RESUMO

Environmental exposure to active pharmaceutical ingredients (APIs) can have negative effects on the health of ecosystems and humans. While numerous studies have monitored APIs in rivers, these employ different analytical methods, measure different APIs, and have ignored many of the countries of the world. This makes it difficult to quantify the scale of the problem from a global perspective. Furthermore, comparison of the existing data, generated for different studies/regions/continents, is challenging due to the vast differences between the analytical methodologies employed. Here, we present a global-scale study of API pollution in 258 of the world's rivers, representing the environmental influence of 471.4 million people across 137 geographic regions. Samples were obtained from 1,052 locations in 104 countries (representing all continents and 36 countries not previously studied for API contamination) and analyzed for 61 APIs. Highest cumulative API concentrations were observed in sub-Saharan Africa, south Asia, and South America. The most contaminated sites were in low- to middle-income countries and were associated with areas with poor wastewater and waste management infrastructure and pharmaceutical manufacturing. The most frequently detected APIs were carbamazepine, metformin, and caffeine (a compound also arising from lifestyle use), which were detected at over half of the sites monitored. Concentrations of at least one API at 25.7% of the sampling sites were greater than concentrations considered safe for aquatic organisms, or which are of concern in terms of selection for antimicrobial resistance. Therefore, pharmaceutical pollution poses a global threat to environmental and human health, as well as to delivery of the United Nations Sustainable Development Goals.

The occurrence and dissemination of antibiotic resistant genes (ARGs) that are associated with clinical pathogens and the evaluation of associated risks are still under-investigated in developing countries under tropical conditions. In this context, cultivable and molecular approaches were performed to assess the dissemination of bacteria and the antibiotic resistance genes in aquatic environment in Kinshasa, Democratic Republic of the Congo. Cultivable approach quantified ß-lactam, carbapenem resistant, and total Escherichia coli and Enterobacteriaceae in river sediments and surface waters that receive raw hospital effluents. The molecular approach utilized Quantitative Polymerase Chain Reaction (qPCR) to quantify the total bacteria and the richness of relevant bacteria (Escherichia coli, Enterococcus, and Pseudomonas), and antibiotic resistance genes (ARGs: blaOXA-48, blaCTX-M, blaIMP, blaTEM) in sediment samples. Statistical analysis were employed to highlight the significance of hospital contribution and seasonal variation of bacteria and ARGs into aquatic ecosystems in suburban municipalities of Kinshasa, Democratic Republic of the Congo. The contribution of hospitals to antibiotic resistance proliferation is higher in the dry season than during the wet season (p < 0.05). Hospital similarly contributed Escherichia coli, Enterococcus, and Pseudomonas and ARGs significantly to the sediments in both seasons (p < 0.05). The organic matter content correlated positively with E. coli (r = 0.50, p < 0.05). The total bacterial load correlated with Enterococcus, and Pseudomonas (0.49 < r < 0.69, p < 0.05). Each ARG correlated with the total bacterial load or at least one relevant bacteria (0.41 < r < 0.81, p < 0.05). Our findings confirm that hospital wastewaters contributed significantly to antibiotic resistance profile and the significance of this contribution increased in the dry season. Moreover, our analysis highlights this risk from untreated hospital wastewaters in developing countries, which presents a great threat to public health.

An epidemiological survey conducted among users of water points and medical institutions in the N'djili Kilambu neighborhood of Kinshasa in Democratic Republic of the Congo, indicates that waterborne diseases have already affected more than 60% of the patients admitted to local clinics between 2013 and 2017. In order to raise public and political awareness about this hazardous health issue resulting from the lack of safely managed sanitation systems, this study investigates the microbial quality of drinking water from local water resources. Water samples were collected from nine wells and streams used as drinking sources, and analyzed for Fecal Indicator Bacteria (FIB), including Escherichia coli, Enterococcus, and Total Coliforms. Physicochemical parameters (pH, electrical conductivity, O2, and soluble ions (Na+, K+, PO43-, SO42-, NO3-, NO2-) were also analyzed. Except for NO3- and NO2-, the average concentrations of the physicochemical parameters and dissolved ions generally meet the guidelines for drinking/domestic water quality. By contrast, the results reveal high levels of FIB in the water samples collected during both dry and wet seasons. The contamination is significantly higher during the wet season compared to dry season, due to increased runoff, open defecation practices, and more frequent overflow of onsite sanitation systems and septic tanks.

