RESUMO

Research on the markers of autoimmune response in multiple sclerosis (MS) is still of great importance. The aim of our study was the evaluation of plasma 20S constitutive proteasome, 20S immunoproteasome, and cathepsin S concentrations as potential biomarkers of a relapsing-remitting type of MS (RRMS). Surface plasmon resonance imaging (SPRI) biosensors were used for the evaluation of protein concentrations. Plasma 20S constitutive proteasome, 20S immunoproteasome, and cathepsin S concentrations were significantly higher in RRMS patients compared to the control group. All three parameters were characterized by excellent usefulness in differentiating MS patients from healthy individuals (AUC equal to or close to 1.000). The plasma concentration of analyzed parameters was not correlated with severity of disability in the course of RRMS (EDSS value), the number of years from the first MS symptoms, the number of years from MS diagnosis, or the number of relapses within the 24-month observational period. Our study has shown that plasma concentrations of 20S constitutive proteasome, 20S immunoproteasome, and cathepsin S have promising potential in differentiating RRMS patients from healthy individuals. All of the analyzed parameters were found to be independent of the time of MS relapse and the severity of neurological symptoms. Hence, their potential as highly sensitive and independent circulating markers of RRMS suggests a stronger association with immunological activity (inflammatory processes) than with the severity of the disease.

The aim of the study was to assess the zinc (Zn), copper (Cu) and selenium (Se) content in freshwater fish from Poland. Selected species of raw, smoked and pickled fish were evaluated by atomic absorption spectrometry (AAS). The concentration of Zn, Cu and Se in the examined fish ranged from 1.5 to 49.9 mg/kg, 0.01 to 2.8 mg/kg and 30.9 to 728.2 µg/kg, respectively. One serving of every fish product covered the recommended dietary allowance (RDA) of Zn by 5.38-65.0%, of Cu by 0.42-11.4% and of Se by 12.3-198.6%. A cluster analysis allowed us to distinguish European eel (raw and smoked) based on the Zn content. Additionally, based on the Cu and Se content, pickled common whitefish was differentiated from other species and types of fish products. The discriminant analysis model of smoked fish enabled their classification with a 70% accuracy. Regarding Zn, all forms of the European eel as well as raw and smoked vendace can be considered a source of this element. None of the tested fish can be considered a source of Cu. All products are safe for human consumption with regard to the Zn and Cu content. Almost each form of every species of fish can be considered a source of Se. However, the Se content should be monitored in smoked and pickled common whitefish, pickled bream and pickled vendace.

INTRODUCTION: Cigarette smoking is one of the most common habits of the modern world. According to a NATPOL PLU study, every third adult Pole is dependent on nicotine. Tobacco smoke contains about 5,000 components, of which over 1,000 are very toxic chemical substances (3,4-benzopyrene, heavy metals, free radicals, hydrogen cyanide, nitrogen oxides and N-nitrosamines). Exposure to tobacco smoke is an example of a complex, with a significant number of interactions. THE AIM OF THE STUDY: To assess the concentration of copper in the urine of smokers. Based on the results, an attempt was made to determine whether smoking can affect the level of copper in the body. MATERIAL AND METHODS: The study involved 170 healthy volunteers, 99 smokers and 71 non-smokers (control group). The age of patients in both groups were in the range of 20-60 years. The mean age for men and women was 41 years. The average length of cigarette smoking was 18 years for women and 21 years for men, and the number of cigarettes smoked 1-40 / 24. The urine concentrations of Cu were determined by atomic absorption spectrometry (AAS) and serum creatinine kinetic method using a set of BIOLAB. Cu concentration in urine was expressed in mg / g creatinine. RESULTS: Smokers were found to have reduced levels of copper in the urine, depending on sex, age and brand of cigarettes. In male smokers, copper concentration in the urine was dependent on age and time of smoking, whereas among women this relationship was not observed. CONCLUSIONS: Cigarette smoking significantly influences the level of copper in the urine. Both female and male smokers showed reduced levels of copper in the urine, which may indicate its increased accumulation in the body. Excessive accumulation of copper is very dangerous since it may exhibit toxic effects towards many organs and systems.

