OBJECTIVE: To assess the economic burden of acute stroke, and to determine the average cost of acute stroke care for a single hospital stay in a public tertiary care hospital. METHODS: The cross-sectional study was conducted at the Medical Teaching Institute, Bacha Khan Medical Complex, Swabi, Pakistan, from May 16 to September 19, 2022, and comprised patients of either gender who were hospitalised with an acute stroke for the first time. All costs incurred during the care of the patients were measured using the micro-costing methodology, and the association of the cost with other variables was evaluated. Data was analysed using SPSS 24. RESULTS: Of the 34 patients, 24(70.6%) were males and 10(29.4%) were females. The overall mean age was 66+/-13.00 years. The mean length of hospital stay was 4+/-3.00 days. The mean total cost was 18,156+/-9,068 Pakistani rupees, which was the equivalent of 76.89+/-38.4 United States dollars. The cost of the first day of admission was the highest, declining per day as the stay progressed, and imaging/laboratory investigations formed the highest component of the overall cost (p<0.001). CONCLUSIONS: The cost of acute stroke care was found to be high even in a public hospital. The length of hospital stay was the most important determinant of the overall cost.

In aging kidneys, a decline of function resulting from extracellular matrix (ECM) deposition and organ fibrosis is regarded as "physiological." Whether a direct link between high salt intake and fibrosis in aging kidney exists autonomously from arterial hypertension is unclear. This study explores kidney intrinsic changes (inflammation, ECM derangement) induced by a high-salt diet (HSD) in a murine model lacking arterial hypertension. The contribution of cold shock Y-box binding protein (YB-1) as a key orchestrator of organ fibrosis to the observed differences is determined by comparison with a knockout strain (Ybx1ΔRosaERT+TX). Comparisons of tissue from mice fed with normal-salt diet (NSD, standard chow) or high-salt diet (HSD, 4% NaCl in chow; 1% NaCl in water) for up to 16 mo revealed that with HSD tubular cell numbers decrease and tubulointerstitial scarring [periodic acid-Schiff (PAS), Masson's trichrome, Sirius red staining] prevails. In Ybx1ΔRosaERT+TX animals tubular cell damage, a loss of cell contacts with profound tubulointerstitial alterations, and tubular cell senescence was seen. A distinct tubulointerstitial distribution of fibrinogen, collagen type VI, and tenascin-C was detected under HSD, transcriptome analyses determined patterns of matrisome regulation. Temporal increase of immune cell infiltration was seen under HSD of wild type, but not Ybx1ΔRosaERT+TX animals. In vitro Ybx1ΔRosaERT+TX bone marrow-derived macrophages exhibited a defect in polarization (IL-4/IL-13) and abrogated response to sodium chloride. Taken together, HSD promotes progressive kidney fibrosis with premature cell aging, ECM deposition, and immune cell recruitment that is exacerbated in Ybx1ΔRosaERT+TX animals.NEW & NOTEWORTHY Short-term experimental studies link excessive sodium ingestion with extracellular matrix accumulation and inflammatory cell recruitment, yet long-term data are scarce. Our findings with a high-salt diet over 16 mo in aging mice pinpoints to a decisive tipping point after 12 mo with tubular stress response, skewed matrisome transcriptome, and immune cell infiltration. Cell senescence was aggravated in knockout animals for cold shock Y-box binding protein (YB-1), suggesting a novel protective protein function.

