RESUMO

The removal of pollutants from water bodies is crucial for the well-being of humanity and is a topic of global research. Researchers have turned their attention to green synthesized nanoparticles for wastewater treatment due to their eco-friendly nature, biocompatibility, and cost-effectiveness. This work demonstrates the efficient removal of organic dye and both gram-positive and gram-negative bacteria from water bodies using copper-doped cerium oxide nanoparticles synthesized withMurraya Koenigiiextract. Characterized via various methods, the 15% copper doped cerium oxide nanoparticles (Cu 15% NPs) exhibited maximum Congo red dye adsorption (98% degradation in 35 min). Kinetic analysis favoured a pseudo-second-order model, indicating the chemical nature of adsorption. Equilibrium adsorption isotherms aligned with the Langmuir model, indicating homogenous monolayer dye adsorption on the doped adsorbent. The maximum uptake of adsorbate,Qmobtained from Langmuir model for Cu 15% NPs was 193 mg g-1. The study also showed enhanced antibacterial activity againstBacillus subtilis, Staphylococcus aureus, Escherichia coliandPseudomonas aeruginosafor Cu-doped ceria, attributed to generation of reactive oxygen species (ROS) induced by the redox cycling between Ce3+and Ce4+. This substantiated that the green synthesized copper doped cerium oxide nanoparticles are potential candidates for adsorptive removal of Congo red dye and as antibacterial agents.

OBJECTIVES: To determine the predictive performance of the urine Congo red point-of-care test for the identification of preeclampsia in women presenting with suspected preeclampsia. METHODS: A prospective multi-center cohort study was conducted to include women with suspected preeclampsia (n = 244). The urine Congo red test was determined (score range 1-8). The diagnosis of preeclampsia was based on criteria proposed by The American College of Obstetricians and Gynecologists. The primary outcome was the predictive performance (sensitivity, specificity, negative and positive predictive values, as well as likelihood ratios) of the Congo red kit test for the diagnosis of preeclampsia. RESULTS: Fifty-four percent (131/244) of women with suspected preeclampsia subsequently developed preeclampsia. The sensitivity and specificity of the urine Congo red test were 49.6% and 94.7%, respectively, when using a cutoff for Congo red ≥4. The test had a significant positive correlation with the level of urine protein (Pearson correlation 0.61, p-value <.01). Intra- and inter-observer reliabilities were good (intra-class correlation coefficient and Cohen's kappa coefficient of 0.88 and 0.75, respectively; p < .01). CONCLUSION: The urine Congo red kit test has a high positive predictive performance for the identification of preeclampsia with high reproducibility. This test may be used as a bed side test to rule-in the diagnosis of preeclampsia in women presenting with suspected preeclampsia.