INTRODUCTION: Cigarette smoking is one of the most common habits of the modern world. The World Health Organization (WHO) showed that about 30% of the world's population over the age of 15 smoked cigarettes. That gives the number of 1.1 billion smokers. It is known that the habit of smoking is a serious risk to human health and life. Cigarette smoke induces the formation of reactive oxygen species (ROS). ROS can cause direct damage of DNA chain and leads to the mutagenic and carcinogenic effect. THE AIM OF THE STUDY: To evaluate the concentration of 8-hydroxy-2'-deoxyguanosine (8-OHdG) in the urine of smokers. Based on the obtained results an attempt to determine whether the concentration of 8-OH-dG in the urine can be an indicator of genotoxicity of tobacco smoke. MATERIAL AND METHODS: The study was conducted in 83 healthy volunteers, 63 smokers and 20 non-smokers (control group). The age of patients in both groups were in the range 20-60 years. Average age of women and men in the control group and the test group was 34 years. Duration of smoking was 1-30 years, the number of cigarettes smoked from 1 to over 20 per day. In the urine the concentration of 8-OH-dG was determined using Bioxytech® 8-OH-dG-EIA™Kit (manufactured by OXIS International, Inc., Portland, USA) and the concentration of creatinine using in vitro diagnostic kit BIOLABO. The concentration of 8-OHdG in urine was expressed in ng/g creatinine. RESULTS: More than 3-fold increase in the concentration of 8-OHdG in the urine of smokers was found. The increase in the concentration of 8-OHdG in the urine depended on age, the amount of smoked cigarettes per 24 hours, duration of smoking and cigarette brands. CONCLUSIONS: The increase in the concentration of 8-OHdG in the urine of smokers, both women and men indicates DNA damage and the genotoxicity action of cigarette smoke. According to our previously conducted studies and examinations of other authors, it can be suggested that the concentration of 8-OHdG in urine may be considered as the sensitive and specific marker of oxidative DNA damage.

BACKGROUND: Dietary habits and adequate dietary intake of antioxidants in the diet may be one of the most important environmental factors for the prevention of Multiple Sclerosis (MS). OBJECTIVES: The aim of this study was to estimate selenium (Se) concentration, glutathione peroxidase (GSH-Px) activity and total antioxidant status (TAS) in the serum of patients with MS and the influence of dietary habits on the status. METHODS: 101 patients with relapsing-remitting MS (aged 18-58 years), as well as control group of 63 healthy people (aged 19-65 years) were studied. Food-frequency questionnaires were implemented to collect the dietary data. Se concentration in the serum samples was determined by atomic absorption spectrometry. GSH-Px activity and TAS in examined serum was measured using the ready-made sets of tests by Randox Laboratories Ltd., UK. RESULTS: Serum Se concentration and GSH-Px activity in the serum of patients with MS (55.2±16.2 µg/L, 6676.1±2386.4 U/L; respectively) were significantly decreased (p<0.01, p<0.05; respectively) compared with control group (79.2±20.6 µg/L, 8029.9±2650.1 U/L; respectively). A significant correlation (r=0.39, p<0.01) was observed between Se concentration and GSH-Px activity in the serum of examined patients. TAS value in the serum of patients with MS (1.03±0.37 mmol/L) was also significantly lower (p<0.01) than in healthy volunteers (1.48±0.41 mmol/L). Frequent consumption of poultry, bakery products, pulses and fish seemed to increase serum Se concentration in the group of patients; whereas frequent consumption of butter, wholegrain bread, sweet beverages and sugar was found to accompany with lower values of Se in the serum. We have observed significant decrease TAS (p<0.05, p<0.01; respectively) in the serum of smokers and those patients who received immunomodulatory drugs (0.95±0.39 mmol/L, 0.92±0.34 mmol/L; respectively) compared with no-smoking patients and not taking immunomodulators (1.14±0.33 mmol/L, 1.31±0.31 mmol/L; respectively). CONCLUSIONS: Serum Se concentration, GSH-Px activity and TAS value were significantly lower in patients with relapsing-remitting MS compared with healthy volunteers. Dietary habits have a significant influence on Se status. Smoking cigarettes and intake of immunomodulatory drugs therapy have a negative impact on TAS of examined patients.

