This study describes which antigens of Fasciola hepatica are present in the feces of patients with chronic fascioliasis and in the feces of rats infected experimentally with F. hepatica metacercariae. Using a Western blot assay technique with hyperimmune serum obtained from excretory-secretory antigens of adult F. hepatica, we found in the patients' feces antigens of possible diagnostic interest, with molecular weights of 14, 19, 20, 23, 25, 32, 46, 51, and 62 kilodaltons (kDa). In addition, we showed that the peptides of 14, 20, 23, and 51 kDa are also recognized by the majority of the sera from chronic patients. We used affinity chromatography to purify the antigens present in the feces of rats that had been infected for 6 to 12 weeks, using ES78 monoclonal antibody bound to CNBr-activated Sepharose 4B. Through that approach, we identified six polypeptides, of 11, 14, 26, 32, 47, and 51 kDa; three more polypeptides, of 17, 24, and 66 kDa, could only be identified in the feces of rats that had been infected for 10 to 12 weeks. Our results suggest that these polypeptides could be antigens common to both parasitic stages. This is particularly true for the polypeptides of 14, 24, 26, and 51 kDa, because they reacted with the immune sera, the human sera, and the ES78 monoclonal antibody. These polypeptides could be important markers for acute and chronic fascioliasis.

The monoclonal antibody ES78 was used in a sandwich immunosorbent assay (Sandwich ELISA) for the detection of antigens in sera and faeces in the course of Fasciola hepatica infection in 10 experimentally infected sheep. All infected sheep had circulating antigens in the first week post-infection (WPI). Antigenemia was detectable until WPI 3 in four infected sheep, WPI 4 in five infected sheep and in only one sheep by WPI 5. The detection of coproantigens (Fa(g)) was possible in five infected sheep at WPI-4, in four sheep at WPI-5 and in one sheep only at WPI-6. This technique was compared to an indirect ELISA for the detection of antibodies using excretory secretory antigens of F. hepatica. A significant correlation was found between Fa(g) and egg output and also with adult worm numbers. Our method demonstrated that the diagnosis of active fasciolosis in sheep is possible during all periods of infection.

The monoclonal antibody-based sandwich immunoassay (mAb Sandwich ELISA) was used to evaluate the effectiveness of triclabendazole treatment by the detection of coproantigens in Fasciola hepatica naturally infected sheep. Twelve sheep (2 to 5 years of age) were separated into two groups. The first group (three sheep) remained untreated; the other group (nine sheep) was treated with a single dose of 5% triclabendazole at 10 mg kg-1 of body weight. All but one of the treated group had negative optical density values (OD492) after two weeks of treatment, while seven sheep intermittently shed eggs during the course of the study. In all but one of the treated sheep, no F. hepatica infection; the one positive ELISA in 5th week after treatment according to the mAb Sandwich, had one fluke in the liver. The results of the parasitological examinations, as well as OD492 values obtained by the mAb Sandwich ELISA for the detection of coproantigens are described. The findings at necropsy, of the treated group in comparison to the untreated group are shown. The mAb Sandwich ELISA could be a useful and accurate method with which to monitor the efficacy of flukicides in F. hepatica natural infections.

The standardization of an ultramicroELISA for the detection of IgG antibodies anti excretory-secretory antigens of Fasciola hepatica (UME-Fasciola) is described. It was studied a considerable group of sera of which 56 were from patients with fascioliasis, 168 from patients with other parasitic diseases, and 300 from sound persons that were used as negative controls. As regards the parasitology test considered as "Gold Standard", the UME-Fasciola showed a sensitivity of 100%, an specificity of 98%, and predictive values for positives and negatives of 90.3% and 100%, respectively. Cross-reaction was only observed with the sera from patients infected with Opistorchis felineus. On comparing the UME-Fasciola with the conventional ELISA, it was obtained a concordance index of 95.5%.

A monoclonal antibody-based sandwich immunoassay (mAb sandwich ELISA) was developed for the detection of Fasciola hepatica antigen in the faeces of cattle. The assay was applied to samples from 100 cattle infected with F hepatica, 56 animals with parasitologically proven infections of other parasites and 100 uninfected animals. F hepatica antigen was detected in all the faecal samples from animals with fasciolosis, but none of the samples from the uninfected animals or from those with other parasitic infections had significant levels of F hepatica antigens. The results indicate that the mAb sandwich ELISA is a rapid, simple and useful method for the diagnosis of active F hepatica infection in cattle.