OBJECTIVE: To discuss the influence of Sailuotong (, SLT) on the Neurovascular Unit (NVUs) of amyloid precursor protein (APP)/presenilin-1(PS1) mice and evaluate the role of gas supplementation in activating blood circulation during the progression of Alzheimer's disease (AD). METHODS: The mice were allocated into the following nine groups: (a) the C57 Black (C57BL) sham-operated group (control group), (b) ischaemic treatment in C57BL mice (the C57 ischaemic group), (c) the APP/PS1 sham surgery group (APP/PS1 model group), (d) ischaemic treatment in APP/PS1 mice (APP/PS1 ischaemic group), (e) C57BL mice treated with aspirin following ischaemic treatment (C57BL ischaemic + aspirin group), (f) C57BL mice treated with SLT following ischaemic treatment (C57BL ischaemic + SLT group), (g) APP/PS1 mice treated with SLT (APP/PS1 + SLT group), (h) APP/PS1 mice treated with donepezil hydrochloride following ischaemic treatment (APP/PS1 ischaemic + donepezil hydrochloride group) and (i) APP/PS1 mice treated with SLT following ischaemic treatment (APP/PS1 ischaemic + SLT group). The ischaemic model was established by operating on the bilateral common carotid arteries and creating a microembolism. The Morris water maze and step-down tests were used to detect the spatial behaviour and memory ability of mice. The hippocampus of each mouse was observed by haematoxylin and eosin (HE) and Congo red staining. The ultrastructure of NVUs in each group was observed by electron microscopy, and various biochemical indicators were detected by enzyme-linked immunosorbent assay (ELISA). The protein expression level was detected by Western blot. The mRNA expression was detected by quantitative real-time polymerase chain reaction (qRT-PCR). RESULTS: The results of the Morris water maze and step-down tests showed that ischemia reduced learning and memory in the mice, which were restored by SLT. The results of HE staining showed that SLT restored the pathological changes of the NVUs. The Congo red staining results revealed that SLT also improved the scattered orange-red sediments in the upper cortex and hippocampus of the APP/PS1 and APP/PS1 ischaemic mice. Furthermore, SLT significantly reduced the content of Aß, improved the vascular endothelium and repaired the mitochondrial structures. The ELISA detection, western blot detection and qRT-PCR showed that SLT significantly increased the vascular endothelial growth factor (VEGF), angiopoietin and basic fibroblast growth factor, as well as the levels of gene and protein expression of low-density lipoprotein receptor-related protein-1 (LRP-1) and VEGF in brain tissue. CONCLUSIONS: By increasing the expression of VEGF, SLT can promote vascular proliferation, up-regulate the expression of LRP-1, promote the clearance of Aß and improve the cognitive impairment of APP/PS1 mice. These results confirm that SLT can improve AD by promoting vascular proliferation and Aß clearance to protect the function of NVUs.

This paper presents the formation of a self-healing hydrogel prepared by carboxyethyl modification of chitosan and crosslinking with oxidized sodium alginate. Concurrently, the incorporation of Ca2+ facilitated the formation of "calcium bridges" through intricate coordination with carboxyl moieties, bolstering the attributes of the hydrogel. Various characterization methods, including scanning electron microscopy, texture analysis, and rheological measurements, demonstrated that the introduction of carboxyethyl groups resulted in a more compact hydrogel network structure and improved the hardness and elasticity. The addition of Ca2+ helped to further enhance the mechanical performance of the hydrogel and increase its thermal stability. Then, the adsorption capacity was also investigated, showing adsorption capacities of 46.17 mg/g methylene blue and 46.44 mg/g congo red for carboxyethyl chitosan/oxidized sodium alginate hydrogel, a four-fold increase for congo red versus chitosan/oxidized sodium alginate hydrogel. In addition, the adsorption behavior of CEC/OSA/2%Ca2+ hydrogel can be well described by pseudo-second-order kinetic model and Langmuir adsorption isothermal model. Compared to traditional hydrogels, CEC/OSA/2%Ca2+ hydrogel shows superior mechanical strength, enhanced thermal stability, and improved adsorption capacity, which can effectively adsorb not only methylene blue but also congo red. These advancements demonstrate our hydrogel's innovative properties.

The human-adapted enteric bacterial pathogen Shigella causes millions of infections each year, creates long-term growth effects among pediatric patients, and is a leading cause of diarrheal deaths worldwide. Infection induces watery or bloody diarrhea as a result of the pathogen transiting the gastrointestinal tract and infecting the epithelial cells lining the colon. With staggering increases in antibiotic resistance and the current lack of approved vaccines, standardized research protocols are critical to studying this formidable pathogen. Here, methodologies are presented to examine the molecular pathogenesis of Shigella using in vitro analyses of bacterial adherence, invasion, and intracellular replication in colonic epithelial cells. Prior to infection analyses, the virulence phenotype of Shigella colonies was verified by the uptake of the Congo red dye on agar plates. Supplemented laboratory media can also be considered during bacterial culturing to mimic in vivo conditions. Bacterial cells are then used in a standardized protocol to infect colonic epithelial cells in tissue culture plates at an established multiplicity of infection with adaptations to analyze each stage of infection. For adherence assays, Shigella cells are incubated with reduced media levels to promote bacterial contact with epithelial cells. For both invasion and intracellular replication assays, gentamicin is applied for various time intervals to eliminate extracellular bacteria and enable assessment of invasion and/or the quantification of intracellular replication rates. All infection protocols enumerate adherent, invaded, and/or intracellular bacteria by serially diluting infected epithelial cell lysates and plating bacterial colony forming units relative to infecting titers on Congo red agar plates. Together, these protocols enable independent characterization and comparisons for each stage of Shigella infection of epithelial cells to study this pathogen successfully.