Research on the markers of immunoregulatory response in multiple sclerosis (MS) is still of great importance. The aim of our study was the evaluation of leptin, fibronectin, and UCHL1 concentrations as potential biomarkers of a relapsing-remitting type of MS (RRMS). Surface Plasmon Resonance Imaging (SPRI) biosensors were used for the evaluation of proteins concentrations in 100 RRMS patients and 46 healthy volunteers. Plasma leptin, fibronectin, and UCHL1 concentrations were significantly higher in RRMS patients compared to the control group (p < 0.001, respectively). UCHL1 concentration evaluation revealed the highest diagnostic sensitivity (100%) and negative predictive value (100%) in differentiating MS patients from healthy individuals. There was no significant difference in the UCHL1 concentrations depending on the patient's sex, the presence of relapse within the last 24 months, and the EDSS value (p > 0.05, respectively). In RRMS patients UCHL1 concentration positively correlated with fibronectin levels (r = 0.3928; p < 0.001). In the current cohort of patients plasma UCHL1 concentration was independent of the time of MS relapse and the severity of neurological symptoms. Thus current study may indicate that plasma UCHL1, besides leptin and fibronectin, also could be a promising high-sensitive potential biomarker of relapsing-remitting type of MS. However, these results should be validated with a larger group of patients, taking into account neuroimaging and cerebrospinal fluid analysis data, and by comparing them to patients with other neurological diseases as a control group.

Phenolic acids are an important component of honey. Literature data indicate their pro-health properties and diversified content in different varieties. Therefore, the aim of our study was to evaluate the content of phenolic acids in bee honey. The material for the research was 49 samples of honey obtained from beekeepers from Poland. Selected phenolic acids were determined by HPLC with PDA detection. Additionally, total phenolic content (TPC), color intensity, color on the Pfund scale, water content, electrical conductivity, and FRAP were assessed. A higher trans-ferulic acid content is accompanied by a stronger free radical scavenging ability. It was shown that buckwheat honeys are characterized by a high TPC value (196.59 mg GAE/100 g), color intensity (2109.2 mAU), color on the Pfund scale (159.8 mm Pfund), and high activity in the FRAP assay (0.403 equivalent of µmol Fe2+/mL). The median obtained in the DPPH test for this honey variety was 41.1%. Moreover, the highest median of 4-hydroxybenzoic acid (3.129 mg/100 g) in buckwheat honey was shown. Buckwheat honeys have promising antioxidant properties and should be included in diets low in antioxidants.

The choices of food products and their nutritional value have a significant impact on nutritional status and body composition parameters. Popular and low-cost indicators of nutritional status, such as BMI, may not reflect the actual condition of the body. The aim of the study was to assess the consumption of energy as well as selected nutrients, such as protein, fats, carbohydrates, unsaturated fatty acids, lactose, starch, and dietary fiber, and to evaluate body mass index (BMI) and the parameters of the body composition among young women. Next, correlations were made between these nutrients and the parameters, such as percent body fat (PBF), visceral fat area (VFA), and fitness score (FS). The study group included 478 young women. To assess their consumption, the participants completed 72 h dietary interviews. In addition, the participants had a body composition analysis performed by bioelectric impedance technology by the InBody 720 Body Composition Analyzer (InBody, South Korea). The average energy value of the diets was about 1480 kcal. Most diets contained adequate portions of protein, fat, and carbohydrates. The diets were deficient in terms of unsaturated fatty acids and fiber. We showed that up to 37% of women with normal BMI had excess body fat. Moreover, the % energy from plant protein consumption negatively correlated with PBF and BMI (r = -0.10, p = 0.029, r = -0.10, p = 0.037, respectively) and dietary fiber with PBF, BMI, VFA, and FS (r = -0.17, p = 0.0003, r = -0.12, p = 0.01, r = -0.18, p = 0.0001, r = 0.09, p = 0.041, respectively). Education on the proper consumption of basic nutrients, including plant-based protein and dietary fiber, seems to be essential in preventing overweight and obesity.

The aim of the study was to assess arsenic (As), cadmium (Cd), lead (Pb), and mercury (Hg) contamination of freshwater fish from Poland. Selected species of raw, smoked, and pickled freshwater fish (n = 212) were evaluated by atomic absorption spectrometry (AAS) and inductively coupled plasma - mass spectrometry (ICP-MS). Assessment of health risk associated with intake of investigated elements present in fish was performed. Contamination of fish with As, Cd, Hg and Pb ranged as follows: As 23.3-59,290.1 µg/kg, Cd 0.02-97.0 µg/kg, Hg 9.04-606.3 µg/kg, and Pb 0.04-171.4 µg/kg. Consumption of selected species of freshwater fish, especially smoked fish products, may pose a non-carcinogenic and also carcinogenic health risk.