Although the United Nations General Assembly recognized in 2010 the right to safe and clean drinking water and sanitation as a human right that is essential to the full enjoyment of life and all other human rights, the contamination of water supplies with faecal pathogens is still a major and unsolved problem in many parts of the world. In this study, faecal indicator bacteria (FIB), including Escherichia coli (E. coli) and Enterococcus (ENT), were quantified over the period of June/July 2014 and June/July 2015 to assess the quality of hospital effluents (n = 3: H1, H2 and H3) and of rivers receiving wastewaters from the city of Kinshasa, Democratic Republic of Congo. The water and sediment samples from the river-receiving systems were collected in, upstream and downstream of the hospital outlet pipe (HOP) discharge. The analysis of E. coli and ENT in water and sediment suspension was performed using the cultural membrane filter method. The FIB characterization was performed for general E. coli, Enterococcus faecalis(E. faecalis) and human-specific Bacteroides by PCR using specific primers. The results revealed very high FIB concentration in the hospital effluent waters, with E. coli reaching the values of 4.2 × 10(5), 16.1 × 10(5) and 5.9 × 10(5) CFU 100 mL(-1), for the hospital effluents from H1, H2, and H3, respectively; and Enterococcus reaching the values of 2.3 × 10(4), 10.9 × 10(4) and 4.1 × 10(4) CFU 100 mL(-1), respectively. Interestingly, the FIB levels in the water and sediment samples from river-receiving systems are spatially and temporally highly variable and present in some samples with higher values than the hospital effluents. The PCR assays for human-specific Bacteroides HF183/HF134 further indicate that more than 98% of bacteria were from human origin. The results of this research therefore confirm the hypothesis of our previous studies, indicating that in developing countries (e.g., Democratic Republic of Congo and South India), the hospital effluent waters can be a significant source of the deterioration of the bacteriological quality for urban rivers. The approach used in this investigation can be further used to decipher the pollution of water resources by human faecal contamination. The results of this research will help to better understand the microbiological pollution problems in river-receiving systems and will guide municipality decisions on improving the urban water quality.

Wastewater treatment plants (WWTP) receive the effluents from various sources (communities, industrial, and hospital effluents) and are recognized as reservoir for antibiotic-resistance genes (ARGs) that are associated with clinical pathogens. The aquatic environment is considered a hot-spot for horizontal gene transfer, and lake sediments offer the opportunity for reconstructing the pollution history and evaluating the impacts. In this context, variation with depth and time of the total bacterial load, the abundance of faecal indicator bacteria (FIB; E. coli and Enterococcus spp. (ENT)), Pseudomonas spp., and ARGs (blaTEM, blaSHV, blaCTX-M, blaNDM, and aadA) were quantified in sediment profiles of different parts of Lake Geneva using quantitative PCR. The abundance of bacterial marker genes was identified in sediments contaminated by WWTP following eutrophication of the lake. Additionally, ARGs, including the extended-spectrum ß-lactam- and aminoglycoside-resistance genes, were identified in the surface sediments. The ARG and FIB abundance strongly correlated (r ≥ 0.403, p < 0.05, n = 34) with organic matter and metal concentrations in the sediments, indicating a common and contemporary source of contamination. The contamination of sediments by untreated or partially treated effluent water can affect the quality of ecosystem. Therefore, the reduction of contaminants from the source is recommended for further improvement of water quality.