Until now, no specific and effective treatment exists for coronavirus disease 2019 (COVID-19). Since honey and Nigella sativa (HNS) have established antiviral, antibacterial, antiinflammatory, antioxidant, and immunomodulatory properties, we tested their efficacy for this disease in a multicenter, placebo-controlled, and randomized clinical trial at four medical care facilities in Pakistan. RT-PCR confirmed COVID-19 adults showing moderate or severe disease were enrolled in the trial. Patients were randomly assigned in a 1:1 ratio to receive either honey (1 g kg-1 day-1 ) and Nigella sativa seeds (80 mg kg-1 day-1 ) or a placebo for up to 13 days along with standard care. The outcomes included symptoms' alleviation, viral clearance, and 30-day mortality in the intention-to-treat population. Three hundred and thirteen patients, 210 with moderate and 103 with severe disease, underwent randomization from April 30 to July 29, 2020. Among the moderate cases, 107 were assigned to HNS, whereas 103 were assigned to the placebo group. Among the severe cases, 50 were given HNS, and 53 were given the placebo. HNS resulted in ~50% reduction in time taken to alleviate symptoms as compared to placebo (moderate cases: 4 vs. 7 days, Hazard Ratio [HR]: 6.11; 95% Confidence Interval [CI]: 4.23-8.84, p < 0.0001 and for severe cases: 6 vs. 13 days, HR: 4.04; 95% CI: 2.46-6.64; p < 0.0001). HNS also cleared the virus earlier than placebo in both moderate cases (6 vs. 10 days, HR: 5.53; 95% CI: 3.76-8.14, p < 0.0001) and severe cases (8.5 vs. 12 days, HR: 4.32; 95% CI: 2.62-7.13, p < 0.0001). HNS further led to a better clinical score on day 6 with normal activity resumption in 63.6% vs. 10.9% among moderate cases (OR: 0.07; 95% CI: 0.03-0.13, p < 0.0001) and hospital discharge in 50% versus 2.8% in severe cases (OR: 0.03; 95% CI: 0.01-0.09, p < 0.0001). In severe cases, the mortality rate was less than 1/4th in the HNS group than in placebo (4% vs. 18.87%, OR: 0.18; 95% CI: 0.02-0.92, p = 0.029). No HNS-related adverse effects were observed. HNS, compared with placebo, significantly improved symptoms, expedited viral load clearance, and reduced mortality in COVID-19 patients. This trial was registered on April 15, 2020 with ClinicalTrials.gov Identifier: NCT04347382.

The aim of the study was to compare three moulting procedures and their effect on productive performance, egg quality, and antibody response of leghorn hens. For this, a total of 324 laying hens were distributed into three treatment groups having 12 replicates of 9 birds each according to completely randomized design. Treatments consisted of three moulting procedures based on feed and light restriction. The targeted weight at the end of moulting was 1450-1470 g. As soon as the moulting procedure is complete, the comparative analysis of post-moult productive performance (feed intake, egg production, egg weight, egg mass, feed per dozen eggs, feed per kg egg mass, livability), egg quality characteristics (egg weight, egg length and width, shape index, surface area, volume, albumen height, weight, Haugh unit score, yolk width, height, index, egg shell pore number, shell weight, thickness, breaking strength), and antibody response against Newcastle disease and avian influenza (H-9) were evaluated. Birds subjected to moulting procedure 3 (8 days fasting and gradual decrease in light) showed improvement in productive performance, egg geometry and quality traits, and antibody response against Newcastle disease virus. Birds experienced moulting procedure 1 (11 days fasting) had improved feed intake, egg production, and livability. However, birds moulted with procedure 2 (6 days fasting) revealed intermediate result in all the studied parameters. In conclusion, moulting through feed and light restriction with 8 days fasting and gradual reduction in light has potential to improve performance of leghorn hens.

Present study evaluated the effect of lighted incubation on pre- and post-hatch performance of Japanese quail. In a completely randomized design, 1200 eggs were evenly divided into 4 treatments groups having six replicates (each tray was considered as replicate), 50 eggs each. Different dichromatic lights (Green + Red; GR, Green + Blue; GB, and Blue + Red; BR) of 250 lux were provided during incubation for 12 h daily and effects of these lights very evaluated on hatching results and post-hatch growth. After hatch, 600 quail chicks were divided into 4 treatments, 6 replicates, and 25 birds each. Regarding hatching traits, better hatchability was found in the GR group compared to GB, BR, and dark group; while early embryonic mortality was lower in BR, GB, and dark group than GR; mid embryonic mortality was lower in dark group and late embryonic mortality was noted in the GR group than those of other treatment groups. In addition, moisture loss during incubation was minimum in BR and dark groups; however, chick spread was better in the GR group. In terms of growth performance, weight gain was better in the GR group; feed intake in dark, feed conversion ratio in BR, and livability were better in BR and GR group. In morphometrics, keel and shank length were higher in all the colored groups (GB, BR, and GR) whereas body length, wing spread, shank circumference, drumstick length, and circumference were higher in the GR group. Regarding serum chemistry, glucose, albumin, and globulin levels were higher in the GR group. It was concluded that under the experimental conditions, GR light at the prenatal stage to Japanese quail eggs positively influenced hatching performance and post-hatch growth.