The aim of this study was to estimate the consumption of selected dietary components with antioxidant properties, undertake body composition analysis, assess skin hydration and lubrication, and establish the relationships between the above parameters. The study was carried out on 172 young women. The consumption of ingredients (vitamins A, C, D and E, and Cu, Mn, Zn) was assessed using the Diet 6.0 program, body composition was assessed using electrical bioimpedance and skin hydration and lubrication were assessed using the corneometric and sebumetric methods, respectively. About one-third of students showed insufficient consumption of vitamin C, vitamin E and zinc, while about 99% showed insufficient vitamin D levels. The highest degree of hydration was observed in the areas of the eyelids, neckline and chin. The greatest amount of sebum was found in the area of the nose and forehead. Low positive correlations between hydration or lubrication and Cu, vitamin A and vitamin E were observed. In conclusion, to properly moisturize and lubricate the skin, young women should eat products that are rich in ingredients with antioxidant properties, in particular fat-soluble vitamins A and E, but also copper.

The aim of the present study is to investigate, based on the rat model of moderate and relatively high human exposure to cadmium (Cd), whether zinc (Zn) supplementation may prevent Cd-induced disorders in bone metabolism. For this purpose, male Wistar rats received Cd (5 and 50mg/l) or/and Zn (30 and 60mg/l) in drinking water for 6 and 12 months. Bone densitometry and biochemical markers of bone turnover were used to assess the effects of Cd or/and Zn. Bone mineral content (BMC) and density (BMD) were measured in the femur. Serum osteocalcin (OC) and alkaline phosphatase in trabecular (bT-ALP) and cortical (bC-ALP) bone were determined as bone formation markers, and carboxy-terminal cross-linking telopeptides of type I collagen (CTX) in serum were measured as bone resorption marker. Serum concentration of calcium (Ca) and its renal handling, as well as Zn and Cd concentrations in the serum/blood, urine and femur were evaluated as well. The exposure to 5 and 50mg Cd/l (0.340+/-0.026 and 2.498+/-0.093mg Cd/kg body wt/24h, respectively), in a dose and duration dependent manner, affected bone turnover (inhibited bone formation and stimulated its resorption) and disturbed bone mineralization (decreased BMC, BMD and Zn concentration). Zn supply at the concentration of 30 and 60mg/l (1.904+/-0.123 and 3.699+/-0.213mg/kg body wt/24h, respectively) during Cd exposure influenced the Cd-induced disorders in bone metabolism. Zn administration to the Cd-exposed rats enhanced the bone ALP activity and prevented Cd-induced bone resorption, but had no statistically significant effect on BMC and BMD; however, mean values of the densitometric parameters in the rats receiving both Cd and Zn were higher than in those treated with Cd alone. Moreover, Zn supplementation at both levels of Cd exposure was found to prevent Cd accumulation in the femur and the Cd-induced decrease in bone Zn concentration. The results of the present study allow the conclusion that Zn supplementation during Cd exposure may partly protect from disorders in bone metabolism. The influence of Zn may be accompanied by its ability to prevent Cd-induced Zn deficiency and to decrease Cd accumulation in bone tissue. The findings seem to indicate that enhanced dietary intake of Zn in subjects chronically exposed to moderate and relatively high Cd levels may have a protective influence on the skeleton.

