BACKGROUND: The current method to evaluate the autonomic balance after renal denervation (RDN) relies on heart rate variability (HRV). However, parameters of HRV were not always predictive of response to RDN. Therefore, the complexity and disorder of heart rhythm, measured by entropy of entropy (EoE) and average entropy (AE), have been used to analyze autonomic dysfunction. This study evaluated the dynamic changes in autonomic status after RDN via EoE and AE analysis. METHODS: Five patients were prospectively enrolled in the Global SYMPLICITY Registry from 2020 to 2021. 24-h Holter and ambulatory blood pressure monitoring (ABPM) was performed at baseline and 3 months after RDN procedures. The autonomic status was analyzed using the entropy-based AE and EoE analysis and the conventional HRV-based low frequency (LF), high frequency (HF), and LF/HF. RESULTS: After RDN, the ABPM of all patients showed a significant reduction in blood pressure (BP) and heart rate. Only AE and HF values of all patients had consistent changes after RDN (p < 0.05). The spearman rank-order correlation coefficient of AE vs. HF was 0.86, but AE had a lower coefficient of variation than HF. CONCLUSIONS: Monitoring the AE and EoE analysis could be an alternative to interpreting autonomic status. In addition, a relative change of autonomic tone, especially an increasing parasympathetic activity, could restore autonomic balance after RDN.

Surface spectral reflectance is useful for color reproduction. In this study, the reconstruction of spectral reflectance using a conventional camera was investigated. The spectrum reconstruction error could be reduced by interpolating camera RGB signals, in contrast to methods based on basis spectra, such as principal component analysis (PCA). The disadvantage of the interpolation method is that it cannot interpolate samples outside the convex hull of reference samples in the RGB signal space. An interpolation method utilizing auxiliary reference samples (ARSs) to extrapolate the outside samples is proposed in this paper. The ARSs were created using reference samples and color filters. The convex hull of the reference samples and ARSs was expanded to enclose outside samples for extrapolation. A commercially available camera was taken as an example. The results show that with the proposed method, the extrapolation error was smaller than that of the computationally time-consuming weighted PCA method. A low cost and fast detection speed for spectral reflectance recovery can be achieved using a conventional camera.

The recovery of surface spectral reflectance using the quadcolor camera was numerically studied. Assume that the RGB channels of the quadcolor camera are the same as the Nikon D5100 tricolor camera. The spectral sensitivity of the fourth signal channel was tailored using a color filter. Munsell color chips were used as reflective surfaces. When the interpolation method or the weighted principal component analysis (wPCA) method is used to reconstruct spectra, using the quadcolor camera can effectively reduce the mean spectral error of the test samples compared to using the tricolor camera. Except for computation time, the interpolation method outperforms the wPCA method in spectrum reconstruction. A long-pass optical filter can be applied to the fourth channel for reducing the mean spectral error. A short-pass optical filter can be applied to the fourth channel for reducing the mean color difference, but the mean spectral error will be larger. Due to the small color difference, the quadcolor camera using an optimized short-pass filter may be suitable as an imaging colorimeter. It was found that an empirical design rule to keep the color difference small is to reduce the error in fitting the color-matching functions using the camera spectral sensitivity functions.

The spectrum of light captured by a camera can be reconstructed using the interpolation method. The reconstructed spectrum is a linear combination of the reference spectra, where the weighting coefficients are calculated from the signals of the pixel and the reference samples by interpolation. This method is known as the look-up table (LUT) method. It is irradiance-dependent due to the dependence of the reconstructed spectrum shape on the sample irradiance. Since the irradiance can vary in field applications, an irradiance-independent LUT (II-LUT) method is required to recover spectral reflectance. This paper proposes an II-LUT method to interpolate the spectrum in the normalized signal space. Munsell color chips irradiated with D65 were used as samples. Example cameras are a tricolor camera and a quadcolor camera. Results show that the proposed method can achieve the irradiance independent spectrum reconstruction and computation time saving at the expense of the recovered spectral reflectance error. Considering that the irradiance variation will introduce additional errors, the actual mean error using the II-LUT method might be smaller than that of the ID-LUT method. It is also shown that the proposed method outperformed the weighted principal component analysis method in both accuracy and computation speed.