The study aims to evaluate dichromatic light with different intensities during incubation on pre- and post-hatch performance of Japanese quail. In completely randomized design, 600 quail hatching eggs were evenly divided into 4 treatments, 3 replicates, and 50 eggs in each. These eggs were provided with lighted incubation [dichromatic light; green-red (GR) with three different intensities 150, 250, and 350 lx] with lighting schedule (12L: 12D), and one treatment was the complete dark period during incubation and considered as control group. After hatch, 300 quail chicks were equally divided into four treatments (3 replicates and 25 chicks in each). Effects of GR light with different intensities during incubation were evaluated on hatching traits and post-hatch performance of Japanese quails. Regarding hatching traits, hatchability was better in all the treated groups (150, 250, and 350 lx of GR light during incubation) compared to complete dark. In terms of embryonic mortality, early embryonic mortality was lower in the 250 lx group, mid embryonic mortality in dark, and late embryonic mortality in the 150 lx group. Furthermore, an ideal chick spread and growth performance (weight gain, feed intake, feed conversion ratio, and liveability) was reported in 250 lx group compared to other treated groups. Serum chemistry values were higher in dark group compared to 150, 250, and 350 lx groups. Pre-slaughter weight was higher in 250 and 350 lx group than 150 lx and dark group, whereas carcass weight was better in 350 lx group compared to dark. In conclusion, stimulation of dichromatic light (green + red) at 250 lx to Japanese quail eggs positively influenced hatching traits and post-hatch growth performance of Japanese quail.

RESUMO

Biliary cystadenoma, a rare potentially malignant hepatic cystic lesion, is characterized by multiloculations and septations. It is common in middle-aged females (about 5% of nonparasitic liver cysts); only 12 cases are described in children. We report a rare case of hepatic biliary cystadenoma in a 3-year-old girl, with a gradually increasing lump in the right upper abdomen. Complete excision with a healthy liver margin was done.

Autosomal primary microcephaly (MCPH) is a heterogenetic disorder that affects brain's cerebral cortex size and leads to a reduction in the cranial vault. Along with the hallmark feature of reduced head circumference, microcephalic patients also exhibit a variable degree of intellectual disability as well. Genetic studies have reported 28 MCPH genes, most of which produce microtubule-associated proteins and are involved in cell division. Herein this study, 14 patients from seven Pashtun origin Pakistani families of primary microcephaly were analyzed. Mutation analysis was performed through targeted Sanger DNA sequencing on the basis of phenotype-linked genetic makeup. Genetic analysis in one family found a novel pathogenic DNA change in the abnormal spindle microtubule assembly (ASPM) gene (NM_018136.4:c.3871dupGA), while the rest of the families revealed recurrent nonsense mutation c.3978G>A (p.Trp1326*) in the same gene. The novel reported frameshift insertion presumably truncates the protein p.(Lys1291Glyfs*14) and deletes the N-terminus domains. Identification of novel ASPM-truncating mutation expands the mutational spectrum of the ASPM gene, while mapping of recurrent mutation c.3978G>A (p.Trp1326*) will aid in establishing its founder effect in the Khyber Pakhtunkhwa (KPK) inhabitant population of Pakistan and should be suggestively screened for premarital counseling of MCPH susceptible families. Most of the recruited families are related to first-degree consanguinity. Hence, all the family elders were counseled to avoid intrafamilial marriages.