This study was conducted to assess potential human health risks presented by pathogenic bacteria in a protected multi-use lake-reservoir (Lake Ma Vallée) located in west of Kinshasa, Democratic Republic of Congo (DRC). Water and surface sediments from several points of the Lake were collected during summer. Microbial analysis was performed for Escherichia coli, Enterococcus (ENT), Pseudomonas species and heterotrophic plate counts. PCR amplification was performed for the confirmation of E. coli, ENT, Pseudomonas spp. and Pseudomonas aeruginosa isolated from samples. The results reveal low concentration of bacteria in water column of the lake, the bacterial quantification results observed in this study for the water column were below the recommended limits, according to WHO and the European Directive 2006/7/CE, for bathing water. However, high concentration of bacteria was observed in the sediment samples; the values of 2.65 × 10(3), 6.35 × 10(3), 3.27 × 10(3) and 3.60 × 10(8) CFU g(-1) of dry sediment for E. coli, ENT, Pseudomonas spp. and heterotrophic plate counts, respectively. The results of this study indicate that sediments of the Lake Ma Vallée can constitute a reservoir of pathogenic microorganisms which can persist in the lake. Possible resuspension of faecal indicator bacteria and pathogens would affect water quality and may increase health risks to the population during recreational activities. Our results indicate that the microbial sediment analysis provides complementary and important information for assessing sanitary quality of surface water under tropical conditions.

An adventive population of the exotic parasitoid wasp, Trissolcus japonicus (Ashmead) (Hymenoptera: Scelionidae), discovered in Michigan in 2018, is a promising biological control agent of the invasive Halyomorpha halys (Stål) (Hemiptera: Pentatomidae). Following its discovery, field releases of Tr. japonicus were conducted over 2 yr in southern Michigan, to test how release size or release frequency impacts establishment. Sentinel eggs of H. halys and of the native Podisus maculiventris (Say) (Hemiptera: Pentatomidae) were used alongside yellow sticky cards to monitor parasitoids. In 2019 and 2020, 7,200 Tr. japonicus were released at 16 sites. Monitoring between 2019 and 2021 yielded only 49 individuals. The captures suggest reproductive activity and overwintering success in the field but do not allow for evaluation of best release methods. Parasitism by native parasitoids was below 7%, which is similar to other states and unlikely to provide sufficient control of H. halys. The placement of sentinel eggs or sticky traps either in the lower or middle canopy of trees did not influence parasitoid capture rates. Frozen and fresh H. halys sentinel eggs were attacked at the same rate, but more native parasitoids emerged from frozen eggs. We did not find signs of nontarget effects on P. maculiventris thus parasitism rates overall were very low. These results could indicate dispersal of Tr. japonicus from the release sites or slow population growth. The latter may be due to the relatively low densities of H. halys in Michigan or may stem from the small founding size of our laboratory colony.

This study characterized the fecal indicator bacteria (FIB), including Escherichia coli (E. coli) and Enteroccocus (ENT), disseminated over time in the Bay of Vidy, which is the most contaminated area of Lake Geneva. Sediments were collected from a site located at ∼500 m from the present waste water treatment plant (WWTP) outlet pipe, in front of the former WWTP outlet pipe, which was located at only 300 m from the coastal recreational area (before 2001). E. coli and ENT were enumerated in sediment suspension using the membrane filter method. The FIB characterization was performed for human Enterococcus faecalis (E. faecalis) and Enterococcus faecium (E. faecium) and human specific bacteroides by PCR using specific primers and a matrix-assisted laser desorption ionization-time of flight mass spectrometry (MALDI-TOF MS). Bacterial cultures revealed that maximum values of 35.2 × 10(8) and 6.6 × 10(6)CFU g(-1) dry sediment for E. coli and ENT, respectively, were found in the sediments deposited following eutrophication of Lake Geneva in the 1970s, whereas the WWTP started operating in 1964. The same tendency was observed for the presence of human fecal pollution: the percentage of PCR amplification with primers ESP-1/ESP-2 for E. faecalis and E. faecium indicated that more than 90% of these bacteria were from human origin. Interestingly, the PCR assays for specific-human bacteroides HF183/HF134 were positive for DNA extracted from all isolated strains of sediment surrounding WWPT outlet pipe discharge. The MALDI-TOF MS confirmed the presence of general E. coli and predominance E. faecium in isolated strains. Our results demonstrated that human fecal bacteria highly increased in the sediments contaminated with WWTP effluent following the eutrophication of Lake Geneva. Additionally, other FIB cultivable strains from animals or adapted environmental strains were detected in the sediment of the bay. The approaches used in this research are valuable to assess the temporal distribution and the source of the human fecal pollution in aquatic environments.