Gait stability has been measured by using many entropy-based methods. However, the relation between the entropy values and gait stability is worth further investigation. A research reported that average entropy (AE), a measure of disorder, could measure the static standing postural stability better than multiscale entropy and entropy of entropy (EoE), two measures of complexity. This study tested the validity of AE in gait stability measurement from the viewpoint of the disorder. For comparison, another five disorders, the EoE, and two traditional metrics methods were, respectively, used to measure the degrees of disorder and complexity of 10 step interval (SPI) and 79 stride interval (SI) time series, individually. As a result, every one of the 10 participants exhibited a relatively high AE value of the SPI when walking with eyes closed and a relatively low AE value when walking with eyes open. Most of the AE values of the SI of the 53 diseased subjects were greater than those of the 26 healthy subjects. A maximal overall accuracy of AE in differentiating the healthy from the diseased was 91.1%. Similar features also exists on those 5 disorder measurements but do not exist on the EoE values. Nevertheless, the EoE versus AE plot of the SI also exhibits an inverted U relation, consistent with the hypothesis for physiologic signals.

The complexity and the disorder of a 1/fα noise time series are quantified by entropy of entropy (EoE) and average entropy (AE), respectively. The resulting EoE vs. AE plot of a series of 1/fα noises of various values of α exhibits a distinct inverted U curve. For the 1/fα noises, we have shown that α decreases monotonically as AE increases, which indicates that α is also a measure of disorder. Furthermore, a 1/fα noise and a cardiac interbeat (RR) interval series are considered equivalent as they have the same AE. Accordingly, we have found that the 1/fα noises for α around 1.5 are equivalent to the RR interval series of healthy subjects. The pink noise at α = 1 is equivalent to atrial fibrillation (AF) RR interval series while the white noise at α = 0 is more disordered than AF RR interval series. These results, based on AE, are different from the previous ones based on spectral analysis. The testing macro-average F-score is 0.93 when classifying the RR interval series of three groups using AE-based α, while it is 0.73 when using spectral-analysis-based α.

BACKGROUND: Total motile sperm count (TMSC) and curvilinear velocity (VCL) are two important parameters in preliminary semen analysis for male infertility. Traditionally, both parameters are evaluated manually by embryologists or automatically using an expensive computer-assisted sperm analysis (CASA) instrument. The latter applies a point-tracking method using an image processing technique to detect, recognize and classify each of the target objects, individually, which is complicated. However, as semen is dense, manual counting is exhausting while CASA suffers from severe overlapping and heavy computation. METHODS: We proposed a simple frame-differencing method that tracks motile sperms collectively and treats their overlapping with a statistical occupation probability without heavy computation. The proposed method leads to an overall image of all of the differential footprint trajectories (DFTs) of all motile sperms and thus the overall area of the DFTs in a real-time manner. Accordingly, a theoretical DFT model was also developed to formulate the overall DFT area of a group of moving beads as a function of time as well as the total number and average speed of the beads. Then, using the least square fitting method, we obtained the optimal values of the TMSC and the average VCL that yielded the best fit for the theoretical DFT area to the measured DFT area. RESULTS: The proposed method was used to evaluate the TMSC and the VCL of 20 semen samples. The maximum TMSC evaluated using the method is more than 980 sperms per video frame. The Pearson correlation coefficient (PCC) between the two series of TMSC obtained using the method and the CASA instrument is 0.946. The PCC between the two series of VCL obtained using the method and CASA is 0.771. As a consequence, the proposed method is as accurate as the CASA method in TMSC and VCL evaluations. CONCLUSION: In comparison with the individual point-tracking techniques, the collective DFT tracking method is relatively simple in computation without complicated image processing. Therefore, incorporating the proposed method into a cell phone equipped with a microscopic lens can facilitate the design of a simple sperm analyzer for clinical or household use without advance dilution.

An internally focusing design is developed for a compact endoscopic lens to have optical performance close to the diffraction limit. When changing the working distance from infinity to a predetermined short distance, the overall system length and the field of view remain constant. It is numerically demonstrated that the diffraction-limit-like performance is maintained over a wide range of working distances, showing a novel capability of adjustable focuses. The design concept is numerically proven to be feasible and can enable very high-resolution examination for doctors checking the positions of biological entities through the immediate capture of clear images at different working distances. The corresponding lens configuration, PNN, is shown to be useful in achieving the high-resolution performance once the number of lens elements is four, where P and N are used to denote positive and negative optical power, respectively, for the lens groups. A corresponding tolerance analysis is also presented.