Breynia distachia is a plant of genus Breynia belonging to family Phyllanthaceae. This study was conducted to isolate and examine the anti-inflammatory attributes of the roots of Breynia distachia. Methanol extract from roots were prepared by simple maceration. For phytochemical studies, isolation, purification, structure elucidation, metal analysis, total phenolic content, and solubility test were done by chromatographic and spectroscopic techniques. Anti-inflammatory activity was evaluated by cotton pallet edema model and carrageenan paw edema model, and antioxidant potential was evaluated by DPPH, FRAP, and ABTS antioxidants assays. Metal analysis of BD.Me revealed the presence of Na > Mg > K > Mn > Fe = Zn in respective order. Four phytochemicals such as gallic acid, quercetin, sinapic acid, and p-coumaric acid are found in Breynia distachia. Quercetin is present in relatively larger quantity, and shows antioxidant activity by reducing the ferric iron to ferrous iron. Novel distachionate shows high antioxidant activity in ABTS assay by reducing reactive oxygen species. Quantitative or qualitative analysis performed by HPLC indicates the ascending peaks or presence of secondary products (metabolites) respectively. Histopathology analysis of liver, spleen, heart, and kidney was done, revealing mild inflammations in spleen and liver, and no cytotoxicity in heart and kidney. Oral administration of BD.Me and ditachionate significantly inhibits the carrageenan and cotton pellet-induced paw edema in 1st and 2nd h with (ns = p > 0.05) than control. After 3rd, 4th, 5th, and 6th h, BD.Me and ditachionate showed inhibition of paw edema in a highly significant (*** = p < 0.001) manner as compared to control. In cotton-pellet edema model, distachionate shows a %inhibition of 57.3% at a dose level of 5 mg/kg. Docking values obtained from distachionate-COX-2 complex suggest a potent inhibitor evaluated for this protein. The distachionate shows effective anti-inflammatory activity. Methanol extracts of roots showed significant lipoxygenase inhibitory activity by IC50 values of 155.7 ± 0.55 and 132.9 ± 0.33 µg/mL. Data from various in vitro and in vivo models suggest that novel distachionate isolated from Breynia distachia shows strong antioxidant and anti-inflammatory activities; it should be further studied for the exploration of its medicinal potential.

Semen traits play the vital role in determining the fertility of a broiler breeder flock; however, it can be influenced by several factors. This experiment was carried out to assess some of these factors affecting the semen. A total of 89 male birds and 960 hens of 20-week-old broiler breeder (2215 g ± 7.5%) were divided into two main groups; one was kept in cages (AIC) and another group was kept on deep litter floor (AIF), while both these groups were subjected to AI. The male birds of aforementioned groups (44 males and 480 females) were further divided into 4 sub-groups (11 males and 120 females) to execute different semen collection frequencies i.e., 2, 3, 4, and 5th days' interval. The impact of time duration between semen collection and insemination on sperm kinematics was monitored. The quantitative and qualitative analysis of semen including sperm concentration and sperm kinematics of the collected semen was conducted through a computer-assisted sperm analyzer (CASA) and ONGO machine (working on the CASA principle). Resultantly, the data revealed that the studied parameters of semen were deteriorated with the progression of age of male birds, while the group of males with standard body weight produced the best semen quantitatively and qualitatively followed by overweight particularly during the post peak phase (46-65 = 20 weeks). Although the 3rd day, semen collection frequency was found better for quality, the higher quantity of semen was achieved when males were being collected at the intervals of 4th and 5th day respectively regardless of housing systems. Significant decline in sperm kinematics was recorded with the progression of semen holding duration at the temperature of poultry farm. Furthermore, the highest contamination of E. coli, Salmonella, and Mycoplasma gallisepticum was recorded in the reproductive tract of hens and semen of the AIF group as compared to AIC. Thus, conclusion can be settled that the semen properties are significantly affected by age, body weight, and semen collection intervals in both housing systems, while sperm kinematics is being disrupted with the progression of holding duration. Although housing systems could affect the semen insignificantly, yet lesser contamination was recorded in semen and in the reproductive tract of hens of AIC.