The co-occurrence of heavy metals, antibiotic-resistant bacteria (ARB) and antibiotic-resistance genes (ARGs) from hospital effluents spreading into the river receiving systems and evaluating associated risks are topics of scientific interest and still under-studied in developing countries under tropical conditions. To understand the selectors of the ARGs, we examined the occurrence of heavy metals (Cr, Co, Ni, Cu, Cd, Pb and Zn), associated ARB (ß-lactam-resistant Escherichia coli, ß-lactam-resistant Enterobacteriaceae, and carbapenem-resistant Enterobacteriaceae) and ARGs (blaOXA, blaCTX-M, blaIMP, blaTEM) in water and sediments from two sub-urban rivers receiving urban and hospital effluent waters in the Democratic Republic of the Congo (DRC). High abundances of ARB and ARGs were observed in all sediment samples. All the metal contents correlated negatively with grain size (- 0.94 ≤ r ≤ - 0.54, p < 0.05) except for Ni and positively with organic matter content and total copies of 16 s rRNA (0.42 ≤ r ≤ 0.79, p < 0.05), except for Ni and Zn. The metals had a significant positive correlation with the faecal indicator Enterococcus except for Ni and Cd (0.43 ≤ r ≤ 0.67, p < 0.05). Carbapenem-resistant Enterobacteriaceae correlated negatively with Zn (r = - 0.44, p < 0.05) and positively with all the rest of toxic metals (0.58 ≤ r ≤ 1.0, p < 0.05). These results suggested that some metals had a great influence on the persistence of ARB and ARGs in sediments. Overall, this study strongly recommends the managing urban wastewater to preserve water resources used for human and agricultural purposes. Additionally, we recommend the utilizing biological indicators (faecal indicator bacteria, ARB, ARGs) when investigating urban wastewater pollutions.

In many suburban municipalities of developing countries, the household drinking water comes mainly from groundwater including, wells, streams and springs. These sources are vulnerable because poor hygienic conditions and sanitation prevail causing persistence and recurrent waterborne diseases. In this research, a survey study on water resource use and an epidemiological survey of waterborne diseases were conducted among users of water points and medical institutions in suburban communes of Selembao and Kimbanseke (Kinshasa, the Democratic Republic of the Congo). In addition, physicochemical (temperature, pH, O2, electrical conductivity, and soluble ions: Na+, K+, PO4 3-, SO4 2-, NO3 -, NO2 -) and bacteriological (FIB: faecal indicator bacteria) analyses of water from 21 wells and springs were performed according to the seasonal variations. FIB included Escherichia coli (E. coli), Enterococcus and Total Coliforms. The survey results indicate that more than 75% of the patients admitted to local medical institutions between 2016 and 2019 are affected by waterborne diseases, including typhoid fever, amoebic dysentery, diarrhoea, gastroenteritis disorders and cholera. Except for NO3 - in some sites, the water physicochemical parameter values are within WHO permissible limits for drinking/domestic water quality. On the contrary, the results revealed high FIB levels in water from unmanaged wells and springs during rainy and dry seasons. The microbiological pollution was significantly higher in the rainy season compared to the dry season. Interestingly, no FIB contamination was observed in water samples from managed/developed wells. The results from this study will guide local government decisions on improving water quality to prevent recurrent waterborne diseases.

