RESUMO

Objectives: To investigate the clinical correlation between the expression of serum tumor necrosis factor-α (TNF-α) and inducible nitric oxide synthase (iNOS) as well as cognitive impairment and disease burden in patients with first-episode schizophrenia. Methods: A total of 102 patients with first-episode schizophrenia admitted to the Department of Psychiatry, Hainan Province Anning Hospital from June 2018 to June 2020 were randomly selected as the observation group, and 102 healthy people who underwent physical examination in our hospital during the same period were selected as the control group. Enzyme-linked immunosorbent assay (ELISA) was used to detect the expression level of serum TNF-α in each group, and the colorimetric method was adopted for detecting the expression level of serum iNOS in each group; Moreover, cognitive function of patients was assessed by MATRICS Consensus Cognitive Battery (MCCB), disease burden was assessed by Family Burden of Disease Scale (FBS), and the correlation between the expression of serum TNF-α and iNOS as well as cognitive impairment and disease burden was analyzed by Pearson method. Results: The serum levels of TNF-α, iNOS, trail making test scores and FBS scores of the observation group were significantly higher than those of the control group (p<0.05), while the scores of symbolic coding, verbal memory, maze, visual memory, and semantic fluency were significantly lower than those of the control group (p<0.05); Pearson correlation analysis results showed that the serum levels of serum TNF-α and iNOS in the observation group were positively correlated with the trail making test and FBS scores respectively (p<0.05), and negatively correlated with the scores of symbolic coding, verbal memory, maze, visual memory, and semantic fluency (p<0.05). Conclusions: Patients with first-episode schizophrenia have remarkably increased expression levels of serum TNF-α and iNOS, which is associated with cognitive impairment and disease burden.

RESUMO

Experimental studies have shown that overexpression of Rap guanine nucleotide exchange factor 1 (C3G) plays pro-survival and anti-apoptotic roles through molecule phosphorylated extracellular signal-regulated kinase1/2 (p-ERK1/2) in cardiomyocytes. However, it is still unclear if silencing of C3G may increase cell survival inhibition and apoptosis in cardiomyocytes, and whether C3G silence induced injuries are reduced by the overexpression of C3G through regulation of p-ERK1/2 and pro-apoptotic molecule Bax. In this study, the rat-derived H9C2 cardiomyocytes were infected with C3G small hairpin RNA interference recombinant lentiviruses, which silenced the endogenous C3G expression in the cardiomyocytes. Then, contrary experiments were conducted using C3G overexpression. The cell proliferation and apoptosis were analyzed in the cardiomyocytes which were treated with or without hypoxia/reoxygenation (H/R). Silencing of C3G leaded to significant increase in cell survival inhibition and apoptosis, combined with aggravated the injuries induced by H/R. Overexpression of C3G reduced the injuries induced by the silencing of C3G in the cardiomyocytes via regulation of p-ERK1/2 and Bax. In conclusion, our results provide new experimental evidence that silencing of C3G can increase cell survival inhibition and apoptosis in cardiomyocytes via regulation of p-ERK1/2 and Bax.

RESUMO

A simple route for the preparation of ZnS nanocrystal/reduced graphene oxide (ZnS/RGO) by a hydrothermal synthesis process was achieved. The chemical composition, morphology, and structural characterization reveal that the ZnS/RGO composite is composed of sphalerite-phased ZnS nanocrystals uniformly dispersed on functional RGO sheets with a high specific surface area. The ZnS/RGO composite was utilized as an anode in the construction of a high-performance lithium-ion battery. The ZnS/RGO composite with appropriate RGO content exhibits a high reversible specific capacity (780 mA h g-1), excellent cycle stability over 100 cycles (71.3% retention), and good rate performance at 2C (51.2% of its capacity when measured at a 0.1C rate). To further investigate this ZnS/RGO anode for practical use in full Li-ion cells, we tested the electrochemical performance of the ZnS/RGO anode at different cut-off voltages for the first time. The presence of RGO plays an important role in providing high conductivity as well as a substrate with a high surface area. This helps alleviate the typically problems associated with volume expansion and shrinkage during prolonged cycling. Additionally, the RGO provides multiple nucleation points that result in a uniformly dispersed film of nanosized ZnS that covers its surface. Thus, the high surface area RGO enables high electronic conductivity and fast charge transfer kinetics for ZnS lithiation/delithiation.

