BACKGROUND: Phenylketonuria (PKU) is an autosomal recessive genetic disease, caused by the phenylalanine hydroxylase (PAH) deficiency in the metabolic pathway, which prevents phenylalanine from being converted into tyrosine, leading to a large amount of phenylalanine discharged from the urine. Therefore, it is necessary to establish a simple, fast, accurate and reliable PKU molecular diagnostic method for clinical diagnosis. METHODS: We established a novel diagnostic method by combining a single-tube multiplex PCR technique with molecular hybridization technique. The method was verified by DNA sequencing technology. The established new technology successfully detected 9 common PAH gene mutations in the Chinese population. RESULTS: Double-blind analysis indicated that the diagnostic accuracy and specificity of the PKU sample were all 100%. Frequencies of single mutation R111X, R176X, Ex6-96A, R241C, R243Q, R252Q, Y356X, V399 V and R413P genotypes were 8, 0.5, 16.5, 1.5, 27, 4.5, 13, 10.5, 8.5% respectively. CONCLUSIONS: The established method of combing single-tube multiplex PCR with molecular hybridization technology can accurately and rapidly detect PAH gene mutations in Chinese and is suitable for screening of large PKU populations with clinical samples.

Inspiration for molecular design and construction can be derived from mathematically based structures. In the quest for new materials, the adaptation of new building blocks can lead to unexpected results. Towards these ends, the quantitative single-step self-assembly of a shape-persistent, Archimedean-based building block, which generates the largest molecular sphere (a cuboctahedron) that has been unequivocally characterized by synchrotron X-ray analysis, is described. The unique properties of this new construct give rise to a dilution-based transformation into two identical spheres (octahedra) each possessing one half of the molecular weight of the parent structure; concentration of this octahedron reconstitutes the original cuboctahedron. These chemical phenomena are reminiscent of biological fission and fusion processes. The large 6 nm cage structure was further analyzed by 1D and 2D NMR spectroscopy, mass spectrometry, and collision cross-section analysis. New routes to molecular encapsulation can be envisioned.

RESUMO

A three-dimensional, highly symmetric, terpyridine-based, spherical complex was synthesized via the coordination of four novel, trisdentate ligands and six Ru(2+) ions, and it exhibits excellent stability over a wide range of pH values (1-14). Structural confirmation was obtained by NMR and ESI-TWIM-MS.

RESUMO

The design and construction of the first multicomponent stepwise assembly of a -based (tpy=terpyridine), three-dimensional, propeller-shaped trismacrocycle, 8, are reported. Key steps in the synthesis involve the preparation of a hexaterpyridinyl triptycene and its reaction with dimeric, 60°-directional, bisterpyridine-Ru(II) building blocks. Characterization includes ESI- and ESI-TWIM-MS and TEM, along with 1D and 2D (1) H NMR spectroscopy.

A series of metal-organic framework {Ln(BCPBA)(H2O)}n {Ln = Nd (1), Sm (2), Eu (3), Tb (4), Dy (5)}; {[Ln(BCPBA)(H2O)](H2O)}n {Ln = Pr (6), Gd (7)} have been synthesized through the hydrothermal synthesis method. These compounds possess non-interpenetrating 3D networks with 10.1438 Å× 17.9149 Å rhombic channels along the [001] direction. The results of temperature-dependent magnetic susceptibility measurements indicate that compounds 4 and 7 exhibit Ln(III)Ln(III) antiferromagnetic interactions, while compound 5 exhibits Ln(III)Ln(III) ferromagnetic interactions. Frequency dependent out-of-phase signals were observed in alternating current (ac) magnetic susceptibility measurements which indicate that they have slow magnetic relaxation characteristics. The luminescent properties of 1, 2, 3, 4, and 5 are also discussed. Due to the good match between the lowest triplet state of the ligand and the resonant energy level of the lanthanide ion, compound 4 has longer fluorescence lifetime (τ1 = 400.0000 ms, τ2 = 1143.469 ms) and higher quantum yield (Φ = 42%) compared with other compounds.