When UV-curable material is used with a spherical glass lens, it is classified as a hybrid lens and can provide a new degree of freedom in designing a lens system. This method is used to explore the optical limitation of an ultrawide-angle imaging system of a folded structure, which is utilized to achieve a compact volume. The f-number and field of view of the system are 2.0 and 190°, respectively. Diffraction-limited performance is achieved within a field of view of 80° with Strehl ratios greater than approximately 80%. Even at the maximum field of view of 190°, the Strehl ratio is nearly 75%. The lens configuration is NNPNN, where N and P denote negative and positive optical power, respectively; this configuration uses only two aspherical hybrid lens surfaces. Comparisons of different optimizations are also provided.

A sense of meaning and purpose in life-known in Japan as one's ikigai-can lead to better health outcomes, an improved sense of well-being, and longer life as people age. The design of socially assistive robots, however, has so far focused largely on the more hedonic aims of supporting positive affect and happiness through interactions with robots. To explore how social robots might be able to support people's ikigai, we performed (1) in-depth interviews with 12 'ikigai experts' who formally support and/or study older adults (OAs)' ikigai and (2) 5 co-design workshop sessions with 10 such experts. Our interview findings show that expert practitioners define ikigai in a holistic way in their everyday experience and practice, incorporating physical, social, and mental activities that relate not only to the individual and their behaviors, but also to their relationships with other people and to their connection with the broader community (3 levels of ikigai). Our co-design workshops showed that ikigai experts were overall positive towards the use of social robots to support OAs' ikigai, particularly in the roles of an information-provider and social enabler that connects OAs to other people and activities in their communities. They also point out areas of potential risk, including the need to maintain OAs' independence, relationships with others, and privacy, which should be considered in design. This research is the first to explore the co-design of social robots that can support people's sense of ikigai-meaning and purpose-as they age.

BACKGROUND: Ikigai (meaning or purpose in life) is a concept understood by most older adults in Japan. The term has also garnered international attention, with recent academic attempts to map it to concepts in the Western well-being literature. In addition, efforts to use social and home robots to increase well-being have grown; however, they have mostly focused on hedonic well-being (eg, increasing happiness and decreasing loneliness) rather than eudaimonic well-being (eg, fostering meaning or purpose in life). OBJECTIVE: First, we explored how Japanese older adults experience ikigai and relate these to concepts in the Western well-being literature. Second, we investigated how a home robot meant to promote ikigai is perceived by older adults. METHODS: We used a mixed methods research design-including 20 interviews with older adults, a survey of 50 older adults, and 10 interviews with family caregivers. For interviews, we asked questions about older adults' sources of ikigai, happiness, and social support, along with their perception of the robot (QT). For surveys, a number of well-being scales were used, including 2 ikigai scales-ikigai-9 and K-1-and 6 Patient-Reported Outcomes Measurement Information System scales, measuring meaning and purpose, positive affect, satisfaction with participation in social roles, satisfaction with participation in discretionary social activities, companionship, and emotional support. Questions related to the perception and desired adoption of the robot and older adults' health status were also included. RESULTS: Our results suggest that health is older adults' most common source of ikigai. Additionally, although self-rated health correlated moderately with ikigai and other well-being measures, reported physical limitation did not. As opposed to social roles (work and family), we found that ikigai is more strongly related to satisfaction with discretionary social activities (leisure, hobbies, and friends) for older adults. Moreover, we found that older adults' sources of ikigai included the eudaimonic aspects of vitality, positive relations with others, contribution, accomplishment, purpose, and personal growth, with the first 3 being most common, and the hedonic aspects of positive affect, life satisfaction, and lack of negative affect, with the first 2 being most common. However, the concept of ikigai was most related to eudaimonic well-being, specifically meaning in life, along the dimension of significance. Finally, we found that Japanese older adults have high expectations of a home robot for well-being, mentioning that it should support them in a multitude of ways before they would likely adopt it. However, we report that those with the highest levels of meaning, and satisfaction with their leisure life and friendships, may be most likely to adopt it. CONCLUSIONS: We outline several ways to improve the robot to increase its acceptance, such as improving its voice, adding functional features, and designing it to support multiple aspects of well-being.

We develop and test a thermally activated state transition technique for ultraweak force measurement. As a force sensor, the technique was demonstrated on a classical Brownian bead immersed in water and restrained by a bistable optical trap. A femto-Newton-level flow force imposed on this sensor was measured by monitoring changes in the transition rates of the bead hopping between two energy states. The treatment of thermal disturbances as a requirement instead of a limiting factor is the major feature of the technique, and provides a new strategy by which to measure other ultraweak forces beyond the thermal noise limit.