This study evaluated the effect of amino acids balanced at a specific lysine ratio with exogenous protease supplementation and reduced CP levels in the broiler diet. In total, 480 straight-run broiler chicks (Ross-308) were used for the study. A completely randomized design in the factorial arrangement was applied, with 4 treatments of 6 replicates with 20 birds each. Treatments consisted of 2 lysine ratios (100% and 110%; factor 1) without or with exogenous protease supplementation (200 g/ton; factor 2). Twenty percent reduction of CP from standard requirements of Ross-308 (18.4% vs 23% in starter and 17.2% vs 21.5% in grower phase) was made in all the treatment diets. Growth performance (feed intake, weight gain, feed conversion ratio, livability), carcass traits (live and carcass weight, carcass yield, heart, liver, gizzard, leg quarter, breast weight, and abdominal fat percentage), gut morphology (villus height, crypt depth, and villus height and crypt depth ratio), and serum metabolites (total protein, albumin, and uric acid) were evaluated. A significant interaction (p ≤ 0.05) for lysine ratio and protease was noted in body weight gain, feed conversion ratio, carcass traits, and gut morphology throughout the experimental period yet a non-significant (p > 0.05) influence was observed on serum metabolites. It was concluded that the broiler diet with a net reduction of 20% CP from Ross-308 standards, balanced for amino acids at lysine ratio 110% and supplemented with exogenous protease, could be used to improve growth performance, intestinal health, and carcass traits.

This study evaluated the effect of environmental enrichment on the performance, behaviour and welfare aspects; blood biochemistry; carcass and meat quality traits of broiler chickens. A total of 450 straight run broiler chicks (Ross-308) were divided into 5 treatment groups having 6 replicates of 15 birds each under a completely randomised design. Treatments were environmental enrichment (EE) tools and consisted of C = control group; R = red ball for EE; G = green ball for EE; B = blue ball for EE and M = mirror for EE. These environmental enrichment tools were provided throughout the experimental period (0 to 35 days). Mean feed intake per bird was higher in all treatment groups except the blue balls group; weight gain and feed conversion ratio were better in the green and blue ball groups. Broiler chickens reared under different environmental enrichment were more active and they exhibited maintenance behaviour (preening, dust bathing and wing stretching, or scratching) more frequently. Regarding welfare traits, lower incidence of toe damage, footpad dermatitis and hock burn was observed in birds having different environmental enrichment tools as compared to the control group. The birds reared with red balls as environmental enrichment showed the lowest values for glucose, cholesterol, total protein, albumin and globulin amongst all the treatment groups. Birds reared with green balls had the highest body weight at slaughter, dressed weight, carcass yield and liver weight. Breast meat of environmentally enriched treated groups was lighter and had lower ultimate pH. It was concluded that the addition of environmental enrichment tools (visual, structural and plastic) motivates the birds for physical activities and improves the performance of broiler chickens.

An experimental group (Angew. Chem., Int. Ed., 2021, 60, 7013-7017) has recently demonstrated the synthesis of two-dimensional (2D) RuSe2, which was shown to hold promise for hydrogen evolution due to enhanced HER performance. Herein, we studied the effects of external factors (strain, electric field, and atom adsorption) on the electronic and magnetic properties of T'-RuX2 (X = S, Se) sheets using DFT+U calculations. The estimated carrier effective mass results show that electrons are heavier than holes for T'-RuX2 sheets. The band-gap increases when the bi-axial strain increases from -5% to -1% (-5% to 1%) for T'-RuS2(RuSe2) and then decreases beyond these strain points. We found that the T'-RuX2 sheets exhibit a semiconductor to metal transition under a maximum electric field strength of 10 V nm-1. It is revealed that the magnetic moment can be achieved in T'-RuX2via adsorption of Li, Na, and K atoms. The findings show that the AFM state is the preferred magnetic ground state for T'-RuS2 with adsorbed Li and Na, whereas FM is the magnetic ground state for the remaining atm-RuX2 systems. Interestingly, an indirect to direct transition of the band-gap for T'-RuS2 with adsorbed K was found whereas the remaining T'-RuX2 with adsorbed Li, Na and K atoms showed either half-metallic or metallic electronic properties. Our results can extend the application of T'-RuX2 sheets in actuating, optoelectronic and spintronic fields.