Polydopamine (PDA) and metal-organic skeleton HKUST-1 were co-deposited on the base membrane of hexamethylenediamine (HDA)-crosslinked polyetherimide (PEI) ultrafiltration membrane as the interlayer, and high-throughput organic solvent nanofiltration membrane (OSN) was prepared by interfacial polymerization and solvent activation reaction. The polyamide (PA) layer surface roughness from 28.4 nm in PA/PEI to 78.3 nm in PA/PDA-HKUST-10.6/PEI membrane, reduced the thickness of the separation layer from 79 to 14 nm, and significantly improved the hydrophilic, thermal and mechanical properties. The flux of the PA/PDA-HKUST-10.6/PEI membrane in a 0.1 g/L Congo Red (CR) ethanol solution at 0.6 MPa test pressure reached 21.8 L/(m2·hr) and the rejection of CR was 92.8%. Solvent adsorption test, N, N-dimethylformamide (DMF) immersion experiment, and long-term operation test in ethanol showed that the membranes had high solvent tolerance. The solvent flux test demonstrated that, under the test pressure of 0.6 MPa, the flux of different solvents ranked as follows: methanol (56.9 L/(m2·hr)) > DMF (39.6 L/(m2·hr)) > ethanol (31.2 L/(m2·hr)) > IPA (4.5 L/(m2·hr)) > N-hexane (1.9 L/(m2·hr)). The ability of the membranes to retain dyes in IPA/water dyes solution was also evaluated. The flux of the membrane was 30.4 L/(m2·hr) and the rejection of CR was 91.6% when the IPA concentration reached 50%. This OSN membrane-making strategy is economical, environment-friendly and efficient, and has a great application prospect in organic solvent separation systems.

OBJECTIVES: Preeclampsia (PE) poses a serious threat to the health of the pregnant woman and her developing fetus due to the difficulty in diagnosing the condition. The disease can develop and worsen suddenly without noticeable signs and symptoms. Thus, there is an urgent need for a simple Point of Care Test (POCT) that improves accessibility to testing and can be used as an aid in the diagnosis of PE. CercaTest Red is a noninvasive detection kit for impending preeclampsia using urine from pregnant women. This is especially pertinent for women who have limited access to secondary/tertiary healthcare as those in remote settings, low-income countries or simply lack of out of hours laboratory services. METHODS: The kit employs an absorptive column that separates Congo red dye bound to urinary misfolded protein from pregnant women and unbound dye. When a solution of Congo red dye pre-mixed with urine is loaded onto the absorptive matrix in a detection cuvette, the presence (positive) or absence (negative) of misfolded proteins can be determined based on the color of eluate collected in the lower section of the cuvette. 190 and 937 pregnant women who were >18 years old at the gestational age of ≥20 weeks were enrolled for the feasibility and validation cohort, respectively. The consistency between CercaTest Red and clinical diagnosis of PE according to the American College of Obstetricians and Gynecologist (ACOG) Guidelines was analyzed using the kappa statistic. RESULTS: The POCT has a limit of detection (LoD) of human urinary misfolded proteins equivalent to 0.45 µM of denatured human serum albumin, with high reproducibility and stability. An accuracy of 96.84% for diagnosis of preeclampsia with a Kappa statistic of 0.746 (p < 0.001) was validated in a cohort of 937 subjects. CONCLUSION: This test is easy to use, cost-effective and portable with short turnaround time and no laboratory instrument requirement. In the future, the test may have the potential to become quantitative using spectroscopy (Chinese Clinical Trial Registry No. ChiCTR1800017692).