Understanding the prevalence and polymorphism of antibiotic resistance genes in soil bacteria and their potential to be transferred horizontally is required to evaluate the likelihood and ecological (and possibly clinical) consequences of the transfer of these genes from transgenic plants to soil bacteria. In this study, we combined culture-dependent and -independent approaches to study the prevalence and diversity of bla genes in soil bacteria and the potential impact that a 10-successive-year culture of the transgenic Bt176 corn, which has a blaTEM marker gene, could have had on the soil bacterial community. The bla gene encoding resistance to ampicillin belongs to the beta-lactam antibiotic family, which is widely used in medicine but is readily compromised by bacterial antibiotic resistance. Our results indicate that soil bacteria are naturally resistant to a broad spectrum of beta-lactam antibiotics, including the third cephalosporin generation, which has a slightly stronger discriminating effect on soil isolates than other cephalosporins. These high resistance levels for a wide range of antibiotics are partly due to the polymorphism of bla genes, which occur frequently among soil bacteria. The blaTEM116 gene of the transgenic corn Bt176 investigated here is among those frequently found, thus reducing any risk of introducing a new bacterial resistance trait from the transgenic material. In addition, no significant differences were observed in bacterial antibiotic-resistance levels between transgenic and nontransgenic corn fields, although the bacterial populations were different.

In Lake Geneva, Switzerland, the most Hg-contaminated sediments have been found in the Vidy Bay where high Hg contents largely exceeds the background levels of Lake Geneva sediments. This contamination has been attributed to the discharge of a waste water treatment plant (WWTP). Total Hg (THg) and monomethylmercury (MMHg) were determined in bulk sediment and in three different grain size fractions (i: clay and silt, ii: fine-coarse sand, iii: and very coarse sand and gravel) collected close to the outlet pipe of a WWTP in order to verify whether the standardized procedures of sediment treatment is adequate for this setting and, by extension, for similar contaminated sites. THg was homogeneously distributed in the different grain size fractions and was correlated to organic matter content (R(2) = 0.6). MMHg was homogeneously distributed in the two finer grain fractions (Φ < 0.063 mm; 0.063 mm < Φ <1 mm). The results of this study suggested that the analysis of the bulk sediment seems to be more appropriate for the assessment of the content and spatial distribution of Hg in freshwater sediments contaminated by WWTPs.

The contamination of water resource and food chain by persistent organic pollutants (POPs) constitutes a major environmental and human health concern worldwide. The aim of this study was to investigate the levels of POPs in irrigation water, soil and in Amaranthus viridis (A. viridis) from different gardening sites in Kinshasa to evaluate the potential environmental and human health risks. A survey study for the use of pesticides and fertilizers was carried out with 740 market gardeners. The levels of POPs (including organochlorine pesticides (OCPs), polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs), and polycyclic aromatic hydrocarbons (PAHs)) were analyzed in irrigation water and 144 vegetable samples collected from different gardening sites. The assessment of potential human health risk was estimated by calculating daily intake and toxic equivalency to quantify the carcinogenicity. The results show highest PAH levels in A. viridis from all studied sites. The concentrations of the sum of seven PCBs (Σ7PCBS) congeners in analyzed plants ranged between 15.89 and 401.36 ng g-1. The distributions of OCPs in both water and A. viridis were congener specific, chlorpyrifos-ethyl and p,p'-DDE were predominantly detected. Among PBDEs, only BDE47 was quantified with noticeable concentration in A. viridis, while no PBDEs were detected in irrigation water. Higher estimated daily intake values indicate that consuming leafy vegetables might associate with increased human health risks. However, calculated incremental lifetime cancer risk values indicates no potential carcinogenic risk for the local population. The results of this study provide important information on A. viridis contamination by POPs and strongly recommend implementing the appropriate measures to control the use of chemicals used in studied gardening areas. Thus in Kinshasa, urban agriculture control programs for POPs and fertilizers is very important in order to protect the public health through direct and dietary exposure pathways.

