine the components in the complex mixture (tea). Leaves from two populations of this species were used in the preparation of infusions at two concentrations: 6g.L-1 and 24g.L-1, besides using distilled water as a negative control and 9.6% glyphosate as a positive control. Six groups of four bulbs of A. cepa were used, each group corresponding to one of the treatments. The bulbs were rooted in distilled water and then transferred to the respective treatments where they remained for 24 hours (the bulbs of the negative control remained in distilled water). The rootlets were collected, fixated in ethanol-acetic acid (3:1) for 24 hours and stored in 70% alcohol. 4,000 cells were analyzed per group of bulbs, and the calculated mitotic indices were submitted to statistical analysis using the c 2 test. The aqueous extracts of C. sinensis have as main components flavonoids and phenolic acids. The values of the mitotic index decreased with increasing concentration of the extracts relative to the control in water. The results indicated that the orange aqueous extracts in the studied concentrations in both populations have antiproliferative and genotoxic effects on the cell cycle of A. cepa.


A espécie Citrus sinensis (laranjeira) (L.) Osbeck é usada na medicina popular como infusão (chá) para o tratamento de irritações nervosas, insônia, espasmos, tosse convulsa, gripes e problemas gastrointestinais. O objetivo desse estudo foi avaliar o potencial antiproliferativo e genotóxico dos extratos aquosos de Citrus sinensis pelo teste de Allium cepa L., bem como determinar os componentes na mistura complexa (chá). Folhas de duas populações dessa espécie foram utilizadas no preparo das infusões em duas concentrações: 6g.L-1 e 24g.L-1, além de serem usados água destilada como controle negativo e glifosato 9,6% como controle positivo. Utilizaram-se seis grupos de quatro bulbos de A. cepa, cada grupo correspondendo a um dos tratamentos. Os bulbos foram enraizados em água destilada e então transferidos para os respectivos tratamentos onde permaneceram por 24 horas (os bulbos do controle negativo permaneceram em água destilada). As radículas foram coletadas, fixadas em etanol-ácido acético (3:1) por 24 horas e armazenadas em álcool 70%. Foram analisadas 4,000 células por grupo de bulbos, e os índices mitóticos calculados submetidos à análise estatística pelo teste c 2. Os extratos aquosos de C. sinensis têm como componentes principais flavonoides e ácidos fenólicos. Os valores dos índices mitóticos diminuíram com o aumento das concentrações dos extratos, em relação ao controle em água. Os resultados indicaram que os extratos aquosos de laranjeira nas concentrações estudadas das duas populações possuem ação antiproliferativa e genotóxica sobre o ciclo celular de A. cepa.

Los contaminantes del aire han sido, y siguen siendo, los principales factores que contribuyen a enfermedades pulmonares obstructivas crónicas (EPOC), enfermedades cardiovasculares (ECV), asma y cáncer. El ozono, los óxidos de azufre, monóxido de carbono, óxidos de nitrógeno y el material particulado constituyen los contaminantes del aire. En este estudio se realizó la cuantificación de hidrocarburos aromáticos policíclicos (HAP) presentes en muestras de material particulado fracción respirable PM2.5 del aire de la ciudad de Pamplona (Norte de Santander, Colombia). Inicialmente la materia orgánica presente en el material particulado PM2.5 se extrajo por ultrasonido, seguido de la concentración por rotaevaporación, obteniéndose el extracto global, parte del cual se sometió a un proceso de fraccionamiento en una columna de silicagel. Finalmente se obtienen cuatro fracciones. La cuantificación de los HAP presentes en el material particulado PM2.5, el extracto global y las cuatro fracciones se realizó en un cromatógrafo de gases Agilent 6890 plus con detector FID. Entre los HAP identificados se encuentran: naftaleno, fluoreno, fenantreno, benzo(a)antraceno, pireno, dibenzo(a,h)antraceno e indeno(1,2,3,cd)pireno, estos HAP son compuestos tóxicos, mutágenos y carcinógenos para los animales y los seres humanos según la agencia internacional de investigación del cáncer (IARC).


Air pollutants have been. and still are. the main factor that contribute to chronic obstructive pulmonary disease (COPD), cardiovascular disease (CVD), asthma and cancer. Ozone, sulfur oxides, carbon monoxide, nitrogen oxides and particulate matter are air pollutants. Quantification of polycyclic aromatic hydrocarbons (PAHs) present in samples of particulate matter breathable fraction PM2.5 air of the city of Pamplona (Norte de Santander, Colombia) was performed in this study. Initially, organic material present in the particulate material PM2.5 was extracted through ultrasound, followed by concentration through a rotary evaporator, obtaining the overall extract, part of which was subjected to a division process in a silica gel column, Finally four fractions were obtained. Quantification of PAHs present in the PM2.5 particulate material, global extract and the four fractions was carried out in an Agilent 6890 gas chromatograph plus with FID detector. Among the HAPs identified the following are included: naphthalene, fluorene, phenanthrene, benzo(a)anthracene, pyrene, dibenzo(a,h)anthracene and indene(1,2,3,cd-pyrene). These PAHs are toxic compounds, mutagenic and carcinogenic for animals and humans according to the International Agency for Research on Cancer (IARC).