The Klebsiella pneumoniae type 3 fimbriae are mainly composed of MrkA pilins that assemble into a helixlike filament. This study determined the biomechanical properties of the fimbriae and analyzed 11 site-directed MrkA mutants to identify domains that are critical for the properties. Escherichia coli strains expressing type 3 fimbriae with an Ala substitution at either F34, V45, C87, G189, T196, or Y197 resulted in a significant reduction in biofilm formation. The E. coli strain expressing MrkAG189A remained capable of producing a normal number of fimbriae. Although F34A, V45A, T196A, and Y197A substitutions expressed on E. coli strains produced sparse quantities of fimbriae, no fimbriae were observed on the cells expressing MrkAC87A. Further investigations of the mechanical properties of the MrkAG189A fimbriae with optical tweezers revealed that, unlike the wild-type fimbriae, the uncoiling force for MrkAG189A fimbriae was not constant. The MrkAG189A fimbriae also exhibited a lower enthalpy in the differential scanning calorimetry analysis. Together, these findings indicate that the mutant fimbriae are less stable than the wild-type. This study has demonstrated that the C-terminal ß strands of MrkA are required for the assembly and structural stability of fimbriae.

In optical tweezers applications, tracking a trapped particle is essential for force measurement. One of the most popular techniques for single-particle tracking is achieved by analyzing the forward and backward light pattern, scattered by the target particle trapped by a trap laser beam, of an additional probe-laser beam with different wavelength whose focus is slightly apart from the trapping center. However, the optimized focal offset has never been discussed. In this paper, we investigate the tracking range and sensitivity as a function of the focal offset between the trapping and the probe-laser beams. As a result, the optimized focal offsets are a 3.3-fold radius ahead and a 2.0-fold radius behind the trapping laser focus in the forward tracking and the backward tracking, respectively. The experimental result agrees well with a theoretical prediction using the Mie scattering theory.

BACKGROUND/PURPOSE: Acinetobacter baumannii is an important nosocomial pathogen. To better understand the role of CsuA/BABCDE pilus of A. baumannii in virulence, bacterial biofilm formation, adherence and carbohydrate-mediated inhibition were conducted. METHODS: CsuA/BABCDE pilus-producing (abbreviated Csu pilus) operon of A. baumannii ATCC17978 was cloned for analysis of biofilm formation on an abiotic plastic plate, bacterial adherence to respiratory epithelial human A549 cells and carbohydrate-mediated inhibition. The carbohydrates used for inhibition of biofilm formation and adherence to A549 cells included monosaccharides, pyranosides, and mannose-polymers. RESULTS: The Csu pilus of A. baumannii ATCC17978 was cloned and expressed into a non-pilus-producing Escherichia coli JM109, and was knocked out as well. The recombinant Csu (rCsu) pilus on E. coli JM109/rCsu pilus-producing clone observed by both electro-microscopy and atomic force microscopy showed abundant, while Csu-knockout A. baumannii ATCC17978 mutant appeared less or no pilus production. The E. coli JM109/rCsu pilus-producing clone significantly increased biofilm formation and adherence to A549 cells; however, the Csu-knockout mutant dramatically lost biofilm-making ability but, in contrast, increased adherence. Moreover, both of biofilm formation and adherence could be significantly inhibited by d-mannose and methyl-α-d-mannopyranoside in Csu pilus-producing E. coli JM109, whereas in A. baumannii ATCC17978, high concentration of carbohydrates was required for the inhibition, suggesting that Csu pilus is sensitive to d-mannose. CONCLUSION: This is the first study confirming that Csu pilus of A. baumannii belongs to mannose-sensitive type 1 pilus family and contributes to biofilm formation and bacterial adherence to human epithelial cells.