OBJECTIVE: Dietary habits and adequate intake of antioxidants in the diet-for example, copper (Cu) and zinc (Zn)-may be an environmental factor in the occurrence of multiple sclerosis (MS). The aim of this study was to estimate the influence of dietary habits on the concentration of Cu, Zn in the serum, and the effect of Cu-to-Zn ratio on the ability status of patients with relapsing-remitting MS. METHODS: This was an observational case-control study that included 101 individuals with MS and 68 healthy individuals (controls). Food frequency questionnaires were used to collect dietary data. Serum concentrations of Cu and Zn were determined by the electrothermal and flame atomic absorption spectrometry method, respectively. The ratio of Cu to Zn was calculated and compared with the Expanded Disability Status Scale of patients. RESULTS: The concentration of Zn was significantly lower in the serum of individuals with MS (0.776 ± 0.195 mg/L) than in the control group (0.992 ± 0.315 mg/L). The ratio of Cu to Zn was higher in the examined patients (1.347 ± 0.806) than in the healthy volunteers (1.012 ± 0.458). Lower ability status (P < 0.05) was revealed in patients with an abnormal ratio of Cu to Zn, particularly, in cerebellar function, pyramidal tracts, and emotional conditions. Selected dietary habits have a significant influence on Cu and Zn concentration in the serum of patients with MS. CONCLUSIONS: Lower serum concentrations of Zn and higher ratio of Cu to Zn in patients with MS can suggest a relationship between MS and oxidative stress. Products that are a source of Zn should be included in the diet, which can improve the clinical condition of people with MS.

Antioxidants can play a significant role in chronic inflammatory process. The aim of this study was to evaluate the content of selenium (Se), zinc (Zn), copper (Cu), and total antioxidant status (TAS) of patients with chronic tonsillitis (CT). The study group consisted of 84 patients with CT from 18 to 62 years old and the control group of 67 healthy people aged 19-65 years. Se, Zn, and Cu concentration in serum samples were determined by atomic absorption spectrometry. Serum TAS was measured spectrophotometrically, using the test by Randox Laboratories-Us Ltd. The mean content of Se and Zn in the serum of patients with CT (61.122 ± 12.73 µg/L, 0.887 ± 0.26 mg/L, respectively) was lower compared to the control group (77.969 ± 12.73 µg/L, 0.993 ± 0.32 mg/L, respectively). The mean serum concentration of Cu in patients with CT (1.219 ± 0.35 mg/L) was higher compared to its serum concentration in healthy people (1.033 ± 0.37 mg/L). Serum TAS of patients with CT (1.171 ± 0.33 mmol/L) was lower in comparison with healthy volunteers (1.333 ± 0.42 mmol/L). The serum concentration of Se, Zn, and TAS in patients with CT was lower, whereas the concentration of Cu was higher compared to healthy volunteers. Smoking has an influence on reducing the concentration of Se and TAS of patients with CT.

INTRODUCTION: Although much is known about cytokines and adhesion molecules during an active course of multiple sclerosis (MS), there is limited information about their serum levels during remission. OBJECTIVE: This study aimed to (1) compare peripheral levels of tumor necrosis factor-α (TNF-α), soluble interleukin-2 receptor α (sIL-2Rα), soluble intercellular adhesion molecule-1 (sICAM-1), and soluble E-selectin (sE-selectin) in MS patients during clinical remission with those of healthy controls and (2) explore possible relationships between the levels of these cytokines and adhesion molecules and neurological impairment. METHODS: Initially, 92 patients with relapsing-remitting multiple sclerosis (RRMS) who were in clinical remission and 30 healthy controls were recruited for this study. The severity of neurological impairment was assessed with the Expanded Disability Status Scale (EDSS). Serum concentrations of TNF-α, sIL-2Rα, sICAM-1, and sE-selectin were determined using the sandwich enzyme-linked immunosorbent (ELISA) technique and compared between patients and controls. In a subset of RRMS patients (n = 67), the levels of these cytokines and adhesion molecules were compared between subgroups of patients based on scores on the EDSS subscales, which measure disability level for specific neurological functions. RESULTS: The MS patients' TNF-α, sICAM-1, and sE-selectin levels were markedly lower than those of the controls, while their sIL-2Rα level was higher. The serum sICAM-1 concentration was positively associated with EDSS total score (ρ = .291, p = .017) as well as with the EDSS pyramidal (ρ = .267, p = .029) and cerebellar subscores (ρ = .303, p = .013). In the patients with cerebellar deficits and severe brain stem dysfunction, sICAM-1 levels were upregulated. CONCLUSION: Although a decreased sICAM-1 concentration was observed in RRMS patients in remission as compared to healthy controls, sICAM-1 seemed to reflect neurological impairment and clinical disability. These data suggest that increasing serum sICAM-1 levels may be associated with progression of cerebellar or brain stem perturbations. However, further studies are required to confirm these findings in a larger population of RRMS patients.