Background: Transthyretin amyloidosis (ATTR) is a significant cause of cardiomyopathy and other morbidities in the elderly and Black Americans. ATTR can be treated with new disease-modifying therapies, but large shortfalls exist in its diagnosis. The objective of this study was to test whether TTR amyloid can be detected and imaged in the conjunctiva using a novel small-molecule fluorescent ocular tracer, with the implication that ATTR might be diagnosable by a simple eye examination. Methods: Three approaches were used in this study. First, AMDX-9101 was incubated with in vitro aggregated TTR protein, and changes in its excitation and emission spectra were quantified. Second, a cadaver eye from a patient with familial amyloid polyneuropathy type II TTR mutation and a vitrectomy sample from an hATTR patient were incubated with AMDX-9101 and counterstained with Congo Red and antibodies to TTR to determine whether AMDX-9101 labels disease-related TTR amyloid deposits in human conjunctiva and eye. Last, imaging of in vitro aggregated TTR amyloid labeled with AMDX-9101 was tested in a porcine ex vivo model, using a widely available clinical ophthalmic imaging device. Results: AMDX-9101 hyper-fluoresced in the presence of TTR amyloid in vitro, labeled TTR amyloid deposits in postmortem human conjunctiva and other ocular tissues and could be detected under the conjunctiva of a porcine eye using commercially available ophthalmic imaging equipment. Conclusions: AMDX-9101 enabled detection of TTR amyloid in the conjunctiva, and the fluorescent binding signal can be visualized using commercially available ophthalmic imaging equipment. Translational Relevance: AMDX-9101 detection of TTR amyloid may provide a potential new and noninvasive test for ATTR that could lead to earlier ATTR diagnosis, as well as facilitate development of new therapeutics.

The majority of environmental researchers are becoming increasingly concerned with the manufacture of inexpensive adsorbents for the detoxification of industrial effluents. To address one of the significant and well-known pollution issues with certain drains that act as hotspots and contribute to coastal pollution in Alexandria, this study aims to develop an economical, ecologically friendly sorbent. This study assessed the efficacy of a biomass-coated magnetic composite and a magnetic active adsorbent for the removal of two dyes from an industrially contaminated sewer using a wetland plant (Phragmites australis). Using magnetic biosorbent, the biosorption of Xylenol orange and Congo red ions from polluted drain discharge in Abu Qir Bay was evaluated in the current study. Using scanning electron microscopy imaging and Fourier transform infra-red analysis; the surface function and morphology of the nano-biosorbent were examined. At room temperature, the effects of initial dye concentration, pH, contact time, and nano-biosorbent concentration have all been investigated. The greatest percentages that nano-biosorbent can remove from Congo red and Xylenol orange are 97% and 47%, respectively. The removal of the initial Congo red concentration varied from 42 to 97%, while the removal of the initial Xylenol orange concentration varied from 30 to 47%. The adsorption capacity was shown to be strongly pH-dependent; capacity dose as pH value increased, with pH 10 being the ideal pH for Congo red and pH 6 being the ideal pH value for Xylenol orange. The adsorption capacity for Congo red varied between 0.96 and 3.36 and the adsorption capacity for Xylenol orange varied between 0.18 and 17.58. The removal capacity decreased from 3.36 to 0.96 mg/g when the biosorbent dosage was increased from 0.05 to 0.5 g/L for Congo red, in case of Xylenol orange, the removal capacity increased from 0.18 to 17.58 mg/g when the biosorbent dosage was increased from 0.05 to 0.5 g/L. The removal capacity of Congo red increases quickly with time and varied from 1.66 to 1.88 of contact time; while the removal capacity of Xylenol orange varied between 3.08 and 4.62 of contact time. For the dyes under study, kinetics and adsorption equilibrium were examined. Within 180 min, the equilibrium was attained because to the quick adsorption process. For Congo red and Xylenol orange, the highest adsorption capacities were 3.36 and 17.58 mg g-1, respectively. The equilibrium data were assessed using a number of isotherm models, including Langmuir, Freundlich, BET, and Tempkin, while the kinetic data were examined using a variety of kinetic models, including pseudo-first- and pseudo-second-order equations. The pseudo-second-order equation provides the greatest accuracy for the kinetic data and Langmuir model is the closest fit for the equilibrium data.