Toxocara canis is a commun ascarid of dogs worldwide. The ingestion of helminths eggs by humans may cause visceral larva. Knowing the prevalence of T. canis in domestic dogs from Havana City (17.9%), it was decided to study for the first time the contamination of soil in the city with canine nematode ova, and thus, understand the magnitude of the rich for our population, thus, mainly children, for developing the disease three soil samples were taken in different residential areas from 15 municipalities of Havana City, and they were processed by Willis flotation technique (1921). Nineteen out of 45 samples were positive to T. canis eggs accounting for 42.2% of prevalence. Only 3 municipalities were found to have negative results. 38.2% of egg were embryonate. Findings are discussed.

A study on the prevalence of Toxocara canis in domestic dogs of City of Havana was carried out for the first time in Cuba. 22 dogs were selected in each municipality, totalling 330 as the study universe. Dogs' feces samples were processed through direct examination using Willis' flotation method (1921). The results attained showed a 17.9% prevalence of Toxocara canis, a high figure constituting an important risk factor for transmission to humans. This was influenced by the age of the animals and the hygienic conditions of the houses, significant factors from the epidemiological point of view. Sex did not play a significant role.

RESUMO

Four antigenic polypeptides present in excretion-secretion antigens which were common to Fasciola hepatica somatic and tegumentary antigens, were identified by Western blotting and purified by affinity chromatography. Monoclonal antibody ES78 was used to that purpose. Molecular weights calculated for these polypeptides ranged from 37 to 13 kd; they proved to be highly reactive with sera from animals experimentally infected by Fasciola hepatica in periods as early as the second week of infection, with maximal values in weeks 6 and 10.

The presence of Dirofilaria immitis excretory-secretory (ES) products was detected in the urine of infected dogs using a coagglutination assay. Urine samples from 30 naturally infected dogs were positive. Seventeen of them were microfilaremic, whereas 13 had become amicrofilaremic after receiving 2 courses of diethylcarbamazine. Urine samples from 20 dogs infected with other parasites, Dipetalonema reconditum (7), Toxocara canis (5), and Ancylostoma caninum (8), and urine samples from 20 healthy dogs were negative. The assay detected 200 ng/ml or more of ES products. This assay is simple, easy to perform with minimum training, and requires no equipment. Therefore it should be useful to detect canine filariasis under field conditions.

In sera from patients with fascioliasis the enzyme-linked immunosorbent assay (ELISA) was used to detect antibody using excretory-secretory products (ES) from Fasciola hepatica adult worms. The specificity of ES-ELISA (with OD values greater than 0.38) allowed the differentiation among fascioliasis, schistosomiasis, clonorchiasis, and other human parasite infections.

Se describe el primer hallazgo de microfilarias de Dipetalonema reconditum (Grassi, 1890) en un perro en Cuba. La identificacion se baso en el estudio morfologico de 50 microfilarias aisladas mediante la tecnica de Knott, de la sangre de un perro de Ciudad de La Habana, en 1982.El estudio de la mitad del material se hizo con microfilarias fijadas despues de la concentracion y la otra mitad tenidas con Giemsa.Nuestro trabajo demuestra que, ademas de los datos del largo y ancho, la presencia del cuerpo refractil y de la cola en forma de gancho son caracteristicas que permiten una inmediata identificacion de estas microfilarias

Dado el hallazgo en Cuba y por primera vez en America del Angiostrongylus cantonensis nos propusimos determinar la presencia de este parasito en las provincias habaneras.Se colectaron 2.720 moluscos y 20 ratas.Se obtuvieron 16 especies de moluscos de los cuales, 10 fueron positivos. En el 90% de las ratas capturadas se encontraron adultos de A. cantonensis. En el 100% y 89,5% de los municipios de C. Habana y Habana, respectivamente, se hallaron moluscos con larvas de segundo y tercer estadio o ambos inclusive. Se reporta una nueva subespecie de moluscos positivos. (Bradybaena similaris honkonensis) y una nueva especie de rata (Rattus rattus)