Previous research in human-robot interaction has explored using robots to increase objective and hedonic aspects of well-being and quality of life, but there is no literature on how robots might be used to support eudaimonic aspects of well-being (such as meaning in life). A sense of meaning has been shown to positively affect health and longevity. We frame our study around the Japanese concept of ikigai, which is widely used with Japanese older adults to enhance their everyday lives, and is closely related to the concept of eudaimonic well-being (EWB) known in Western countries. Using a mixed-methods and exploratory approach, including interviews with 17 older adults and the collection of 100 survey responses, we explored how older adults in the US experience a sense of meaning, and if and how a social robot could be used to help foster this sense. We find that meaning for older adults is often obtained by helping others, through family connections, and/or through activities of daily life, and that sources of meaning often differ based on the older adults' living situation. Assessing how meaning compares to happiness and social connection, we highlight general similarities and differences, and also find that living situation influences older adults' sources of happiness, desire for social connection, and barriers to well-being, in addition to companionship and happiness having a weaker correlation with meaning for those who live alone than for those who live with others. Additionally, we evaluated initial perceptions of a social robot (QT) meant to enhance ikigai and overall well-being, finding mostly positive perceptions, though those who live alone also reported being less willing to adopt a social robot into their homes. Using both data collected on older adults' meaning and the potential use of QT to support meaning, we make several design recommendations with regards to using robots to enhance ikigai, such as by prompting daily reflecting, enhancing family bonds, and suggesting new experiences and volunteer opportunities.

Exercise stress testing (EST) has limited power in diagnosing obstructive coronary artery disease (CAD). The heart rate variability (HRV) analysis might increase the sensitivity of CAD detection. This study aimed to evaluate the correlation between short-term HRV and myocardial ischemia during EST, including the acceleration, maximum, and recovery stages of heart rate (HR). The HRV during EST from 19 healthy (RHC) subjects and 35 patients with CAD (25 patients with insignificant CAD (iCAD), and 10 patients with significant CAD (sCAD)) were compared. As a result, all HRV indices decreased at the maximum stage and no significant differences between iCAD and sCAD were found. The low-frequency power of heart rate signal (LF) of the RHC group recovered relatively quickly from the third to the sixth minutes after maximum HR, compared with that of the sCAD group. The relative changes of most HRV indices between maximum HR and recovery stage were lower in the sCAD group than in the RHC group, especially in LF, the standard deviation of all normal to normal intervals (SDNN), and the standard deviation in the long axis direction of the Poincaré plot analysis (SD2) indices (p < 0.05). The recovery slope of LF was significantly smaller in the sCAD group than in the RHC group (p = 0.02). The result suggests that monitoring short-term HRV during EST provides helpful insight into the cardiovascular autonomic imbalance in patients with significant CAD. The relative change of autonomic tone, especially the delayed sympathetic recovery, could be an additional marker for diagnosing myocardial ischemia.

This study investigated the structural and mechanical properties of Klebsiella pneumoniae type 3 fimbriae, which constitute a known virulence factor for the bacterium. Transmission electron microscopy and optical tweezers were used to understand the ability of the bacterium to survive flushes. An individual K. pneumoniae type 3 fimbria exhibited a helix-like structure with a pitch of 4.1 nm and a three-phase force-extension curve. The fimbria was first nonlinearly stretched with increasing force. Then, it started to uncoil and extended several micrometers at a fixed force of 66 ± 4 pN (n = 22). Finally, the extension of the fimbria shifted to the third phase, with a characteristic force of 102 ± 9 pN (n = 14) at the inflection point. Compared with the P fimbriae and type 1 fimbriae of uropathogenic Escherichia coli, K. pneumoniae type 3 fimbriae have a larger pitch in the helix-like structure and stronger uncoiling and characteristic forces.

This study proposes a method for creating a light field with controlled distribution of transverse momentum (TM) by displaying a hologram only in an azimuth region that centers at θ(0) and has a range of Δθ of a spatial light modulator in holographic optical tweezers. This study utilized ray optics to analyze the TM of the resultant field, revealing that the direction of the TM is determined by the center angle of the azimuth region and that the magnitude of the TM is proportional to sin(Δθ/2), without regarding the intensity. The relationship was verified experimentally. In addition, this study demonstrated moving particles along a designed path and depleting particles by the fields.

RESUMO

We develop light-driven optoelectronic tweezers based on the organic photoconductive material titanium oxide phthalocyanine. These tweezers function based on negative dielectrophoresis (nDEP). The dynamic manipulation of a single microparticle and cell patterning are demonstrated by using this light-driven optoelectronic DEP chip. The adaptive light patterns that drive the optoelectronic DEP onchip are designed by using Flash software to approach appropriate dynamic manipulation. This is also the first reported demonstration, to the best of our knowledge, for successfully patterning such delicate cells from human hepatocellular liver carcinoma cell line HepG2 by using any optoelectronic tweezers.