RESUMO

Integrin ß1 subunit and its downstream molecule integrin-linked kinase and focal adhesion kinase have been confirmed to be essential to cell survival and inhibition of apoptosis and hypoxia/reoxygenation (H/R)-induced injuries in cardiomyocytes. However, it is still unclear whether CrkL [v-crk avian sarcoma virus CT-10 oncogene homolog (Crk)-like], which acts also as a component of the integrin pathway, could also affect H/R-induced injuries in the cardiomyocytes. The rat-derived H9C2 cardiomyocytes were infected with a CrkL small hairpin RNA interference recombinant lentivirus, which knockdowns the endogenous CrkL expression in the cardiomyocytes. Apoptosis, cell proliferation and survival were examined in the H9C2 cardiomyocytes treated with either H/R or not. Results showed that knockdown of CrkL could significantly increase apoptosis and inhibition of the cell proliferation and survival and deteriorate the previously mentioned injuries induced by H/R. In contrast, overexpression of human CrkL could relieve the exacerbation of the previously mentioned injuries induced by CrkL knockdown in the H9C2 cardiomyocytes via regulation of Bax and extracellular signal-regulated kinase1/2 (p-ERK1/2). In conclusion, these results confirmed that knockdown of CrkL could deteriorate H/R-induced apoptosis and cell survival inhibition in rat-derived H9C2 cardiomyocytes via Bax and downregulation of p-ERK1/2. It implies that CrkL could mitigate H/R-induced injuries in the cardiomyocytes.

RESUMO

In the present study, alfalfa canopy reflectance was researched at alfalfa squaring period under different irrigation amount at the hutubi county grassland ecological station. Determining the spectral diagnostic model of alfalfa leaf moisture content was determined by spectrometry. The results showed that (1) The spectral reflectance of alfalfa canopy gradually decreases with the increase in the leaf water content in the near infrared. (2) The spectral inversion model of alfalfa leaf moisture content established by normalized reflectance spectra is superior to the original reflectance spectra, and the prediction model established in the 1,344-1,660 nm band has the lowest average relative error (7.8%). (3) In this study, the spectral diagnostic model of the leaf moisture content is: Y=0.962 - 7.560X1451 + 5.295X1473. The spectral prediction model of the alfalfa leaf moisture content can provide a basis for decision making for scientific irrigation of alfalfa.

RESUMO

Salinity stress significantly hampers cotton seed germination and seedling growth. Employing plant growth regulators stands out as an effective strategy to mitigate salt stress. In this study, we assessed the impact of varying concentrations of natural composite salt conditions (0%, 0.6%, and 1.2%) on cotton seed germination, seedling growth, and physiology. Additionally, we explored the effects of compound sodium nitrophenolate (CSN: 2 mg·L-1 and 10 mg·L-1), 24-epibrassinolide (EBR: 0.02 mg·L-1 and 0.1 mg·L-1), and gibberellic acid (GA: 60 mg·L-1 and 300 mg·L-1), against a control (CK: distilled water) group. The results indicate that with an increase in the composite salt concentration, the germination potential (GP) and germination rate (GR) of cotton seeds gradually decrease. Simultaneously, the fresh weight and root vitality of seedlings also correspondingly decrease, while the degree of membrane lipid peroxidation increases. Under high-salt (1.2%) conditions, soaking treatments with CSN and EBR significantly enhance both GP (45-59% and 55-64%) and GR (30-33% and 39-36%) compared to the CK. However, the GA treatment does not increase the GP and GR of cotton. Moreover, under high-salt (1.2%) conditions, CSN and EBR treatments result in the increased activities of superoxide dismutase (56-66% and 71-80%), peroxidase (20-24% and 37-51%), and catalase (26-32% and 35-46%). Consequently, cotton exhibits a relatively good tolerance to natural composite salts. Soaking treatments with CSN and EBR effectively improve cotton germination by enhancing antioxidant enzyme activities, thereby strengthening cotton's tolerance to salt stress. These findings offer new insights for enhancing the salt tolerance of cotton.