The early detection of damaged (partially broken) outdoor insulators in primary distribution systems is of paramount importance for continuous electricity supply and public safety. Unmanned aerial vehicles (UAVs) present a safer, autonomous, and efficient way to examine the power system components without closing the power distribution system. In this work, a novel dataset is designed by capturing real images using UAVs and manually generated images collected to overcome the data insufficiency problem. A deep Laplacian pyramid-based super-resolution network is implemented to reconstruct high-resolution training images. To improve the visibility of low-light images, a low-light image enhancement technique is used for the robust exposure correction of the training images. A different fine-tuning strategy is implemented for fine-tuning the object detection model to increase detection accuracy for the specific faulty insulators. Several flight path strategies are proposed to overcome the shuttering effect of insulators, along with providing a less complex and time- and energy-efficient approach for capturing a video stream of the power system components. The performance of different object detection models is presented for selecting the most suitable one for fine-tuning on the specific faulty insulator dataset. For the detection of damaged insulators, our proposed method achieved an F1-score of 0.81 and 0.77 on two different datasets and presents a simple and more efficient flight strategy. Our approach is based on real aerial inspection of in-service porcelain insulators by extensive evaluation of several video sequences showing robust fault recognition and diagnostic capabilities. Our approach is demonstrated on data acquired by a drone in Swat, Pakistan.

Light in terms of photo- and scoto-periods is the key ambient factor affecting the physiology of birds through establishing normal biological clock and circadian rhythms. In natural incubation light significantly influences embryonic development, however, at commercial setups eggs are incubated under a dark environment. Presently not a single commercial poultry hatchery is using light during incubation; hence, comprehensive studies are needed to address the industry for considering light as a potential embryonic growth stimulant. In the present study, white Light-emitting diodes (LEDs; 5000 K) were installed in the incubator and 250 lx light intensity was provided for 0, 12, and 24 h per day during the whole incubation period. A total of 900 broiler hatching eggs (Hubbard classic; from 58 weeks old parents) were randomly allocated to 3 treatment groups, having 5 replicates of 60 eggs each, a tray was considered as replicate during incubation and these eggs were incubated under standard incubation protocols. After hatching, a total of 300 chicks were picked and divided into 3 described treatments (0, 12, and 24 h of photo-stimulation to eggs during incubation) having 5 replicates of 20 birds each. The results indicated that incubation of eggs under 12 and 24 h of lighting significantly improved (P ≤ 0.05) hatch window, hatchability % (0.0002), a hatch of fertile % (0.001), and carcass yield % (0.0454). Embryonic mortality, dead germs, and dead in shell embryos were lower in eggs incubated under 12 h light. Significantly better FCR (0.0006), stress susceptibilities such as H/L ratio (0.0227), and physical asymmetry (0.0065) were observed among the birds incubated under 12 h light (P ≤ 0.05). In conclusion, an appropriate light stimuli (12 h) may help to improve hatching traits and post-hatch performance of commercial broiler.

Ultraviolet-sensitive syndrome is a rare skin disorder characterised by heterogeneous phenotypic spectrum of skin freckling, telangiectasia and acute sunburn. It usually has an autosomal recessive pattern. So far, only 18 patients from nine different families (Japanese, French, Israeli, Iranian and Pakistani) have been reported in scientific literature. Its precise prevalence is still unknown, but, according to an estimate, its prevalence ratio is 1:100,000 worldwide. Until now, only three genes have been reported to be involved in the syndrome; the Excision Repair Cross-Complementing, Group 6, the Excision Repair Cross-Complementing, Group 8 and the UV-Stimulated Scaffold Protein A (UVSSA). Among these genes, the last one is reported to be more prevalent among different ethnicities, including Pakistani. Physiologically, most of the syndrome genes are involved in the transcription-coupled nucleotide excision pathway. In order to reduce the disease severity, the patients are advised to use medicated skin moisturisers or sun-blocks, sunglasses and gloves, while going out in the sun to avoid sun exposure. The current narrative review was planned to discuss the molecular genetics and the mutational spectrum of the syndrome, and to describe the differential diagnosis of various related disorders in order to facilitate clinical researchers.