In situ microcosm study investigated both the kinetics of plant material mass loss and qualitative and quantitative aspects of DNA content by researching leaf degradation of two specific varieties of tomato (Admiro and Palmiro) in freshwater column incubated for 40 days. A two-compartment first order model fitted both tomato dry matter and DNA content mass loss well. The composite half-decrease times were, respectively, 1.13 ± 0.51 and 1.16 ± 0.47 days for Palmiro and Admiro. The composite half-disappearance times of total DNA in Palmiro and Admiro tomato leaves were, respectively, 0.92 ± 0.31 and 0.88 ± 0.26 days. Genomic analysis indicates that before having been released, a significant amount of DNA may be degraded in plant tissues decomposing in water column. The results of this study confirm the hypothesis that release of plant DNA in aquatic environments can be caused by intracellular nuclease activities in the plants cells and by enzymatic degradation of cell structures by residual microbial activities in leaves.

Unsaturated soil columns were used to examine the transport of the plasmid pLEPO1 and plant DNA (transplastomic tobacco DNA), both carrying an antibiotic resistance gene (aadA gene), and the capacity of bacteria to incorporate the gene in their genome after its passage through the soil. Soil columns containing a top leaf layer had sterile water percolated through them at a rate of 0.5mLh(-1). DNA from column leachate water was extracted and analyzed. Quantitative measurements included total DNA concentrations in the water and the transformation frequencies of Acinetobacter sp. BD413 by DNA in the column effluent. Qualitative measurements included the relative degradation of DNA after passage in the columns by agarose gel electrophoresis and the potential of effluent DNA to transform bacteria, leading to the production of antibiotic-resistant bacteria. The presence of aadA gene in the leachate water of soil columns suggests the mobility of DNA in unsaturated soil medium. The extent of DNA degradation was found to be proportional to its residence time in the soil column while a fraction of DNA was always able to incorporate into the Acinetobacter genome under all conditions studied. These results suggest that biologically active transgenic DNA might be transported downward by rain in unsaturated soils.

RESUMO

Over the past decades, the rising antibiotic resistance bacteria (ARB) are continuing to emerge as a global threat due to potential public health risk. Rapidly evolving antibiotic resistance and its persistence in the environment, have underpinned the need for more studies to identify the possible sources and limit the spread. In this context, not commonly studied and a neglected genetic material called extracellular DNA (eDNA) is gaining increased attention as it can be one of the significant drivers for transmission of extracellular ARGS (eARGs) via horizontal gene transfer (HGT) to competent environmental bacteria and diverse sources of antibiotic-resistance genes (ARGs) in the environment. Consequently, this review highlights the studies that address the environmental occurrence of eDNA and encoding eARGs and its impact on the environmental resistome. In this review, we also brief the recent dedicated technological advancements that are accelerating extraction of eDNA and the efficiency of treatment technologies in reducing eDNA that focuses on environmental antibiotic resistance and potential ecological health risk.

The occurrence and dissemination of toxic metals, antibiotic resistant bacteria and their resistance genes (ARGs) in the aquatic ecosystems of sub-Saharan African countries are still understudied, despite their potential to threat human health and aquatic organisms. In this context, the co-contamination and seasonal distribution of toxic metals and ARG in river sediments receiving untreated urban sewages and hospital effluents from Kinshasa, the capital city of the Democratic Republic of the Congo were investigated. ARGs including ß-lactam resistance (blaCTX-M and blaSHV), carbapenem resistance (blaVIM, blaIMP, blaKPC, blaOXA-48 and blaNDM) and total bacterial load were quantified by using quantitative polymerase chain reaction (qPCR) in total DNA extracted from sediment. The amount of toxic metals in sediments was quantified using Inductively Coupled Plasma Mass Spectrometry (ICP-MS). The results highlight high abundance of 16S rRNA and ARGs copy numbers in sediment samples. Strong pollution of rivers by toxic metals was found, with max values (mg kg-1) of 81.85(Cr), 5.09(Co), 33.84(Ni), 203.46 (Cu), 1055.92(Zn), 324.24(Pb) and 2.96(Hg). Results also highlight the high abundance of bacterial markers (8.06 × 109-2.42 × 1012 16S rRNA/g-1 DS) as well as antibiotic resistance genes (up to 4.58 × 108 ARG. g-1 DS) in the studied rivers. Significant correlations were observed between (i) metals (except Cd and Hg) and organic matter (R > 0.60, p < 0.05); and (ii) ARGs (except blaNDM) and 16S rRNA (R > 0.57, p < 0.05) suggesting a tight link between (i) metal contamination and anthropogenic pressure and (ii) microbial contamination of river and dissemination of antibiotic resistance. Results demonstrated that multi-diffuse pollution originating from human activity contribute to the spread of toxic metals and ARGs into the aquatic ecosystems.