RESUMO

Excessive salt content in soil has adverse effects on cotton production, especially during the germination and seedling stages. γ-aminobutyric acid (GABA) is an important active substance that is expected to improve the resistance of plants to abiotic stresses. This study focused on two cotton cultivars (Gossypium hirsutum L.: Tahe 2 and Xinluzhong 62) and investigated the impact of exogenous GABA (0, 1, 2, 3, and 4 mM) on seed germination, seedling growth, and related morphological, physiological, and biochemical indicators under salt stress (150 mM NaCl). The results showed that salt stress significantly reduced the germination rate and germination index of cotton seeds (decreased by 20.34% and 32.14% for Tahe 2 and Xinluzhong 62, respectively), leading to decreased seedling height and biomass and causing leaf yellowing. Salt stress induced osmotic stress in seedlings, resulting in ion imbalance (marked reduction in K+/Na+ ratio) and oxidative damage. Under salt stress conditions, exogenous GABA increased the germination rate (increased by 10.64~23.40% and 2.63~31.58% for Tahe 2 and Xinluzhong 62, respectively) and germination index of cotton seeds, as well as plant height and biomass. GABA treatment improved leaf yellowing. Exogenous GABA treatment increased the content of proline and soluble sugars, with varying effects on betaine. Exogenous GABA treatment reduced the Na+ content in seedlings, increased the K+ content, and increased the K+/Na+ ratio (increased by 20.44~28.08% and 29.54~76.33% for Tahe 2 and Xinluzhong 62, respectively). Exogenous GABA treatment enhanced the activities of superoxide dismutase and peroxidase, and reduced the accumulation of hydrogen peroxide and malondialdehyde, but had a negative impact on catalase activity. In conclusion, exogenous GABA effectively improved cotton seed germination. By regulating osmoprotectant levels, maintaining ion homeostasis, and alleviating oxidative stress, GABA mitigated the adverse effects of salt stress on cotton seedling growth.

RESUMO

Different textured soils (sandy loam, silty clay loam and clay) from Manas County, Xinjiang were researched with indoor spectral reflectance, continuum removal was used to process soil spectra curve, and correlation analysis was made about normalized spectral reflectance and water content for modeling. The results show that different textured soils have reflectance in a order that clay>silty clay loam>sandy loam; the critical points of field capacity in sandy loam, silty clay loam and clay were 20.01%, 24.10% and 30.43% respectively, and water content was inversely proportional to spectral reflectance below such critical points while proportional above the points. Within 1390-1623 nm band, the negative correlation coefficients of soil water content and normalized spectra reflectance show better negative correlation and reaching significant levels, R2 of the model established for soil water content prediction exceeded mostly 0.8, and the average relative error of the model was 10%. The model could accurately reflect the soil moisture content, Its advantages such as accuracy, non-destruction and rapidness provide a new approach to measuring soil water content.

RESUMO

In this paper, the decay law of dissolved oxygen in the culture solution of cotton in the seedling stage, bud stage, and blossoming and boll-forming stage was studied via hydroponic cotton experiments, and then, the oxygen consumption and its rule of cotton were explored. The results showed that with the increase of the cultivation time, the dissolved oxygen concentration of the cotton at seedling stage, bud stage, and blossoming and boll-forming stage showed an exponential function decrease. The dissolved oxygen concentration of the culture medium decreased from about 7.5 mg·L-1 to a relatively stable dissolved oxygen concentration, which was 4.92 mg·L-1, 1.64 mg·L-1, and 1.82 mg·L-1, respectively. When CK is about 7.1 mg·L-1, a relative equilibrium point between low oxygen absorption rate of cotton and oxygen supply is reached. The sudden drop points of the instant oxygen consumption rate of cotton seedling stage and bud stage are at about 5.15 mg·L-1 and 5.89 mg·L-1, respectively, while the sudden drop point was not obvious at the blossoming and boll-forming stage. The budding cotton can absorb oxygen when the dissolved oxygen in the culture solution is at a higher level. The blossoming and boll-forming stage cotton is more resistant to hypoxia stress.

RESUMO

The abuse of antibiotics would seriously affect human health and has become of worldwide critical concern, thus it is urgent to develop an environmentally friendly and nontoxic fluorescent probe for antibiotics sensing. In this work, a lead-free Cs2ZnCl4 perovskite nanocrystals (PNCs) probe was fabricated for sensing norfloxacin (NOR) employing a modified ligand-assisted reprecipitation method. The prepared Cs2ZnCl4 PNCs probe had strong blue emission around 440 nm, and the characteristics of PNCs were systematically characterized by X-ray photoelectric spectroscopy (XPS), Fourier transforms infrared spectroscopy (FTIR), transmission electron microscope (TEM) and powder X-ray diffraction (XRD). The results revealed that the fluorescence intensity of the Cs2ZnCl4 PNCs was significantly enhanced after the introduction of norfloxacin. The Cs2ZnCl4 PNCs can be used as a fluorescent probe to selectively and sensitively detect norfloxacin in the concentration range from 0.2 to 50.0 µM, with a correlation coefficient (R2) of 0.9954 and the limit of detection (LOD, 3σ) of 0.1499 µM. The preparation and application of a lead-free perovskite fluorescent probe for norfloxacin would promote the application of perovskite fluorescent probes in biochemical assays.