In the present study, hybrid activation of sodium peroxydisulfate (PS) by hydrodynamic cavitation and ultraviolet radiation was investigated for Congo Red (CR) degradation. Experiments were conducted using the Box-Benken design on inlet pressure (2-6 bar), PS concentration (0-50 mg. L-1) and UV radiation power (0-32 W). According to the results, at the optimum point where the pressure, PS concentration and UV radiation power were equal to 4.5 bar, 30 mg. L-1 and 16 W respectively, 92.01% of decolorization was achieved. Among the investigated processes, HC/UV/PS was the best process with the rate constant and synergetic coefficient of 38.6 × 10-3 min-1 and 2.76, respectively. At the optimum conditions, increasing the pollutant concentration from 20 mg. L-1 to 80 mg. L-1 decrease degradation rate from 92.01 to 45.21. Presence of natural organic mater (NOM) in all concentrations inhibited the CR degradation. Quenching experiments revealed that in the HC/UV/PS hybrid AOP free radicals accounted for 63.4% of the CR degradation, while the contribution of sulfate (SRs) and hydroxyl radicals (HRs) was 53.1% and 46.9%, respectively.

In this study, a new nanocomposite of hydroxyapatite (HA)/Mg-Al layered double hydroxide (LDH) was successfully formed via a facile co-precipitation method and applied to adsorb three anionic dyes of alizarin red S (ARS), Congo red (CR), and reactive red 120 (RR120) differing in the number of SO3- groups from aqueous solution. Based on a combination of characterization analysis and adsorption experiments, HA/Mg-Al LDH nanocomposite showed better adsorption performance than HA and Mg-Al LDH. Using XRD and TEM analyses, the crystallinity and the presence of nanoparticles were confirmed. According to the SEM investigation, the Mg-Al LDH layers in the nanocomposite structure were delaminated, while HA nanorods were formed at the surface of Mg-Al LDH nanoparticles. The higher BET surface area of the novel HA/Mg-Al LDH nanocomposite compared to HA and Mg-Al LDH provided its superior adsorption performance. Considering an effective amount of adsorbent dosage, pH 5 was selected as the optimum pH for each of the three dye solutions. According to the results from the study of contact time and initial concentration, the pseudo-second-order kinetic (R2 = 0.9987, 0.9951, and 0.9922) and Langmuir isotherm (R2 = 0.9873, 0.9956, and 0.9727) best fitted the data for ARS, CR, and RR120, respectively. Anionic dyes with different numbers of SO3- groups demonstrated distinct adsorption mechanisms for HA and Mg-Al LDH nanoparticles, indicating that the adsorption capacity is influenced by the number of SO3- groups, with HA/Mg-Al LDH nanocomposite offering superior performance toward dyes with higher numbers of SO3- groups. Furthermore, ΔH° less than 40 kJ/mol, positive ΔS°, and negative ΔG° accompanied by the mechanism clarifying show physical spontaneous adsorption without an external source of energy and increase the randomness of the process during the adsorption, respectively. Finally, the regeneration study demonstrated that the nanocomposite could be utilized for multiple adsorption-desorption cycles, proposing the HA/Mg-Al LDH as an economically and environmentally friendly adsorbent in the adsorption of anionic dyes in water treatment processes.

Polysaccharides from natural products play multiple roles and have extensive bioactivities in life process. Bioactivities of polysaccharides (e.g., Lentinan, Schizophyllan, Scleroglucan, Curdlan, Cinerean) have a close relation to their chain conformation. Compared to other types of polysaccharides, the conformation of ß-glucan has been studied more. The major research methods of conformation of polysaccharides from natural products (Congo red experiment, circular dichroism spectrum, viscosity method, light scattering method, size exclusion chromatography, atomic force microscope), corresponding experimental schemes, and the external factors affecting polysaccharide conformation were reviewed in this paper. These research methods of conformation have been widely used, among which Congo red experiment and viscosity method are the most convenient ones to study the morphological changes of polysaccharide chains.