Superbenzene porphyrin conjugates find wide range of applications from nonlinear optical materials to semiconductors. Herein, we report the synthesis and characterization of 5,15-bis(3,5-di-tert-butylphenyl)-10,20-bis(pentaphenylphenyl)phenylporphyrin and its Zinc-metallated complex. Oxidative planarization of 5,15-bis(3,5-di-tert-butylphenyl)-10,20-bis(pentaphenylphenyl)phenylporphyrin and its metallated complex was carried out by using NOBF4 as an oxidizing agent. The formation of superbenzene porphyrin conjugates validates its Scholl type reactions. The laboratory-synthesized porphyrin conjugates were characterized experimentally using spectroscopic techniques such as 1H NMR, 13C NMR, electron spin resonance, and ultraviolet-visible spectroscopy for structural conformation. In addition, density functional theory calculations were carried out to validate the experimental results. The theoretical and experimental results show that the 4-(pentaphenylphenyl)phenyl ligand increases the stability, optical properties, and rate of planarization of synthesized porphyrins. The conjugates exhibited intense and distant electronic communication between two hexabenzocoronene sites, taking advantage of porphyrin as a π-spacer. The π-radical cation has also been found to be an intermediate in oxidative C-C bond formation. NICS calculations support such a conclusion.

PINA is a novel ATPase and DNA helicase highly conserved in Archaea, the third domain of life. The PINA from Sulfolobus islandicus (SisPINA) forms a hexameric ring in crystal and solution. The protein is able to promote Holliday junction (HJ) migration and physically and functionally interacts with Hjc, the HJ specific endonuclease. Here, we show that SisPINA has direct physical interaction with Hjm (Hel308a), a helicase presumably targeting replication forks. In vitro biochemical analysis revealed that Hjm, Hjc, and SisPINA are able to coordinate HJ migration and cleavage in a concerted way. Deletion of the carboxyl 13 amino acid residues impaired the interaction between SisPINA and Hjm. Crystal structure analysis showed that the carboxyl 70 amino acid residues fold into a type II KH domain which, in other proteins, functions in binding RNA or ssDNA. The KH domain not only mediates the interactions of PINA with Hjm and Hjc but also regulates the hexameric assembly of PINA. Our results collectively suggest that SisPINA, Hjm and Hjc work together to function in replication fork regression, HJ formation and HJ cleavage.

This study examined the analysis of personal and production performance of broiler farming in 10 cities of central Punjab (Lahore, Sheikhupura, Kasur, Okara, Sargodha, Sahiwal, Gujranwala, Gujrat, Nankana, and Kamalia) of Pakistan. These districts are densely populated and renowned for broiler production. Primary data were collected through a survey of 100 poultry farmers. Personal and production information were collected from the respondents. Descriptive statistics was employed in the form of averages and percentages. The comparison among different districts was made through one-way ANOVA technique followed by Tukey's HSD test. Parameters were observed significantly in experience of farmer, age, number of sheds, capacity of farm, and flock/annuum during the survey of Gujranwala and Kasur, while the high percentage of graduate farmers was observed in Lahore and Sheikhupura regions. Significant results were observed during the production performance feed efficiency and mortality in Okara and Kasur as well as maximum administration of both live and killed vaccine in Lahore through drinking water in Nankana. In the districts of Sheikhupura and Kamalia, farmers due to lake of education left their dead birds on the surface ground. In conclusion, this study pointed out the exact trend of modern broiler farming and its price fluctuation in Punjab (Pakistan) as well as the reason behind the losses to the farmers.
