RESUMO

Campylobacter jejuni subsp. jejuni is a leading bacterial cause of human gastroenteritis. C. jejuni strain P4549 was isolated from an asymptomatic rhesus monkey, Macaca mulatta. We report the genome sequences have a circular chromosome of 1,729,940 bp and two plasmids of 50,482 bp and 7,259 bp, respectively.

RESUMO

Sphingobacterium multivorum is a ubiquitous environmental microbe in nature, particularly abundant in soil and water sources. Recently, S. multivorum has been considered to be a potential opportunistic pathogen in humans. We report the 5,9540,064-bp genome sequence of S. multivorum strain FW102, isolated from drinking fountain water and predicted to have multiple copies of ß-galactosidase genes.

RESUMO

Delftia tsuruhatensis is a Gram-negative rod-shaped aerobic bacterium with environmental remediation functions. D. tsuruhatensis strain HA60 was isolated from a commercial nano-particle product, nano-hydroxyapatite. We report that the genome of D. tsuruhatensis strain HA60 has a circular genome of 6,922,195 base pairs with a G+C content of 66.45%.

RESUMO

We report the circularized 6,427,509-bp genome of Pseudomonas aeruginosa isolated from the inner ear of a laboratory mouse (Mus musculus) diagnosed with otitis media. The genome of P. aeruginosa NCTR 501 has a circular chromosome of 6,420,288 bp and two plasmids of 4,042 and 3,179 bp, respectively.

Parabacteroides distasonis is an anaerobic bacterium with ambivalent health effects. P. distasonis strain GP102 was isolated from the cecum content of a morbid pregnant laboratory guinea pig (Cavia porcellus). The genome consists of one circular 5.39-Mbp chromosome with a G + C content of 44.79%.

Clostridium perfringens is a leading cause of human gastroenteritis. The genome sequence of C. perfringens CP201 from an asymptomatic rhesus monkey, Macaca mulatta, consists of one chromosome (3,241,413 bp; 30.22% G + C) and one plasmid (92,983 bp; 28.43% G + C), encoding 2,935 genes, 94 tRNAs, 30 rRNAs, and 1 CRISPR array.

The ROP16 kinase of Toxoplasma gondii is injected into the host cell cytosol where it activates signal transducer and activator of transcription (STAT)-3 and STAT6. Here, we generated a ROP16 deletion mutant on a Type I parasite strain background, as well as a control complementation mutant with restored ROP16 expression. We investigated the biological role of the ROP16 molecule during T. gondii infection. Infection of mouse bone marrow-derived macrophages with rop16-deleted (ΔROP16) parasites resulted in increased amounts of IL-12p40 production relative to the ROP16-positive RH parental strain. High level IL-12p40 production in ΔROP16 infection was dependent on the host cell adaptor molecule MyD88, but surprisingly was independent of any previously recognized T. gondii triggered pathway linking to MyD88 (TLR2, TLR4, TLR9, TLR11, IL-1ß and IL-18). In addition, ROP16 was found to mediate the suppressive effects of Toxoplasma on LPS-induced cytokine synthesis in macrophages and on IFN-γ-induced nitric oxide production by astrocytes and microglial cells. Furthermore, ROP16 triggered synthesis of host cell arginase-1 in a STAT6-dependent manner. In fibroblasts and macrophages, failure to induce arginase-1 by ΔROP16 tachyzoites resulted in resistance to starvation conditions of limiting arginine, an essential amino acid for replication and virulence of this parasite. ΔROP16 tachyzoites that failed to induce host cell arginase-1 displayed increased replication and dissemination during in vivo infection. We conclude that encounter between Toxoplasma ROP16 and the host cell STAT signaling cascade has pleiotropic downstream effects that act in multiple and complex ways to direct the course of infection.

Gordonia alkaliphila is a little known mesophilic Gram-positive rod-shaped bacterium. We report the 3.85-Mbp genome sequence of G. alkaliphila strain WW102, isolated from wastewater at a research center with multiple-species laboratory animal facilities. The genome predicted FadD32 gene clusters that are involved in the biosynthesis of mycolic acids as found in Mycobacterium tuberculosis.

RESUMO

Serotype 3 Streptococcus pneumoniae is associated with major invasive pneumococcal diseases in humans. We report the circularized 2.0-Mbp complete genome sequence of invasive serotype 3 S. pneumoniae strain B1900, untypeable by multilocus sequence typing (MLST). This strain was isolated from the brain of an infant rhesus monkey (Macaca mulatta) that suddenly died of meningitis with no clinical symptoms.

Metabacillus litoralis is part of the newly proposed genus Metabacillus. The bacterium was first isolated from a Yellow Sea tidal flat in 2005. As of May 2022, there are five genomic assemblies deposited in GenBank. We report the 5.2-Mbp genome sequence of M. litoralis strain NCTR108, from commercial tattoo ink.

Terrisporobacter glycolicus is an emerging obligate anaerobic pathogen. We report the 3.9-Mbp genome sequence of T. glycolicus strain WW3900, which was isolated from wastewater at a research center with laboratory animal facilities. The genome sequence predicted a biosynthetic gene cluster encoding an S-adenosylmethionine enzyme and other synthetic genes associated with potential antimicrobial producers.

Ralstonia pickettii is an emerging global opportunistic pathogen. Here, we report the 5.3-Mbp draft genome sequence of R. pickettii NCTR106, isolated from milk carton paperboard obtained from a commercial paper mill. The genome sequence carries two beta-lactamase genes similar to those reported in R. pickettii isolates collected from a hospital.

The purpose of this study was to identify factors affecting medial meniscus extrusion and cartilage degeneration in medial meniscus root tears (MMRT) and to determine the optimal cut-off values for the factors that predict disease severity. We retrospectively evaluated 86 consecutive patients diagnosed with an isolated MMRT by magnetic resonance imaging (MRI) examinations and arthroscopic procedures for 2 years. Patient-specific factors such as age, sex, the time between injury and MRI (mTIME), the time between injury and surgery (sTIME), the time between MRI and surgery (dTIME), Kellgren-Lawrence (KL) grades, and the mechanical tibiofemoral axis angle (mTFA) were documented. Regression analyses and receiver operating characteristic (ROC) curve analyses were performed. The mTIME was only positively correlated with meniscal extrusion (r = 0.425, p < 0.001). The patients who had grades 3 and 4 cartilage lesions had only significantly higher KL grades and longer sTIME than the patients who had lower grades of cartilage lesion (6.5 months [interquartile range (IQ): 3.0-12.0) vs. 2.5 months [IQ: 0.9-3.9]; p < 0.001). The cut-off value of mTIME and sTIME were 2.5 and 6 months. Relapse times greater than 2.5 months and 6 months after a specific event were associated with a 7.8-fold increased risk for meniscus extrusion and an 18.2-fold increased risk for cartilage lesions, respectively. The risk of medial extrusion of the meniscus and the severity of articular cartilage lesions increased with time after an injury. The period of time from 2.5 to 6 months after traumatic event might be a critical window for intervention in the patients with MMRT to perform the repair in the status which the meniscus did not extrude more than 3 mm and the cartilage lesion did not progress more than grade 3. This study is a retrospective and uncontrolled case series and reflects level IV of evidence.