This present work targets the production of an eco-friendly adsorbent (hereinafter KA/CEL) from kaolin clay functionalized with cellulose extract obtained from peanut shells. The adsorbents were used for decolorization of two different types of organic dyes (cationic: methylene blue, MB; anionic: Congo red, CR) from an aqueous environment. Several analytical methods, including Brunauer-Emmett-Teller (surface properties), Fourier Transforms infrared (functionality), scanning electron microscope, Energy dispersive X-Ray (morphology), and pHpzc test (surface charge), were used to attain the physicochemical characteristics of KA/CEL. The Box-Behnken Design (BBD) was applied to determine the crucial factors affecting adsorption performance. These included cellulose loading at 25 %, an adsorbent dose of 0.06 g, solution pH set at 10 for MB and 7 for CR, a temperature of 45 °C, and contact times of 12.5 min for MB and 20 min for CR dye. The adsorption data exhibited better agreement with the pseudo-second-order kinetic and Freundlich models. The Langmuir model estimated the monolayer capacity to be 291.5 mg/g for MB and 130.7 mg/g for CR at a temperature of 45 °C. This study's pivotal finding underscores the promising potential of KA/CEL as an effective adsorbent for treating wastewater contaminated with organic dyes.

In recent years, the growing concern over the presence of toxic aquatic pollutants has prompted intensive research into effective and environmentally friendly remediation methods. Photocatalysis using semiconductor quantum dots (QDs) has developed as a promising technology for pollutant degradation. Among various QD materials, indium phosphide (InP) and its hybrid with zinc sulfide (ZnS) have gained considerable attention due to their unique optical and photocatalytic properties. Herein, InP and InP/ZnS QDs were employed for the removal of dyes (crystal violet, and congo red), polyaromatic hydrocarbons (pyrene, naphthalene, and phenanthrene), and pesticides (deltamethrin) in the presence of visible light. The degradation efficiencies of crystal violet (CV) and congo red (CR) were 74.54% and 88.12% with InP, and 84.53% and 91.78% with InP/ZnS, respectively, within 50 min of reaction. The InP/ZnS showed efficient performance for the removal of polyaromatic hydrocarbons (PAHs). For example, the removal percentage for naphthalene, phenanthrene, and pyrene was 99.8%, 99.6%, and 88.97% after the photocatalytic reaction. However, the removal percentage of InP/ZnS for pesticide deltamethrin was 90.2% after 90 min light irradiation. Additionally, advanced characterization techniques including UV-visible spectrophotometer (UV-Vis), photoluminescence (PL), X-ray diffractometer (XRD), energy-dispersive spectrometer (EDS) elemental mapping, transmission electron microscopy (TEM), and thermogravimetric analysis (TGA) were used to analyze the crystal structure, morphology, and purity of the fabricated materials in detail. The particle size results obtained from TEM are in the range of 2.28-4.60 nm. Both materials (InP and InP/ZnS) exhibited a spherical morphology, displaying distinct lattice fringes. XRD results of InP depicted lattice planes (111), (220), and (311) in good agreement with cubic geometry. Furthermore, the addition of dopants was discovered to enhance the thermal stability of the fabricated material. In addition, QDs exhibited efficacy in the breakdown of PAHs. The analysis of their fragmentation suggests that the primary mechanism for PAHs degradation is the phthalic acid pathway.

Amyloid is an extracellular deposition of Congo red positive material which shows apple green birefringence under polarized light. A cytopathologist can uncommonly encounter such cases. Among the reported cases, a fine-needle aspiration (FNA) of amyloid is frequently misinterpreted as acellular nondiagnostic material. We report a case of amyloidoma of the right upper arm in a 68-year-old man with history of renal transplantation for diabetic nephropathy who presented with loss of appetite and weight loss. Physical exam showed a 7 cm hard nodular subcutaneous mass in the right upper arm. FNA yielded abundant acellular, irregular fragments of dense material, which was Congo red positive with apple green birefringence by polarized light, consistent with amyloid. Further subtyping of the amyloid by mass spectrometry, showed AIns (insulin)-type amyloid deposition. After further questioning, the patient admitted to injecting insulin at the same site for many years. Awareness of the cytological features is important for diagnosis. This is especially important when dealing with uncommon sites and without adequate clinical information.