The pollution assessment and the evaluation of potential risks in the Atlantic Coastal Region of the Democratic Republic of the Congo are still very limited. Consequently, the present study investigates for the first time the concentrations of heavy metals and persistent organic pollutants (organochlorine pesticides (OCPs), polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs), and polycyclic aromatic hydrocarbons (PAHs)) in river, estuary, and marine sediments from this area. The results highlighted high concentrations of Cr, Zn, As, and Pb exceeding the probable effect level (PEL) on aquatic life. Zn was the most dominant element detected at a range of 180-480 mg kg-1 in marine sediment, 132-382 mg kg-1 in estuary sediment, and 121-687 mg kg-1 in river sediment. Total PCBs (∑7 × 4.3) ranged from 1995 to 20,156 µg kg-1, 2013-12,058 µg kg-1, and 1861-36,417 µg kg-1 in marine, estuary, and river sediments, respectively. Total PCBs (∑7 × 4.3) were above PEL for all sediments, suggesting potential adverse effects on benthic organisms. The OCP, PBDE, and PAH levels were low to moderate for all sediments. Taking into consideration, the concentrations of Zn, Pb, PCBs, and DDTs, probable environmental risks, are present.

This work constitutes a part of a wider study examining the degradation and release of plant DNA into the environment. Microcosm studies investigated the kinetics of leaf and DNA content degradation in a specific variety of tomato (Admiro) after incubation in sediments over 30 days at 20, 10, and 4 degrees C. Temperature and microorganisms have been found to play a key role in the decomposition of plant material in freshwater sediment. A two-compartment first-order function fitted well both tomato leaf matter degradation and DNA content mass loss. Genomic analysis indicated that before having been released, an important part of DNA may be degraded inside plant tissues during decomposition in sediments. PCR amplification demonstrated that, after having been released, DNA can both be rapidly adsorbed onto sediment particles and persist as dissolved extracellular DNA in the water column.

The origin and distribution of microbial contamination in Lake Geneva's most polluted bay were assessed using faecal indicator bacteria (FIB). The lake is used as drinking water, for recreation and fishing. During 1 year, water samples were taken at 23 points in the bay and three contamination sources: a wastewater treatment plant (WWTP), a river and a storm water outlet. Analyses included Escherichia coli, enterococci (ENT), total coliforms (TC), and heterotrophic plate counts (HPC). E. coli input flux rates from the WWTP can reach 2.5 x 10(10) CFU/s; those from the river are one to three orders of magnitude lower. Different pathogenic Salmonella serotypes were identified in water from these sources. FIB levels in the bay are highly variable. Results demonstrate that (1) the WWTP outlet at 30 m depth impacts near-surface water quality during holomixis in winter; (2) when the lake is stratified, the effluent water is generally trapped below the thermocline; (3) during major floods, upwelling across the thermocline may occur; (4) the river permanently contributes to contamination, mainly near the river mouth and during floods, when the storm water outlet contributes additionally; (5) the lowest FIB levels in the near-surface water occur during low-flow periods in the bathing season.