Large quantities of organic dyes are discharged into the environment, causing serious damage to the ecosystem. Therefore, it is urgent to develop inexpensive adsorbents to remove organic dyes. A novel cellulose-based aerogel (MPPA) with 3D porous structure was prepared by using cassava residue (cellulose) as basic construction blocks, doping ferroferric oxide (Fe3O4) for magnetic separation, and applying polyethyleneimine (PEI) as functional material for highly efficient and selective capture of Congo red (CR). MPPA exhibited porous network structure, numerous active capture sites, nontoxicity, high hydrophilicity, and excellent thermal stability. MPPA showed superior adsorption property for CR, with an equilibrium adsorption capacity of 2018.14 mg/g, and still had an adsorption property of 1189.31 mg/g after five recycling procedures. In addition, MPPA has excellent selectivity for CR in four binary dye systems. The adsorption behavior of MPPA on CR was further explored using a multilayer adsorption model, EDR-IDR hybrid model and AOAS model. Electrostatic potential and independent gradient models were used to further verify the possible interaction between MPPA and CR molecules. In conclusion, MPPA is a promising adsorbent in the field of treating anionic dyes.

BACKGROUND: Amyloid, presenting as a mass, is termed amyloidoma. Among the reported cases, fine-needle aspiration (FNA) of amyloid is often misinterpreted as acellular nondiagnostic material. METHODS: A computer search of all FNAs was performed and cases diagnosed as amyloidoma were identified. RESULTS: Among 11,956 cases and 20,634 FNAs, there were six cases and 12 FNAs of amyloidoma. One case with mucin/myxoid matrix was misinterpreted as amyloid, which on our review was Congo red negative. All five other cases of amyloidoma were adequate for evaluation. The smears showed most of the aspirated contents in the middle of the slide and it did not spread when smeared. The amyloid was present as large chunks of waxy, smooth, orangophilic/cyanophilic fragments on Papanicolaou stain and as basophilic fragments on Diff-Quik stain in a clean background. In cases with lymphoma/myeloma, there were admixed lymphocytes and/or plasma cells. Unlike fibrous tissue, amyloid aspirates well and provides adequate material for interpretation. The clean background distinguishes it from mucin/myxoid matrix. Congo red stain was positive with apple green birefringence in all five cases. Further subtyping by mass spectrometry showed AL (κ) type in three patients and AIns (insulin) type in one patient. In one patient with lymphoma, the subtyping was not done. CONCLUSION: FNA of amyloidoma is rare (0.04%), but an optimal method for diagnosis and subtyping, avoiding unwanted surgical interventions. Although mistaken for fibrous tissue, which aspirates poorly, abundant acellular orangophilic/cyanophilic material on FNA should raise a suspicion for amyloid. Unlike mucin/myxoid matrix, amyloid does not smear the background.

A woman in her 60s with rheumatoid arthritis was admitted with fever and abdominal pain. Laparoscopic examination with the differential diagnosis of peritoneal neoplasm and infection revealed granulomatous phlebitis in the resected greater omentum. Amorphous eosinophilic deposits observed in the resected tissue exhibited focal, weak positivity for Congo red but were strongly positive for thioflavin S, confirming their focal amyloid properties. Marked degeneration of elastic fibers was also evident. Electron microscopy revealed deposits around the affected elastic fibers. Immunohistochemistry revealed the deposition of epidermal growth factor-containing fibulin-like extracellular matrix protein 1 (EFEMP1) along with T-cell-predominant lymphocytic inflammation. The definitive diagnosis was granulomatous enterocolic lymphocytic phlebitis (ELP) associated with EFEMP1 deposition exhibiting focal amyloid properties (EFEMP1/AEFEMP1), supported by proteomics analysis. This type of vasculitis is similar to amyloid-ß-related angiitis of the central nervous system. Thus, we speculate that granulomatous ELP also results from an immune response that recognizes EFEMP1/AEFEMP1 deposits as foreign material and attempts to remove them. Confirmation of EFEMP1/AEFEMP1 deposition with Congo red staining is challenging, particularly in the presence of inflammation, and warrants comprehensive evaluation.

The primary aim of this investigation was to synthesise novel adsorbent by incorporating greenly synthesized zinc oxide nanoparticles into chitosan matrix (G-ZnO-Cs). The production of ZnO Nanoparticles via a green approach involved the utilization of extracts derived from Nymphaeaceae fronds. This assertion was substantiated by the application of Field Emission Scanning Electron Microscopy (FESEM) and X-ray Diffraction (XRD) analytical techniques. Several Analytical methods such as Fourier Transform Infrared spectroscopy (FT-IR), Energy Dispersive X-ray Analysis (EDAX), FESEM, Thermogravimetric Analysis (TGA), XRD, Brunauer-Emmett-Teller (BET) analysis, and point-of-zero charge determination were used to characterize G-ZnO-Cs. Further study investigates the impact of five key processing parameters, namely pH, interaction duration, G-ZnO-Cs dosage, temperature, and initial concentration of dyes, on the removal of three organic dyes Reactive Blue 19 (RB 19), Reactive Orange 16 (RO 16), and Congo Red (CR) The adsorption process of Reactive Blue 19 (RB 19), Reactive Orange 16 (RO 16), and Congo Red (CR) dyes on G-ZnO-Cs were determined to comply to the pseudo-second-order (PSO) and Langmuir models, as determined through equilibrium and kinetic experiments. The highest adsorption capabilities for RB 19, RO 16 and CR dye were revealed to be 219.6 mg/g, 129.6 mg/g, and 118.8 mg/g, respectively. The elimination success rate of the fixed-bed column approach for treating huge volumes was highlighted in the conducted research. Moreover, the G-ZnO-Cs composite exhibited significant reusability due to its ability to undergo elution and simultaneous regeneration processes.

Preeclampsia is a disorder that can occur during pregnancy and is one of the leading causes of death among pregnant women. This disorder occurs after the 20th week of pregnancy and is characterized by arterial hypertension, proteinuria, fetoplacental, and multiple organ dysfunctions. Despite the long history of studying preeclampsia, its etiology and pathogenesis remain poorly understood, and therapy is symptomatic. One of the factors of the disorder is believed to be misfolded proteins that are prone to form amyloid aggregates. The CRD tests, utilizing the binding of the amyloid-specific dye Congo red to urine components, demonstrate high efficiency in diagnosing preeclampsia. However, these tests have also been found to be positive in other disorders with proteinuria, presumably associated with concomitant amyloidosis. To assess the limitations of the CRD tests, we examined urine congophilia and protein components mediating Congo red positivity in patients with proteinuria, including preeclampsia, amyloid and non-amyloid nephropathies. We stained the urine samples and calculated congophilia levels. We also assessed the contribution of large protein aggregates to congophilia values using ultracentrifugation and determined the molecular weights of congophilic urinary proteins using centrifugal concentrators. All proteinuric groups demonstrate positive results in the CRD tests and congophilia levels were more than two times higher compared with the control non-proteinuric groups (p <0.01). There was a strong correlation between urine protein excretion and congophilia in amyloid nephropathy (rs = 0.76), non-amyloid nephropathies (rs = 0.90), and preeclampsia (rs = 0.90). Removal of large aggregates from urine did not affect the congophilia levels. Separation of urine protein fractions revealed congophilic components in the range of 30-100 kDa, including monomeric serum albumin. Our results indicate limitations of CRD tests in preeclampsia diagnostics in women with renal disorders and underscore the need for further research on the mechanisms of Congo red binding with urine components.
